Abstract:The effect of regulatory system on the engineered biosynthetic pathway in chassis cells remains incompletely understood in microorganisms. Acyl-CoAs function as key precursors for the biosynthesis of various natural products and the dominant donors for protein acylation. The polyphenol pinosylvin, with high antimicrobial and antifungal activities, is biosynthesized with malonyl-CoA as its direct precursors. But correlation between lysine malonylation and pinosylvin biosynthesis remains unknown. Herein, we found that the malonyl-CoA-driven lysine malonylation plays an important role in interaction between the engineered pathway of pinosylvin synthesis and E. coli chassis cell. Oversupply of malonyl-CoA leads to an increase in malonylation level of global proteome as well as the enzymes in the artificial pathway, thereby decreasing yield of pinosylvin. The results revealed that the intricate balance of cellular acyl-CoA concentrations is critical for the yields of acyl-CoA-derived natural products. We next modified the enzymes in the biosynthetic pathway to adjust their acylation level and successfully improved the yield of pinosylvin. Our study uncovers the effect of protein acylation on the biosynthetic pathway, helps optimization of synthetic constructs, and provides new strategies in metabolic engineering and synthetic biology at the protein post-translational level.

Abstract:Adipic acid (AA) is a key industrial dicarboxylic acid intermediate used in nylon manufacturing. Unfortunately, the traditional process technology is accompanied by serious environmental pollution. Given the growing demand for adipic acid and the desire to reduce its negative impact on the environment, considerable efforts have been devoted to developing more green and friendly routes. This Review is focused on the latest advances in the sustainable preparation of AA from biomass-based platform molecules, including 5-hydroxymethylfufural, glucose, γ-valerolactone, and phenolic compounds, through biocatalysis, chemocatalysis, and the combination of both. Additionally, the development of state-of-the-art catalysts for different catalytic systems systematically is discussed and summarized, as well as their reaction mechanisms. Finally, the prospects for all preparation routes are critically evaluated and key technical challenges in the development of green and sustainable processes for the manufacture of AA are highlighted. It is hoped that the green adipic acid synthesis pathways presented can provide insights and guidance for further research into other industrial processes for the production of nylon precursors in the future.

Abstract:Adipic Acid (AA) is a valued platform chemical compound, which can be used as a precursor of nylon-6,6. Due to the generation of an enormous amount of nitric oxide metabolites and the growing depletion of oil resources as a result of AA production from a mixture of cyclohexanol and cyclohexanone, the microbial methods for synthesizing AA have attracted significant attention. Of the several AA-producing pathways, the reverse adipate degradation pathway in Thermobifida fusca (Tfu RADP) is reported to be the most efficient, which has been confirmed in Escherichia coli. In this study, the heterologous Tfu RADP was constructed for producing AA in S. cerevisiae by co-expressing genes of Tfu_0875, Tfu_2399, Tfu_0067, Tfu_1647, Tfu_2576, and Tfu_2576. The AA titer combined with biomass, cofactors and other by-products was all determined after fermentation. During batch fermentation in a shake flask, the maximum AA titer was 3.83 mg/L, while the titer increased to 10.09 mg/L during fed-batch fermentation in a 5-L bioreactor after fermentation modification.

Abstract:Adipic acid is one of the most important industrial dicarboxylic acids and is used mainly as a precursor to nylon‐6,6. Currently, commercial adipic acid is produced primarily from benzene by a chemical route that is associated with environmental, health, and safety concerns. Herein, we report a new process to produce adipic acid from an inexpensive renewable feedstock, sugar beet residue by combining an engineered Escherichia coli strain and Re‐based chemical catalysts. The engineered E. coli converted d‐galacturonic acid to mucic acid, which was precipitated easily with acid, and the mucic acid was further converted to adipic acid by a deoxydehydration reaction catalyzed by an oxorhenium complex followed by a Pt/C‐catalyzed hydrogenation reaction under mild conditions. A high selectivity to the free acid products was achieved by tuning the acidity of the Re‐based catalysts. Finally, adipic acid was produced directly from sugar beet residue that was hydrolyzed enzymatically with engineered E. coli and two chemical catalysts in a yield of 8.4 %, which signifies a new route for the production of adipic acid.

Abstract:The pursuit of a greener adipic acid synthesis process, aiming to replace the highly polluting nitric acid oxidation method, has been a longstanding endeavor in both academic and industrial spheres. In this study, we present a reaction-controlled Pickering interfacial catalysis system, enabling the efficient oxidation of cyclohexene to adipic acid using 30 wt% hydrogen peroxide without the involvement of organic solvents. Following the reaction, the system spontaneously undergoes demulsification, facilitating easy recovery of catalyst particles via simple filtration, followed by product separation through cooling-induced crystallization. Employing high speed microscopic imaging for in-situ characterization of interfacial properties, we elucidate the influence of reaction conditions on interfacial properties and catalytic performance. The reaction-controlled Pickering interfacial catalysis system demonstrated herein offers a novel and industrially feasible strategy for developing green adipic acid synthesis route.

Abstract:For decades, plants have been the subject of genetic engineering to synthesize novel, value-added compounds. Polyhydroxyalkanoates (PHAs), a large class of biodegradable biopolymers naturally synthesized in eubacteria, are among the novel products that have been introduced to make use of plant acetyl-CoA metabolic pathways. It was hoped that renewable PHA production would help address environmental issues associated with the accumulation of nondegradable plastic wastes. However, after three decades of effort synthesizing PHAs, and in particular the simplest form polyhydroxybutyrate (PHB), and seeking to improve their production in plants, it has proven very difficult to reach a commercially profitable rate in a normally growing plant. This seems to be due to the growth defects associated with PHA production and accumulation in plant cells. Here, we review major breakthroughs that have been made in plant-based PHA synthesis using traditional genetic engineering approaches and discuss challenges that have been encountered. Then, from the point of view of plant synthetic biology, we provide perspectives on reprograming plant acetyl-CoA pathways for PHA production, with the goal of maximizing PHA yield while minimizing growth inhibition. Specifically, we suggest genetic elements that can be considered in genetic circuit design, approaches for nuclear genome and plastome modification, and the use of multiomics and mathematical modeling in understanding and restructuring plant metabolic pathways.

Abstract:The linear C6 dicarboxylic acid adipic acid is an important bulk chemical in the petrochemical industry as precursor of the polymer nylon-6,6-polyamide. In recent years, efforts were made towards the biotechnological production of adipate from renewable carbon sources using microbial cells. One strategy is to produce adipate via a reversed β-oxidation pathway. Hitherto, the adipate titers were very low due to limiting enzyme activities for this pathway. In most cases, the CoA intermediates are non-natural substrates for the tested enzymes and were therefore barely converted. We here tested heterologous enzymes in Escherichia coli to overcome these limitations and to improve the production of adipate via a reverse β-oxidation pathway. We tested in vitro selected enzymes for the efficient reduction of the enoyl-CoA and in the final reaction for the thioester cleavage. The genes encoding the enzymes which showed in vitro the highest activity were then used to construct an expression plasmid for a synthetic adipate pathway. Expression of paaJ, paaH, paaF, dcaA, and tesB in E. coli BL21(DE3) resulted in the production of up to 36 mg/L of adipate after 30 h of cultivation. Beside the activities of the pathway enzymes, the availability of metabolic precursors may limit the synthesis of adipate, providing another key target for further strain engineering towards high-yield production of adipate with E. coli.

Abstract:Biomass-derived furans offer sustainable routes to adipic acid (AA), a key chemical in Nylon-6,6 synthesis. In this work, we show that tetrahydrofuran-2,5-dicarboxylic acid (THFDCA) is a viable precursor for AA production, achieving up to 89% yield in a metal-free system containing HI and molecular H-2 in a propionic acid solvent at 160 degrees C. Reactivity studies demonstrate that the interplay between HI, H-2, and the solvent is essential for effective THFDCA ring opening. By measuring the reaction orders of HI and molecular H-2 and calculating an acid-base equilibrium constant in a nonaqueous solvent, we show that HI plays a multifaceted role in the reaction by acting both as a proton source and an iodide source to selectively cleave C-O bonds without overhydrogenation of carboxylic acid groups. Using reactivity studies, kinetic measurements, and first-principles computational insights, we demonstrate that metal-free activation of molecular H-2 plays a key role in the reaction, following HI mediated cleavage of the etheric C-O bond in THFDCA.

Abstract:Fumaric acid is an industrially important platform chemical, which has been widely used in food, chemicals, agriculture and pharmaceutical industries. Due to the rising price of raw materials in traditional petrochemical method and demand for sustainable development, fermentative fumaric acid production has attracted great attention. In this context, various strains with enhanced fumaric acid production or elimination of by-products were developed through mutagenesis or metabolic engineering. In addition, renewable organic wastes together with the fermentation process optimization have also been widely investigated. Accordingly, this review will comprehensively summarize the achievements of fermentative fumaric acid production in recent years and major obstacles occurring in industrialization to provide insights and perspective for future microbial fermentation of fumaric acid.

Abstract:The biobased production of adipic acid, a precursor in the production of nylon, is of great interest in order to replace the current petrochemical production route. Glucose-rich lignocellulosic raw materials have high potential to replace the petrochemical raw material. A number of metabolic pathways have been proposed for the microbial conversion of glucose to adipic acid, but achieved yields and titers remain to be improved before industrial applications are feasible. One proposed pathway starts with lysine, an essential metabolite industrially produced from glucose by microorganisms. However, the drawback of this pathway is that several reactions are involved where there is no known efficient enzyme. By changing the order of the enzymatic reactions, we were able to identify an alternative pathway with one unknown enzyme less compared to the original pathway. One of the reactions lacking known enzymes is the reduction of the unsaturated α,β bond of 6-amino-trans-2-hexenoic acid and trans-2-hexenedioic acid. To identify the necessary enzymes, we selected N-ethylmaleimide reductase from Escherichia coli and Old Yellow Enzyme 1 from Saccharomyces pastorianus. Despite successful in silico docking studies, where both target substrates could fit in the enzyme pockets, and hydrogen bonds with catalytic residues of both enzymes were predicted, no in vitro activity was observed. We hypothesize that the lack of activity is due to a difference in electron withdrawing potential between the naturally reduced aldehyde and the carboxylate groups of our target substrates. Suggestions for protein engineering to induce the reactions are discussed, as well as the advantages and disadvantages of the two metabolic pathways from lysine. We have highlighted bottlenecks associated with the lysine pathways, and proposed ways of addressing them.

Abstract:Petrochemical based polymers, paints and coatings are cornerstones of modern industry but our future sustainable society demands greener processes and renewable feedstock materials. A challenge is to access platform monomers from biomass resources while integrating the principles of green chemistry in their chemical synthesis. We present a synthesis route starting from biomass-derived furfural towards the commonly used monomers maleic anhydride and acrylic acid, implementing environmentally benign photooxygenation, aerobic oxidation and ethenolysis reactions. Maleic anhydride and acrylic acid, transformed into sodium acrylate, were isolated in yields of 85 % (2 steps) and 81 % (4 steps), respectively. With minimal waste and high atom efficiency, this biobased route provides a viable alternative to access key monomers.

Abstract:As high-performance monomers for the manufacture of polyamide materials, mid- and long-chain dicarboxylic acids (DCAi, i ≥ 6) have received extensive attention from researchers. Biosynthesis is gradually replacing chemical synthesis due to its outstanding advantages in the industrial production of mid- and long-chain dicarboxylic acids, which is mostly achieved by using the strong terminal oxidation ability of nonmodel microorganisms such as Candida tropicalis to oxidize hydrophobic substrates such as alkanes. Here, we first summarize the metabolic pathways of oxidative alkane conversion into dicarboxylic acid by terminally oxidizing unconventional yeasts and the corresponding metabolic engineering strategies. Then, we summarize the research progress on new dicarboxylic acid production processes. Finally, the future development directions in the biosynthesis of mid- and long-chain dicarboxylic acids are prospected from synthetic biology and bioprocess engineering, which can also provide a reference for the synthesis of other biobased chemicals and biomaterials.

Abstract:With the gradual depletion of petroleum resources, the one-step synthesis of acrylic acid/ methyl acrylate from acetic acid/methyl acetate and formaldehyde via aldol reaction, instead of propylene selective oxidation, has stimulated researchers' interest. Current research in this field focus on catalyst modification, structure–activity relationships, catalytic mechanism and kinetics, and separation process design. Abstract:Adipic acid (AA) is an important precursor for the production of Nylon-66, and renewable routes for AA are being explored. Previously we have shown the use of HI and molecular H-2 to be an effective combination for the selective ring-opening of tetrahydrofuran-2,5-dicarboxylic acid (THFDCA) in organic acids without the need for a metal catalyst. However, the use of HI could deter this system from large scale use for renewable AA. In this work, we employ zeolites as an effective proton source in combination with iodide salts (NaI), which obtains high AA rates in a similar metal-free pathway. Kinetic measurements coupled with zeolite characterization suggest that a dominant mechanism for THFDCA activation is bulk-phase protonation promoted by in situ ion-exchange via the metal cation from the iodide salt. Zeolites can be regenerated multiple times with minimal loss in reactivity, which enhances the commercial applicability of the system.
