Abstract:Quinoline derivatives, aryl hydrazines and hydrazide compounds have significant biological activities. Benzo[h]quinoline derivatives were synthesized from 1-naphthylamine, substituted benzaldehyde and methyl pyruvate by Doebner-Miller reaction. After the ester was reduced, the corresponding aldehyde was obtained by oxidation. Benzo[h]quinolinium and quinoline hydrazide compounds were synthesized by reacting benzo[h]quinoline formaldehyde with hydrazine salts and hydrazides, respectively. The preliminary activity data showed that most of the compounds exhibited significant inhibitory activities against cycle protein 25B (CDC 25B) and protein tyrosine phosphatase PTP 1B.

Polycyclic Aromatic Compounds

Abstract:Natural compounds including bis-heterocyclic moieties displayed several applications in pharmaceutical, agrochemical, and industrial fields. Di-thiazoles are effective core of several anticancer drugs such as bleomycin. Derived from importance of di-thiazoles, herein, we synthesized novel series of di-thiazoles from condensation reaction of thiazole thiosemicarbazone derivative with α-haloketones. Physical features and chemical structures of new compounds formed were discussed and assured based on their spectral and conformational analysis. The cytotoxic potency of newly synthesized thiazole derivatives against colon (HCT-116) and hepatocellular (HepG2) carcinoma-cell lines was determined by MTT assay. Compounds 7a, 9a, 9c, and 9e revealed marked anticancer activity especially against (HCT-116). Molecular docking study was executed over chosen compounds referring to expected promising features against LDH-5, which appeared agree with biological results. This study was utilized to predict prior binding-efficiency of active thiazole compounds.

Abstract:Ten novel benzotriazole compounds were synthesized. Their chemical structures were confirmed by 1 H NMR, IR, and elemental analyses, coupled with three selected single-crystal structures (compounds A2 , B3 , and B5 ). Their antimycotic and antitumor activities were also investigated. The title compounds showed some antitumor activities, especially in the case of A3 and A4 , which showed the most potent activity of propagation inhibition in liver and galactophore cancer cells.

Abstract:A series of novel 2-aryl-benzimidazole derivatives of dehydroabietic acid were synthesized and characterized by IR, 1H NMR, 13C NMR, MS and elemental analyses. All the target compounds were evaluated for their in vitro cytotoxic activity against SMMC-7721, MDA-MB-231, HeLa and CT-26 cancer cell lines and the normal hepatocyte cell line QSG-7701 through MTT assays. Among them, compound 6j displayed the most potent cytotoxic activity with IC50 values of 0.08 ± 0.01, 0.19 ± 0.04, 0.23 ± 0.05 and 0.42 ± 0.07 μM, respectively, and substantially reduced cytotoxicity against QSG-7701 cells (5.82 ± 0.38 μM). The treatment of SMMC-7721 cells with compound 6j led to considerable inhibition of cell migration ability. The influence of compound 6j on cell cycle distribution was assessed on SMMC-7721 cells, exhibiting a cell cycle arrest at the G2/M phase. Moreover, tubulin polymerization assays and immunofluorescence assays elucidated that compound 6j could significantly inhibit tubulin polymerization and disrupt the intracellular microtubule network. A molecular docking study provided insight into the binding mode of compound 6j in the colchicine site of tubulin. In addition, compound 6j was found to induce apoptosis of SMMC-7721 cells, an increase of intracellular ROS level and a loss of mitochondrial membrane potential in a dose-dependent manner. These findings provided new molecular scaffolds for the further development of novel antitumor agents targeting tubulin polymerization.

Abstract:The therapeutic potential of benzimidazole has brought them widespread recognition as a class of heterocyclic compounds having nitrogen-containing rings. In recent years, there has been a lot of focus on synthesizing these molecules in order to reveal a broad range of biological actions. Therefore, a novel series of 1H-benzo[ d ]imidazole-5-carboxamide derivatives were designed, synthesized, and investigated for their antioxidant and enzyme inhibitory activities. The structural elucidation of synthesized compounds was accomplished by a variety of spectroscopic techniques, such as elemental analysis, 1 H and 13 C NMR, and LC-MS. Using the DPPH and FRAP techniques, the antioxidant activity of newly synthesized compounds was assessed. With IC 50 values of 81.45, 72.14, and 77.35 μM, respectively, in the DPPH assay and 86.07, 75.02, and 81.14 μM, respectively, in the FRAP assay, the compounds 10c, 10f, and 10 g shown have significant antioxidant activity in comparisons with reference drugs ascorbic acid and Trolox. Furthermore, the inhibitory activity of xanthine oxidase (XO) and lipoxygenase (LOX) enzymes was assessed for the most potent molecules. Compounds 10c, 10f, and 10 g show superior inhibitory effects than reference drugs allopurinol and baicalein, with IC 50 values of 19.52, 13.95, and 15.83 μM, respectively, against LOX and 26.14, 18.43, and 22.05 μM, respectively, against the XO enzyme. Moreover, studies using molecular docking were conducted to gain a deeper understanding of the interactions between the 3NM8 and 1N8Q protein and the most effective compounds, 10c, 10f, and 10 g. While compared to the reference medications, the studied compounds showed substantial docking scores and binding affinities, as indicated by docking studies. Through the use of drug-likeness and structure-activity relationships (SAR), an association between the newly synthesized compounds' biological and physicochemical properties was established.

Abstract:A series of quinoline derivatives were synthesized and their immunosuppressive activity and cytotoxicity were evaluated with a T-cell functional assay and MTT method, respectively. Most of 5,7-dimethoxyquinolin-4-yl ortho-substituted benzoate derivatives (18, 22, 24, 28, 31 and 37) showed a quite stronger inhibitory activity compared to other analogs. Among the synthesized compounds, 5,7-dimethoxyquinolin-4-yl 2,6-dichlorobenzoate (22) and 5,7-dimethoxyquinolin-4-yl 4-methylbenzenesulfonate (40) exhibited a potent inhibitory activity without significant cytotoxicity at 10μM concentration. The preliminary mechanism of the active compounds 22 and 40 was further clarified based on the fluorescence activated cell sorter (FACS) assay, and the compounds exerted immunosuppressive activity via inhibiting the T cell activation in a dose dependent manner.

Abstract:A novel series of six compounds, benzisoxazole tethered 1,2,4-triazoles were synthesized in five steps with good yields and characterized. 1H NMR, 13C NMR and mass spectrometry confirmed the formation of the derivatives. Antimicrobial activity was evaluated for the synthesized derivatives against three bacterial strains (Escherichia coli, Mycobacterium smegmatis and Staphylococcus aureus) and a fungal strain (Candida albicans). Similarly, anticancer activity was assessed against MCF-7 cell lines. The biological analysis disclosed that, out of the six compounds, derivative 6c showed better antimicrobial and anticancer activity with MIC of 12.5 μg/mL against Staphylococcus aureus and Mycobacterium smegmatis; and IC50 of 35.57 μM against MCF-7 cancer cell line. Theoretical evaluation provides an insight about ligand interaction with proteins, highest occupied molecular orbital (HOMO), lowest unoccupied molecular orbital (LUMO), electronegativity, hardness, softness, electrophilicity and neucleophilicity of the ligands via molecular docking and density functional theory (DFT) studies; this presents probable evidence for the biological activity of the derivatives. Molecular Dynamics (MD) simulation of the ligand 6c with protein HDAC7 (PDB ID: 3ZNR) was studied.

Abstract:Tubulin-targeting drugs have increasingly become the focus of anticancer drugs research. Twenty-five novel benzimidazole grafted benzsulfamide-containing pyrazole ring derivatives were synthesized and evaluated for bioactivity as potential tubulin polymerization inhibitors. Among them, compound 30 showed the most excellent inhibition against tubulin assembly (IC50 = 1.52 mu M) and in vitro growth inhibitory activity against a panel of four human cancer cell lines (IC50 = 0.15, 0.21, 0.33 and 0.17 mu M, respectively for A549, Hela, HepG2 and MCF-7). It could also validly induce A549 cell apoptosis, cause cell cycle arrest in G2/M phase and disrupt the cellular microtubule network. These results, along with molecular docking data, provided an important basis for further optimization of compound 30 as a potential anticancer agent.

Anti-Cancer Agents in Medicinal Chemistry

Abstract:Novel substituted triptycene bisquinones and 1, 4-anthracenediones were synthesized and screened for their anti-cancer activities. A number of analogs were synthesized utilizing various synthetic transformations and found to elicit interesting antitumor effects. Analogs included water-soluble pro-drugs and ammonium salts. These potent antitumor drugs are DNA topoisomerase inhibitors that induce DNA strand breaks, inhibit DNA, RNA and protein syntheses and reduce tumor cell proliferation in the nanomolar range in vitro. They induce cytochrome c release, caspase-9, -3 and -8 activities, poly(ADP)-ribose polymerase-1 (PARP) cleavage, and internucleosomal DNA fragmentation by a mechanism which involves caspase-2 activation but not Fas signaling. Moreover, these drugs remain effective in multidrug-resistant tumor cells and have the advantage of blocking nucleoside transport and inducing a rapid loss of mitochondrial transmembrane potential. Based on their effects in tumor cells and isolated mitochondria, it is hypothesized that these drugs might, directly and indirectly, target components of the permeability transition pore to induce mitochondrial permeability transition and the release of proapoptotic factors. This review provides a summary of synthetic efforts and mechanistic endeavor.

Abstract:A new series of linear dimeric compounds mimicking naturally occurring annonaceous acetogenins have been synthesized by bivalent analogue design, and their cytotoxicities have been evaluated against the growth of cancer cells by MTT method. Most of these compounds show selective action favored to human cancer cell lines over normal cell lines, and compound 9 with bis-terminal benzoquinone functionality exhibits an IC50=0.40μM against MCF7 cell lines. This work mentions that appropriate conformational constraints might be a useful optimizing tool for this unique class of anticancer compounds.

Abstract:A series of novel compounds (8a-21b) were designed and synthesized based on 1H-benzo[d]imidazole. Their biological activities were evaluated as potential tubulin polymerization inhibitors and anticancer agents. Among all the compounds, compound 18b showed the most potent in vitro growth inhibitory activity against A549, MCF-7, K562 cancer cell lines, with IC50 values of 0.12 mu M, 0.15 mu M, 0.21 mu M, respectively. And, compound 18b also exhibited significant tubulin polymerization inhibitory activity (IC50 = 2.1 mu M), which was comparable with the positive control. Furthermore, docking simulation and 3D-QSAR modeling of inhibitor analogues provide determination of the probable binding model and an important basis that compound 18b with potent inhibitory activity in tumor growth may be a potential anticancer agent.

Abstract:Abstract: Heterocyclic compounds are fundamental building blocks for developing novel bioactive compounds. Due to their extensive uses in both industrial and synthetic organic chemistry, quinoline and benzimidazole have recently become important heterocycles. Clinical trials have investigated quinoline and benzimidazole analogues to treat a variety of illnesses, including cancer, bacterial and fungal infection, DNA damage, etc. Medicinal chemists are paying attention to nitrogen-containing hybrid heterocyclic compounds that have a wide range of therapeutical potential with lesser adverse effects. Many efforts have been made to find new and more efficient ways to synthesize these molecules. However, microbial resistance is becoming a major threat to the scientific community; hence, the necessity for the discovery and development of novel antimicrobial drugs with novel modes of action is becoming highly significant. One strategy to overcome this problem is to produce hybrid molecules by combining two or more bioactive heterocyclic moieties in a single molecular platform. Based on established research data on quinoline-bearing benzimidazole derivatives, it can be concluded that both moieties are used for the synthesis of promising therapeutically active agents. This present review comprises the synthetic approaches of biologically active quinolines containing benzimidazole derivatives with their structure-activity relationship studies to provide an overview of the work done on quinoline derivatives to the medicinal chemist for future research.

Abstract:In this study, a series of novel β-carboline condensed imidazolium derivatives (7a-7y) were designed and synthesized by incorporating imidazolium salt structures into β-carboline. The cytotoxicity of compounds 7a-7y was evaluated in various cancer cell lines, including lung cancer (A549), gastric cancer (BGC-823), mouse colon cancer (CT-26), liver cancer (Bel-7402), and breast cancer (MCF-7), using the MTT assay. Most compounds exhibited significant activity against one or more of the cancer cell lines. Notably, compounds 7 g, 7o, 7r, 7 s, 7u, 7v, 7x, and 7w showed the highest cytotoxic activity (IC50 < 2 μM) in the tested tumor cell lines. Compound 7x demonstrated cytotoxic activities of 1.3 ± 0.3 μM (for BGC-823), 2.4 ± 0.4 μM (against A549), 7.8 ± 0.9 μM (for Bel-7402), and 9.8 ± 1.4 μM (against CT-26). The chick chorioallantoic membrane assay revealed significant anti-angiogenic potential of compound 7x. Molecular imprinting studies suggested the anti-angiogenic effect of compound 7x might be attributed to inhibition of VEGFR2 kinase. Molecular docking and molecular dynamics further indicate that its activity may be primarily associated with the potential inhibition of VEGFR2. Our research outcomes have provided valuable lead compounds for the development of novel antitumor drugs and have offered beneficial insights for subsequent drug design and optimization.

Abstract:A series of 1-sulfonyl indolines was synthesized and evaluated for antiproliferative activity. The most potent compounds 9a and 9e showed significant cytotoxicity (IC50 in the range of 0.055–0.105 and 0.039–0.112μM, respectively) against four human cancer cell lines HCT116, PC3, HepG2 and SK-OV-3. The structure–activity relationship of this series of sulfonamides, including the influence of azaheterocycle rings, substituent at the different positions of indoline, and the cyclopropane moiety, was described.

Abstract:The N , N ′-dimethylation of a diphenylsquaramide induces a conformational change in the orientation of the phenyl rings. This has been exploited to create a series of bis-urea, -thiourea and -squaramide anionophores. The compounds were shown to bind to Cl − using proton NMR titration techniques and to transport H + /Cl − through the lipid bilayers, whereas a non-methylated analogue displayed limited transport activity. Despite their potency in transport studies, the series had a negligible impact on cancer cell viability.
