Abstract:Optical coherence tomography (OCT) suffers from speckle noise, causing the deterioration of image quality, especially in high-resolution modalities like visible light OCT (vis-OCT). The potential of conventional supervised deep learning denoising methods is limited by the difficulty of obtaining clean data. Here, we proposed an innovative self-supervised strategy called Sub2Full (S2F) for OCT despeckling without clean data. This approach works by acquiring two repeated B-scans, splitting the spectrum of the first repeat as a low-resolution input, and utilizing the full spectrum of the second repeat as the high-resolution target. The proposed method was validated on vis-OCT retinal images visualizing sublaminar structures in outer retina and demonstrated superior performance over conventional Noise2Noise and Noise2Void schemes. Abstract:An improved sparse representation algorithm is proposed for reducing speckle noise in optical coherence tomography (OCT) images. To reduce the dictionary training time, the orthogonal matching pursuit algorithm (OMP) is replaced by a piecewise orthogonal matching pursuit algorithm (ST-OMP), and a threshold standard is set to train multiple atoms simultaneously to improve the tracking efficiency. Normal, diabetic macular edema, and fundus neovascularization membrane retina OCT images were tested using the proposed algorithm and other denoising algorithms. The results show that the proposed algorithm exhibits an excellent denoising ability. The details and edge information in the OCT images were well preserved by the proposed algorithm. Meanwhile, the image processing time was reduced by half with the proposed algorithm compared with the traditional K-SVD algorithm.

Abstract:A deconvolution method for image enhancement suitable for spectral domain optical coherence tomography (SD-OCT) is proposed. The method includes Wiener filtering for spectral deconvolution and successive homomorphic filtering for spatial deconvolution, aiming to minimize blurring effect of axial point spread function (PSF) and its depth dependence. Tunable factor in Wiener filtering is optimized for tradeoff between compensation ratio and signal-to-noise ratio (SNR). In vivo OCT imaging of a fresh shrimp is done and image reconstructions are performed. Comparing results demonstrates the proposed method suppresses the coherent noises in OCT image while compensating sensitivity fall-off with increased averaged SNR and recovers structural information at deep depths. Contrast-to-noise ratio (CNR) and averaged SNR in the reconstructed image based on the proposed method in contrast to those without deconvolution are improved by 3.4 dB and 4.6 dB, respectively. Abstract:Optical coherence tomography (OCT) is an important imaging technique extensively applied in medical sciences. However, OCT images often suffer from speckle noise, which is a kind of multiplicative noise inherited in coherent imaging systems. A speckle noise reduction algorithm with total variation (TV) regularization is proposed to restore speckled OCT images. It constructs the regularization parameter and utilizes the tuning function. The proposed algorithm realizes sufficient speckle noise reduction and delicate edge preservation. Simulations demonstrate the performance of the proposed algorithm with respect to visual effects, processing time and image quality metrics of signal-to-noise ratio (SNR), equivalent number of looks (ENL), contrast-to-noise ratio (CNR), relative mean square error (RMSE) and edge preservation factor. Compared with some classical and typical despeckling algorithms, the proposed algorithm exhibits good results in edge preservation, recovery error and time efficiency, and presents better SNR, ENL and CNR. The applicability of the proposed algorithm with regard to OCT in-device preprocessing is discussed in details. Abstract:Optical coherence tomography (OCT) has been an important and powerful tool for biological research and clinical applications. However, speckle noise significantly degrades the image quality of OCT and has a negative impact on the clinical diagnosis accuracy. In this paper, we propose a novel speckle noise suppression technique which changes the spatial distribution of sample beam using a special optical chopper. Then a series of OCT images with uncorrelated speckle patterns could be captured and compounded to improve the image quality without degradation of resolution. Typical signal-to-noise ratio improvement of ∼6.4 dB is experimentally achieved in tissue phantom imaging with average number n = 100. Furthermore, compared with conventional OCT, the proposed technique is demonstrated to view finer and clearer biological structures in human skin in vivo, such as sweat glands and blood vessels. The advantages of low cost, simple structure and compact integration will benefit the future design of handheld or endoscopic probe for biomedical imaging in research and clinical applications.

Abstract:Optical coherence tomography (OCT) is a powerful biomedical imaging technology that relies on the coherent detection of backscattered light to image tissue morphology in vivo. As a consequence, OCT is susceptible to coherent noise (speckle noise), which imposes significant limitations on its diagnostic capabilities. Here we show a method based purely on light manipulation that is able to entirely remove the speckle noise originating from turbid samples without any compromise in resolution. We refer to this method as Speckle-Free OCT (SFOCT). Using SFOCT, we succeeded in revealing small structures that are otherwise hidden by speckle noise when using conventional OCT, including the inner stromal structure of a live mouse cornea, the fine structures inside the mouse pinna, sweat ducts, and Meissners corpuscle in the human fingertip skin. SFOCT has the potential to markedly increase OCTs diagnostic capabilities of various human diseases by revealing minute features that correlate with early pathology.

Abstract:Optical coherence tomography (OCT) has proven to be an effective and safe diagnostic tool in clinical settings due to its unique advantages. Nonetheless, the OCT images are unavoidably corrupted with speckle noise. Despeckling has become a vital preliminary step in OCT based image processing and analysis tasks. The supervised deep learning (DL) based denoising algorithms have recently attracted much attention due to their promising performance, but the lack of the noise-free OCT images renders it difficult to provide the effective supervision for DL model training. This paper has presented a self-supervised OCT image despeckling approach which does not depend on the reference clean image and combines the disentangled representation strategy with speckle noise estimation for the effective denoising. The proposed algorithm firstly disentangles the corrupted image into the speckle noise and clean image. Then, the multiple generated clean images are multiplied by the disentangled speckle noise to synthesize the speckle-corrupted image for self-supervised learning. To make the generated clean image and extracted speckle noise more statistically realistic, a collaborative masking strategy and a noise calibration module are applied to explore the spatial correlation of the clean image and refine speckle noise. The proposed method has been validated on two public datasets. Experimental results demonstrate that our method has superior speckle reduction performance to some traditional and DL-based despeckling approaches in terms of visual evaluation and quantitative metrics. Particularly, our method provides contrast-to-noise ratio (CNR) and signal-to-noise ratio (SNR) improvements of at least 0.35 dB and 14.37 dB over the compared methods, respectively. Additionally, the retinal image classification results provide further evidence for the exceptional despeckling performance of our approach against other compared methods. It is obvious that the developed method has significant advantages in keeping the fidelity of retinal structure and enhancing the speckle suppression level. Meanwhile, the presented method holds certain reference value for improving diagnostic efficiency in clinical.

Abstract:Optical coherence tomography (OCT) is widely used in clinical diagnosis due to its non-invasive, real-time, and high-resolution characteristics. However, the inherent speckle noise seriously degrades the image quality, which might damage the fine structures in OCT, thus affecting the diagnosis results. In recent years, supervised deep learning-based denoising methods have shown excellent denoising ability. To train a deep denoiser, a large number of paired noisy-clean images are required, which is difficult to achieve in clinical practice, since acquiring a speckle-free OCT image requires dozens of repeated scans and image registration. In this research, we propose a self-supervised strategy that helps build a despeckling model by training it to map neighboring pixels in a single noisy OCT image. Adjacent pixel patches are randomly selected from the original OCT image to generate two similar undersampled images, which are respectively used as the input and target images for training a deep neural network. To ensure both the despeckling and the structure-preserving effects, a multi-scale pixel patch sampler and corresponding loss functions are adopted in our practice. Through quantitative evaluation and qualitative visual comparison, we found that the proposed method performs better than state-of-the-art methods regarding despeckling effects and structure preservation. Besides, the proposed method is much easier to train and deploy without the need for clean OCT images, which has great significance in clinical practice.

Abstract:Optical coherence tomography (OCT) has been extensively utilized in the field of biomedical imaging due to its non-invasive nature and its ability to provide high-resolution, in-depth imaging of biological tissues. However, the use of low-coherence light can lead to unintended interference phenomena within the sample, which inevitably introduces speckle noise into the imaging results. This type of noise often obscures key features in the image, thereby reducing the accuracy of medical diagnoses. Existing denoising algorithms, while removing noise, tend to also damage the structural details of the image, affecting the quality of diagnosis. To overcome this challenge, we have proposed a speckle noise (PSN) framework. The core of this framework is an innovative dual-module noise generator that can decompose the noise in OCT images into speckle noise and equipment noise, addressing each type independently. By integrating the physical properties of noise into the design of the noise generator and training it with unpaired data, we are able to synthesize realistic noise images that match clear images. These synthesized paired images are then used to train a denoiser to effectively denoise real OCT images. Our method has demonstrated its superiority in both private and public datasets, particularly in maintaining the integrity of the image structure. This study emphasizes the importance of considering the physical information of noise in denoising tasks, providing a new perspective and solution for enhancing OCT image denoising technology.

Abstract:In most coherent imaging modality, speckle noise is a major cause that blurs the boundary of tissues and degrades the image contrast. By utilizing the unique properties of supercontinuum (SC) generated by noise-like pulses (NLPs) and a simple multi-frame averaging technique, we achieved significant speckle reduction in spectral domain optical coherence tomography (SD-OCT). We quantitatively compared the speckle of our proposed method with those of conventional swept source OCT (SS-OCT) and SD-OCT based on commercial light sources. The experimental results show that SC pumped by NLPs combined with noncoherent averaging method achieves better denoising performance in terms of contrast to noise ratio (CNR).

Abstract:Deconvolution methods are not feasible in spectral domain optical coherence tomography (SD-OCT) system due to the linear space variant property of its axial point spread function (PSF). In order to improve the axial resolution and enhance the imaging quality of the SD-OCT system, a Lucy-Richardson algorithm based deconvolution method is implemented with the numerically compensated A-scan signals and the effective axial PSF in this paper. The main factors decreasing the value of PSF along the axial direction and blurring the images, as well as the deconvolution process, are theoretical studied. With the developed SD-OCT system, the axial modulation function of the PSF is retrieved by curve fitting of the measured peak values of the PSF at different axial positions. After compensating the A-scan signal with the modulation function, deconvolution is performed with the measured effective axial PSF. In vivo imaging on typical samples is conducted and reconstruction is done based on the deconvolution method. The comparing results demonstrate that the proposed deconvolution method not only enhances the axial resolution but also improves the depth dependent sensitivity fall-off in SD-OCT system.

In this present study, we discussed about the nephroprotective and cardioprotective and antioxidant effect of Trianthema portulacastrum Linn in experimental animals. Biochemical parameters and histopathological studies were analysed and the results are discussed to show the potent effect of the Siddha medicinal plant Trianthema Portulacastrum Linn.

Abstract:Properties of hypotonically-activated Cl- channels in isolated rat hepatocytes were studied by the patch-clamp whole-cell technique. Hypotonic stress (140-150 mosmol kg 1 H2O) induced a hyperpolarization of the membrane of hepatocytes in the presence of an inwardly oriented Cl gradient, but had no effect on the membrane potential in the absence of Cl. An increase in the hypotonically-induced conductance was significantly inhibited by 4-acetamido-4'-isothiocyanatostilbene-2, 2'-disulfonic acid (SITS; 50 microM), but not by Ba2+ (1 mM). Pre-incubation with arachidonic acid (20 microM) significantly inhibited the hypotonically-activated conductance. The combined application of a cyclo-oxygenase inhibitor, indomethacin (50 microM) and a lipoxygenase inhibitor, esculetin (100 microM) also inhibited the conductance, whereas quinacrine (200 microM), a phospholipase A2 inhibitor, significantly induced a large steady conductance. Outward Cl- currents, but not cationic currents, were elicited by the hypotonic stress. The current did not show any rapid time-dependent inactivation during the voltage clamp of 0.1 s. The combined application of arachidonic acid, indomethacin and esculetin inhibited the hypotonically-activated Cl- currents. The present study has shown that the Cl- channel is activated by exposure to hypotonic stress and closed by an increase in arachidonic acid concentration in isolated rat hepatocytes.
Speckle Reduction for Ophthalmic OCT Images Based on Wavelet Filtering Technique

Abstract:Speckle is a major factor that degrades the contrast and resolution of ophthalmic optical coherence tomography images. In this paper, we present a speckle reduction method for ophthalmic OCT images based on wavelet decomposition and reconstruction. Here, the Otsu algorithm is introduced for the calculation of a threshold to distinguish signals and speckles roughly, and the correlations of neighboring wavelet coefficients and the coefficients of the corresponding locations in different decomposition levels are used to suppress the speckle noise. The experimental results show that our method can improve the contrast of the ophthalmic OCT images significantly without unbearable image sharpness loss.
A Dynamic Wavelet Filtering for Anterior Chamber Optical Coherence Tomographic Images

Abstract:Speckle noise is a common phenomenon in Optical Coherence Tomography (OCT) images. This paper describes a dynamic filtering approach for anterior chamber OCT images to reduce the speckle noise in wavelet domain. The approach proposed segments the OCT image into some parts and identifies if the parts have region of interest (ROI), which includes the anterior chamber tissues. Then, it suppresses noise with three different suppression strategies in part with ROI. For the part without ROI, it sets the discrete wavelet transform coefficients of this part to zero. Here, the sampling-based sub-band adaptive algorithm is used to distinguish the ROI; and the correlations of neighboring wavelet coefficients and the coefficients of the corresponding locations in adjacent decomposition levels are used to suppress the noise. The performance improvement over the previously published method is quantified in terms of noise suppression, image structural preservation and visual quality. The numerical values of the image quality metrics along with the qualitative analysis results indicated that the approach proposed has better performance.

Abstract:Speckle noise is problematic in optical coherence tomography (OCT). With the fast scan rate, swept source OCT scans the same position in the retina for multiple times rapidly and computes an average image from the multiple scans for speckle reduction. However, the eye movement poses some challenges. In this paper, we propose a new method for speckle reduction from multiply-scanned OCT slices. The proposed method applies a preliminary speckle reduction on the OCT slices and then registers them using a global alignment followed by a local alignment based on fast iterative diamond search. After that, low rank matrix completion using bilateral random projection is utilized to iteratively estimate the noise and recover the underlying clean image. Experimental results show that the proposed method achieves average contrast to noise ratio 15.65, better than 13.78 by the baseline method used currently in swept source OCT devices. The technology can be embedded into current OCT machines to enhance the image quality for subsequent analysis.

Abstract:By combining the advantages of compressive sensing optical coherence tomography (OCT) and full-depth OCT in terms of imaging time and imaging depth, we demonstrate how compressive sampling and dispersion encoding can be simultaneously used to reconstruct a full-depth OCT image. Moreover, by considering the image processing speed, we propose a two-step compressive dispersion encoding (TCDE) method, in which a large dispersion imbalance is introduced between the reference arm and the sample arm and two iterations are performed. The first iteration selects the signals with higher intensity and then removes their conjugate items and incoherent aliasing artifacts caused by undersampling based on the least squares method. The second iteration selects the signals with lower intensity. Experimental results show that nearly the same conjugate inhibition ratio can be obtained with 50% sampled data and 100% sampled data using the TCDE method. Full-depth images of a glass slide, an onion, and a live fish eye are obtained from 50% and 100% sampled data using the TCDE method. For a 1.4 mm x 3.6 mm fish eye image, the conjugate items are reduced by 33.2 and 31.7 dB using 50% sampled data and 100% sampled data, respectively. (C) 2018 Optical Society of America
