Abstract:The geometry of the medical titanium surface micro pattern, the micro- and nanostructures within the pattern, and the roughness of the structure are key parameters affecting the integration of the implant surface with the bone tissue. The femtosecond laser is an effective method for surface modification of implants , but achieving effective control of the depth of the pattern and the inner surface topography and roughness is a crucial processing challenge. In this study, the evolution of ablation pit morphology with process parameters was analyzed to establish the correspondence between process parameters and the ablation mechanism. A femtosecond laser pulse sequence method is proposed to increase the machining depth from 24 μm to 38 μm while reducing the surface roughness from Sa = 2.2 μm to Sa = 0.5 μm by regulating the liquid phase migration as well as the thermal phase change process without affecting the processing efficiency. And the surface roughness can be reduced from Sa = 2.2 μm to <0.1 μm with appropriate pulse sequence parameters.

Abstract:Femtosecond laser modification of titanium surfaces was performed to produce microstructured hydrophilic and biocompatible surface layers. Biocompatible nano/microcoatings were prepared for the first time by dry femtosecond laser imprinting of hydroxylapatite nano/micropowder onto VT6 titanium surfaces. In these experiments HAP was first deposited onto the titanium surfaces and then softly imprinted by multiple femtosecond laser pulses into the laser-melted surface metal layer. The surface relief was modified at the nano- and microscales depending on the incident laser fluence and sample scanning speed. Wetting tests demonstrated that the wetting properties of the pristine Ti surface can be tuned through its laser modification in both the hydrophobic and hydrophilic directions.

Abstract:The osteo-bioactive potential of biomaterials can be modulated by altering material properties such as chemical composition, surface topography, and geometry. The correlation between the physicochemical properties of biomaterials and their osteo-bioactivity is highly complex. As a material widely used in bone repair, the structure-activity/dose-effect relationship between titanium (Ti) surface features and osteo-bioactivity remains unclear. To quantitatively enhance the osteogenic activity of Ti, we employed femtosecond laser (FSL) technology to create Ti surfaces with gradient changes in bioactive (nucleation) sites. Based on the apatite deposition ability on the etched surfaces, the quantitative relationship between the osteo-bioactivity of Ti surfaces and their characteristic parameters was systematically explored. Concurrently, classical molecular dynamics (MD) simulations were utilized to investigate the aggregation behavior of calcium and phosphate ions on the Ti surfaces with different sites. The findings from mineralization experiments revealed that the type and density of bioactive site could influence the deposition of apatite. It further identifies TiO2 and Ti-OH as the crucial bioactive sites, with basic Ti-OH exhibiting superior efficacy as a bioactive site compared to its acidic counterpart. Moreover, the bioactive (nucleation) site densities should maintain at a minimum of 2.33 ± 0.55/nm2 for TiO2 and 2.50 ± 0.59/nm2 for -OH, ensuring satisfactory osteo-bioactivity on the Ti surface. The results provide the atomic/molecular features of Ti surfaces that effectively foster apatite deposition and bioactivity, promoting the rapid progression of titanium-based bone repair materials.

Abstract:After nearly half a century of development under the guidance of the osseointegration theory, the major dilemmas for current implant dentistry are the implant associated infection and insufficient osseointegration. Moreover, biological aging of titanium (Ti) implants also brings great uncertainty to clinical results. In the present study, a novel nano-micro-hierarchical topography pattern is created by sandblasting and dual acid-etching on a Ti surface. The physico-chemical properties of the surfaces were characterized by scanning electron microscopy, contact angle measurement, X-ray photoelectron spectroscopy and X-ray diffraction. The effects of the hierarchical surfaces on osteoprogenitor cell growth and bacterial activities were separately evaluated. The optimized nano-micro-hierarchical Ti surface exhibits surprisingly topography-dependent antibacterial capacity via inhibiting bacterial adhesion of several species in the early stage and better osteogenesis ability than the microscaled surface. Aging studies demonstrate that, compared with the surface with a microscale structure, the nano-micro-hierarchical Ti surface has greater anti-aging ability manifested as being more capable to retain hydrophilicity and bioactivity during aging. Furthermore, the present study reveals that the biological aging of the Ti implant is attributed to two decisive factors during the aging period: the progressively thickened amorphous TiO2 layer by autoxidation and the unavoidable accumulation of hydrocarbons on the Ti implant surface.

Abstract:Surface microstructure and chemical composition of the implant are very important for its osseointegration in vivo. In this paper, a hierarchical micropattern covered with calcium phosphate (Ca/P phase) was obtained on titanium (Ti) implant surface by femtosecond lasers (FSL) irradiation in hydroxyapatite suspension. The hierachical micropattern as well as Ca/P phase increased osteoblastic cell adhesion. Higher expression of osteogenic markers (osteocalcin, osteopontin, and runt related transcription factor-2) on the surface treated by FSL of 2.55 J/cm(2) indicated the favorable effect of laser treatment on cell differentiation. In vivo studies were carried out to evaluate the effect of laser treatment and Ca/P deposition on the osseointegration. It showed that the binding capacity between bone and FSL-treated Ti implants was obviously stronger than that between bone and polished or sand blasting and acid etching (SLA) Ti implants. Bone trabecula surrounded the FSL-treated implants without fibrous tissue after 8-week implantation. Also, higher bone mineral density was seen surrounding the FSL-treated implants. Our in vitro and in vivo studies demonstrated that the FSL induced micropattern and Ca/P phase had positive effects on the acceleration of early osseointegration of Ti implants with bone tissue.

Abstract:In this study, a femtosecond laser with a wavelength of 800 nm was used to modify the surface of a titanium alloy bone scaffold created via selective laser melting (SLM). The outcomes demonstrated that the surface morphology of the bone scaffold after femtosecond laser treatment was micro-nano morphology. The hydrophobic structure of the scaffold was changed into a super-hydrophilic structure, improving the surface roughness, which was highly helpful for osteoblast adhesion and differentiation. The femtosecond laser surface treatment in vitro samples produced a thick layer of hydroxyapatite (HAP) with improved surface bioactivity. The effectiveness of osseointegration and interstitial growth of the specimens treated with the femtosecond laser surface were found to be better when bone scaffolds were implanted into the epiphysis of the tibia of rabbits. As a result, femtosecond laser therapy dramatically enhanced the surface activity of bone scaffolds and their capacity to integrate with the surrounding bone tissues, serving as a trustworthy benchmark for future biological scaffold research.

Abstract:While pure titanium is a material of choice for medical applications for its excellent mechanical and chemical properties, further improving its surface-related functionalities by surface texturing is also promising. In the present work, we experimentally investigate the fabrication, as well as the resulting functionalities of surface wettability and biocompatibility, of precise mesh-type surface textures on pure titanium by picosecond pulsed laser ablation operating at 1064 nm wavelength. Specifically, the dependence of microgroove morphology on average power and scanning speed is evaluated to yield optimized laser processing parameters, which are utilized to fabricate high precision mesh-type surface textures with uniform feature size and limited thermal effects on pure titanium. Subsequent performance evaluation tests demonstrate that the mesh-type surface textures induce a beneficial effect on the biocompatibility with respect to BMSC cells due to the enhanced hydrophilicity.

Abstract:In this paper, the surface of the titanium alloy bone scaffold manufactured by selective laser melting (SLM) was modified by femtosecond laser with a wavelength of 800 nm. The results showed that femtosecond laser treatment of bone scaffold presented micro-nano surface morphology. The surface roughness of the scaffold was improved, and the hydrophobic structure was transformed into the super hydrophilic structure, which was very beneficial to the adhesion and differentiation of osteoblasts. In vitro bioactivity experiments, femtosecond laser surface treatment (AM+FS) samples formed a dense layer of hydroxyapatite (HA) on the surface, with better surface bioactivity. The bone scaffolds were implanted into rabbit tibial epiphysis, and the bone tissue growth of the AM+FS sample was better than that of the additive manufacturing (AM) sample at the same time points. In conclusion, femtosecond laser treatment significantly improves the surface activity of bone scaffolds and the ability to integrate with surrounding bone tissues, which provides a reliable reference for research in biological scaffolds.

Abstract:Surface modification of titanium implant is processed by microarc oxidation and hydrothermal treatment. A porous surface with a biologically active bone-like apatite layer was formed. The apatite layer consists of very fine crystals and high crystallinity and is integrated with the titanium alloy substrate with a graded structure without a distinct interface. Fabrication of multifunctional titanium surfaces by producing hierarchical surface patterns using laser based ablation methods

Abstract:Textured implant surfaces with micrometer and sub-micrometer features can improve contact properties like cell adhesion and bacteria repellency. A critical point of these surfaces is their mechanical stability during implantation. Therefore, strategies capable to provide both biocompatibility for an improved implant healing and resistance to wear for protecting the functional surface are required. In this work, laser-based fabrication methods have been used to produce hierarchical patterns on titanium surfaces. Using Direct Laser Writing with a nanosecond pulsed laser, crater-like structures with a separation distance of 50 µm are produced on unpolished titanium surfaces. Directly on this texture, a hole-like pattern with 5 µm spatial period is generated using Direct Laser Interference Patterning with picosecond pulses. While the smaller features should reduce the bacterial adhesion, the larger geometry was designed to protect the smaller features from wear. On the multifunctional surface, the adherence of E. Coli bacteria is reduced by 30% compared to the untreated reference. In addition, wear test performed on the multiple-scale patterns demonstrated the possibility to protect the smaller features by the larger craters. Also, the influence of the laser treatment on the growth of a titanium oxide layer was evaluated using Energy Dispersive X-Ray Spectroscopy analysis.

Abstract:Abstract Zirconia implants have great application prospects due to their favorable biocompatibility and esthetic properties. However, the percentage of osseointegration of zirconia implants is lower than that of titanium implants. This study, structures bioactive surfaces on zirconia using a femtosecond laser combined with hydroxyapatite (HA) crystals and evaluates the surface morphology, functional groups, crystal phase, mechanical performance, and cell response. The results revealed that calcium phosphate deposition coincided with the establishment of micro‐grooves on the zirconia surface. The crystal composition and flexural strength of zirconia showed no significant changes after the surface treatment. Furthermore, the modified surface promoted cell adhesion and proliferation through adjustments in cell morphology. The findings suggest that the combination of femtosecond laser and HA crystals offers a clean, rapid, and controllable approach to preparing bioactive surfaces for zirconia implants.

Abstract:Cell-material interactions during early osseointegration of the bone-implant interface are critical and involve crosstalk between osteoblasts and osteoclasts. The surface properties of titanium implants also play a critical role in cell-material interactions. In this study, femtosecond laser treatment and sandblasting were used to alter the surface morphology, roughness and wettability of a titanium alloy. Osteoblasts and osteoclasts were then cultured on the resulting titanium alloy disks. Four disk groups were tested: a polished titanium alloy (pTi) control; a hydrophilic micro-dislocation titanium alloy (sandblasted Ti (STi)); a hydrophobic nano-mastoid Ti alloy (femtosecond laser-treated Ti (FTi)); and a hydrophilic hierarchical hybrid micro-/nanostructured Ti alloy [femtosecond laser-treated and sandblasted Ti (FSTi)]. The titanium surface treated by the femtosecond laser and sandblasting showed higher biomineralization activity and lower cytotoxicity in simulated body fluid and lactate dehydrogenase assays. Compared to the control surface, the multifunctional titanium surface induced a better cellular response in terms of proliferation, differentiation, mineralization and collagen secretion. Further investigation of macrophage polarization revealed that increased anti-inflammatory factor secretion and decreased proinflammatory factor secretion occurred in the early response of macrophages. Based on the above results, the synergistic effect of the surface properties produced an excellent cellular response at the bone-implant interface, which was mainly reflected by the promotion of early ossteointegration and macrophage polarization.

Abstract:Near-threshold femtosecond laser surface treatment of titanium produces functional nanostructures, which has received much attention in applications and scientific researches. However, researches on the mechanism of near-threshold temporally-shaped femtosecond laser ablation of titanium are scarce, because the difficulty of investigating the modification mechanism of surface properties by sub-pulses below the ablation threshold. In this study, the double-pulse femtosecond laser ablation with various pulse separation was investigated. We found that the maximum ablation area appeared around 1 ps pulse separation at different fluences, and the ablation enhancement weakened with the increase of the fluence. The ultrafast pump–probe microscopy technique and ultrafast thermoelasticity model were used to investigate the mechanism. The result of pump–probe microscopy technique indicated that the enhancement of the area was caused by the decrease of transient reflectivity in the early stages which facilitated the subsequent sub-pulse energy absorption. The theoretical results indicate that the maximum ablation enhancement occurred at 1 ps, which is due to the combination of thermal and mechanical effects. The maximum of long axis diameter of periodic structure also appeared at 1 ps in multi-point processing. These findings have significant implications for the modulation of titanium surface nanostructures and providing theoretical support for processing.

Abstract:The bioactivity and biocompatibility play key roles in the success of dental and orthopaedic implants. Although most commercial implant systems use various surface microstructures, the ideal multi-scale topographies capable of controlling osteointegration have not yielded conclusive results. Inspired by both the isotropic adhesion of the skin structures in tree frog toe pads and the anisotropic adhesion of the corrugated ridges on the scales of Morpho butterfly wings, composite micro/nano-structures, including the array of micro-hexagons and oriented nano-ripples on titanium alloy implants, were respectively fabricated by microsecond laser direct writing and femtosecond laser-induced periodic surface structures, to improve cell adherence, alignment and proliferation on implants. The main differences in both the bioactivity in simulated body fluid and the biocompatibility in osteoblastic cell MC3T3 proliferation were measured and analyzed among Ti-6Al-4V samples with smooth surface, micro-hexagons and composite micro/nano-structures, respectively. Of note, bioinspired micro/nano-structures displayed the best bioactivity and biocompatibility after in vitro experiments, and meanwhile, the nano-ripples were able to induce cellular alignment within the micro-hexagons. The reasons for these differences were found in the topographical cues. An innovative functionalization strategy of controlling the osteointegration on titanium alloy implants is proposed using the composite micro/nano-structures, which is meaningful in various regenerative medicine applications and implant fields.
Titanium Surfaces with a Laser-Produced Microchannel Structure Enhance Pre-Osteoblast Proliferation, Maturation, and Extracellular Mineralization In Vitro

Abstract:The clinical success of dental titanium implants is profoundly linked to implant stability and osseointegration, which comprises pre-osteoblast proliferation, osteogenic differentiation, and extracellular mineralization. Because of the bio-inert nature of titanium, surface processing using subtractive or additive methods enhances osseointegration ability but limits the benefit due to accompanying surface contamination. By contrast, laser processing methods increase the roughness of the implant surface without contamination. However, the effects of laser-mediated distinct surface structures on the osteointegration level of osteoblasts are controversial. The role of a titanium surface with a laser-mediated microchannel structure in pre-osteoblast maturation remains unclear. This study aimed to elucidate the effect of laser-produced microchannels on pre-osteoblast maturation. Pre-osteoblast human embryonic palatal mesenchymal cells were seeded on a titanium plate treated with grinding (G), sandblasting with large grit and acid etching (SLA), or laser irradiation (L) for 3–18 days. The proliferation and morphology of pre-osteoblasts were evaluated using a Trypan Blue dye exclusion test and fluorescence microscopy. The mRNA expression, protein expression, and protein secretion of osteogenic differentiation markers in pre-osteoblasts were evaluated using reverse transcriptase quantitative polymerase chain reaction, a Western blot assay, and a multiplex assay, respectively. The extracellular calcium precipitation of pre-osteoblast was measured using Alizarin red S staining. Compared to G- and SLA-treated titanium surfaces, the laser-produced microchannel surfaces enhanced pre-osteoblast proliferation, the expression/secretion of osteogenic differentiation markers, and extracellular calcium precipitation. Laser-treated titanium implants may enhance the pre-osteoblast maturation process and provide extra benefits in clinical application.

Abstract:In this paper, four array patterns were first designed by observing the structural features of the surface microstructures of pig bones and tree frog paws on the titanium alloy surface bionically. Then, the optimal parameters for laser processing were determined experimentally, and the optimized processing parameters were used to prepare micron-scale bumps on the titanium alloy surface and to investigate the relationship between the weaving height and the processing times. Finally, multiple acid etching was used to prepare nanoscale holes on the surface of the titanium alloy. It was found that the multiple acid etching could not only prepare nanoscale holes on the surface of the titanium alloy, but could also well eliminate the slag left on the surface of titanium alloy by laser ablation. Based on the above study, this paper also analyzed the effect of micro-nano structure on the friction properties of the titanium alloy surface from three aspects—theory, hardness test and friction coefficient test—and experimentally analyzed the effect of single-factor and multi-factor coupling of structural features on the friction properties of the titanium alloy surface. It was found that the optimal mean friction coefficient was 0.0902, corresponding to the characteristic values of 0.66 for the shape, 200 μm for the edge length and 60 μm for the height.

Abstract:Smart surfaces with switchable wettability have aroused enormous interest, due to their multifunction in chemistry, biology and medicine. In this paper, a controllable hydrophilic titanium surface with either micro-protrusion or micro-groove is processed by femtosecond laser. The relationship between laser processing parameters of defocus value (Df), scanning speed (Vs), power (Po) and the shape/size of structures are systematically studied. Results show that Df and Vs mainly affect the shape of structures, while Vs and Po influence size. The surface structures can change from micro-groove to micro-protrusion, for the first time, by adjusting processing parameters. After laser processing, morphology and chemical composition are analyzed. It is found that the oxygen content in the middle area of micro-protrusion is higher than the recast layer of micro-groove. The formation mechanism can be attributed to the photo-thermal effect and Marangoni convection effect. Furthermore, the surface roughness and contact angle (CA) measurement results show that the surface roughness of micro-groove array is greater than micro-protrusion array, while micro-protrusion array can promote hydrophilicity compared with micro-groove array. Moreover, the micro-protrusion array with high scanning interval (△) can maintain the hydrophilicity better than micro-groove array even after 90 days. The selected area modification of femtosecond laser processing can potentially be applied to improve biological activity in titanium implants.

Investigations on femtosecond laser-induced surface modification and periodic micropatterning with anti-friction properties on Ti6Al4V titanium alloy

Abstract:Titanium alloys have a wide application in aerospace industries as it has greater strength and low density, but it has poor tribological properties. To improve its friction and wear performance, in present work, a femtosecond laser is used to directly irradiate the Ti6Al4V titanium alloy surface in air conditioning, which results in localized ablation and the formation of periodic microstructures but also a strong pressure wave, propagating the material inside. Through the optimization of processing parameters, surface modification and periodic micropatterning with effective anti-friction properties were successfully induced on the surface. After a treatment of femtosecond laser-induced surface modification (FsLSM), the surface microhardness was improved by 16.6% and compressive residual stress reached -746 MPa. Besides, laser-induced periodic surface structures (LIPSS) with a titanium oxide outer coating were fabricated uniformly on the titanium alloy surface. Rotary ball-on-disk wear experiments revealed that the average coefficient of friction (COF) and wear mass loss of the specimen with FsLSM treatment were largely reduced by 68.9% and 90% as compared to that of untreated specimens, respectively. It was analyzed that the reason for the remarkable wear resistance was attributed to the comprehensive action of the generation of LIPSS, the titanium oxide outer coating, high amplitude compressive residual stress and gradient grain size distribution on the subsurface during the laser surface treatment. Since the findings here are broadly applicable to a wide spectrum of engineering metals and alloys, the present results offer unique pathways to enhancing the tribological performance of materials.

Abstract:Micro/nano structured surface has been shown to effectively enhance the biological performance of titanium alloy implants, exhibiting faster postoperative recovery. However, a common drawback of bio-functionalized surfaces featuring structured characteristics is their susceptibility to wear. This study proposed a novel micro/nano surface modification method using ultrasonic milling and anodization to enhance both biocompatibility and wear resistance of the implant surface. First, hierarchical structures are fabricated on the titanium surface by utilizing ultrasonic milling and anodization to generate micro-scale and nano-scale structures, respectively. Linear reciprocating tests, contact angle tests, and surface morphology observation were conducted to investigate the surface properties. Then, cell proliferative and adhesive abilities were tested on different modified surfaces by using mouse osteoblasts. Finally, nonosilver coatings were created on the structured surface by vapor deposition and the antibacterial performance was tested using staphylococcus aureus. The experiment results reveal a significant improvement in wear resistance, biological properties, and antibacterial capabilities of the processed surface, demonstrating the great application potential of the proposed method for implant surface modification.

Abstract:Laser texturing seems to be a promising technique for reducing bacterial adhesion on titanium implant surfaces. This work aims to demonstrate the possibility of obtaining a functionally orientated surface of titanium implant elements with a specific architecture with specific bacteriological and photocatalytic properties. Femtosecond laser-generated surface structures, such as laser-induced periodic surface structures (LIPSS, wrinkles), grooves, and spikes on titanium, have been characterised by XRD, Raman spectroscopy, and scanning electron microscopy (SEM). The photocatalytic activity of the titanium surfaces produced was tested based on the degradation effect of methylene blue (MB). The correlation between the photocatalytic activity of TiO2 coatings and their morphology and structure has been analysed. Features related to the size, shape, and distribution of the roughness patterns were found to influence the adhesion of the bacterial strain on different surfaces. On the laser-structurised surface, the adhesion of Escherichia coli bacteria were reduced by 80% compared to an untreated reference surface.
