Abstract:With the development of nanotechnology, it has been discovered that some nanomaterials have the activity of mimicking enzymes. This type of inorganic nanomaterial with characteristics similar to natural enzymes is called nanoenzyme. Compared with natural enzymes, nanoenzymes have advantages such as low deactivation, good stability, low production and storage costs, surface modification, and large-scale preparation. They have a wide range of applications in fields such as human health, environmental safety, and biosensing. MoS$_2$ can replace natural peroxidase to catalyze the decomposition of H$_2$O$_2$ and is considered to have peroxidase like activity, making it a typical nanoenzyme material. In addition, the Fenton reaction between Fe$_2^+$ and H$_2$O$_2$ can also cause H$_2$O$_2$ to decompose. Both MoS$_2$ and Fe$_2^+$ can cause the decomposition of H$_2$O$_2$, resulting in the production of hydroxyl radicals (OH). Hydroxyl radicals can catalyze the oxidation of chromogenic substrates 3,3',5,5'-tetramethylbenzidine (TMB), generating blue single electron oxidation products (oxTMB) and characteristic absorption peaks at 652 nm. Based on this characteristic, we doped Fe$_2^+$ into MoS$_2$ to obtain Fe-MoS$_2$ composite materials. Firstly, natural glucose oxidase is used to decompose glucose into glucose lactone and hydrogen peroxide. Then, Fe-MoS$_2$ composite material is added, and TMB is added dropwise to observe the blue turning of the mixed solution. Based on the above principle, colorimetric detection of glucose can be achieved. In addition, under infrared irradiation, the activity of peroxidase like enzymes in Fe-MoS$_2$ is enhanced.

Abstract:An electrochemical glucose biosensor was developed by immobilizing glucose oxidase (GOx) on a glass carbon electrode that was modified with molybdenum disulfide (MoS2) nanosheets that were decorated with gold nanoparticles (AuNPs). The electrochemical performance of the modified electrode was investigated by cyclic voltammetry, and it is found that use of the AuNPs-decorated MoS2 nanocomposite accelerates the electron transfer from electrode to the immobilized enzyme. This enables the direct electrochemistry of GOx without any electron mediator. The synergistic effect the MoS2 nanosheets and the AuNPs result in excellent electrocatalytic activity. Glucose can be detected in the concentration range from 10 to 300 μM, and down to levels as low as 2.8 μM. The biosensor also displays good reproducibility and long-term stability, suggesting that it represents a promising tool for biological assays.
Controllable doping of Fe atoms into MoS2 nanosheets towards peroxidase-like nanozyme with enhanced catalysis for colorimetric analysis of glucose

Abstract:MoS2 nanosheets were doped with Fe elements simply via the one-pot hydrothermal route resulting in the peroxidase-like nanozymes with dramatically enhanced catalysis. It was discovered that the resulted Fe-MoS2 could display controllable catalysis activities depending on the Mo-to-Fe molar ratios, among which the highest one was achieved at 3/1 with catalysis efficiency over three folds higher than that of pristine MoS2. The greatly improved catalysis performances of Fe-MoS2 nanozyme are thought to be resulted from the synergistic effects of the doped Fe atom and Mo-S sites. On the one hand, herein, the Fe sites might serve as the electron transferring mediators to facilitate the peroxidase-like catalytic reactions, as confirmed by the cytochrome C-based electron transportation experiments. On the other hand, the Mo-S sites might conduct the catalytic behavior of Fenton-like reactions, as revealed by the hydroxyl radicals-capturing experiments. Furthermore, the feasibility of practical applications for the developed Fe-MoS2 nanozyme based colorimetric methods was demonstrated for glucose in blood sample. Importantly, an insight can be thereby obtained into the design of different nanozymes with enhanced catalysis designed by doping heteroatoms like Fe, promising for the catalysis-based applications in the biomedical analysis, food safety, and environmental monitoring fields.

Abstract:Photoelectrochemical detection represents a unique signal transducing modality, where the photogenerated charge carriers are modulated by the redox reactions of molecular targets over the electrode surface. In this work, we report a novel glucose sensor based on hydrothermally-grown, single-crystalline TiO2 nanowires with surface-functionalized glucose oxidase (GOx), which can oxidize glucose to gluconic acid. The key feature of this glucose sensor design is that the photogenerated holes over the TiO2 anode are utilized to form O-2, which directly serves as an efficient electron shuttling mediator between the enzymatic redox center of GOx and the sensor surface, thus leading to an increase of the photocurrent. As a proof-of-concept, the GOx-functionalized TiO2 (TiO2-GOx) nanowires exhibit a high sensitivity of similar to 0.9 nM in the detection of glucose in buffer. The capability of detecting glucose in mouse serum is also demonstrated. Our work suggests the potential of developing these nanowire-based photoelectrochemical biosensors for convenient, efficient and low-cost biomarker detection and disease diagnosis.

Abstract:The authors have discovered that vanadium disulfide (VS2) nanosheets, synthesized by a hydrothermal method, exert stable peroxidase-like activity. The catalytic activity, with H2O2 as a cosubstrate, follows Michaelis-Menten kinetics and varies with temperature, pH value and H2O2 concentration. Two-dimensional VS2 sheets acting as peroxidase (POx) mimics can replace horseradish peroxidase due to their availability, robustness, and reusability. The POx-like activity of VS2 sheets was exploited to design a colorimetric glucose assay by using 3,3',5,5'-tetramethylbenzidine as a substrate and by working at an analytical wavelength of 652 nm. The assay covers the 5 to 250 μM glucose concentration range with a 1.5 μM detection limit. It was applied to the analysis of glucose in fruit juice. In our perception, the peroxidase-like nanozyme out of the family of transition metal dichalcogenides presented here has a wide scope in that it may stimulate promising biocatalytic applications in biotechnology and analytical chemistry. PtS2 nanosheets as a peroxidase-mimicking nanozyme for colorimetric determination of hydrogen peroxide and glucose

Abstract:Using an ultrasonication-assisted liquid exfoliation method, we have synthesized PtS2 nanosheets with good reproducibility. Herein, intrinsic peroxidase-like activity was for the first time demonstrated for PtS2 nanosheets, which can catalyze H2O2 oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) to generate a colored solution. The catalytic mechanism of PtS2 nanosheets was investigated, which indicated that acceleration of the electron transfer between TMB and H2O2 was the main reason for the peroxidase-like activity of PtS2 nanosheets. Based on these observations, we exploited PtS2 nanosheets integrated into dopamine-functionalized hyaluronic acid (HA-DA) hydrogel microspheres by droplet microfluidics to construct PtS2 nanosheet- and PtS2@HA-DA microsphere-based sensors for highly sensitive determination of H2O2. When coupled with glucose oxidase, we further developed two glucose sensors based on the above two methods. Among them, the linearity of the PtS2 nanosheet-based spectrophotometry was in the range of 0.5 to 150 μM and the limit of detection as low as 0.20 μM. The linearity of the microsphere-based colorimetry was in the range 200 to 12,000 μM with a detection limit of 29.95 μM. Both of the glucose sensors can be applied to the determination of glucose in human serum with reliable results and reproducibility.

Abstract:In the present work, WO3 nanosheets(WO3 NSs) were prepared by a facile method at room temperature. The obtained WO3 NSs showed peroxidase-like activity, which could catalyze 3,3',5,5'-tetramethylbenzidine(TMB) to form a blue oxidation product(ox TMB) in the presence of H2O2. Based on this, convenient and sensitive colorimetric methods for the detection of H2O2 and glucose were established. The linear ranges for detecting H2O2 and glucose were 1–200 µmol/L and 1–100 µmol/L, respectively. The limits of the detection of H2O2 and glucose were as low as 0.79 and 0.96 µmol/L, respectively. This method was also successfully applied to the detection of glucose in urine samples. The detection result was consistent with that of the value detected by the clinical method, indicating the potential in clinical diagnosis and biomedical detection.

Hemin-porous G-C3n4 Hybrid Nanosheets As an Efficient Peroxidase Mimic for Colorimetric and Visual Determination of Glucose.

Abstract:A method is described for colorimetric determination of glucose by using hemin-porous graphitic carbon nitride (g-C3N4) hybrid nanosheets as a peroxidase mimic. The porous g-C3N4 nanosheets were prepared by a combination of one-pot self-polymerization, pyrolysis and liquid-phase exfoliation. The hemin-porous g-C3N4 hybrid nanosheets were prepared via in-situ deposition. It is shown that the hybrid composite has improved dispersibility, stability, and peroxidase-mimicking activity in the 3,3′,5,5′–tetramethylbenzidine (TMB)/H2O2 system. This is deemed to be the result of the synergistic effect of hemin and porous g-C3N4 nanosheets. Based on these advantages of the nanosheets, a simple, low-cost, sensitive and selective colorimetric method was established for the determination of glucose at pH values around 7. Best performed at a wavelength of 652 nm, the assay has a linear response in the 10.0 μM to 500 μM glucose concentration range (R2 = 0.9942) and a 1.94 μM limit of detection. This method was successfully applied to the determination of glucose in (spiked) human serum samples. In our perception, the hybrid is a robust peroxidase mimic for use in POx-based assays as needed in medical diagnosis and environmental analysis.
3D V2O5-MoS2/rGO nanocomposites with enhanced peroxidase mimicking activity for sensitive colorimetric determination of H2O2 and glucose

Abstract:In this work, we reported a novel nanozyme (3D V2O5-MoS2/rGO) by decorating MoS2 nano-flowers and V2O5 nanoparticles on reduced graphene oxide (rGO). The 3D V2O5-MoS2/rGO nanocomposites exhibited intrinsic peroxidase mimicking activity and catalyzed the oxidation of 3, 3', 5, 5'-tetramethylbenzidine (TMB) to produce a blue colored product in the presence of H2O2. Compared with horseradish peroxidase (HRP), 3D V2O5-MoS2/rGO nanocomposites displayed high catalytic velocity (Vmax) and affinity (Km) for substrates (H2O2 and TMB). The study of the catalytic mechanism showed that the reduction of V5+ and the oxidation of S2- in the 3D V2O5-MoS2/rGO nanocomposites accelerate electron transfer between H2O2 and TMB, which enhanced the peroxidase mimicking activity of 3D V2O5-MoS2/rGO nanocomposites. The as-synthetized 3D V2O5-MoS2/rGO could be used for the colorimetric detection of H2O2 in the range of 20.00-800.00 μM with the LOD of 12.40 μM (3σ/S). Moreover, the 3D V2O5-MoS2/rGO could also be used for the detection of glucose in the range of 4.00-300.00 μM with the LOD of 3.99 μM (3σ/S). In addition, the as-synthetized novel peroxidase mimics has good applicability for sensitive colorimetric determination of glucose in human blood samples and artificial urine samples, and has broad application prospects as a multi-functional sensing platform in clinical diagnosis.

Abstract:In this article, for the first time, two-dimensional hybrid mesoporous Fe2O3–graphene (mFe2O3–G) nanostructures were developed as a peroxidase mimetic with catalytic activities superior to those of mFe2O3, G, and previously reported Fe-based peroxidase mimetics. The high-surface-area mFe2O3 not only offers a large number of catalytically active sites, but also facilitates the diffusion of 3,3,5,5-tetramethylbenzidine (TMB) and H2O2 toward G surface. On the other hand, G is π-rich and thus favors the adsorption and enrichment of TMB within these pores. These synergistic effects lead to highly improved catalytic performances. Based on these findings, a simple, rapid, and highly sensitive and selective optical detector of glucose has been developed and demonstrated in buffer solution with a pretty low detection limit of 0.5μM. In addition, this nanosensor is reusable and can also be used for glucose detection in diluted serum.
Peroxidase-mimetic activity of a nanozyme with uniformly dispersed Fe3O4 NPs supported by mesoporous graphitized carbon for determination of glucose

Abstract:A Fe3O4/mesoporous graphitized carbon (Fe3O4/m-GC) composite was prepared through a facile calcination method with iron-based metal–organic frameworks (Fe-MOFs) as a sacrificial template. After carbonization, the Fe3O4 nanoparticles were uniformly dispersed in the mesoporous carbon support, resulting in spatial structural stability. The mesoporous carbon support obtained was highly graphitized and exhibited eminent electrical conductivity, which accelerated the electron transfer between the Fe3O4 nanoparticles by Fe(II)/Fe(III) redox cycles and m-GC by C = Csp2/C-Csp3 redox cycles, leading to the excellent peroxidase-mimetic activity of Fe3O4/m-GC. Km values for tetramethylbenzidine (TMB) and H2O2 were 26.8 and 15.8 times lower than that of natural horseradish peroxidase, respectively. Abstract:To enhance the catalytic activity of two-dimensional layered materials as versatile materials, the modification of transition metal dichalcogenide nanosheets such as MoS2 by doping with heteroatoms has drawn great interests. However, few reports are available on the study of the enzyme-like activity of doped MoS2. In this study, a facile in situ hydrothermal method for the preparation of various ultrathin transition metals (Fe, Cu, Co, Mn, and Ni) doped MoS2 nanosheets has been reported. Through the density functional theory (DFT) and steady-state kinetic analysis, the Co-doped MoS2 nanosheets exhibited the highest peroxidase-like catalytic activity among them. Furthermore, a typical colorimetric assay for H2O2 was presented based on the catalytic oxidation of colorless 3,3′,5,5′-tetramethylbenzidine (TMB) to a blue product (oxTMB) by Co-MoS2. The proposed colorimetric method showed excellent tolerance under extreme conditions and a broad linear range from 0.0005 to 25 mM for H2O2 determination. Concerning the practical application, in situ detection of H2O2 generated from SiHa cells was also fulfilled, fully confirming the great practicability of the proposed method in biosensing fields.

Abstract:Enzymes are still indispensable for bio-assaying methods in biomolecule detection by far. The unsatisfied long-term instability, high cost, and susceptibility to the physical environment of natural enzymes are obvious weak points. Here, we developed peroxidase-like heterostructured nanozyme, vertically arraying molybdenum disulfide nanosheets on a substrate layer of nitrogen-doped reduced graphene oxide (MoS2/N-rGO), with a well-pleasing stability that is characterized by the retained enzymatic activity and maintained structure after 2 years of casual storage at ambient temperatures or 80 cycles of catalytic reaction. The catalytic kinetics of the as-prepared heterostructured nanozyme was superior to some reported nanozymes and even horse radish peroxidase, which was demonstrated due to the defect-rich MoS2 with Mo and S vacancies and nitrogen-doped rGO experimentally and theoretically. The vertically heterostructured nanozyme exhibited adequate analytical performance in sensitive and quantitative detection of glucose and glutathione (GSH), with a large dynamic sensing range and extremely low limit of detection (0.02 and 0.12 μM (3σ/slope) for glucose and GSH, respectively). We hope this inspired artificial nanozyme will contribute to the future development in sensitive detection of other biomolecules in physiological conditions.

Abstract:A facile one-step route was presented to prepared NH2-MIL-88B(Fe) and Fe3O4 modified reduced graphene oxide nanoparticles (NH2-MIL-88B(Fe)@MRGO) as mimic enzymes. The as-prepared NH2-MIL-88B(Fe)@MRGO exhibits a higher affinity for hydrogen peroxide and thus possessed a prominent peroxidase-mimic activity. Under the most favorable conditions, the hybrid nanomaterials with high catalytic velocity (2.57 × 10−7 M s−1) and affinity (Km = 0.0091 mM) for substrates of H2O2 display a great glucose detection performance in the range of 20–800 μM with a Limit of detection (LOD) of 3.16 μM (S/N = 3).

Abstract:Well-dispersed Prussian blue (PB) and Au nanoparticles (Au NPs) loaded three dimensional MoS2 nanoflowers (PB-Au@MoS2 NFs) was synthesized by a simple and economical method. The structure, morphology and composition of the hybrid were characterized by XRD, SEM and EDS. Similar to the reported literature, MoS2 nanoflowers showed peroxidase-like activity in catalyzing the oxidation of 3,3',5,5'-tetramethylbenzidine (TMB). This peroxidase-mimicking activity could be enhanced with the introduction of PB and Au NPs. Herein, PB-Au@MoS2 NFs could be used to establish a new platform for the determination of H2O2 and glucose by the chromogenic reaction. Wide linear ranges with 0-15 μM and 0-120 μM for H2O2 and glucose detection were finally obtained. The detection limits were as low as 0.25 μM and 3 μM (with signal to noise ratio of 3), respectively. The established platform was also used successfully for the determination of glucose in human serum and fruit juice samples with excellent sensitivity and stability.

Abstract:Abstract High-performance glucose biosensors are highly desired for healthcare. To meet these demands, glucose biosensors, particularly enzyme-free glucose biosensors, have received much attention. Two-dimensional materials, e.g., graphene, with high surface area, excellent electrical properties, and good biocompatibility, have been the main focus of biosensor research in the last decade. This review presents the recent progress made in enzyme-free glucose biosensors based on MoS2 nanocomposites. Two different techniques for glucose detections are introduced, with an emphasis on electrochemical glucose biosensors. Challenges and future perspectives of MoS2 nanocomposite glucose biosensors are also discussed.

Abstract:Electrochemical oxidation of glucose is the guarantee to realize nonenzymatic sensing of glucose, but greatly hindered by the slow kinetics of its oxidation process. Herein, various nanomaterials were designed as catalysts to accelerate glucose oxidation reaction. However, how to effectively build an excellent platform for promoting the glucose oxidation is still a great challenge. In our work, 1D CaMoO4 and NiMoO4 nanofibres with same morphologies and sub-microstructures were fabricated by electrospinning technique in the first time, and explored to modify the detection electrodes of nonenzymatic glucose sensors. The electrochemical results indicated that the NiMoO4 based sensor exhibited a good catalytic activity toward glucose including the low response potential (0.5 V), high sensitivity(193.8 mA mM(-1) cm(-2)) with a linear response region of 0.01-8 mM, low detection limit (4.6 mu M) and fast response time (2 s), all of which are superior to the corresponding values of CaMoO4 nanofibres and even higher than those of most reported NiO and Co3O4 catalysts, which is due to the NiMoO4 nanofibres are not only advantageous to electron transfer, but can mediated the electrocatalytic reaction of glucose. Abstract:The low activity of nanozymes, which work as an alternative to natural enzymes, limits their applications in the fabrication of biosensors, drawing increasing attention aimed at improving their catalytic capacity. In this work, the peroxidase-like activity of MoS2 nanosheets (NSs) was dramatically enhanced through DNA modification, and was 4.3-times higher than that of bare MoS2 NSs. Such an enhancement of catalytic activity was mainly ascribed to the increased affinity of the DNA/MoS2 NSs toward the substrate, TMB, further accelerating electron transfer from TMB to H2O2. On the basis of DNA-tuned MoS2 NS nanozyme activity, a colorimetric sensing platform was developed for the facile detection of carcinoembryonic antigen (CEA) in a sensitive manner. Interestingly, a convenient, affordable, and instrument-free portable test kit was fabricated to visually monitor CEA via rooting the aptamer/MoS2 NS system into an agarose hydrogel. Importantly, our work illuminates the feasibility of using DNA to enhance the catalysis of nanozymes and their application potential in the label-free, portable, and visual detection of aptamer-targeted biomolecules.
