Abstract:Zwitterionic materials have moieties possessing cationic and anionic groups. This molecular structure leads to unique properties that can be the solutions of various application problems. A typical example is that zwitterionic carboxybetaine (CB) and sulfobetaine (SB) materials resist nonspecific protein adsorption in complex media. Considering the vast number of cationic and anionic groups in the current chemical inventory, there are many possible structural variations of zwitterionic materials. The diversified structures provide the possibility to achieve many desired properties and urge a better understanding of zwitterionic materials to provide design principles. Molecular simulations and modeling are a versatile tool to understand the structure–property relationships of materials at the molecular level. This progress report summarizes recent simulation and modeling studies addressing two fundamental questions regarding zwitterionic materials and their applications as biomaterials. First, what are the differences between zwitterionic and nonionic materials? Second, what are the differences among zwitterionic materials? This report also demonstrates a molecular design of new protein‐resistant zwitterionic moieties beyond conventional CB and SB based on design principles developed from these simulation studies.

Abstract:Zwitterionic polymers have attracted research attention in recent years owing to their unique molecular structures. In the same repeat unit, positive and negative charges are simultaneously located on a pair of cationic and anionic groups; therefore, zwitterionic polymers have a large dipole moment and numerous charged groups. Although the molecular chain of the zwitterionic polymer can be maintained in an electrically neutral state overall, the coexistence of the oppositely charged groups confers extremely high polarity and excellent hydrophilicity to the polymer. At the same time, the electricality of the polymer can be further regulated by the environmental pH and salt ions, which greatly broadens the scope of applications in different fields. This review introduces various structures of zwitterionic polymers and analyzes the reasons why zwitterionic polymers exhibit pH responsiveness, anti-polyelectrolyte effects, and superior electrical conductivity. The application fields are also summarized by generalizing the research status of zwitterionic polymers, including applications in antifouling coatings, drug delivery, wastewater treatment, and sensors, etc.

Abstract:Polymer chain architectures play a crucial role in the physical properties of polymers and this unique phenomenon has been recognized as the topological effects.As one of the most representative architectures,macrocyclic polymers characterized by the endless topology have received extensive attention due to their distinct physical properties as compared to the linear counterparts.To understand these differences and unravel the underlying mechanisms,there is a long pursuit to efficiently fabricate macrocyclic polymers.To date,both ring-closing and ring-expansion strategies have been developed,which drastically elevates the accessibility of macrocyclic polymers.The improved availability of macrocyclic polymers enables the further investigation of the biomedical applications and the preliminary results suggest that macrocyclic polymers outperform their linear analogs in terms of improving gene delivery efficiency,elevating blood circulation time,and enhancing colloidal stability of nanoparticles.

Abstract:Biomedical polymers have been extensively developed for promising applications in a lot of biomedical fields, such as therapeutic medicine delivery, disease detection and diagnosis, biosensing, regenerative medicine, and disease treatment. In this review, we summarize the most recent advances in the synthesis and application of biomedical polymers, and discuss the comprehensive understanding of their property-function relationship for corresponding biomedical applications. In particular, a few burgeoning bioactive polymers, such as peptide/biomembrane/microorganism/cell-based biomedical polymers, are also introduced and highlighted as the emerging biomaterials for cancer precision therapy. Furthermore, the foreseeable challenges and outlook of the development of more efficient, healthier and safer biomedical polymers are discussed. We wish this systemic and comprehensive review on highlighting frontier progress of biomedical polymers could inspire and promote new breakthrough in fundamental research and clinical translation.

Abstract:The design of advanced functional materials with nanometer- and micrometer-scale control over their properties is of considerable interest for both fundamental and applied studies because of the many potential applications for these materials in the fields of biomedical materials, tissue engineering, and regenerative medicine. The layer-by-layer deposition technique introduced in the early 1990s by Decher, Moehwald, and Lvov is a versatile technique, which has attracted an increasing number of researchers in recent years due to its wide range of advantages for biomedical applications: ease of preparation under "mild" conditions compatible with physiological media, capability of incorporating bioactive molecules, extra cellular matrix components and biopolymers in the films, tunable mechanical properties, and spatio-temporal control over film organization. The last few years have seen a significant increase in reports exploring the possibilities offered by diffusing molecules into films to control their internal structures or design "reservoirs", as well as control their mechanical properties. Such properties, associated with the chemical properties of films, are particularly important for designing biomedical devices that contain bioactive molecules. In this review, we highlight recent work on designing and controlling film properties at the nanometer and micrometer scales with a view to developing new biomaterial coatings, tissue engineered constructs that could mimic in vivo cellular microenvironments, and stem cell "niches".
Janus Polymerization: A Novel Approach towards Topology Design

Abstract:Janus polymerization combines cationic and anionic polymerizations at two ends of a single propagating chain involving a living/controlled chain-growth polymerization and a self-triggered stepwise polycondensation, which provides a facile way to synthesize novel polymers with sophisticated topologies in one step directly from monomers. Di-/Multi-block copolymers, branch polymers, mikto-arm star copolymers and cylindrical polymer brushes are synthesized using rare earth triflates catalysts assisted by various epoxy initiators owing to their specific position at the middle of the chain. This work reviews recent progress on Janus polymerization in our group. Specifically, the characteristic of rare earth metal catalysts and mechanism of Janus polymerization are discussed in detail where tripedal crows are employed to illustrate the mechanism. New methodologies for the construction of topological polymers are introduced, together with the unique and prominent properties, such as thermoplastic properties, self-assembly and self-healing behaviors and the corresponding applications in self-sealing films for plants and electrostatic spinning. A new Janus system is put forward which affords a facile synthetic method for functional polyesters by incorporating a modifiable monomer 3,3-bis(chloromethyl)oxacyclobutane. Finally, the development of Janus polymerization including new catalytic systems and monomers is also prospected. The concept of Janus polymerization makes significant breakthroughs into traditional cognitions about incompatible cationic and anionic polymerization mechanisms in one system which motivates the power in the catalysis and synthesis of polymer chemistry.

Abstract:Surface engineering of biomedical nanoparticles is of great importance to maintain nano-stability, resist nonspecific biomolecular adsorption so as to enhance in vivo circulation time. Cell membrane mimicking zwitterions can form superhydrophilic antifouling surface which helps to maintain colloidal stability and resist immune elimination via electrostatically induced hydration. The stealthy surface prolongs the blood circulation time to reach "passive targeting", further "active targeting" is also available if combined with stimuli-responsive or bioactive molecules. Hence "zwitteration" has been developed as a new surface engineering strategy of biomedical nanoparticles. This review mainly talks about surface engineering of inorganic nanoparticles with small molecular and polymeric zwitterions, zwitterionic polymeric assemblies as drug delivery system and zwitterionic polymeric prodrugs. Introduction of special properities and applications of mixed-charge materials is also involved.

Abstract:Hyperbranched polymers (HBPs), an important subclass of dendritic macromolecules, are highly branched, three-dimensional globular nanopolymeric architectures. Attractive features like highly branched topological structures, adequate spatial cavities, numerous terminal functional groups and convenient synthetic procedures distinguish them from the available polymers (the linear, branched, and crosslinking polymers). Due to their unique physical/chemical properties, applications of HBPs have been explored in a large variety of fields. In particular, HBPs exhibit unique advantages in the biological and biomedical systems and devices. Firstly, the way to prepare HBPs usually only involves simple one-pot reactions and avoids the complicated synthesis and purification procedures, which makes the manufacturing process more convenient, thus reducing production costs. Secondly, the large number of end-groups of HBPs provides a platform for conjugation of the functional moieties and can also be employed to tailor-make the properties of HBPs, enhancing their versatility in biological applications. Thirdly, HBPs possess excellent biocompatibility and biodegradability, controlled responsive nature, and ability to incorporate a multiple array of guest molecules through covalent or noncovalent approaches. All of these features of HBPs are of great significance for designing and producing biomaterials. To date, significant progress has been made for the HBPs in solving some of the fundamental and technical questions toward their bioapplications. The present review highlights the contribution of HBPs to biological and biomedical fields with intent to aid the researchers in exploring HBPs for bioapplications.

Abstract:The catechol plays a variety of important roles in biological processes, prompting researchers to include them in the design of biomimetic biomedical materials. The low molecular weight and good water solubility of the catechol group (or its derivatives) make it a good candidate for functionalizing biomaterials that can be typically achieved by grafting it onto a polymer chain. To fully harness the powerful capabilities of catechols, one can think beyond grafting to linear polymer chains, towards hyperbranched polymers, wherein at least one of the branches comprises at least one catechol moiety. In recent years, a number of approaches have been developed to synthesize multifunctional hyperbranched polymers with catechol functionalities for bioadhesives and surface coatings. This review article focuses on the main synthetic approaches applied for introducing the important and versatile catechol building blocks within the (hyper)branched structure polymers. In addition, the applications of these polymers in biomedical fields are highlighted as well.

Abstract:This review summarizes recent research on the design of polymer material systems based on biomimetic concepts and reports on the medical devices that implement these systems. Biomolecules such as proteins, nucleic acids, and phospholipids, present in living organisms, play important roles in biological activities. These molecules are characterized by heterogenic nature with hydrophilicity and hydrophobicity, and a balance of positive and negative charges, which provide unique reaction fields, interfaces, and functionality. Incorporating these molecules into artificial systems is expected to advance material science considerably. This approach to material design is exceptionally practical for medical devices that are in contact with living organisms. Here, it is focused on zwitterionic polymers with intramolecularly balanced charges and introduce examples of their applications in medical devices. Their unique properties make these polymers potential surface modification materials to enhance the performance and safety of conventional medical devices. This review discusses these devices; moreover, new surface technologies have been summarized for developing human-friendly medical devices using zwitterionic polymers in the cardiovascular, cerebrovascular, orthopedic, and ophthalmology fields. Abstract:Nanocarriers play an important role in drug delivery for disease treatment. However, nanocarriers face a series of physiological barriers after administration such as blood clearance, nonspecific tissue/cell localization, poor cellular uptake, and endosome trapping. These physiological barriers seriously reduce the accumulation of drugs in target action site, which results in poor therapeutic efficiency. Although polyethylene glycol (PEG) can increase the blood circulation time of nanocarriers, its application is limited due to the ″PEG dilemma″. Zwitterionic polymers have been emerging as an appealing alternative to PEG owing to their excellent performance in resisting nonspecific protein adsorption. Importantly, the diverse structures bring functional versatility to zwitterionic polymers beyond nonfouling. This review focuses on the structures and characters of zwitterionic polymers, and will discuss and summarize the application of zwitterionic polymers for drug delivery. We will highlight the strategies of zwitterionic polymers to address the physiological barriers during drug delivery. Finally, we will give some suggestions that can be utilized for the development of zwitterionic polymers for drug delivery. This review will also provide an outlook for this field. Our aim is to provide a comprehensive and systemic review on the application of zwitterionic polymers for drug delivery and promote the development of zwitterionic polymers.

Abstract:Polyampholyte polymers are a subclass of zwitterionic materials that are composed of mixtures of charged monomer subunits. These materials have properties that mimic those of the more widely characterized zwitterionic polymers as long as their polymer chemistry has been well controlled. In particular, overall neutral polyampholyte polymers have been shown to have resistance to nonspecific protein adsorption and protein conjugation capability. This has been demonstrated for both polymer brush and hydrogel-based platforms. In this review, our recent work demonstrating these characteristics will be summarized and placed into the broader context of zwitterionic materials.

Abstract:Zwitterionic polymers by virtue of their unique chemical and physical attributes have attracted researchers in recent years. The simultaneous presence of positive and negative charges in the same repeat unit renders them of various interesting properties such as superhydrophilicity, which has significantly broadened their scope for being used in different applications. Among polyzwitterions of different architectures, micro- and/or nano-gels have started receiving attention only until recently. These 3D cross-linked colloidal structures show peculiar characteristics in context to their solution properties, which are attributable either to the comonomers present or the presence of different electrolytes and biological specimens. In this review, a concise yet detailed account is provided of the different synthetic techniques and application domains of zwitterion-based micro- and/or nanogels that have been explored in recent years. Here, the focus is kept solely on the "polybetaines," which have garnered maximum research interest and remain the extensively studied polyzwitterions in literature. While their vast application potential in the biomedical sector is being detailed here, some other areas of scope such as using them as microreactors for the synthesis of metal nanoparticles or making smart membranes for water-treatment are discussed in this minireview as well.

Abstract:Currently, many types of non-linear topological structure polymers, such as brush-shaped, star, branched and dendritic structures, have captured much attention in the field of gene delivery and nanomedicine. Compared with linear polymers, non-linear topological structural polymers offer many advantages, including multiple terminal groups, broad and complicated spatial architecture and multi-functionality sites to enhance gene delivery efficiency and targeting capabilities. Nevertheless, the complexity of their synthesis process severely hampers the development and applications of nonlinear topological polymers. This review aims to highlight various synthetic approaches of non-linear topological architecture polymers, including reversible-deactivation radical polymerization (RDRP) including atom-transfer radical polymerization (ATRP), nitroxide-mediated polymerization (NMP), reversible addition-fragmentation chain transfer (RAFT) polymerization, click chemistry reactions and Michael addition, and thoroughly discuss their advantages and disadvantages, as well as analyze their further application potential. Finally, we comprehensively discuss and summarize different non-linear topological structure polymers for genetic materials delivering performance both in vitro and in vivo, which indicated that topological effects and nonlinear topologies play a crucial role in enhancing the transfection performance of polymeric vectors. This review offered a promising guideline for the design and development of novel nonlinear polymers and facilitated the development of a new generation of polymer-based gene vectors.

[Shape-memory polymers for biomedical engineering applications]

The mechanism responsible for shape memory effect was analyzed in reference to the polymer structure. Also introduced and reviewed were the characteristics of some shape-memory polymers (polyurethane polycaprolactone and polylactide) and their applications in medical engineering.

Abstract:The rapid development of supramolecular polymer chemistry and constitutional dynamic chemistry over the last decades has made tremendous impact on the emergence of dynamic covalent polymers. These materials are formed through reversible covalent bonds, endowing them with adaptive and responsive features that have resulted in high interest throughout the community. Owing to their intriguing properties, such as self-healing, shape-memory effects, recyclability, degradability, stimuli-responsiveness, etc., the materials have found multiple uses in a wide range of areas. Of special interest is their increasing use for biomedical applications, and many examples have been demonstrated in recent years. These materials have thus been used for the recognition and sensing of biologically active compounds, for the modulation of enzyme activity, for gene delivery, and as materials for cell culture, delivery, and wound-dressing. In this review, some of these endeavors are discussed, highlighting the many advantages and unique properties of dynamic covalent polymers for use in biology and biomedicine.
