Abstract:A photoelectrochemical (PEC) biosensor based on SnO2 nanoparticles (SnO2 NPs) was developed and applied for phosphatidylcholine (PC) detection in soybean oil. SnO2 NPs were grown on an indium tin oxide (ITO) electrode, polythionine (PTh) was electropolymerized on the surface of ITO/SnO2 NPs, and choline oxidase (ChOx) was immobilized to prepare the ITO/SnO2 NPs/PTh/ChOx electrode. The developed PEC biosensor can detect PC under visible light irradiation. The experimental conditions for PC detection were as follows: 1.8 mg mL-1 ChOx concentration, 0.5 V bias voltage, 18 mW cm-2 light intensity, and pH 6. The PEC biosensor had a detection limit of 0.005 mM (S/N = 3) and a detection range from 0.03 mM to 4 mM. This PEC biosensor based on SnO2 NPs was applied to detect PC in soybean oil. The recovery rate tested by the standard addition method was 95.2-107.4%. These findings were consistent with the results obtained by high-performance liquid chromatography (HPLC). Therefore, the proposed PEC biosensor based on SnO2 NPs has excellent reproducibility, stability, and great potential applications in the PEC analysis of PC in soybean oil.

Abstract:A bienzyme photoelectrochemical (PEC) biosensor was explored for the sensitive determination of phosphatidylcholine (PC) in vegetable oil based on phospholipase D (PLD) and choline oxidase (ChOx) immobilized on an indium tin oxide (ITO) electrode. The bienzyme electrode is based on the modification with SnO2 nanoparticles (NPs) (SnO2 NPs) and polythionine (PTh). SnO2 NPs were obtained by using a hydrothermal method. Electropolymerization was performed to load the PTh layer on the ITO/SnO2 NPs electrode surface. PLD and ChOx were immobilized on the surface of modified ITO electrode using embedding and cross-linking methods. The prepared ITO/SnO2 NPs/PTh/ChOx/PLD electrode was finally applied as bienzyme PEC biosensor for PC detection. Under optimized conditions, a good linear relationship was obtained in the range of 0.01 mmol/L to 5 mmol/L with a detection limit of 0.002 mmol/L (S/N = 3). The proposed PEC biosensor exhibited remarkable reproducibility, selectivity, and stability. The prepared bienzyme biosensor was used to detect PC in soybean oil by acceptable recoveries. The results were consistent with those of liquid chromatography, confirming the suitability of this PEC sensing strategy for the rapid and sensitive determination of PC in vegetable oil.
Determination of phospholipids in soybean oil using a phospholipase-choline oxidase biosensor based on g-C3N4-TiO2 nanocomposite material

Abstract:In this paper, we constructed a photoelectrochemical biosensor with ITO electrode as the reaction platform, g-C3N4 and TiO2 as the reaction substrate, Nafion solution as the cross-linking agent, dropwise addition of choline oxidase (ChOx) and optimized its preparation and detection conditions, and finally used this photoelectrochemical biosensor for the detection of Phosphatidylcholine (PC) in actual soybean gross oil. The experiments showed that the PC concentration showed a good linear relationship with the photocurrent response value in the range of 0.05–280 mg/L, and the limit of detection was 0.05 mg/L (S/N = 3). Under the optimized conditions, the sensor had the highest photocurrent response value and the best sensitivity for the artificially simulated gross oil; Phosphatidyl ethandamine (PE), Phosphatidic acid (PA) and Phosphatidyl inositol (PI) showed no significant interference for the detection of PC, and the sensor showed high selectivity for PC detection. The results showed that the relative photocurrent value of the sensor remained at a high level during the storage time of about 32 days, which proved that the electrode storage stability was good, and provided a theoretical basis for the establishment of a fast, accurate and low-cost method for the determination of the PC content, which could more accurately reflect the rule of change of PC in the catalytic reaction of soybean oil degumming, and further realise the precise and moderate processing of fats and oils.

Abstract:In the field of oil refining, the presence of excessive residual phosphorus in crude oil can significantly impact its quality, thereby emphasizing the necessity for compact and convenient testing equipment. This study primarily focuses on developing of self-powered biosensor (SPB) using immobilizing Choline Oxidase with a photoactive ternary nanocomposite complex (CHOx-BiOI-rGO-Fe3O4 NPs-ITO) as the anode and utilizing a Pt electrode as the cathode. The successful preparation of the ternary composite photoelectrode for the anode was confirmed through a range of characterization techniques, including X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), Transmission electron microscopy (TEM), Scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), N2 absorption/desorption, Dynamic light scattering (DLS), and Ultraviolet-visible diffuse reflection spectrometer (UV-vis DRS). The electrochemical and photoelectrochemical properties were assessed using an electrochemical workstation, revealing a significant enhancement photoelectrical responsiveness attributed to the formation of heterojunction structures. The SPB exhibited a remarkable linear relationship between the instantaneous photocurrent and phosphatidylcholine (PC) concentration, with a regression equation of I (μA) = 39.62071C (mM) + 3.47271. The linear range covered a concentration range of 0.01-10 mM, and the detection limit (S/N = 3) was determined to be 0.008 mM. It demonstrated excellent reproducibility and storage stability, positioning it a promising alternative to High-performance liquid chromatography (HPLC) for accurate quantification of PC content in rhodotorula glutinis oil. The standard recovery PC content ranged from 98.48% to 103.53%, with a relative standard deviation (RSD) ranging from 1.4% to 2.4%. This research presents a convenient and precise detection device that has the potential to address the issue of lagging detection in the oil refining process.

Abstract:Owing to their large specific surface, favorable electrical conductivity, and excellent electrocatalytic capabilities, two-dimensional transition metal dichalcogenides have received considerable attention in the field of biosensors. On the basis of these properties, we developed a portable and disposable enzyme-based biosensor for paraoxon detection using a metallic MoS2 nanosheets modified screen-printed electrode (SPE). The exfoliated ultrathin metallic MoS2 nanosheets can accelerate the electron transfer on electrode surface and contribute to the immobilization of acetylcholinesterase (AChE) via the cross-linking of glutaraldehyde. Electrodeposited gold nanoparticles (AuNPs) on SPE were used to immobilize MoS2 nanosheets through the interaction between Au atoms on AuNPs and S atoms on MoS2. Using acetylcholine as the substrate, AChE can catalyze the formation of electroactive thiocholine and further generate the redox current. In the presence of paraoxon, the activity of AChE can be inhibited, making the related electrochemical signals weaken. Under the optimized conditions, this electrochemical biosensor exhibited a favorable linear relationship with the concentration of paraoxon from 1.0 to 1000 μg L-1, with the detection limit of 0.013 μg L-1. Furthermore, this developed biosensor was successfully applied to detect paraoxon in pretreated apple and pakchoi samples, which can provide a reliable method for the rapid analysis of organophosphorus pesticides in agricultural products.

Abstract:A novel, rapid and convenient competitive immunoassay for ultrasensitive detection of chloramphenicol residues in shrimp and honey was established combined with flow injection chemiluminescence. The carboxylic resin beads were used as solid phase carriers to load with more coating antigen due to their larger specific surface area and good biocompatibility. The surface of the silica dioxide nanoparticles was modified with aldehyde group to combine with more horseradish peroxidase and the chloramphenicol antibody. There was a competitive process between the chloramphenicol in solution and the immobilized coating antigen to combine with the limited binding site of antibody to form the immunocomplex. Silica dioxide nanoparticles played an important role in enhancing chemiluminescence signal, because the horseradish peroxidase on SiO2 effectively catalyzed the system of luminol-PIP-H2O2. Under optimal conditions, the chemiluminescence intensity decreased linearly with the logarithm of the chloramphenicol concentration in the range of 0.0001 to 100 ng mL−1 and the detection limit (3σ) was 0.033 pg mL−1. This immunosensor demonstrated acceptable stability, high specificity and reproducibility. The horseradish peroxidase-silica dioxide nanoparticle-chloramphenicol antibody complex successfully prepared in this article was firstly applied to the detection of chloramphenicol, and had extremely important meanings for the application of nanoparticles and enzymatic catalysis in the field of chemiluminescence.
Overoxidized polypyrrole / overoxidized poly(3, 4-ethylenedioxythiophene) / graphene quantum dots-chitosan as enhanced electrochemical sensing platform for luteolin determination in peanut shells

Abstract:A novel electrochemical sensor was constructed based on an overoxidized polypyrrole / overoxidized poly(3, 4-ethylenedioxythiophene) /graphene quantum dots-chitosan composite films modified glass carbon electrode (oPPy/oPEDOT/ GQDs-CS/GCE), for the accurate determination of luteolin in peanut shells. The oPPy/ oPEDOT/ GQDs-CS/GCE was prepared by layer-by-layer assembly methods. After incubation in NaOH-EtOH solution, the current of luteolin on the composite electrode increased from 0 to ~30 μA, the Rct value decreased by ~80 fold, and the specific surface area (A) increased by ~7 fold. Based on a three-level four-factor Box-Behnken Design (BBD), the optimal preparation conditions for the composite electrode were as follows: mass ratio of CS to GQDs of 5, polymerization cycle number for EDOT of 4, polymerization cycle number for Py of 4, with peak current increased to 43.21 μA (RSD=3.51%, n=3). A morphology with multi-channels and grooves was observed on the surface of the incubated composite film by SEM analysis. Additional -C=N- and -SO/-SO2 groups formed were observed by XPS and FTIR analysis. The electrochemical sensor constructed exhibited good anti-interference ability, repeatability, and stability. The linear detection range recorded by the current responses of the composite electrode to luteolin was 0.01 to 10.0 μM (R2=0.991) and the limit of detection was 7.5 nM (S/N=3). The observed content of luteolin in peanut shell was 0.769% , which was close to the result from High Performance Liquid Chromatography (HPLC) analysis (0.808%). This agreement indicated that the constructed electrochemical sensor based on oPPy/oPEDOT/GQDS-CS/GCE was capable of accurate detection of luteolin in peanut shells.

Abstract:Photoelectrochemical(PEC) biosensors have shown great promise in bioanalysis and diagnostic applications in recent years. In this work, the CuO/Cu 2 O nanowire array(CuO/Cu 2 O Nanowire) supported on copper foam was prepared as a photocathode for detection of tyrosinase though quinone-chitosan conjugation chemistry method. The in-situ generated quinones that were the catalytic product of tyrosinase acted as electron acceptors, which were captured by the chitosan deposited on the surface of the electrode. Direct immobilization of electron acceptor on the electrode surface improved the photocurrent conversion efficiency and thus sensitivity. The as-prepared biosensor can realize a rapid response in a wide linear range of 0.05 U/mL to 10 U/mL with the detection limit as low as 0.016 U/mL of tyrosinase. The current work provides a new perspective to design and develop highly sensitive and selective PEC biosensor.

Abstract:A sensitive photoelectrochemical (PEC) biosensor for detection of organophosphorus pesticides (OPs) using the nanocomposite of CdSe@ZnS quantum dots (QDs) and graphene deposited on the ITO coated glass electrode as a photoactive electrode is presented. The integration of CdSe@ZnS/graphene nanocomposite with biomolecules acetylcholinesterase (AChE) as a biorecognition element yields a novel biosensing platform. Under visible light irradiation, the AChE-CdSe@ZnS/graphene nanocomposite can generate a stable photocurrent and the photocurrent is found to be inversely dependent on the concentration of OPs. Under the optimal experimental conditions, the photocurrents were proportional to the logarithm of paraoxon and dichlorvos within the concentration range of 10(-12)-10(-6) M. The detection limits (LOD) of the proposed biosensor for paraoxon and dichlorvos are as low as 10(-14) M and 10(-12) M. The photoelectrochemical biosensor shows good sensitivity, reproducibility, stability, and could be successfully applied to detection of OPs in real fruit samples.

Abstract:A novel photoelectrochemical (PEC) sensor based on the sensitive interface of photosensitive electrode has been developed for the enzyme-free PEC response of hydrogen peroxide (H2O2). Polythionine (PTh) was polymerized on tin dioxide nanoparticles (SnO2 NPs) modified Indium Tin Oxide (ITO) electrode by scanning voltammetry. Characterization by scanning electron microscopy, Fourier transform infrared spectroscopy, and ultra-violet-visible spectroscopy showed that PTh was successfully polymerized on the surface of the photosensitive electrode and did not change the structure of SnO2 NPs. The PTh modified SnO2 NPs photosensitive electrode has excellent photoelectric performance, further enhancing the photocurrent response of H2O2. The calibration curve was linear in the range of 0.03-8 mM with a detection limit of 0.003 mM. The PEC sensor was successfully applied to detect H2O2 residues in dried bean curds. The results showed no remarkable difference compared with the reference iodometry method, suggesting that this alternative method can be used for the quality control analysis of dried bean curds.

Abstract:In this study, a novel organic photophotochemical transistor (OPECT) biosensing platform was proposed for dual-mode detection of CEA. The dual-mode detecting system is achieved benefit from the exceptional photoelectric performance of MIL-53(Fe) -NH 2 @ZnIn 2 S 4 (MNZ) and the peroxidase enzyme (POD) activity of ZIF-67/Cu 0.76 Co 2.24 O 4 (ZIF-67/CuCoO). Ab2- ZIF-67/CuCoO probe was immobilized on a 96-well plate by enzyme-linked immunosorbent assay, which accelerated the oxidation of 3,3,5,5-tetramethylbenzidine (TMB) by hydrogen peroxide and thus successfully realized visual detection of CEA. Additionally, in OPECT biosensing mode, the oxidized form of TMB (oxTMB·) serves as a consumptive agents of the electron donor AA, which cause the photocurrent change of MNZ heterojunction, leading to a decrease in the channel current of poly (ethylenedioxythiophene): poly (styrene sulfonate) (PEDOT: PSS) organic transistor. The integration of nanobiocatalysts and the OPECT system demonstrates excellent detection performance for CEA, with a detection limit as low as 3.24 fg mL −1 and expanded their prospective applications for clinical detection of nucleic acids, proteins, and other tumor markers.

Highly-sensitive organophosphorous pesticide biosensors based on nanostructured films of acetylcholinesterase and CdTe quantum dots

Abstract:The optical transducer of CdTe semiconductor quantum dots (QDs) has been integrated with acetylcholinesterase enzyme (AChE) by the layer-by-layer (LbL) assembly technique, resulting in a highly sensitive biosensor for detection of organophosphorus pesticides (OPs) in vegetables and fruits based on enzyme inhibition mechanism. The detection limits of the proposed biosensors are as low as 1.05 × 10(-11) M for paraoxon and 4.47 × 10(-12) M for parathion, which are significantly better than those of the conventional GC/MS methods or amperometric biosensors (0.5 nM). These biosensors are used for quick determination of low concentrations of OPs in real vegetable and fruit samples and exhibit satisfactory reproducibility and accuracy. Moreover, the stock stability of the biosensors are very good due to the stabilizing environment for the enzyme in the nanostructures made by LbL technique. Many advantages provided by these biosensors, like fluorescent change recognized by naked eyes and mass production with low cost, will facilitate future development of rapid and high-throughput screening of OPs.
Simplifying the complexity: Single enzyme (choline oxidase) inhibition-based biosensor with dual-readout method for organophosphorus pesticide detection

Abstract:Organophosphorus pesticides (OPs) are widely used in agricultural production, but their residues could cause pollution to the environment and living organisms. In this paper, a simple dual-readout method for OPs detection was proposed based on ChOx single enzyme inhibition. Firstly, ChOx can catalyze the production of H 2 O 2 from choline chloride (Ch-Cl). Bifunctional iron-doped carbon dots (Fe-CDs) with good peroxidase-like activity and superior fluorescence properties can catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) to blue oxidized TMB (oxTMB) by H 2 O 2 formed, and oxTMB could quench the fluorescence of Fe-CDs. In light of the fact that OPs exhibited activity in inhibiting ChOx, less H 2 O 2 and the decreasing oxTMB led to a result that the fluorescence of the system recovered and the solution became lighter in blue color. Moreover, the process of ChOx inhibition by OPs was analyzed by molecular docking technique and it was found that OPs interact with key amino acid residues catalyzed by ChOx (Asn510, His466, Ser101, His351, Phe357, Trp331, Glu312). Finally, a dual-mode (colorimetry and fluorescence) sensor was created for the detection of OPs with the detection limit of 6 ng/L, and was successfully used in the quantitative determination of OPs in actual samples with satisfactory results.

Abstract:The photoelectrochemical properties of free-base-porphyrin-functionalized zinc oxide nanoparticles were studied. A universal photoelectrochemical biosensing platform was constructed on indium tin oxide (ITO) by using the functional nanohybrid. The nanohybrid was synthesized by means of dentate binding of ZnO nanoparticles with carboxylic groups of 4,4', 4 ", 4 "'-(21H, 23H-porphine-5,10,15,20-tetrayl)tetrakis(benzoic acid) (TCPP), and characterized with scanning electron microscopy, contact angle measurement, and spectral techniques. The nanohybrid-coated ITO electrode showed an efficient photocurrent response under irradiation at a wavelength of 360 nm, which could be greatly improved upon addition of cysteine by its oxidation at + 0.3 V. The possible mechanism was that cysteine acts as a sacrificial electron donor to scavenge the photogenerated holes that locate on the excited state of TCPP, which then injects the photoexcitation electrons into the conduction band of ZnO nanoparticles, thereby transferring photoinduced electrons to the ITO electrode. Based on this enhanced photocurrent signal, a novel method for photoelectrochemical detection of cysteine was developed with a linear range of 0.6 to 157 mu molL(-1) in physiological media. The detection limit was 0.2 mu molL(-1) at a signal-to-noise ratio of 3. The novel strategy of cysteine analysis could provide an alternative method for monitoring biomolecules and extend the application of porphyrin-functionalized semiconductor nanoparticles.
In-situ growth of SnO2 nanoparticles on Nb2CTx nanosheets as highly sensitive electrochemical sensing platform for organophosphorus pesticide detection

Abstract:In this study, the SnO2/Nb2CTx MXene nanocomposite containing 0D/2D interfaces was prepared by situ growth strategy of one-step hydrothermal method. A SnO2/Nb2CTx MXene based acetylcholinesterase (AChE) biosensor was constructed for pesticide detection. Highly conductive Nb2CTx MXene, acting as substrate material, restrained the agglomeration of nanoparticles (NPs) and accelerated electron migration due to the confinement effect and well-known accordion-like layered structure. In addition, SnO2 anchored on both sides of the Nb2CTx MXene nanosheets effectively provided a large surface area, abundant surface groups and active sites, which preserved numbers of electrons at the interface of the heterojunction. The SnO2/Nb2CTx MXene hybrids with outstanding conductivity, good biocompatibility and structural stability were beneficial for AChE immobilization. Under the optimized conditions, as-fabricated electrochemical biosensor demonstrated superior performance with linear detection range of 5.1 × 10-14 - 5.1 × 10-7 M for chlorpyrifos, along with the limit of detection (LOD) down to 5.1 × 10-14 M (calculated for 10% inhibition). Furthermore, it is highly expected that this biosensor can be applied for the detection of other organophosphorus pesticides in the environment, providing an effective nanoplatform in biosensing field.

Abstract:A novel visible light photoelectrochemical (PEC) platform coupled with enzyme-inhibition for rapid and sensitive determination of organophosphates (OPs) was constructed based on a dual-functional Cd0.5Zn0.5S-reduced graphene oxide (Cd0.5Zn0.5S-rGO) nanocomposite. Due to the inherent biocompatibility of the Cd0.5Zn0.5S-rGO nanocomposite, acetylcholinesterase (AChE) immobilized on the Cd0.5Zn0.5S-rGO modified electrode can hydrolyze acetylthiocholine chloride into thiocholine, which could increase the photocurrent of the enzyme electrode, and the further inhibition of OPs on the enzyme electrode could decrease the photocurrent response. Based on the notable change in the PEC response of the AChE-Cd0.5Zn0.5S-rGO modified electrode and using Dursban as a model, a simple and effective way for PEC monitoring of OPs is proposed, which showed a wide linear range of 0.001-1 μg mL(-1) with a low detection limit of 0.3 ng mL(-1) (S/N = 3). Moreover, the biosensor was successfully challenged with water samples, demonstrating a new method for rapid and sensitive screening/evaluating exposure to organophosphorus pesticides and other hazardous substances.
MOF-derived nanozyme CuOx@C and its application for cascade colorimetric detection of phytosterols

Abstract:Phytosterols (PSs), a class of naturally occurring bioactive lipid compounds, have been found to possess a significant cholesterol-lowering effect. In developing countries, the consumption of rapeseed oil is the primary pathway of PS intake for the general population. However, developing low-cost, real-time, and high-throughput screening techniques for PSs remains a challenge. Here, a Cu-based nanocomposite CuOx@C was synthesized via a simple method of the calcination of HKUST-1 and systematically characterized by scanning electron microscopy, transmission electron microscopy, X-ray diffraction, and X-ray photoelectron spectroscopy. The CuOx@C demonstrated excellent peroxidase-like (POD-like) activity, functioning as a peroxidase mimic to facilitate the catalysis of 3,3',5,5'-tetramethylbenzidine (TMB) into its oxidized form (oxTMB), thereby initiating a discernible color response. On the basis of this discovery, a CuOx@C-based colorimetric method for detecting total sterols in rapeseed was successfully constructed via cascade reactions. After optimizing the conditions, the high-throughput screening of total sterols in rapeseed could be completed in only 21 min, which significantly facilitated the sensing of PSs. A linear range of 0.6-6 mg/g was achieved for the detection of total sterols in rapeseed samples, thereby satisfying the requirements for detection. In addition, due to the high stability of CuOx@C and the specificity of cholesterol oxidase, the developed method had excellent stability and selectivity toward PSs, indicating that this work has huge prospects for commercial application. This innovative work overcomes the limitation of the instrumental method and provides a portable and reliable tool for total sterols detection. It can also facilitate the development of oilseeds with a high content of PSs.

Abstract:The photoelectrochemical behavior of SnO2 nanoporous film electrode sensitized from composite PbS/RuL2 (NCS)2 was investigated by photoelectrochemical methods and UV - vis absorption spectroscopy. The results showed that the photosensitization of composite PbS and RuL2(NCS)2 for SnO2 nanoporous film electrode largely improved the conversion efficiency of light to electricity and was superior to the photosensitization of PbS or of RuL2(NCS)2 respectively. The charge transfer mechanism of compositely sensitized electrode was discussed in detail.

Abstract:The accurate determination of organophosphorus pesticide residues is of great importance for human disease monitoring and environmental safety. Numerous detection methods exist, among which sensitive monitoring of organophosphorus compounds using electrochemical sensors has gradually become a research hotspot. This paper used acetylcholinesterase (AChE) as an indicator anchored on a zinc oxide-reduced graphene oxide (ZnO-rGO) composite rich in active sites, in which green non-toxic zinc oxide (ZnO) nanomaterials were uniformly distributed on the reduced graphene for rapid detection of organophosphorus. The effects of different ratios of ZnO to reduced graphene on the performance of ZnO-rGO nanocomposites were investigated. The AChE/ZnO-rGO biosensor detects organophosphorus by electrochemical inhibition of acetylcholinesterase in the presence of organophosphorus. The developed electrochemical biosensor has high selectivity and good linearity, and the ZnO-rGO nanocomposite as a matrix for immobilization of acetylcholinesterase and detection of organophosphorus has the potential for highly sensitive pesticide detection.
