Abstract:Enzymatic biosensor (EBS) is the most promising substitute for traditional analytical methods on determination of organophosphorus pesticides (OPs). The merits of simple, inexpensive and miniature make it potential for environmental monitoring. Enzymatic determination of OPs is often based on cholinesterase (acetylcholineaterase and butyrylcholinesterase) and organophosphorus hydrolase. Various methods have been applied for enzyme immobilization in the construction of EBS for determination of OPs. Since the chemical and physical properties of the polymers may be tailored for particular needs, they gained importance as the substrate for enzyme immobilization. Application of single-use screen-printed enzymatic electrodes are most intensively developed procedures.

Abstract:A novel electrochemical microbial biosensor for the rapid monitoring of p-nitrophenyl-substituted organophosphates (OPs) compounds based on glass carbon electrode (GCE) modified with both ordered mesopore carbons (OMCs) and cell surface-expressed organophosphorus hydrolase (OPH) (OPH-bacteria/OMCs/GCE) was described in this paper. The genetically engineered Escherichia coli strain surface displayed mutant OPH (S5) with improved enzyme activity and favorable stability was constructed using a newly identified N-terminal of ice nucleation protein as an anchoring motif, which can be used directly without further time-consuming enzyme-extraction and purification, thereafter greatly improved the stability of the enzyme. Compared to OPH-bacteria modified GCE (OPH-bacteria/GCE), the OPH-bacteria/OMCs/GCE not only significantly enhanced the current response but also reduced the oxidation overpotential towards oxidizable p-nitrophenol (p-NP), which was the hydrolysate of p-nitrophenyl-substituted OPs. Under the optimized experimental conditions, at +0.84V (vs. SCE), the current–time curve was performed with varying OPs concentration. The current response was linear with paraoxon concentration within 0.05–25μM. Similarly, linear range of 0.05–25μM was found for parathion, and 0.08–30μM for methyl parathion. The low limits of detection were evaluated to be 9.0nM for paraoxon, 10nM for parathion and 15nM for methyl parathion (S/N=3). Thus, a highly specific, sensitive and rapid microbial biosensor was established, which holds great promise for on-site detection of trace p-nitrophenyl-substituted OPs.
Functional Cell Surface Displaying of Acetylcholinesterase for Spectrophotometric Sensing Organophosphate Pesticide

Abstract:The inhibition of acetylcholinesterase (AChE) activity based methods have been widely used for rapid detection of organophosphate (OP) pesticide residues. However, the sensitivity of the enzyme toward pesticides is not desirable and complex purification steps increase production cost. In this study, we optimized and synthesized the cDNA of carp AChE based on the preferred codon using Saccharomyces cerevisiae, which was ligated into expression vector pYD1. The AChE was successfully presented on yeast surface through a-agglutinin mediated display system. In shake-flask after induction with galactose in YPD medium for 24 h, this strain expressed AChE activity at a high level of 0.15 U/OD600/ml. The correct location of AChE was confirmed by immunofluorescence analysis. The whole cell catalyst was applied to visible spectrophotometry detection for OP pesticides based on inhibition rate. The inhibition rate was linear with paraoxon concentration in the range of 0.5 ng/ml-10 mu g/ml and parathion concentration in the range of 5 ng/ml-10 mu g/ml. The low detection limit was 0.136 ng/ml paraoxon and 3.72 ng/ml parathion (S/N = 3). The proposed method is applicable to analyzing OP pesticides in real samples. Therefore, the recombinant strain developed could be an efficient and affordable biomaterial for rapid and accurate detection of OP pesticide residues.
An Ultra-Sensitive Acetylcholinesterase Biosensor Based on Reduced Graphene Oxide-Au Nanoparticles-Β-cyclodextrin/prussian Blue-Chitosan Nanocomposites for Organophosphorus Pesticides Detection.

Abstract:This work reports a novel, ultrasensitive, and selective sensing platform based on a direct electrodeposition of electrochemical reduced graphene oxide (ERGO)-Au nanoparticles (AuNPs)-β-cyclodextrin (β-CD) and Prussian blue-chitosan (PB-CS) on glass carbon electrode (GCE) for efficiently fixed acetylcholinesterase (AChE) to fabricate organophosphorus pesticides (OPs) biosensor. The PB-CS not only effectively catalyzed the oxidation of thiocholine (TCh), but also shifted its oxidation potential from 0.68 to 0.2V, and accordingly the sensitivity of the biosensor was obviously improved. The synergistic effect between ERGO and AuNPs significantly promoted the electron transfer between PB and GCE, and remarkably enhanced the electrochemical oxidation of TCh. Besides, β-CD could interact with substrate by reversible bonding, which is contribute to increase the enrichment of the substrate and improve the selectivity and sensitivity of the biosensor. The integration of ERGO-AuNPs-β-CD with PB-CS provided an advantageous and high-performance platform for sensing applications. Based on the inhibition of OPs on AChE activity, the sensor showed wide linear ranges of 7.98−2.00×103pgmL−1 and 4.3−1.00×103pgmL−1 with low detection limits of 4.14pgmL−1 and 1.15pgmL−1 for malathion and carbaryl, respectively. The proposed biosensor exhibited short response time, good stability and high sensitivity, which can be used for direct analysis of practical samples.

Abstract:The accurate determination of organophosphorus pesticide residues is of great importance for human disease monitoring and environmental safety. Numerous detection methods exist, among which sensitive monitoring of organophosphorus compounds using electrochemical sensors has gradually become a research hotspot. This paper used acetylcholinesterase (AChE) as an indicator anchored on a zinc oxide-reduced graphene oxide (ZnO-rGO) composite rich in active sites, in which green non-toxic zinc oxide (ZnO) nanomaterials were uniformly distributed on the reduced graphene for rapid detection of organophosphorus. The effects of different ratios of ZnO to reduced graphene on the performance of ZnO-rGO nanocomposites were investigated. The AChE/ZnO-rGO biosensor detects organophosphorus by electrochemical inhibition of acetylcholinesterase in the presence of organophosphorus. The developed electrochemical biosensor has high selectivity and good linearity, and the ZnO-rGO nanocomposite as a matrix for immobilization of acetylcholinesterase and detection of organophosphorus has the potential for highly sensitive pesticide detection.

Abstract:BACKGROUND Texture analysis can be used to identify and discriminate between objects in an image and has already been used for the characterization of diseases. The present study evaluated the potential of texture analysis for monitoring tumor cell viability during treatment with an anti-vascular drug. MATERIALS AND METHODS Texture analysis of T2-weighted magnetic resonance images of C3H mammary carcinomas in mice was used to study tumor cell viability during treatment with the anti-vascular drug combretastatin A-4 disodium phosphate (CA4DP). Texture analysis was done by MaZda, and boot strap statistics were used to analyze the results. RESULTS The results revealed significant differences in the textures of T2-weighted images when non-tumor tissue and tumor tissue were compared. Furthermore, selected texture parameters were significantly different when image data from before and after CA4DP administration were analyzed. CONCLUSION Texture analysis is helpful in the diagnosis of different types of tissues, especially when these tissues can not be recognized by human observers. Some functional changes, such as perfusion changes in the tumor, can also be detected by texture analysis.
Simplifying the complexity: Single enzyme (choline oxidase) inhibition-based biosensor with dual-readout method for organophosphorus pesticide detection

Abstract:Organophosphorus pesticides (OPs) are widely used in agricultural production, but their residues could cause pollution to the environment and living organisms. In this paper, a simple dual-readout method for OPs detection was proposed based on ChOx single enzyme inhibition. Firstly, ChOx can catalyze the production of H 2 O 2 from choline chloride (Ch-Cl). Bifunctional iron-doped carbon dots (Fe-CDs) with good peroxidase-like activity and superior fluorescence properties can catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) to blue oxidized TMB (oxTMB) by H 2 O 2 formed, and oxTMB could quench the fluorescence of Fe-CDs. In light of the fact that OPs exhibited activity in inhibiting ChOx, less H 2 O 2 and the decreasing oxTMB led to a result that the fluorescence of the system recovered and the solution became lighter in blue color. Moreover, the process of ChOx inhibition by OPs was analyzed by molecular docking technique and it was found that OPs interact with key amino acid residues catalyzed by ChOx (Asn510, His466, Ser101, His351, Phe357, Trp331, Glu312). Finally, a dual-mode (colorimetry and fluorescence) sensor was created for the detection of OPs with the detection limit of 6 ng/L, and was successfully used in the quantitative determination of OPs in actual samples with satisfactory results.

Porous-Reduced Graphene Oxide for Fabricating an Amperometric Acetylcholinesterase Biosensor

Abstract:This work reports a sensitive amperometric biosensor for organophosphate pesticides (OPs) fabricated through modifying glassy carbon electrode with acetylcholinesterase (AChE) immobilized on porous-reduced graphene oxide (pRGO). The pRGO sheets can not only provide high surface area but also facilitate the diffusion and mass transport of reactants. The as-prepared biosensor shows high affinity to acetylthiocholine (ATCl) with a Michaelis–Menten constant value of 0.73mM. Furthermore, based on the inhibition of the enzymatic activity (immobilized AChE) caused by the model compound of carbaryl (one kind of pesticides), it is found that the inhibition activity of carbaryl is proportional to its concentration ranging from 0.001 to 0.05μgmL−1. The developed biosensor shows a detection limit of 0.5ngmL−1 for OPs detection and exhibits good performance such as reproducibility and stability, which makes it possible to provide a new and promising tool for the analysis of enzyme inhibitors.

Abstract:An ultrasensitive amperometric acetylcholinesterase (AChE) biosensor was fabricated by controlled immobilization of AChE on gold nanoparticles/poly(dimethyldiallylammonium chloride) protected Prussian blue (Au-PDDA-PB) nanocomposite modified electrode surface for the detection of organophorous pesticide. The Au-PDDA-PB membrane served as an excellent matrix for the immobilization of enzyme, which not only enhanced electron transfer but also possessed a relatively large surface area. In addition, the surface hydrophilicity of the Au-PDDA-PB nanocomposite was finely controlled in the static water contact angle range of 25.6-78.1° by adjusting the ratio of gold nanoparticles to PDDA-PB. On an optimized hydrophobic surface, the AChE adopts an orientation with both good activity and stability, which has been proven by electrochemical methods. Benefit from the advantages of the Au-PDDA-PB nanocomposite and the good activity and stability of AChE, the biosensor shows significantly improved sensitivity to monocrotophos, a typical highly toxic organophorous pesticide, with wide linear range (1.0-1000 pg/mL and 1.0-10 ng/mL) and an ultra-low detection limit of 0.8 pg/mL. The biosensor exhibits accuracy, good reproducibility and stability. This strategy may therefore provide useful information for the controlled immobilization of protein and the design of highly sensitive biosensors.

Abstract:A sensitive and flexible detection method for organophosphorus pesticides (OPs) detection is a crucial request to avoid their further expanded pollution. Herein, an acetylcholinesterase (AChE) electrochemical sensor, based on the co-modification of ZIF-8 and graphene (GR), was constructed for the detection of OPs. ZIF-8/GR composite can provide a stable and biocompatible environment for the loading of AChE and can accelerate the chemical reaction on the electrode surface. After optimization, the linear detection range of the constructed AChE-CS/GR/ZIF-8/GCE sensor for ICP was 0.5–100 ng/mL (1.73–345.7 nM), and the limit of detection was 0.18 ng/mL (0.62 nM). Moreover, high sensitivity and high specificity of the sensor were also achieved in actual cabbage and tap water samples. Therefore, it has great potential for the application of organophosphorus pesticide residue analysis.

Abstract:Owing to their large specific surface, favorable electrical conductivity, and excellent electrocatalytic capabilities, two-dimensional transition metal dichalcogenides have received considerable attention in the field of biosensors. On the basis of these properties, we developed a portable and disposable enzyme-based biosensor for paraoxon detection using a metallic MoS2 nanosheets modified screen-printed electrode (SPE). The exfoliated ultrathin metallic MoS2 nanosheets can accelerate the electron transfer on electrode surface and contribute to the immobilization of acetylcholinesterase (AChE) via the cross-linking of glutaraldehyde. Electrodeposited gold nanoparticles (AuNPs) on SPE were used to immobilize MoS2 nanosheets through the interaction between Au atoms on AuNPs and S atoms on MoS2. Using acetylcholine as the substrate, AChE can catalyze the formation of electroactive thiocholine and further generate the redox current. In the presence of paraoxon, the activity of AChE can be inhibited, making the related electrochemical signals weaken. Under the optimized conditions, this electrochemical biosensor exhibited a favorable linear relationship with the concentration of paraoxon from 1.0 to 1000 μg L-1, with the detection limit of 0.013 μg L-1. Furthermore, this developed biosensor was successfully applied to detect paraoxon in pretreated apple and pakchoi samples, which can provide a reliable method for the rapid analysis of organophosphorus pesticides in agricultural products.

Abstract:Electrochemical reduced β-cyclodextrin dispersed graphene (β-CD-graphene) was developed as a sorbent for the preconcentration and electrochemical sensing of methyl parathion (MP), a representative nitroaromatic organophosphate pesticide with good redox activity. Benefited from the ultra-large surface area, large delocalized π-electron system and the superconductivity of β-CD-graphene, large amount of MP could be extracted on β-CD-graphene modified electrode via strong π–π interaction and exhibited fast accumulation and electron transfer rate. Combined with differential pulse voltammetric analysis, the sensor shows ultra-high sensitivity, good selectivity and fast response. The limit of detection of 0.05ppb is more than 10 times lower than those obtained from other sorbent based sensors. The method may open up a new possibility for the widespread use of electrochemical sensors for monitoring of ultra-trace OPs.
Disposable Amperometric Acetylcholinesterase Biosensor for the Detection of Organophosphorus Pesticides

Abstract:A rapid, simple, disposable and inexpensive acetylcholinesterase (AChE) amperometric biosensor for the detection of organophosphorus pesticides was developed by simple adsorption of the enzyme on screen-printed electrodes. The biosensor consisted of an Ag/AgCl reference electrode and a graphite working electrode. The mixture of graphite and the 7,7,8,8-tetracyanoquinodimethane (TCNQ) was printed on electrodes. The detection of organophosphorus pesticides was done with acetylthiocholine chloride (ATCh) as substrate. The biosensor was used to detect the inhibitory effect of organophosphorus pesticides on ACHE activity. The 1 mu l of enzyme solution containing 0.1 U AChE and 1% bovine serum albumin (BSA) were simply dropped on the working electrode surface. The biosensor operated at a potential of 300 mV vs. Ag/AgCl in a pH 7.2 0.1 mol/L phosphate buffer and 0.1 mol/L KCl. We obtained a calibration plot of the percentage inhibition versus the logarithm of parathion methyl concentration following an incubation time of 10 min in parathion methyl solution. The lowest detectable amount of parathion methyl was 0.026 ppm. The amperometric biosensor based on acetylcholinesterase was disposable and low cost (about 1 yuan RMB).

Abstract:In this work, a convenient ratiometric fluorescent platform was designed to measure organophosphorus pesticides (OPs) based on acetylcholinesterase (AChE), acetylthiocholine (ATCh), manganese dioxide nanosheets (MnO 2 ), near-infrared carbon dots (RCDs) and o-phenylenediamine (OPD). In this platform, a direct oxidation of OPD by MnO 2 generated the luminescent product 2,3-diaminophenolazine (DAP) through intrinsic oxidase activity, while RCDs served as a fluorescent reference indicator. In the presence of AChE and ATCh, the enzymatic hydrolysate thiocholine (TCh) would reduce MnO 2 nanosheets to Mn 2+ , leading to the quenching of DAP fluorescence. On the other hand, OPs can inhibit the catabolism of ATCh by AChE thus acting as a recognizer of OPs. According to these reactions, OPs were quantitatively analyzed by the intensity ratio of fluorescence emitted from RCDs and DAP ( F 560 / F 676 ). The constructed platform can detect OPs with the range of 0.2–0.6 μM with a detection limit of 4.3 nM.

Abstract:Based on a screen printed carbon electrode which contained a sensitive film prepared by crosslinking acetylcholine esterase (AChE) with glutaraldehyde on the surface of working electrode, an amperometric enzyme biosensor was taken on the determination of organophosphorus (OP) pesticide in water environment. Cyclic voltammetry was applied to assess the coherence of prepared screen printed electrodes. The results show that the variation is less than 10%. The sensor sensitivity was optimized with respect to content of enzyme, crosslinker and substrate. The best sensitivity is obtained using 0.2% of glutaraldehyde, 0.5 mg/mL of AChE and 10 mmol/L of acetylthiocholine chloride (ATChCl). Amperometric I-t curve was conducted to detect the decrease rate of the current after the addition of two kind of OPs (terbufos and parathion), respectively. The detection limit for two kind of OPs are found to be 1 ng/mL with the linear range 1 ng/mL - 10 000 ng/mL. Cyclic voltammetry was conducted to detect parathion on the sensors which immobilized AChE by physical adsorption. The results show that it can achieve a better sensitivity than amperometric I-t curve.
Highly sensitive turn-on electrochemical sensing of organophosphorus pesticides by integration of homogeneous reaction and heterogeneous catalytic signal amplification

Abstract:Enzyme-inhibited electrochemical sensor is a promising strategy for detecting organophosphorus pesticides (OPs). However, the poor stability of enzymes and the high oxidation potential of thiocholine signal probe limit their potential applications. To address this issue, an indirect strategy was proposed for highly sensitive and reliable detection of chlorpyrifos by integrating homogeneous reaction and heterogeneous catalysis. In the homogeneous reaction, Hg2+ with low oxidation potential was employed as signal probe for chlorpyrifos detection since its electroactivity can be inhibited by thiocholine, which was the hydrolysate of acetylthiocholine catalyzed by acetylcholinesterase. Additionally, Co,N-doped hollow porous carbon nanocage@carbon nanotubes (Co,N-HPNC@CNT) derived from ZIF-8@ZIF-67 was utilized as high-performance electrode material to amplify the stripping voltammetry signal of Hg2+. Thanks to their synergistic effect, the sensor exhibited outstanding sensing performance, excellent stability and good anti-interference ability. This strategy paves the way for the development of high-performance OP sensors and their application in food safety.
Selective Detection of Hypertoxic Organophosphates Pesticides Via PDMS Composite Based Acetylcholinesterase-Inhibition Biosensor.

Abstract:We report on a pair of highly sensitive amperometric biosensors for organophosphate pesticides (OPs) based on assembling acetylcholinesterase (AChE) on poly(dimethylsiloxane) (PDMS)-poly(diallydimethylemmonium) (PDDA)/gold nanoparticles (AuNPs) composite film. Two AChE immobilization strategies are proposed based on the composite film with hydrophobic and hydrophilic surface tailored by oxygen plasma. The twin biosensors show interesting different electrochemical performances. The hydrophobic surface based PDMS-PDDAN AuNPs/choline oxidase (ChO)/AChE biosensor (biosensor-1) shows excellent stability and unique selectivity to hypertoxic organophosphate. At optimal conditions, this biosensor-1 could measure 5.0 x 10(-10) g/L paraoxon and 1.0 x 10(-9) g/L parathion. As for the hydrophilic surface based biosensor (biosensor-2), it shows no selectivity but can be commonly used for the detection of most OPs. Based on the structure of AChE, it is assumed that via the hydrophobic interaction between enzyme molecules and hydrophobic surface, the enzyme active sites surrounded by hydrophobic amino acids face toward the surface and get better protection from OPs. This assumption may explain the different performances of the twin biosensors and especially the unique selectivity of biosensor-1 to hypertoxic OPs. Real sample detection was performed and the omethoate residue on Cottomrose Hibiscus leaves was detected with biosensor-1.

Abstract:Precisely detecting organophosphorus pesticides (OPs) is paramount in upholding human safety and environmental preservation, especially in food safety. Herein, an electrochemical acetylcholinesterase (AChE) sensing platform entrapped in chitosan (Chit) on the glassy carbon electrodes (GCEs) decorated with Pt/MoS2/Ti3C2 MXene (Pt/MoS2/TM) was constructed for the detection of chlorpyrifos. It is worth noting that Pt/MoS2/TM possesses good biocompatibility, remarkable electrical conductivity, environmental stability and large specific surface area. Besides, the heterostructure formed by the composite of TM and MoS2 improves the conductivity and maintains the original structure, which is conducive to improving the electrochemical property. The coordination effect between the individual components enables the even distribution of functional components and enhances the electrochemical performance of the biosensor (AChE-Chit/Pt/MoS2/TM). Under optimal efficiency and sensitivity, the AChE-Chit/Pt/MoS2/TM/GCE sensing platform exerts comparable analytical performance and a wide concentration range of chlorpyrifos from 10-12 to 10-6 M as well as a low limit of detection (4.71 × 10-13 M). Furthermore, the biosensor is utilized to detect OPs concerning three kinds of fruits and vegetables with good feasibility and recoveries (94.81% to 104.0%). This work would provide a new scheme to develop high-sensitivity sensors based on the two-dimensional nanosheet/laminar hybrid structure for practical applications in environmental monitoring and agricultural product detection.
