Abstract:Anthrax spores have been a determining risk to human beings and animals across the world. As such, the qualitative monitoring toward dipicolinic acid (DPA), the unique biomarker of bacillus anthracis, is highly desired and being moved forward in health management. Herein, a breathing zeolite-like supramolecular assembly (ZSA), with the sra topology and 1D hexagon channel, is designed by controlling the bridging angle of the ditopic ligand after systematic analysis in topology. The solvent-dependent dynamic behavior is illustrated by powder X-ray diffraction and reflected in tunable luminescent emission based on solvent polarity parameters. According to the structural result and theoretical analysis, Eu3+ is anchored within the framework to form a ratiometric luminescent sensor successfully because of the abundant potential active site. After DPA addition, the resulting composite shows a sensitive and selective response in the linear range of 0-7.0 μM stemmed from absorbance energy-transfer emission and preferential coordination. The work we presented here may enrich the sensing platforms containing lanthanides and expand the potential application of ZSAs in clinical analysis.

Abstract:The fabrication of luminescent materials with lanthanide cations encapsulated within MOF pores is currently of interest because luminescent materials are used in numerous applications. In this study, a distinctive strategy via postsynthetic modification (PSM) of a novel metal-organic framework [Cu(HCPOC)(2)](n)(Cu1) (H2CPOC = 5-(40-carboxyphenoxy) nicotinic acid) and terbium ions (Tb3+) for sensing hydrogen sulfide (H2S) is reported. The obtained composite Tb3+@ Cu1 emits a weak typical Tb3+ ion emission and strong ligand-centred emission. Interestingly, H2S, as a strong electron donor, can strongly enhance the luminescence of Tb-3+ through its superior affinity for Cu2+ ions. The composite Tb3+@Cu1 was designed as a luminescent turn-on ratiometric probe for H2S detection, showing high sensitivity and selectivity. The detection limit of Tb3+@Cu1 (1.20 mM) is far below that of Cu1 (13.25 mM). Moreover, a similar ligand, 5-(40-carboxyphenyl) nicotinic acid (H2CPC), was used to synthesize a fascinating structure, [Cu-5(CPC)(2)(HCPC)(2)(OH-)(4)](n) (Cu2), which was quite similar to that of the famous semiconductor MoS2, with the advantageous support of an organic linker between layers, and it showed a band gap of 2.45 eV.

Abstract:Fluorescent materials with lanthanide cations encapsulated in MOFs are currently used in numerous applications, especially in biosensors. Therefore, herein, two novel composites were designed and developed based on a Tb(iii)&Yb(iii)-functionalized Cu(ii)-coordination polymer, possessing higher thermal and water stability and fascinating fluorescence properties. The first bimetallic composite Tb@Cu-Hcbpp demonstrated broad ligand-centered emission and weak typical Tb3+ ion emission; moreover, it was used as an excellent ratiometric fluorescent sensor for the metabolic product NMF of DMF in the human body (LOD = 0.02 mu M). In addition, the Yb3+ ions were doped into Tb@Cu-Hcbpp to improve the fluorescence performance of the green Tb3+ ion emission. Among the series of Tb1-xYbx@Cu-Hcbpp samples (x = 0.05, 0.10, 0.15, 0.20, 0.25, 0.30, 0.35 and 0.40), Tb0.85Yb0.15@Cu-Hcbpp showed maximum enhanced fluorescence intensity (almost 9.6 times that of the pure terbium system), but exhibited high fluorescence quenching efficiency for methylglyoxal (MGO), which could be used for the sensitive detection of MGO (LOD = 0.25 mu M). Furthermore, the developed biosensors were successfully applied for the detection of NMF and MGO in urine and serum samples, and satisfactory results were obtained, showing good potential of these biosensors in practical applications such as in disease diagnosis and biochemical research.

Abstract:Anthrax bacillus is a very dangerous zoonotic pathogen that seriously endangers public health. Rapid and accurate qualitative and quantitative detection of its biomarkers, 2,6-dipicolinic acid (DPA), is crucial for the prevention and treatment of this pathogenic bacterium. In this work, a novel Cd-based MOF (TTCA-Cd) has been synthesized from a polycarboxylate ligand, [1,1':2',1″-terphenyl]-4,4',4″,5'-tetracarboxylic acid (H4TTCA), and further doped with Tb(III), forming a dual-emission lanthanide-functionalized MOF hybrid (TTCA-Cd@Tb). TTCA-Cd@Tb can be developed as a high-performance ratiometric fluorescent sensor toward DPA with a very low detection limit of 7.14 nM and high selectivity in a wide detection range of 0-200 μM, demonstrating a big advancement and providing a new option for the detection of DPA.

Abstract:A novel Tb-doped Eu-based metal–organic framework (Eu-MOF@Tb) has been developed by incorporating hexanuclear europium cluster and 2,2′-bipyridine-5,5′-dicarboxylic acid as well as coordination with Tb(III). Owing to the diverse coordination status of Tb(III) and Eu(III) in MOF, antenna effect emission from Tb(III) can be invoked by dipicolinic acid (DPA), but the luminescence originating from Eu(III) remains unchanged. Taking advantage of this phenomenon, a ratiometric luminescent method for detection of DPA, a biomarker for Bacillus subtilis spores, was developed through differential sensitization toward lanthanide ions. This analysis method allowed for the detection of DPA in the 0.2–10 μM concentration range, with a detection limit of 60 nM. It was further validated by spiked recoveries (89.3–110%) of real-world samples with RSD values in the range 3.9–11%. Alongside this, a paper indicator test was prepared for naked-eye detection of DPA via a dose-sensitive color evolution from red to green under UV light. The effectiveness of the proposed approach was explored in the detection of bacterial spores in real biological and environmental samples and indicated great potential for applications as a real-time monitoring system against the anthrax threat.

Abstract:Covalent organic frameworks (COFs) with full π-conjugation can be obtained by tuning the building blocks and covalent bonds. Meanwhile, lanthanide ions (Ln3+) have attracted attention for their unique luminescent properties. In this study, we coordinated Tb ions in COF (TQBQ-COF), thereby constructing a fluorescent sensor for the detection of 2,6-pyridinedicarboxylic acid (DPA) as an anthrax biomarker. The prepared Tb3+@TQBQ-COF shows an ultra-low detection limit for DPA (3.58 nM), which is much lower than the required level of DPA for Bacillus anthracis to produce anthrax (60 μM). In addition, the prepared sensors showed good detection performance in detecting anionic pollutants when DPA was used as a photosensitizer. Our work provides a simple method for the detection of DPA.

Abstract:A ratiometric fluorescent sensor based on luminescent bio-metal-organic framework was prepared by exchanging both Tb3+ and Eu3+ cations into anionic bio-MOF-1. Due to a highly efficient energy transfer from Tb3+ to Eu3+ (>89%), emission color of Tb/Eu@bio-MOF-1 was orange-red even though Tb3+ was the dominant content in this Tb/Eu co-doping material. More interestingly, this energy transfer process could be modulated by dipicolinic acid (DPA), an unique biomarker for bacillus spores. With DPA addition, corresponding DPA-to-Tb3+ energy transfer was gradually enhanced while the energy transfer from Tb3+ to Eu3+ was significantly weakened. By regulating the energy transfer process in Tb/Eu@bio-MOF-1, visual colorimetric sensing of DPA in porous MOF was realized for the first time. Detection limit of Tb/Eu@bio-MOF-1 for DPA was 34nM, which was much lower than an infectious dosage of Bacillus anthracis spores (60μM) for human being. Besides, Tb/Eu@bio-MOF-1 showed a remarkable selectivity over other aromatic ligands and amino acids. More importantly, this porous ratiometric sensor worked equally well in human serum. These particularly attractive features of Tb/Eu@bio-MOF-1 made the direct, rapid and naked-eye detection of DPA for practical application possible.

Abstract:Sensitive, efficient and rapid detection of 2, 6-pyridinedicarboxylic acid (DPA), commonly labeled as the main biology marker for bacillus anthracis, is of far-reaching consequence to eliminate potential threats. In this work, a 3D Eu-MOF ([Eu(DHBDC)1.5(DMF)2]·DMF) that has outstanding thermal and water stability was synthesized. The Eu-MOF showed characteristic fluorescence at 506nm caused by ligand π-π* transition. The quantum calculation showed that there was a large energy difference between the 5D0 level of Eu3+ and the triplet energy level of 2,5-dihydroxy terephthalic acid ligand (DHBDC2-), which was why the coordination compound did not exhibit the characteristic fluorescence of Eu3+. Interestingly, it effectively restored the red fluorescence of Eu3+ (593, 618 and 699 nm) and showed a dual-emission fluorescence response at 506 and 593 nm when the DPA existed. Therefore, based on the high detection sensitivity of Eu-MOF (the detection limit is 1.3 μM) and outstanding anti-interference ability, it is expected to become a prospective ratiometric fluorescence sensor for the detection of DPA. In addition, combined with density functional theory (DFT) and a variety of other analytical methods, the detection mechanism of DPA by Eu-MOF was investigated in detail.

Abstract:Luminescent rare-earth (RE) complexes usually present outstanding advantages of long-lived emissions and high emission efficiency in aggregate and solid states. However, the combination of aggregation-induced emission (AIE)-based materials and RE complexes with excellent luminescent properties is rarely reported. Herein, AIE luminogens (AIEgens)-based Tb3+ complex (TPE-Tb) was designed and synthesized for the first time, which can self-assemble into novel TPE-Tb nanoparticles to be used as ratiometric fluorescent probe for sensitive and rapid detection of anthrax spore biomarker in aqueous solution or actual spores. The results indicated that the limit of detection (LOD) of this ratiometric fluorescent probe was as low as 0.187 nM and 1.64 × 104 spores/mL in water solution or actual spore samples, respectively. This work explored a new method for the combination of AIEgens-based materials and RE complexes by coordination mode and provided a versatile nanoprobe candidate for anthrax biomarker detection.

Abstract:Rare earth (RE) complexes have found a variety of applications in materials science and biomedicine because of their unique luminescence properties. However, the poor stability and solubility in water of multicomponent RE assemblies significantly limit their practical applications. We rationally designed and developed a novel Eu3+/Tb3+ supramolecular assembly hybrids (Eu/Tb-SAH) by supramolecular host-guest recognition and coordination recognition with the excellent characteristics of water dispersion stability, biocompatibility and luminous properties. As anthrax spore biomarker, 2,6-pyridinedicarboxylic acid (DPA) can coordinate with Tb3+ and sensitize Tb3+, resulting in a proportional change of fluorescence intensity and lifetime on the ms timescales, thereby realizing rapid and sensitive detection of DPA in water media or actual spores. To confirm our prediction, accurate and selective detection of DPA was achieved with Eu/Tb-SAH as a nanoprobe through steady-state ratiometric fluorescence and time-resolved technology, of which the limit of detection (LOD) are 27.3 nM and 1.06 nM, respectively. This was obviously lower than the amount of anthrax spores infecting the human body (60 μM). Besides, the filter paper was used to carry out visual detection of DPA and read the corresponding data through smart phones. This work paves a new way to fabricate luminescent RE nanomaterials and provides new ideas for the design of ratiometic lifetime imaging biosensors in the meantime.

Abstract:Bacillus anthracis spores have a unique biomarker of calcium dipicolinate (CaDPA). In this work, we reported a composite nanostructure for the optical sensing of DPA, with Eu (III)-doped metal-organic framework (MOF) as supporting lattice, a rhodamine-derived dye as sensing probe, respectively. By means of XRD, IR, TGA and photophysical analysis, this composite structure was carefully discussed. It was found that rhodamine absorption and emission were enhanced by DPA, while Eu emission was quenched by DPA. As a consequence, two sensing skills were observed from this composite structure, which are colorimetric sensing based on absorption spectra and ratiometric fluorescent sensing based on emission spectra. Linear sensing response was observed for both sensing channels with a warning signal at DPA concentration higher than 140 μM. Good selectivity was confirmed with a low LOD value of 0.52 μM. The sensing mechanism was revealed as the combination of emission turn-on effect triggered by DPA-released protons and emission turn-off effect originated from electron-transfer from EuBTC to DPA. This composite structure showed its advantage of naked eye detection and two sensing skills with linear response.

Abstract:A stable 3D metal-organic framework (MOF) with formula [Tb(L)(H2O)2·3H2O]n (H3L = 5-(4′-carboxylphenoxy)isophthalic acid) (1) has been synthesized solvothermally and characterized using spectroscopic techniques and crystallographically. The 3D MOF is constructed from 1D Tb(III)-based chains where the overall structure is 3D 5-connected bnn topology framework with Schläfli symbol 46.64. The MOF exhibits displays intense green emission on excitation at 273 nm and had been used as turn-off luminescent sensor and exhibits sensitive and selective luminescence detectability for sulfamethazine (SMZ) (Ksv = 1.28 × 103 M−1 and LOD = 1.43 ppm). The MOF 1 also possesses excellent stability and recyclability to detect SMZ. The plausible mechanistic pathway for the detrioration in the luminescent intensity of the MOF in presence of antibiotics have been explained with the help of theoretical calculations and Hirshfeld surface analyses.

Abstract:2,6-pyridine dicarboxylic acid (DPA) is an exceptional biomarker of notorious anthrax spores. Therefore, the rapid, sensitive, and selective quantitative detection of DPA is extremely significant and urgent. This paper reports a Zn(II) metal–organic framework with the formula of {[Zn6(NDA)6(DPBT)3] 2H2O·3DMF}n (MOF-1), which consists of 2,6-naphthalenedicarboxylic acid (2,6-NDA), 4,7-di(4-pyridyl)-2,1,3-benzothiadiazole (DPBT), and Zn(II) ions. Structural analysis indicated that MOF-1 is a three-dimensional (3D) network which crystallized in the monoclinic system with the C2/c space group, revealing high pH, solvent, and thermal stability. Luminescence sensing studies demonstrated that MOF-1 had the potential to be a highly selective, sensitive, and recyclable fluorescence sensor for the identification of DPA. Furthermore, fluorescent test paper was made to detect DPA promptly with color changes. The enhancement mechanism was established by the hydrogen-bonding interaction and photoinduced electron transfer transition between MOF-1 and DPA molecules.

Abstract:One Zn-MOF, namely, {[Zn(L) 0.5 (bpy) 0.5 (H 2 O)]·H 2 O·DMF} n ( LCU-123 ), was synthesized from an amide-containing carboxylic acid N,N''-(3,5-dicarboxylphenyl)benzene-1,4-dicarbox-amide (H 4 L) and 4,4'-bipyridine. The carboxylate ligand and 4,4'-bipyridine connect Zn(II) centers to form two-dimensional layers. Adjacent 2D layers are stacked to form a stable supramolecular structure with one-dimensional channels. Luminescence measurements indicated that the complex exhibited excellent multiple sensing activities toward biomarker (2-methoxyethoxy)acetic acid (MEAA) and antibiotic tetracycline hydrochloride (TC). It is worth noting that the LOD for MEAA and TC were as low as 18.030 and 0.552 μ M, respectively. Investigations on the sensing mechanism indicated that the luminescence quenching of MEAA was due to the weak interaction and dynamic quenching mechanism, while the quenching sensing of TC was attributed to competitive absorption and PET effect. The recoveries for the sensing analytes are satisfactory from 102.8 to 109.8% (toward MEAA in simulated urine) and 99.3 to 106.0% (toward TC in water), indicating the complex possesses acceptable reliability for monitoring the above two analytes in real samples.

Abstract:Development of sensitive and prompt approaches for reliable determination of dipicolinic acid (DPA) is of great importance for human health and public security. Herein, we synthesize a bimetallic lanthanide metal-organic framework (Tb/Eu-MOF) by using 2-hydroxyterephthalic acid as ligand and lanthanide ions (Tb3+ and Eu3+) as central metal ions. The as-synthesized Tb/Eu-MOF possesses multiple recognition sites toward DPA. In the existence of DPA, the DPA can displace the chelated water molecules and then coordinate with Tb3+ and Eu3+ to sensitize their luminescence, causing that the fluorescence of Tb3+ (546 nm) and Eu3+ (617 nm) raises while that of ligand (432 nm) remains constant. Based on the multiple fluorescence signal responses of Tb/Eu-MOF toward DPA, a multi-ratiometric fluorescence sensor by using the (F546/F432) and (F617/F432) as output signals is developed to detect DPA. Besides, an intrinsic signal amplification strategy is used to achieve more sensitive sensing of DPA by integrating the fluorescence signals of Tb3+, Eu3+, and ligand with (F546 + F617)/F432 as the output signal. Benefitting from the signal amplification and multiple fluorescence responses, this method shows high sensitivity and obvious fluorescence color variation toward DPA detection. This strategy offers a prompt, sensitive, and reliable method for visual sensing of DPA.

Abstract:The rapid development of functionalization of lanthanide metal organic frameworks (Ln-MOFs) offers the potential for biomarkers sensing. It is closely associated with biology science and clinic medicine. In this work, a porous and luminescent terbium metal-organic framework TbTATB has been designed and solvothermally synthesized. The PXRD results demonstrated the commendable stability of this sensor. Enhanced luminescence of TbTATB origin from Tb3+ was observed in the presence of Zn2+, which is a kind of biologically important ion. This turn-on pattern was utilized for sensing Zn2+. TbTATB exhibited excellent sensitivity and selectivity towards Zn2+ in aqueous solution with the detection limit of 10.5 nM. What's more, it has fairly acceptable bio-compatibility which was fully verified by cytotoxicity test. In the state of coexistence with living cells, visible increased green light of TbTATB emerged after the addition of Zn2+, while the survival state of cells was hardly disturbed. Markedly, this is the first time that a luminescent Tb-MOF can act as a sensor for sensing and imaging of Zn2+ ions simultaneously. What's more, the results also guide a new application direction of MOFs and supply a new way for the detection of other analytes in biological systems.
