Abstract:Modern biotechnology,especially genetic engineering,is one of the effective methods for alfalfa to improve its forage yield and quality,while a prerequisite for using this technology is the efficient solutions to tissue culture and regeneration system of Alfalfa.The bottleneck of alfalfa genetic engineering attributes to lack of stalbe and hige frequency regeneration,because the tissue culture and regeneration are highly affected by genotype of alfalfa and environmental conditions.A solution for rapid and highly efficient tissue regeneration of alfalfa NFF2was established by studying the influence of hormone concentration,and different combinations of medium on alfalfa callus induction and differentiation in the experiment.The result showed that the best medium for callus induction was the modified SH(SH2K)with 1.0 mg/L 2,4-D+0.2mg/L KT,and the best subculture medium for callus induction was BOi2Y;additionally,the best differentiation medium for callus induction was MSO,while the best medium for plant development of alfalfa was 1/2 MS.

Abstract:Current research needs to be more focused on agronomical plants to effectively utilize the knowledge obtained from model plant species. Efforts to improve legumes have long employed common breeding tools. Recently, biotechnological approaches facilitated the development of improved legumes with new traits, allowing them to withstand climatic changes and biotic stress. Owing to its multiple uses and profits, alfalfa (<i>Medicago sativa</i> L.) has become a prominent forage crop worldwide. This review provides a comprehensive research summary of tissue culture-based genetic transformation methods, which could be exploited for the development of transgenic alfalfa with agronomically desirable traits. Moreover, advanced bio-imaging approaches, including cutting-edge microscopy and phenotyping, are outlined here. Finally, characterization and the employment of beneficial microbes should help to produce biotechnologically improved and sustainable alfalfa cultivars.

Abstract:The high frequent regeneration system of 4 alfalfa cultivars were obtained and the effects of genotype,explants and medium on dedifferentiation and differentiation were studied.The results showed that the callus differentiation rate of WL-323 was 76.3%,it is the optional genotype.The best optimal explants was hypocotyls.The optimal dedifferentiation medium was SH+4.0 mg/L 2,4-D+0.5⒐mg/L 6-BA.The optional subculture medium was MSO+0.5 mg/L NAA+1.0 mg/L 6-BA+1.0 mg/L AgNO3.The optimal differentiation medium was MS.

Abstract:Background: The Alxa Region in Inner Mongolia, northwest China is a desert grassland area. Wild alfalfa growing in this area is a precious wild plant resource that can provide resources for improving the stress resistance of alfalfa varieties. Alfalfa is a cross-pollinated plant and its genes are commonly heterozygous. Therefore, establishing a complete tissue culture regeneration system for a single plant is an effective method of protecting wild plant resources. This study used the bud and leaf of wild alfalfa from the Alxa Region as explants to establish an efficient and stable tissue culture regeneration system. Methods: Two kinds of wild alfalfa tissues were selected as explants. Different concentrations and types of plant growth hormones and regulators were tested, to determine the optimal medium for callus induction, differentiation and rooting culture, including 6-Benzylaminopurine (6-BA), Kinetin (KT), 2, 4-Dichlorophenoxyacetic acid (2, 4-D), Naphylacetic acid (NAA), Indole-3-butyric acid (IBA) and activated carbon (AC). Result: The callus induction rate was 93.3% when the bud of wild alfalfa was used as an explant for propagation. With a basic medium (Murashige and Skoog medium with Ca2+, Mg2+ and Mn2+ halved) and hormone regulator (6-BA, KT, NAA, AC), the differentiation rate and rooting rate reached as high as 66.67% and 93.33%, respectively. When leaves were used as explants, the callus induction rate and differentiation rate were 100% and 73.33%, respectively. In the rooting culture process, adding IBA (0.2 mg/L) and AC (0.5 mg/L) to the medium improved the rooting rate of wild alfalfa. Both buds and leaves can be used as explants for plant tissue regeneration and culture, which can help expand and preserve wild alfalfa resources.

Abstract:In vitro methods have environmental advantages of preparing bio additives as harmful chemicals such as fertilizers, herbicides, pesticides are used. In addition, obtaining biomass does not depend on seasonality and a long growing season. In this series of experiments, an accelerated method for obtaining a sterile alfalfa culture was developed by adding 1% potassium humate to the explant culture. From 4 varieties of alfalfa, 3 cell lines were selected, characterized by heterogeneity of callus tissues. 1 - line morphogenic structures; 2 - line - without meristematic foci and brown and dark brown in color, which were not further differentiated; 3- line forming polymerogenic tissues. In addition, it was possible to induce denser tissues from loose callus by passaging on the Risting medium in the Ray khan variety. Structured tissues with meristematic foci induced on Risting medium were lyophilized and the dried biomass was prepared for further biochemical analyses. Thus, we have optimized the conditions for obtaining biomass from alfalfa culture and carried out the selection of cell lines, and we assume that callus lines, upon receipt of positive biochemical analyzes, can be used as feed additives.

Abstract:To identify the aluminum stress induced genes of alfalfa,a forward subtractive cDNA library was constructed from Al-treated and non-treated seedling roots by suppression subtractive hybridization using the alfalfa(Medicago sativa)aluminum tolerance cultivar Xiaoguan.Colony PCR suggested that the inserts were between 250 ～1200 bp and the library was suitable for the following work.Forty clones were randomly selected and secquenced,and 34 unique ESTs were obtained.BLASTX showed that 25 ESTs found the homology.One EST coding for pectinesterase which could catalyse pectin and resulted in more Al binding sites might play the role in alfalfa aluminum detixitifcation.The other 24 ESTs might be involed in plant antioxidative stress,signal transduction,development and energy metaboly,code for peroxidase,calcium-dependent protein kinase,neutral/alkaline invertas,protein phosphatase regulator,protein transporter,cytochrome P450,acetylhexosaminidase ect.These physiological responses were observed to be similar to those occuring under other stresses.Nine ESTs had no homology from NCBI.

Abstract:Alfalfa is a valuable high-protein fodder crop suitable for the preparation of various types of feed (green mass, hay, haylage, silage). Due to the increasing aridity of the climate, there is a need to expand its crops in the northern regions of the country. Currently, it is necessary to create alfalfa varieties with a good adaptation to modern environmental conditions, to improve the development of seed production of this crop. Often alfalfa plants are highly susceptible to disease damage, damage by pests. Fungal diseases (fusarium, brown spotting) during the epiphytotic period can also reduce the productivity of alfalfa herbage and its quality by 30% or more. Methods of selection, polycross, and hybridization were used to create a fusarium-resistant, brown-spotted alfalfa source material. Long-term work has made it possible to identify and create sources of alfalfa resistance to these diseases. In breeding nurseries, the most promising samples for further breeding were P 67 (2), LG 2, SU 9032, No. 27, No. 28, SU 85. In the competitive test and the control nursery, the best according to the results of the previous years (2014–2020 and earlier), samples of SGP 387, SGP 12, C 63-11 can be considered superior to the standard in productivity by an average of 15%, resistance to fusarium, brown spotting by 15–25%.
Identification of Crucial Genes and Regulatory Pathways in Alfalfa against Fusarium Root Rot

Abstract:Fusarium root rot, caused by Fusarium spp. in alfalfa (Medicago sativa L.), adversely impacts alfalfa by diminishing plant quality and yield, resulting in substantial losses within the industry. The most effective strategy for controlling alfalfa Fusarium root rot is planting disease-resistant varieties. Therefore, gaining a comprehensive understanding of the mechanisms underlying alfalfa's resistance to Fusarium root rot is imperative. In this study, we observed the infection process on alfalfa seedling roots infected by Fusarium acuminatum strain HM29-05, which is labeled with green fluorescent protein (GFP). Two alfalfa varieties, namely, the resistant 'Kangsai' and the susceptible 'Zhongmu No. 1', were examined to assess various physiological and biochemical activities at 0, 2, and 3 days post inoculation (dpi). Transcriptome sequencing of the inoculated resistant and susceptible alfalfa varieties were conducted, and the potential functions and signaling pathways of differentially expressed genes (DEGs) were analyzed through gene ontology (GO) classification and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis. Meanwhile, a DEG co-expression network was constructed though the weighted gene correlation network analysis (WGCNA) algorithm. Our results revealed significant alterations in soluble sugar, soluble protein, and malondialdehyde (MDA) contents in both the 'Kangsai' and 'Zhongmu No. 1' varieties following the inoculation of F. acuminatum. WGCNA analysis showed the involvement of various enzyme and transcription factor families related to plant growth and disease resistance, including cytochrome P450, MYB, ERF, NAC, and bZIP. These findings not only provided valuable data for further verification of gene functions but also served as a reference for the deeper explorations between plants and pathogens.

Abstract:Plant tissue culture is the finest method to propagate in large scale and to protect the rare, endangered and important plant species. It is in urgent to concentrate on conservation of rare, endangered medicinal and commercially important plant species. But the success rate of propagation in this method is attenuate in particular with a few medicinal plants. Also the researchers are facing lot of troubles with propagation of plant tissues and acclimatization of in vitro raised plants in the natural habitat. There are many reasons behind these problems. This review covered all that difficulties right from laboratory construction up to field adaptation of tissue cultured plants along with remedies to elucidate all the complications in this technique.

Abstract:(...) The addition of Al further decreased growth of acid-sensitive (AS) callus but not acid-tolerant (AT) callus. This suggests that plants from the AT germplasm rapidly express Al tolerance at the tissue level. With increasing time in culture, the AT germplasm also expresses acid tolerance in culture. The differences in callus growth shown by genotypes within germplasms indicates the cell culture procedure can be used to screen genotypes for acid or Al tolerance. Significant genotypic variability for acid, Al tolerance is still present within the germplasm, suggesting that continued progress from selection should still be possible

Fractal and Topological Analyses and Antioxidant Defense Systems of Alfalfa (Medicago sativa L.) Root System under Drought and Rehydration Regimes

Abstract:Alfalfa (Medicago sativa L.) is a nutritious forage plant in dryland regions with strong drought tolerance and broad adaptability. To understand the architectural characters and physiological functions of the root systems in response to different drought stress and rehydration, two experiments were conducted in a glasshouse. Experiment 1 used three alfalfa cultivars and four soil moisture treatments in a split-plot design with four replicates to investigate the response of root systems of different alfalfa cultivars to drought stress and subsequent rehydration. Experiment 2 used one alfalfa cultivar and four soil moisture treatments in a randomized complete block (RCB) design with three replicates to measure reactive oxidant species and antioxidant defense systems of alfalfa root. The soil moisture treatments included (1) 75–80% of the water-holding capacity (CK); (2) 60–65% of the water-holding capacity and subsequent rehydration to 75–80% of the water-holding capacity; (3) 45–50% of the water-holding capacity and subsequent rehydration to 60–65% and 75–80% of the water-holding capacity; and (4) 30–35% of the water-holding capacity and subsequent rehydration to 45–50%, 60–65% and 75–80% of the water-holding capacity. Our results demonstrated that plants had higher root biomasses (mean = 0.77 g), fractal dimensions (1.47), fractal abundances (3.84), and branching ratios (0.39) under severe water deficit and rehydration than under less stressful soil moisture conditions. Plant root systems tended to exhibit herringbone branching (Topological Index = 0.63) when soil moisture was at 60–65% water-holding capacity with the soil being then rehydrated, whereas they tended to have a smaller lateral root length, root surface area, root volume, and branching ratio and larger root diameter under more favorable soil moisture conditions for less stressful soil moisture treatments. Severe water stress and subsequent considerably improved superoxide (O2−) generation rate, the content of hydrogen peroxide (H2O2) and Malondialdehyde (MDA), and non-enzyme antioxidant glutathione (GSH), and reduced the content of ascorbate (AsA) and activity of catalase (CAT), ascorbate peroxidase (APX), and glutathione peroxidase (GPX). It is concluded that Medicago sativa L. cv Zhaodong (ZD) and cv Aohan (AH) exhibited higher drought resistances than cv Golden empress (GE) using topological and fractal analyses. This could be a key contributor to altering optimum soil moisture conditions and screening alfalfa cultivars based on root system architecture and antioxidant defense systems. The molecular functions warrant further investigation which could help explain the observed root phenotypes after drought conditions and rehydration.

Abstract:Objective: The study is to establish the key techniques of tissue culture of Lavandula angustifolia L. Method: Using stem with buds of Lavandula angustifolia L. as explant, the effect of disinfection time on contamination rate, types and ratio of plant regulator on propagation index and rooting, types of transplanting substance on plantlet survival rate were studied. Results and conclusion: The proper disinfection time of 0.1 % HgCl2 was 15min, by which the survival and contamination rate of plantlets respectively were 83.3 % and 25.0 %; MS +1.0mg/L BA + 0.3 mg/L IBA was the proper medium for propagation, which propagation index was 4.92; White + 0.4 mg/L NAA was the proper medium for rooting, which rooting rate and the average number of roots per plantlet respectively were 66.7% and 9.4; the survival rate of plantlets on transplanting substance which were peat and perlite mixed with an equal volume was 66.7 %.

Abstract:As plants are sessile, they cannot escape exposure to multiple environmental stresses. Over the course of evolution, a range of morphological and physiological adaptations has been developed in order to maximize their chances of survival. Alfalfa (Medicago sativa L.) is the most extensively cultivated forage legume in the world, while its yield and growing range are limited by abiotic stress. Physiology, genetic and molecular level studies have revealed complex regulatory processes that coordinate stress adaptation and tolerance, which are integrated at various levels. This review summarizes the current body of knowledge regarding the physiological, transcriptional, metabolic and genetic/epigenetic basis of the alfalfa stress response, along with the key signal transduction processes involved. Gathering the knowledge together will contribute to a more systematic understanding of the adaption to stress response in alfalfa, as well as provide guidance towards improving the crop through breeding.

Abstract:● This review systematically raises the subject concept of alfalfa biology. ● The discipline of alfalfa biology has been divided into six major sections. ● The recent advances from the perspective of discipline system have been reviewed. Alfalfa ( Medicago sativa ) is the main leguminous forage crop with great ecologic and economic value. The research of alfalfa in various fields has exploded, but has not been included in a systematic framework. This paper summarizes the status of global alfalfa research over the past 10 years, raise the subject concept of alfalfa biology, and review the recent advances from the perspective of discipline system as germplasm resources, multiomics and biotechnology, environmental biology, symbiotic nitrogen fixation, biological breeding and cultivation. This paper proposes the key unsolved scientific and technical issues in alfalfa biology, and hope to appeal the research interest of more plant scientists and to promote the development of alfalfa industry.

Abstract:The perennial legume alfalfa (Medicago sativa L.) is of high value in providing cheap and high-nutritive forages. Due to a lack of tillage during the production period, the soil in which alfalfa grows prunes to become compacted through highly mechanized agriculture. Compaction deteriorates the soil's structure and fertility, leading to compromised alfalfa development and productivity. However, the way alfalfa responses to different levels of soil compaction and the underlying molecular mechanism are still unclear. In this study, we systematically evaluated the effects of gradient compacted soil on the growth of different cultivars of alfalfa, especially the root system architecture, phytohormones and internal gene expression profile alterations. The results showed that alfalfa growth was facilitated by moderate soil compaction, but drastically inhibited when compaction was intensified. The inhibition effect was universal across different cultivars, but with different severity. Transcriptomic and physiological studies revealed that the expression of a set of genes regulating the biosynthesis of lignin and flavonoids was significantly repressed in compaction treated alfalfa roots, and this might have resulted in a modified secondary cell wall and xylem vessel formation. Phytohormones, like ABA, are supposed to play pivotal roles in the regulation of the overall responses. These findings provide directions for the improvement of field soil management in alfalfa production and the molecular breeding of alfalfa germplasm with better soil compaction resilience.
