Abstract:We report a facile, green, and high yielding approach for the synthesis and stabilization of relatively monodisperse gold nanoparticles using typical nonionic biosurfactant ethoxylated sterol (BPS-30). The obtained gold nanoparticles capped by BPS-30 not only showed remarkable stability after several months of storage under ambient conditions, but also exhibited exciting stability in high concentrations of electrolyte aqueous solutions such as acid, base, salt, and even physiological phosphate buffered saline (PBS). Derjaguin-Landau-Verwey-Overbeek (DLVO) theory was introduced to interpret the relationship between the capping mechanism and the excellent stability of the nanoparticles. UV-vis absorption spectroscopy was used to determine the yield of the gold nanoparticles. Moreover, the catalytic efficiency of the gold nanoparticles was evaluated by using the reduction of 4-nitroaniline by potassium borohydride in aqueous solution.
Multidentate Zwitterionic Chitosan Oligosaccharide Modified Gold Nanoparticles: Stability, Biocompatibility and Cell Interactions

Abstract:Surface engineering of nanoparticles plays an essential role in their colloidal stability, biocompatibility and interaction with biosystems. In this study, a novel multidentate zwitterionic biopolymer derivative is obtained from conjugating dithiolane lipoic acid and zwitterionic acryloyloxyethyl phosphorylcholine to the chitosan oligosaccharide backbone. Gold nanoparticles (AuNPs) modified by this polymer exhibit remarkable colloidal stabilities under extreme conditions including high salt conditions, wide pH range and serum or plasma containing media. The AuNPs also show strong resistance to competition from dithiothreitol (as high as 1.5 M). Moreover, the modified AuNPs demonstrate low cytotoxicity investigated by both MTT and LDH assays, and good hemocompatibility evaluated by hemolysis of human red blood cells. In addition, the intracellular fate of AuNPs was investigated by ICP-MS and TEM. It showed that the AuNPs are uptaken by cells in a concentration dependent manner, and they can escape from endosomes/lysosomes to cytosol and tend to accumulate around the nucleus after 24 h incubation but few of them are excreted out of the cells. Gold nanorods are also stabilized by this ligand, which demonstrates robust dispersion stability and excellent hemocompatibility. This kind of multidentate zwitterionic chitosan derivative could be widely used for stabilizing other inorganic nanoparticles, which will greatly improve their performance in a variety of bio-related applications.

Abstract:Parkinson's disease (PD) is a common neurodegenerative disease characterized by a substantial decrease of dopaminergic neurons in the substantia nigra pars compacta. The neurological deterioration during PD can be, in part, attributed to elevated levels of reactive oxygen species (ROS). Radical scavengers have previously been shown to protect dopaminergic cells from toxic effects in vitro. Hence, new approaches need to be investigated to improve the administration of antioxidants in order to provide neuroprotection. Polymers exhibiting catechol structures offer one such approach due to their interesting physicochemical properties. In the present study a photocrosslinkable dopamine-containing poly(β-amino ester) (DPAE) was synthesized from poly(ethylene glycol) diacrylate (PEGDA) and dopamine hydrochloride using Michael type addition. A water-in-oil emulsion technique was used to photo-crosslink the polymer into spherical microparticles. DPAE microspheres featured excellent scavenging properties towards 1,1-Diphenyl-2-picryl-hydrazyl (DPPH) radicals in a dose dependent manner and could even reduce the dissolved oxygen content of physiological solution. Furthermore, the concentrations required for radical scavenging were shown to be non-toxic towards dopaminergic SH-SY5Y cells as well as primary astrocytes and primary embryonic rat ventral midbrain cultures.

Abstract:Electrochemically mediated atom transfer radical polymerization was used to develop and synthesize carboxylic acid‐containing double hydrophilic block copolymer (DHBC), and DHBC was used to stabilize gold nanoparticles (AuNPs), to investigate the stability of DHBC@ AuNPs with a long interacting chain in different environments and the catalytic degradation performance. In this study, the gold nanoparticles (Au NPs) are capped with a novel, well‐defined double hydrophilic block copolymer, poly (ethylene glycols)‐block‐poly(mono‐2‐(methacryloyloxy) ethyl succinate) (PEG‐b‐PSEMA). The PEG‐b‐PSEMA, with long side chains and relatively narrow dispersity index, is successfully prepared by aqueous electrochemically mediated atom transfer radical polymerization using a PEG‐Br macroinitiator (Mn≈2000 g mol−1). By simply mixing PEG‐b‐PSEMA and Au precursor before adding reductants, coulombic interactions occur between the Au precursor and the carboxylate group of the PSEMA, as a result dense Au NPs are formed. This enables the Au NPs to remain dispersed even in the presence of temperature variations, pH adjustments, and incubation in organic solvents. Au NPs were endowed by PEG‐b‐PSEMA with more remarkable catalytic activity in organic solvents of 4‐nitrophenol into 4‐aminophenol than by sodium citrate (Au@citrate) and MeO‐PEG‐OH (Au@PEG). In addition, PEG‐b‐PSEMA‐stabilized Au NPs (Au@PEG‐b‐PSEMA) were also tested for the dye degradation property against Rhodamine‐B in only pH 10 basic solution, and over 99% of dye degradation can be achieved in 9 min. PEG‐b‐PSEMA induces coulombic interactions with Au NPs, providing outstanding stability for catalytic applications in organic or specific acidic‐base environments.

Abstract:Polysarcosine (PS), a non-ionic hydrophilic polypeptoid whose structure is similar to polypeptides, bearing repeating units of natural alpha-amino acid, has been used to stabilize gold nanoparticles (AuNPs) due to its excellent hydrophilicity and biocompatibility. Disulfide functionalized polysarcosines with different molecular weight were synthesized and used to cap AuNPs by traditional ligand exchange. The grafting of PS on AuNPs was evidenced by Fourier transform infrared (FTIR) spectroscopy and the alternation of surface zeta potential. The polysarcosine coated AuNPs (Au@PS) showed good stabilities in wide pH range and saline condition. They had strong resistance to ligand competition of dithiothreitol (DTT). They showed good stability in serum, with a molecular weight dependent interaction pattern with proteins. The Au@PS had very low cytotoxicity and cell uptake in vitro. Based on the results in vitro, polysarcosine with molecular weight of 5 kD with the best ability to stabilize AuNPs was used for in vivo test. The Au@PS had a longer circulation time in blood after intravenous injection than that of Au@PEG, indicating a better stealth-like property of polysarcosine. The Au@PS did not cause obvious toxicity in vivo, suggesting potential applications in disease diagnosis and therapy. (C) 2016 Elsevier Inc. All rights reserved.

Abstract:We report a biocompatible polysiloxane containing amphiphilic diblock copolymer, poly(ethylene oxide)-block-poly(gamma-methacryloxypropyltrimethoxysilane) (PEO-b-PgammaMPS), for coating and stabilizing nanoparticles for biomedical applications. Such an amphiphilic diblock copolymer that comprises both a hydrophobic segment with "surface anchoring moiety" (silane group) and a hydrophilic segment with PEO (M(n) = 5000 g/mol) was obtained by the reversible addition-fragmentation chain transfer (RAFT) polymerization using the PEO macromolecular chain transfer agent. When used for coating paramagnetic iron oxide nanoparticles (IONPs), copolymers were mixed with hydrophobic oleic acid coated core size uniformed IONPs (D = 13 nm) in cosolvent tetrahydrofuran. After being aged over a period of time, resulting monodispersed IONPs can be transferred into aqueous medium. With proper PgammaMPS block length (M(n) = 10 000 g/mol), polysiloxane containing diblock copolymers formed a thin layer of coating (approximately 3 nm) around monocrystalline nanoparticles as measured by transmission electron microscopy (TEM). Magnetic resonance imaging (MRI) experiments showed excellent T(2) weighted contrast effect from coated IONPs with a transverse relaxivity r(2) = 98.6 mM(-1) s(-1) (at 1.5 T). Such thin coating layer has little effect on the relaxivity when compared to that of IONPs coated with conventional amphiphilic copolymer. Polysiloxane containing diblock copolymer coated IONPs are stable without aggregation or binding to proteins in serum when incubated for 24 h in culture medium containing 10% serum. Furthermore, a much lower level of intracellular uptake by macrophage cells was observed with polysiloxane containing diblock copolymers coated IONPs, suggesting the reduction of nonspecific cell uptakes and antibiofouling effect.

Abstract:In this work, a highly stable nanocomposite film based on embedding gold nanoparticles (nanoAus) into a poly(3,4-ethylenedioxythiophene) (PEDOT) modified Pt electrode was fabricated and explored for dopamine sensing. The PEDOT film was synthesized in ionic liquids electrolyte-1-butyl-3methylimidazolium tetrafluoroborate ([bmim][BF4]). Citrate coated nanoAus were immobilized on the polymer matrix via electrostatic interactions to facilitate the mass transport and weaken the capacitive current. This modified electrode took advantage of the high stability and excellent permselectivity of PEDOT, and exhibited a wide linear response to dopamine from 6.0 x 10(-6) to 0.013 M. The detection limit was 0.2 mu M (s/n=3) and the amperometric response time was 2.5 s with chronoamperometry. Over the 8 months period of this study, the nanocomposite modified electrode still retained 85% of the original current response to dopamine. The highly stable modified electrode with improved sensitivity and selectivity could provide an ideal matrix for commercial applications.

Abstract:Narrow disperse poly(divinylbenzene-co-glycidyl methacrylate) (P(DVB-co-GMA)) microspheres with reactive epoxy group were prepared by distillation precipitation copolymerization of divinylbenzene (DVB) and glycidyl methacrylate (GMA) with benzoyl peroxide (BPO) as initiator in neat acetonitrile. The epoxy group was modified with ethylenediamine (EDA) for transferring to amino group, which was used as a stabilizer for the gold metallic nanocolloids during the in situ reduction of gold chloride trihydrate (HAuCl4) with sodium borohydride (NaBH4) as a reductant. The catalytic properties of the microsphere-stabilized gold nanocolloids (P(DVB-co-GMA)-NH2@Au) were investigated by the reduction of aqueous 4-nitrophenol (4-NP) to 4-aminophenol (4-AnP) with NaBH4 as reductant. The resultant microspheres were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), Fourier-transform infrared spectra (FT-IR), elemental analysis (EA), Zeta potential, X-ray photoelectron spectroscopy (XPS), inductively coupled plasma (ICP) mission spectrum, and ultraviolet–visible spectroscopy.

Abstract:Gold nanoparticles coated polystyrene/reduced graphite oxide (AuNPs@PS/RGO) microspheres have been successfully prepared via a facile process, and the decorative gold nanoparticles could prevent the aggregation of RGO by electrostatic repulsive interaction, and lead to high dispersibility of the composite. The prepared composite has a highly increased conductivity of 129 S m(-1) due to the unique electrical properties of citrate reduced gold nanoparticles. Being employed as an electrochemical sensor for detection of dopamine, the modified electrode exhibits remarkable sensitivity (3.44 mu A/mu M) and lower detection limit (5 nM), with linear response in a range of 0.05-20 mu M. Moreover, valid response to dopamine obtained in present work also indicates the prospective performances of AuNPs@PS/RGO microspheres to other biological molecules, such as nucleic acids, proteins and enzymes. Abstract:Poly(N-isopropylacrylamide)-b-[poly(ethyl acrylate)-g-poly(2-(diethylamino)ethyl methacrylate)] (PNIPAM-b-(PEA-g-PDEAEMA)) double hydrophilic graft copolymers were employed to prepare stable colloidal gold nanoparticles in situ with controllable size in aqueous media without any external reducing agent. PDEAEMA side chains served as reduction agent and stabilizer and PNIPAM segment acted as a hydrated layer to enhance the stability of gold nanoparticles. These gold nanoparticles showed a remarkable colloidal stability without any observable flocculation or aggregation for at least 2 months and they were characterized by UV-vis, XRD, TEM and AFM in detail. The size of gold nanoparticles can be tuned by adjusting the length of PDEAEMA side chains and the molar ratio of [DEAEMA]/[AuCl(4) over bar]. Both the increasing of the length of PDEAEMA side chains and the decreasing of [DEAEMA]/[AuCl(4) over bar] molar ratio resulted in the fall of size. (C)2009 Elsevier Ltd. All rights reserved.
Dopamine Receptor-Mediated Binding and Cellular Uptake of Polydopamine-Coated Nanoparticles

Abstract:Polydopamine (PDA)-coated nanoparticles (NPs) are emerging carriers of therapeutic agents for nanomedicine applications due to their biocompatibility and abundant entry to various cell types, yet it remains unknown whether their cellular entry engages cell-surface receptors. As monomeric dopamine (DA) is an endogenous ligand of dopamine receptor and raw ingredient of PDA, we elucidate the interaction between polyethylene glycol-stabilized, PDA-coated gold NPs (@PEG NPs) and dopamine receptors, particularly D2 (D2DR). After proving the binding of @PEG NPs to recombinant and cellular D2DR, we employ antibody blocking, gene knockdown, and gene overexpression to establish the role of D2DR in the cellular uptake of @PEG NPs in vitro. By preparing a series of PEG-coated AuNPs that contain different structural analogues of DA ( NPs), we demonstrate that catechol and amine groups collectively enhance the binding of NPs to D2DR and their cellular uptake. By intravenously injecting @PEG NPs to Balb/c mice, we reveal their in vivo binding to D2DR in the liver by competitive inhibition and immunohistochemistry together with their preferential association to D2DR-rich resident Kupffer cells by flow cytometry, a result consistent with the profuse expression of D2DR by resident Kupffer cells. Catechol and amine groups jointly contribute to the preferential association of NPs to D2DR-rich Kupffer cells. Peroxidase-like activity of gold nanoparticles stabilized by hyperbranched polyglycidol derivatives over a wide pH range

Abstract:The aim of this work was to carry out comparative studies on the peroxidase-like activity of gold nanoparticles (AuNPs) stabilized with low molecular weight hyperbranched polyglycidol (HBPG-OH) and its derivative modified with maleic acid residues (HBPG-COOH). The influence of the stabilizer to gold precursor ratio on the size and morphology of nanoparticles obtained was checked, and prepared nanoparticles were characterized by means of transmission electron microscopy and UV-Vis spectroscopy. The results indicated the divergent effect of increasing the concentration of stabilizers (HBPG-OH or HBPG-COOH) on the size of the nanostructures obtained. The gold nanoparticles obtained were characterized as having intrinsic peroxidase-like activity and the mechanism of catalysis in acidic and alkaline mediums was consistent with the standard Michaelis-Menten kinetics, revealing a strong affinity of AuNPs with 2, 2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 3, 3', 5, 5'-tetramethylbenzidine (TMB), and significantly lower affinity towards phenol. By comparing the kinetic parameters, a negligible effect of polymeric ligand charge on activity against various types of substrates (anionic or cationic) was indicated. The superiority of steric stabilization via the application of tested low-weight hyperbranched polymers over typical stabilizers in preventing salt-induced aggregation and maintaining high catalytic activity in time was proved. The applied hyperbranched stabilizers provide a good tool for manufacturing gold-based nanozymes, which are highly stable and active over a wide pH range.

Abstract:Abstract Owing to their remarkable properties, gold nanoparticles are applied in diverse fields, including catalysis, electronics, energy conversion and sensors. However, for catalytic applications of colloidal gold nanoparticles, the trade-off between their reactivity and stability is a significant concern. Here we report a universal approach for preparing stable and reactive colloidal small (~3 nm) gold nanoparticles by using multi-dentate polyoxometalates as protecting agents in non-polar solvents. These nanoparticles exhibit exceptional stability even under conditions of high concentration, long-term storage, heating and addition of bases. Moreover, they display excellent catalytic performance in various oxidation reactions of organic substrates using molecular oxygen as the sole oxidant. Our findings highlight the ability of inorganic multi-dentate ligands with structural stability and robust steric and electronic effects to confer stability and reactivity upon gold nanoparticles. This approach can be extended to prepare metal nanoparticles other than gold, enabling the design of novel nanomaterials with promising applications.

Abstract:A versatile ethanol-mediated oxidative polymerization of dopamine is demonstrated for the effective surface modification of nanomaterials. The presence of ethanol is found to significantly slow down the polymerization rate of dopamine and make the surface modification of nanomaterials with polydopamine more controllable in comparison to the water-phase polymerization. Various nanomaterials with different morphologies and surface properties, including one-dimensional (1-D) CNTs and iron oxide nanorods, 2-D nanodiscs, silver nanocubes and magnetite particles, were successfully modified by a layer of PDA with a controllable thickness from 5 to 100 nm, giving rise to PDA-shelled nanocomposites with well-defined structures and excellent water dispersibility. As exemplified by the case of magnetite particles, the PDA coating can dramatically reduce the cytotoxicity of nanomaterials and enhance their biocompatibility. This method is facile and particularly suitable for the surface engineering of nanomaterials, and thus promising for designing various functional nanostructures for a broad range of applications, such as drug delivery, protein purification, enzyme immobilization, and chemo/biosensing.

Abstract:Polymeric graft polyphosphazene containing 4-aminobenzoic acid diethylaminoethyl ester (DEAAB) as hydrophobic side groups was rationally designed and named PDEP. PDEP can self-assemble into a nanovesicle in water. More importantly, when compared with the amphiphile poly[(methoxy-poly(ethylene glycol))(ethyl p-aminobenzoate)]phosphazene (PEP) copolymer containing benzene rings and the amphiphile poly[(methoxy-poly(ethylene glycol)(N,N-diisopropylethylenediamine)]phosphazene (PDP) copolymer containing tertiary amino groups, the coexistence of benzene and tertiary amino groups in PDEP enabled it to effectively load water-soluble small-molecule doxorubicin hydrochloride (DOXHCl) into the vesicle and efficiently induce in situ transformation of gold tetrachloroaurate (HAuCl4) to gold nanoparticles (AuNPs) as both a reductant and a stabilizer. By optimizing the reduction conditions, such as the temperature, reaction time, and hydrophobic group in polymer/HAuCl4 molar ratio, the AuNP complex PDEP vesicles significantly inhibited the DOXHCl burst release at pH 7.4 while displaying a fast release responsive to pH 5.5.
Gold Nanoparticles Stabilized by δ‐Cyclodextrin in aqueous media: Characterization and Evaluation of their Catalytic Properties in the Reduction of 4‐Nitrophenol

Abstract:The synthesis of gold nanoparticles stabilized by cyclomaltononaose (δ‐CD) in aqueous phase was performed. Protection of the gold nanoparticles by standard native cyclodextrins such as α‐CD, β‐CD and γ‐CD has also been considered for comparison. All of these colloidal suspensions were fully characterized by FT‐IR, DLS, UV‐Vis spectroscopy, TEM, XPS and also NMR experiments. Finally, their catalytic activity was evaluated in the reduction of 4‐nitrophenol to 4‐aminophenol in the presence of an excess of sodium borohydride. Gold nanoparticles stabilized by δ‐CD presented good activity and exhibited better long‐term stability. This study highlighted the fact that the obtention of the best catalytic activity corresponds to not only a compromise between the size of the nanoparticles and the interaction of the substrate with the metal nanoparticles surface, but also the supramolecular interactions between the substrate and the cyclodextrin.

Abstract:Gold nanoparticles (GNPs) are promising nanomaterials for various biomedical applications owing to their remarkable optical properties and biocompatibility. However, their interfacial properties require modification for practical use in such applications. Herein, a simple method for modifying the surface of GNPs with polydopamine (PDA) to serve as a scaffold for the subsequent polymerization of hyperbranched polyglycerol (HPG) is reported. GNPs were first coated with PDA (GNP-PDA), and then ring-opening chemistry was used at this interface to modify GNP-PDA with HPG (GNP-PDA-HPG). The produced GNP-PDA-HPG shows not only excellent dispersibility in a salt-containing solution but also strong resistance to aggregation in high- and low-pH solutions, even after 10 days. Moreover, we demonstrate a one-pot method for functionalizing GNP-PDA with HPG and either COOH or trimethylammonium. Finally, we conjugated the trimethylammonium-functionalized GNP-PDA-HPG with fluorescent nanodiamonds to investigate the photothermal ability of the functional GNPs.

Abstract:This work originated from the need to functionalize surfactant-coated inorganic nanoparticles for biomedical applications, a process that is limited by excess unbound surfactant. These limitations are connected to the bioconjugation of targeting molecules that are often in equilibrium between the free aliquot in solution and that which binds the surface of the nanoparticles. The excess in solution can play a role in the biocompatability in vitro and in vivo of the final nanoparticles stock. For this purpose, we tested the ability of common surfactants – monothiolated polyethylene glycol and amphiphilic polymers - to colloidally stabilize nanoparticles as excess surfactant is removed and compared them to newly appearing multidentate surfactants endowed with high avidity for inorganic nanoparticles. Our results showed that monothiolated polyethylene glycol or amphiphilic polymers have an insufficient affinity to the nanoparticles and as the excess surfactant is removed the colloidal stability is lost, while multidentate high-avidity surfactants excel in the same regard, possibly allowing improvement in an array of nanoparticle applications, especially in those stated.

Abstract:Gold nanoparticles (AuNPs) play a vital role in biotechnology, medicine, and diagnostics due to their unique optical properties. Their conjugation with antibodies, antigens, proteins, or nucleic acids enables precise targeting and enhances biosensing capabilities. Functionalized AuNPs, however, may experience reduced stability, leading to aggregation or loss of functionality, especially in complex biological environments. Additionally, they can show non-specific binding to unintended targets impairing assay specificity. Within this work, citrate-stabilized and silica coated AuNPs (GNPs and SiGNPs, respectively) have been coated using N,N-dimethylacrylamide-based copolymers to increase their stability and enable their functionalization with biomolecules. AuNPs stability after modification has been assessed by a combination of techniques including spectrophotometric characterization, nanoparticle tracking analysis, transmission electron microscopy and functional microarray tests. Two different copolymers were identified to provide a stable coating of AuNPs while enabling further modification through click chemistry reactions, due to the presence of azide groups in the polymers. Following this experimental design, AuNPs decorated with ssDNA and streptavidin were synthesized and successfully used in a biological assay. In conclusion, a functionalization scheme for AuNPs has been developed that offers ease of modification, often requiring single steps and short incubation time. The obtained functionalized AuNPs offer huge flexibility, as the functionalization protocol can be personalized to match requirements of multiple assays.
