Abstract:Reversibly photo-cross-linkable pH-responsive block copolymer poly(ethylene oxide)-b-poly((2-(diethylamino)ethyl methacrylate-co-4-methyl-[7-(methacryloyl)oxyethyloxy] coumarin)) (PEO-b-P(DEA-co-CMA)) was synthesized via atom transfer radical polymerization (ATRP). Block copolymer nanogels could be easily prepared by first photo-cross-linking of the micelles at pH>7 and then adjusting the solution to pH<7. The photo-cross-linking was proved to be reversibly controlled under alternative irradiation of UV light at 365nm and 254nm. As a result, the cross-linking degrees and sizes of the nanogels can be easily controlled by alternatively UV light irradiation. Finally, the nanogels can serve as nanoreactors for the synthesis of gold nanoparticles. The protonated DEA units were first coordinated with HAuCl4, and then the electrostatically bounded AuCl4− anions were reduced to gold nanoparticles by NaBH4. The nanogel-supported gold nanoparticles were used in chemical catalysis. The pH-responsive photo-cross-linked nanogels have been characterized using dynamic light scattering, transmission electron microscopy, UV–vis spectra and 1H NMR spectroscopy measurements, respectively.
Mussel-Inspired Polydopamine: A Biocompatible And Ultrastable Coating For Nanoparticles In Vivo

Abstract:Bioinspired polydopamine (PDA) has served as a universal coating to nanoparticles (NPs) for various biomedical applications. However, one remaining critical question is whether the PDA shell on NPs is stable in vivo. In this study, we modified gold nanoparticles (GNPs) with finely controlled PDA nanolayers to form uniform core/shell nanostructures (GNP@PDA). In vitro study showed that the PDA-coated GNPs had low cytotoxicity and could smoothly translocate to cancer cells. Transmission electron microscopy (TEM) analysis demonstrated that the PDA nanoshells were intact within cells after 24 h incubation. Notably, we found the GNP@PDA could partially escape from the endosomes/lysosomes to cytosol and locate close to the nucleus. Furthermore, we observed that the PDA-coated NPs have very different uptake behavior in two Important organs of the liver and spleen: GNP@PDA in the liver were mainly uptaken by the Kupffer cells, while the GNP@PDA in the spleen were uptaken by a variety of cells. Importantly, we proved the PDA nanoshells were stable within cells of the liver and spleen for at least six weeks, and GNP@PDA did not show notable histological toxicity to main organs of mice in a long time. These results provided the direct evidence to support that the PDA surface modification can serve as an effective strategy to form ultrastable coatings on NPs in vivo, which can improve the intracellular delivery capacity and biocompatibility of NPs for biomedical application.
Optimized polydopamine coating and DNA conjugation onto gold nanorods for single nanoparticle bioaffinity measurements.

Abstract:Gold nanorods (NRs) have attracted a great deal of interest for a variety of biomedical and sensing applications. However, developing robust methods for biofunctionalizing NRs has continued to be challenging, especially for NR-DNA conjugates. This is due to the presence of cetyltrimethylammonium bromide (CTAB), which plays an essential role in controlling the anisotropic particle growth. In this article, we systematically explore the growth of a polydopamine (PDA) layer on a range of NR surfaces, comparing different polyelectrolyte and alkanethiol coatings as well as direct CTAB displacement. This revealed that the PDA layer thickness and growth rate is strongly dependent on the underlying nanorod functionalization chemistry and allowed us to establish a preferred route for the creation of stable, non-aggregated suspensions of PDA-coated NRs. The utility of this platform was then demonstrated by self-assembling packed monolayers of single-stranded DNA on the outer surface. Both the surface attachment and bioactivity of the resulting NR-DNA conjugates was then demonstrated by performing bulk solution and single nanoparticle imaging fluorescence measurements.

Abstract:Gold nanoparticles (AuNPs) play a vital role in biotechnology, medicine, and diagnostics due to their unique optical properties. Their conjugation with antibodies, antigens, proteins, or nucleic acids enables precise targeting and enhances biosensing capabilities. Functionalized AuNPs, however, may experience reduced stability, leading to aggregation or loss of functionality, especially in complex biological environments. Additionally, they can show non-specific binding to unintended targets, impairing assay specificity. Within this work, citrate-stabilized and silica-coated AuNPs (GNPs and SiGNPs, respectively) have been coated using N,N-dimethylacrylamide-based copolymers to increase their stability and enable their functionalization with biomolecules. AuNP stability after modification has been assessed by a combination of techniques including spectrophotometric characterization, nanoparticle tracking analysis, transmission electron microscopy and functional microarray tests. Two different copolymers were identified to provide a stable coating of AuNPs while enabling further modification through click chemistry reactions, due to the presence of azide groups in the polymers. Following this experimental design, AuNPs decorated with ssDNA and streptavidin were synthesized and successfully used in a biological assay. In conclusion, a functionalization scheme for AuNPs has been developed that offers ease of modification, often requiring single steps and short incubation time. The obtained functionalized AuNPs offer considerable flexibility, as the functionalization protocol can be personalized to match requirements of multiple assays.

Abstract:Polyethylene glycol (PEG) and poly(PEGMA) conjugated nanodiamond (ND) have been synthesized via “grafting to” and “grafting from” methods, respectively. In “grafting to” method, hydroxyl groups on ND surface were firstly oxidized to carboxyl groups, and then reacted with thionyl chloride to form acyl chloride groups. The acyl chloride functionalized ND (ND–COCl) was subsequently reacted with poly(ethylene glycol) monomethyl ether (mPEG) in the presence of triethylamine to generate mPEG conjugated ND (ND–mPEG). On the other hand, in “grafting from” method, ND–OH was modified with 2-bromoisobutyryl bromide (ND–Br), and then poly(PEG methyl ether methacrylate) (Poly(PEGMA)) chains were linked on the ND surface through surface-initiated atom transfer radical polymerization (ATRP) using ND–Br as the initiator and Cu(Br)/N,N,N′,N″,N″-pentmethyl diethylenetriamine (PMDETA) as the catalyst and ligand. The polymer conjugated ND particles were characterized using transmission electron microscopy (TEM), Fourier transform infrared (FT-IR) spectroscopy, thermal gravimetric analysis (TGA) and X-ray photoelectron spectroscopy (XPS). TGA analyses demonstrated that the polymer weight ratios through “grafting to” and “grafting from” methods were 29.8% and 34.4%, respectively. The mPEG and poly(PEGMA) conjugated ND nanoparticles exhibited enhanced dispersibility in organic media. More importantly, due to the relative high graft ratios and molecular weight, poly(PEGMA) functionalized ND was also dispersed well in water. Given the excellent physicochemical and biological properties of PEG and ND, the methods described in current work might be useful for the preparation of functional ND nanoparticles for potential biomedical applications.

Abstract:Gold nanorods coated with Polydopamine demonstrate exceptional photothermal and photocatalytic characteristics in both UV‐vis and NIR. How the thickness of the PDA shell affected the overall performance of these nanocomposites is examined and the underlying mechanism responsible for their exceptional capabilities is uncovered. This study highlights the synergistic potential of these nanocomposites and provides valuable insights for their strategic development across various fields and applications. Localized surface plasmon resonance (LSPRs) shown by gold nanorods (AuNRs) has several applications in photocatalysis, sensing, and biomedicine. The combination of AuNRs with Polydopamine (PDA) shells results in a strong photo‐thermal effect, making them appealing nanomaterials for biomedical applications. However, the precise roles and relative contributions of plasmonic effects in gold, and light‐to‐heat conversion in PDA are still debated. Herein, a hybrid nanoplatform made by an AuNR core surrounded by a polydopamine (PDA) shell is synthesized, and its photocatalytic behavior is studied. Synthesis is based on a seed‐mediated growth followed by the further self‐polymerization of dopamine hydrochloride (DA) on the surface of the AuNRs, and the effect of the thickness of the PDA shell on the plasmon response of the composite is the main examined parameter. Photocatalytic performance is tested toward Rhodamine 6G (Rh6G), with the nanocomposites achieving better performance than bare AuNRs and bare PDA nanoparticles. The degradation of 54% of Rh6G initial concentration is achieved within 60 min of irradiation with a catalyst concentration of 7.4 μg mL−1. Photodegradation kinetics, time‐resolved spectroscopy, and finite‐element‐method simulations of plasmons show that AuNRs plasmons, coupled with the low thermal conductivity of PDA, provide slow thermalization, while enhancing the charge carrier transfer.

Abstract:Mussel-inspired polydopamine (PDA) chemistry was employed for the surface modification of ordered mesoporous carbons (OMCs), improving the hydrophilicity, binding ability toward uranium ions, as well as enriching chemical reactivity for diverse postfunctionalization by either surface grafting or surface-initiated polymerization. Uniform PDA coating was deposited on the surface of CMK-3 type OMCs via self-polymerization of dopamine under mild conditions. Surface properties and morphology of the PDA-coated CMK-3 can be tailored by adjusting the dopamine concentration and coating time, without compromising the meso-structural regularity and the accessibility of the mesopores. Due to high density of −NH groups (4.7 μmol/m2 or 2.8 group/nm2) and −OH groups (9.3 μmol/m2 or 5.6 group/nm2) of the PDA coating, the modified CMK-3 showed improved hydrophilicity and superior adsorption ability toward uranyl ions (93.6 mg/g) in aqueous solution. Moreover, with the introduction of α-bromoisobutyryl bromide (BiBB) initiator to the PDA-coated CMK-3, we demonstrated for the first time that activators regenerated by electron transfer for atom transfer radical polymerization (ARGET ATRP) can be conducted for controlled growth of polymer brushes from the surface of OMCs. Thus, PDA chemistry paves a new way for surface modification of OMCs to create a versatile, multifunctional nanoplatform, capable of further modifications toward various applications, such as environmental decontamination, catalysis, and other areas.

Abstract:Gold nanoparticles (AuNPs) have received great attention in a number of fields ranging from the energy sector to biomedical applications. As far as the latter is concerned, due to rapid renal clearance and a short lifetime in blood, AuNPs are often encapsulated in a poly(lactic-co-glycolic acid) (PLGA) matrix owing to its biocompatibility and biodegradability. A better understanding of the PLGA polymers on the AuNP surface is crucial to improve and optimize the above encapsulation process. In this study, we combine a number of computational approaches to explore the adsorption mechanisms of PLGA oligomers on a Au crystalline NP and to rationalize the PLGA coating process toward a more efficient design of the NP shape. Atomistic simulations supported by a recently developed unsupervised machine learning scheme show the temporal evolution and behavior of PLGA clusterization by tuning the oligomer concentration in aqueous solutions. Then, a detailed surface coverage analysis coupled with free energy landscape calculations sheds light on the anisotropic nature of PLGA adsorption onto the AuNP. Our results prove that the NP shape and topology may address and privilege specific sites of adsorption, such as the Au {1 1 1} crystal planes in selected NP samples. The modeling-based investigation suggested in this article offers a solid platform to guide the design of coated NPs.
Gold nanorods with an ultrathin anti-biofouling siloxane layer for combinatorial anticancer therapy

Abstract:Despite the wide utility of gold nanorods (GNRs) in biomedical fields, only a few methods for modifying or coating the surface of GNRs suitable for biomedical applications are available. In this study, we report a new facile method that enables formation of an ultra-thin (nanometre-thickness) siloxane layer on GNRs with anti-biofouling properties and ligand functionalisation ability. A triblock random copolymer, poly(TMSMA- r -PEGMA- r -NAS), was used to coat GNRs. An ultrathin polymeric shell was formed surrounding GNRs through acid-catalysed crosslinking of silicates of TMSMA. The polymer-coated GNRs (p-GNRs) exhibited high colloidal stability in biological solutions of high ionic strength and long-term stability superior to that of PEG 2k -S-GNRs. The functionalities of NAS were demonstrated using two methods for conjugating targeting ligands and loading doxorubicin via electrostatic interactions. The ligand-specific cancer-targeting ability and combinatorial chemo-photothermal anticancer effects were validated in vitro and in vivo , suggesting their potential utility in various fields.

Dopamine Receptor-Mediated Binding and Cellular Uptake of Polydopamine-Coated Nanoparticles

Abstract:Polydopamine (PDA)-coated nanoparticles (NPs) are emerging carriers of therapeutic agents for nanomedicine applications due to their biocompatibility and abundant entry to various cell types, yet it remains unknown whether their cellular entry engages cell-surface receptors. As monomeric dopamine (DA) is an endogenous ligand of dopamine receptor and raw ingredient of PDA, we elucidate the interaction between polyethylene glycol-stabilized, PDA-coated gold NPs (@PEG NPs) and dopamine receptors, particularly D2 (D2DR). After proving the binding of @PEG NPs to recombinant and cellular D2DR, we employ antibody blocking, gene knockdown, and gene overexpression to establish the role of D2DR in the cellular uptake of @PEG NPs in vitro. By preparing a series of PEG-coated AuNPs that contain different structural analogues of DA ( NPs), we demonstrate that catechol and amine groups collectively enhance the binding of NPs to D2DR and their cellular uptake. By intravenously injecting @PEG NPs to Balb/c mice, we reveal their in vivo binding to D2DR in the liver by competitive inhibition and immunohistochemistry together with their preferential association to D2DR-rich resident Kupffer cells by flow cytometry, a result consistent with the profuse expression of D2DR by resident Kupffer cells. Catechol and amine groups jointly contribute to the preferential association of NPs to D2DR-rich Kupffer cells. PEG Branched Polymer for Functionalization of Nanomaterials with Ultralong Blood Circulation

Abstract:Nanomaterials have been actively pursued for biological and medical applications in recent years. Here, we report the synthesis of several new poly(ethylene glycol) grafted branched-polymers for functionalization of various nanomaterials including carbon nanotubes, gold nanoparticles (NP) and gold nanorods (NRs), affording high aqueous solubility and stability for these materials. We synthesize different surfactant polymers based upon poly-(g-glutamic acid) (gPGA) and poly(maleic anhydride-alt-1-octadecene) (PMHC18). We use the abundant free carboxylic acid groups of gPGA for attaching lipophilic species such as pyrene or phospholipid, which bind to nanomaterials via robust physisorption. Additionally, the remaining carboxylic acids on gPGA or the amine-reactive anhydrides of PMHC18 are then PEGylated, providing extended hydrophilic groups, affording polymeric amphiphiles. We show that single-walled carbon nanotubes (SWNTs), Au NPs and NRs functionalized by the polymers exhibit high stability in aqueous solutions at different pHs, at elevated temperatures and in serum. Morever, the polymer-coated SWNTs exhibit remarkably long blood circulation (t1/2 22.1 h) upon intravenous injection into mice, far exceeding the previous record of 5.4 h. The ultra-long blood circulation time suggests greatly delayed clearance of nanomaterials by the reticuloendothelial system (RES) of mice, a highly desired property for in vivo applications of nanomaterials, including imaging and drug delivery.

Abstract:Inspired by the mechanism of mussel adhesion, polydopamine (PDA), a versatile polymer for surface modification has been discovered. Owing to its unique properties like extraordinary adhesiveness, excellent biocompatibility, mild synthesis requirements, as well as distinctive drug loading approach, strong photothermal conversion capacity and reactive oxygen species (ROS) scavenging facility, various PDA-modified nanoparticles have been desired as drug carriers. These nanoparticles with diverse nanostructures are exploited in multifunctions, consisting of targeting, imaging, chemical treatment (CT), photodynamic therapy (PDT), photothermal therapy (PTT), tissue regeneration ability, therefore have attracted great attentions in plenty biomedical applications. Herein, recent progress of PDA-modified nanoparticle drug carriers in cancer therapy, antibiosis, prevention of inflammation, theranostics, vaccine delivery and adjuvant, tissue repair and implant materials are reviewed, including preparation of PDA-modified nanoparticle drug carriers with various nanostructures and their drug loading strategies, basic roles of PDA surface modification, etc. The advantages of PDA modification in overcoming the existing limitations of cancer therapy, antibiosis, tissue repair and the developing trends in the future of PDA-modified nanoparticle drug carriers are also discussed.

A polydopamine-based coating technology to manufacture probes for multimodality imaging.

Abstract:1120 Objectives The development of polydopamine (PD) coating technique has enabled many applications, and it inspires the development of multifunctional nanoprobes for molecular imaging. In addition to its superior optical behavior, PD also posesses good chelating property for transition metal ions. Furthermore, the coating layer provides a platform for further functionalization, for instance, molecules can be anchored onto the polymer layer through Schiff-type reactions or Michael addition reaction. Taking the advantages of such properties, we have developed a technology based on PD coating, to transform nanoplatforms into multimodal probes with the potential application in molecular imaging. Methods Gold nanoparticles with the size of 18 nm were first manufactured. Then the nanoparticles were mixed with dopamine hydrochloride in a mixed solution of water, ethanol and Tris buffer, and was stirred in air. A solution of copper sulfate was incubated with dopamine and added halfway of the reaction. After coating, polyethylene glycol or folic acid which had been functionalized with amine or thiol groups was added for further functionalization. The coated nanoparticles were separated, and various characterizations were performed. MTT tests were done to test the cytotoxicity. Moreover, ex vivo photoacoustic experiments were performed. Results The morphology of the nanoprobe was characterized by transmission electron microscopy and the coating layer with the thickness ranging from 4 nm to 15 nm was visible. Infrared red spectrum proved that the functionalized molecules were anchored onto the surface. The absorption spectra showed peaks at 536 nm and 595 nm. The molar ration between gold and copper is 20:1, which was determined by atomic absorption spectrum, and the presence of copper was also proved by X-ray photoelectron spectrum. Conclusions PD coating technology shows great potential in transforming nanomaterial to multifunctional probes for molecular imaging, with its innate chelating capability, optical behaviors and functionalization potentials. Abstract:Polydopamine (PDA), the final oxidation product of dopamine or other catecholamines, attracted much attention as versatile coatings that can be used to cover the surface of almost all materials with a conformal layer of adjustable thickness ranging from a few to about 100 nm. These PDA layers can be subsequently modified with molecules carrying nucleophilic groups or with metallic nanoparticles from solutions containing metallic cations. However, during the deposition of PDA film on the surfaces, the reaction products that are simultaneously obtained from the oxidation of catecholamines in solution precipitate. Hence, some recent effort has been devoted to produce PDA in the form of nanoparticles. The aim of this short review is to give a comprehensive description of the synthesis methods yielding of PDA nanoparticles in the absence or in the presence of templating agents (polymers, polyelectrolytes, surfactants, proteins, and even some small organic molecules). We will also describe the use of thin PDA layers to coat already synthesized nanoparticles or nanotubes. Finally, several first applications of the obtained PDA nanoparticles will be described.

Versatile Polydopamine Platforms: Synthesis and Promising Applications for Surface Modification and Advanced Nanomedicine

Abstract:As a mussel-inspired material, polydopamine (PDA), possesses many properties, such as a simple preparation process, good biocompatibility, strong adhesive property, easy functionalization, outstanding photothermal conversion efficiency, and strong quenching effect. PDA has attracted increasingly considerable attention because it provides a simple and versatile approach to functionalize material surfaces for obtaining a variety of multifunctional nanomaterials. In this review, recent significant research developments of PDA including its synthesis and polymerization mechanism, physicochemical properties, different nano/microstructures, and diverse applications are summarized and discussed. For the sections of its applications in surface modification and biomedicine, we mainly highlight the achievements in the past few years (2016–2019). The remaining challenges and future perspectives of PDA-based nanoplatforms are discussed rationally at the end. This timely and overall review should be desirable for a wide range of scientists and facilitate further development of surface coating methods and the production of PDA-based materials.

Rapidly-Deposited Polydopamine Coating via High Temperature and Vigorous Stirring: Formation, Characterization and Biofunctional Evaluation

Abstract:Polydopamine (PDA) coating provides a promising approach for immobilization of biomolecules onto almost all kinds of solid substrates. However, the deposition kinetics of PDA coating as a function of temperature and reaction method is not well elucidated. Since dopamine self-polymerization usually takes a long time, therefore, rapid-formation of PDA film becomes imperative for surface modification of biomaterials and medical devices. In the present study, a practical method for preparation of rapidly-deposited PDA coating was developed using a uniquely designed device, and the kinetics of dopamine self-polymerization was investigated by QCM sensor system. It was found that high temperature and vigorous stirring could dramatically speed up the formation of PDA film on QCM chip surface. Surface characterization, BSA binding study, cell viability assay and antibacterial test demonstrates that the polydopamine coating after polymerization for 30 min by our approach exhibits similar properties to those of 24 h counterpart. The method has a great potential for rapid-deposition of polydopamine films to modify biomaterial surfaces.

Abstract:Well known for the adhesive property, mussel-inspired polydopamine (PDA) has been shown to enhance performance in a wide range of adsorption-based applications. However, imparting porous nanostructures to PDA materials for enhanced loading capacities has not been demonstrated even when surfactants were present in the synthesis. Herein, we report on the preparation of mesoporous PDA particles (MPDA) based on the assembly of primary PDA particles and Pluronic F127 stabilized emulsion droplets on water/1,3,5-trimethylbenzene (TMB) interfaces. The key to the formation of this new type of the MPDA structure is the full utilization of the π-π stacking interactions between PDA structures and the π-electron-rich TMB molecules. Remarkably, this method presents a facile approach for MPDA particles with an average diameter of ∼90 nm, slit-like pores with a peak size of ∼5.0 nm as well as hollow cavities. When used as the adsorbent for a model dye RhB, the MPDA particles achieved an ultrahigh RhB adsorption capacity of 1100 μg mg<sup>-1</sup>, which is significantly higher than that for the PDA-reactive dyes with Eschenmoser structure. Moreover, it was demonstrated that the cavity space in MPDA can facilitate high volumetric uptake in a capillary filling/stacking manner via the π-π interactions. These developments pave a new avenue on the mechanism and the designed synthesis of functional PDA materials by organic-organic composite assembly for advanced adsorption applications.

Abstract:Graphene nanoparticles (GNPs) can serve as a versatile platform for the development of drug-delivery systems by means of suitable functionalization strategies. By using a green physical method, we prepared GNPs formed by N < 10 stacked graphene layers with an average lateral size of about 102 nm. These GNPs are naturally endowed with carboxyl groups decorating their edges that facilitates further functionalization. Thus, the cell permeable peptide (CPP) poly-arginine-11 (R11) was grafted on the GNPs to obtain the peptide-functionalized GNPs (R11@GNP). Moreover, the preparation of a non-covalent complexes with 1-pyrene carboxylic acid (PyCA) demonstrates that the GNPs surface can be loaded with small molecules for drug delivery purposes. The structure of R11@GNP and PyCA@GNP supramolecular systems as well as the evidence of the covalent and non-covalent bond, respectively, was investigated. The thorough analysis of the functionalized GNPs by UV-vis, FTIR and Raman spectroscopies, Dynamic Light Scattering, and Z-potential measurements illustrates the potential of well-established and easily available techniques to achieve a structural characterization at the molecular level. Fluorescence spectroscopy allows collecting further evidence of the formation of stable π-π complexes between GNPs and PyCA, and provides a first test of the thermally induced release of the absorbed molecules.

Facile Approach to Functionalize Nanodiamond Particles with V-Shaped Polymer Brushes

Abstract:Nanodiamond (ND) particles were functionalized with V-shaped polymer brushes of polystyrene and poly(t-butyl methacrylate) (PtBMA) through the reaction of surface carboxylic groups of NDs toward epoxy functionalities located in the middle of the polymer precursor, an ABC-type triblock copolymer. The block copolymer was prepared through sequential radical polymerizations of tBMA, glycidyl methacrylate (GMA), and styrene mediated by a reversible addition-fragmentation chain transfer process, in which the lengths of different segments are well-controlled by virtue of the living nature of the reaction. The polymerization product, PtBMA-b-PGMA-b-PS, carried a short block of PGMA in the middle, which was used for subsequent reaction with -COOH on the convex surface of the NDs. This grafting-onto approach through a center-linking process functionalized nanoparticles with V-shaped polymer brushes possessing an exact 1: 1 molar ratio of different arms. Furthermore, ND particles with amphiphilic functionalities were prepared after hydrolysis of the PtBMA segment. The obtained polymer grafted ND was characterized by electron microscopy (TEM and SEM), NMR and IR spectroscopy, and TGA. The product not only formed stable dispersions in organic solvents such as tetrahydrofuran, toluene, and chloroform but also self-assembled at oil-water interfaces to form flat films or large droplets of water-in-oil and oil-in-water. The mechanism of self-assembly at liquid-liquid interfaces is discussed.
