Abstract:Quantitative descriptive sensory analysis(QDSA),gas chromatography-ion mobility spectrometry(GC-IMS)and Heracles NEO electronic nose(E-nose)combined with chemometrics were used for differential analysis of volatile compounds in essential oils(EOs)extracted from sweet orange peels(blood and navel)using cold pressing(CP),microwave-assisted hydrodistillation(MAHD)or spinning cone column(SCC).The results indicated that significant variations in flavor composition were found among the three EOs.The CP oil had the highest sensory preference with stronger fruity,sweet,and floral aromas.The MAHD oil had stronger fatty,green,and woody aromas,and the SCC oil had a lower aroma intensity.A total of 64 volatile components were identified by GC-IMS,terpenes,alcohols and aldehydes being the major ones,followed by acids,esters and other compounds.The fingerprint indicated that the contents of terpenes in the CP and MAHD oils were significantly higher than that in the SCC oil,whereas the contents of alcohols and aldehydes in the MAHD and SCC oils were significantly higher than those in the CP oil.Based on relative odor activity value(ROAV),β-myrcene and linalool were identified as the major contributors to the flavor of sweet orange EO.Moreover,using GC-IMS combined with partial least square discriminant analysis(PLS-DA)and variable importance in the projection(VIP),13 differential volatile compounds were identified as aroma markers to distinguish among the 6 oils.Additionally,E-nose combined with principal component analysis(PCA)and discriminant factor analysis(DFA)also successfully distinguished among the 6 EOs according to their volatile compounds.

Abstract:Objective: To compare the difference of volatile components in bergamot by different sample collection methods.Methods: Volatile components in bergamot were collected by headspace sampling and steam distillation and analyzed by GC-MS.Results: There were some differences between the above methods. There were 19 components identified from extract by steam distillation; whereas 17 components were identified from extract by headspace sampling. There were 13 common components a-mong the identified compounds in the samples collected with different method and the relative contents of the two most impor-tant components were basically identical. Conclusion: More variety of components can be identified in the samples collected with headspace sampling. More volatile components could be identified if the two collection methods were combined.

Abstract:Objective:To determine and analyze the volatile compounds of twelve common species of bee pollen.Methods:The volatile compounds in twelve common species of bee pollen were analyzed and compared by GC-MS combined with simultaneous distillation and solvent extraction(SDE).Results:A total of 74 volatile components were determined in samples,including of 41 relatively common components and 33 relatively specific components.The analyzed compounds were mainly composed of alkanes, alkenes, alkynes, aldehydes, ketones, alcohols, lipids, benzenes, phenols and carboxylic acids,et al.,and pollen samples had quite different profiles.Relevant specific ingredients were characterized in pollen of Fagopyrum esculentum L, Helianthus annuus L., Nelumbo nucifera Gaertn,Rosa rugosa,Schisandra chinensis,and Vicia faba L.Conclusion:The volatile compounds tends to be species-specific.A relative food safety of bee pollen was indicated,as prohibited substances in daily used essences and pesticide residues were nearly inexistence in pollen volatiles.

Abstract:Fresh Mentha haplocalyx is a well known traditional Chinese medicinal material (CMM) used in both China and America. This paper reports analysis of the volatile components of fresh Mentha haplocalyx by comprehensive two-dimensional gas chromatography (GCxGC) and high-resolution time-of-flight mass spectrometry (HR-TOF-MS), a combination that provides almost complete chemical separation with elemental composition determination of analytes. 163 ketones and terpenes, including menthol and menthone, were tentatively identified, including enantiomers. This study suggests that GCxGCxHR-TOF-MS is suitable for routine identification of target compounds and enantiomers in CMM.

Abstract:OBJECTIVE To develnp a quantitative method for the determination of seven different components of volatile oil in Elsholtzia blanda,and compare primarily their contents extracted with different methods.METHODS The contents of seven components were analyzed by headspace gas chromatography with INNOWAX capillary column(0.53 mm×30 m,5μm).The column temperature raised by programming,the intial temperature was 60℃,and was raised to 90℃at the rate of 30℃·min~(-1)for 30 min and then was raised to 180℃at the rate of 5℃·min~(-1).The injector temperature was set at 200℃,the detector temperature of FID was at 240℃. RESULTS Seven components were completely separated.The calibration curves ofα-pinene,β-pinene,1,8-cineol,camphor,linalool, bornyl acetate,borneol,were linear within the range of 33～330,32～320,111～1110,238～2380,140～1400,65～650,53～530 mg·L~(-1),the correlation coefficient were 0.9999,0.9998,0.9998,0.9996,0.9997,0.9995,0.9995,and their average recover- ies(n=9)were 96.6%,97.0%,97.8%,99.9%,96.5%,97.4% and 97.1%,respectively.CONCLUSION The method is simple, accurate and sensitive,and can be used for quality control of E.blanda and its further reseach.

Abstract:Volatile oils of fresh myrica rubra fruits from muye,dongkui and heitan were analyzed by GC-MS and compared. 55 compositions were identified from muye myrica rubra fruit,and the main components was monoterpene category. 49 compositions were identified from dongkui myrica rubra fruit,and the main components were sesquiterpene compounds and saturated group. 48 compositions were identified from Heitan myrica rubra fruit,and the main components were sesquiterpene compounds and saturated group. Aromatic and putrescible properties of muye myrica rubra originated from the monoterpene possibly.

Abstract:The volatile oils from leaves of different Myrica rubra varieties were analyzed and compared by GC-MS.The forteen same components were detected in the three kinds of Myrica rubra leaves.The contents of the same components accounted for 53.3%,48.4%,55.5% of Muyemei,Heitanmei,Dongkuimei overall volatile oils repectively.The essential oil contained multiple terpenes in the Myrica rubra leaves.The components had a role of preservation of Myrica rubra fruit.The relative content of terpenes in the Dongkuimei leaves was the highest.This could explaine why the period of keep-freshness of Dongkuimei fruits was relatively longer.The relative content of aroma substance in the Muyemei leaves was the highest.This was the reason why the aroma of Muyemei fruits was the most full-bodied.

Abstract:Objective:To study the chemical components of volatile oil from the root of Angelica dahurica Benth.et Hook.f.ex Franch.et Sav.(Xing′anbaizhi)and compare the components difference of volatile oil with that of A.dahurica var.dahurica cv.Qibaizhi(Qibaizhi).Methods:The volatile oil was extracted by water-steam distillation and analyzed by GC-MS with capillary gas chromatographic column.Results:Two hundred forty-four chromatographic peaks were detected,among which 76 compounds have been identified.The contents of them made up 86.13% of the total volatile oil of Xing′anbaizhi.The main components in the volatile oil of Xing′anbaizhi were tetradecanol(19.43%),α-limonene(15.25%),3-carene(10.94%),1-dodecanol(5.74%)and 1R-α-pinene(3.85%).Conclusion:The two species contained 38 common components,which amounted 34.54% of the total volatile oil of Xing′anbaizhi.The contents of sesquiterpenes and their derivates in Xing′anbaizhi were lower than that of Qibaizhi.

Abstract:OBJECTIVE To study the chemical constituents of ethanolic extract from herbs of Dicliptera chinensis. METHOD The compounds were separated by silica gel column chromatography, preparation TLC and reverse phase HPLC, their structures were identified by the spectroscopic methods of UV, NMR and MS. RESULT Seven compounds were isolated from ethyl acetate extract. Their structure were identified as octasulphur (1), secoisolariciresinol dimethyl ether diacetate (2), 5-methoxy-4, 4'-di-O-methyl secolariciresinol (3), chinensinaphthol methyl ester (4), loliolide (5), beta-sitosterol 3-O-beta-D-glucopyranoside (6) and stigmasterol 3-O-beta-D-glucopyranoside (7). CONCLUSION All the compounds except 6 were obtained from the plants of Dicliptera for the first time.

Abstract:The volatile oil from Vaccinium bracteatum Thumb.leaves was analyzed by GC-MS.65 peaks were separated and 49 compounds were identified,which represented about 91.94 % of the total content.The main chemical constituents were terpenes 54.75 %,open chain alkanes 6.93 %,cycloparaffins 20.13 %,olefins 0.60 %,aromatics 1.60 %,phenol,ether and alcohol 3.39 %,aldehyde and ketone 2.14 %,ester 1.15 %,heterocyclic compounds 0.47 % and amines 0.78 %.Nerolidol 20.01 %,(Z,Z,Z)-1,5,9,9-tetramethyl-1,4,7-cycloundecatriene 17.99 % and caryophyllene 9.59 % were the main components in the identified terpenes.12 kinds of industrial production flavors were found from volatile oil in Vaccinium bracteatum leaves,and five components have insecticide and antibacterial activity.

Abstract:OBJECTIVE:To determine the structures and contents of the volatile components of flowers of Fritillaria thunbergii, and investigate the effects of operation modes on its volatile components.METHOD:The volatile oils were first obtained by the hydrodistillation assay and then submitted to gas chromatography-time-of flight mass spectrometry (GC-TOF-MS) analysis.RESULT:More than 60 peaks were resolved, and 39 of which were identified quantitatively and qualitatively based on high-resolution spectra and compounds library screening. Among these identified components, the octadecatrienoic acid methyl esters were major components in the unprocessed flowers, while some aromatic aldehydes and ketones, such as benzeneacetaldehyde and 1-(2-hydroxy-5-methylphenyl)-ethanone, were prominent components in the flowers both dried in the fluidized bed and in shadow. In addition, the flowers dried in the fluidized bed were more fragrant than other flowers.CONCLUSION:The component and contents closely related to their processing mode, and the fluided bed drying may be a best choice to process the flowers of F. thunbergii.

Abstract:A one-step ultrasound/microwave-assisted solid-liquid-solid dispersive extraction procedure was used for the simultaneous determination of eight neonicotinoids (dinotefuran, nitenpyram, thiamethoxam, clothianidin, imidacloprid, acetamiprid, thiacloprid, imidaclothiz) in dried Dendrobium officinale by liquid chromatography combined with electrospray ionization triple quadrupole tandem mass spectrometry in multiple reaction monitoring mode. The samples were quickly extracted by acetonitrile and cleaned up by the mixed dispersing sorbents including primary secondary amine, C-18, and carbon-GCB. Parameters that could influence the ultrasound/microwave-assisted extraction efficiency such as microwave irradiation power, ultrasound irradiation power, temperature, and solvent were investigated. Recovery studies were performing well (70.4-113.7%) at three examined spiking levels (10, 50, and 100 mu g/kg). Meanwhile, the limits of quantification for the neonicotinoids ranged from 0.87 to 1.92 mu g/kg. The method showed good linearity in the concentration range of 1-100 mu g/L with correlation coefficients >0.99. This quick and useful analytical method could provide a basis for monitoring neonicotinoid insecticide residues in herbs.

Abstract:Objective: Scrophularia ningpoensis hemsl is a kind of traditional Chinese medicinal materials with high medicinal purposes value and wide application. It had important meanding to analyze its volatile oil consitituents on expanding its utilization and increasing the added value. Methods: The volatile compounds of scrophularia ningpoensis hemsl were isolated with steam distillation and identified by capillary GC/MS and the relative contents of the constitons were detemined by area normalization method. Results: 64 volatile components were identified and determined, accounting for 96.89% of the total area of the peaks. Conclusion: The main volatile compounds are: furfural, 5-methyl-2-furan carboxaldehyde, Z-5-methyl-6-heneicosen-ll-one, palmitic acid, furfuryl alcohol, acetylfuran, terpineol, m-Cresol, 2-Acetylpyrrole, o-Guaiacol, phenylethyl alcohol, (2Z)-2, 4-pentadien-1-ol, 3-ethyl, isovaleric acid, Spiro[2.4]heptan-4-one, phenylethyl alcohol, 4-ethyl-2-methoxy-phenol, 4-Vinylguaiacol and sobutyric acid. © 2011 IEEE.
Combined Analysis of Volatile Compounds and Extraction of Floral Fragrance Genes in Two Dendrobium Species

Abstract:Many species of the Dendrobium genus are traditional Chinese herbal medicine and ornamental plants. Flower fragrance is one of the most important horticultural ornamental characters and plays a crucial role in the ecology, economy, and aesthetics of plants. However, the volatile constituents and key regulatory genes related to floral biosynthesis are poorly understood. In this experiment, the flowers from two species of Dendrobium with high-scent smells, Dendrobium moniliforme (L.) Sw. (D. moniliforme), and light-scent smells, Dendrobium nobile “H1” (D. “H1”), were selected. The aim of this study was to explore the key gene expression profiles of floral biosynthesis by combining volatile constituent determination and transcriptome analysis in two different Dendrobium species. Physiological determination results showed that 60 volatile compounds were identified in D. moniliforme and 52 volatile compounds were identified in D. ‘H1’ flowers in four flowering stages, and the full bloom stage was the most complicated stage because there were 41 and 33 volatile compounds, respectively. These compounds belong to terpenes, aromatics, fatty acids, nitrogenous compounds, ketones, alcohols, and alkanes, respectively. The components identified in the gynandrium and petals revealed that the petals were probably the most important sites affecting the release of volatiles. The relative content of terpene compounds was the highest, with 77.25% (D. moniliforme) and 50.38% (D. “H1”), respectively. Transcriptome analysis showed that differentially expressed genes (DEGs) were highly enriched in terpenoid backbone biosynthesis and that four linalool synthetase (LIS) genes were up-regulated in high-scent smell species. This study is helpful to explore the key genes of flower fragrance and provides a theoretical basis for further understanding of the regulatory molecular functions of floral synthesis and release, as well as for the cultivation of new aromatic species.

Abstract:Volatile oil,extracted from the leaves of Mangifera persiciformis by steam distillation,was analyzed by gas chromatography-mass spectrometry(GC-MS)for the first time.The relative contents in the volatile oil were determined by peak area normalization.38 components were identified,which accounted for 94.52% of the volatile oil of the leaves.The major compositions were Germacrene D(18.33%),Ledol(9.46%),Caryophyllene(9.04%),δ-Cadinene(5.73%),1(R)-α-Pinene(5.32%),β-Elemene(4.90%),α-Gurjunene(4.26%),Phenol,2-(1,1-dimethylethyl)-5-methyl-(3.78%),allo-aromadendrene(3.45%),δ-Elemene(3.41%),Selina-3,7(11)-diene(3.27%),Spathulenol(3.20%),and many of the components had biological activity.This study will provide theoretic basis for the multiple utilization and developments of the leaves of M.persiciformis.

Abstract:The effects of three sample pretreatment methods,i.e.the solvent extraction(SE),the solid-phase microextraction(SPME)and the microwave-assisted solid-phase microextraction(MAE-SPME)were investigated on the analysis of strawberry volatiles with the gas chromatography-mass spectrometry(GC-MS)method.By MAE-SPME,85 volatile components were detected,of which,53 components were esters and the rest were aldehydes,ketones,alcohols,organic acids,ethers,alkenes,aromatics,heterocyclic compound and so on.Meanwhile,26 and 16 volatile components were detected with SE and SPME respectively.The results indicated that MAE-SPME is a better method for extracting aroma components from fruist.

Abstract:The flowers of Elaeagnus angustifolia L. have been used as a homologous variety in China, whose quality seriously relies on the compositions during the flowering period. Unfortunately, studies on the variations of volatile compounds during the flowering season are rarely reported. Herein, a gas chromatography-mass spectrometry-based untargeted metabolomic methodology was proposed for the comprehensive analysis of volatile compounds in E. angustifolia flowers to classify various flowering stages. Samples from four flowering stages were collected, including the initial bloom stage, pre-full bloom stage (70–80% of flowers), full bloom stage, and ending of the bloom stage. Simultaneous distillation extraction was used for the extraction of volatile compounds in the flowers, which was then analyzed by a newly developed chemometric data analysis tool, autoGCMSDataAnal. An advantage of the developed methodology is that compounds can be accurately screened and identified. Finally, 59 compounds that showed significant difference among four flowering stages were screened and 31 compounds were identified. Sample clustering results from principal component analysis and hierarchical clustering analysis suggested that flowers from the pre-full bloom stage and full bloom stage may be more suitable when used as raw materials for industrial products.

Abstract:Objective To study the chemical constituents in the whole herb of Dicliptera chinensis.Methods Separation and purification were conducted by silica gel column chromatographic and preparative TLC methods,the structures were identified by IR,NMR,and MS.Results Thirteen compounds were isolated from petroleum ester extract fraction.Their structures were identified as hexatriacontanol(Ⅰ),stearic acid(Ⅱ),lupenone(Ⅲ),lupeol(Ⅳ),4-sitost-4-en-3-one(Ⅴ),stigmast-5-en-7-oxo-3β-yl palmitate(Ⅵ),β-sitosterol(Ⅶ),oleanolic acid(Ⅷ),3β,6β-stigmast-4-en-3,6-diol(Ⅸ),6β-hydroxy-stigmast-4-en-3-one(Ⅹ),3β-hydroxy-stigmast-5-en-7-one(Ⅺ),dehydrovomifoliol(ⅩⅡ),and vomifoliol(ⅩⅢ).Conclusion All the compounds except Ⅱ and Ⅶ are obtained from the plants of Dicliptera Juss.for the first time.
