Abstract:An improved process for synthesis of esmolol hydrochloride is described. p-Hydroxycinnamic acid from p-hydroxybenzaldehyde was hydrogenated,esterified with methanol alkylated with epichlorohydrni in PTC conditions and followed by isopropylamination to give the title compound in 38.9% overall yield.

Abstract:Celiprolol hydrochloride, a beta-blocker drug, has been synthesised from 4-chloronitrobenzene. The route involved hydrolysis of the chloride and acetylation of the phenol, reduction of the nitro group, and acylation of theamine. This was followed by Fries rearrangement of the acetyl group, Williamson etherification with epichlorohydrin, followed by opening of the epoxide ring and salt formation. The overall yield of celipropol was 39.1% on the basis of 4-chloronitrobenzene. The Fries rearrangement was substantially improved. The process is suitable for industrial application because of its convenient and environmentally friendly reaction conditions.

Abstract:An improved synthetic process for 3-benzyloxy-4-n-butylaniline hydrochloride the key intermediate of nequinate which is an anticoccidal drug to parasites of Eimeria,was described.The process was started with 3-aminophenol,via acylation,Fries rearrangement,benzylation,hydrolysis,and Huang-Minlon reduction,to give the title compound.The optimal reaction conditions and molar yield were as follows:acylation,130—140℃ for 3 h;Fries rearrangement,140℃ for 5 h,77.9%(based on 3-aminophenol);benzylation,70℃ for 2 h and 80℃ for 4 h,81.3%;hydrolysis and Huang-Minlon reduction,reflux for 90 min,78.2%.The structures of the main compounds were confirmed by 1H NMR.The process is much of industrial value because of high yields,cheap and available materials,moderate reaction conditions and convenient operations.

Abstract:The synthetic method of polyester monomer 5-hydroxylevulinic acid(5-HLA) was improved and simplified.Using levulinic acid(LA) derived from biomass as raw material,5-HLA was synthesized via modified bromination and hydrolysis processes.In the bromization reaction of LA,the reaction conditions such as reaction temperature,drop time of Br_2 and reaction medium were optimized.The side product was reused via rearrangement of methyl 3-bromolevulinate to methyl 5-bromolevulinate(5-MBL) and the yield of 5-MBL was thus improved to 45% from an earlier reported value of 30%.Then pure 5-HLA in 30% yield was obtained via a simplified one-step hydrolysis of 5-MBL followed by concentration of the hydrolysate,extraction of 5-HLA in a continuous manner with stirring,and recrystallization in chloroform.Compared with the two-step method,the processing time was shortened and the yield of 5-HLA was improved.

Abstract:OBJECTIVE:To synthesize epalrestat and to improve the method of synthesis.METHODS:Glycine reacted with carbondisulfide,then with ClCH(2)COONa to give 3-carboxymethylrhodanine. PhCHO reacted with CH(3)CH(2)CHO in NaOH/EtOH solution to produce 2-methylcinnamaldehyde.3-carboxymethylrhodanine and 2-methylcinnamaldehyde were treated with NH(3).H(2)O to obtain epalrestat.RESULT:The described method was effective in synthesis of Epalrestat and the yield was higher than that of in references.CONCLUSION:The results suggest that this method is suitable for industrial production.

Abstract:A procedure for synthesis of 2-methoxyphenothiazine (1) has been developed, starting from resorcinol and aniline by condensation, following methylation and cyclization. p-Toluenesulfonic acid and polysubstituted aromatics were employed as the catalyst of condensation and the solvent of cyclization, respectively, to improve the yield. The use of parallel experiments, statistical experimental design, and multivariate modeling made the total yield of the procedure as high as 74.2%. J. Heterocyclic Chem., (2012).

Abstract:Simvastatin is a new antihyperlipidaemic agent. It can be synthesized with the material of lovastatin, hydroxy protecting agent of trimethylchlorosilane, and t-butyl lithium instead of pyrrole lithium. The total yield is about 78%.The structure is identified by UV,IR,MS,NMR spectrums.This method is very easy and the cost is very low, but the yield is high. Most of organic solvents can be recovered in order to be used next time. So this method will have perfect industrialization foreground.

Abstract:Methyl 3-oxo-2,6,6-trimethylcyclohex-1-ene-1-carboxylate was synthesised over eight steps from the starting material of citral instead of α-ionone. KMnO4, HC(OEt)3 and O2/TEMPO/CuCl were substituted for NaIO4, CH3I and pyridinium chlorochromate, respectively. Every step was simplified and gave a 48% overall yield. The procedure is suitable for large scale production.

Abstract:In this paper, a new angiotensin Ⅱ receptor antagonist, (5-methyl-2-oxo-1,3-dioxol-4-yl) methyl-4-(1-hydroxy-1-methylethyl)-2-propyl-1-[(2′-1H-tetrazol-5-yl-biphenyl-4-yl) methyl] imdazole-5-carboxylate(Olmesartan medoxomil), was synthesized using tartaric acid as starting material via nitration, cyclization, esterifcation, Grignard reaction, N-alkylation, hydrolyzation, O-alkylation and N-deprotection in 32.7% overall yield. The structure of title compound has been characterized by MS(mass spectrum), (()~1H)-NMR (nuclear magnetic resonance) and IR (ifrared spectrum). The condensation of 4-(1-hydroxy-1-methylethyl)-2-propylimidazole-5-carboxylic acid ethyl ester with 5-[4′-(bromomethyl) biphenyl-2-yl]-1-(triphenylmethyl) tetrazole by means of potassium hydroxide in DMF, instead of potassium tert-butoxide in dimethylacetamide or sodium hydroxide in DMF, gives the corresponding N-alkylated product, which is hydrolyzed with sodium hydroxide, instead of lithium hydroxide, in dioxane-water to the desired acid. The improved process is more practical.

Abstract:A highly stereocontrolled synthesis of the C18–C28 ketone fragment of the 16-membered plecomacrolide micromonospolide B has been accomplished. The C21–C23 syn–anti stereotriad is secured by the anti-selective aldol condensation of the ephedrine-derived chiral propionate with (E,E)-hexa-2,4-dienal and Sharpless asymmetric allylic epoxidation–regioselective reductive epoxide ring opening, respectively. The overall yield of this 14-step sequence is 18.4% and the target C18–C28 ketone was obtained in enantiomerically pure form.

Abstract:Ezetimibe is a novel selective cholesterol absorption inhibitor which was developed by both Schering-Plough and Merck. An improved process for the preparation of ezetimibe was studied and was described as follows. N-(4-(benzyloxy)benzylidene)-4-fluorobenzenamine (Compound 2) was prepared from 4-hyd-roxybenzaldehyde, 4-fluorobenzenamine and benzyl chloride in one pot. 5-(4-Fluorophenyl)-5-oxopentanoic acid was reacted with pivaloyl chloride and directly condensed with (S)-4-phenyloxazolidin-2-one promoted with LiCl, and then reduced by CBS/BH3 to get (4S)-3-[(5S)-5-(4-fluoror)-5-hydrox-1-oxo-pentyl]-4-phenyl-oxazolidin-2-one (Compound 4). It was condensed with compound 2 catalyzed with Lewis acid after being protected by TMS to afford the condensation intermediate. Finally it was cyclized by TBAF·3H2O and deprotected with Pd/C to get the title compound with a total yield of 56%. Compared with the current technology, the present process shows advantage in high yield, less raw material consumed, and easy production on industrial scale. The specific rotation and melting point of the prepared title compound is the same as that reported in literature,and the structure of the prepared title compound and some intermediates were confirmed by MS and 1H-NMR.

Abstract:A new practical method for the synthesis of a non-cross-linked polystyrene supported 2-phenylimino-2-oxazolidine chiral auxiliary is reported. This method started from the same material as in a previous synthesis, N-Boc-L-tyrosine ethyl ester, but the overall yield was 42.5%, which was higher than the copolymerisation method.

Abstract:Benzophenone,as a starting material,went through a series of reactions,such as Darzens,methanolysis and hydrolysis to give 2-hydroxy-3-methoxy-3,3-diphenylpropionic acid(5).The latter reacted with L-proline methyl ester hydrochloride(7) and a diasterotropic resolution was carried out to provide the key intermediate(S)-2-hydroxy-3-methoxy-3,3-diphenylpropionic acid(6).Finally,the target product ambrisentan(1) was obtained from 2-methylsulfonyl-4,6-dimethylpyrimidine(8) and 6 via a nucleophilic substitution with an overall yield of 26.5%.

Abstract:An optimized two-step synthesis of 2,5-bis(4-aminophenyl)-1,3,4-oxadiazole has been established.The reaction of 4-nitrobenzoic acid and semicarbazide hydrochloride at 150-160 ℃ for 2 h,using polyphosphoric acid as the dehydrating reagent,afforded 2,5-bis(4-nitrophenyl)-1,3,4-oxadiazole in 89% yield.This was subsequently reduced with Na2S in DMF at 100 ℃ for 4 h,to give the title compound in 85% yield.The as-prepared title compound is sufficiently pure for further uses.

Abstract:(E)-4-bromo-3-methyl-2-butenoic acid was conveniently synthesized from 3-methyl-2-butenoic acid in 29% overall yield.In this procedure,the title compound was easily obtained by hydrolysis of (E)-trimethylsilyl 4-bromo-3-methyl-2-butenoate,which could be isolated from its mixture of Z-/E-isomer by distillation in vacuo.The isomeric mixture was prepared via the esterification of 3-methyl-2-butenoic acid with chlorotrimethylsilane,followed by bromination with NBS.

Abstract:(S)-Atenolol ((S)-2-(4-(2-Hydroxy-3-(isopropylamino)propoxy)phenyl)acetamide) has been synthesized in >99% enantiomeric excess (ee) with the use of Candida antarctica lipase B from Syncozymes (Shanghai, China), in a kinetic resolution of the corresponding racemic chlorohydrin. A catalytic amount of base was used in deprotonation of the phenol building block. The enantiopurity of the chlorohydrin building block remained unchanged upon subsequent amination to yield the final drug. All four steps in the synthesis protocol have been optimized compared to previously reported methods, which makes this new protocol more sustainable and in accordance with green chemistry principles. The overall yield of (S)-atenolol was 9.9%, which will be further optimized.

Abstract:2-(5-Amino-1,2,4-thiadiazol-3-yl)-2-(Z)-methoxyiminoacetic acid 2-benzothiazolyl thioester,an important intermediate of the fourth generation cephalosporins,was efficiently synthesized by reacting 2-(5amino-1,2,4-thiadiazol-3-yl)-2-(Z)-methoxyiminoacetic acid(Ⅰ) with 2,2′-dibenzothiazole disulfide(Ⅱ) in the presence of triphenylphosphine.An improved procedure suitable for industrial production was established.Using 1,2dichloroethane as solvent,triphenylphosphine as reducer and triethylamine as catalyst,n(Ⅰ)∶n(Ⅱ)∶n(triphenylphosphine)=1.0∶1.0∶1.0,the product was obtained at room temperature with a yield of 98.1%.The purity of the product without further purification is 98.7% by HPLC method.This procedure was a suitable alternative to the traditional processes due to easy handling,high yield and low cost.

Abstract:A process is reported for stereoselectively hydrolyzing one enantiomer of rac-3-cyano-2-carboxyethyl-5-methylhexanoic acid ethyl ester for(S)-2-cyano-5-methyl-2-hexanoic acid ethyl ester,an important chiral intermediate of(S)-(+)2-aminomethyl-5-methylhexanoic acid(pregabalin).Our route begins with the Knoevenagel condensation of isovaleraldehyde and diethyl malonate,followed by the addition reaction with sodium cyanide to give the key intermediate Ⅰ.Compound Ⅰ can be stereoselectively hydrolyzed to form(3S)-2-carboxyethyl-3-cyano-5-methylhexanoic acid.GC assay:e.e.p=99%.After synthetic method was improved and optimized,the yields were increased from 78.0% to 88.5% for Ⅰ and from 90.0% to 99.0% for Ⅱ,respectively.

Abstract:Baclofen hydrochloride,an antispastic agent,was synthesized from 4-chlorobenzaldehyde and malonic acid by Knoevenagel-Doebner condensation and esterification to give methyl 3-(4-chlorophenyl) acrylate,which was subjected to Michael addition,hydrolysis,catalytic hydrogenation and then salt formation with an overall yield of about 52%.
