Abstract:Anthraquinone〔1,2-C〕methylisoxazole was prepared from 2-ethyl-1-nitro- anthraquinone through oxidation by oleum. The structure of obtained title compound was characterized by 1H NMR and MS. The optimum synthesis conditions and separation conditions for TLC were given in this paper.

Abstract:3-(3-Chloro-1,2,5-thiadiazol-4-yl)pyridine,which is a key intermediate of selective M1 receptor agonist xanomeline,was synthesized from 3-pyridine carboxaldehyde,sodium cyanide,and sulfur chloride.This procedure has advantage of convenient operations and mild reaction conditions.The yield is improved from 40% to 52% and the process is easy to be popularized in industry.

Abstract:A new method for synthesis of synthesis 6-hydroxy-3, 4-dihydro-2(lH) quinolinone was investigated with higher yield and simpler operation. The title compound was prepared starting from 3-chloropropionyl chloride, via cyclizing, nitration, reduction, diazotization and hydrolysis. The total yield is 67.7%, and the yield of each step is between 86.5% -99.6%. 6-Hydroxy-3, 4-dihydro-2(1H)quinolinone was characterized by MS and 1H NMR.

Abstract:The synthetic method-for 2-(2-bromophenyl)oxazoline was modified, The condensation reaction of N-(beta-hydroxyethyl)-o-bromobenzamide to corresponding oxaozoline is performed by using SOCl2,; not NaH reagent, with a elevated yield of up to 84%, The experimental results show that the reaction temperature has a great influence on the, oxazoline yield, The oxazoline yield drop obviously with,the rising of the reaction temperature and at room temperature hardly any oxazoline product formed, Based on the results obtained, it is presumed that ehe oxazoline ring was formed via an imide tautermer intermediate.

Abstract:15-Cyano-12-oxopentadecano-15-lactone was synthesized in 59% total yield starting from 2-nitrocyclododecanone by Michael addition to acrylaldehyde, followed by reaction with trimethylsilylcyanide, hydrolysis, ring-expansion, and Nef reaction. A two-step, one-pot synthesis of intermediate 2-hydroxy-4-(1-nitro-2-oxycyclododecyl)butanenitrile from 3-(1-nitro-2-oxocyclododecyl)propanal was developed and the conditions for the Nef reaction were studied. 15-Cyano-12-oxopentadecano-15-lactam was synthesized in 40% total yield starting from 2-nitrocyclododecanone by Michael addition to acrylaldehyde, followed by Strecker reaction, ring-expansion, and Nef reaction. The conditions for the Strecker and Nef reactions were studied. The structures of the target compounds, intermediates, and by-product were characterized by 1R, H-1- and C-13-NMR, and elemental analysis or MS.

Abstract:Xanthates are widely used in organic synthesis for the construction of various molecules and have numerous applications as bioactive compounds. Here, we present their preparation via a straightforward photocatalytic decarboxylative xanthylation process involving readily available oxime esters and xanthate dimers. This approach provides access to a wide variety of xanthate compounds in moderate to good yields, including xanthates derived from relevant pharmaceutical and bioactive molecules.

Abstract:An efficient synthetic method for multi-substituted xanthones was developed. The reaction of diaryliodonium salts and salicylates was employed for the preparation of the xanthones. This method proceeded through an intermolecular etherification-acylation to give target heterocycles in good yields (up to 91%). Multi-substituted xanthones were gained by shifting the substituent of salicylates or diaryliodonium salts (C) 2017 Chinese Chemical Society and Institute of Materia Medica, Chinese Academy of Medical Sciences. Abstract:Allylamines have long been recognized as valuable synthons because of their excellent reactivity in organic synthesis. Here, an efficient amination reaction of allenyl ethers via copper/Lewis acid synergistic catalysis has been established, providing straightforward access to diverse functionalized Z-oxalylamines and E -halogenated oxalylamines in good to excellent yields with high regio- and stereoselectivities. The developed method tolerates more than 100 examples that include late-stage functionalization of bioactive molecules, and features gram-scale synthesis of oxalylamines with high turnover number (TON > 1000) under mild and simple conditions. The applicability of the protocol is further demonstrated with the construction of drug molecules.

Abstract:A series of xanthones with the dibenzo-γ-pyrone framework were synthesized through the palladium-catalyzed carbonylation/C–H activation sequence from the ortho diazonium salts of diaryl ethers in moderate to excellent yields. After screening the reaction condition, the optimal condition was 5mol% tetrakis (triphenylphosphine) palladium (0) as the catalyst, potassium carbonate as the base, and toluene as the solvent in the presence of catalytic amount of tetrabutyl ammonium bromide under carbon monoxide atmosphere.

Abstract:Aim To synthesize xanthotoxin in a simple way.Methods Xanthotoxin was synthesized from parogallol through nine steps with PCC used.The key compound in this synthesis is 2-chloro-2′-hydroxy-3′,4′-dimethoxy-5′-chloromethyl acetophenone,which was obtained from 2-chloro-2′-hydroxy-3′,4′-dimethoxyacetophenone through chloromethylation.Oxidation of 6,7-dimethoxy-5-hydromethylbenzofuran with the selective oxidant PCC yielded the 6,7-dimethoxy-5-formyl benzofuran.After Knoevenagel reaction of 6-hydroxy-7-methoxy-5-formyl benzofuran with malonic acid and decarboxylation,xanthotoxin is obtained.Its chemical structure was identified with ~1H-NMR,MS and IR.Results The structure was determined by spectrum.The overall yield was 9.0% and its purity was more than 99%.Conclusions Xanthotoxin can be synthesized in a practical way.The cost of the total synthesis is lower than the former one and the yield is definitely increased.

Abstract:A highly efficient ruthenium-catalyzed stereospecific N-demethylative rearrangement of isoxazolidines to synthetically useful N-H-1,3-oxazinanes is described. 1,3-Oxazinanes are useful building blocks, which can be further converted to N-H-1,3-aminoalcohols in one step. This new method was used in a three-step gram-scale total synthesis of HPA-12 in an overall 24% yield.

Abstract:An optimized two-step synthesis of 2,5-bis(4-aminophenyl)-1,3,4-oxadiazole has been established.The reaction of 4-nitrobenzoic acid and semicarbazide hydrochloride at 150-160 ℃ for 2 h,using polyphosphoric acid as the dehydrating reagent,afforded 2,5-bis(4-nitrophenyl)-1,3,4-oxadiazole in 89% yield.This was subsequently reduced with Na2S in DMF at 100 ℃ for 4 h,to give the title compound in 85% yield.The as-prepared title compound is sufficiently pure for further uses.

Abstract:Oxasulfuron was synthesized by the reaction of 2-(oxetan-3-oxycarbonyl) -phenylsulfonamide(3) with phenyl N-(4,6-dimethylpyrimidin-2-yl) carbamate(6) . Using 2-chlorosulfonylbenzoyl chloride,3-hydroxyoxetane and ammonia as starting materials,compound(3) was conveniently obtained. Meantime,amidation of phenyl chlorocarbonate afforded the intermediate(6) ,while triethylamine was selected as a more efficient deacid reagent. The total yield was 41%,and the purity of the target product was up to 97.4%.

Abstract:A facile solid phase synthesis of 2-substituted 1,3-oxazin-6-ones using polymer-supported Meldrum's acid has been reported. Reaction of the resin-bound cyclic malonic acid ester with triethyl orthoformate and subsequent double substitution with amide, afforded the corresponding polymer-supported acylaminomethylene cyclic malonic acid ester, which upon thermal treatment led to 1, 3-oxazin-6-ones in good yields and with high purity.

Abstract:Phenoxy acetophenones were usually employed as β-O-4' lignin models for chemical conversion. Herein, an iridium-catalyzed dehydrogenative annulation between 2-aminobenzylalcohols and phenoxy acetophenones was demonstrated to prepare valuable 3-oxo quinoline derivatives, which are hard to prepare using previous methods. This operationally simple reaction tolerated a wide scope of substrates and enabled successful gram-scale preparation.

Abstract:N-Substituted-5-aryl-1,3,4-oxadiazole-2-amine was synthesized by a three-step reaction in overall yields of 42.0%~68.7%.The products were characterized by IR,~1H NMR,MS and elemental analysis.Furthermore,the single crystal X-ray study of 5-phenyl-1,3,4-oxadiazole-2-amine(4a) indicated that this compound belonged to 1,3,4-oxadiazole derivatives,not to 1,4-dihydro-1,2,4,5-tetrazine derivatives.
