Abstract:Single-nanoparticle detection has received tremendous interest due to its significance in fundamental physics and biological applications. Here, we demonstrate an optical nanofibre-enabled microfluidic sensor for the detection and sizing of nanoparticles. Benefitting from the strong evanescent field outside the nanofibre, a nanoparticle close to the nanofibre can scatter a portion of the field energy to the environment, resulting in a decrease in the transmitted intensity of the nanofibre. On the other hand, the narrow and shallow microfluidic channel provides a femtoliter-scale detection region, making nanoparticles flow through the detection region one by one. By real-time monitoring of the transmitted intensity of the nanofibre, the detection of a single polystyrene (PS) nanoparticle as small as 100 nm in diameter and exosomes in solution is realised. Based on a statistical analysis, the mean scattering signal is related to the size of the nanoparticle. Experimentally, a mixture of nanoparticles of different diameters (200, 500, and 1000 nm) in solution is identified. To demonstrate its potential in biological applications, high-throughput counting of yeasts using a pair of microchannels and dual-wavelength detection of fluorescently labelled nanoparticles are realised. We believe that the developed nanoparticle sensor holds great potential for the multiplexed and rapid sensing of diverse viruses. Abstract:In this paper, a novel method is presented for creating a 3-dimensinal sub-diffraction effective observation volume based on microsphere photonic nanojet and fluorescence depletion effect. Using microsphere to achieve photonic nanojet focusing effect, the proposed method applies the radially polarized plane wave to excite fluorescence and the azimuthally polarized beam to obtain fluorescence depletion field. The effective detection volume of photonic nanojet can reach 0.002μm3 (2aL). Compared with conventional confocal microscopy, this effective detection volume represents a reduction of almost 2 orders of magnitude. With simple configuration based on cost-effective microspheres, the proposed method is theoretically proved to be a potential tool for the fluorescence correlation spectroscopy (FCS) to have large analysis range and to investigate single molecule at high concentrations.

Abstract:Achieving nanometer-scale resolution remains challenging in expansion microscopy due to photon loss. To address this concern, here we develop a multi-color expansion stimulated emission depletion technique based on small-molecule probes to realize high labeling density and intensity. Our method substantially lowers the barrier to visualizing diverse intracellular proteins and their interactions in three dimensions. It enables us to achieve sub-10-nm resolution in structures such as microfilaments, lysosomes, and mitochondria, providing new insights into cell biology.

Abstract:The orientation of a single molecule provides valuable intimation on fundamental biological processes. We report a technique for the simultaneous estimation of single-molecule 2D position and 2D orientation with ultra-high localization precision (similar to 2-nm precision with similar to 500 photons under a typical 100-nm diameter of excitation beam pattern), which is also compatible with tracking in living cells. In the proposed method, the theoretical precision limits are calculated, and the localization and orientation performance along with potential applications are explored using numerical simulations. Compared to other camera-based orientation measurement methods, it is confirmed that the proposed method can obtain reasonable estimates even under very weak signals (similar to 15 photons). Moreover, the maximum likelihood estimator (MLE) is found to converge to the theoretical limit when the total number of photons is less than 100. Abstract:We experimentally demonstrated that the microsphere can discern the details of the object whose sizes are below the conventional diffractive limit and such super-resolution capability can be reinforced if semi-immersing the corresponding microspheres in liquid droplet, producing a sharper contrast and a comparatively smaller magnification factor. The microsphere is considered as a channel that connects the near-field evanescent wave and the transmission one in far field. A conjecture based on this is proposed to explain the mechanism of super-resolution and the corresponding phenomenon.

Abstract:Whispering gallery mode based optical microcavities are important for highly sensitive optical sensing. However, the current experimental realizations are strongly dependent on high-resolution tunable lasers and evanescent coupling, which are too cumbersome and too expensive for portable devices. Herein we experimentally demonstrate a cost-effective and robust approach to detect and size a single nanoparticle with far-field laser emissions. By placing a limacon microdisk close to a spiral microdisk, chiral resonances have been successfully generated. In contrast to previous research, here the internal chirality is strongly correlated with the far-field patterns (FFPs) and thus can be transduced to far-field emissions. Once a nanoparticle is attached to the limacon microdisk, the asymmetrical backscattering at the notch of the spiral can be averaged by the symmetrical scattering of the nanoparticle. Consequently, the internal chirality and the corresponding FFPs are changed. By measuring a far-field directional laser emission, nanoparticles with a radius of similar to 50 nm have been successfully detected and sized without employing any spectral information. As a narrow-linewidth tunable laser is not used in our experiment and microdisks lasers may be electrically driven, this research will provide a new path to cost-effective, portable, highly sensitive optical sensors. Abstract:Single-nanoparticle detection and sizing is demonstrated using a nanofiber pair in an aqueous environment. The sizing of nanoparticles with a single radius (100 nm) and of mixed nanoparticles with different radii (100 nm and 170 nm) are both realized, and the experimental results agree well with predictions of Rayleigh-Gans scattering, by taking the inhomogeneous field distribution of the nanofibers into account.

Abstract:Fluorescence sensing and fluorescence correlation spectroscopy (FCS) are powerful methods to detect and characterize single molecules; yet, their use has been restricted by expensive and complex optical apparatus. Here, we present a simple integrated design using a self-assembled bi-dimensional array of microspheres to realize multi-focus parallel detection scheme for FCS. We simultaneously illuminate and collect the fluorescence from several tens of microspheres, which all generate their own photonic nanojet to efficiently excite the molecules and collect the fluorescence emission. Each photonic nanojet contributes to the global detection volume, reaching FCS detection volumes of several tens of femtoliters while preserving the fluorescence excitation and collection efficiencies. The microspheres photonic nanojets array enables FCS experiments at low picomolar concentrations with a drastic reduction in apparatus cost and alignment constraints, ideal for microfluidic chip integration.

Abstract:Detection of nanoscale objects is highly desirable in various fields such as early-stage disease diagnosis, environmental monitoring and homeland security. Optical microcavity sensors are renowned for ultrahigh sensitivities due to strongly enhanced light-matter interaction. This review focuses on single nanoparticle detection using optical whispering gallery microcavities and photonic crystal microcavities, both of which have been developing rapidly over the past few years. The reactive and dissipative sensing methods, characterized by light-analyte interactions, are explained explicitly. The sensitivity and the detection limit are essentially determined by the cavity properties, and are limited by the various noise sources in the measurements. On the one hand, recent advances include significant sensitivity enhancement using techniques to construct novel microcavity structures with reduced mode volumes, to localize the mode field, or to introduce optical gain. On the other hand, researchers attempt to lower the detection limit by improving the spectral resolution, which can be implemented by suppressing the experimental noises. We also review the methods of achieving a better temporal resolution by employing mode locking techniques or cavity ring up spectroscopy. In conclusion, outlooks on the possible ways to implement microcavity-based sensing devices and potential applications are provided.

Abstract:Sizing individual nanoparticles and dispersions of nanoparticles provides invaluable information in applications such as nanomaterial synthesis, air and water quality monitoring, virology, and medical diagnostics. Several conventional nanoparticle sizing approaches exist; however, there remains a lack of high-throughput approaches that are suitable for low-resource and field settings, i.e., methods that are cost-effective, portable, and can measure widely varying particle sizes and concentrations. Here we fill this gap using an unconventional approach that combines holographic on-chip microscopy with vapor-condensed nanolens self-assembly inside a cost-effective hand-held device. By using this approach and capturing time-resolved in situ images of the particles, we optimize the nanolens formation process, resulting in significant signal enhancement for the label-free detection and sizing of individual deeply subwavelength particles (smaller than λ/10) over a 30 mm(2) sample field-of-view, with an accuracy of ±11 nm. These time-resolved measurements are significantly more reliable than a single measurement at a given time, which was previously used only for nanoparticle detection without sizing. We experimentally demonstrate the sizing of individual nanoparticles as well as viruses, monodisperse samples, and complex polydisperse mixtures, where the sample concentrations can span ∼5 orders-of-magnitude and particle sizes can range from 40 nm to millimeter-scale. We believe that this high-throughput and label-free nanoparticle sizing platform, together with its cost-effective and hand-held interface, will make highly advanced nanoscopic measurements readily accessible to researchers in developing countries and even to citizen-scientists, and might especially be valuable for environmental and biomedical applications as well as for higher education and training programs.

Abstract:Single nanoparticle detection is of critical importance in many applications. Optical microcavities featuring high-Q factors and small mode volumes have been widely investigated in sensing applications. In general, the microcavity sensing depends mainly on reactive (i.e., dispersive) interactions, resulting in a resonance wavelength shift or mode splitting, which essentially responds to the real part of the polarizability of the targets. In the first part of this talk, we report the experimental demonstration of single nanoparticle detection using either resonance mode broadening or microcavity Raman laser splitting. In the second part, a dissipative sensing method is demonstrated to detect single lossy nanoparticles. This dissipative sensing method holds great potential in detecting nanoparticles of high absorption or ultralow polarizabilities, such as carbon nanotubes and metal nanoparticles, and in characterizing nanoparticle properties in combination with the reactive sensing method.
Fast Size Estimation of Single-Levitated Nanoparticles in a Vacuum Optomechanical System.

Abstract:Optical trapping of single nanoparticles in vacuum has various applications in both precise measurements and fundamental physics. However, to date, the number and size of randomly loaded nanoparticles in an optical trap is difficult to determine unless in vacuum. In this Letter, an efficient method for nanoparticle size estimation in an optical tweezer system before the evacuation of air was proposed and demonstrated experimentally, using scattering light from levitated particles. The particle radii deduced from the scattering light power in our proposal and from the kinetic theory of particles in gas match well (with the differences of less than 10 % ). For sample particles with radii ranging within 50–100 nm, we also provide a preselection rule based on this method, where over half of the trapped particles are verified as single particles. Such a particle analysis method is applicable also for the size estimation of levitated diamond particles, gold particles, and other plasmonic particles and can be applied to discovering novel scattering effects.

Abstract:We demonstrate an optical micro-particle size detection technique based on phase sensing by a fiber interferometer through phase-generated carrier (PGC) modulation/demodulation. Particle diameters were resolved from phase shift due to particle-induced optical scattering. Polystyrene nanoparticles, air bubbles and yeast cells in a microfluidic channel were tested using this technique, and particle diameters ranging from 0.7 to 5.5 μm can be resolved in real-time. In comparison with existing amplitude-sensing techniques which require tens of milliwatts of laser irradiance, phase-sensing through PGC can successfully utilize probe laser powers as low as 220 μW to measure the test particle sizes. We further constructed a theoretical model based on phase scattering and PGC demodulation, which obtained good agreement between experimental data and calculated phase shift as a function of particle time-of-flight. This technique may be applied to a wide range of potential applications, ranging from real-time analysis of clinically relevant cell samples, to contamination control of processing fluids used in the semiconductor industry.

Abstract:Micro-nano particles, indispensable components in various industrial and scientific applications, exhibit size-dependent properties. Accurate granular information is critical for manufacturing products involving these particles. This study presents a novel detection method that merges image and laser modes to achieve precise size and shape determination of micro-nano particles. The proposed method integrates adaptive image analysis with laser particle size fitting to ensure accurate feature detection. Experimental results using polystyrene samples demonstrate that the fusion mode outperforms both the laser and image modes in terms of size and shape accuracy. The errors of D10, D50, and D90 of the fusion mode are 3.9%, 1%, and 11.36%, respectively, which are significantly lower than those obtained using the laser or image mode alone. Additionally, the fusion mode retains the particle shape information from the image mode. The fusion model offers a promising approach for dual-mode detection of micro-nano particles, demonstrating enhanced robustness compared to single-mode methods. Furthermore, the study introduces an image processing method based on a BP neural network classification model that accurately classifies and detects agglomerated and non-agglomerated particles. The classification accuracy reaches 94.98%, and the particle size information remains within the acceptable range.

Abstract:In recent years, Janus microspheres technology has been widely applied and developed in various fields. However, existing studies often rely on electron microscopy images, treating Janus microspheres as spherical to observe their submicron structures. This approach falls short in meeting the demand for precise measurement of the specific position and orientation of microspheres at the single-molecule scale. Here, we propose a standardized processing procedure that integrates multiple image processing algorithms to measure the position of Janus microspheres through the image capture. Our approach enhances the two-dimensional planar positioning accuracy of non-spherical Janus microspheres in micro-nano optical measurement systems. Utilizing this method, we can conduct a further research on the structure and motion characteristics of the microsphere at the single-particle scale. This method has been validated on a transmission illumination optical tweezers platforms, successfully calibrating the central position of the optical trap. Our work offers a greater convenience for measuring non-spherical Janus microspheres in liquid environments. It also provides a fresh perspective for subsequent studies on the motion trajectories and force dynamics of heterogeneous microspheres, showcasing its broad potential for application.

Abstract:The ability to detect and size individual nanoparticles with high resolution is crucial to understanding the behaviour of single particles and effectively using their strong size-dependent properties to develop innovative products. We report real-time, in situ detection and sizing of single nanoparticles, down to 30 nm in radius, using mode splitting in a monolithic ultrahigh-quality-factor ( Q ) whispering-gallery-mode microresonator. Particle binding splits a whispering-gallery mode into two spectrally shifted resonance modes, forming a self-referenced detection scheme. This technique provides superior noise suppression and enables the extraction of accurate particle size information with a single-shot measurement in a microscale device. Our method requires neither labelling of the particles nor a priori information on their presence in the medium, providing an effective platform to study nanoparticles at single-particle resolution.

Abstract:Accurate determination of nanoparticle size is paramount in various fields, including molecular imaging, food safety, drug delivery, and nanobiotechnology. Conventional methods face limitations in accurately resolving different size distributions. Here, we introduce an approach to measure size distribution by combining photometry and tracking analysis of single particles. Our approach is based on a plasmonic dark field imaging system to image and track individual nanoparticles. Scattering intensity, spot size, and diffusion coefficient are quantified and fed into the machine learning model to establish the relationship with particle sizes. Compared with conventional methods, our approach shows improved accuracy in size measurement. To verify the universality of our method, we further demonstrate the sizing capability at diverse and complex conditions, enabling us to discriminate particle size differences within 20 nm in polydisperse systems.
