Abstract:In this study, we have described a facile process for fabrication of multifunctional composite hydrogel, in which sodium alginate was subjected to cross-linking using Ca2+ derived from ZnO/CaCO₃/Ag composite nanospheres. The ZnO/CaCO₃/Ag composite nanospheres were prepared based on our previously reported AA-[Zn(OH)₄]2- composite nanosphere reaction conducted with silver and calcium salt following hydrothermal method, that led to the disintegration and release of Ca2+ under acidic conditions for application as a cross-linking agentto catalyze reaction with sodium alginate. Ag nanoparticles were well-dispersed in the multifunctional composite hydrogel, exhibiting excellent antibacterial activity. Additionally, polydopamine (PDA) with photothermal effect was also added to obtain a multifunctional composite hydrogel, and this hydrogel showed photothermal conversion performance and facilitated the release of Ag+ to achieve the rapid antibacterial effect. Simultaneously, PDA NPs could scavenge free radicals and improve cell adhesion. All such features would promote wound healing. The potent antimicrobial activity of the prepared composite hydrogel was demonstrated in the mouse model of S. aureus infection, and biosafety of the hydrogel was confirmed by conducting histopathological examination in the mouse model. This type of multifunctional hydrogel wound dressing with photosensitive and antibacterial properties presents with broad applications and prospects in antibacterial treatment.

Abstract:Hydrogels are materials consisting of a network of hydrophilic polymers. Due to their good biocompatibility and hydrophilicity, they are widely used in biomedicine, food safety, environmental protection, agriculture, and other fields. This paper summarizes the typical complex materials of photocatalysts, photosensitizers, and hydrogels, as week as their antibacterial activities and the basic mechanisms of photothermal and photodynamic effects. In addition, the application of hydrogel-based photoresponsive materials in microbial inactivation is discussed, including the challenges faced in their application. The advantages of photosensitive antibacterial complex hydrogels are highlighted, and their application and research progress in various fields are introduced in detail.

Abstract:Antibacterial hydrogels have attracted significant attention due to their diverse applications, efficient antimicrobial properties, and adaptability to various environments and requirements. However, their relatively fragile structure, coupled with the potential for environmental toxicity when exposed to their surroundings for extended periods, may significantly limit their practical application potential. In this work, a composite hydrogel was synthesized with outstanding mechanical features and antibacterial capability. The hydrogel was developed through the combination of the eco-friendly and enduring antibacterial agent, lignin silver nanoparticles (Lig-Ag NPs), with polyvinyl alcohol (PVA) and sodium alginate (SA), in varying proportions. The successful synthesis of the hydrogel and the dispersed distribution of Lig-Ag NPs within the hydrogel were confirmed by various analytical techniques, including field emission scanning electron microscopy (SEM), energy-dispersive spectroscopy (EDS), mercury intrusion porosimetry (MIP), Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD), and X-ray photoelectron spectroscopy (XPS). The formation of multiple hydrogen bonds between Lig-Ag NPs and the composites contributed to a more stable and dense network structure of the hydrogel, consequently enhancing its mechanical properties. Rheological tests revealed that the hydrogel exhibited an elastic response and demonstrated outstanding self-recovery properties. Significantly, the antibacterial hydrogel demonstrated effectiveness against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus), achieving a <5% survival of bacteria within 12 h. This study presented a green and straightforward synthetic strategy for the application of antibacterial composite hydrogels in various fields.

Abstract:Antibacterial hydrogels have gained significant attention as appealing materials, but their weak structures largely limit their practical applications. In this study, chitosan (CS)-based double-network antibacterial hydrogels were developed, where polyacrylamide (pAAm) attributed to the high mechanical property and [2-(methacryloyloxy)ethyl]trimethylammonium chloride (MTAC) exerted the strong antibacterial activity, structured as CS/p(AAm-MATC). The structure and morphology of hydrogels with a ratio of pAAm and MTAC of 5:5 were preferred and analyzed using scanning electron microscopy, Fourier transform infrared spectroscopy, solid-state NMR spectroscopy, and X-ray diffraction, confirming the successful synthesis. The hydrogels had remarkable compression resistance, withstanding a high strain of 85% with excellent shape recovery. Rheological tests revealed that the samples exhibited characteristic behaviors of hydrogels, with the storage modulus surpassing the loss modulus and increasing with the angular frequency. Furthermore, the composite hydrogels had excellent antibacterial efficacy against Gram-positive (Listeria monocytogenes) and Gram-negative (Escherichia coli) bacteria, mainly attributed to the presence of quaternary ammonium groups in MTAC polymers. These hydrogels, with outstanding mechanical and antibacterial properties, hold promising potential for diverse applications, such as wastewater treatment.Graphical AbstractA tough double-network hydrogel is created by a combination of physical and chemical crosslinking methods, exerting high mechanical and enhanced antibacterial properties.

Abstract:Hydrogel materials are the preferred candidates for antimicrobial materials due to their high water absorption, superior biocompatibility and structural diversity. In this work, we present for the first time that diamine-alkyl and 1,3-dibromo-2-propanol can be rapidly cross-linked to prepare a series of new cationic polymer gels by polycondensation reactions under a strongly alkaline environment. The presence of C−N+ bonds are demonstrated in the formation of dendritic structured polymers by XPS analysis. In addition, the physicochemical properties of b-PHIs hydrogels were evaluated and it was found that the variation of diamine alkyl chain length could modulate the storage modulus, water uptake properties, thermal stability and zeta-potential of the gels. The increase in hydroxyl content and the C−N+/C-C ratio is evidenced by the elevated water absorption properties of the gels and the variations in zeta potential, respectively. In addition, the antibacterial performance evaluation showed that gels had good antibacterial activity against S. aureus and E. coli, with antimicrobial rates of b-PHOI as high as 96% and 97%, respectively. This work pioneers a brand-new idea for the simple and rapid synthesis of branched cationic polymers with low cytotoxicity in the field of antimicrobial biomaterials and proceeds a new attempt in the directional modulation of the physical properties of the materials.

Abstract:Multifunctional materials have received considerable attention as they could integrate different functional components in one‐single platform. In this study, novel chitosan/Fe3O4/TiO2@TiO2 nanowire microspheres having extracellular matrix‐like fibrous surface and photothermal antibacterial property were synthesized through in situ hydrothermal growth of TiO2 nanowires on chitosan/Fe3O4/TiO2 microspheres. It is found that the microspheres were spherical in morphology with a diameter of 100–300 μm and exhibited a hierarchical and nanofibrous feature. Their surface was mainly constructed by numerous TiO2 nanowires with a diameter of 10–20 nm. In vitro biological evaluation indicates that the chitosan/Fe3O4/TiO2@TiO2 nanowire microspheres significantly enhanced attachment and proliferation of human dermal microvascular endothelial cells compared with chitosan/Fe3O4/TiO2 nanocomposite microspheres due to the presence of nanofibrous surface. Moreover, the microspheres showed photo‐thermal antibacterial property to inhibit the growth of bacteria due to the presence of Fe3O4 component. This article is protected by copyright. Abstract:Bacterial infections are a great threat to public health. The development of materials which prevent biofilm formation, and exhibit antibacterial properties, but are otherwise biocompatible, is a major research goal. In this study, a novel hybrid hydrogel with aggregation-induced emission (AIE) behavior was prepared through the self-assembly of commercial N-(9-fluorenylmethoxycarbonyl)-L-phenylalanine (Fmoc-F) and berberine chloride (BBR). BBR self-assembled into nanoparticles which dispersed throughout the gel network, and endowed the hydrogel with AIE functionality. Fmoc-F/BBR hydrogel displayed enhanced antibacterial and anti-biofilm activity against both Gram-negative (Escherichia coli) and Gram-positive (Staphylococcus aureus) bacteria when used as the active agent in the photodynamic therapy (PDT) process. Additionally, negligible cytotoxicity and low hemolytic activity were displayed. In vivo infection studies showed that Fmoc-F/BBR hydrogel facilitated the repair of wounds in rats infected with S. aureus. Therefore, Fmoc-F/BBR hydrogel provides potential applications in biomedicines, and especially in wound healing.

Abstract:A series of antibacterial hydrogels were fabricated from an aqueous solution of AgNO3, gelatin and carboxymethyl chitosan (CM-chitosan) by radiation-induced reduction and crosslinking at ambient temperature. The nanosilver particles were in situ synthesized accompanying with the formation of gelatin/CM-chitosan hydrogel. Transmission Electron Microscope and UV–vis analysis have verified the formation and homogeneous distribution of nanosilver particles in the hydrogel matrix. The nanosilver/gelatin/CM-chitosan hydrogels possessed interconnected porous structure, had a compressive modulus of 44 to 56kPa, and could absorb 62 to 108 times of deionized water to its dry weight. Furthermore, the hydrogels were found to have sound antibacterial effect on Escherichia coli (E. coli), and their antibacterial ability could be significantly enhanced by the increasing of AgNO3 content. The comprehensive results of this study suggest that nanosilver/gelatin/CM-chitosan hydrogels have potential as an antibacterial wound dressing.

Abstract:A polyoxometalates‐based hydrogel loaded with copper sulfide nanoparticles is synthesized for antimicrobial treatment of wound surfaces. The POM‐based hydrogel is not only well biocompatible but also efficient in its antimicrobial properties by synergizing with low‐temperature photothermal therapy and chemodynamic therapy. This research can provide valuable insights into the development of non‐antibiotic materials. Bacterial infection of wound surfaces has posed a significant threat to human health and represents a formidable challenge in the clinical treatment. In this study, a novel antimicrobial hydrogel utilizing POM is synthesized as the primary component, with gelatin and sodium alginate as the structural framework. The resultant hydrogel demonstrates exceptional mechanical properties and viscoelasticity attributed to the hydrogen‐bonded cross‐linking between POM and gelatin, as well as the ionic cross‐linking between sodium alginate and Ca2+. In addition, the integration of CuS nanoparticles conferred photothermal properties to the hydrogel system. To address the concerns regarding the potential thermal damage to the surrounding normal cells, this study employs a LT‐PTT combined with CDT approach to achieve the enhanced antimicrobial efficacy while minimizing the inadvertent harm to the healthy cells. The findings suggested that POM‐based hydrogels, serving as an inorganic–organic hybrid material, will represent a promising antimicrobial solution and offer valuable insights for the development of the non‐antibiotic materials.

Abstract:Hydrogels have received considerable attention due to their biocompatibility and desirable physical characteristics. Nonetheless, due to their open structure, hydrogels are prone to dehydration in air, resulting in a loss in their elasticity and function. Herein, we report a facile yet effective method for the modification of hydrophobic hydrogel surfaces by using bioinspired amphiphilic Janus silica particles, which are obtained by modifying hydrophilic polydopamine and hydrophobic 1H,1H,2H,2H-perfluorodecanthiol on their two sides via a templating method. With the coating of amphiphilic Janus silica particles, the water contact angles of poly(ethylene imine)-polyacrylamide and polydopamine-polyacrylamide hydrogels significantly increase to 96° and 97°, respectively. Furthermore, we demonstrate that the hydrophobic modification of the hydrogels by Janus silica particles improves the water retention capability, and the overall mechanical properties of bulk hydrogels are not compromised. In addition, we show that hydrogels coated with Janus silica particles not only exhibit hydrophobic surfaces but also have photothermal antibacterial capabilities. Consequently, this study provides a facile method for the fabrication of hydrogels with hydrophobic surfaces, which could potentially be applied to biomedical materials.

Injectable Photothermal PDA/Chitosan/β‐Glycerophosphate Thermosensitive Hydrogels for Antibacterial and Wound Healing Promotion

Abstract:Controlling infections while reducing the use of antibiotics is what doctors as well as researchers are looking for. As innovative smart materials, photothermal materials can achieve localized heating under light excitation for broad‐spectrum bacterial inhibition. A polydopamine/chitosan/β‐glycerophosphate (PDA/CS/β‐GP) temperature‐sensitive hydrogel with excellent antibacterial ability was synthesized in the study. Initially, the hydrogel has good biocompatibility. In vitro experiments reveal its non‐cytotoxic property when co‐cultured with gingival fibroblasts and non‐hemolytic capability. Concurrently, the in vivo biocompatibility is confirmed through liver and kidney blood markers and staining of key organs. Crucially, the hydrogel has excellent photothermal conversion performance, which can realize the photothermal conversion of hydrogel up to 3 mm thickness. When excited by near‐infrared light, localized heating is attainable, resulting in clear inhibition impacts on both Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli), with the inhibition rates of 91.22% and 96.69%, respectively. During studies on mice's infected wounds, it is observed that the hydrogel can decrease S. aureus' presence in the affected area when exposed to near‐infrared light, and also lessen initial inflammation and apoptosis, hastening tissue healing. These findings provide valuable insights into the design of antibiotic‐free novel biomaterials with good potential for clinical applications. This article is protected by copyright. Abstract:Supramolecular hydrogels serve as an excellent platform to enable in-situ reactive oxygen species (ROS) generation while maintaining controlled localized conditions thereby mitigating cytotoxicity. Herein, we demonstrate hydrogel formation using guanosine-5'-monophosphate (GMP) with tetra(4-carboxylphenyl) ethylene (1) to exhibit aggregation induced emission (AIE) and tunable mechanical strength in the presence of divalent metal ions such as Ca2+, Mg2+, Fe2+. Addition of divalent metal ions leads to the structural transformation in the metallogels, M-1GMP. Further, the incorporation of Fe2+ ions into the hydrogel (Fe-1GMP) promotes Fenton reaction that could be upregulated upon adding ascorbic acid (AA) demonstrating antibacterial efficacy via the ROS generation. In vitro studies with AA loaded Fe-1GMP demonstrate excellent bacterial killing efficacy against E. coli, S. aureus and vancomycin-resistant enterococci (VRE) strains. Finally, in vivo studies involving topical administration of Fe-1GMP on Balb/c mice with skin infections further suggest the potential antibacterial efficacy of the hydrogel. Taken together, the hydrogel with its unique combination of mechanical tunability, ROS generation capability and antibacterial efficacy, can be used for biomedical applications, particularly in wound healing and infection control.

Abstract:Bacterial infectious diseases and bacterial-infected environments have been threatening the health of human beings all over the world. In view of the increased bacteria resistance caused by overuse or improper use of antibiotics, antibacterial biomaterials are developed as the substitutes for antibiotics in some cases. Among them, antibacterial hydrogels are attracting more and more attention due to easy preparation process and diversity of structures by changing their chemical cross-linkers via covalent bonds or noncovalent physical interactions, which can endow them with various specific functions such as high toughness and stretchability, injectability, self-healing, tissue adhesiveness and rapid hemostasis, easy loading and controlled drug release, superior biocompatibility and antioxidation as well as good conductivity. In this review, the recent progress of antibacterial hydrogel including the fabrication methodologies, interior structures, performances, antibacterial mechanisms, and applications of various antibacterial hydrogels is summarized. According to the bacteria-killing modes of hydrogels, several representative hydrogels such as silver nanoparticles-based hydrogel, photoresponsive hydrogel including photothermal and photocatalytic, self-bacteria-killing hydrogel such as inherent antibacterial peptides and cationic polymers, and antibiotics-loading hydrogel are focused on. Furthermore, current challenges of antibacterial hydrogels are discussed and future perspectives in this field are also proposed.
Synergistic photothermal enhancement of the antibacterial activity of fibroin hydrogel by dual-directional crosslink strategy for efficient solar steam generation

Abstract:Silk fibroin (SF) has attracted wide attention due to its excellent biocompatibility and versatile adjustability. The regeneration and reutilization of silk fibroin can efficiently utilize waste cocoons and silk. However, the regenerated SF hydrogels suffer from inadequate mechanical properties and lack inherent antibacterial capabilities, limiting their application in seawater desalination. This study achieved a novel hydrogel (STS@Fe) with photothermal antimicrobial activity, good mechanical property, and excellent durability using a green dual-directional crosslink and meso-reconstruction strategy. The metal-phenolic networks (MPN) were synthesized utilizing the natural plant polyphenol tannic acid (TA) and Fe 3 O 4 nanoparticles. This research incorporated MPN into dissolved and regenerated silk fibroin and sodium alginate solution, enhanced mechanical properties through cryogenic salt precipitation, and prepared STS@Fe hydrogel. The inherent photothermal properties of Fe 3 O 4 nanoparticles and TA complex, coupled with their synergistic effect under near-infrared radiation, can confer excellent photothermal-enhanced antibacterial activity to the fibroin hydrogel. The tensile strength of STS@Fe hydrogel is enhanced 31 times than that of traditional fibroin hydrogel, and its evaporation rate is 4.77 times higher than that of pure water evaporation. The hydrogel has an efficient seawater desalination rate and outstanding antibacterial properties in real seawater conditions. Moreover, The STS@Fe shows effective treatment capabilities for dyeing wastewater and oily saline water.

Abstract:After skin tissue trauma, wound infections caused by bacteria posed a great threat to skin repair. However, resistance to antibiotics, the current treatment of choice for bacterial infections, greatly affected the efficiency of anti-infection and wound healing. Therefore, there has been a critical need for the development of novel antimicrobial materials and advanced therapeutic methods to aid in skin repair. In this paper, rGO-PDA@ZIF-8 nanofillers were prepared by coating graphene oxide (GO) with dopamine (DA), followed by in situ growth of zeolite imidazolate framework-8 (ZIF-8). Using polyvinyl alcohol (PVA) and chitosan quaternary ammonium salt (CS) as matrix materials, along with polyethylene glycol (PEG) as a pore-forming agent, and rGO-PDA@ZIF-8 as an antimicrobial nano-filler, we successfully prepared rGO-PDA@ZIF-8/PVA/CS composite hydrogels with a directional macroporous structure using bidirectional freezing method and phase separation technique. This hydrogel exhibited excellent mechanical properties, good solubility and water retention capabilities. In addition, the hydrogel demonstrated excellent biocompatibility. Most notably, it not only exhibited excellent bactericidal effect against E. coli and S. aureus (99.1 % and 99.0 %, respectively) under the synergistic effect of intrinsic antibacterial activity and photothermal antibacterial, but also exhibited the ability to promote wound healing, making it a promising candidate for wound healing applications.
Polydopamine-intercalated montmorillonite to mitigate polymerization inhibition for preparation of photothermal antibacterial and hemostatic hydrogel

Abstract:The mechanical properties of hydrogels with in situ introduced polydopamine (PDA) will always be unsatisfactory owing to the polymerization inhibition of the catechol structure. Therefore, it is necessary to find a way to mitigate the inhibition. In this work, PDA-inserted montmorillonite (MMT) was first prepared. The micro-spacing of the MMT nanolayers limited the oxygen content which prevented the complete oxidation of the dopamine adsorbed between the MMT nanosheet layers and retained the catechol moiety. Then, polydopamine montmorillonite polyacrylamide (PMP) hydrogel was prepared under green LED irradiation. The hydrogel exhibited excellent mechanical properties and adhesive strength, as well as excellent photothermal properties and antimicrobial ability. After irradiation by NIR laser, the PMP hydrogel can be heated up and maintained at 50 °C within 2 min, and the antimicrobial rate reached 98 %. Meanwhile, PMP hydrogel presents excellent biocompatibility and hemostatic ability. Therefore, PMP hydrogel has a great potential in the field of photothermal therapy and hemostasis.

Cotton Fabric-Reinforced Hydrogels with Excellent Mechanical and Broad-Spectrum Photothermal Antibacterial Properties

Abstract:Antibacterial hydrogel wound dressings hold great potential in eliminating bacteria and accelerating the healing process. However, it remains a challenge to fabricate hydrogel wound dressings that simultaneously exhibit excellent mechanical and photothermal antibacterial properties. Here we report the development of polydopamine-functionalized graphene oxide (rGO@PDA)/calcium alginate (CA)/Polypyrrole (PPy) cotton fabric-reinforced hydrogels (abbreviated as rGO@PDA/CA/PPy FHs) for tackling bacterial infections. The mechanical properties of hydrogels were greatly enhanced by cotton fabric reinforcement and an interpenetrating structure, while excellent broad-spectrum photothermal antibacterial properties based on the photothermal effect were obtained by incorporating PPy and rGO@PDA. Results indicated that rGO@PDA/CA/PPy FHs exhibited superior tensile strength in both the warp (289 ± 62.1 N) and weft directions (142 ± 23.0 N), similarly to cotton fabric. By incorporating PPy and rGO@PDA, the swelling ratio was significantly decreased from 673.5% to 236.6%, while photothermal conversion performance was significantly enhanced with a temperature elevated to 45.0 °C. Due to the synergistic photothermal properties of rGO@PDA and PPy, rGO@PDA/CA/PPy FHs exhibited excellent bacteria-eliminating efficiency for S. aureus (0.57%) and E. coli (3.58%) after exposure to NIR for 20 min. We believe that the design of fabric-reinforced hydrogels could serve as a guideline for developing hydrogel wound dressings with improved mechanical properties and broad-spectrum photothermal antibacterial properties for infected-wound treatment.

Abstract:Bacterial infected environments and resulting bacterial infections have been threatening the human health globally. Due to increased bacterial resistance caused by improper and excessive use of antibiotics, antibacterial biomaterials are being developed as alternatives to antibiotics in some cases. Herein, an advanced multifunctional hydrogel with excellent antibacterial properties, enhanced mechanical properties, biocompatibility and self‐healing performance, was designed through freezing‐thawing method. This hydrogel network is composed of polyvinyl alcohol (PVA), carboxymethyl chitosan (CMCS), protocatechualdehyde (PA), ferric iron (Fe) and an antimicrobial cyclic peptide actinomycin X2 (Ac.X2). The double dynamic bonds among protocatechualdehyde (PA), ferric iron (Fe) and carborxymethyl chitosan containing coordinate bond (catechol‐Fe) as well as dynamic Schiff base bonds and hydrogen bonds endowed the hydrogel with enhanced mechanical properties. Successful formation of hydrogel was confirmed through ATR and XRD, and structural evaluation through SEM analysis, whereas mechanical properties were tested with electromechanical universal testing machine. The resulting PVA/CMCS/Ac.X2/PA@Fe (PCXPA) hydrogel has favorable biocompatibility and excellent broad‐spectrum antimicrobial activity against both S. aureus (95.3%) and E. coli (90.2%) compared with free‐soluble Ac.X2, which exhibited subpar performance against E. coli reported in our previous studies. This work provides a new insight on preparing multifunctional hydrogels containing antimicrobial peptides as antibacterial material.
