Abstract:Drug resistance and increasing dangers during antibiotic treatment have brought a new eternal task for the research of effective antibacterial agents or therapeutics. In this work, we used Cu, N-doped carbon dots (Cu, N-CDs) to modify graphene oxide (GO) nanosheets and then loaded to chitosan (CS) hydrogels via electrostatic interaction to form Cu, N-CDs@GO-CS hydrogel nanoplatforms to treat Staphylococcus aureus and Escherichia coli. The excellent antibacterial activity is from the combined effects of hyperthermia and reactive oxygen species generated under near-infrared (NIR) laser irradiation of the Cu, N-CDs@GO-CS hydrogel, which shows excellent antibacterial activity compared with the CS hydrogel or the Cu, N-CDs@GO-CS hydrogel without NIR laser irradiation. Moreover, the inherent antibacterial nature of the CS hydrogel or the Cu, N-CDs@GO-CS hydrogel was used to treat bacteria-infected wounds in mice, which also protected the wound area from second infection. In vivo experiments demonstrate favorable wound healing results and have no significant harmful side effects to the major organs in mice. Overall, this work demonstrates that the antibacterial Cu, N-CDs@GO-CS hydrogel offers significant prospect as an antibacterial reagent for wound healing.
Flower-like CuS/graphene Oxide with Photothermal and Enhanced Photocatalytic Effect for Rapid Bacteria-Killing Using Visible Light

Abstract:Pathogenic bacteria have been threatening the daily life of human beings. More effective methods without causing drug-resistance of bacteria need to be developed to fight against these pathogens. Herein, flower-like CuS/-graphene oxide (GO) hybrids have been successfully synthesized via simple one-pot hydrothermal process. GO worked as an excellent electron acceptor to transport the photogenerated electrons from CuS, which can suppress the recombination of hole-electron pairs efficiently, thus enhancing the photocatalytic property. In addition, the morphology of CuS and GO with high specific surface area and the increased defect in GO also improved photocatalytic performance of the hybrid. Owing to the synergy of photothermal, enhanced photocatalytic effect and released Cu ions, CuS/GO exhibited outstanding antibacterial efficacy under visible light irradiation for 15 min. Additionally, the hybrid showed great biocompatibility to L929 cell. Hence, the synthesized CuS/GO would be a promising antibacterial material for daily life including rapid water disinfection and wounds sterilization.

Abstract:We have constructed biocompatible antibacterial nanoplatforms on the basis of a hybrid hydrogel via simple electrostatic interaction embedded with thin layer graphene oxide (GO) sheets modified with zinc oxide quantum dots (ZnO QDs). The materials possess favorable antibacterial efficacy, thereby providing excellent killing effect for Escherichia coli (Gram-negative) and Staphylococcus aureus (Gram-positive) compared with materials that have single diagnosis and treatment mode. The excellent performance stems from the combined effects of hyperthermia produced under the near-infrared irradiation of GO sheets, reactive oxygen species, the release of Zn2+ from ZnO QDs under acidic environment, and the antibacterial activity of hydrogel. The synergy of antibacterial nanoplatforms can be used for wound anti-inflammatory in vivo indicated by the wound healing results. The hybrid hydrogel causes no evident side effects to major organs in mice during wound healing. Therefore, the biocompatible multimodal therapeutic nanoplatforms have great potential for antibacterial activity and wound healing.

Abstract:Charge transfer and separation play a critical role in the photocatalytic efficiency of photoresponsive materials and their subsequent applications. Herein, for the first time, we constructed a homo-heterostructured S-Cu-FC/CuS modified carboxymethyl cellulose (CMC) hydrogel doped with graphene oxide (GO) (S-Cu-FC/CuS/GO@CMC) through an in situ ionic complexation reaction, which exhibited enhanced photocatalytic performance under 808 nm near-infrared (NIR) light irradiation. On the one hand, photo-excited charges can rapidly transfer across the homo-heterojunction interface with copper atoms in S-Cu-FC/CuS as both donors and acceptors. On the other hand, the interface between GO and S-Cu-FC/CuS also favors the fast transfer of photogenerated charges. Hence, with the assistance of a photothermal effect, the enhanced photocatalytic performance of S-Cu-FC/CuS/GO endowed the hydrogel with robust sterilization ability for killing 99.5% S. aureus and 100.0% E. coli under 10 min NIR light irradiation. An in vivo test disclosed that this hydrogel could also accelerate the healing of bacteria-infected wounds.

Abstract:The research on biocompatible materials which exhibit remarkable antibacterial capability has always attracted lots of interests. Among numerous antibacterial materials, CuS micro-/nanomaterial has gained attention as a promising option due to its low toxicity and efficiently photothermal effect. In this study, CuS nanotube/chitin composite hydrogel was prepared by crosslinking with chitin. Due to the prominent photothermal conversion efficiency of CuS nanotube which was up to 64.3 %, the as-prepared composite hydrogel showed enhanced photothermal effect under near-infrared (NIR) illumination. The composite hydrogel also showed a typical peroxidase-like activity which can convert H 2 O 2 into an extremely oxidative and toxic hydroxyl radicals (•OH). The prominent photothermal effect and peroxidase-like activity made the composite hydrogel perform well in antibacterial against Escherichia coli with NIR activation. The possible mechanism of NIR-activated antibacterial from CuS nanotube/chitin composite hydrogel was discussed as well. The results of this work indicate that the CuS/chitin composite hydrogel with enhanced photothermal effect could be a new NIR-activated antibacterial agent used for sterilization.

Abstract:In this study, catechol-conjugated chitosan (CSS) was synthesized to prepare silver nanoparticles in aqueous solution as both a reducing and stabilizing agent. The entire reaction process complied with the principles of green chemistry. The stability and physicochemical properties of CSS-coated silver nanoparticles (CSS–Ag NPs) were well characterized by a series of techniques. A narrow diameter distribution and excellent stability were observed in the target CSS–Ag NPs. Determination of the minimum inhibitory concentration (MIC) and the disk diffusion test were applied to evaluate the antibacterial activities with respect to quantity and quality, respectively. The cytotoxic effects on HepG2 cells were also evaluated with a series of assays. The resulting CSS–Ag NPs combined the unique properties of chitosan and silver nanoparticles, showing effective antibacterial activities and low cytotoxicity. The effective toxic concentration of CSS–Ag NPs towards E. coli and S. aureus is too low to damage HepG2 cells. The results demonstrated that CSS–Ag NPs could be a potential candidate for use in biological and pharmaceutical areas to prevent infections caused by microorganisms.
Chitosan-based GOx@Co-MOF composite hydrogel: A promising strategy for enhanced antibacterial and wound healing effects

Abstract:A novel composite hydrogel was synthesized via Schiff base reaction between chitosan and di-functional poly(ethylene glycol) (DF-PEG), incorporating glucose oxidase (GOx) and cobalt metal-organic frameworks (Co-MOF). The resulting CS/PEG/GOx@Co-MOF composite hydrogel was characterized using Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), thermogravimetric analysis (TGA), and energy-dispersive X-ray spectroscopy (EDS). The results confirmed successful integration and uniform distribution of Co-MOF within the hydrogel matrix. Functionally, the hydrogel exploits the catalytic decomposition of glucose by GOx to generate gluconic acid and hydrogen peroxide (H2O2), while Co-MOF gradually releases metal ions and protects GOx. This synergy enhanced the antibacterial activity of the composite hydrogel against both Gram-positive (S. aureus) and Gram-negative bacteria (E. coli), outperforming conventional chitosan-based hydrogels. The potential of the composite hydrogel in treating wound infections was evaluated through antibacterial and wound healing experiments. Overall, CS/PEG/GOx@Co-MOF hydrogel holds great promise for the treatment of wound infections, paving the way for further research and potential clinical applications.
Novel Graphene Quantum Dots Modified NH2-MIL-125 Photocatalytic Composites for Effective Antibacterial Property and Mechanism Insight

Abstract:Bacterial infection threatens human health, so the design of efficient and stable photocatalytic antibacterial materials has attracted widespread attention. In this regard, a novel photocatalyst GQDs/NH2-MIL-125 (GQDs: graphene quantum dots) was prepared by the solvothermal method for photocatalytic disinfection. Since the addition of GQDs enhanced the absorption of visible light and also accelerated the transfer of photogenerated electrons, the photocatalytic performance of GQDs/NH2-MIL-125 was significantly improved. 5 wt% GQDs loading sample (2-GQDs/NH2-MIL-125) exhibited the best antibacterial properties. The killing efficiency by 2-GQDs/NH2-MIL-125 against E. coli and S. aureus (10(7) CFU center dot mL(-1)) increased from 40 % and 75 % by NH2-MIL-125 to 92 % and 93 % within 60 min, respectively. Moreover, through the single-wavelength antibacterial experiments, it was found that the introduction of GQDs led to a wide-range-responsive antibacterial effect. Through photocatalytic trapping experiments and ESR analysis, the dominant roles of superoxide radicals (center dot O-2(-)) were confirmed in the antibacterial process. Finally, by studying the state of bacteria via TEM, confocal fluorescent images, and gel electrophoresis, it was found that the active substances not only destroyed the cell membranes but also further destroyed DNA to achieve the purpose of completely killing bacteria. This work provides a new method for the rapid disinfection of pathogenic microorganisms using GQDs-related photocatalysts.

Abstract:Bacterial infections can significantly impede wound healing and pose a serious threat to the patient's life. The excessive use of antibiotics to combat bacterial infections has led to the emergence of multi-drug-resistant bacteria. Therefore, there is a pressing need for alternative approaches, such as photothermal therapy (PTT), to address this issue. In this study, for the first time, CuS NPs with photothermal properties were synthesized using sericin as a biological template, named CuS@Ser NPs. This method is simple, green, and does not produce toxic and harmful by-products. These nanoparticles were incorporated into a mixture (XK) of xanthan gum and konjac glucomannan (KGM) to obtain XK/CuS NPs composite hydrogel, which could overcome the limitations of current wound dressings. The composite hydrogel exhibited excellent mechanical flexibility, photothermal response, and biocompatibility. It also demonstrated potent antibacterial properties against both Gram-positive and negative bacteria via antibacterial experiments and accelerated wound healing in animal models. Additionally, it is proved that the hydrogel promoted tissue regeneration by stimulating collagen deposition, angiogenesis, and reducing inflammation. In summary, the XK/CuS NPs composite hydrogel presents a promising alternative for the clinical management of infected wounds, offering a new approach to promote infected wound healing.

Abstract:In response to the wide spread of microbial contamination induced by bacterial pathogens, the development of novel materials with excellent antibacterial activity is of great interest. In this study, novel antibacterial chitosan (CS) and polyhexamethylene guanidine hydrochloride (PHGC) dual-polymer-functionalized graphene oxide (GO) (GO-CS-PHGC) composites were designed and easily fabricated. The as-prepared materials were characterized by Fourier transform infrared (FTIR), X-ray photoelectron spectrometer (XPS), field emission scanning electron microscopy (FE-SEM), transmission electron microscope (TEM), thermogravimetric analysis (TGA) and Raman spectroscopy. Their antibacterial capability towards bacterial strains was also studied by incubating both Gram-negative bacteria and Gram-positive bacteria in their presence. More significantly, the synergistic antibacterial action of the three components was assayed, and the findings implied that the as-prepared GO-CS-PHGC shows enhanced antibacterial activity when compared to its single components (GO, CS, PHGC or CS-PHGC) and the mixture of individual components. Not only Gram-negative bacteria but also Gram-positive bacteria are greatly inhibited by GO-CS-PHGC composites. The minimum inhibitory concentration (MIC) value of GO-CS-PHGC against E. coli was 32 μg/mL. With the powerful antibacterial activity as well as its low cost and facile preparation, GO-CS-PHGC has potential applications as a novel antibacterial agent in a wide range of biomedical uses.

Abstract:Microbial infection has been a major threat to human health worldwide. With a dramatic increase in bacterial resistance, it is critical to develop nonantibiotic-dependent therapies to conquer the bacterial drug-resistance. In this work, we constructed Cu2Se/Bi2S3 nanocomposites used as both chemodynamic therapy and photothermal therapy agents. It had synergistic bactericidal efficiency based on peroxidase-like activity and near-infrared (NIR)-induced hyperthermia, which made Cu2Se/Bi2S3 nanocomposites possess good anti-Escherichia coli and anti-Staphylococcus aureus ability with inappreciable cytotoxicity. Moreover, it was further manifested that S. aureus infected wound treating with Cu2Se/Bi2S3 NCs presented excellent healing outcome.

Facile Synthesis of Thermo-Sensitive Composite Hydrogel with Well Dispersed Ag Nanoparticles for Application in Superior Antibacterial Infections

Abstract:In this study, we have described a facile process for fabrication of multifunctional composite hydrogel, in which sodium alginate was subjected to cross-linking using Ca2+ derived from ZnO/CaCO₃/Ag composite nanospheres. The ZnO/CaCO₃/Ag composite nanospheres were prepared based on our previously reported AA-[Zn(OH)₄]2- composite nanosphere reaction conducted with silver and calcium salt following hydrothermal method, that led to the disintegration and release of Ca2+ under acidic conditions for application as a cross-linking agentto catalyze reaction with sodium alginate. Ag nanoparticles were well-dispersed in the multifunctional composite hydrogel, exhibiting excellent antibacterial activity. Additionally, polydopamine (PDA) with photothermal effect was also added to obtain a multifunctional composite hydrogel, and this hydrogel showed photothermal conversion performance and facilitated the release of Ag+ to achieve the rapid antibacterial effect. Simultaneously, PDA NPs could scavenge free radicals and improve cell adhesion. All such features would promote wound healing. The potent antimicrobial activity of the prepared composite hydrogel was demonstrated in the mouse model of S. aureus infection, and biosafety of the hydrogel was confirmed by conducting histopathological examination in the mouse model. This type of multifunctional hydrogel wound dressing with photosensitive and antibacterial properties presents with broad applications and prospects in antibacterial treatment.
Photothermal Hydrogel Composites Featuring G4-Carbon Nanomaterial Networks for Staphylococcus aureus Inhibition

Abstract:Microbial infections represent a significant health risk, often leading to severe complications and, in some cases, even fatalities. As a result, there is an urgent need to explore innovative drug delivery systems and alternative therapeutic techniques. The photothermal therapy has emerged as a promising antibacterial approach and is the focus of this study. Herein, we report the successful synthesis of two distinct supramolecular composite hydrogels by incorporating graphene oxide (GO) and single-walled carbon nanotubes (SWNTs) into guanosine quadruplex (G4) based hydrogels containing covalently bound β-cyclodextrin (β-CD). The G4 matrix was synthesized through a two-step process, establishing a robust network between G4 and β-CDs, followed by the encapsulation of either GO or SWNTs. Comprehensive characterization of these composite hydrogels were conducted using analytical techniques, including circular dichroism, Raman spectroscopy, rheological investigations, X-ray diffraction, and scanning electron microscopy. A notable discovery from the conducted research is the differential photothermal responses exhibited by the hydrogels when exposed to near-infrared laser irradiation. Specifically, SWNT-based hydrogels demonstrated superior photothermal performance, achieving a remarkable temperature increase of up to 52 °C, in contrast to GO-based hydrogels, which reached a maximum of 34 °C. These composite hydrogels showed good cytotoxicity evaluation results and displayed synergistic antibacterial activity against Staphylococcus aureus, positioning them as promising candidates for antibacterial photothermic platforms, particularly in the context of wound treatment. This study offers a valuable contribution to the development of advanced and combined therapeutic strategies for combating microbial infections and highlights the potential of carbon nanomaterial-enhanced supramolecular hydrogels in photothermal therapy applications.

Abstract:Metal chalcogenides have been intensively investigated as antibacterial agents due to their unique structures and superior photoactivities. Herein, various structures of copper sulfide (CuS), a metal chalcogenide, such as microspheres (MSs), nanosheets (NSs), and nanoparticles (NPs), were developed in this work for antibacterial applications. A hydrothermal process was utilized to synthesize CuS MSs, CuS NSs, and CuS NPs. Under simulated solar light and near-infrared (NIR) light irradiation, the antibacterial behaviors, reactive oxygen species (ROS) production, and light-driven antibacterial mechanisms of CuS MSs, CuS NSs, and CuS NPs were demonstrated with the bacterium Escherichia coli (E. coli). Bacterial growth curves and ROS generation tests indicated that CuS NSs and CuS NPs had higher light-driven antibacterial activities than that of CuS MSs. ROS of hydroxyl (·OH) and superoxide anion radicals (O2-) were investigated via an electron spin resonance (ESR) spectroscopic analysis by respectively incubating CuS MSs, CuS NSs, and CuS NPs with E. coli under simulated solar light irradiation. Furthermore, E. coli incubated with CuS NPs and CuS NSs showed substantial bacterial degradation after NIR laser irradiation, which was attributed to their photothermal killing effects. Light-driven antibacterial mechanisms of CuS NSs and CuS NPs were investigated, and we discovered that under simulated solar and NIR light irradiation, CuS NSs and CuS NPs produced photoinduced electrons, and the copper ions and photoinduced electrons then reacted with atmospheric moisture to produce hydroxide and superoxide anion radicals and heat, resulting in bacterial mortality.

Abstract:Due to the increasing resistance of pathogens to antibiotics, novel antibacterial nanocomposite materials have been attracting increasing research interest. Herein, we report a facile synthetic method for a novel highly antibacterial nanocomposite based on graphene oxide (GO) nanosheets decorated with cuprous oxide by a two-step method. The as-prepared nanocomposites were comprehensively characterized by X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), energy-dispersive X-ray (EDX) spectroscopy, field emission scanning electron microscopy (FESEM) and transmission electron microscopy (TEM). The nanocomposite was investigated for antibacterial activity against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) using the medium dilution method and the disc diffusion method. The results showed that the synthesized Cu2O-GO has good antibacterial activities. At the nanocomposites concentration of 100 mu g/mL, the inhibition rate against E. coli reached 68%, while the inhibition rate against. S. aureus reached 32%.
Review on chitosan-based antibacterial hydrogels: Preparation, mechanisms, and applications

Abstract:Chitosan (CS) is known for its remarkable properties, such as good biocompatibility, biodegradability, and renewability, in addition to its antibacterial and biological activities. However, as CS is insoluble in water, it displays limited antibacterial performance under neutral and physiological conditions. A viable solution to this problem is grafting chemically modified groups onto the CS framework, thereby increasing its solubility and enhancing its antibacterial effect. Herein, the antibacterial action mechanism of CS and its derivatives is reviewed, confirming the prevalent use of composite materials comprising CS and its derivatives as an antibacterial agent. Generally, the antimicrobial ability of CS-based biomaterials can be enhanced by incorporating supplementary polymers and antimicrobial agents. Research on CS-based composite biomaterials is ongoing and numerous types of biomaterials have been reported, including inorganic nanoparticles, antibacterial agents, and CS derivatives. The development of these composite materials has considerably expanded the application of CS-based antibacterial materials. This study reviews the latest progress in research regarding CS-based composite hydrogels for wound repair, tissue engineering, drug release, water purification, and three-dimensional printing applications. Finally, the summary and future outlook of CS-based antibacterial hydrogels are presented in anticipation of a broader range of applications of CS-based antibacterial hydrogels.

Polyvinyl alcohol/chitosan quaternary ammonium salt composite hydrogel with directional macroporous structure for photothermal synergistic antibacterial and wound healing promotion

Abstract:After skin tissue trauma, wound infections caused by bacteria posed a great threat to skin repair. However, resistance to antibiotics, the current treatment of choice for bacterial infections, greatly affected the efficiency of anti-infection and wound healing. Therefore, there has been a critical need for the development of novel antimicrobial materials and advanced therapeutic methods to aid in skin repair. In this paper, rGO-PDA@ZIF-8 nanofillers were prepared by coating graphene oxide (GO) with dopamine (DA), followed by in situ growth of zeolite imidazolate framework-8 (ZIF-8). Using polyvinyl alcohol (PVA) and chitosan quaternary ammonium salt (CS) as matrix materials, along with polyethylene glycol (PEG) as a pore-forming agent, and rGO-PDA@ZIF-8 as an antimicrobial nano-filler, we successfully prepared rGO-PDA@ZIF-8/PVA/CS composite hydrogels with a directional macroporous structure using bidirectional freezing method and phase separation technique. This hydrogel exhibited excellent mechanical properties, good solubility and water retention capabilities. In addition, the hydrogel demonstrated excellent biocompatibility. Most notably, it not only exhibited excellent bactericidal effect against E. coli and S. aureus (99.1 % and 99.0 %, respectively) under the synergistic effect of intrinsic antibacterial activity and photothermal antibacterial, but also exhibited the ability to promote wound healing, making it a promising candidate for wound healing applications.
Synthesis of a Two-Dimensional Molybdenum Disulfide Nanosheet and Ultrasensitive Trapping of Staphylococcus Aureus for Enhanced Photothermal and Antibacterial Wound-Healing Therapy

Abstract:Photothermal therapy has been widely used in the treatment of bacterial infections. However, the short photothermal effective radius of conventional nano-photothermal agents makes it difficult to achieve effective photothermal antibacterial activity. Therefore, improving composite targeting can significantly inhibit bacterial growth. We inhibited the growth of Staphylococcus aureus (S. aureus) by using an extremely low concentration of vancomycin (Van) and applied photothermal therapy with molybdenum disulfide (MoS2). This simple method used chitosan (CS) to synthesize fluorescein 5(6)-isothiocyanate (FITC)-labeled and Van-loaded MoS2-nanosheet hydrogels (MoS2-Van-FITC@CS). After modifying the surface, an extremely low concentration of Van could inhibit bacterial growth by trapping bacteria synergistically with the photothermal effects of MoS2, while FITC labeled bacteria and chitosan hydrogels promoted wound healing. The results showed that MoS2-Van-FITC@CS nanosheets had a thickness of approximately 30 nm, indicating the successful synthesis of the nanosheets. The vitro antibacterial results showed that MoS2-Van-FITC with near-infrared irradiation significantly inhibited S. aureus growth, reaching an inhibition rate of 94.5% at nanoparticle concentrations of up to 100 μg/mL. Furthermore, MoS2-Van-FITC@CS could exert a healing effect on wounds in mice. Our results demonstrate that MoS2-Van-FITC@CS is biocompatible and can be used as a wound-healing agent.

Abstract:The wound healing in the presence of bacterial infections is a complex process and prolonged challenge with severe implications for human health. To address this issue, injectable thermo-sensitive hydrogels as wound dressings have garnered considerable attention as a potential solution. In this study, we have developed an injectable and antibacterial thermosensitive hydrogel for wound healing, based on chitosan (CS) and β-glycerophosphate (β-GP) and modified with a range of cinnamaldehyde (CA). The CS/β-GP/CA gels demonstrate a phase transition at body temperatures, and exhibit good biocompatibility. Compared with the unmodified CS/β-GP hydrogels, incorporating CA can promote endothelial cell growth and the antibacterial property of the hydrogel system, which may lead to faster wound healing and tissue regeneration. Our findings suggest that the CS/β-GP/CA thermosensitive hydrogel emerges as a promising candidate for an injectable wound dressing in wound treatments.
