Abstract:Polymeric prodrugs have attracted great interest in the field of anticancer drug delivery owing to their integrated advantages of prodrugs and nanoparticles. However, the ambiguous chemical composition of polymeric prodrugs is still a major obstacle to their clinical applications. In this study, an amphiphilic poly(ethylene glycol) derivative with multiple thiols was first synthesized on a large scale through the melt co-polycondensation of oligo(ethylene glycol), 1,4-butanediol and mercaptosuccinic acid. Then an adamantane modified doxorubicin (DOX) with a pH-sensitive hydrazone bond and a beta-cyclodextrin derivative with multiple acryloyls were precisely synthesized. Finally, novel supramolecular polymeric prodrug (SPP) micelles with a precise drug molecular structure and drug loading content were facilely prepared using the above three components via simultaneous self-assembly, a thiol-ene "click" reaction and a host-guest supramolecular interaction in water. The resultant pH-sensitive SPP micelles with a stable core-crosslinked structure and strong drug "bonding" show long blood circulation time, effective cellular uptake and fast DOX release in tumor cells, which significantly improved the antitumor efficacy and security compared with free DOX. This work established a promising drug delivery platform for cancer chemotherapy.

Abstract:Cancer has become a major public health problem, and chemotherapy is one of the main methods for treating cancer. However, traditional anticancer drugs have shortcomings such as poor selectivity, short half-life, and large toxic side effects. Recently, polymeric prodrugs with therapeutic moieties covalently bonding to polymers have attracted considerable interests in anticancer field, which can self-assemble into nanodrugs with improved the durability, targeting and bioavailability comparing to traditional anticancer drugs. Nevertheless, the chemical composition of polymeric prodrugs is complex due to the polydispersity of a polymer, which greatly limits their clinical translation. In order to obtain polymeric prodrugs with accurate chemical composition, we designed and synthesized an amphiphilic supramolecular polymeric prodrug based on host-guest interaction, which can self-assemble into nano-sized micelles in aqueous medium for anticancer chemotherapy. The novel supramolecular polymeric prodrug we report in this work can combine the general advantages of polymeric prodrug and small molecular prodrug, which should have a bright future in clinical chemotherapy.
Hydrogen Sulfide-Specific and NIR-Light-Controllable Synergistic Activation of Fluorescent Theranostic Prodrugs for Imaging-Guided Chemo-Photothermal Cancer Therapy

Abstract:Theranostic prodrugs are promising for cancer medicine; however, the inability to activate these systems exclusively at the desired tumor location compromises the specificity and efficacy of cancer treatment. Here, we developed a fluorescent theranostic nanoprodrug with synergistic hydrogen-sulfidespecific and near-infrared (NIR)-light-controllable activation for imaging-guided chemo-photothermal cancer therapy. This nanoprodrug system was fabricated by the inclusion of hydrogen sulfide (H2S)activatable small molecule to the theranostic prodrug and a photothermal transducer in the interior of a NIR-light-responsive container. The resultant nanoprodrug could be activated specifically, evidenced by a "turn on" of NIR fluorescence in H2S-rich cancers; thus, enabling accurate cancer location and identification of when and where to implement NIR irradiation for efficient conversion of photoenergy into heat. Such a prominent photothermal effect guarantees photothermal ablation of cancers and the release of the H2S-preactivated DOX for cancer chemotherapy. By utilizing this H2S-responsive and NIR-light-controllable nanoprodrug, complete suppression of tumors was realized under the guidance of the NIR fluorescence imaging. Thus, our study provides a novel strategy toward theranostic prodrug systems that enable precision cancer treatment with high specificity and efficacy.

Abstract:Unimolecular micelles have attracted more interests in tumor chemotherapy owing to their superior stability. Here, GSH-triggered disintegrable hyperbranched polymer prodrug (HBPP-DOX) was designed via the self-condensing vinyl polymerization (SCVP) of reduction-sensitive monomer prodrug (MA-SS-DOX) with reduction-sensitive inimer (MA-SS-Br), in which polyethylene glycol monomethyl ether methacrylate (PEGMA) was used as comonomer to introduce hydrophilic brushes. The proposed HBPP-DOX with a high drug content of 32% could easily self-assemble into unimolecular micelles, which could be completely disintegrated and release drug in the tumor cells triggered by the high GSH level therein, showing excellent tumor-specific cytotoxicity with enhanced tumor growth inhibition.

Abstract:Poly(ethylene glycol) (PEG) is widely utilized as a hydrophilic coating to extend the circulation time and improve the tumor accumulation of polymeric micelles. Nonetheless, PEGylated micelles often activate complement proteins, leading to accelerated blood clearance and negatively impacting drug efficacy and safety. Here, we have crafted amphiphilic block copolymers that merge hydrophilic sulfoxide-containing polymers (psulfoxides) with the hydrophobic drug 7-ethyl-10-hydroxylcamptothecin (SN38) into drug-conjugate micelles. Our findings show that the specific variant, PMSEA-PSN38 micelles, remarkably reduce protein fouling, prolong blood circulation, and improve intratumoral accumulation, culminating in significantly increased anti-cancer efficacy compared with PEG-PSN38 counterpart. Additionally, PMSEA-PSN38 micelles effectively inhibit complement activation, mitigate leukocyte uptake, and attenuate hyperactivation of inflammatory cells, diminishing their ability to stimulate tumor metastasis and cause inflammation. As a result, PMSEA-PSN38 micelles show exceptional promise in the realm of anti-metastasis and significantly abate SN38-induced intestinal toxicity. This study underscores the promising role of psulfoxides as viable PEG substitutes in the design of polymeric micelles for efficacious anti-cancer drug delivery.
The design and synthesis of dextran-doxorubicin prodrug-based pH-sensitive drug delivery system for improving chemotherapy efficacy

Abstract:Tumor cells show acidic conditions compared with normal cells, which further inspires scientist to build nanocarrier responsive to tumor microenvironment (TME) for enhancing tumor therapeutic efficacy. Here, we report a pH-sensitive and biocompatible polyprodrug based on dextran-doxorubicin (DOX) prodrug (DOXDT) for enhanced chemotherapy. High-density DOX component was covalently decorated on the nanocarrier and the drug molecules could be effectively released in the acidic tumor tissue/cells, improving chemotherapy efficacy. Specifically, a dextran-based copolymer was preliminarily prepared by one-step atom transfer radical polymerization (ATRP); then DOX was conjugated on the copolymer component via pH-responsive hydrazone bond. The structure of DOXDT can be well-controlled. The resulting DOXDT was able to further self-assemble into nanoscale micelles with a hydration diameter of about 32.4 nm, which presented excellent micellar stability. Compared to lipid-based drug delivery system, the DOXDT prodrug showed higher drug load capacity up to 23.6%. In addition, excellent stability and smaller size of the nanocarrier contributed to better tissue permeability and tumor suppressive effects in vivo. Hence, this amphipathic DOXDT prodrug is promising in the development of translational DOX formulations, which would be widely applied in cancer therapy.Biocompatible polyprodrug based on dextran-doxorubicin (DOXDT) with high drug loading and pH-triggered release for enhanced chemotherapy.

Abstract:Combination chemotherapy has attracted more intense attention in tumor treatment, in which the drug-loading mode and drug proportion of the nano drug co-delivery system (NDCDS) played an important role in the anti-tumor efficacy. Here, GSH-triggered degradable hyperbranched polymer coprodrugs were designed via the self-condensing vinyl polymerization (SCVP) of the polymerizable prodrug monomers and poly(ethylene glycol) methacrylate (PEGMA) with a GSH-sensitive inimer. The drug proportion of the polymer coprodrugs could be easily adjusted by altering the feeding ratio for a better synergistic effect. The proposed polymer coprodrugs could form unimolecular micelles and be degraded completely to release the two parent drugs (DOX and CPT) responding to the high-level GSH in the tumor cells with a very low drug premature leakage in the intercellular environment and a low mis-release in the normal cells. The optimized one, HBPP-DOX/CPT-5, possessed a low CI 50 of 0.11, showing promising potential in safe and effective tumor combination chemotherapy.

Abstract:Recently, the global trend in the field of nanomedicine has been toward the design of highly sophisticated drug delivery systems with specific targeting and synergistic therapeutic functions for improving therapeutic efficacy. But offering sophistication generally increases their complexity that might be disadvantageous in pharmaceutical development. We hypothesize that using a macromolecular prodrug with a dual role will be conductive to integrating its dual function into self-targeted multidrug co-delivery and combination cancer therapy. In this paper, the on-off switching function-responsive, macromolecular methotrexate (MTX) prodrug-self-targeted, controlled-/sustained-release, and high drug-loading hydroxylcamptothecin (HCPT) drug nanospheres were prepared and characterized. The self-targeting system can co-deliver multi-drug to different action sites with distinct anticancer mechanisms to specifically target folate receptors-overexpressing cancer cells with synergistic therapeutic efficiency.

Abstract:Hydrogen sulfide is essential in numerous physiological and pathological processes and has emerged as a promising cancer imaging and signaling molecule and a potentially versatile therapeutic agent. However, the endogenous levels of hydrogen sulfide remain insufficient to perform its biological functions, and thus, developing novel strategies that amplify hydrogen sulfide signals at lesion sites is of increasing interest. In this work, a nanoplatform (SNP) based on hydrogen sulfide-responsive self-immolative poly(thiocarbamate) with localized hydrogen sulfide signal amplification capability is developed to encapsulate a hydrogen sulfide-responsive fluorescent probe (e.g., hemicyanine dye; p-Cy) or an anticancer prodrug (e.g., doxorubicin; p-DOX) to form a nanoprobe (SNPp-Cy) or nanomedicine (SNPp-DOX) for cancer imaging and therapy, respectively. SNPp-Cy exhibits a low detection limit for hydrogen sulfide, enabling ultrasensitive detection of small (<2 mm) tumors in female mice. In addition, SNPp-DOX can effectively inhibit the growth of DOX-resistant human breast cancer xenograft, lung metastasis, and patient-derived xenograft tumors in female mice.
Hypoxia-responsive block copolymer polyprodrugs for complementary photodynamic-chemotherapy

Abstract:The inherent hypoxic microenvironment of solid tumors has an important influence on tumor growth, distant metastasis, and invasiveness. The heterogeneous distribution of hypoxic regions inside tumors limits the therapeutic efficacy of O2-assisted therapeutic strategy (e.g. photodynamic therapy (PDT)). On the other hand, the hypoxia-activable prodrugs cannot work effectively in the regions with enough O2 concentration. To address the issues, we prepare a block copolymer polyprodrug consisting of polyethylene glycol (PEG) and copolymerized segments of nitroimidazole-linked camptothecin (CPT) methacrylate and 5,10,15,20-tetraphenylporphyrin (TPP)-containing methacrylate monomers for complementary photodynamic-chemotherapy. The polyprodrug can self-assemble into polymeric micelles in aqueous solution with suitable size and high stability. After intravenous injection, the polyprodrug micelles show tumor accumulation. Followed by light irradiation (650 nm) at tumor sites, TPP moieties induce singlet oxygen (1O2) production in the oxygen-rich area to exert PDT and cause transformation of the oxygen-rich areas into hypoxia. Simultaneously, in the hypoxic areas, the hypoxia-responsive polyprodrugs can be activated to release free CPT due to the cleavage of nitroimidazole linkages. The polyprodrug micelles with the segments for PDT and hypoxia-activable CPT efficiently suppress the growth of HeLa tumors. The well-defined polyprodrug amphiphiles offer an effective strategy to overcome the disadvantages of single treatment of PDT or hypoxia-responsive prodrugs for complementary photodynamic-chemotherapy of cancers.

Abstract:The cancer drug delivery process involves a series of biological barriers, which require the nanomedicine to exhibit different, even opposite properties for high therapeutic efficacy. The prevailing design philosophy, i.e., integrating these properties within one nanomedicine via on-demand property transitions such as PEGylation/dePEGylation, complicates nanomedicines' composition and thus impedes clinical translation. Here, polyzwitterionic micelles of poly(tertiary amine-oxide)-block-poly(ε-caprolactone) (PTAO-PCL) amphiphiles that enable all the required functions are presented. The zwitterionic nature and unique cell membrane affinity confer the PTAO micelles long blood circulation, efficient tumor accumulation and penetration, and fast cellular internalization. The mitochondrial targeting capability allows drug delivery into the mitochondria to induce mitochondrial dysfunction and overcome tumor multidrug resistance. As a result, the PTAO/drug micelles exhibit potent anticancer efficacy. This simple yet multipotent carrier system holds great promise as a generic platform for potential clinical translation.

Acid-responsive Endosomolytic Polymeric Nanoparticles with Amplification of Intracellular Oxidative Stress for Prodrug Delivery and Activation.

Abstract:Prodrug strategy especially in the field of chemotherapy of cancers possesses significant advantages reducing the side toxicity of anticancer drugs. However, high-efficiency delivery and in situ activation of prodrugs for tumor growth suppression are still a great challenge. Herein, we report rationally engineered pH-responsive endosomolytic polymeric micelles for the delivery of an oxidation-activable prodrug into the cytoplasm of cancer cells and amplification of intracellular oxidative stress for further prodrug activation. The prepared block copolymers consist of a poly(ethylene glycol) (PEG) block and a segment grafted by endosomolytic moieties and acetal linkage-connected cinnamaldehyde groups. The amphiphilic diblock copolymers can self-assemble to form micelles in water for loading the oxidation-activable phenylboronic pinacol ester-caged camptothecin prodrug (ProCPT). The obtained micelles can release free cinnamaldehyde under acidic conditions in tumor tissues and endo/lysosomes followed by efficient endosomal escape, which further induces enhancement of intracellular reactive oxygen species (ROS) to activate the prodrugs. Simultaneously, intracellular glutathione (GSH) can be reduced by quinone methide that was produced during prodrug activation. The ProCPT-loaded micelles can finally achieve efficient tumor accumulation and retention as well as effective tumor growth inhibition. More importantly, hematological and pathological analysis of toxicity reveals that the ProCPT-loaded micelles do not cause obvious toxic side effects toward important organs of mice. A positive immunomodulatory microenvironment in tumor tissue and serum can be detected after treatment with ProCPT-loaded micelles. Therefore, the endosomolytic ProCPT-loaded micelles exert synergistic therapeutic effects toward tumors through amplification of intracellular oxidative stress and activation of the prodrugs.
Engineering Thio-/seleno-ether Linkers into Zwitterionic Small Molecule Nano-prodrugs for Traceable Cancer Theranostics

Abstract:Given the over-production of glutathione (GSH) and hydrogen peroxide (H 2 O 2 ) in tumor cells, redox-responsive linkers such as disulfide bonds have been widely used to develop site-specific prodrug system, but may suffer from their poor sensitivity that results in low efficacy. Herein, we report the development of a class of redox-triggered small molecule prodrugs (RhB-S(Se)-CPT), by conjugating zwitterionic rhodamine B (RhB) and camptothecin (CPT) with thio-/seleno-ether linkers. The amphiphilic prodrug can self-assemble into stable nanoparticles in water, and zwitterionic RhB is located on the surface of RhB-S(Se)-CPT nanoparticles, tracking the distribution and kinetics of drug delivery in real time, and reducing the non-specific binding to in vivo components, thus decreasing the side effects of nanomedicines. The thio-/seleno-ether linkers enable selective and rapid release of CPT in response to a redox heterogeneous intratumoral microenvironment. Interestingly, selenoether bonds exhibit significant advantages over thioether in terms of CPT release rate, intracellular toxicity and therapeutic efficacy against large tumors (~ 200 mm 3 ) in mice. These results provide insights to the rational design of subsequent redox-responsive nanoprodrug system, which may contribute to the material selection and structural design of next-generation nanomedicines for tumor theranostics.

A Co-delivery System Based on a Dimeric Prodrug and Star-Shaped Polymeric Prodrug Micelles for Drug Delivery

Abstract:Chemotherapy is one of the commonly used therapies for the treatment of malignant tumors. Insufficient drug-loading capacity is the major challenge for polymeric micelle–based drug delivery systems of chemotherapy. Here, the redox-responsive star-shaped polymeric prodrug (PSSP) and the dimeric prodrug of paclitaxel (PTX) were prepared. Then the dimeric prodrug of PTX (diPTX, diP) was loaded into the core of the star-shaped polymeric prodrug micelles of PSSP by hydrophobic interaction forming the redox-responsive prodrug micelles of diPTX@PSSP for intracellular drug release in tumor cells. The hydrodynamic diameter of diPTX@PSSP nanoparticles was 114.3 nm ± 2.1 (PDI = 0.219 ± 0.016), and the micelles had long-term colloidal stability and the drug-loading content (DLC) of diPTX and PTX is 16.7 and 46.9%, respectively. The prepared micelles could broke under the reductive microenvironment within tumor cells, as a result, the dimeric prodrug of diP and polymeric prodrug micelles of PSSP were rapidly disassembled, leading to the rapid release of intracellular drugs. In vitro release studies showed that under the condition of reduced glutathione (GSH) (10 mM), the release of PTX was significantly accelerated with approximately 86.6% released within 21 h, and the released PTX in cytoplasm could promote the disintegration of microtubules and induce cell apoptosis. These results indicated that the new type of this reduction-sensitive nanodrug delivery system based on dimeric prodrug@polymeric prodrug micelles would be a promising technology in chemotherapy.

Abstract:The field of anticancer nanomedicine seeks to boost the arsenal’s exploitation with intelligent performance. One opportunistic choice comes from the fabrication of amphiphilic polymer micelles that are reversibly crosslinked. Here, hydrogen-bond (H-bond)-driven core-crosslinked supramolecular polymer micelles (FUS/ICG@PEDD) are constructed, co-loading α,ω-functionalized symmetrical H-bonding prodrug 5-fluorouracil-acetic acid–SS–5-fluorouracil-acetic acid (FUS) and a dual photothermal/photodynamic agent (indocyanine green, ICG). Such core-crosslinked nanomedicine is characterized by enhanced drug loading content, crosslinking-prompted stability, pH-responsive charge reversal, and smart drug release, which can facilitate the engineering of synergistic chemo/photothermal/photodynamic therapies (CT/PTT/PDT). To do so, an amphiphilic diblock copolymer, PEG-b-P(DAPA-co-DEAEMA) (denoted as PEDD), is developed to serve as a drug delivery vehicle, with hydrophilic PEG [poly(ethylene glycol)] and hydrophobic P(DAPA-co-DEAEMA) [poly(diaminopyridine acrylamide-co-2-(diethylamino)ethyl methacrylate], bearing randomly dispersed dual functionalities: pH-responsive charge-reversal DEAEMA and H-bonding DAP motifs. Thanks to the specific DAP/FUS H-bonding interactions and two-terminal FU structure of the prodrug, core-crosslinked nanomedicine FUS/ICG@PEDD is thus enabled. In vitro and in vivo investigations indeed reveal remarkable antitumor efficacy. We believe that such H-bonded nanomedicines can fuel the development of intelligent nanomedicines with value in cancer therapy.

Disulfide bonds as a molecular switch of enzyme-activatable anticancer drug precise release for fluorescence imaging and enhancing tumor therapy

Abstract:Enzyme-activatable drug delivery systems have been developed for cancer diagnosis and therapy. However, targeted intracellular drug delivery is a challenge for precisely tumor imaging and therapy due to the increased stability of copolymer nanoparticles (NPs) is accompanied by a notable decrease in enzyme degradation. Herein, disulfide bond was designed as an enzyme-activatable molecular switch of SS-P(G2)2/DOX NPs. The copolymer NPs consists of polyvinylpyrrolidone (PVP) with disulfide bonds in the center and enzyme-degradable peptide dendrites (Phe-Lys) to form dendritic-linear-dendritic triblock copolymers (TBCs). The amphiphilic TBCs could be split into two identical amphiphilic diblock copolymers (DBCs) by glutathione (GSH) in cancer cells specifically while maintaining the same hydrophilic-lipophilic equilibrium. This structural transformation significantly reduced the stability of copolymer NPs and enhanced sensitivity of DOX release by cathepsin B-activated. Subsequently, the released DOX acted as an indicator of fluorescence imaging and chemotherapy drug for cancer cells. The polymeric NPs achieved excellent drug-loaded stability and prolonged blood circulation in vivo, and realized fluorescence imaging and specific cancer cell killing capabilities by responding to the overexpression of GSH and cathepsin B in tumor cells. Furthermore, the copolymer NPs demonstrated excellent blood compatibility and biosafety. Therefore, a novel strategy based on one tumor marker acting as the switch for another tumor microenvironment responsive drug delivery system could be designed for tumor intracellular imaging and chemotherapy.
Hydrogen Sulfide Delivery System Based on Salting-Out Effect for Enhancing Synergistic Photothermal and Photodynamic Cancer Therapies

Abstract:The simultaneous application of photothermal therapy (PTT) and photodynamic therapy (PDT) offers substantial advantages in cancer treatment. However, their synergistic anticancer efficacy is often limited by tumor hypoxia, and thermotolerance induced by high expression of heat shock proteins (HSP). Fortunately, hydrogen sulfide (H2S), known for its direct cytotoxic effect on tumor cells, has been recognized for its ability to enhance PTT and PDT. The effectiveness of H2S in these therapies is challenged by its low loading efficiency, poor stability, and short diffusion distance. To address these issues, a nanoscale emulsion drop template created through the salting-out effect is employed to construct a robust H2S delivery system. Polydopamine (PDA), chosen for its interfacial polymerization tendency and excellent photothermal conversion rate, is utilized as a carrier for the H2S donor (ADT) and Zinc phthalocyanine (ZnPc) to fabricate a novel nanomedicine termed APZ NPs. The temperature-responsive APZ NPs are designed to release H2S during the PTT process. Elevated H2S levels promoted vasodilation, thereby enhancing the enhanced permeability and retention effect (EPR) of APZ NPs within solid tumors. This strategy effectively alleviated tumor hypoxia by disrupting the mitochondrial respiratory chain and mitigated tumor cell heat tolerance by inhibiting HSP expression.
Tumor-specific activatable biopolymer nanoparticles stabilized by hydroxyethyl starch prodrug for self-amplified cooperative cancer therapy

Abstract:Rationale: Chemodynamic therapy (CDT) is an emerging tumor-specific therapeutic strategy. However, the anticancer activity of CDT is impeded by the insufficient Fenton catalytic efficiency and the high concentration of glutathione (GSH) in the tumor cells. Also, it is challenging to eliminate tumors with CDT alone. Thus, simple strategies aimed at constructing well-designed nanomedicines that can improve therapeutic efficiency of CDT and simultaneously incorporate extra therapeutic modes as helper are meaningful and highly required. Method: Tailored to specific features of tumor microenvironment (TME), in this study, we developed a biosafe, stable and TME-activated theranostic nanoplatform (P(HSD-Cu-DA)) for photoacoustic imaging (PAI) and self-amplified cooperative therapy. This intelligent nanoplatform was fabricated following a simple one-pot coordination and polymerization strategy by using dopamine and Cu2+ as precursors and redox-responsive hydroxyethyl starch prodrugs (HES-SS-DOX) as stabilizer. Results: Interestingly, the pre-doped Cu2+ in polydopamine (PDA) framework can endow P(HSD-Cu-DA) NPs with tumor-specific CDT ability and remarkably enhance NIR absorption of PDA. PAI and biodistribution tests proved such nanoplatform can effectively accumulate in tumor tissues. Following enrichment, massive amounts of toxic hydroxyl radicals (·OH, for CDT) and free DOX (for chemotherapy) were generated by the stimulation of TME, which was further boosted by local hyperthermia. Concomitantly, in the process of activating these therapeutic functions, GSH depletion triggered by disulfide bond (-SS-) breakage and Cu2+ reduction within tumor cells occurred, further amplifying intratumoral oxidative stress. Importantly, the framework structure dominated by bioinspired polydopamine and clinical-used HES guaranteed the long-term biosafety of in vivo treatment. As a result, the mutual promotion among different components yields a potent tumor suppression outcome and minimized systemic toxicity, with one dosage of drug administration and laser irradiation, respectively. Conclusion: This work provides novel insights into designing efficient and tumor-specific activatable nanotherapeutics with significant clinical translational potential for cancer therapy.
