Abstract:A series of amphiphilic pH-responsive poly (ethylene glycol) methyl ether-b-(poly lactic acid-co-poly (β-amino esters)) (MPEG-b-(PLA-co-PAE)) block copolymers with different PLA/PAE ratios were designed and synthesized via a Michael-type step polymerization. The molecular structures of the copolymers were confirmed with (1)H NMR and gel permeation chromatography (GPC). These amphiphilic copolymers were shown to self-assemble into core/shell micelles in aqueous solution at low concentrations, and their critical micelle concentrations (CMC) in water were 1.2-9.5 mg/L. The pH-responsive PAE segment was insoluble at pH 7.4, but it became positively charged and soluble via protonation of amino groups at pH lower than 6.5. The average particle size and zeta potential of micelles increased from 180 nm and 15 mV to 220 nm and 40 mV, respectively, when the pH decreased from 7.4 to 5.0. Doxorubicin (DOX) was loaded into the core of these micelles with a high drug loading of 18%. The in vitro DOX release from the micelles was significantly accelerated when solution pH decreased from 7.4 to 5.0. DOX release in the first 10 h appeared to follow Fickian diffusion mechanism. Toxicity test showed that the copolymers had low toxicity whereas the DOX-loaded micelles remained high cytotoxicity for HepG2 cells. The results indicate the pH-sensitive MPEG-b-(PLA-co-PAE) micelle may be a potential hydrophobic drug delivery carrier for cancer targeting therapy with sustained release.

Abstract:Polymeric prodrugs have attracted great interest in the field of anticancer drug delivery owing to their integrated advantages of prodrugs and nanoparticles. However, the ambiguous chemical composition of polymeric prodrugs is still a major obstacle to their clinical applications. In this study, an amphiphilic poly(ethylene glycol) derivative with multiple thiols was first synthesized on a large scale through the melt co-polycondensation of oligo(ethylene glycol), 1,4-butanediol and mercaptosuccinic acid. Then an adamantane modified doxorubicin (DOX) with a pH-sensitive hydrazone bond and a beta-cyclodextrin derivative with multiple acryloyls were precisely synthesized. Finally, novel supramolecular polymeric prodrug (SPP) micelles with a precise drug molecular structure and drug loading content were facilely prepared using the above three components via simultaneous self-assembly, a thiol-ene "click" reaction and a host-guest supramolecular interaction in water. The resultant pH-sensitive SPP micelles with a stable core-crosslinked structure and strong drug "bonding" show long blood circulation time, effective cellular uptake and fast DOX release in tumor cells, which significantly improved the antitumor efficacy and security compared with free DOX. This work established a promising drug delivery platform for cancer chemotherapy.
Oxime Linkage: a Robust Tool for the Design of Ph-Sensitive Polymeric Drug Carriers.

Abstract:Oxime bonds dispersed in the backbones of the synthetic polymers, while young in the current spectrum of the biomedical application, are rapidly extending into their own niche. In the present work, oxime linkages were confirmed to be a robust tool for the design of pH-sensitive polymeric drug delivery systems. The triblock copolymer (PEG-OPCL-PEG) consisting of hydrophilic poly(ethylene glycol) (PEG) and hydrophobic oxime-tethered polycaprolactone (OPCL) was successfully prepared by aminooxy terminals of OPCL ligating with aldehyde-terminated PEG (PEG-CHO). Owing to its amphiphilic architecture, PEG-OPCL-PEG self-assembled into the micelles in aqueous media, validated by the measurement of critical micelle concentration (CMC). The MTT assay showed that PEG-OPCL-PEG exhibited low cytotoxicity against NIH/3T3 normal cells. Doxorubicin (DOX) as a model drug was encapsulated into the PEG-OPCL-PEG micelles. Drug release study revealed that the DOX release from micelles was significantly accelerated at mildly acid pH of 5.0 compared to physiological pH of 7.4, suggesting the pH-responsive feature of the drug delivery systems with oxime linkages. Flow cytometry and confocal laser scanning microscopy (CLSM) measurements indicated that these DOX-loaded micelles were easily internalized by living cells. MTT assay against HeLa cancer cells showed DOX-loaded PEG-OPCL-PEG micelles had a high anticancer efficacy. All of these results demonstrate that these polymeric micelles self-assembled from oxime-tethered block copolymers are promising carriers for the pH-triggered intracellular delivery of hydrophobic anticancer drugs.
Folic Acid Grafted and Tertiary Amino Based Ph-Responsive Pentablock Polymeric Micelles for Targeting Anticancer Drug Delivery.

Abstract:Increasing target to tumor sites and reducing accumulation at normal tissue sites of anticancer drugs are essential to improve the cancer chemotherapy efficiency. In this study, we have developed a novel pentablock polymeric poly(ethylene glycol)-b-(poly(2-(diethylamino) ethyl methacrylate)-b-poly (hydroxyethyl methacrylate)-g-folic acid)2 [PEG-b-(PDEAEMA-b-PHEMA-g-FA)2] micelles as anticancer drug nanocarrier. The carriers could target tumor cells rapidly, and response to the tumor sites pH to control drug release. The critical micelle concentration (CMC) of the intermediates copolymers was 4.37-7.08mg/L, which indicated that the self-assembled micelles had comparatively good internal circulation stability. The drug loaded micelles were prepared using dialysis method, resulting in an average particle size of below 120nm, and the drug loading content and entrapment efficiency were 21% and 48% respectively. The pH-responsiveness and in vitro drug release of the micelles were studied, and the results showed a higher doxorubicin (DOX) cumulative amount at pH5.0 (~90%) compared to pH7.4 (~20%) owing to the protonation of the tertiary amino groups. In vitro cytotoxicity and endocytosis experiments showed that the tumor-suppressing effect of drug-loaded micelles was close to those of free DOX. The loaded DOX could be delivered into the cancer cells in a short time, and about 80% of the tumor cells were killed after 48h incubation. The results indicate that the pentablock polymeric micelles have the potential to be applied for targeting anticancer drug delivery and control release.

Abstract:In this study, an novel amphiphilic block copolymer P[PEGMA-b-(DEMA-co-APMA)]-FA and its cross-linker uracil-(CH2)(6)-uracil (U-(CH2)(6)-U) were synthesized and used as the targeted and pH-responsive nanocarriers for anticancer drug delivery. The hydrophobic block of the copolymer contains adenine (A) and tertiary amine moieties and the hydrophilic block is terminated with a targeting ligand folic acid (FA). Under neutral pH, the hydrophobic chain segments of the copolymer are cross-linked by U-(CH2)(6)-U through the A-U nucleobase pairing based on complementary multiple hydrogen bonding, and the copolymer forms stable micelles with their mean diameter of around 170 urn in water. While under acidic pH, the micelles dissociate as a result of protonation of tertiary amines and disruption of the A-U nucleobase pairing. Flow cytometry and fluorescent microscope observation show that, when loaded with an anticancer drug DOX, the micelles can preferably enter folate receptor (FR)-positive cancer cells and kill the cells via intracellular release of the anticancer drug. Cytotoxicity tests (MTT tests) indicate that the micelles with FA on their surfaces exhibit higher cytotoxicity toward FR-positive cells than those without FA. This study provides useful insights on designing and improving the applicability of copolymer micelles for other targeted drug delivery systems.
PDEAEMA-based pH-sensitive amphiphilic pentablock copolymers for controlled anticancer drug delivery

Abstract:The synthesis of a series of PDEAEMA-based pH-sensitive amphiphilic pentablock copolymers poly(methyl methacrylate)-b-poly(2-(diethylamino)ethyl methacrylate)-b-poly(ethylene glycol)-b-poly(2-(diethylamino) ethyl methacrylate)-b-poly(methyl methacrylate) [PEG-b-(PDEAEMA-b-PMMA)(2)] with different compositions proceeded via the combination of a bromination reaction and continuous activators regenerated by electron transfer atom transfer radical polymerization (ARGET ATRP). All the copolymers were characterized by H-1 NMR and gel permeation chromatography (GPC). The amphiphilic copolymers can self-assemble into micelles in aqueous solution, and the CMC values were comparatively low (2.40-2.80 mg L-1). The pK(b) buffering region, particle sizes, zeta potentials and optical transmittance were measured to investigate the pH-sensitivity of the polymeric micelles. The size and morphology of the self-assembled blank and DOX-loaded micelles were determined by transmission electron microscopy (TEM) and dynamic light scattering (DLS). The in vitro release rate was sharply increased by decreasing the pH from 7.4 to 5.0, due to the swelling of micelles at lower pH caused by the protonation of tertiary amine groups of PDEAEMA. The in vitro cytotoxicity of DOX-loaded micelles against Hela cells were measured and compared with free DOX, suggesting that the blank micelles provide low cytotoxicity and the DOX-loaded micelles provided high cytotoxicity nearly that of free DOX. The results indicate that this new kind of amphiphilic copolymers could serve as promising nanocarriers for controlled anticancer drug delivery.

Abstract:Novel redox-responsive amphiphilic cationic multi-block copolymers PEG 2000 – PLA 3000 – PEI 1200 – PLA 3000 – PEG 2000 and PEG 2000 – PLA 3000 – PEI 1800 – PLA 3000 – PEG 2000 were synthesized and self-assembled into micelles for co-delivery of siRNA and hydrophobic doxorubicin (DOX). The chemical structure and molecular weight of the copolymers were characterized by 1 H nuclear magnetic resonance and gel permeation chromatography, respectively. The copolymeric micelles were examined by dynamic light scattering, and their size, zeta potential and critical micelle concentration were determined. The in vitro drug release analyses indicated that reductive environment can trigger the release of DOX and siRNA by breaking the micelles. MTT assay demonstrated that the DOX/siRNA-loaded micelles are capable of inhibiting proliferation of SGC7901 cells. The results of fluorescence microscopy and flow cytometry verify the simultaneous delivery of DOX and siRNA from the nanomicellar particles into SGC7901 cells. The reduction-responsive cationic copolymers will provide a platform for constructing drug/gene delivery system toward cancer therapy.
Poly(2-(diethylamino)ethyl Methacrylate)-Based, Ph-Responsive, Copolymeric Mixed Micelles for Targeting Anticancer Drug Control Release

Abstract:We have demonstrated a novel drug delivery system to improve the selectivity of the current chemotherapy by pH-responsive, polymeric micelle carriers. The micelle carriers were prepared by the self-assembly of copolymers containing the polybasic poly(2-(diethylamino) ethyl methacrylate) (PDEAEMA) block. The mixed copolymers exhibited a comparatively low critical micelle concentration (CMC; 1.95-5.25 mg/L). The resultant mixed micelles were found to be <100 nm and were used to encapsulate the anticancer drug doxorubicin (DOX) with pretty good drug-loading content (24%) and entrapment efficiency (55%). Most importantly, the micelle carrier exhibited a pH-dependent conformational conversion and promoted the DOX release at the tumorous pH. Our in vitro studies demonstrated the comparable level of DOX-loaded mixed micelle delivery into tumor cells with the free DOX (80% of the tumor cells were killed after 48 h incubation). The DOX-loaded mixed micelles were effective to inhibit the proliferation of tumor cells after prolonged incubation. Overall, the pH-responsive mixed micelle system provided desirable potential in the controlled release of anticancer therapeutics.
One-pot Synthesis of Ph and ROS Dual Responsive Nanogels for Anti-Cancer Drug Delivery

Abstract:Synthetic polymers that can change their physicochemical properties in response to physiological stimuli have attracted particular interest for constructing smart drug delivery systems. In the present work, a novel oxidation-responsive block copolymer bearing thioether pendants was designed and synthesized by ring-opening alternating copolymerization (ROAP) of phthalic anhydride (PA) and 2-(methylthio)ethyl glycidyl ether (MTG) monomer using mPEG-OH as the macroinitiator and a phosphazene base, t-BuP1, as the catalyst. The obtained mPEG-b-P(PA-alt-MTG) copolymer could self-assemble into spherical micelles in water with an average size of about 30 nm and a critical micelle concentration value of 7.44 × 10−3 mg/mL. 1H NMR results indicated that the hydrophobic thioether group can be selectively oxidized to hydrophilic sulfoxide by H2O2, leading to disassociation of the micelles as illustrated by the TEM observation. Nile Red, as a model drug, was loaded into the copolymer micelles. The intensity of the fluorescence emission decreased sharply after addition of H2O2, indicating an oxidation-triggered drug release. Moreover, the drug-loaded micelles could be effectively internalized into MCF-7 cells, as indicated by the confocal laser scanning microscopy (CLSM) and flow cytometry (FCM) studies. MTT assay further demonstrated that the synthesized copolymer showed good biocompatibility towards L929 cell, even at a concentration up to 0.5 mg/mL. In sum, this kind of copolymer may have great potential applications in oxidation responsive drug delivery.

Abstract:The cancer drug delivery process involves a series of biological barriers, which require the nanomedicine to exhibit different, even opposite properties for high therapeutic efficacy. The prevailing design philosophy, i.e., integrating these properties within one nanomedicine via on-demand property transitions such as PEGylation/dePEGylation, complicates nanomedicines' composition and thus impedes clinical translation. Here, polyzwitterionic micelles of poly(tertiary amine-oxide)-block-poly(ε-caprolactone) (PTAO-PCL) amphiphiles that enable all the required functions are presented. The zwitterionic nature and unique cell membrane affinity confer the PTAO micelles long blood circulation, efficient tumor accumulation and penetration, and fast cellular internalization. The mitochondrial targeting capability allows drug delivery into the mitochondria to induce mitochondrial dysfunction and overcome tumor multidrug resistance. As a result, the PTAO/drug micelles exhibit potent anticancer efficacy. This simple yet multipotent carrier system holds great promise as a generic platform for potential clinical translation.

Abstract:The pH-triggered biodegradable micelles based on poly(beta-amino ester)-graft-12-acryloyloxy dodecyl phosphorylcholine (PAE-graft-ADPC) were explored as an efficient acid-triggered anticancer drug delivery system. A pH-responsive biodegradable polymer was synthesized by using hydrophilic 12-acryloyloxy dodecyl phosphorylcholine (ADPC) and poly(beta-amino ester) (PAE) via Michael addition reaction. Micelles with phosphorylcholine shells were prepared in aqueous solution and doxorubicin (DOX) was loaded into the core of the micelles with high efficiency (73%). Dynamic light scattering (DLS) and transmission electron microscopy (TEM) demonstrated that the size of the drug-loaded micelles was about 230 nm. DOX-loaded micelles were stable in the physiological environment, whereas they disassembled at pH 6.5 which is similar to the extracellular pH of most tumors. In vitro drug release showed that DOX release from micelles was faster at pH 6.5 than at 7.4. Intracellular uptake demonstrated that DOX was delivered into the cells effectively after the cells were incubated with DOX-loaded micelles at pH 6.5. Moreover, an in vitro cytotoxicity study indicated that the growth of HepG2 cells was inhibited remarkably when the cells were preincubated with DOX-loaded micelles at pH 6.5. The results indicated that the pH-triggered biodegradable micelles could serve as promising drug carriers for cancer therapy.
pH-sensitive polycarbonate micelles for enhanced intracellular release of anticancer drugs: a strategy to circumvent multidrug resistance

Abstract:In this study, we have synthesized two novel amphiphilic diblock copolymers with aldehyde groups via organocatalytic ROP of a functionalized cyclic carbonate monomer (MTC-Bz) using polyethylene oxide (PEG) as the macroinitiator. The polymers were covalently conjugated with an anti-tumor drug doxorubicin (DOX) via a pH-sensitive Schiff-base linkage. The resulting conjugates formed micelles in phosphate-buffered saline (PBS) (pH 7.4) with an average size of about 100 nm and narrow size distribution. The surface charge of the micelles was close to zero. The micelles were stable in both PBS and cell culture media containing 10% FBS up to 5 days. The results obtained from the in vitro release study indicated that DOX release from the micelles was pH-dependent, being faster at pH 5.0 (the endolysosomal environment) than pH 7.4 (the extracellular environment). Human breast cancer MCF-7 cells and DOX-resistant MCF-7/Adr cells were employed to investigate the cellular uptake and cytotoxicity of DOX-loaded micelles. The confocal microscopy and flow cytometry studies showed that the uptake of DOX-loaded micelles by MCF-7 cells was similar to that of free DOX. In sharp contrast, the uptake of DOX-loaded micelles by MCF-7/Adr cells was significantly higher than that of free DOX. The polymers showed no toxicity to MCF-7 and MCF-7/Adr cells. The DOX-loaded micelles killed the cells efficiently. In particular, they were more potent against drug-resistant MCF-7/Adr cells than free DOX due to the higher cellular drug accumulation and pH-triggered intracellular drug release, providing a strategy to navigate around drug resistance. These DOX-conjugated micelles can be a promising carrier for the delivery of anticancer drugs with amine functional groups.

Abstract:In the present study, an amphiphilic polymer was prepared by conjugating methoxy poly(ethylene glycol) (mPEG) with tetraphenylethene (TPE) via disulfide bonds (Bi(mPEG-S-S)-TPE). The polymer could self-assemble into micelles and solubilize hydrophobic anticancer drugs such as paclitaxel (PTX) in the core. Combining the effect of TPE, mPEG, and disulfide bonds, the Bi(mPEG-S-S)-TPE micelles exhibited excellent AIE feature, reduced protein adsorption, and redox-sensitive drug release behavior. An in vitro intracellular uptake study demonstrated the great imaging ability and efficient internalization of Bi(mPEG-S-S)-TPE micelles. The excellent anticancer effect and low systemic toxicity were further evidenced by the in vivo anticancer experiment. The Bi(mPEG-S-S)-TPE micelles were promising drug carriers for chemotherapy and bioimaging.

Abstract:The surface/interface state of nanomaterials plays a key role on their biomedical applications. Nanotechnology offers a versatile means to develop nanoparticles with well-defined architecture. In this study, mesoporous silica nanoparticles were firstly loaded with an anticancer drug (doxorubicin, DOX), which were then decorated with a cationic oligomer (low molecular weight polyethyleneimine, LPEI) to acquire an increased surface charge. The resulting particles were further self-assembled with negative charged bovine serum albumin (BSA) as natural protein nanoblocks to offer surface charge tunability. The resulting mesoporous nanohybrids (MDPB) acquired charge-reversal ability, which presented negative charge under biological conditions (beneficial to biocompatibility), while displaying a positive charged state under acidic conditions mimicking the tumor extracellular microenvironment (favoring cell uptake or tumor penetration). Furthermore, the nanohybrids not only allowed for an effective loading of DOX drug, but also accelerated its release under acidic tumor microenvironments in a sustainable way. In vitro biological study indicated that the DOX-free nanoparticles were biocompatible, while MDPB exerted good cytotoxicity against cancer cells, suggesting their promise for therapeutic delivery application.

Abstract:A new type of salt/pH dual-responsive micelles based on supramolecular amphiphilic brush copolymers, poly(2-hydroxyethyl metharylate)-g-(poly(epsilon-caprolactone)-adenine:uracil-poly(ethylene glycol)) (PHEMA-g-(PCL-A:U-PEG)) was developed for the anticancer drug delivery owing to the fact that the tumor tissues show low pH and high salt concentration. The supramolecular structure of brush copolymer was confirmed by variable-temperature H-1 NMR and Fourier transform infrared spectroscopy (FTIR). Doxorubicin (DOX) as a model anticancer drug was efficiently loaded into the supramolecular micelles (up to 70%) due to the compact structure of brush polymer. The cumulative release profile of the DOX-loaded micelles showed a low level of drug release (about 20 wt% in 25 h) at pH 7.4 with a low salt concentration, and was significantly accelerated at a lower pH (5.0) and a high salt concentration (over 70 wt% in 6 h), exhibiting an salt/pH dual-responsive controlled drug release capability. Methyl tetrazolium (MTT) assay showed that DOX-loaded micelles had high anticancer efficacy against Hela cancer cells and blank micelles had a very low cytotoxicity. These supramolecular brush copolymer micelles possess many favorable traits, such as low cytotoxicity and excellent biodegradability, adequate drug loading capacity, and rapid drug release in response to the intracellular level of pH and salt concentration, which endow them as a promise candidate for delivering anticancer drugs.

Abstract:The therapeutic efficacy of anticancer drug delivery systems is severely truncated by their low tumor target ability and inefficient drug release at the target site. Herein, we report the design and synthesis of a novel dual-responsive hyaluronic acid-grafted poly(ionic liquid) block copolymer that was able to self-assemble into spherical nanoparticles in water. Then, the doxorubicin-loaded block copolymer produced particles of 100 nm in size as examined using transmission electron microscopy and dynamic light scattering. The hyaluronic acid-grafted block copolymer micelles were efficiently taken up by CD44-overexpressing cancer cells as indicated by the cell counting kit-8 assays, confocal laser scanning microscopy, and flow cytometry experiments. Therefore, the as-prepared block copolymer micelles had good biocompatibility, low toxicity, and high selectivity, thus offering promise for the treatment of various CD44-overexpressing cancers.

Abstract:Stimuli-responsive drug delivery systems (DDSs) are expected to realize site-specific drug release and kill cancer cells selectively. In this study, a pH-responsive micelle was designed utilizing the pH-sensitivity of borate bonds formed between dopamine and boronic acid. First, methyl (polyethylene glycol)-block-polycaprolactone (mPEG-PCL) was conjugated with 4-cyano-4-(thiobenzoylthio)pentanoic acid (CTP) to obtain a macroinitiator. Two different segments poly(dopamine methacrylamide) (PDMA) and poly(vinylphenylboronic acid) (PVBA) were then grafted to the end of mPEG-PCL. Two triblock copolymers, mPEG-PCL-PDMA and mPEG-PCL-PVBA, were then obtained by reversible addition-fragmentation transfer (RAFT) polymerization. These copolymers and their mixture self-assembled in aqueous solution to form micelles that were able to load hydrophobic anticancer drug doxorubicin (DOX). These two-component micelles were found to be pH-sensitive, in contrast to the one-component micelles. Furthermore, MTT studies showed that the micelles were almost nontoxic. The DOX-loaded micelles showed cytotoxicity equivalent to that of DOX at high concentration. In vivo antitumor experiments showed that this pH-sensitive polymeric micellar system had an enhanced therapeutic effect on tumors. These two-component boronate-based pH micelles are universally applicable to the delivery of anticancer drugs, showing great potential for cancer therapy.

Abstract:Nano-drug carriers have been widely used in chemotherapy to improve the efficacy of chemotherapeutic drugs. Polymer nano carriers have more potential to enrich the chemotherapeutic drugs in the target tumor site and controlled release. Herein, a series of amphiphilic copolymers (ITT) were synthesized by melt polymerization with the ROS-cleavable thioketal cross-linker, imidazole groups and polyethylene glycol conjugated triphenylphosphonium (PEG-TPP) segments, the proportions of the actual imidazole groups to PEG-TPP segments for which were 5.2:1, 2.6:1 and 1.8:1, respectively. The amphiphilic copolymers could self-assemble into cationic micelles with hydrophobic imidazole groups as core and hydrophilic PEG-TPP segments as shell in deionized water. The cationic micelles with an average particle size of 124.6-187.3 nm showed a drug loading content (DLC) of 5.65%-6.10%. The in vitro release studies revealed the pH/ROS dual-responsibility of DOX-loaded ITT micelles,and the release behavior of DOX from micelles strongly depended on the content of imidazole groups in ITT micelles. Typically, the release of encapsulated DOX were 27.2%, 30.0% and 34.1% from DOX-loaded ITT-1 micelles, in PBS (pH 6.5) containing 0, 100, and 500 μM H2O2, respectively. The release of encapsulated DOX were 20.3%, 27.2% and 42.5% from DOX-loaded ITT-1 micelles, in PBS (pH 7.4), PBS (pH 6.5) and PBS (pH 5.0), respectively. DOX-loaded micelles effectively inhibited the growth of 4T1 cells through being internalized into 4T1 cells, and releasing DOX into the mitochondrion, with the cell viability was less than 65% when DOX concentration was 2.5 μg·mL-1. This work presented a novel mitochondrial targeting and pH/ROS responsive polymer nanocarrier for potential chemotherapy.

pH-Sensitive Dextran-Based Micelles from Copper-Free Click Reaction for Antitumor Drug Delivery

Abstract:There remains a need to develop new strategies to fabricate dextran-based biocompatible drug delivery systems for safe and effective chemotherapy. Herein, a copper-free azide-propiolate ester click reaction was introduced for dextran modification to fabricate a pH-sensitive dextran-based drug delivery system. A pH-sensitive dextran-based micelle system, self-assembled from amphiphilic dextran-graft-poly(2-(diisopropylamino)ethyl methacrylate-co-2-(2',3',5'-triiodobenzoyl)ethyl methacrylate) or dextran-g-P(DPA-co-TIBMA), is reported for effective chemotherapy. The amphiphilic dextran-g-P(DPA-co-TIBMA) was prepared via reversible addition-fragmentation chain-transfer (RAFT) polymerization and copper-free azide-propiolate ester click reaction. Doxorubicin (DOX)-loaded dextran-g-P(DPA-co-TIBMA) micelles were prepared through self-assembly of DOX and dextran-g-P(DPA-co-TIBMA) in aqueous solution, and had a mean diameter of 154 nm and a drug loading content of 9.7 wt %. The release of DOX from DOX-loaded dextran-g-P(PDPA-co-TIBMA) micelles was slow at pH 7.4, but was greatly accelerated under acidic conditions (pH 6 and 5). Confocal laser scanning microscopy and flow cytometry experiments showed that the dextran-g-P(DPA-co-TIBMA) micelles could effectively deliver and release DOX in human breast cancer cell line (MCF-7 cells). MTT assay showed that dextran-g-P(DPA-co-TIBMA) exhibited excellent biocompatibility while DOX-loaded dextran-g-P(DPA-co-TIBMA) micelles have good antitumor efficacy in vitro. The in vivo therapeutic studies indicated that the DOX-loaded dextran-g-P(PDPA-co-TIBMA) micelles could effectively reduce the growth of tumor with little body weight reduction.

Abstract:In this work, well-defined pH-responsive methoxy poly(ethylene glycol)-block-poly(e-caprolactone)-block-poly[2-(dimethylamino)ethyl methacrylate]-g-7-propinyloxy coumarin triblock amphiphilic copolymers (mPEG-b-PCL-b-PDMAEMA-g-PC) were synthesized using a combination of atom transfer radical polymerization (ATRP), ring opening polymerization (ROP) and click chemistry. The chemical structures and compositions of these copolymers were characterized using Fourier transform infrared spectroscopy (FT-IR) and proton nuclear magnetic resonance (1H NMR). The molecular weights of the copolymers were obtained using 1H NMR spectroscopy and gel permeation chromatography (GPC) measurements. Subsequently, the polymers could self-assemble into micelles which were investigated using dynamic light scattering (DLS), transmission electron microscopy (TEM), and fluorescence spectroscopy. The pH-responsive self-assembly behavior of these triblock copolymers in water were investigated at different pH values of 5 and 7.4 for controlled doxorubicin release, the results indicated that the release rate of DOX could be effectively controlled by altering the pH, DOX was sealed in neutral surroundings and DOX release was triggered in acidic surroundings. CCK-8 assays and confocal laser scanning microscopy (CLSM) against HeLa cells indicated that the micelles had no associated cytotoxicity, possessed good biodegradability and biocompatibility, and identified the location of the DOX in HeLa cells. The DOX-loaded micelles possessed high cytotoxicity to HeLa cells and exhibited inhibition of the proliferation of HeLa cells. Moreover, these flexible micelles with an on–off switched drug release may offer a promising pattern to deliver a wide variety of hydrophobic payloads to tumor cells for cancer therapy.
