Abstract:Mechanical forces play crucial role in biology by regulating several cellular properties, such as morphology, proliferation, migration, and their interaction with the surrounding microenvironment. Precise mechano-regulation is important since any distortion may lead to changes in the cellular behavior and result in diseases such as cancer. Cell adhesion molecules such as integrins and cadherins are essential elements of these mechanotransduction processes at cell-cell junctions. Piconewton-level molecular tensile forces are normally associated with these adhesion molecules. However, it has been challenging to precisely measure these forces, especially considering the complexity of the junction environment. In our lab, we have developed a type of DNA-based tension probe that uses a DNA hairpin module to sense the force and a lipid anchor to modify onto live-cell membranes. Here, we demonstrate that by monitoring changes in the fluorescence lifetime, we can quantitatively image the tensile force distributions of multiple pairs of ligand-receptor at cell-cell junctions. Lifetime imaging is advantageous since it is independent on the concentration of the probe and is not limited by the spectral width of visible light. The ability of the probe to accommodate multiple hairpins further enables the opportunity to sense a large range of forces at the same time. After validating this novel tension probe by imaging E-cadherin-mediated forces between adjacent epithelial cells, we have further used the probe to monitor the mechanical dynamics of epithelial-to-mesenchymal transition, a key biological process that controls cellular migration and metastatic properties. We believe these fluorescence lifetime-based tension probes will be a widely useful tool for investigating the role of intercellular mechanical stimuli on regulating cellular behavior.
