Abstract:Background: Cysteine (Cys) is the major intracellular thiol and plays a key role in human pathology. Furthermore, endogenous sulfur dioxide (SO2) is produced in mammals. Abnormal levels of SO2 are commonly associated with a variety of respiratory, cardiovascular, and neurological diseases. Therefore, given their important role in life activities, it is significant to construct a fluorescent probe that can detection between Cys and SO2. Results: We have designed and synthesized a two-site fluorescent probe CUM with coumarin derivative and benzaldehyde molecules, which can detect and differentiate between Cys and SO2 through dual excitation wavelengths. Its carbon-carbon double bond reacts with Cys and undergoes a nucleophilic reaction, emitting green fluorescence at 520 nm, while SO32- reacts with benzaldehyde molecules in the probe CUM and undergoes a blue fluorescence at 460 nm. SO32- reacts with the benzaldehyde molecule of probe CUM and fluoresces blue at 460 nm. Thus, the probe CUM with two reaction sites can distinguish between Cys and SO2 and shows good selectivity and fast reaction speed. In addition, we successfully utilized probe CUM to image Cys and SO2 in human breast cancer cells (MDA-MB-231). Significance: This work provides an effective method for the molecular design of coumarin-based fluorescent probes. Probe CUM as a promising and reliable tool for the meticulous discrimination and quantification of Cys and SO2 in diverse biological matrices, thereby opening up new avenues for various biological systems.

Abstract:Differentiation of biologically important thiols, such as cysteine (Cys), homocysteine (Hcy), and glutathione (GSH) is still a challenging task. Herein, we present a novel fluorescent chemodosimeter capable of selectively detecting Cys over other biothiols including Hcy and GSH and other amino acids by a facile thiol-Michael addition/transcyclization rearrangement cascade click process. The unique trans-cyclization step is critical for the selectivity as a result of the kinetically favorable formation of a six-membered ring with the Cys Michael adduct. Moreover, the probe adopts a dis-tinctive dual quenching mechanism-photoinduced electron transfer (PET) and photoinduced intramolecular charge transfer (ICT) to deliver a drastic turn-on fluorescence response only at the Cys-selective transcylization step. The judicious selection of strong electron-withdrawing naphthalimide fluorophore with maleimide group enhances the electrophilicity and thus reactivity for the cascade process leading to fast detection and ultrasensitivity with a detection limit of 2.0 nm (S/N=3). The probe has demonstrated its practical utility potential in Cys imaging in live cells.
Upconversion-based chiral nanoprobe for highly selective dual-mode sensing and bioimaging of hydrogen sulfide in vitro and in vivo

Abstract:Chiral assemblies have become one of the most active research areas due to their versatility, playing an increasingly important role in bio-detection, imaging and therapy. In this work, chiral UCNPs/CuxOS@ZIF nanoprobes are prepared by encapsulating upconversion nanoparticles (UCNPs) and CuxOS nanoparticles (NPs) into zeolitic imidazolate framework-8 (ZIF-8). The novel excited-state energy distribution-modulated upconversion nanostructure (NaYbF4@NaYF4: Yb, Er) is selected as the fluorescence source and energy donor for highly efficient fluorescence resonance energy transfer (FRET). CuxOS NP is employed as chiral source and energy acceptor to quench upconversion luminescence (UCL) and provide circular dichroism (CD) signal. Utilizing the natural adsorption and sorting advantages of ZIF-8, the designed nanoprobe can isolate the influence of other common disruptors, thus achieve ultra-sensitive and highly selective UCL/CD dual-mode quantification of H2S in aqueous solution and in living cells. Notably, the nanoprobe is also capable of in vivo intra-tumoral H2S tracking. Our work highlights the multifunctional properties of chiral nanocomposites in sensing and opens a new vision and idea for the preparation and application of chiral nanomaterials in biomedical and biological analysis.
Chiral MoSe2 Nanoparticles for Ultrasensitive Monitoring of Reactive Oxygen Species In Vivo

Abstract:Reactive oxygen species (ROS) are involved in neurodegenerative diseases, cancer, and acute hepatitis, and the early diagnosis of these diseases by quantification of ROS is critical for the curable treatment of patients. Here, a novel probe was developed based on chiral molybdenum diselenide (MoSe2) nanoparticles (NPs) modified by the fluorescence molecule, cyanine 3 (Cy3). Chiral MoSe2 NPs showed intensive circular dichroism (CD) signals at 390 nm and 550 nm, whereas the fluorescence signal of Cy3 at 560 nm was quenched by MoSe2 NPs. With the presence of ROS, the novel probe reacted with ROS and then oxidated rapidly, resulting in decreased CD signals and the recovery of Cy3 fluorescence signals. With the help of ROS‐related CD and fluorescent dual‐mode signals, the limit of detection (LOD) of CD signals and fluorescence signals in living cells was 0.0093 nmol/106 cells and 0.024 nmol/106 cells, respectively. The high selectivity and sensitivity to ROS in complex biological environments was attributed to the Mo4+ and Se2− oxidation reaction on the surface of the NPs. Furthermore, chiral MoSe2 NPs was able to monitor the levels of ROS in vivo by fluorescence signals. Collectively, this strategy offers a new approach for ROS detection and has the potential to inspire others to explore chiral nanomaterials as potential biosensors to investigate biological events. Abstract:A fluorescent probe has been designed using cyanine phenothiazine and 7-nitro-1,2,3-benzoxadiazole (NBD) for selective detection of Cys-SO2 components. The probe utilizes the NBD structure to achieve specificity towards Cys and employs a reaction mechanism between the double bond of cyanine and phenothiazine with SO32- to achieve selectivity towards SO2. Importantly, the NBPI phenothiazine structure incorporates a large C-O bond energy attached to NBD, effectively eliminating interference from Hcy and ensuring highly selective response to Cys. The optimized design of the probe enables excellent linearity and extremely low detection limits for Cys-SO2 components in vitro experiments. The probe NBPI allows separate detection of Cys-SO2 in the presence of both components. Furthermore, the probe NBPI demonstrated successful imaging of endogenous and exogenous Cys and SO2 in cell studies.

Abstract:The hemicyanine hybrid containing the 7-(diethylamino)coumarin (ACou) donor attached to the cationic indolenium (Ind) acceptor through a vinyl linkage (ACou-Ind) represents a classic ratiometric fluorescent probe for detecting nucleophilic analytes, such as cyanide and reactive sulfur species (RSS), through addition reactions that disrupt dye conjugation to turn off red internal charge transfer (ICT) fluorescence and turn on blue coumarin emission. The chemosensing mechanism for RSS detection by ACou-Ind suggested in the literature has now been revised. Our studies demonstrate that thiolates react with ACou-Ind through conjugate addition to afford C4-SR adducts that lack coumarin fluorescence due to photoinduced electron transfer quenching by the electron-rich enamine intermediate. Thus, ACou-Ind serves as a turn-off probe through loss of red ICT fluorescence upon RSS addition. The literature also suggests that blue coumarin emission of thiolate adducts is enhanced in the presence of reactive oxygen species (ROS) due to ROS-mediated cellular changes. Our studies predict that such a scenario is unlikely and that thiolate adducts undergo oxidative deconjugation in the presence of H2O2, the pervasive ROS. Under basic conditions, H2O2 also reacts directly with ACou-Ind to generate intense coumarin fluorescence through an epoxidation process. The relevance of our chemosensing mechanism for ACou-Ind was assessed within live zebrafish, and implications for the utility of ACou-Ind for unraveling the interplay between RSS and ROS are discussed.

Abstract:A novel dual-mode cholesterol biosensor was developed based on gold nanorods-carbon quantum dots-Cholesterol oxidase (AuNRs-CQDs-COD) complex synthesized by the cross-linking method. The specific reaction to/against cholesterol and cholesterol oxidase can produce hydrogen peroxide. Due to the etching effect between the hydrogen peroxide and AuNRs, the absorption peaks of AuNRs would drift so that the fluorescence quenching occurred because of the electronic charge transfer between the hydrogen peroxide and CQDs. The spectrum properties, structure and micromorphology of AuNRs-CQDs-COD complex were characterized by transmission electron microscope (TEM), Fourier transform infrared (FTIR), Fluorescence and Ultraviolet–Visible absorption spectroscopy, respectively. There was a good linear correlation between the fluorescence response and absorption peak shift of AuNRs-CQDs-COD probe in cholesterol solution within the concentration in the range 0.1–1 mM, and the limit of detection (LOD) is 0.028 μM. This dual-mode cholesterol biosensor based on AuNRs-CQDs-COD complex has good anti-interference and selectivity.

Abstract:A practical strategy of introducing ortho-methoxyl group was explored to achieve the fluorescence-enhancing and bathochromic-shift bi-functional optimization. It was tested in the Cys sensing ISOPH-X series, thus the successful case, ISOPH-2, was obtained. It realized the optimization in a simple and compatible way. The corresponding strategy was basically established during the confirmation of checkpoints including applicable steadiness (over 24 h), wide pH range (7.0-9.0), rapid response (20 min), good biocompatibility, high sensitivity (LOD = 0.072 nm), high selectivity and biological monitoring of Cys in living cells as well as C. elegans. In this work, the o-methoxyl introduction strategy led to a 15 nm red shift and a near 4-fold fluorescence enhancement. This strategy could be combined with the double bond-introducing approach. Compared with reported strategies, by breaking the dilemma between red shift and strong fluorescent intensity, this strategy might offer beneficial information for exploiting better sensors with more fluorophores and mechanisms for their targets.

Abstract:HClO and H2S, as two kinds of crucial small biomolecules, are endowed various roles in biological organisms. The redox balance between HClO and H2S is closely related to the physiological and pathological processes. Thus, it is significant to monitor the redox process between HClO and H2S. Inspired by the advantages of ratiometric fluorescent probes, we firstly developed a reversible ratiometric fluorescent probe (BT-Se) for HClO and H2S via combination of phenyl selenide as the response group and 2-(2'-hydroxyphenyl)benzothiazole dye as the fluorophore. The proposed probe BT-Se could detect HClO with well-separated dual emission (110 nm), fast response, good selectivity and sensitivity owing to the oxidation reaction of the Se atom induced by HClO. Moreover, only H2S could effectively recover the fluorescence of the detection system to the original state via H2S induced-reduction of selenoxide. Cell imaging studies demonstrated that the probe BT-Se was capable of ratiometric monitoring the changes of intracellular HClO/H2S, which suggested that it has great potential for researching the biological functions of HClO and H2S.

Abstract:The quantification of cysteine (Cys) levels in the organisms holds paramount significance in biological research and disease diagnosis, which can give the correlation between abnormal Cys levels and diseases. In this study, two fluorescent probes, designated as DEA-OH and DEA-AC, featuring a coumarin-aurone backbone specifically engineered for Cys detection, were meticulously designed and synthesized. The diethylamino coumarin-aurone probe DEA-OH and the acrylate-substituted probe DEA-AC demonstrated remarkable sensitivity in detecting cysteine by means of copper displacement (DEA-OH) and acrylate hydrolysis mechanisms (DEA-AC) with fluorescence detection limits of 7.25 μM and 1.65 μM, respectively. Moreover, the fluorescence peak wavelength of the two probes displayed a linear relationship with solvent polarity in the ET (30) range of 30-65 kcal•mol-1, indicating the potential for monitoring changes in environmental polarity within this ET (30) range. The outstanding attributes exhibited by DEA-AC including superior photostability, remarkable selectivity, and swift response (kinetic rate constant: 0.00747 s-1), coupled with the exceptional anti-interference ability, have significantly broadened its scope of applications, for example detecting alterations in Cys within biological systems.
Enhanced Sulfite-Selective Sensing and Cell Imaging with Fluorescent Nanoreactors Containing a Ratiometric Lipid Peroxidation Sensor

Abstract:The detection of SO2 and its derivatives is indispensable for monitoring atmospheric, water quality, and biological fluctuation of oxidative stress and metabolism of biothiols within native cellular contexts. In this article, the brush copolymer nanoreactors containing amine-terminated PDMS were used to encapsulate the fluorescent indicator C11-BDP, forming sulfite-sensitive nanoreactors (ssNRs). Surprisingly, the ssNRs were found to be highly selective to sulfite over a range of reactive oxygen/nitrogen/sulfur species and anions, which was not observed with freely dissolved indicators. The ssNRs showed a rapid response (t95 = 65 s), an excellent detection limit (0.7 μM), and a very high sensitivity (ca. 1000-fold ratiometric intensity change) to sulfite. For cellular studies, the ssNRs exhibited negligible toxicity and could be endocytosed into endosomes and lysosomes. Dual-emissive fluorescence measurements of hydroxyl radicals using a coumarin-activated silica nanohybrid probe

Abstract:This work reports a novel dual-emissive fluorescent probe based on dye hybrid silica nanoparticles for ratiometric measurement of the hydroxyl radical (˙OH). In the probe sensing system, the blue emission of coumarin dye (coumarin-3-carboxylic acid, CCA) immobilized on the nanoparticle surface is selectively enhanced by ˙OH due to the formation of a coumarin hydroxylation product with strong fluorescence, whereas the emission of red fluorescent dye encapsulated in the silica nanoparticle is insensitive to ˙OH as a self-referencing signal, and so the probe provides a good quantitative analysis based on ratiometric fluorescence measurement with a detection limit of 1.65 μM. Moreover, the probe also shows high selectivity for ˙OH determination against metal ions, other reactive oxygen species and biological species. More importantly, it exhibits low cytotoxicity and high biocompatibility in living cells, and has been successfully used for cellular imaging of ˙OH, showing its promising application for monitoring of intracellular ˙OH signaling events.

Abstract:Butyrylcholinesterase (BChE) mainly contributing to plasma cholinesterase activity is an important indicator for routinely diagnosing liver function and organophosphorus poisoning in clinical diagnosis, but its current assays are scarce and frequently suffer from some significant interference and instability. Herein, we report a redox-controlled fluorescence nanoswtich based on reversible disulfide bonds, and further develop a fluorometric assay of BChE via thiol-triggered disaggregation-induced emission. Thiol-functionalized carbon quantum dots (thiol-CQDs) with intense fluorescence is found to be responsive to hydrogen peroxide, and their redox reaction transforms thiol-CQDs to nonfluorescent thiol-CQD assembly. The thiols inverse this process by a thiol-exchange reaction to turn on the fluorescence. The fluorescence can be reversibly switched by the formation and breaking of disulfide bonds caused by external redox stimuli. The specific thiol-triggered disaggregation-induced emission enables us to assay BChE activity in a fluorescence turn-on and real-time way using butyrylthiocholine iodide as the substrate. As-established BChE assay achieves sufficient sensitivity for practical determination in human serum, and is capable of avoiding the interference from micromolar glutathione and discriminatively quantifying BChE from its sister enzyme acetylcholinesterase. The first design of reversible redox-controlled nanosiwtch based on disulfide expands the application of disulfide chemistry in sensing and clinical diagnostics, and this novel BChE assay enriches the detection methods for cholinesterase activity.

Abstract:Hydrogen peroxide (H2O2) and hydrogen sulfide (H2S) play a key role in many cellular signaling pathways. The efficient and accurate in situ detection of H2O2 and H2S released from living cells has attracted extensive research interests. Herein, we presented a portable colorimetric and electrochemical dual-mode sensor for detection of cellular secreted H2O2 and H2S based on MOF-818 nanozyme. MOF-818 containing trinuclear copper centers not only possesses excellent peroxidase-like activity under 37 °C and acidic conditions, but also exhibits outstanding electrocatalytic reduction activity to H2O2 (reduction peak potential at −0.08 V vs Ag/AgCl). Meanwhile, the copper sites of MOF-818 can be coordinated with sulfide, reducing the catalytic activity of MOF-818. As a result, the colorimetric and electrochemical sensing of H2O2 and H2S were established based on “signal-on” and “signal-off” strategies, respectively. Additionally, the combination of a smartphone and a miniature electrochemical analyzer enables dual-mode colorimetric and electrochemical sensing platform for in situ detection of H2O2 and H2S released from live cells. This portable dual-mode sensor based on MOF nanozyme not only opens a new path for point-of-care testing, but also has potential applications in the field of cell biology and clinical diagnosis.
