Abstract:Chiral supramolecules have great potential for use in chiral recognition, sensing, and catalysis. Particularly, chiral supramolecular biocoordination polymers (SBCPs) provide a versatile platform for characterizing biorelated processes such as chirality transcription. Here, we selectively synthesize homochiral and heterochiral SBCPs, composed of chiral naphthalene diimide ligands and Zn ions, from enantiomeric and mixed R-ligands and S-ligands, respectively. Notably, we find that the chiral self-sorted SBCPs exhibit multifunctional properties, including photochromic, photoluminescent, photoconductive, and chemiresistive characteristics, thus can be used for various sensors. Specifically, these materials can be used for detecting hazardous amine materials due to the electron transfer from the amine to the SBCP surface and for enantioselectively sensing a chiral species naproxen due to the different binding energies with regard to their chirality. These results provide guidelines for the synthesis of chiral SBCPs and demonstrate their versatility and feasibility for use in various sensors covering photoactive, chemiresistive, and chiral sensors.

Abstract:Herein we report that few-atom silver nanoclusters (Ag NCs) can be effective energy acceptors for upconversion phosphors (UCPs). A luminescence resonance energy transfer (LRET) probe for biothiols was constructed by decorating UCPs with dithiol-stabilized Ag NCs. Owing to the unique properties of ultrasmall NCs, properties which bridge the gap between those of small molecules and those of nanoparticles, the use of approximately 1.9 nm Ag NCs as energy acceptors endows the probe with high energy-transfer efficiency, good biocompatibility, and flexibility. The UCP-Ag NC nanoprobe enables rapid and robust target assay in solutions. It was also uploaded into living cells and used to detect intracellular biothiol levels with high discrimination. Moreover, the probe shows transportability in vivo and can be used for tissue imaging. The facile growth of few-atom metal NCs on diverse templates may enable the development of various nanoprobes combining UCPs and metal NCs.

Abstract:Chiral inorganic nanomaterials have emerged as a highly promising area of research in nanoscience due to their exceptional light–matter interaction and vast potential applications in chiral sensing, asymmetric catalysis, enantiomer separation, and negative-index materials. We present an overview of the latest advances in chiral inorganic nanomaterials including chiral individual nanoparticles, chiral assemblies, and chiral film-based sensors over the past ten years. Additionally, we discuss the challenges and future perspectives for developing chiral nanomaterials in biosensing applications.

Abstract:Merocyanines adsorbed into the mesopores of mSiO2 shell of NaYF4: 20% Yb, 2% Er, 0.2% Tm nanocrystals are demonstrated as ratiometric upconversion luminescence (UCL) probe for highly selective detection of HS(-) in living cells through inhibition of energy transfer from the UCL of the nanocrystals to the absorbance of the merocyanines. Abstract:A highly sensitive and selective upconversion near-infrared (NIR) fluorescence and colorimetric dual readout hydrogen sulfide (H2S) nanoprobe was constructed based on the excellent NIR fluorescence emission performance of upconversion nanomaterials (UCNPs), the specific recognition effect of synergistically synthesized gold nanoflowers (trypsin-stabled AuNFs (Try-AuNFs)) and the effective NIR fluorescence quenching capability. In this assay, the sensing strategy included three processes. First of all, the synthesized UCNPs can emit 803 nm NIR fluorescence when they were excited by 980 nm excitation light. Secondly, as a result of the principle of fluorescence resonance energy transfer (FRET), Try-AuNFs can effectively quench the NIR fluorescence of UCNPs at 803 nm, which can effectively improve the signal-to-background ratio of nanoprobes, thereby improving the sensitivity of the probes. Thirdly, in the presence of H2S, the Try protective layer on the surface of Try-AuNFs was specifically penetrated, which will subsequently cleave Try-AuNFs via the strong S-Au bond. As such, the NIR fluorescence of UCNPs will be restored, achieving high selectivity and sensitivity detection of H2S. Under optimized conditions, the linear response range of H2S was 0.1-300 μM, and the detection limit was 53 nM. It is worth noting that the Try on the surface of Try-AuNFs via the synergistic effect can increase the steric hindrance of the probe, and this can effectively prevent the interaction between the probe with biothiols (cysteine (Cys), homocysteine (Hcy)) and other natural amino acids (non-thiol-containing) with resultant in the high selectivity regarding the detection of H2S in human serum, which is unlikely to be achieved by AuNFs synthesized by the gold seed method (Se-AuNFs). This work not only provided a new type of UCNPs fluorescence quencher and recognition unit, but also exemplified that the use of the physical properties (steric hindrance) of protein ligands on the surface of nanoflowers can improve the specificity of the probe. This will provide new ideas for the design of other nanoprobes.

Abstract:The core-shell NaGdF4:Yb,Er@NaGdF4:Yb,Nd upconversion nanoparticles (UCNPs) were successfully obtained with the method of co-precipitation, and the water-solubility of UCNPs was improved by the ligand exchange reaction between nitrosyl tetrafluoroborate (NOBF4) and nanoparticles. The IR-783 dye with negative charge and NOBF4-UCNPs with positive charge can bind together by electrostatic action to sensitize UCNPs through the energy transfer from IR-783 to UCNPs. However, with the presence of Na2S (a commonly used H2S donor), a highly selective reaction between H2S and IR-783, which destoried the structure of IR-783 and blocked the energy transfer, thus led to the quenching of luminescent intensity. Based on this, a sensing system for determination of H2S has been constructed successfully. The linear range of H2S detection by this system is 0.5-15 μM, and the detection limit is 34.17 nM. Furthermore, the dye-sensitized core-shell NaGdF4:Yb,Er@NaGdF4:Yb,Nd upconversion nanoprobe was applied to real sample analysis with satisfactory results.

Abstract:Lanthanide-doped upconversion nanoparticles (Ln-UCNPs) are a new type of nanomaterials with excellent fluorescence properties, which are well applied in fluorescent biosensing. Herein we developed a multifunctional probe based on the surface engineering of core-shell structure UCNPs with polyacrylic acid (PAA). The developed PAA/UCNPs probe could be highly selective to detect and respond to Cu2+ at different pH. Cu2+ could easily combine with the carboxylate anion of PAA to quench the fluorescence of UCNPs. Therefore, we creatively proposed a fluorescent array sensor (PAA/UCNPs-Cu2+), in which the same material acted as the sensing element by coupled with pH regulation for pattern recognition of 5 thiols. It could also easily identify the chiral enantiomer of cystine (l-Cys-and d-Cys), and distinguish their mixed samples with different concentrations, and more importantly, it could be combined with urine samples to detect actual level of homocysteine (Hcys) to provide a new solution for judging whether the human body suffers from homocystinuria.

Abstract:The fabrication of lanthanide upconversion nanocomposites as probes has become a new research hotspot due to its special advantages via utilizing upconversion luminescence (UCL) as a detection signal. Herein, a hybrid organic dye modified upconversion nanophosphor is successfully developed as a nanoprobe for cysteine/homocysteine. Yolk-shell structured upconversion nanoparticles (YSUCNP) with lanthanide upconversion nanophosphor as moveable core and silica as mesoporous shell are synthesized, and a colorimetric chemodosimeter for cysteine/homocysteine is accommodated in the hollow cavities. Thus, cysteine/homocysteine can be quantitatively detected on the basis of luminescent resonance energy transfer (LRET) in a UCL turn-off pattern. The dye-loaded YSUCNP possess good dispersibility in aqueous solution; thus detection of the targeted molecule can be achieved in pure water. Cellular experiments carried out with laser-scanning upconversion luminescence microscopy further demonstrate that the dye-loaded YSUCNP can serve as an intracellular nanoprobe to detect cysteine/homocysteine. Moreover, this dye-loading protocol can be developed as a common approach to construct other chemodosimeter-modified UCNP hybrid nanoprobes, as proved by a UCL turn-on style sensor for cyanide.
Realizing highly chemoselective detection of H2S in vitro and in vivo with fluorescent probes inside core-shell silica nanoparticles

Abstract:Hydrogen sulfide (H2S) is an appealing signaling molecule that plays fundamental roles in health and disease. However, H2S-mediated selective chemical transformations for the construction of imaging probes are limited, retarding the interrogation of H2S-related biological processes. Here, we present an alternative approach for engineering a new generation of efficient probes with a nonchemoselective moiety as a building block. To demonstrate our design concept, we developed a sulfoxide-functionalized BODIPY that exhibited a substantial redshift in its absorption and emission spectra upon reduction with H2S. However, such a probe also showed reactivity toward various competing biothiols under aqueous buffer conditions. To achieve high chemoselectivity, we used core-shell silica nanoparticles as an encapsulation matrix to confine the designed molecule probe within their interiors. The inherent molecular-size sieving character of the porous silica shell was capable of impeding competing biothiols from accessing the molecule probe within the core while allowing the specific reaction with the small target H2S. Thus, this strategy avoided disturbance from coexisting biothiols and achieved highly chemoselective detection in ratiometric and near-infrared (NIR) turn-on fluorescence modes. In light of these promising features, together with fast responsiveness and favorable cellular uptake, such a silica nanocomposite was successfully used to detect the endogenous production of H2S in estrogen-induced cardiomyocytes and living mouse model. To our knowledge, the approach reported here is the first to exploit the usefulness of common thiol-sensitive moieties for building chemoselective probes.
A dual-modality hydrogen sulfide-specific probe integrating chemiluminescence with NIR fluorescence for targeted cancer imaging

Abstract:Taking apart in numerous physiological and pathological activities, hydrogen sulfide (H 2 S) has been selected as an excellent target spot for the early diagnosis of cancer. So far, there are many mature probes that apply single optical imaging to monitor endogenous H 2 S. Nevertheless, a single modality is not an ideal method to afford accurate diagnostic information in comprehensive biological organisms. Herein, we developed a dual-modal imaging probe BWS. This designed probe showed a specific response to H 2 S with both chemiluminescence and NIR fluorescence light-up. The concurrence of fluorescence and chemiluminescence signal provided "double insurances" for highly accurate monitoring of H 2 S. Satisfactorily, this dual-modal imaging probe performed precise visualization of endogenous H 2 S in living cells and in vivo . We envisaged that this chemiluminescence/fluorescence real-time dual-modality strategy for H 2 S detection will expand and optimize the multimodal imaging methods for efficient diagnosis and treatment of cancer.

Chiral MoSe2 Nanoparticles for Ultrasensitive Monitoring of Reactive Oxygen Species In Vivo

Abstract:Reactive oxygen species (ROS) are involved in neurodegenerative diseases, cancer, and acute hepatitis, and the early diagnosis of these diseases by quantification of ROS is critical for the curable treatment of patients. Here, a novel probe was developed based on chiral molybdenum diselenide (MoSe2) nanoparticles (NPs) modified by the fluorescence molecule, cyanine 3 (Cy3). Chiral MoSe2 NPs showed intensive circular dichroism (CD) signals at 390 nm and 550 nm, whereas the fluorescence signal of Cy3 at 560 nm was quenched by MoSe2 NPs. With the presence of ROS, the novel probe reacted with ROS and then oxidated rapidly, resulting in decreased CD signals and the recovery of Cy3 fluorescence signals. With the help of ROS‐related CD and fluorescent dual‐mode signals, the limit of detection (LOD) of CD signals and fluorescence signals in living cells was 0.0093 nmol/106 cells and 0.024 nmol/106 cells, respectively. The high selectivity and sensitivity to ROS in complex biological environments was attributed to the Mo4+ and Se2− oxidation reaction on the surface of the NPs. Furthermore, chiral MoSe2 NPs was able to monitor the levels of ROS in vivo by fluorescence signals. Collectively, this strategy offers a new approach for ROS detection and has the potential to inspire others to explore chiral nanomaterials as potential biosensors to investigate biological events. Abstract:We present an approach for constructing a H2S-specific nanoprobe by the entrapment of a small molecule probe within the hydrophobic interior of surface cross-linked micelles (SCMs), endowing the designed nanoprobes with good water solubility and biocompatibility. Importantly, the obtained nanoprobes displayed good responsiveness to H2S in both ratiometric fluorescence and light-up NIR emission modes, thus enabling accurate identification of H2S-rich colorectal cancer cells.
Dual-Mode Scattering Nanoprobes for Imaging Hydrogen Sulfide in Living Cells

Abstract:Hydrogen sulfide (H2S), a gaseous signaling agent, was considered to play an essential role in the regulation of many biological systems. Herein, we design a surface-enhanced Raman scattering (SERS) and plasmon resonance Rayleigh scattering (PRRS) dual-mode nanoprobe for real-time imaging of H2S in living cells. To realize the dual-mode imaging, we fabricated a core–shell nanostructure with built-in electromagnetic hot spots by embedded Raman dyes between a gold nanostar (AuNS) core and a silver shell. When exposed to sulfide, the Ag shell will rapidly transform into Ag2S, resulting in the variation of PRRS caused by the change of the dielectric constant and the reduction of the SERS signal resulted from the blocking effect of Ag2S. There was well-defined linearity between the dual signals and the sulfide concentrations from 1 to 100 nM. The limit of detection (LOD) was 87 pM for the SERS mode and 53 pM for the PRRS mode, respectively. Complementary performances were proclaimed between the two proposed sensing modes, i.e., the PRRS mode sensing based on single nanoprobes can provide local concentration information of H2S in single cells, while the SERS signal can directly provide the average concentration information of H2S in single cells. Abstract:Nanoprobes based on lanthanide-doped upconversion nanoparticles (UCNPs) exhibit promising potential in bioimaging and biosensing due to their unique optical properties. However, conventional UCNP nanoprobes based on the dye quenching effect are still limited in biosensing due to their low upconversion efficiency. The advent of dye-sensitized upconversion has resulted in nanoprobes with significantly enhanced efficiency; however, these still suffer from a high initial emissive background. In view of this, herein, we have constructed a dye-quenched/sensitized switching upconversion nanoprobe for high-contrast imaging of the pH-related tumor microenvironment. Under normal conditions, the luminescence of the nanoprobe at 540 nm (UCL 540 ) was significantly quenched by the employed dye. Upon being triggered by an acid, the dye would switch to its fluorescent form to sensitize the luminescence of UCNPs, affording a significant enhancement of UCL 540 . The switching from dye-quenched UCL to dye-sensitized UCL jointly enables the detection of a high signal-to-background ratio (high up to 50), allowing for high-contrast mapping of the tumor specific acidic microenvironment in vivo . We believe that this nanoplatform holds considerable promise for acid-related sensing.

Abstract:Detection of Cu2+ ions and study of their subcellular distribution in physiological processes are of considerable significance because of their potential environmental and biological applications. Some fluorescence based sensors have been developed for selective detection of Cu2+ ions, based on organic fluorescent probes that specifically bind to Cu2+ ions. However, these sensors are not suitable for detection in biological samples due to the short penetration depth of UV/visible light used to excite the fluorescent probes. The use of near-infrared (NIR) light can afford penetration depths of an order of magnitude greater than that of visible light, however, a material that can convert NIR light to visible light is required. A facile method has been developed for in-depth detection of Cu2+ ions based on fluorescence upconversion. A mesoporous silica shell is coated on upconversion nanoparticles (UCNPs) and a Cu2+ ion sensitive fluorescent probe, rhodamine B hydrazide, is incorporated into the mesoporous silica. Upon excitation by a NIR light, the UCNPs emit visible light to excite the Cu2+-sensitive fluorescent probe. Because of the unique optical properties of UCNPs and their ability to convert NIR light to visible light, this is a feasible method for sensitive and in-depth detection of Cu2+ ions in a complex biological or environmental sample due to the low autofluorescence and the high penetration depth of NIR light.
Synthesis and applications of a corrole-based dual-responsive fluorescent probe for separate detection of hydrazine and hydrogen sulfide

Abstract:Here, a corrole-based dual-responsive fluorescent probe DPC-DNBS was rationally designed and synthesized for the separate detection of hydrazine (N2H4) and hydrogen sulfide (H2S) with high selectivity and sensitivity. The probe DPC-DNBS is intrinsically none fluorescent due to PET effect, however, addition of increasing amount of N2H4 or H2S to DPC-DNBS turned on an excellent NIR fluorescence centered at 652 nm and thereby provided a colorimetric signaling behavior. The sensing mechanism was verified by HRMS, 1H NMR and the DFT calculations. Common metal ions and anions do not interfere with the interactions of DPC-DNBS with N2H4 or H2S. Furthermore, the presence of N2H4 does not affect the detection of H2S; however, the presence of H2S interferes with the detection of N2H4. Hence, quantitative detection of N2H4 must occur in an H2S-free environment. The probe DPC-DNBS displayed some fascinating merits in separate detection of these two analytes, including large Stokes shift (233 nm), fast response (15 min for N2H4, 30 s for H2S), low detection limit (90 nM for N2H4, 38 nM for H2S), wide pH range (6-12) and outstanding biological compatibility. Significantly, DPC-DNBS was utilized to detect hydrazine in real water, soil and food samples. And its favorable performances for separate detection N2H4 and H2S were successfully demonstrated in HeLa cells and zebrafish, indicating its value of practical application in biology.

Abstract:The rational design of effective tools capable of monitoring hydrogen sulfide (H2S) is of great importance to fully understand its physiological and pathological functions. Herein, a self-assembled fluorescent nanoprobe with both aggregation-induced emission (AIE) and excited-state intramolecular proton transfer (ESIPT) characteristics for H2S detection has been successfully developed via a modified nanoprecipitation method. The nanoprobe features high water dispersibility, a large Stokes shift (ca. 100 nm), good biocompatibility as well as high selectivity towards H2S over other putative interferents. Live cell imaging experiments demonstrate that the nanoprobe, with good cell-membrane permeability, can facilitate the visualization of exogenous and endogenous H2S levels. Moreover, the nanoprobe has also been found capable of detecting H2S in zebrafish. This nanoprobe with AIE and ESIPT characteristics can provide a novel method for the development of fluorescent probes for monitoring biological processes.
Ratiometric Fluorescent Nanosensors for Selective Detecting Cysteine with Upconversion Luminescence.

Abstract:Fluorescent sensors based on upconversion (UC) luminescence have been considered as a promising strategy to detect bio-analyte due to their advantages in deep penetration, minimum autofluorescence, and ratiometric fluorescent output. A prototype of nanosensors combined with mesoporous silica coated upconversion nanoparticles (UCNPs) and a fluorescein-based fluorescent probe loaded in pores was therefore designed to detect cysteine (Cys). The silica shell provided loading space for the probe and enabled the nanosensors to disperse in water. In the presence of Cys, the fluorescent probe was transformed into 5(6)-carboxyfluorescein with an emission band centering at 518nm which was secondarily excited by the light at around 475nm from NaYF4:Yb3+, Tm3+ UCNPs driven by 980nm near-infrared (NIR) laser. The intensity ratio between green and blue luminescence (I518/I475) grew exponentially with increasing concentrations of Cys over a range of 20–200μmolL−1. The response of the nanosensors towards Cys was recognizable with naked eyes by luminescence color change. Evidences suggest that these nanosensors are capable of sensing Cys in aqueous solution and distinguishing Cys from homocysteine (Hcy) with kinetically-controlled selectivity. The system was further employed to detect Cys in human serum and the result was in agreement with it tested by high performance liquid chromatography with acceptable recovery.

Abstract:Hydrogen sulfide (H2S), a third gas signaling molecule, is considered to play a vital role in the development and treatment of diseases. To elucidate the intricate role of H2S in the organism and its participation in disease processes, there is an urgent need to visualize the dynamics of H2S. However, most currently available molecular probes have limitations in terms of sensitivity, specificity, and precision. In this study, the safe and biocompatible upconversion nanosensor NaYF4:Yb3+, Tm3+@NaYF4 anchored with Ag2O was successfully fabricated for H2S detection with an ultralow detection limit at 0.93 ng/mL. NaYF4:Yb3+, Tm3+@NaYF4@Ag2S is formed through in situ sulfuration reaction, and second near-infrared (NIR-II) fluorescence can be recorded upon the presence of H2S under 808 nm excitation. The results demonstrate the exceptional detection linearity and high specificity for H2S quantification. Additionally, NaYF4:Yb3+, Tm3+@NaYF4@Ag2O possesses a safe nature in normal and cancer cells. This nanosensor presents a NIR fluorescence imaging strategy for highly sensitive and specific detection of H2S, which has promises to be a practical tool for biomedical applications.

Abstract:A novel dual‐modal imagingprobe in the second near‐infrared (NIR‐II) window is developed with "double assurances" for the accurate imaging and monitoring of H2S in vivo. High hydrogen sulfide (H2S) expression has been shown to play a crucial role in tumor growth and proliferation in colon cancer. So far, most probes used for monitoring H2S offer single‐modal imaging, which makes the collection of accurate information for tumor diagnosis difficult. In the present study, a novel dual‐modal imaging probe in the second near‐infrared (NIR‐II, 950‐1700) window is proposed and constructed by coating a uniform shell of polydopamine (PDA) on the surface of cuprous oxide (Cu2O)‐conjugated downconversion nanoparticles (DCNPs). Of note, DCNP‐Cu2O@PDA is in the "on" state, with emission centered at 1550 nm (irradiation using a 980nm laser) in non‐neoplastic tissues. Colon tumors with endogenous H2S overexpression can induce the in situ transformation of Cu2O to copper sulfide (Cu2‐XS). Owing to the competitive absorption between Cu2‐XS and DCNPs at a wavelength of 980 nm, the generated DCNP‐Cu2‐XS@PDA exhibits a quenched NIR‐II fluorescence (FL) signal. Meanwhile, Cu2‐XS exhibits a strong absorbance at 1250 nm, enhancing the NIR‐II photoacoustic (PA) signal accompanied by NIR‐II FL attenuation. The switching in PA and FL signals at the tumor site offered "double assurances" for the accurate monitoring of H2S.
