Abstract:Label-free vibrational imaging by stimulated Raman scattering (SRS) provides unprecedented insight into real-time chemical distributions. Specifically, SRS in the fingerprint region (400–1800 cm −1 ) can resolve multiple chemicals in a complex bio-environment. However, due to the intrinsic weak Raman cross-sections and the lack of ultrafast spectral acquisition schemes with high spectral fidelity, SRS in the fingerprint region is not viable for studying living cells or large-scale tissue samples. Here, we report a fingerprint spectroscopic SRS platform that acquires a distortion-free SRS spectrum at 10 cm −1 spectral resolution within 20 µs using a polygon scanner. Meanwhile, we significantly improve the signal-to-noise ratio by employing a spatial-spectral residual learning network, reaching a level comparable to that with 100 times integration. Collectively, our system enables high-speed vibrational spectroscopic imaging of multiple biomolecules in samples ranging from a single live microbe to a tissue slice.
Broadband Quantum Spectroscopy at the Fingerprint Mid-Infrared Region

Abstract:Numerous molecules exhibit unique absorption bands in the “fingerprint” infrared (IR) region, allowing chemical identification and detection. This makes IR spectroscopy an essential tool in biomedical diagnostics, sensing, and material characterization. However, measurements in the IR range are challenging due to the relatively poor performance, need for cryogenic cooling, and high cost of IR light sources and photodetectors. Here, we demonstrate that the mid-IR fingerprints of the sample can be revealed from measurements in the near-IR range using conventional silicon photodetectors. Our approach relies on the quantum interference of correlated photon pairs produced in strong frequency nondegenerate parametric down-conversion. Our technique simultaneously measures the absorption coefficient and refractive index in the broad “fingerprint” region with high accuracy. As a proof-of-concept, we perform spectroscopy of nitrous oxide gas in the 7.4 to 8.4 μm wavelength range, with the detection in the 865 to 877 nm range. This work extends the applicability of quantum interferometry to a range of broad practical appeals, such as detection of atmosphere pollutants, breath diagnostics, chemical safety, and others.

Abstract:Fingerprints are the best form of personal identification for criminal investigation purposes. We present a line-scanning Raman imaging system and use it to detect fingerprints composed of β-carotene and fish oil on different substrates. Although the line-scanning Raman system has been used to map the distribution of materials such as polystyrene spheres and minerals within geological samples, this is the first time to our knowledge that the method is used in imaging fingerprints. Two Raman peaks of β-carotene (501.2, 510.3 nm) are detected and the results demonstrate that both peaks can generate excellent images with little difference between them. The system operates at a spectra resolution of about 0.4 nm and can detect β-carotene signals in petroleum ether solution with the limit of detection of 3.4×10(-9) mol/L. The results show that the line-scanning Raman imaging spectroscopy we have built has a high accuracy and can be used in the detection of latent fingerprints in the future.

Abstract:We investigate the recognition of fingerprints from the Fourier spectrum. The inherent properties of fingerprints allow a feature extraction and data reduction in the spatial frequency domain. The Fourier representation allows fingerprints to be distinguished from a small and spatially well-defined area. This suggests various schemes to detect the significant information in order to optimize the trade-off between sensitivity and robustness. We show illustrative results which confirm the usefulness of this approach. In addition, the classification of fingerprints from their plane wave spectra allows the design of compact systems, where the Fourier transformation is performed optically, while detection and post-processing is done by electronics. This provides the advantage that both optics and electronics are used in an optimum way to minimize the physical size of the system, as well as the computational load to interpret the detected signal.

Abstract:This article aims to overview infrared (IR) spectroscopy. Simultaneously, it outlines mid-infrared (MIR), near-infrared (NIR), and far-infrared (FIR) or terahertz (THz) spectroscopy separately, and compares them in terms of principles, characteristics, advantages, and applications. MIR spectroscopy is the central spectroscopic technique in the IR region, and is mainly concerned with the fundamentals of molecular vibrations. NIR spectroscopy incorporates both electronic and vibrational spectroscopy; however, in this review, I have chiefly discussed vibrational NIR spectroscopy, where bands due to overtones and combination modes appear. FIR or THz spectroscopy contains both vibrational and rotational spectroscopy. However, only vibrational FIR or THz spectroscopy has been discussed in this review. These three spectroscopy cover wide areas in their applications, making it rather difficult to describe these various topics simultaneously. Hence, I have selected three key topics: hydrogen bond studies, applications of quantum chemical calculations, and imaging. The perspective of the three spectroscopy has been discussed in the last section.

Abstract:Fourier transform infrared (FTIR) spectroscopy is a widely used characterization technique for studying chemical compositions and light-matter interactions in the infrared range. However, it remains challenging to use conventional FTIR equipment to characterize materials that are environmentally sensitive and/or have rough surfaces due to their dynamic response to external stimuli (e.g., humidity) and light scattering from the sample surface. Here, we describe an approach enabling quantitative infrared spectroscopy measurements of these challenging samples, including transmittance, reflectance, and emissivity. We designed and 3D-printed a miniaturized environmental chamber that can be directly inserted into the sample holder of a conventional integrating sphere to perform total transmission and reflection measurements in controlled environments. Moreover, a calibration method was developed to exclude light scattering from rough surfaces of the sample and the distortion created by infrared windows. To illustrate the potential application of this method, we have performed quantitative infrared measurements, both specular and diffusive, on a sapphire standard reference with a specular surface, as well as a textile sample that has a rough surface and is dynamically responsive to changes in humidity. The calibrated results measured with the equipped chamber match those measured without the IR windows, demonstrating the viability of our environmental chamber and the proposed calibration method. This quantitative infrared spectroscopy measurement technique may help advance fields such as metasurfaces and adaptive textiles, where samples are often rough, dynamically responsive, and/or environmentally sensitive.
Fast infrared chemical imaging with a quantum cascade laser

Abstract:Infrared (IR) spectroscopic imaging systems are a powerful tool for visualizing molecular microstructure of a sample without the need for dyes or stains. Table-top Fourier transform infrared (FT-IR) imaging spectrometers, the current established technology, can record broadband spectral data efficiently but requires scanning the entire spectrum with a low throughput source. The advent of high-intensity, broadly tunable quantum cascade lasers (QCL) has now accelerated IR imaging but results in a fundamentally different type of instrument and approach, namely, discrete frequency IR (DF-IR) spectral imaging. While the higher intensity of the source provides a higher signal per channel, the absence of spectral multiplexing also provides new opportunities and challenges. Here, we couple a rapidly tunable QCL with a high performance microscope equipped with a cooled focal plane array (FPA) detector. Our optical system is conceptualized to provide optimal performance based on recent theory and design rules for high-definition (HD) IR imaging. Multiple QCL units are multiplexed together to provide spectral coverage across the fingerprint region (776.9 to 1904.4 cm(-1)) in our DF-IR microscope capable of broad spectral coverage, wide-field detection, and diffraction-limited spectral imaging. We demonstrate that the spectral and spatial fidelity of this system is at least as good as the best FT-IR imaging systems. Our configuration provides a speedup for equivalent spectral signal-to-noise ratio (SNR) compared to the best spectral quality from a high-performance linear array system that has 10-fold larger pixels. Compared to the fastest available HD FT-IR imaging system, we demonstrate scanning of large tissue microarrays (TMA) in 3-orders of magnitude smaller time per essential spectral frequency. These advances offer new opportunities for high throughput IR chemical imaging, especially for the measurement of cells and tissues.
Detection of illicit substances in fingerprints by infrared spectral imaging

Abstract:FTIR and Raman spectral imaging can be used to simultaneously image a latent fingerprint and detect exogenous substances deposited within it. These substances might include drugs of abuse or traces of explosives or gunshot residue. In this work, spectral searching algorithms were tested for their efficacy in finding targeted substances deposited within fingerprints. "Reverse" library searching, where a large number of possibly poor-quality spectra from a spectral image are searched against a small number of high-quality reference spectra, poses problems for common search algorithms as they are usually implemented. Out of a range of algorithms which included conventional Euclidean distance searching, the spectral angle mapper (SAM) and correlation algorithms gave the best results when used with second-derivative image and reference spectra. All methods tested gave poorer performances with first derivative and undifferentiated spectra. In a search against a caffeine reference, the SAM and correlation methods were able to correctly rank a set of 40 confirmed but poor-quality caffeine spectra at the top of a dataset which also contained 4,096 spectra from an image of an uncontaminated latent fingerprint. These methods also successfully and individually detected aspirin, diazepam and caffeine that had been deposited together in another fingerprint, and they did not indicate any of these substances as a match in a search for another substance which was known not to be present. The SAM was used to successfully locate explosive components in fingerprints deposited on silicon windows. The potential of other spectral searching algorithms used in the field of remote sensing is considered, and the applicability of the methods tested in this work to other modes of spectral imaging is discussed.

Abstract:The Fourier-Transform-Infrared spectrometry (FTIR) is a very valuable technique for identifying organic substances. FTIR is more versatile in comparison to disperse infrared spectrometry, as it combines speed and sensitivity. FTIR is even superior to GC/MS: 1. Easy differentiation between isomers is possible. 2. Functional group information can be achieved instead of mass fragments. 3. GC fractions can be saved for further analysis. The FTIR instrument has no prisma or grating. It is equipped with a Michelson interferometer, which gives much more beam energy on the examination object. Most beneficial is the use of a broadband beam source, which impinges simultaneously all frequencies on the sample and on the detector. All these qualities allow the registration of fast chemical and biological processes or the observation of all reactions by volatizing and evaporation at high temperatures. IR spectrometer can run high quality infrared spectra in short time (almost 100/second) with high sensitivity. This is the basic requirement for the "on line" coupling of the IR-system with a GC. The GC peaks are analysed by IR beams with the help of a "light pipe". It is possible to examine all kinds of substances with diffuse and attenuated total reflectance methods. In special cases the change for binding of proteins and kinetic and structural processes in the flowing blood can be measured in vivo. Another use of FTIR-techniques is possible in combination with a microscope. Samples with a square aera of 8 microns can be examined. In criminalistics and forensic toxicology little particles as hairs, fibers, foams, adherents of powder in clothes, splinters of lacquer and paints etc. can be detected and identified by transmission or reflection. In conclusion, the FTIR spectrometry offers a versatility to some measurement problems that is often superior, faster and easier than other techniques such as NMR and MS.
High-speed Fourier-transform infrared spectroscopy with phase-controlled delay line

Abstract:Fourier-transform infrared spectroscopy (FTIR) is the golden standard of mid-infrared (MIR) molecular spectroscopic analysis through optically-encoded vibrational signatures. Michelson-type FTIR and MIR dual-comb spectrometers allow us to simultaneously investigate multiple molecular species via the broadband and high-resolution spectroscopic capabilities. However, these are not applicable to high-speed measurements due to the low temporal resolution which is fundamentally limited by the signal-to-noise ratio (SNR). In this study, we develop a high-speed FTIR spectroscopy technique called phase-controlled Fourier-transform infrared spectroscopy (PC-FTIR) that has the capability to measure MIR absorption spectra at a rate of above 10 kHz. PC-FTIR demonstrates the high scan rate with a high SNR for various spectral bandwidths by arbitrarily adjusting the instrumental spectral resolution. As a proof of principle demonstration, we measure high-speed mixing dynamics of two liquids at a rate of 24 kHz. We also measure MIR spectra of gas-phase molecules with higher spectral resolution at a rate of 12 kHz. This high-speed MIR spectrometer could be especially useful for measuring non-repetitive fast phenomena and acquiring a large amount spectral data within a short time.

Abstract:The mechanical detection of photothermal expansion from infrared (IR) absorption with an atomic force microscope (AFM) bypasses Abbe's diffraction limit, forming the chemical imaging technique of AFM-IR. Here, we develop a Fourier transform AFM-IR technique with peak force infrared microscopy and broadband femtosecond IR pulses. A Michelson interferometer creates a pair of IR pulses with controlled time delays to generate photothermal signals transduced by AFM to form an interferogram. A Fourier transform is performed to recover IR absorption spectra. We demonstrate the Fourier transform AFM-IR microscopy on a polymer blend and hexagonal boron nitride. An intriguing observation is the vertical asymmetry of the interferogram, which suggests the presence of multiphoton absorption processes under the tip-enhancement and femtosecond IR lasers. Our method demonstrates the feasibility of time-domain detection of the AFM-IR signal in the mid-IR regime and paves the way toward multiphoton vibrational spectroscopy at the nanoscale below the diffraction limit.

Abstract:We applied Hilbert-Huang Transform to analyze terahertz spectra in order to improve the state-of-art detection sensitivity of inconspicuous spectral fingerprints for materials with low concentrations and remote sensing. We conducted experiments on ammonia gas and water vapor with terahertz time domain spectroscopy. The temporal terahertz signal is decomposed into a series of intrinsic mode functions using Hilbert-Huang Transform. By comparing the frequency and absorption spectra obtained from the original signals and their intrinsic mode functions, we have showed the validity of low-order intrinsic mode functions and demonstrated that basically the first two orders of functions are capable to show the spectral fingerprints clearly. Therefore, Hilbert-Huang Transform has the potential to become an effective supplementary tool for terahertz spectroscopy.

Abstract:Fourier transform infrared spectrometer can carry out qualitative and quantitative analysis of samples. Therefor it is widely used in medicine and chemical industry, geology, mining, petroleum, coal and other fields. Since the characteristic frequency region and fingerprint region of the infrared spectrum play an important role in distinguishing compounds with similar structures, the locating accuracy of spectral peaks directly affects the performance of subsequent sample analysis. In order to realize the peak searching of infrared spectrum, this paper proposes a method based on piece-wise polynomial fitting to fit the spectral data, and designs a peak searching method based on derivation on this basis. Taking the polystyrene infrared wavelength standard material as the sample, the experiment verifies that the spectral fitting and peak searching method proposed in this paper can obtain the required wave number indication error and wave number repeatability, having good performances.

Abstract:A new method for representing fingerprint feature by visualization of infrared spectral data was proposed. Using the multivariate statistical method based on Bayes rules, raw analytical data were transformed into a new subspace by mathematical projection. Subsequently, the transformed data were visualized into the virtual chemical fingerprint plots by means of two-dimensional grayscale images. The proposed method was applied to identify herbal medicine-DangGui from three origins. The results showed that it can effectively extract the image characteristics from infrared spectral data, and the different geographical origins of DangGui samples can be accurately identified by the virtual chemical fingerprint plots. Therefore, it provides a new technical tool for identifying complicated chemical substance system.
