Abstract:A general and stereoselective synthesis of (Z)-ketoeneynes 9 and 11a-f was established by using a Pd(O)-Cu(I)-catalyzed cross-coupling of the labile (Z)-ketoenol triflate 3 with 1-alkynes under carefully controlled reaction conditions. Isomerization of the coupling products into the more stable (E)-ketoeneynes 8 and 12a-f was observed and could be minimized by earring out the coupling reaction in CH3CN at low temperature using Et3N as the base. Abstract:Cross-electrophile coupling reactions of two Csp3-X bonds remain challenging. Herein we report an intramolecular nickel-catalyzed cross-electrophile coupling reaction of 1,3-diol derivatives. Notably, this transformation is utilized to synthesize a range of mono- and 1,2-disubstituted alkylcyclopropanes, including those derived from terpenes, steroids, and aldol products. Additionally, enantioenriched cyclopropanes are synthesized from the products of proline-catalyzed and Evans aldol reactions. A procedure for direct transformation of 1,3-diols to cyclopropanes is also described. Calculations and experimental data are consistent with a nickel-catalyzed mechanism that begins with stereoablative oxidative addition at the secondary center.

Abstract:The direct conversion of alkyl esters to ketones has been hindered by the sluggish reactivity of the starting materials and the susceptibility of the product towards subsequent nucleophilic attack. We have now achieved a cross-coupling approach to this transformation using nickel, a bulky N-heterocyclic carbene ligand, and alkyl organoboron coupling partners. 65 alkyl ketones bearing diverse functional groups and heterocyclic scaffolds have been synthesized with this method. Catalyst-controlled chemoselectivity is observed for C(acyl)-O bond activation of multi-functional substrates bearing other bonds prone to cleavage by Ni, including aryl ether, aryl fluoride, and N-Ph amide functional groups. Density functional theory calculations provide mechanistic support for a Ni-0/Ni-II catalytic cycle and demonstrate how stabilizing non-covalent interactions between the bulky catalyst and substrate are critical for the reaction's success.

Abstract:We have developed a nickel- nickel/bisphosphin-ecatalyzed stereoconvergent cross-coupling reaction of epoxides with alkenylboronic acids. Racemic spiroepoxyoxindoles were converted to chiral homoallylic alcohols bearing quaternary carbon stereogenic centers via a stereoablative enantioconvergent transformation. The subsequently fabricated oxindoles-carrying quaternary carbon products were obtained in good yields and enantioselectivity. A wide range of substrates and alkenylboronic acids was tolerated under the catalytic system. This reaction provided a rare example of a nickel-catalyzed enantioselective cross-coupling reaction of tertiary alkyl electrophiles and an enantiocon-vergent transformation of racemic epoxides, beneficial as a low-cost, sustainable, and efficient catalyst in the preparation of chiral oxindole-containing natural and pharmaceutical compounds.

Abstract:Chiral nickel complexes promoted enantioselective reductive alkenylation of a range of conjugated enones, using alkenyl bromides, triflates, and tosylates. The neutral condition was compatible with sensitive groups and azaheteroaryl rings. Importantly, alkenyl groups in products can be readily converted to functionalized alkyl groups via iron-catalyzed reductive hydrofunctionalization. Some examples of asymmetric alkenylation of N-enoyl pyrroles and indoles were also included.

Abstract:The preparation of enantioenriched diarylmethanol derivatives is described using nickel‐catalyzed electrochemical cross‐couplings between various alkyl/aryl aldehydes and aryl iodides. Performed in an electrochemical cell equipped with an iron anode and a nickel cathode, this electrocatalytic variant led to the scalemic targeted products in the presence of 2,2‐bis((4R,5S)‐4,5‐diphenyl‐4,5‐dihydrooxazol‐2‐yl)acetonitrile (L2), as enantiopure cyano‐bis(oxazoline) ligand. X‐ray structure analysis of a pre‐catalyst, for instance the [Ni(II)(L2)2] complex, with L2 as an anionic bisoxazolinate ligand, confirms the chemical formulation of one nickel surrounded by two ligands. The redox behavior of the new Ni complexes generated in situ was first assessed by cyclic voltammetry showing a redox wave at ca. ‐1.5 V that can be assigned to the two‐electron reduction of the Ni(II) center to the Ni(0) state. Oxidative addition between the electrogenerated Ni(0) complex and aryl iodide was evidenced. An intense current was observed in presence of a mixture of the two substrates pertaining an electrocatalytic process. Interestingly, we found that the sacrificial iron anode plays a crucial role in the catalytic mechanism.

Abstract:α-Tertiary aliphatic amides are key elements in organic molecules, which are abundantly present in natural products, pharmaceuticals, agrochemicals, and functional organic materials. Enantioconvergent alkyl-alkyl bond-forming process is one of the most straightforward and efficient, yet highly challenging ways to build such stereogenic carbon centers. Herein, we report an enantioselective alkyl-alkyl cross-coupling between two different alkyl electrophiles to access α-tertiary aliphatic amides. With a newly-developed chiral tridentate ligand, two distinct alkyl halides were successfully cross-coupled together to forge an alkyl-alkyl bond enantioselectively under reductive conditions. Mechanistic investigations reveal that one alkyl halides exclusively undergo oxidative addition with nickel versus in-situ formation of alkyl zinc reagents from the other alkyl halides, rendering formal reductive alkyl-alkyl cross-coupling from easily available alkyl electrophiles without preformation of organometallic reagents.

Abstract:Enol ether structural motifs exist in many highly oxygenated biologically active natural products and pharmaceuticals. The synthesis of the geometrically less stable Z-enol ethers is challenging. An efficient Z-selective oxidative isomerization process of allyl ethers catalyzed by a cobalt(II) (salen) complex using N-fluoro-2,4,6-trimethylpyridinium trifluoromethanesulfonate (Me3NFPY•OTf) as an oxidant has been developed. Thermodynamically less stable Z-enol ethers were prepared in excellent yields with high geometric control. This methodology also demonstrates the effectiveness in controlling the Z-selective isomerization reaction of diallyl ethers at room temperature. This catalytic system provides an alternative pathway to extend the traditional reductive isomerization of allyl ethers.

Abstract:Abstract Enantioenriched N -alkylindole compounds, in which nitrogen is bound to a stereogenic sp 3 carbon, are an important entity of target molecules in the fields of biological, medicinal, and organic chemistry. Despite considerable efforts aimed at inventing methods for stereoselective indole functionalization, straightforward access to a diverse range of chiral N -alkylindoles in an intermolecular catalytic fashion from readily available indole substrates remains an ongoing challenge. In sharp contrast to existing C–N bond-forming strategies, here, we describe a modular nickel-catalyzed C–C coupling protocol that couples a broad array of N -indolyl-substituted alkenes with aryl/alkenyl/alkynyl bromides to produce chiral N -alkylindole adducts in single regioisomeric form, in up to 91% yield and 97% ee. The process is amenable to proceed under mild conditions and exhibit broad scope and high functional group compatibility. Utility is highlighted through late-stage functionalization of natural products and drug molecules, preparation of chiral building blocks.

Abstract:Metal-catalysed cross-coupling has played a central role in modern chemical synthesis for decades. Unactivated alkenes, including light olefins, are manufactured on enormous scale in the petroleum industry and represent ideal starting materials for the preparation of pharmaceuticals, agrochemicals and materials. However, enantioselective cross-coupling of unactivated alkenes remains a challenging and long-standing goal. Here we report a highly enantio- and regioselective three-component cross-coupling of unactivated alkenes with aryl (or alkenyl) triflates and organometallics (or reductant) to construct diverse C sp 3 stereocentres by nickel catalysis. Specifically, selective carbonickelation and in situ trapping with nucleophiles enable efficient hydrofunctionalization and dicarbofunctionalization of unactivated alkenes in a directing group-free manner. Nickel catalysts bearing bulky C 2 -symmetric chiral N-heterocyclic carbene ligands were crucial for attaining high reactivity and selectivity. This strategy offers a general, modular and divergent platform for rapidly upgrading feedstock alkenes to various value-added molecules and is expected to inspire the development of other challenging enantioselective alkene cross-couplings.

Abstract:Asymmetric catalytic versions of electricity-driven processes hold immense potential for the sustainable preparation of chiral compounds. However, the involvement of anodic oxidative cross-dehydrogenative coupling events between two distinct nucleophiles makes it challenging for a chiral catalyst to regulate the stereochemistry of the products. Our current electrocatalytic strategy for enantioconvergent cross-dehydrogenative α- and γ-nitroalkylation via radical-based pathways produces an array of enantioenriched nitroesters without supplementary stoichiometric oxidants. Mechanistic investigations reveal that the nickel catalyst plays a key role in both the electrochemical activation of the substrates and the stereoselectivity-defining events, affording the electrochemically generated Lewis acid-bound α-carbonyl radicals to interact with in situ-generated nitronate anions in a stereoselective manner. This electrocatalytic approach enables transformations that are highly challenging under thermal conditions, such as umpolung reactivity with readily available substrates, all-carbon quaternary stereocenter creation, and the control of remote stereochemistry.
