Abstract:A highly regio- and stereoselective hydrocyanation of 1,3-enynes was implemented by nickel/diphosphine catalysts. A wide range of highly regio- and stereoselective alkenyl nitriles were efficiently prepared. In this transformation, both the tethered alkene and the ligand play key roles in the reactivity and selectivity. The origin of regioselectivity was understood preliminarily by DFT studies.

Abstract:The development of a catalytic multicomponent reaction by orthogonal activation of readily available substrates for the streamlined di-functionalization of alkynes is a compelling objective in organic chemistry. Alkyne carboalkynylation, in particular, offers a direct entry to valuable 1,3-enynes with different substitution patterns. Here, we show that the synthesis of stereodefined 1,3-enynes featuring a trisubstituted olefin is achieved by merging alkynes, alkynyl bromides, and redox-active N-(acyloxy)phthalimides through nickel-catalyzed reductive alkylalkynylation. Products are generated in up to an 89% yield as single regio- and E isomers. Transformations are tolerant of diverse functional groups and the resulting 1,3-enynes are amenable to further elaboration to synthetically useful building blocks. With olefin-tethered N-(acyloxy)phthalimides, a cascade radical addition/cyclization/alkynylation process can be implemented to obtain 1,5-enynes. This study underscores the crucial role of redox-active esters as superior alkyl group donors compared with haloalkanes in reductive alkyne dicarbofunctionalizations.

Abstract:A novel synthesis of acyclic cis-enediynes 2 has been established by an acid-catalyzed rearrangement of 1,2-diyn-2-propen-1-ols 1 possessing a C-3-aryl group in the presence of water, alcohols, or thiols. Reactivity of allyl alcohols and regio- and cis/trans diastereoselectivity of the allylic migration were examined. In the presence of(+/-)-10-camphorsulfonic acid (CSA), the parent allyl alcohol 5 and the C-3-methyl-substituted 9 failed to give enediynes, whereas the C-3-aryl-substituted 12 and 29 underwent the allylic rearrangement to provide predominantly cis-enediynes 16 and 31 at room temperature or below. Under similar acidic conditions, enediyne alcohol 13 produced 16b and 16d with the same regio and cis/trans diastereoselectivity observed for 12. Allyl alcohol 30, an isomer of 29, also provided enediynes 31c and 32c after a prolonged reaction (90 h) at room temperature in the presence of CSA and EI;OH. These results suggested that the same allylic cations were obtained from allyl alcohols 12 and 13 or 29 and 30 even though the ease of ionization differed for each substrate. Involvement of allylic cations in the product-forming step was confirmed by the finding that chiral allyl alcohols (-)-12 and (-)-18c furnished racemic products. In general, the p-MeOPh-substituted allyl alcohol 29 gave a better regioselectivity than the Ph-substituted 12. In the reactions with alcohols, the regioisomeric ratios were 100:0 (31:33) for 29 and ca. 96:4 (16:17) for 12; the ratios decreased to ca. 90:10 (31:33) for 29 and ca. 10:30 (16:17) for 12 when thiols were used. The cis/trans diastereoselectivity is higher for allyl alcohol 12 (100% for 16 at 20 degrees C) compared to that for 29 (31:32 = 80:20-94:6 at 0 degrees C). Computational calculations at the RHF/3-21G level, carried out on the model compounds and allylic cations, indicated that nucleophilic trapping takes place preferentially at the Cg carbon to form the thermodynamically much more stable enediynes. Under the best reaction conditions (1 equiv of CSA and 2 equiv of EtOH in CH2Cl2, 20 degrees C), a number of acyclic cis-enediynes can be synthesized in three steps from the commercially available alpha-bromocinnamaldehyde (10).

Abstract:A general and stereoselective synthesis of (Z)-ketoeneynes 9 and 11a-f was established by using a Pd(O)-Cu(I)-catalyzed cross-coupling of the labile (Z)-ketoenol triflate 3 with 1-alkynes under carefully controlled reaction conditions. Isomerization of the coupling products into the more stable (E)-ketoeneynes 8 and 12a-f was observed and could be minimized by earring out the coupling reaction in CH3CN at low temperature using Et3N as the base. Abstract:All-carbon quaternary stereocenters constitute a significant portion of natural products and bioactive compounds. Here we disclose a nickel and chiral amine cooperative catalysis to enable a highly enantioselective allylic alkylation reaction between α-branched aldehydes and a wide scope of allyl esters, allowing the all-carbon quaternary stereocenter to be accessed with excellent enantioselectivity (up to 98% ee) and structural diversity. Abstract:NiCl2(PMe3)2-catalyzed reaction of allyl alcohols with silylzinc reagents, including PhMe2SiZnCl, Ph2MeSiZnCl, and Ph3SiZnCl, was performed, achieving allylsilanes in high yields. Aryl- and heteroaryl-substituted allyl alcohols, (E)-3-arylprop-2-en-1-ols, 1-aryl-prop-2-en-1-ols, and (E)-1-phenylpent-1-en-3-ol can be employed in the transformation. A range of functional groups as well as heteroaryl groups were tolerated. Reaction exhibited high regioselectivity and E/Z-selectivity when 1- or 3-aryl-substituted allyl alcohols were used as the substrates. Reaction of chiral allyl alcohol, (S,E)-1-phenylpent-1-en-3-ol, yielded a configuration-inversion product (R,E)-dimethyl(phenyl)(1-phenylpent-1-en-3-yl)silane.

Abstract:An efficient Ni-catalyzed four-component 1,4-carbocarbonylation of 1,3-enynes with activated alkyl halides and arylboronic acids under atmospheric pressure of CO is presented. By tuning the electronic and steric effects of alkyl radicals, both electron-rich and electron-deficient 1,3-enynes were compatible with this cascade. This protocol features mild conditions, broad substrate scope, excellent functional group compatibility and facile gram-scale synthesis, providing a practical approach to the quaternary carbon center-containing allenyl ketones. Mechanistic study revealed that the acyl-Ni II species plays an important role in both the coupling and the alkyl radical generation processes.

Abstract:Allenyl ketones are widely distributed in various bioactive molecules which lead their selective synthesis continues a hot research topic. Currently, most of the developed protocols require multistep and stoichiometric reactions together with noble metal catalysts. Herein, we developed a practical methodology to access allenyl ketones straightforwardly via multi-component coupling of 1,3-enynes, fluoroalkyl halides, arylboronic acids, and CO with nickel as the catalyst. A series of fluoroalkyl unit containing allenyl ketones were produced in moderate to good yields. This new procedure features cheap catalytic system, relatively mild reaction conditions, broad substrate scope, excellent regioselectivity, and attractive synthetic transformations of the obtained products.

Abstract:We herein present the construction of a chiral 1,4-enyne featuring tertiary or quaternary stereogenic center via Pd-catalyzed branch-, enantio-, and diastereoselective allylic C-H alkylation. Alkynyl carbon bearing bulky substituents appeared to exhibit competitive reaction performance, and the desired chiral 1,4-enynes were obtained in up to 93% yield and with up to >20:1 b/l, >20:1 dr, and 98% ee. A gram-scale experiment, the feasible operation of benzothiazole ring, and the preparation of the key intermediate to access (+)-Breynolide and prostaglandin are represented as a demonstration of multifarious synthetic utility in chemical synthesis.

Abstract:Alkenes constitute an enabling motif in organic synthesis, as they can be functionalized to form highly substituted molecules. Z-alkenes are generally challenging to access due to the thermodynamic preference for the formation of E-alkenes compared to Z-alkenes. Dehydrogenation methodologies to selectively form Z-alkenes have not yet been reported. Herein, we report a Z-selective, propargylic dehydrogenation that provides 1,3-enynes through the invention of a Co-catalyzed oxidation system. Observation of a kinetic isotope effect (KIE) revealed that deprotonation of the propargylic position is the rate limiting step. Additionally, isomerization experiments were conducted and confirmed that the observed Z-selectivity is a kinetic effect. A proposed stereomechanistic model for the Z-selectivity is included.

Abstract:1,3-Enynes have been widely recognized as a prevalent synthon for the construction of valuable allenes, 1,3-dienes, and propargylic derivatives. However, significant advancements have primarily focused on elucidating the ionic reaction mechanism. The selective difunctionalization of 1,3-enynes involving radicals remains an outstanding challenge in particular within the realm of radical 1,2-difunctionalizations. Herein, we present a comprehensive account of the regio- and diastereoselective radical multicomponent 1,2-dialkylation of 1,3-enynes via photoexcited Hantzsch ester and chromium co-catalysis to furnish highly valuable homopropargylic alcohols. Our protocol shows excellent compatibility and 1,2-regioselectivity. Remarkably, this innovative approach obviates the need of external photocatalysts, stoichiometric metal reductants, or additional additives, offering a novel strategy for the valuable transformation of 1,3-enynes.

Abstract:Chiral nickel complexes promoted enantioselective reductive alkenylation of a range of conjugated enones, using alkenyl bromides, triflates, and tosylates. The neutral condition was compatible with sensitive groups and azaheteroaryl rings. Abstract:The transition-metal-catalyzed selective hydroarylation of unsymmetrical alkynes represents the state-of-art in organic chemistry, and still mainly relies on the use of precious late-transition-metal catalysts. Reported herein is an unprecedented Mn-I-catalyzed hydroarylation of unsymmetrical 1,3-diyne alcohols with commercially available arylboronic acids with predictive selectivity. This method addresses the challenges in regio-, stereo-, and chemoselectivity. It offers a general, convenient and practical strategy for the modular synthesis of multisubstituted Z-configurated conjugated enynes. This protocol is distinguished by its operational simplicity, complete selectivity, excellent functional-group compatibility, and gram-scale potential. A dimeric Mn-I species, Mn-2(CO)(8)Br-2, was proven to be a much more efficient catalyst precursor than Mn(CO)(5)Br.

Abstract:A regiodivergent hydrophosphorylation of enynes with phosphites has been developed using earth-abundant nickel catalyst. The manipulation of regioselectivity can be achieved by regulating the insertion order of alkyne bonds with (RO) 2 P(O)–Ni–H or R 2 P(O)O–Ni–H species, respectively. Under the Ni/Xantphos catalysis, 4,1-hydrophosphorylation is selectively obtained while the adding of acid can promote reactions towards 1,2-addition. By employing an additional Pd–H catalysis, 2,1-hydrophosphorylation is also an accessible task in one-pot reaction. Mechanistic studies and analysis have also been performed to interpret the origin of the regioselective regulation. This work highlights the arts in accessing different regioisomers by diverting common elementary reaction steps.
