Abstract:A set of two cyclic [2]catenane dimers were self-assembled in aqueous media via selective hydrazone condensation, by means of either one-pot or post-functionalization procedures. Considering that each catenane is composed of two thirty-six member rings, each of these two cyclic [2]catenane dimers can be considered as a ultra-large macrocycle that contains more than fifty atoms in the ring framework. This cyclization methodology relying on catenation thus represents a novel approach for accomplishing ultramacrocyclization in pure water.

Abstract:An efficient, clean and facile protocol for the Knoevenagel condensation of aromatic aldehydes with active methylene compounds catalysed by task specific ionic liquid 1,8-diazabicyclo[5.4.0]-undec-7-en-8-ium acetate in an aqueous medium has been developed. The reactions proceed at room temperature and high to excellent yields were afforded. The work-up procedure is very simple. The ionic liquid could be recycled for 10 runs without noticeably loss of its catalytic activity.

Abstract:A series of purely organic macrocycles and catenanes can be self-assembled by condensing a cationic bisaldehyde compound with a series of dihydrazide linkers in weakly acidic water. On one hand, the macrocycles could be generated as the predominant products under the condition of low concentration or less polar media. In the presence of a guest template, these macrocycles could even be obtained in close to quantitative yields, allowing them to be isolated as pure solid products without the need for chromatographic purification. On the other hand, [2]catenanes could be obtained as the major products in more concentrated solutions or more polar media where hydrophobic effects are enhanced. Once purified, both macrocycles and catenanes exhibit remarkable kinetic stability in both the solid state and neutral aqueous solution at room temperature. By means of selective host-guest recognition, one of the macrocyclic products is capable of resolving a pair of hydrocarbon isomers, namely phenanthrene and anthracene, which have similar properties and can hardly be separated by commonly used approaches.

Abstract:The synthesis of macrocyclic compounds with different sizes and linkages remains a great challenge via transition metal-catalysed intramolecular C–H activation. Herein, we disclose an efficient macrocyclization strategy via Pd-catalysed remote meta-C–H olefination using a practical indolyl template. This approach was successfully employed to access macrolides and coumarins. In addition, the intermolecular meta-C–H olefination also worked well and was exemplified by the synthesis of antitumor drug belinostat from inexpensive and readily available benzenesulfonyl chloride. Notably, catalytic copper acetate and molecular oxygen were used in place of silver salts as oxidants. Furthermore, for the first time, the formation of a macrocyclophane cyclopalladated intermediate was detected through in situ Fourier-Transform Infrared monitoring experiments and ESI-MS.

Abstract:We report the synthesis of macrocycles 1-6 via ring closing metathesis of dienes 7-12. Addition of chlorodicyclohexylborane to thermal and microwave assisted RCM of dienes 8 and 9 markedly improves the yield. The geometric isomers of macrocycles 1-3 and 5 have been assigned using X-ray crystallography and NMR.

Abstract:The promotion of two sulfate or carboxvlate-bearing acyclic cucurbiturils for the water-solubility of arenes and aromatic aldehydes is described. H-1 NMR experiments reveals that sulfate-bearing acyclic cucurbituril (acCB-1) signficantly improves the water-solubility of a number of arenes and aldehydes. For 4,4'-dimethylbiphenvl and biphenyl-4,4'-dicarbaldehyde, the solubility can be improved to 8.9 and 11.2 mmol/L, respectively. F-19 NMR experiments demonstrate that carboxvlate-bearing acyclic cucurbituril can increase the water-solubilities of pentafluorotoluene and hexafluorobenzene to 5.6 and 3.0 mmol/L, respectively. It is also found that the water-solubilization of acCB-1 for aromatic aldehydes can promote their reaction with acylhydrazines to form hvdrozone derivatives. By making use of this promotion, two hydrazone-based macrocvcles can be formed from the coupling reactions of two aromatic dialdehdes and one diacylhydrazine in water.

Abstract:A kind of o-carborane-based macrocycles were synthesized via oxidative coupling of acetyleneterminated o-carborane precursors. The single crystal X-ray diffraction analysis unambiguously revealed their unique porous supramolecular structures, which displayed micropore characteristics. Interestingly, the crystalline phase of macrocyclic CBMC-2 meets special structure parameters through non-covalent interaction. UV–vis, DSC and X-ray analysis indicated that the crystals of macrocycle CBMC-2 could undergo diacetylene topochemical polymerization inside solid by heating. It was confirmed by Raman spectroscopy and solid-state 13C NMR spectroscopy. This work provides a method for synthesizing carborane supramolecular polymers.

Abstract:By performing Friedel-Crafts reaction with a mixture of 1,3,5-tri(furan-2-yl) benzene (TFB) and acetone, we successfully synthesized a cylinder-shaped macrocycle. The isolated yield of 36% is relatively high, considering that twelve C-C bonds are formed simultaneously. The structure of this macrocycle was confirmed using H-1 NMR spectroscopy, mass spectrometry and X-ray crystallography. This macrocycle is able to recognize a variety of cationic guests driven by cation-dipole and CH-pi interactions.

Abstract:A preparative synthesis of previously unknown 21- and 14-membered azamacrocycles via acid-promoted cyclotrimerization or cyclodimerization of three readily available precursors, namely, 1-amino-6-hydroxy-4,6-dimethylhexahydropyrimidine-2-thione, 4-(4-oxopent-2-yl)thiosemicarbazide hydrazone, and 5,7-dimethyl-1,4,5,6-tetrahydro-3H-1,2,4-triazepine-3-thione has been developed. A dramatic dependence of the selectivity of the macrocyclization on the reaction conditions is demonstrated. Thermodynamic aspects of the reactions are discussed based on experimental data and DFT calculation results. Plausible pathways for the formation of the macrocycles are proposed.

Abstract:Macrocycles with up to 100 atoms have been synthesised using two calix[4]arenes as templates: first, (3,5-dialkenyloxy)phenyl groups are attached to the wide rim of a calix[4]arene via urea links, then the alkenyl groups are connected via a metathesis reaction using a tetratosylurea calix[4]arene for their correct prearrangement and finally the urea functions are cleaved to detach the newly formed macrocycles.

Abstract:A macrocycle through a dynamic covalent approach relying on reversible hydrazone formation in acidic aqueous solutions at elevated temperatures is constructed. By decreasing the acidity of the solution and lowering the temperature, the structure becomes kinetically inert. The macrocycle is capable of hosting hydrophobic aromatic guest molecules in water.

Abstract:Ring-in-ring complexation is a novel topological structure in supramolecular chemistry. Two macrocyclic compounds with intramolecular donor-acceptor interactions were designed and synthesized with 4,4'-bi-pyridinium, 2,6-dinaphthol and 2,7-dinaphthol as building blocks. Their associations with cucurbit[8] uril were investigated by means of UV-Vis and H-1 NMR. The spectra results show that intramolecular charge transfer( CT) interaction exsits in aqueous solution of these two macrocycles, and cucurbit[8] uril can bind the two macrocycles. The binding result in the increase of charge transfer(CT) absorption strength and red shift of CT absorption peak, indicating the formation of stable ring-in-ring complexes. These ring-in-ring complexes are new topological structures for cucurbit[8]uril.

Abstract:Macrocycles have attracted much attention due to their specific "endless" topology, which results in extraordinary properties compared to related linear (open chain) molecules. However, challenges still remain in their controlled synthesis with well-defined constitution and geometry. Here, we report the successful application of the (pseudo-)high-dilution method to the conditions of on surface synthesis in ultrahigh vacuum. This approach leads to high yields (up to 84%) of cyclic hyperbenzene ([18]-honeycombene) via an Ullmann-type reaction from 4,4"-dibromo-meta-terphenyl (DMTP) as precursor on a Ag(111) surface. The mechanism of macrocycle formation was explored in detail using scanning tunneling microscopy and X-ray photoemission spectroscopy. We propose that the dominant pathway for hyperbenzene (MTP)(6) formation is the stepwise desilverization of an organometallic (MTP-Ag)(6) macrocycle, which forms via cyclization of (MTP-Ag)(6) chains under pseudo-high-dilution conditions. The high probability of cyclization on the stage of the organometallic phase results from the reversibility of the C-Ag bond. The case is different from that in solution, in which cyclization typically occurs on the stage of a covalently bonded open-chain precursor. This difference in the cyclization mechanism on a surface compared to that in solution stems mainly from the 2D confinement exerted by the surface template, which hinders the flipping of chain segments necessary for cyclization.

Abstract:By taking advantage of a newly developed self-assembly approach based on oxime condensation, we successfully obtained a tetracationic macrocycle bearing two diquats in acidic aqueous media in a high yield. The ring is able to accommodate HPO42- and HCO3- in pure water.

Abstract:Diiodoarenes (or dibromoarenes) reacted with diphenols, catalyzed by Cul/Fe(acac)(3) in the presence of K2CO3 in anhydrous DMSO at 110 degrees C for 7 days under nitrogen atmosphere, to afford macrocyclic aryl ethers effectively. To expand this methodology, a cyclic hepta(p-phenylene oxide) and cyclic deca(pphenylene oxide) were synthesized in one pot. Abstract:This paper describes the synthesis of two series of rigid macrocycles from hydrogen bonding-induced folded aryl amide and hydrazone oligomers that bear two amines or one amine and one aldehyde. The diamines reacted with diacyl chloride to produce amide macrocycles, whereas the latter underwent self-coupling reactions to afford imine macrocycles. DFT calculations revealed that the new macrocycles possess rigid planar conformations and their cavity diameters were estimated to be 1.86 nm-2.75 nm. (C) 2014 Elsevier Ltd. All rights reserved.

Abstract:We have developed a unique approach for the thermally promoted cycloaddition of cyclohexadienone-tethered allenes to form a versatile cyclobutane-fused oxygen-containing tricyclic framework in an environmentally friendly and atomic economic fashion with high regioselectivity. The reaction encompasses a broad substrate scope and functional group tolerance of cyclohexadienone moieties. Moreover, the cycloaddition was also applicable to the late-stage functionalization of pharmaceutically relevant compounds.

Abstract:This paper describes a novel, highly efficient approach to the self-assembly of monomacrocycles and two-layered capsules by using dynamic covalent chemistry. Intramolecular hydrogen-bonding was used to preorganize aromatic amide-based monomers that contain aldehyde and tert-butoxycarbonylamino units. As a result, in the presence of an excess of trifluoroacetic acid (TFA), four monomers could self-couple to produce macrocycles selectively through the formation of three imine or hydrazone bonds. Three dipodal precursors were also prepared by connecting two hydrogen-bonded segments with a flexible linker. In the presence of TFA, these precursors could also self-couple, leading to the exclusive formation of two-layered capsules. As a result of intramolecular hydrogen-bonding, all the macrocycles and capsules were stable in solution and could be purified by simple recrystallization. The new capsules were able to form complexes with linear propylenediammonium derivatives to give unique two-layered pseudo[3]rotaxanes.

Abstract:A series of macrocycles, including crown ethers, cyclodextrins, calixarenes, pillararenes and cucurbiturils, are well known to be able to associate various organic/inorganic/biological guest molecules and ions in their well-defined cyclic cavities to form stable host-guest complexes and supramolecular systems through the cooperative contributions of various non-covalent interactions. When one or more functional groups are attached to the cavity of macrocycles or guest molecules, enhanced and/or controlled host-guest associations may take place, leading to not only improved host-guest binding abilities but also fascinating properties. In this review, some representative contributions in the construction of controllable macrocyclic supramolecular assemblies in aqueous solution are presented with an emphasis on the stimuli-responsive control manner and wide applications of this property.
