Abstract:The optimal fermentation conditions for production of xylanase by engineered Pichia pastoris GS115-ATx in shake-flask cultivation was investigated. The results showed that the best carbon and nitrogen sources for xylanase production were bran and beef extract, respectively. Judging from their effect on xylanase production efficiency, organic nitrogen was better than inorganic nitrogen. The best methanol concentration was 0.5% for inducing xylanase expression. The xylanase activity was increased when 0.5% glycerin was added into the culture every 24 h. In addition, Toween 20, Toween 80, and betaine were found to increase the yield of xylanase, and among them betaine was the optimal stimulator. SDS-PAGE analysis revealed that the molecular weight of xylanase expressed by GS115-ATx was about 19.0 kDa. The xylanase activity of the culture supernatant reached its peak after 96-h methanol induction.

Abstract:proteinic powder of corpuscle was used as material,with enzymatic hydrolysis rate,protein yield,and decolor rate as indexes and by using neutral protease,the key parameters for upgrade the proteinic powder of corpuscle by enzymatic hydrolysis were studied.The obtained key parameters are as follows: temperature 50 ℃,enzyme activity 5 000 u/g,and pH 8.Under the conditions the proteinic powder of corpuscle can be hydrolyzed best and gives high yield of aminosal.Results also show that the liquid can be well decolorized and gives high yield of aminosal under the conditions of pH 4,dosage of active carbon 0.2g/100 mL,temperature 50 ℃ and time 60 min.

Abstract:The fermentation conditions for the production of neutral protease by Bacillus subtilis were investigated using single factor and orthogonal design experiments.The results showed that the optimal fermentation medium in the flasks contained 40 g/L glycerin,30 g/L soymeal,10 g/L bran,4 g/L Na2HPO4,and 0.3 g/L K2HPO4.The optimal fermentation conditions in the shaker were culturing with 6％ inoculum amount at 30 ℃ and 250 r/min.Under this condition,the enzyme activity reached 6 082.7U/mL.To further verify the optimized fermentation condition,the glycerol was fed and batch fermentation was carried out in 15 L fermenter.The enzyme activity reached 11 871 U/mL.
Preparation of Hydrolyzed Protein from Harpodon Nehereus by Enzymatic Hydrolysis Method

Abstract:The preparation conditions of hydrolyzed protein from Harpodon nehereus were studied for increasing the field of hydrolyzed protein. Harpodon nehereus protein was hydrolyzed first by Alcalase and then by Flavourzyme and the debitterization of hydrolysate by activated carbon was also studied. The hydrolysis conditions of Alcalase and Flavourzyme and the debitterization conditions by activated carbon were optimized by orthogonal tests. The molecular weight and amino acid composition of hydrolysate were determined by Sephadex G-75 size-exclusion chromatography and automatic amino acid analyzer, respectively. The results showed that the degree of hydrolysis is up to 17.68 % under optimum conditions. The bitterness of hydrolysate is almost removed by activated carbon. The average molecular weight of hydrolysate is 1 430Da, which indicated that hydrolysate is composed of 13 amino acids, averagely. The content of essential amino acids is 34.9 % of the total amino acids. The content of palatable and sweet taste amino acids is 27.6 % of the total free amino acids.

Abstract:Response surface methodology, which allows for rapid identification of important factors and optimization of them to enhance enzyme production, was employed here to optimize culture conditions for the production of cis-epoxysuccinic acid hydrolase from Bordetella sp. strain 1-3. In the first step, a Plackett-Burman design was used to evaluate the effects of nine variables (yeast extract, cis-epoxysuccinic acid, KH(2)PO(4), K(2)HPO(4).3H(2)O, MgSO(4).7H(2)O, trace minerals solution, culture volume, initial pH and incubation time) on the enzyme production. Yeast extract, cis-epoxysuccinic acid and KH(2)PO(4) had significant influences on cis-epoxysuccinic acid hydrolase production and their concentrations were further optimized using central composite design and response surface analysis. A combination of adjusting the concentration of yeast extract to 7.8 g/l, cis-epoxysuccinic acid to 9.8 g/l, and KH(2)PO(4) to 1.12 g/l would favor maximum cis-epoxysuccinic acid hydrolase production. An enhancement of cis-epoxysuccinic acid hydrolase production from 5.6 U/ml to 9.27 U/ml was gained after optimization.

Abstract:Industrial waste products are of biotechnological interest due to their abundance and can be utilized as protein sources for the production of bioactive peptides through hydrolysis. The utilization of yeast residues from the brewing industry has been demonstrated to be a viable method for obtaining protein hydrolysates. The degree of hydrolysis and soluble total protein of the hydrolysates were evaluated for optimization purposes to achieve a product with a high proportion of soluble organic nitrogen, increased levels of bioactive peptides and amino acids. Both alkaline and enzymatic hydrolysis processes were tested. The hydrolyses were carried out with the aim of large-scale implementation. The treatments were effective in liberating intracellular content and producing biologically functional hydrolysates, with degree of hydrolysis values ranging from 28 to 63% and total protein from 2.95 to 8.28% at the optimum points confirmed by statistical analysis (analysis of variance (ANOVA)). The hydrolysate produced with Alcalase® exhibited the highest peptide intensity, while the hydrolysate produced with Flavourzyme® showed the highest concentration of free amino acids. The peptides were identified by mass spectrometry and were found to have biological activities according to the Biopep database. This study presents a quick and economically feasible approach for the implementation of a pilot project for the reuse of this waste.

Abstract:Biotransformation processes accompanied by whole yeast cells as biocatalyst are a promising area of food industry. Among the chemical sanitizers currently used in food technology, hydrogen peroxide is a very effective microbicidal and bleaching agent. In this paper, permeabilization has been applied to Saccharomyces cerevisiae yeast cells aiming at increased intracellular catalase activity for decomposed H2O2. Ethanol, which is non-toxic, biodegradable and easily available, has been used as permeabilization factor. Response surface methodology (RSM) has been applied in determining the influence of different parameters on permeabilization process. The aim of the study was to find such values of the process parameters that would yield maximum activity of catalase during decomposition of hydrogen peroxide. The optimum operating conditions for permeabilization process obtained by RSM were as follows: 53% (v/v) of ethanol concentration, temperature of 14.8 °C and treatment time of 40 min. After permeabilization, the activity of catalase increased ca. 40 times and its maximum value equalled to 4711 U/g.

Abstract:The objective of the current study was to optimize the fermentation conditions of producing fibrinolytic enzyme through single factor experiment and response surface experiment.The optimal fermentation conditions were obtained as follows:the carbon source was bran,the nitrogen source was yeast extract,the inorganic salt was 0.1% MgSO4,the solvent of medium was bean juice,pH was 4.0.Using 2% bran,2% yeast extract,0.1% MgSO4 as the major factor,choosing the right step,designing response surface experiment,the second-order regression model was obtained fitting the experimental data results.The R2 of the model was 0.8709,and was significant at the 90% of the probability.The second-order model predicted that under the conditions of 2.97% bran,2.30% yeast extract paste,0.147% MgSO4,pH 4,the highest fibrinolytic activity predicted was 410 IU/mL.

Abstract:Many microorganisms produce enzymes which lyse the walls of yeasts, fungi, and bacteria. The proportions of different enzyme activities present in the lytic system, their action patterns, synergism, and dependence on inhibitors, constitute the activity profile of the lytic system. Taken together, the activity profile and process conditions for lysis determine the reaction rate and the distribution of products from lysis of any given type of cells. Kinetics of glucan hydrolysis, proteolysis, and lysis of brewer's yeast were compared for two extracellular yeast-lytic enzyme systems with different properties. The enzyme sources used were filtered culture broths from Cytophaga sp. NCIB 9497 grown in batch culture and from Oerskovia xanthineolytica LL-G109, grown under carbon limitation in continuous culture. Rate and extent of cell hydrolysis, and the accumulation of soluble proteins, peptides, and carbohydrates from the lysed yeast cells, are discussed in terms of the activity profiles and potential applications of the two enzyme systems.

Study on Optimized Conditions for Enzyme Hydrolysis of White Hypsizygus Marmoreus by Response Surface Methodology

Abstract:Objective: In order to get protein hydrolysis fluid with high content of amino acid, we try a new method of developing white hypsizygus marmoreus deeply. Methods: single factor experiment was adopted to determine the technical conditions when using the compound enzyme to hydrolysis white hypsizygus marmoreus and the optimal conditions were determined by response surface with the design-expert software. Results: the single factor experiment showed that the parameters were T=45 ℃, t=5 h, pH 7, water∶material=25∶1. The optimal conditions were T=43.28 ℃, t=5.47 h, pH= 7.08. Under the conditions, the predicted value of hydrolysis was 20.13% , which is close to the actual value 20.37% and the confidence degree of regression equation was 98.76%. Conclusions: The content of amino acid in protein hydrolysis fluid was improved by enzymolysis and the hydrolysis fluid provided a good preparation for white hypsizygus marmoreus quelite.
Effects of a complex phytase-containing enzyme preparation on the rye wort fermentation process

Abstract:A complex of amylases, proteases, and hemicellulases is known to enhance deep conversion of polysaccharides and proteins, especially in the processing of difficult-to-ferment raw materials, such as rye, providing grain wort with soluble carbohydrates, amino acids, and peptides. Grain is also a source of phosphorus, whose bioavailability can be increased by hydrolysing the grain with phytase-containing enzyme preparations. However, their catalytic action during the preparation of grain wort for alcohol production has hardly been studied. This study aimed to investigate the effect of a new complex phytasecontaining enzyme preparation on yeast metabolism and the efficiency of rye wort fermentation. The work was carried out in the Russian Research Institute of Food Biotechnology. The Glucavamorin complex enzyme preparations derived from recombinant strains were the object of our research. The preparations differed in the activity level of the main enzyme, lucoamylase, and minor hemicellulase enzymes, as well as in the presence of phytase. The results confirmed their biocatalytic ability to efficiently hydrolyse polymers of rye grain. An increased content of hemicellulases in Glucavamorin-Xyl improved the rheological properties of rye wort. The greatest effect was achieved with the phytase-containing Glucavamorin-Ply. This preparation improved the phosphorus nutrition of yeast, which increased its biomass by 30% and decreased the level of fermentation by-products by 18–20%. Alcohol yield tended to increase and its strength reached 10.5–10.9% vol. When using a phytase-containing enzyme complex, it was possible to reduce the amount of the main enzyme, glucoamylase, without causing the key fermentation indicators to degrade.

Abstract:Introduction. Microbial biomass is a popular source of food ingredients and feed additives. Its high use has made it focus of many relevant studies. Yeast and fungal biomasses proved to be useful substrates that improve the quality and biological value of functional products. They differ in the content and composition of proteins and polysaccharides. The present research dealt with the enzymatic decomposition of proteins found in a novel fungal and yeast biomass. The research objective was to describe the peptide and amino acid composition of their enzymatic hydrolysates. Study objects and methods. The research featured a new fungal and yeast biomass mix. Aspergillus oryzae is a mycelial fungus and a popular industrial producer of hydrolytic enzymes in food industry. As for the yeast, it was the Saccharomyces cerevisiae strain, which is often used in baking. Results and discussion. The total content of identified amino acids in the fungal and yeast biomass was 306.0 mg/g, which was 1.5 times higher than in the fungal biomass alone. The biomass mix demonstrated a higher biological value of proteins than the yeast biomass. A set of experiments made it possible to compile a scheme for the biocatalytic destruction of polymers in the fungal and yeast biomass under the effect of fungal intracellular and endogenous enzymes. The article also contains a thorough description of the obtained enzymatic hydrolysates with various fractional compositions of peptides and free amino acids. Peptides with the molecular weight in the range of up to 29.0 kDa decreased by 2.1 times after 5 h of hydrolysis and by 10.7 times after 18 h. The designed conditions doubled the release of amino acids and increased the content of low-molecular-weight peptides up to 75.3%. Conclusion. The research provided a new algorithm for the biocatalytic conversion of microbial biomass. Regulating the conditions of enzymatic hydrolysis made it possible to obtain enzymatic hydrolysates with a desired degree of protein degradation. They could serve as peptides and amino acids in functional food and feed products.

Abstract:In this study, optimization of ethanol production from carob pod extract was carried out by immobilized Saccharomyces cerevisiae. Results showed that Ca-alginate concentration and the amount of immobilized cells had significant effects on yield. Optimum conditions for ethanol fermentation were determined to be 2% Ca-alginate concentration, 150 rpm agitation rate, 5% yeast cells entrapped in beads and pH 5.5. After validation experiments; ethanol concentration, yield, production rate and sugar utilization rate were respectively 40.10 g/L, 46.32%, 3.19 g/L/h and 90.66%; and the fermentation time was decreased to 24 h. In addition, the immobilized cells were shown to be reusable for five cycles, though a decrease in yield was observed. Finally, carob pod extract was used for ethanol fermentation by controlled and uncontrolled pH without any enrichment, and the results suggest that carob extract can be utilized effectively by immobilized-cell fermentation without the use of enrichments to facilitate yeast growth.

Abstract:Five proteases(Papain,Alcalase 2.4 L,Neutrase 0.8 L,Protamex,Flavorzyme 1 000 L) were used to hydrolyze the fruit body protein of Coprinus comatus at their suitable conditions.Based on the comparision of degree of hydrolysis(DH),protein recovery(PR),sensory evaluation and antioxidant activities of hydrolysates,Alcalase 2.4 L was the most suitable protease for the protein,for its highest DH and PR,better sensory quality and antioxidant activities.The effects of enzymolysis temperature,pH,time,ratio of enzyme activity to substrate ratio([E]/[S]) and substrate concentration on DH were studied.Response surface methodology was used to optimise the hydrolysis parameters of the fruit body protein of Coprinus comatus by Alcalase 2.4 L.The results showed that the optimum conditions were pH at 9.0,temperature at 54.8 ℃,[E]/[S] at 6271 U/g,time at 4 h,substrate concentration at 1%.At these conditions,the DH was 24.05%.

Abstract:Protein hydrolysates might display diverse bioactivities with potential relevance to human and animal health and food technology. Enzymatic hydrolysis of agro-industrial by-products is increasingly focused. In this study, a crude protease from Bacillus sp. CL18 was applied to obtain antioxidant protein hydrolysates from porcine, bovine, poultry, and fish by-products. The crude enzyme hydrolyzed all the twelve investigated by-products, as detected by increased soluble protein contents after 4 h of proteolysis. Hydrolysates exhibited higher radical-scavenging, Fe 2+ -chelating and reducing power capacities than non-hydrolyzed by-products. Hydrolysis times (0–8 h) and enzyme-to-substrate (E/S) ratios (384, 860, and 1,400 U/g) were assessed to produce antioxidant bovine lung hydrolysates. The highest E/S ratio accelerated both hydrolysis and increases in antioxidant activities; however, it did not result in bioactivities higher than hydrolysates obtained with the intermediate E/S ratio. Optimal antioxidant activities could be reached after 6 h of hydrolysis using 860 U/g. Animal by-products are interesting sources of bioactive protein hydrolysates, which could be produced with a non-commercial bacterial protease. This might represent a promising strategy for the valorization of animal by-products generated in large amounts by the agri-food sector.

Optimization of Enzymatic Hydrolysis by Protease Produced from Bacillus subtilis MTCC 2423 to Improve the Functional Properties of Wheat Gluten Hydrolysates

Abstract:This study is aimed at investigating the reutilizing of gluten protein from the wheat processing industry by Bacillus subtilis MTCC 2423 protease to obtain gluten hydrolysates with high added value. Gluten protein hydrolysis using protease achieved a 34.07% degree of hydrolysis with 5% gluten protein, at a hydrolysis time of 2 h for 1000 U/mL at pH 8.0 and temperature of 40°C. Compared to the wheat gluten, the obtained hydrolysates exhibited enhanced functional attributes, including heightened solubility (43%), increased emulsifying activity (93.08 m2/g), and improved radical scavenging properties. Furthermore, these hydrolysates demonstrated enhanced antioxidant potential, as evidenced by elevated ABTS (2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) of 81.25% and DPPH (2,2-diphenyl-1-picrylhydrazyl) of 56.46% radical scavenging activities and also exhibited a higher α-amylase inhibitory effect of 33.98%. The enhancement in functional characteristics of wheat gluten hydrolysates was observed by Fourier transform infrared spectroscopy. The percentage of free amino acids obtained by protease-mediated hydrolysates increased significantly compared to the unhydrolyzed wheat, which was observed by high-performance liquid chromatography. These findings suggest that wheat gluten hydrolysates hold promising potential as functional and nutritional food ingredients in the food industry, owing to their enhanced functionalities and potential antioxidant and antidiabetic properties.

Abstract:In this study various Aspergillus strains were explored for the production of protease using variety of agricultural byproducts as substrate. Aspergillus viridi was found to be the best among the selected strains with production of 83.0 U/mL of protease when grown on casein broth medium. Various agricultural byproducts were utilized for the growth of A. viridi and for the maximum protease production. Optimal production of enzyme 172.6 U/mL was recorded when PDB medium was supplemented with 3% banana peels powder as compared to 90.7, 84.7, 71.5, 68.7 and 47 U/mL when PDB medium was supplemented with pomegranate peels (3%), mustard oil cake (3%), arabic gum sawdust (2%), linseed oil cake (2%) and rosewood sawdust (3%) respectively. A. viridi could produce 172.6 U/mL when grown on 3% banana peels as sole substrate for growth. The protease production was increased with optimization of substrate water ratio (1:3), incubation time (72 h), inoculum size (2 mL), pH (8) and temperature (35 °C) from 172.6 to 178.1, 180.2, 182.9, 188.7 and 193.1 U/mL. Under optimal conditions, A. viridi produced 202.8 U/mL using agricultural byproducts as sole substrate. The strong potential of protease to remove hair from skin makes this enzyme a suitable candidate for its utilization in leather industry.

Abstract:Yak whey protein concentrates (YWPCs) have good functional properties, but there is still a gap in the study of their peptides. In this study, peptides were obtained by enzymatic hydrolysis, and the bioactivity of each ultrafiltration fraction was evaluated using an optimal process. YWPCs were isolated and purified from yak milk as the raw material. Alkaline protease, trypsin, and papain were used to hydrolyze YWPCs. The protease with the highest degree of hydrolysis (DH) and peptide concentration was selected as the most suitable enzyme. The effects of pH, temperature, time, and the enzyme-to-substrate ratio (E/S) on the DH and peptide concentration were investigated, and response surface methodology was utilized to optimize the hydrolysis process. The hydrolysate was separated using ultrafiltration membranes with molecular weight cut-offs of 10 kDa, 5 kDa, 3 kDa, and 1 kDa. The bioactivity of each ultrafiltration component was analyzed, including the inhibition rates of α-amylase and xanthine oxidase (XOD) activities and the scavenging rates of 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) cation radicals. The results indicated that alkaline protease was the best enzyme for hydrolyzing YWPCs. The peptide concentration in the YWPC hydrolysate was the highest (17.21 mg/mL) at a pH of 8 and a concentration of 7500 U/g, after 2.5 h at 62 °C. The enzymatic hydrolysate was ultrafiltered to yield four peptide fractions, of which the <1 kDa peptides exhibited the highest α-amylase inhibitory activity (22.06%), XOD inhibitory activity (17.15%), and ABTS cationic free radical scavenging rate (69.55%). This demonstrates the potential of YWPC hydrolyzed peptides for hypoglycemic, uric acid-lowering, and antioxidant applications, providing a theoretical basis for the high-value utilization of YWPCs.
