Abstract:One of the fundamental goals of bio-ethanol production is to find the alternative of traditional land-relied feedstock. A kind of macroalgae, Sargassum horneri (S. horneri) was chosen as the bio-source for ethanol production in the present investigation. The bioethanol was achieved by acid-pretreated, hydrolysis, and semi-simultaneous saccharification and fermentation (S-SSF) at small pilot scale. Immobilization of Pichia stipites in calcium alginate was adapted in the S-SSF process. Furthermore, effects of pre-hydrolysis time, cellulase loading, fermentation temperature and fermentation time on the concentration, and yield of bio-ethanol were evaluated by using response surface methodology (RSM). A maximum bioethanol concentration (2.89 g/L), and maximum bioethanol yield (0.11 g/g raw material) at cellulase loading (10 IU/g raw material), pre-hydrolysis time (53 min), SSF temperature (32 °C) and SSF time (14 h) were observed. The optimized bioethanol production data series fit the Gompertz model and modified Logistic model with an R2 value of 0.997. The obtained Gompertz and modified Logistic coefficients indicated that S. horneri can serve as an efficient substrate for bioethanol production. The fermentation residues were used for typical dyeing contamination adsorption, the existence of the residual lignin and exposure functional groups made it an effective adsorbent, which followed the pseudo-second-order kinetics. These results are helpful for the scale-up development of lignocellulosic macroalgae based bioethanol production and fermentation residue use.
Optimization of fermentation conditions for xylanase production by engineered Pichia pastoris

Abstract:The optimal fermentation conditions for production of xylanase by engineered Pichia pastoris GS115-ATx in shake-flask cultivation was investigated. The results showed that the best carbon and nitrogen sources for xylanase production were bran and beef extract, respectively. Judging from their effect on xylanase production efficiency, organic nitrogen was better than inorganic nitrogen. The best methanol concentration was 0.5% for inducing xylanase expression. The xylanase activity was increased when 0.5% glycerin was added into the culture every 24 h. In addition, Toween 20, Toween 80, and betaine were found to increase the yield of xylanase, and among them betaine was the optimal stimulator. SDS-PAGE analysis revealed that the molecular weight of xylanase expressed by GS115-ATx was about 19.0 kDa. The xylanase activity of the culture supernatant reached its peak after 96-h methanol induction.

Abstract:53#, a strain of efficient alginate degrading bacteria, is isolated from rotted kelp using sodium alginate as the only carbon resource. Strain 53# is identified asPaenibacillusagaridevorans based on physiological characteristics and 16S rRNA sequencing. The optimaltion yield condition of strain 53# is identified at pH=8, T=25oC, NaCl 15 g/L, and sodium alginate 15 g/L by orthogonal experiment and analysis, and the highest enzyme activity is 390.53±17.32 U/mL. Strain 53# has the advantages such as being cultivated easily, producing enzyme fast and having high enzyme activity. It can achieve a high efficiency for saccharification of alginate and thus has potential value to be utilized in the production of bioethanol from brown algae.

Abstract:To valorize oleaginous Schizochytrium sp. biomass, a novel sequential process was addressed and developed to simultaneously produce high value DHA-enriched glycerides and low value bulky biodiesels. In this process, Schizochytrium sp. biomass was first treated by alkaline protease; then the resulting biomass was directly subjected to enzymatic ethanolysis catalyzed by liquid enzymes from C. antarctica (CALA), T. lanuginosus (TL), R. miehei (RM) and A. oryzae (PLA). For all four liquid lipases, the alkaline protease treatment on Schizochytrium sp. biomass significantly enhanced total lipid recovery, biodiesel yield and yield of DHA-enriched glycerides. Under optimized conditions, CALA and TL achieved the highest biodiesel yields 76.28-76.66 wt% based on microalgae TFAs while PLA had the lowest (55.25 wt%). Moreover, CALA attained the highest DHA (77.93 wt%) and LC PUFAs (93.08 wt%) contents in glyceride fraction, suggesting a superior performance to concentrate DHA and LC PUFAs in glycerides. C-13 NMR analysis illustrated that non-regiospecificity and unfavorability towards LC PUFAs of CALA made it possible to transesterify saturated fatty acids located in all 3 positions of microalgal oil; thus LC PUFAs were highly enriched in glycerides. Overall, the sequential process using enzymes and ethanol as green media was successfully established for the co-production of high-value microalgal bioproducts rich in LC PUFAs and sustainable biodiesels in a comprehensive manner.

Abstract:Bacillus amyloliquefaciens NB, a glutamate-independent poly-gamma-glutamic acid (gamma-PGA)-producing strain, can directly utilize inulin-containing sustainable materials. However, low gamma-PGA yield and lack of efficient genetic engineering approaches have hindered the industrial use of this strain. Here, we used the CRISPR-Cas9n technique to engineer B. amyloliquefaciens to enhance gamma-PGA production. We engineered three modules involved in inulin hydrolysis, reducing sugars metabolism, and gamma-PGA synthesis in B. amyloliquefaciens. Specifically, overexpresed the native inulin hydrolase CscA and two expression-optimized levanase and en doi nulinase, overexpressed of key genes related to reducing sugar metabolism to increased ATP production, and removed polysaccharide operon epsA-O and gamma-PGA hydrolase cwlO. Finally, the highest production of gamma-PGA (32.14 +/- 0.38 g/L) was obtained in a 7.5 L fed-batch fermenter. Thus, we successfully constructed an ideal candidate strain for efficient gamma-PGA production from inulin, which provides an important research basis for the development of more biobased products.
Optimization of newly isolated Bacillus cereus α-amylase production using orange peels and crab shells and application in wastewater treatment

Abstract:A newly isolated amylolytic strain was identified as Bacillus cereus spH1 based on 16S and 16-23S gene sequencing (Accession numbers OP811441.1 and OP819558, respectively), optimization strategies, using one variable at time (OVAT) and Plackett–Burman design, were employed to improve the alpha-amylase (α-amylase) production. Condition inferred revealed that the optimal physical parameters for maximum enzyme production were 30 °C, pH 7.5, and 12 h of incubation, using tryptone, malt extract, orange ( Citrus sinensis) peels, crab ( Portunus segnis) shells, calcium, and sodium chloride (NaCl) as culture medium. The full factorial design (FFD) model was observed to possess a predicted R 2 and adjusted R 2 values of 0.9788 and 0.9862, respectively, and it can effectively predict the response variables ( p = 0). Following such efforts, α-amylase activity was increased 141.6-folds, ranging from 0.06 to 8.5 U/mL. The ideal temperature and pH for the crude enzyme activity were 65 °C and 7.5, respectively. The enzyme exhibited significant stability, with residual activity over 90% at 55 °C. The maltose was the only product generated during the starch hydrolysis. Moreover, the Bacillus cereus spH1 strain and its α-amylase were used in the treatment of effluents from the pasta industry. Germination index percentages of 143% and 139% were achieved when using the treated effluent with α-amylase and the strain, respectively. This work proposes the valorization of agro-industrial residues to improve enzyme production and to develop a green and sustainable approach that holds great promise for environmental and economic challenges.

Salmeterol enhances pulmonary fibrinolysis in healthy volunteers

Abstract:Objective: Various lung diseases are associated with local activation of coagulation and concurrent inhibition of fibrinolysis. Although salmeterol, a &bgr;2‐adrenoceptor agonist with profound bronchodilatory properties, has been studied extensively, the effects of this compound on the pulmonary hemostatic balance are not elucidated. Design: A single‐blinded, placebo‐controlled study. Setting: University hospital and laboratory. Subjects: A total of 32 human volunteers. Interventions: Subjects inhaled 100 &mgr;g of salmeterol or placebo (t = −30 mins) followed by 100 &mgr;g of lipopolysaccharide (LPS) or normal saline (t = 0 mins; n = 8 per group). Measurements and Main Results: Measurements were performed in bronchoalveolar lavage fluid obtained 6 hrs postchallenge. Inhalation of LPS enhanced pulmonary coagulation as determined by an increase in the concentrations of thrombin–antithrombin complexes, factor VIIa, and soluble tissue factor in bronchoalveolar lavage fluid (all p < .05 vs. saline). LPS concurrently inhibited pulmonary fibrinolysis, as reflected by a decrease in bronchoalveolar lavage fluid plasminogen activator activity together with an increase in plasminogen activator inhibitor type 1 (both p < .05 vs. saline). Moreover, LPS inhalation was associated with a suppression of the anticoagulant protein C pathway, as indicated by an increase in soluble thrombomodulin and decreases in protein C and activated protein C levels in bronchoalveolar lavage fluid (all p < .05 vs. saline). Salmeterol, either with or without LPS inhalation, enhanced fibrinolysis (plasminogen activator activity and tissue‐type and urokinase‐type plasminogen activator levels) but did not influence LPS‐induced changes in coagulation or the protein C pathway. Conclusions: Salmeterol has profibrinolytic properties in the normal lung and when applied in a model of sterile pulmonary inflammation.

Abstract:Sugarcane bagasse (SB) a renewable and abundant source of rich fermentable sugars has been reported extensively to produce microbial enzymes and other valuable products. However, most of the SB-based processes require its pretreatment which is an additional step incurring time and cost. In this study, fermentation of SB by bacterial strains without any pretreatment was carried out to obtain crude cellulase preparation. Enzyme mediated saccharification of SB was carried out and the hydrolysate was analyzed for the presence of reducing sugars and free radical scavenging activity. It was found that 65.86, 42.03 and 88.8 IU mL-1 endoglucanase was produced by Brevibacillus borstelensis UE10, Neobacillus sedimentimangrovi UE25 and B. borstelensis UE27, respectively by fermenting SB. Crude cellulase preparation of UE10, UE25, and UE27 was used to saccharify SB which resulted in 1.14, 1.47, and 1.58 mg mL-1 reducing sugars and 1.08, 0.57, and 1.5 mg mL-1 glucose, respectively. Moreover, the hydrolysate also had radical scavenging activity indicating about its possible application in animal feed. The maximum (35.71 %) amount of bioethanol was produced when UE25 fermented the hydrolysate of SB. The strains UE10 and UE27 produced 31.16 and 27.77 % bioethanol, respectively. Furthermore, Fourier transform infrared spectroscopy, scanning electron microscopy and gravimetric analysis showed the impact of cellulase on structure of SB. Hence, this work expands the understanding of the potential of thermophilic bacterial strains to utilize agricultural waste for the synthesis of valuable products.

Abstract:High-value metabolites, such as enzymes and biofuels, can be produced from various agro-industrial waste containing high percentages of cellulose and hemicellulose. Aspergillus niger ITV02 demonstrates high potential in cellulases production, the key enzyme for converting lignocellulosic materials into fermentable sugars to produce second-generation bioethanol (bioethanol 2G). This study evaluated five lignocellulosic residues of agricultural importance: sugarcane bagasse (SCB), sorghum bagasse (SB), corn stubble (CS), barley straw (BS) and rice husk (RH) as substrates for cellulase production. The temperature, pH and stirring conditions were optimized using a Box–Behnken design to identify the most suitable conditions for cellulase production while minimizing nitrogen concentrations. The results indicate that the best way of propagation A. niger ITV02 is through the use of spores as an inoculant, in conjunction with the use of materials with a high cellulose/lignin ratio, such as CS and SB for the generation of cellulases. These conditions promote the expression of cellulases towards β-glucosidase production, unlike materials with lower cellulose/lignin ratios like BS and RH, which exhibited lower cellulase activity. The optimal conditions for cellulase production by A. niger ITV02 were determined to be 33 °C, pH 5.3, and 200 rpm, resulting in a 1.7-fold increase in Exoglucanase (FPase activity) (from 0.127 to 0.215 U/mL). These findings demonstrate the potential to enhance FPase activity by utilizing substrates with high cellulose/lignin content and implementing optimal operational conditions without the need to raise the nitrogen content of the basal medium, thus mitigating the economic impact of cellulase production.

Abstract:Using PEG-based deep eutectic solvents (PDES), the current study proposes extractive fermentation as a sustainable process integration for the production and purification of α-amylase from Bacillus simplex (ON754233). Glucose: PEG 400 outperformed five PDES in terms of tie lie length (58) and slope value (1.23) against sodium sulphatt. Apple cider pomace was used as a low-cost, sustainable carbon source to produce-amylase, with a maximum enzyme production of 2200.13 U/mL. PDES concentration (20% w/v), salt (12.75 w/v), and apple waste (2.75 g/mL) were all optimized using response surface methodology. When scaled upto 3 L benchtop bioreactor, extractive fermentation was proved to be better technology with maximum recovery of 92.4% with highest partition coefficient (3.59). The partially purified enzyme was further purified using a Sephadex G 100 followed by DEAE-Sephadex anion exchange chromatography with a purity fold of 33. The enzyme was found to be thermostable at the temperature (60 °C), remains alkaline (pH 8), and the activity was stimulated in the presence of Mg 2+ ions. With SDS PAGE electrophoresis, the molecular weight was found to be around 140 kDa. Finally, the enzyme kinetics parameters were evaluated with observed K m (0.00396 mM) and V max (37.87 U/mL). Thus scaling up extractive fermentation entails increasing production capacity with improved extraction efficiency using green solvents.

Abstract:Currently petroleum-based fossil fuels are the main source of the production of energy. The major issue is that all these resources are non-renewable. As the demand of fuel increases day by day, these non-renewable resources may deplete shortly and will not meet the supply criteria according to the requirements. To overcome this problem, the production of bioethanol using wastes biomass such as fruit peels, agricultural waste, municipal and kitchen waste etc. has gained considerable attention. As these agro-wastes mainly contain lignocellulosic biomass, lignocellulolytic bacterial cultures harness the full potential of these substances. In the present study, endoglucanase producing Bacillus cereus ARA-12 was isolated from soil samples. Fermentation parameters to produce endoglucanase were optimized. The optimum production of endoglucanase was achieved in the medium containing carboxymethyl cellulose (15g/L), yeast extract (30g/L), KH2PO4 (1g/L), K2HPO4 (1.45g/L), MgSO4 (0.4g/L), CaCl2 (0.05g/L) and FeSO4 (0.00125g/L). The optimum temperature and pH of the medium were found to be 50°C and 8 respectively. The maximum enzyme production was achieved at agitation speed of 120 rpm after 20 hours of fermentation by using 5% inoculum. Maximum of 35.9 % bioethanol was produced by the action of endoglucanase enzyme on sugarcane bagasse whereas sweet potato, rice bran, banana peel, corn cob, potato peel and corn husk were also found to be potential raw material to produce second-generation biofuel.

Abstract:The paper demonstrates a novel two-peak negative differential resistance (NDR) circuit combining Si-based metal-oxide-semiconductor field-effect-transistor (MOS) and SiGe-based heterojunction bipolar transistor (HBT). Compared to the resonant-tunneling diode, MOS-HBT-NDR has two major advantages in our circuit design. One is that the fabrication of this MOS-HBT-NDR-based application can be fully implemented by the standard BiCMOS process. Another is that the peak current can be effectively adjusted by the controlled voltage. The peak-to-valley current ratio is about 4136 and 9.4 at the first and second peak respectively. It is very useful for circuit designers to consider the NDR-based applications.
Optimizing Exopolysaccharide Production by Bacillus subtilis Using Spoiled Fig and Grape

Abstract:The study aimed to enhance exopolysaccharides (EPSs) production by the bacterial strain Bacillus subtilis ES (OR501464) isolated from sugar cane juice. Spoiled grape and fig extract were utilized as cost-effective substrates for EPS synthesis by B. subtilis ES (OR501464), and the impact of nutritional factors on EPS synthesis was assessed. Among nineteen bacterial isolates evaluated for EPS production, the isolate with the highest EPS yield was identified through a combination of phenotypic and genotypic analyses. The optimization process revealed that the highest EPS yield of 4.7 g/L was achieved in a production medium containing 4% sucrose, 0.1% NaNO 3 , 0.002% Na 2 SO4, and 2% NaCl at 30 °C and pH 9. Additionally, the study explored EPS generation by B. subtilis ES (OR501464) using spoiled grape and fig extract as substrates. The addition of 2% NaCl to spoiled grape extract increased EPS production to 4.357 mg/mL compared to 3.977 mg/mL with grape alone. However, 2% NaCl did not enhance EPSs production in fig waste. Supplementing spoiled fig or grape extract with 0.2 g/L Na 2 SO 4 and 1 g/L NaNO 3 increased EPS production by B. subtilis ES (OR501464). The EPS was analyzed using GC–MS and FTIR spectroscopy for partial characterization. The study found that spoiled figs and grapes can be used as effective substrates for EPS production. The highest yield was achieved by adding 0.2 g/L Na 2 SO 4 and 1 g/L NaNO 3 . This study highlights the use of spoiled figs and grapes to produce valuable biopolymers, promoting sustainable and eco-friendly bioprocessing technologies.

Abstract:Abstract Cellulase is an important enzyme in present-day of industrial biotechnology. The current study is concerned with the production and partial characterization of cellulase enzyme from Bacillus sp. The effect of various fermentation conditions for cellulase production through shake-flask culture was investigated. Maximum enzyme production was obtained after 24 hours of incubation in fermentation medium with pH 3.5 at 35°C under having agitation at 150 rpm while inoculums volume 1% was applied. Enzyme production was 1.91 times higher after optimizing the production condition as compared to the basal media. Cellulase characterization revealed that optimum activity was at pH 5.5 and 50°C for 50 minutes. About 68% of the activity retained after heating the crude enzyme solution at 50°C for 30 minutes. This nature makes cellulase a suitable candidate for current mainstream biomass conversions for sustainable agriculture and industrial processes.

Abstract:The dwindling supply of the petroleum product and its carbon footprint has initiated search for a sustainable fuel and alternate feed-stocks. One such underexplored feedstock is chitin, a waste derived from sea food processing. The limitation of insolubility and crystallinity inherent in chitin is addressed with the chitin hydrolysates. In the present study, a chitinases producing marine isolate was isolated from the sediments of Arabian Sea from a depth of 20 m. In order to increase the expression of the chitinases, sequential optimisation using one factor at a time and Taguchi experimental designs were employed which resulted in a yield of 13.46 U/mL which was 2.62 fold higher than the initial bioprocess condition values. In a two-step refinery protocol, Candida albicans was evolved towards chitooligosaccharides using chemically synthesized hydrolysates. In a fed –batch fermentation design the Candida yielded a 12.8 % conversion of these commercial chitin oligosaccharides into bioethanol in a run time of 48 h. This is the first report demonstrating the potential of Candida to utilise chitin oligosaccharides for the production of bioethanol.

Abstract:Polygalacturonase represents an important enzyme of the pectinase consortium in the food and beverage industry. The present study aims to isolate, screen, and enhance physicochemical parameters for enzyme production under submerged fermentation. A total of 34 fungal strains were obtained from rotten fruits and 70.6 % of them are able to produce polygalacturonase by degrading pectin in pectin screening agar. The fungal isolate (SO5) that exhibited the greatest diameter of the hydrolysis zone (45.3 ± 0.04 mm) was selected for further study and later identified as Aspergillus sp. SO5, based on morphological characteristics using light microscopic observation. Several physicochemical parameters improved for pectinase production including incubation temperature, initial pH, agitation speed, inoculum density, and carbon and nitrogen sources. Under improved conditions, the polygalacturonase activity and fungal biomass increased up to 74.63 and 69.44 %, respectively. The finding also revealed that the improved physicochemical conditions enhance enzyme production and shorten the incubation period from 6 days to 5 days. HIGHLIGHTS This research paper highlighted the capability of indigenous fungus (Aspergillus) to produce polygalacturonase. The finding revealed that the improved physicochemical conditions including agitation speed, inoculum size, the concentration of pectin, and incorporation of yeast extract in the culture medium could enhance polygalacturonase production by up to 74.63 % (34.21 ± 0.49 U/mL). The improved physicochemical conditions not only enhance the polygalacturonase production but shorten the incubation period to obtain optimum enzyme production. GRAPHICAL ABSTRACT
