Abstract:Polarimetry, which seeks to measure the vectorial information of light modulated by objects, has facilitated bionic geolocation and navigation applications. It is a novel and promising field that provides humans with a remote sensing tool to exploit polarized skylight in a similar way to polarization-sensitive animals, and yet few in-depth reviews of the field exist. Beginning with biological inspirations, this review mainly focuses on the characterization, measurement, and analysis of vectorial information in polarimetry for bionic geolocation and navigation applications, with an emphasis on Stokes–Mueller formalism. Several recent breakthroughs and development trends are summarized in this paper, and potential prospects in conjunction with some cutting-edge techniques are also presented. The goal of this review is to offer a comprehensive overview of the exploitation of vectorial information for geolocation and navigation applications as well as to stimulate new explorations and breakthroughs in the field.

Abstract:Biophotonics is a research and technology field that sits at the interface of photonics and biomedical applications, which includes clinical diagnosis, therapeutics, biotechnology, and the environment. The integration of optics and biology has been one of the most active technology areas in the past three decades, not only in curiosity-driven research but also in terms of how these technologies are translated into practice with visible impacts within a relatively short time. A prominent example is the use of lasers and photonics technologies in ophthalmology applications such as laser vision correction, artificial lenses in cataract surgery, and retinal detachment repairs. The adoption of photonics technologies in medicine is significantly faster than other medical devices in a relatively cautious and heavily regulated area, mostly owing to its natural compatibility with minimally invasive procedures. Biophotonics is an interdisciplinary and multidisciplinary area at the same time. Even for a specific application, e.g., laser vision correction, many different photonics technologies may be used in combination: UV laser ablation in cornea reshaping, ultrafast laser ablation for cap cutting, image processing and modeling for procedure planning and design, and real time eye movement tracking. Many optical modalities do not require direct contact with the target, which allows several methods to be multiplexed, e.g., to provide additional sources of diagnostic contrasts or real-time feedback and guidance for therapy.
Polarisation optics for biomedical and clinical applications: a review

Abstract:Many polarisation techniques have been harnessed for decades in biological and clinical research, each based upon measurement of the vectorial properties of light or the vectorial transformations imposed on light by objects. Various advanced vector measurement/sensing techniques, physical interpretation methods, and approaches to analyse biomedically relevant information have been developed and harnessed. In this review, we focus mainly on summarizing methodologies and applications related to tissue polarimetry, with an emphasis on the adoption of the Stokes-Mueller formalism. Several recent breakthroughs, development trends, and potential multi-modal uses in conjunction with other techniques are also presented. The primary goal of the review is to give the reader a general overview in the use of vectorial information that can be obtained by polarisation optics for applications in biomedical and clinical research.

Computational and Mathematical Methods in Medicine

Abstract:Increasingly wider availability of biomedical imaging modalities, such as X-ray computed tomography (CT), magnetic resonance imaging (MRI), ultrasonic imaging, positron emission tomography (PET), single-photon emission computed tomography (SPECT), and optical imaging, have led to significant progresses in both biomedical research and clinical practice. Advancements in imaging technology and image informatics have facilitated and enabled quantitative understanding of the biological structures, functional organisms, and pathological mechanisms. This special issue presents 14 latest research contributions in computational biomedical imaging. The focuses have been on the computational principles, strategies, methods, and algorithms that create, extract, and understand imaging data of biomedical significance. The first group of papers has showcased the development and applications of rigorous optimization techniques in biomedical imaging. The work based on dual unscented Kalman filtering provides a reliable and efficient platform to understand the BOLD signals from fMRI. In the other paper, the robust discrete particle swarm optimization algorithm is adopted to solve the branch-cut phase unwrapping problem of MRI data. Without the need for a regularization penalty term, a semigreed method for bioluminescence tomography reconstruction is fast and stable for small animal imaging. The second group of papers has addressed image segmentation and tissue characterization problems, ever popular among image analysis researchers. The work of L. Li et al. applies machine learning techniques to characterize lymph node metastasis in gastric cancer from gemstone spectral imaging with much improved outcomes, while G. V. Sanchez. Ferrero et al. make new advances in ultrasonic speckle characterization using generalized Gama distribution and generalized Gama mixture model. In recent years, physically meaningful modeling and simulation have become integral components for model-constrained imaging and model-based image understanding. Thus, the third group of papers has continued the stride in this direction. The propagation of myocardial electrical activation is simulated using a monodomain model, without the need of explicit mesh constraints. In similar spirit, one work performs forward modeling of 3D optical molecular imaging with simplified spherical harmonics approximation using extended finite element method. The final group of papers has highlighted some new imaging modalities and their potential use in clinical practice. The further development of ultrasound-based quantitative imaging of Young's modulus with high accuracy is presented in this issue. And the novel Diffusion Kurtosis Imaging shows that more detailed classification of brain tissues is clinical significance. We appreciate the many high quality submissions to this special issue from leading researchers in computational biomedical imaging. Because of space limitation, we can only accept a small number of most exciting works. We sincerely hope that the remaining interesting research outcomes will be soon published in other venues.

Abstract:The rapid growth in laser and photonic technology has resulted in new tools being proposed and developed for use in the medical and biological sciences. Specifically, a discipline known as biomedical optics has emerged which is providing a broad variety of optical techniques and instruments for diagnostic, therapeutic and basic science applications. New laser sources, detectors and measurement techniques are yielding powerful new methods for the study of diseases on all scales, from single molecules, to specific tissues and whole organs. For example, novel laser microscopes permit spectroscopic and force measurements to be performed on single protein molecules; new optical devices provide information on molecular dynamics and structure to perform `optical biopsy' non-invasively and almost instantaneously; and optical coherence tomography and diffuse optical tomography allow visualization of specific tissues and organs. Using genetic promoters to derive luciferase expression, bioluminescence methods can generate molecular light switches, which serve as functional indicator lights reporting cellular conditions and responses in living animals. This technique could allow rapid assessment of and response to the effects of anti-tumour drugs, antibiotics, or antiviral drugs. This issue of Physics in Medicine and Biology highlights recent research in biomedical optics, and is based on invited contributions to the International Conference on Advanced Laser Technology (Focused on Biomedical Optics) held at Cranfield University at Silsoe on 19--23 September 2003. This meeting included sessions devoted to: diffuse optical imaging and spectroscopy; optical coherence tomography and coherent domain techniques; optical sensing and applications in life science; microscopic, spectroscopic and opto-acoustic imaging; therapeutic and diagnostic applications; and laser interaction with organic and inorganic materials. Twenty-one papers are included in this special issue. The first paper gives an overview on the current status of scanning laser ophthalmoscopy and its role in bioscience and medicine, while the second paper describes the current problems in tissue engineering and the potential role for optical coherence tomography. The following seven papers present and discuss latest developments in infrared spectroscopy and diffuse optical tomography for medical diagnostics. Eight further papers report recent advances in optical coherence tomography, covering new and evolving methods and instrumentation, theoretical and numerical modelling, and its clinical applications. The remaining papers cover miscellaneous topics in biomedical optics, including new developments in opto-acoustic imaging techniques, laser speckle imaging of blood flow in microcirculations, and potential of hollow-core photonic-crystal fibres for laser dentistry. We thank all the authors for their valuable contributions and their prompt responses to reviewers' comments. We are also very grateful to the reviewers for their hard work and their considerable efforts to meet tight deadlines.

Abstract:Polarimetry is a powerful optical tool in the biomedical field, providing more comprehensive information on the sub-wavelength micro-physical structure of a sample than traditional light intensity measurement techniques. This review summarizes the concepts and techniques of polarization and its biomedical applications. Specifically, we first briefly describe the basic principles of polarized light and the Mueller matrix (MM) decomposition method, followed by some research progress of polarimetric measurement techniques in recent years. Finally, we introduce some studies on biological tissues and cells, and then illustrate the application value of polarization optical method.

Polaromics: Deriving Polarization Parameters from a Mueller Matrix for Quantitative Characterization of Biomedical Specimen

Abstract:A Mueller matrix is a comprehensive representation of the polarization transformation properties of a sample, encoding very rich information on the microstructure of the scattering objects. However, it is often inconvenient to use individual Mueller matrix elements to characterize the microstructure due to a lack of explicit connections between the matrix elements and the physics properties of the scattering samples. In this review, we summarize the methods to derive groups of polarization parameters, which have clear physical meanings and associations with certain structural properties of turbid media, including various Mueller matrix decomposition (MMD) methods and the Mueller matrix transformation (MMT) technique. Previously, experimentalists have chosen the most suitable method for the specific measurement scheme. In this review, we introduce an emerging novel research paradigm called 'polaromics'. In this paradigm, both MMD and MMT parameters are considered as polarimetry basis parameters (PBP), which are used to construct polarimetry feature parameters (PFPs) for the quantitative characterization of complex biomedical samples. Machine learning techniques are involved to find PFPs that are sensitive to specific micro- or macrostructural features. The goal of this review is to provide an overview of the emerging 'polaromics' paradigm, which may pave the way for biomedical and clinical applications of polarimetry.

Abstract:Polarization imaging and sensing techniques have shown great potential for biomedical and clinical applications. As a novel optical biosensing technology, Mueller matrix polarimetry can provide abundant microstructural information of tissue samples. However, polarimetric aberrations, which lead to inaccurate characterization of polarization properties, can be induced by uneven biomedical sample surfaces while measuring Mueller matrices with complex spatial illuminations. In this study, we analyze the detailed features of complex spatial illumination-induced aberrations by measuring the backscattering Mueller matrices of experimental phantom and tissue samples. We obtain the aberrations under different spatial illumination schemes in Mueller matrix imaging. Furthermore, we give the corresponding suggestions for selecting appropriate illumination schemes to extract specific polarization properties, and then provide strategies to alleviate polarimetric aberrations by adjusting the incident and detection angles in Mueller matrix imaging. The optimized scheme gives critical criteria for the spatial illumination scheme selection of non-collinear backscattering Mueller matrix measurements, which can be helpful for the further development of quantitative tissue polarimetric imaging and biosensing.

Abstract:Faraday rotation of polarised background sources is a unique probe of astrophysical magnetic fields in a diverse range of foreground objects. However, to understand the properties of the polarised sources themselves and of depolarising phenomena along the line of sight, we need to complement Faraday rotation data with polarisation observations over very broad bandwidths. Just as it is impossible to properly image a complex source with limited u-v coverage, we can only meaningfully understand the magneto-ionic properties of polarised sources if we have excellent coverage in $\lambda^2$-space. We here propose a set of broadband polarisation surveys with the Square Kilometre Array, which will provide a singular set of scientific insights on the ways in which galaxies and their environments have evolved over cosmic time.
