Abstract:Three systems included 1:1 beta-cyclodextrin inclusion complex with m-cresol in vacuum, nano water drop and nano acetone drop have been studied using molecular dynamics with pcff force field and without periodic boundary condition. Solvent effects have been discussed. The pre-equilibrated modelling inclusion complex in nano water drop and acetone drop contained two water and two acetone molecules respectively, and during the simulation one water molecule was observed to dissociate from the host in water drop while no dissociation was observed in acetone case. This result well validates the dynamic hydrophobicity of the beta-cyclodextrin. Many analytical results indicate that solvent has an important effect on the stability and dynamic structure of the beta-cyclodextrin inclusion complex.

Due to their excellent biologic and optical properties, cyclodextrins have recently attracted increasingly more attention. In this review the newest development of the supramolecular self-assembly based on cyclodextrin is summarized. The article provided detail information about the synthesis and applications of both of rotaxanes formed by cyclodextrin ring and polymer axes, and supramolecular polymer without polymer backbone.
Viscoelastic Study of Single Chain Cyclized Polymeric Nanoparticles and Linear Polymers Blends

Abstract:It is of great importance to polymerize multivinyl monomers and simultaneously circumvent uncontrollable polymerization process and low gel point. Hereby, a type of divinyl monomer: ethylene glycol dimethacrylate (EGDMA) was successfully homopolymerized by kinetically controlled Cu-0 and Cu-II-meadiated reversible-deactivation radical polymerization (RDRP). The kinetic study showed a well-controlled polymerization behaviour. During the initial stage, a linear chain propagation was observed with a low polydispersity (< 1.35) demonstrating the controlled/living polymerization feature. However, the products obtained during this stage showed a high consumption of pendent vinyl groups (around 28%) based on the analysis by proton nuclear magnetic resonance (H-1-NMR). This high pendent vinyl conversion in the linear chain propagation stage is attributed to the significant involvement of the intramolecular cyclization. The promotion of the intramolecular cyclization and suppression of the intermolecular crosslinking is achieved by the low kinetic chain length and low polymer volume concentration. Given the coexistence of linear chain propagation and high pendent vinyl conversion, the novel single chain cyclized polymer structure was formed during this process, which was further used as polymeric nanoparticles to modify the viscoelasticity of linear poly(methyl methacrylate) (PMMA) melts. The essentially similar chemical structure between the EGDMA and MMA ensures the minimization of enthalpic interactions and dispersion forces between the nanoparticles and linear melts. The oscillatory rheology study and the subsequent time temperature superposition based on the Williams-Landel-Ferry equation demonstrated the coexistence of the segmental relaxation at high frequencies and chain relaxation at low frequencies. The complex viscosity studied as a function of the shear frequency showed the decrease in terminal viscosity with addition of 10 wt% single chain cyclized polyEGDMA nanoparticles. A further decrease was also observed with the addition of 20 wt% nanoparticles, which is not in agreement with Einstein relationship between the complex viscosity and fraction of added particles. This strategy provides a simple and efficient approach to control polymerization of multivinyl monomers, and the unexpected decrease in viscosity of the blend system should in principle open a new avenue to modify polymer melts and molten systems.
Solvent-Free Formation of Cyclodextrin-Based Pseudopolyrotaxanes of Polyethylene Glycol: Kinetic and Structural Aspects

Abstract:Pseudopolyrotaxanes (PPRs) are supramolecular structures consisting of macrocycles able to thread on a linear polymer chain in a reversible, non-covalent way, often referred to in the literature as “molecular necklaces”. While the synthesis and reaction mechanisms of these structures in solution have been widely described, their solvent-free production has received little attention, despite the advantages that this route may offer. We propose in this work a kinetic mechanism that describes the PPR formation in the solid phase as a process occurring in two consecutive stages. This mechanism has been used to investigate the spontaneous formation of a PPR that occurs when grinding α-Cyclodextrin (α-CD) with polyethylene glycol (PEG). In the threading stage, the inclusion of the polymer and subsequent release of the water molecules lodged in the cavity of the macrocycle cause vibrational changes that are reflected in the time-dependence of the FTIR-ATR spectra, while the further assembly of PPRs to form crystals produces characteristic reflections in the XRD patterns, due to the channel-like arrangement of CDs, that can be used to track the formation of the adduct in crystalline form. The effects that working variables have on the kinetics of the reaction, such as temperature, feed ratio, molar mass of the polymer and the introduction of an amorphous block in the polymer structure, have been investigated. The rate constants of the threading step increase with the temperature and the activation energy of the process increases at lower proportions of CD to PEG. This is attributed to the lower degree of covering of the polymer chain with CDs that reduces the hydrogen-bonding driven stabilization between adjacent macrocycles. The formation of crystalline PPR, which takes place slowly at room temperature, is markedly promoted at higher temperatures, with lower proportions of CD favoring both the formation and the growth of the crystals. The molar mass of the polymer does not modify the typical channel-like arrangement of packed PPRs but the conversion into crystalline PPR diminishes when using PEG1000 instead of PEG400. At a microscopic level, the crystals arrange into lamellar structures, in the order of hundreds of nm, embedded in an amorphous-like matrix. The introduction of a polypropylene oxide block in the structure of the polymer (Pluronic L62) renders poorer yields and a considerable loss of crystallinity of the product of the reaction. The methodology here proposed can be applied to the general case of inclusion complexes of CDs with drugs in the solid phase, or to multicomponent systems that contain polymers as excipients in pharmaceutical formulations along with CDs.

Abstract:During synthesis and purification, compounds are exposed to various solvents. These solvents can, at times, assemble in the solid state. This association can arise from intermolecular interactions, allowing solvent molecules to bind with chemical substances and resulting in a unique crystalline structure known as a solvate. Nevertheless, such an association with solvents can substantially alter the properties of the compounds. In the crystalline solid state, the occurrence of solvates is recurrent among mechanically interlocked molecules (MIMs), including rotaxanes; however, solvates are frequently considered incidental or inconsequential. In this context, we sought to investigate the role and relevance of solvent molecules in the crystallization process of rotaxanes by systematically analyzing 24 models, employing the supramolecular cluster as a demarcation tool and using tetralactam-macrocycle based [2]rotaxanes. In general, the most frequently occurring [2]rotaxane solvates are with chloroform and water molecules (∼66%). The occurrence of solvates does not follow any rule or pattern, implying that similar structures can and do contain different solvates in varying proportions. By the crystallization mechanisms proposed, it was feasible to determine at which stage of the crystallization process the solvates are formed, and the energetic and topological contributions of the solvent molecules during crystallization were assessed. The most rotaxanes “trap” solvent molecules during crystallization, and interestingly, in certain instances, these solvent molecules are essential for crystallization, contributing up to 40% of the total stabilization energy of the supramolecular cluster.

Abstract:Cyclodextrins are highly functional compounds with a hydrophobic cavity capable of forming supramolecular inclusion complexes with various classes of molecules including surfactants. The resultant rich nanostructures and their dynamics are an interesting research problem in the area of soft condensed matter and related applications. Herein, we report novel dynamical supramolecular assemblies based on the complexation of β-cyclodextrin with 3 different sulfonic surfactants, which are sodium hexadecylsulfate, sodium dodecylbenzenesulfonate, and myristyl sulfobetaine. It was observed that a β-cyclodextrin : surfactant/2 : 1 molar ratio was ideal for inducing axial growth and imparting large viscosities in the suspensions. Such complexation processes were accompanied by intriguing nanostructural phase behaviors and rheological properties that were very sensitive to the molecular architecture of sulfonic surfactants. The presence of an amino group in the head group of the surfactant allowed for large viscosities that reached 2.4 × 104 Pa s which exhibited gel-like behavior. In contrast, smaller viscosity values with a lower consistency index were observed when a bulky aromatic ring was present instead. DIC microscopy was used to visually probe the microstructure of the systems with respect to sulfonate molecular architecture. Additionally, surface tension measurements, and FTIR and NMR spectroscopies were used to gain insights into the nature of interactions that lead to the complexation and nanostructural characteristics. Finally, mechanics correlating the supramolecular morphologies to the rheological properties were proposed.

Abstract:The cross-linking point of a conventional chemical cross-linking agent is fixed. Therefore, gels that are prepared with a conventional cross-linking agent have poor deformability, strength, shear resistance, and further properties. Some researchers have prepared a new cross-linking agent using cyclodextrin (CD). In a polyrotaxane cross-linking agent, the cross-linking points can slide freely along the molecule chain. The special "slide ring" structure can provide better elongation, strength, and other properties to gels, which can effectively expand the application of the gel's materials. This paper summarizes the preparation methods and applications from different types of CD and compares the improvements of properties (swelling, viscoelastic properties, etc.). In addition, the current results of our group are presented, and some ideas are provided for the development of polyrotaxane cross-linking agents.

Abstract:This paper reports a large volume change in a cholesterol based molecular gel in the presence of multi-amine as the gelling solvents dissociated from the gel in a dynamic manner. The release capability can be controlled by the ratio of multi-amine. The shrinkage capability can be restored and recycled by a gel-sol transformation. The mechanism of the shrinkage was followed by SEM, confocal laser scanning microscopy and X-ray diffraction studies. A structural evolution from lamellar vesicles to a more compact hexagonal structure driven by competitive intermolecular hydrogen bonds was proved to dominate the process of the solvent release. Moreover, the process can be stopped by addition of organic acid such as trifluoroacetic acid, which favors the application of this kind of material for molecular delivery systems.

Abstract:A prototype, based on light-active fluorescent rotor grafted to beta-cyclodextrin, shows a good solvent viscosity-sensitive behavior due to the environment-dependent nonradiative decay. With the reversible photoisomerization of the cyanostilbene unit, the viscosity sensitivity of the molecular rotor could be locked and activated, and the two switchable states can be distinguished by fluorescent signals. This cyclodextrin derivative was threaded to form a novel polypseudorotaxane. Such supramolecular assembly displays a lockable ratiometric fluorescent viscosity sensitivity with two emission channels: one aroused by fluorophore's intramolecular excimer without influenced by viscosity is used to gauge the concentration of the compound, while the other corresponding to the monomer's rotor fluorescence acts as a viscosity-sensitive signal and it can be shut off by UV irradiation.

Abstract:A novel gelation process is demonstrated in self-assemblies of amphiphilic star-shaped macromers of poly(L-lactide)-b-poly(ethylene glycol) bearing methacryloyl terminals with α-cyclodextrins. A physical hydrogel is first formed by inclusion complexation, and subsequently it is changed when a chemically crosslinked hydrogel under UV irradiation; a photoinitiator is added in advance, and this leads to a great improvement in the strength of the hydrogel. The effects of the degree of polymerization of the lactide oligomers as well as the amounts of α-cyclodextrins added on the gelation time are described in detail. The analytical results show that most of the α-cyclodextrins added are still threaded and immobilized onto the network chains after photopolymerization. A study of the gel content and equilibrium swelling behaviors of the crosslinked hydrogel has been performed.

Abstract:We have rationally designed a one-dimensional coordination polymer (1D CP), termed 1D-DGIST-18, that exhibits intrinsic structural flexibility. This 1D CP enables its expansion into a three-dimensional network through supramolecular interactions involving coordinated solvents and/or ligands. The strategic selection of solvents for solvent exchange, prior to drying, significantly influences the structures of 1D-DGIST-18 by removing certain coordinating solvents and modulating π–π stacking. Consequently, a hierarchical porosity emerges, ranging from micro- to meso- to macroporous structures, which is attributed to its inherent structural dynamics. Additionally, the formation of excimers endows 1D-DGIST-18, when immersed in acetone, with 'turn-on' fluorescence, as evidenced by fluorescence decay profiles. These structural transitions within 1D-DGIST-18 are further elucidated using single-crystal X-ray diffractometry. The insights from this study provide a foundation for the design of materials with structural dynamics and tunable properties.

Abstract:Cyclodextrins are a family of oligosaccharides with a toroid shape which exhibit a unique ability of entrapping guest molecules in their internal cavity. Water is the primary guest molecule and is omnipresent in the crystalline phases stabilizing the overall architecture. Despite the presence of water molecules inside the cavity, cyclodextrins provide a hydrophobic environment where poorly soluble molecules can easily fit. In this investigation we put in evidence different types of water in the hydrated {\alpha} , \b{eta} and {\gamma} cyclodextrin crystals. Thermogravimetric measurements identify various binding sites of water and highlight the difference between the crystals equilibrated under various humid atmospheres. We establish by microcalorimetry the limit of solubility versus temperature and measure for the first time the melting temperatures of the hydrated crystals. Dissolution and melting enthalpies are derived and the solubility curves are compared to existing literature. The specific features of each cyclodextrin are underlined.

Abstract:An efficient access to prepare microgels from polyrotaxanes (PRXs) with characteristic thermoresponsiveness and photocontrolled degradation is reported. The microgels were formed by cross-linking the thermally collapsed spherical aggregates from thermoresponsive cyclodextrin PRXs in aqueous solutions above their phase transition temperatures through reaction of glutaraldehyde with acylhydrazine moieties to form the dynamic arylhydrazones. These microgels have uniform spherical morphologies with controllable sizes and tunable cross-linking densities, which retained characteristic thermoresponsive properties in water with cross-linking density-dependent thermally induced deswelling behavior. The spreading and deformation behavior of the microgels on solid substrates was investigated by AFM, which was found to be dominated by the cross-linking density. On the basis of the mechanically interlocked architecture, these cross-linked PRX microgels were stably dispersed in water but can be quickly degraded into small components upon UV cleavage of bulky stoppers. These photodegradable thermoresponsive microgels with dynamic cross-linked structures would hold great potential for biomedical and sensing applications.

Abstract:Desolvation processes, though common in self-assembled biological structures, are rarely evidenced and utilized in the design of crystalline architectures. In this study, we introduce a novel approach using the [Mo8S8O8(OH)8(guest)]2- complex, formed by the self-condensation of four [MoV 2O2S2]2- fragments around a guest unit (MoVIO6H4 or oxalate), as a chaotropic scaffold for crystallizing hybrid organic-inorganic systems with natural cyclodextrins. Our findings reveal that β-cyclodextrin (β-CD) facilitates the formation of host-guest complexes, while α-cyclodextrin (α-CD) induces the formation of a Kagome-type structure with significant voids. These new compounds were thoroughly characterized using X-ray diffraction (both powder and single-crystal), N2 adsorption, elemental and thermogravimetric analysis. Additionally, solution studies using 1H NMR titration and small-angle X-ray scattering (SAXS) demonstrated pre-association of the building units in solution. These results enhance our understanding of the design principles for supramolecular structures composed of inorganic polyanions and cyclodextrins.
Solvent effects on the motion of a crown ether/amino rotaxane

Abstract:Solvents have been recognized as a significant factor for modulating the shuttle of rotaxanes and regulating their functions regarding molecular machines by a lot of published studies. The mechanism of the effects of solvents on the motion of crown ether/amino rotaxanes, however, remains unclear. In this work, a rotaxane, formed by dibenzo-24-crown-8 (C[8]) and a dumbbell-shaped axle with two positively charged amino groups, was investigated at the atom level. Two-dimensional free-energy landscapes characterizing the conformational change of C[8] and the shuttling motions in chloroform and water were mapped. The results indicated that the barriers in water were evidently lower than those in chloroform. By analyzing the trajectories, there was no obvious steric effect during shuttling. Instead, the main driving force of shuttling was verified from electrostatic interactions, especially strong hydrogen bonding interactions between the axle and water, which resulted in the fast shuttling rate of the rotaxane. All in all, the polarity and hydrogen bond-forming ability of solvents are the main factors in affecting the shuttling rate of a crown ether/amino rotaxane. In addition, C[8] would adopt S-shaped conformations during shuttling except for situating in the amino sites with C-shaped ones adopted due to π–π stacking interactions. The results of this research improve the comprehension of the solvent modulation ability for shuttling in crown ether-based rotaxanes and illustrate the effects of structural modifications on motions. These new insights are expected to serve the efficient design and construction of molecular machines.

Abstract:A supramolecular gelator based on the bistable [c2]daisy chain rotaxane is designed and synthesized. The reversible actuation of the [c2]daisy chain renders the formed supramolecular gel with acid/base-responsive switching between gel and monomer solution. The efficient switching process is attributed to the ring-sliding effect of the rotaxane in response to acid/base stimuli.The ring-inhibited hydrogen bonding stacking results in a nearly quantitatively disassembly of the gel network after addition of base which is hard to be realized by traditional heating strategy in hydrogen-bonding-supported gel.

Abstract:4-(4′-Butoxyphenyl)phenyl-β-O-d-glucoside was synthesized as a low-molecular-mass gelator. It was able to immobilize a variety of aqueous and organic solvents in large amounts through the formation of three-dimensional self-assembled fibrillar networks. The morphologies of the aggregates depended on the nature of solvent where they were formed. Planar ribbons were observed in water, while helical ribbons were dominant in toluene and sheets in CHCl3. The xerogel from water exhibited a lamellar structure with a d-spacing of 2.45 nm as demonstrated by 1D-WAXD, indicating a bilayer structure interdigitated with butoxy tails, whereas the xerogel from CHCl3 or toluene yielded a lamellar structure with a d-spacing of 3.05 nm, implying a bilayer structure interdigitated with glycosyl heads. Increasing the content of 1,4-dioxane in water caused a gradual transformation from planar to twisted ribbons and then tubes.

Abstract:Front Cover: Degradable/erodible polyrotaxane solid materials/hydrogels exhibiting physical hydrogen bonds as crosslinks between α‐cyclodextrins are prepared by introducing hyperbranched polyglycerol into the α‐cyclodextrins. The obtained gels recover their original shape upon pulling and releasing. Once the gels are immersed into water, the gels are rapidly eroded in a controlled manner depending on the number of hydrogen bonds and the branched degree. This is reported by Kiyotaka Tanaka and Tooru Ooya in article number 2300294.

Abstract:A series of carbazole-based vinyl-benzoxazole derivatives have been synthesized in order to verify whether X-ray diffraction (XRD) simulation can give more information about intermolecular stacking in the gel phase. It was found that their gelation capabilities were strongly dependent on the length of the alkyl chain. The compounds with shorter alkyl chains have lower critical gelation concentrations (CGCs) in nonpolar alkane and alcohols with longer carbon chains. On the other hand, compounds with long alkyl chains presented small CGCs in polar methanol. Powder XRD structure solution gave more information about intermolecular stacking than the traditional way of analyzing diffraction peaks to derive approximate molecular stacking patterns. The results verified that gelators had a similar head-to-tail π-stacking between aromatic groups in gel phases although different slipping angles existed. Moreover, ordered stacking between the alkyl chains was also present.

Abstract:This study explores the liquid crystalline properties of novel amphiphilic β-cyclodextrin derivatives functionalized with seven oligoethylene glycol chains at the primary face, terminated with either an O-methyl or an O-cyanoethyl group, and fourteen hydrophobic aliphatic chains (elaidic or oleic acids) at the secondary face. These derivatives were designed to study the impact of chain conformation and terminal group polarity on their mesomorphic behavior. Thermal, microscopic, and X-ray diffraction studies revealed that the elaidic derivatives form columnar hexagonal mesophases, with the O-cyanoethyl derivative undergoing a slow, temperature-dependent transition to a bicontinuous cubic phase. The oleic derivatives, although less stable, also exhibit columnar hexagonal phases, but clear differences were observed in the clearing temperatures between these two groups of molecules, and they are also different from analogous derivatives containing no alkene functionalities. This work provides direct insights into the structure–mesomorphic property relationships of amphiphilic cyclodextrins in terms of the geometry and conformation flexibility of the hydrophobic regions, as well as the functional group attached to the end of the polar region.
