Abstract:With quantification techniques of the spinal bone condition, especially with spinal biphotonic absorptiometry, early screening of patients risking subsequent development of osteoporosis complicated with vertebral compression is possible. An investigation was conducted in Lower Normandy, in 386 women who had undergone menopause or an ovariectomy, with ages ranging between 40 and 56 years; 274 were in a peri-menopausal state. We had previously established a curve of the bone mineral content according to age. From this curve, in semilogarhythmic representation, we assessed each patient's risk. This risk is considered as high in 30 p. cent of the patients, non existent in 52 p. cent and 18 p. cent are borderline. The percentage of high risk patients increases with the number of years since menopause. It is hoped that the incidence of osteoporosis will decrease with early screening and preventive therapeutic measures.
Increasing conception rate by addition of a second prostaglandin injection in an Ovsynch and progesterone treatment programme for dairy cows not detected in oestrus before the start of the seasonal mating period

Abstract:To assess the effect on conception rate to first service, 3- and 6-week in-calf rates, final pregnancy rate and the hazard of pregnancy, of addition of a second prostaglandin (PG) injection 24 hours after the first, in a synchrony programme combining Ovsynch and progesterone in pasture-based dairy cows that had not been detected in oestrus before the start of the seasonal mating period.This prospective, negative-controlled study was conducted using anoestrous cows (n = 1,411) from nine spring-calving dairy herds in the Waikato region of New Zealand. Ovaries of cows not detected in oestrus were examined by transrectal ultrasonography and the presence of a corpus luteum determined. All cows were then treated with an intravaginal progesterone-releasing device and injected I/M with gonadorelin (Day −9). Cows were injected I/M with cloprostenol at device removal (Day −2) and on Day −1 were injected with either water (single PG; n = 720), or cloprostenol (double PG; n = 692). On Day 0 cows were injected again with gonadorelin, unless previously detected in oestrus, and were inseminated on Day 1. Based on dated pregnancy diagnosis by transrectal ultrasonography, cows were categorised as conceiving to first insemination, becoming pregnant in the first 3 or 6 weeks, or becoming pregnant by the end of the mating period.Cows in the double PG group had increased conception rate to first service (45.3 (95% CI = 45.1–45.4)% vs. 41.1 (95% CI = 41.0–41.3)%; p < 0.001), 3-week in-calf rate (53.4 (95% CI = 53.6–54.0)% vs. 49.2 (95% CI = 49.0–49.4)%; p < 0.001), and 6-week in-calf rate (67.2 (95% CI = 65.7-68.8)% vs. 63.5 (95% CI = 62.0-65.0)%; p = 0.014) compared to cows in the single PG group. Final pregnancy rate and hazard of pregnancy did not differ between treatment groups (p >0.2).Addition of a second PG injection 24 hours after the first, increased conception rate to first service and the proportion of cows pregnant by 3 and 6 weeks after commencement of the mating period.Improved outcomes can be achieved when treating those cows not detected in oestrus by the start of the seasonal mating period by addition of a second PG injection to the currently recommended treatment protocol for these animals.

The effect of low vs. high serum concentrations of progesterone during early and late stages of follicular development on follicle size and double ovulations of lactating dairy cows

Abstract:The objective of this study was to determine the effect of high vs. low circulating concentrations of progesterone (P4) during early and late stages of ovulatory follicular wave development on ovulatory follicle size and double ovulation rate in lactating dairy cows. Lactating dairy cows were synchronized with prostaglandin F2α (PG; 25 mg of dinoprost tromethamine, i.m.) then gonadotropin releasing hormone (GnRH; 100 µg of gonadorelin; i.m.) 2 d apart to initiate a new cycle. Only cows that started a new cycle were used in this study. On d 7 of the estrous cycle, all cows received GnRH (200 µg; i.m.), were blocked by parity and AI service number and then randomly assigned to four groups. Only animals that ovulated to this 2nd GnRH injection were used in the study. The day of this GnRH injection was considered d0 of the ovulatory follicular wave and treatment. PG and/or CIDRs were utilized to induce high (H) or low (L) circulating concentrations of P4 during either 0 to 4 d (early) and/or 5 to 7 d (late) of the ovulatory wave in a 2 x 2 randomized design, H/H, L/L, L/H and H/L. All cows (n=471) received PG 7 d after initiation of treatment. One d later, CIDRs were removed and a second PG was administered to ensure complete CL regression. GnRH was administered to all cows 56 h after the first of these two PGs to induce ovulation. Ovaries were evaluated by transrectal ultrasonography at time of GnRH administration and 2d later to determine size and number of ovulatory follicles. Variables were analyzed using the GLIMMIX procedure of SAS. There was an effect of treatment on mean diameter of single ovulating follicles (P < 0.001) and percentage of cows with double ovulations (P < 0.001). Mean diameter of single ovulating follicles was smaller for HH (16.8 ± 0.2 mm) compared to HL, LH and LL treatments (18.2 ± 0.3; 17.8 ± 0.2; 19.6 ± 0.3 mm, respectively; P < 0.02). HL and LH had smaller single ovulating follicles compared to LL (P < 0.001). Single ovulating follicles tended to be smaller in LH compared to HL (P = 0.09). Percentage of cows with double ovulations was greater in LL (51%) compared to HH, HL and LH (13, 34, and 32 %, respectively; P < 0.01). Cows in HH had lower rates of double ovulation compared to HL and LH (P < 0.001). An interaction between parity and service number for double ovulation rate was significant (P < 0.02) and determined that primiparous first service cows had lower double ovulation rates compared to primiparous later service cows and multiparous first and later service cows (14 vs. 36, 35, and 40 %, respectively; P < 0.001). In summary, these data indicate that serum concentrations of P4 during specific stages of the follicular wave directly affect size and number of ovulating follicles.

Optimisation of Early Pregnancy Diagnosis and Resynchronisation in Beef Cows

Abstract:The objectives of this study were (1) to investigate the effects of the preovulatory follicle (POF) size on the accuracy of Doppler-based early pregnancy detection, and (2) to determine whether the removal of PGF2α (PGF) treatment during the resynchronisation protocol would affect fertility in beef cows. In Experiment 1, Nelore suckling cows (n = 224) were enrolled in an estradiol-progesterone-based timed artificial insemination (TAI) protocol. At TAI, cows were separated based on the range of POF diameters, as follows: ≤11.0 mm (n = 50), 11.1-12.9 mm (n = 64), 13.0-14.4 mm (n = 62) and ≥14.5 mm (n = 48). On day 22 after TAI, the corpus luteum (CL) blood flow (CLBF) of all cows was examined by colour Doppler ultrasonography to diagnose nonpregnant cows. The cows with the largest POF had the greatest positive predictive value (88.6%; 31 of 35) and diagnostic accuracy (91.7%; 44 of 48). In Experiment 2, Nelore cows (n = 233) were subjected to the same TAI protocol. Fourteen days after TAI, all cows were started on a resynchronisation protocol. Cows diagnosed as nonpregnant based on CLBF, on day 22, received 0.5 mg estradiol cypionate intramuscular (im) and were assigned to receive either 150 μg of PGF (PGF; n = 50) or 2 mL of saline (control; n = 47). Cows treated with PGF had a P/AI of 30.0% compared with a 48.9% P/AI in controls (p = 0.06). Our findings demonstrate that the POF size affects the accuracy of a CLBF-based early pregnancy diagnosis and that the removal of PGF treatment from the resynchronisation protocol tended to increase P/AI of the second TAI.
Endometritis treatment with a PGF2alpha analog does not improve reproductive performance in a large dairy herd in Argentina

Abstract:In Argentina, most dairy cows with endometritis are treated with prostaglandin (PGF(2alpha) or its analogs) and insemination is withheld until there are no signs of endometritis. The objective of the present study was to evaluate if this method of managing endometritis enhances reproductive performance. Three experiments were conducted over 4 years in a large farm in the west of Buenos Aires province. In Experiment 1, half of the cows diagnosed with endometritis (>1.5-fold difference in diameter of uterine horns, as determined by rectal palpation) received standard endometritis management (treatment with tiaprost, a PGF(2alpha) analog, rectal palpation every 20 days, and withholding of AI until endometritis apparently resolved) and the other half was untreated, with AI at the first estrus after the voluntary waiting period. Untreated cows were inseminated and conceived 20 days earlier than treated cows, and the pregnancy rate by Day 90 postpartum was higher in the untreated group. In Experiment 2, cows with endometritis were divided into four groups according to the severity of symptoms; within each group, cows were allocated to treatment or control, as in Experiment 1. Although first service conception rate decreased as endometritis severity increased, reproductive performance in treated versus control cows was similar to that of Experiment 1 (with no interaction due to degree of endometritis). Re-evaluation of the treated cow (to confirm uterine "normality") may have been responsible for the delay in conception in both experiments. The objective of Experiment 3 was to determine the effects of tiaprost treatment on clinically normal postpartum cows (no evidence of endometritis). Tiaprost treatment reduced the interval from calving to conception in multiparous cows, but it delayed conception and reduced the conception rate in primiparous cows. In conclusion, treatment with tiaprost impaired reproductive performance in primiparous cows (in the absence of endometritis). Furthermore, the standard treatment for endometritis (treatment with a prostaglandin analog and withholding insemination until clinical signs abated) impaired reproductive performance and increased costs.

Abstract:In the present study, the kinetics of the prostaglandin F2alpha (PGF2alpha)-metabolite 15-keto-13,14-dihydro-PGF2alpha after a single intramuscular application of various doses of the natural PGF2alpha dinoprost at Day 7 of the cycle in the mare were investigated. Effects of low doses on estrous cycle length and life span of corpus luteum were examined, because release of PGF2alpha is still under discussion to have detrimental influence on success rates of transcervical transfer of equine embryos. Eight Shetland pony mares were each randomly assigned to each of four treatments: (a) 0.8 mg/100 kg (group T1), (b) 0.4 mg/100 kg (group T2), (c) 0.2 mg/100 kg BM dinoprost i.m. (group T3), and (d) 1 ml physiological saline i.m. (group CO). Treatments were administered as single doses on Day 7 of the estrous cycle. Administration of dinoprost caused dose-dependent rises of plasma concentrations of PGF2alpha-metabolite, although values of individual mares showed great variation within groups. Prostaglandin treatments resulted in a distinct decrease of plasma progesterone concentrations to values between 1.6 and 7.9 ng/ml within 24 h. Treatment groups had significantly lower progesterone area under the curve (AUC: T1 942.8+/-175.9, T2 1050+/-181.2 and T3 1117+/-179.8 ng/ml/h) when compared with controls (CO 1601.9+/-227.6; t-test, P<0.05 ). There was a small, but significant negative correlation between AUC of progesterone and of PGF2alpha-metabolite ( R=-0.4; P=0.05 ). Administration of PGF2alpha caused secretion of oxytocin in three (T1, T2) and two (T3) mares out of eight ranging from 19.3 to 63.1 pg/ml. The AUC of oxytocin was positively correlated with AUC of PGF2alpha-metabolite ( R=0.4, P<0.05) and negatively correlated with AUC of progesterone ( R=-0.4, P<0.05). Administration of dinoprost yielded significantly shorter intervals from treatment to estrus and ovulation (values in parentheses), respectively, when compared with controls: T1 3.9+/-0.7 days ( 12.1+/-0.7 days), T2 4.5+/-0.6 ( 12.3+/-0.6 ), T3 4.9+/-0.5 ( 12.3+/-0.6 ), and CO 8.9+/-0.6 days ( 16.5+/-0.8 days) (t-test, P<0.01 ) (Fig. 2). Different doses of PGF2alpha caused similar effects. Data suggest that progesterone concentrations at applications influence efficacy of treatments more than doses administered, as demonstrated by their high correlation with estrous cycle patterns. It is important to note that differences we achieved are gradual and that all mares responded to treatment by luteolysis and premature estrus, regardless of doses applied.

Abstract:The effect of treatment for anestrus in buffaloes with a PGF2α or GnRH injection and vitamin-mineral (Vit-M) supplementation for 1 to 2 months and some factors influencing the treatment effect were studied. In anestrus buffaloes with CL, an injection of PGF2α tended to show higher estrus detection and pregnancy rates within 17 days after treatment than Vit-M supplementation (P<0.10). In those with inactive ovaries, effect of GnRH and Vit-M did not differ. Body condition score of the animals before treatment affected pregnancy rate within 17 days after treatment (P<0.05). Pregnancy rate within 4 months after treatment was adversely influenced by low serum concentrations of calcium (P<0.01) and gastrointestinal parasitic infection before treatment (P<0.05).
Effects of low versus physiologic plasma progesterone concentrations on ovarian follicular development and fertility in beef cattle

Abstract:The objective of this study was to determine the effects of low versus physiologic plasma progesterone concentrations during the ovulatory wave on fertility in cattle. Suckled beef cows (Bos taurus; n=129) and pubertal heifers (Bos taurus; n=150) at random stages of the estrous cycle were given a luteolytic dose of prostaglandin F(2 alpha) (500 microg cloprostenol; PGF) twice, 11 d apart. Ten days after the second PGF treatment, cattle were given estradiol benzoate im (1.5 and 1.0mg for cows and heifers, respectively) and a progesterone-releasing intravaginal device (Cue-Mate) with a single pod containing 0.78 g progesterone (Day 0). Cattle in the low-progesterone group (n = 148) received a luteolytic dose of PGF on Day 0, whereas those in the high-progesterone (i.e., physiologic plasma concentrations) group (n=131) were allowed to retain their corpora lutea. On Day 8, the Cue-Mate was removed, and PGF was given to both groups. Fifty-four hours to 56 h later, cattle received 12.5mg of porcine LH (pLH) im and were concurrently artificially inseminated. The dominant follicle in the low-progesterone group was larger (P<0.001) than that in the high-progesterone group on the day of insemination (14.9+/-0.3mm vs. 12.7+/-0.3mm, mean+/-SEM). At 7 d after ovulation, the low-progesterone group had a larger corpus luteum (24.5+/-0.54 mm vs. 21.9+/-0.64 mm, P<0.01) and higher plasma progesterone concentration (4.0+/-0.3 vs. 3.1+/-0.2, P<0.01) than that of the high-progesterone group. However, pregnancy rates did not differ (79 of 148, 53.4%, and 70 of 131, 53.4%) for low- and high-progesterone groups, respectively). In summary, low circulating progesterone concentrations during the growing phase of the ovulatory follicle resulted in a larger dominant follicle and a larger CL that produced more progesterone, with no significant effect on pregnancy rate.

Abstract:Reproductive performance is crucial for profitability of dual-purpose cow-calf production in the Ecuadorian Amazon. To evaluate three different fixed-time artificial insemination (FTAI) protocols in suckled, dual-purpose cows in the Ecuadorian Amazon. Lactating, Brown Swiss cows (n=301) received 2 mg estradiol benzoate (EB) and an intravaginal device containing 0.5 g of progesterone (P4) on Day 0. They were allocated randomly into the following three treatment groups: Cows in the EB group received 500 μg cloprostenol (PGF2α), and P4 devices were removed on Day 7, and 1 mg EB was administered on Day 8. Cows in the ECP group were treated as those in the EB group, except that they received 0.5 mg estradiol cypionate (ECP) at the time of P4 device removal on Day 7 instead of EB on Day 8. Cows in the J-Synch group had the P4 device removed and PGF2α administered on Day 6. All cows were FTAI on Day 9; cows in the J-Synch group also received 100 μg gonadorelin at that time. Although the diameter of the dominant follicle and the resulting CL were greater (P 0.2) among groups (EB: 51.0%, ECP: 53.0% and J-Synch: 59.4%). The three protocols tested were applied successfully in suckled, dual-purpose cows with no differences in pregnancies per AI. Highlights: Estradiol and progesterone based FTAI protocols can be successfully applied in dual purpose cows in the tropics, with P/AI ≥50%. Proestrus prolongation increased dominant follicle diameter at the time of FTAI and also CL diameter 7 days pos treatment Although differences were not significant, the protocol with the prolonged proestrus (J-Synch) resulted in higher P/AI than the two protocols with short proestrus. Regardless of the protocol used, there was a positive association between estrus manifestation, follicle diameter at the time of FTAI and CL diameter 7 days post treatment.

Use of equine chorionic gonadotropin in lactating dairy cattle: a rapid review

Abstract:Aims: To use an evidence-based approach to evaluate the available research data on the efficacy of equine chorionic gonadotropin (eCG) to improve reproductive performance of dairy cows when added to oestrus synchronisation programmes for the treatment of cows not detected in oestrus. Methods: A rapid literature review was conducted to summarise the existing trial data, primarily the effect of eCG on conception rate (CR) to fixed time artificial insemination (FTAI) from oestrus synchronisation programmes in lactating dairy cows. Relevant papers were identified via databases and auxiliary search strategies, then information was collected using a standardised data collection form. Similar studies were grouped together to create a descriptive summary of CR to FTAI and secondary outcome measures, with the creation of forest plots to visualise results. Results: A total of 15 studies were identified for review: nine examined the administration of 400-500 IU eCG as part of an oestrus synchronisation programme based on intra-vaginal progesterone-releasing devices, gonadotropin-releasing hormone and prostaglandin (P4-GPG programme). The variable methodologies in the trials evaluated posed difficulties in grouping studies for further analysis and meant that there was insufficient data for a full meta-analysis. A single study reported a statistically significant increase in CR to FTAI from the addition to eCG to a P4-GPG programme in cows with anovulatory anoestrous. Four studies evaluated the addition of eCG to P4-GPG programmes in cows without visible oestrous signs; risk differences for CR to FTAI were between -1.7% and 28.8%, with three out of four studies reporting a positive effect of eCG on CR to FTAI, but all 95% CI values crossed the line of no effect. Two studies reported on the effect of eCG on 42-day pregnancy rate following a P4-GPG programme; one reported a risk difference of -4.9% (95% CI = -10.6-0.8%), while the other reported a risk difference of 7.9% (95% CI = 0.8-15%). Conclusions: The results of published studies describing the effect of eCG in P4-GPG programmes on CR to FTAI are inconsistent and further well-designed, adequately powered studies with standardised outcome measures are required to investigate its effect.
Improving Reproduction in Postpartum Dairy Cows : Potential Application of Polyunsaturated Fatty Acids ( PUFA ) and Somatotropin

Abstract:The implementation of methods designed to improve the reproductive performance of postpartum dairy cows has become a priority for many dairy farms. This review will examine two relatively new approaches for improving the reproduction of dairy cows. The first method involves the feeding of polyunsaturated fatty acids (PUFA) to dairy cows. Feeding PUFA decreases uterine secretion of prostaglandin F2á (PGF2á) and may increase the likelihood of pregnancy in cows with poor quality conceptuses. The second method involves the strategic use of recombinant bovine somatotropin (rbST) to improve first service conception rates in dairy cows that are time inseminated. The rbST may increase concentrations of growth factors in the uterus and improve the uterine environment for embryonic growth and the establishment of pregnancy. Both methods have the potential to improve reproduction when practiced in dairy herds. Improving Reproduction with Polyunsaturated Fatty Acids (PUFA) Linoleic and linolenic acid (polyunsaturated fatty acids; PUFA) are essential fatty acids. Dairy cows consume linoleic and linolenic acid in the plant matter that they eat. Linoleic acid (an omega-6 fatty acid) can be converted to arachidonic acid. Linolenic acid (an omega-3 fatty acid) can be converted to eicosapentaenoic acid (EPA) or docosahexaenoic acid (DHA). Most of the linoleic and linolenic acid in the diet is converted to saturated fatty acids in the rumen through a process called biohydrogenation. The saturated fatty acids are not essential fatty acids but can be used for energy or be directly incorporated into milk fat. A small percentage of PUFA pass through the rumen in their native form. Polyunsaturated fatty acids are important nutrients because they can be converted into eicosanoids (bioactive 20 carbon fatty acids). Eicosanoids are hormones that carry out a variety of cellular processes. One type of eicosanoid, prostaglandin F2á (PGF2á), is very familiar to dairy farmers because it causes regression of the corpus luteum in cattle. During a natural estrous cycle, the uterus secretes PGF2á around day 17 to 19 of the estrous cycle. The secretion of uterine PGF2á causes the regression of the corpus luteum and brings the cow back into estrus. The embryo can block PGF2á secretion by the uterus (a process known as maternal recognition of pregnancy) so that pregnant cows do not undergo luteal regression. Pharmaceutical products that cause luteal regression for estrous synchronization (Lutalyse, Estrumate, etc.) are PGF2á or PGF2á analogues and cause luteal regression in the same manner as the endogenous uterine hormone. The content of PUFA in a cow’s body is normally low because PUFA are biohydrogenated to saturated fatty acids in the rumen. The PUFA concentrations of a cow can be increased, however, if PUFA are fed in high concentrations or are fed in a protected form that cannot be degraded by rumen microbes. Increasing the content of PUFA in the cow influences the ability of the uterus to synthesize PGF2á. Surprisingly; the uterus produces less PGF2á when PUFA are fed to dairy cows (Mattos et al., 2002) or PUFA are added to uterine cell cultures (Cheng et al., 2001). This is somewhat opposite to expectations because PUFA are precursors to the prostaglandins. Linoleic acid is the precursor for arachidonic acid which is an immediate precursor of PGF2á. The linoleic acid, however, acts as an inhibitor to the synthesis of PGF2á from arachidonic acid.

Effects of oestrus induction with progestagens or prostaglandin analogues on ovarian and pituitary function in sheep

Abstract:The aim of the current study was to determine possible differences in ovarian and pituitary features explaining lower fertility rates in sheep with oestrus induced with intravaginal progestagens or prostaglandin analogues (group FGA and PGF, n=8 in both) when compared to a control group (group C, n=8). The growth profiles and the mean individual sizes of preovulatory follicles were similar between groups; however, the number of preovulatory follicles per ewe and, consequently, the number of ovulations were higher in groups FGA and PGF (2.3±0.3 and 2.0±0.1, respectively) than in group C (1.4±0.1, P<0.05). However, plasma oestradiol concentrations were similar between groups suggesting a defective function in some preovulatory follicles of groups FGA and PGF. In group FGA, the basal LH levels during the follicular phase were lower (0.21±0.0 ng/mL, P<0.005) than in groups C (0.41±0.1 ng/mL) and PGF (0.55±0.1 ng/mL); the onset of preovulatory discharge being later (21.0±2.3h vs. 12.8±1.5 in C and 14.5±1.5 in PGF; P<0.05 for both). Finally, luteal activity was also found to be affected in group FGA; the rate of progesterone secretion per total luteal tissue was lower (range: 0.46-0.65 ng/mL/cm(2)) than in ewes treated with cloprostenol (2.1-3.3 ng/mL/cm(2)) and control sheep (2.0-3.4 ng/mL/cm(2)).

Abstract:Summary Recently, certain batches of heparin have been associated with an acute, rapid onset of serious side-effects indicative of allergictype reactions.These reports generated significant concern regarding the possible presence of a dangerous contaminant within heparin and highlighted the need to re-assess the purity criteria of heparin preparations for clinical use. Given the nature of the array of all possible contaminants, traditional screening tests cannot safely differentiate between contaminated and uncontaminated heparin preparations. Monoand bi-dimensional NMR spectroscopy are powerful techniques that are able to detect and quantify a wide variety of potential sulfated polysaccharide contaminants. As such, these techniques are powerful tools for the analysis and assessment of heparin preparations.
Effect of peripartum dietary energy supplementation of dairy cows on metabolites, liver function and reproductive variables

Abstract:Multiparous Holstein cows (n=58) were used to study the effects of peripartum dietary supplementation on metabolic status, liver function and reproduction variables. Diets for cows were as follows: (a) no supplementation (CTL), (b) prilled fatty acids as 1.9% of DM (PrFA), (c) calcium salts of long chain n-6 fatty acids as 2.24% of DM (CaLFA) or (d) daily topdressing with 769 g of 65% propylene glycol (PGLY). Supplements were fed during the last 21 days before expected calving except for PGLY that continued until 21 days after parturition. Ovarian activity was monitored by transrectal ultrasonography and days to first ovulation were recorded. Liver biopsies were obtained on day 8 and 21 postpartum and analyzed for triglyceride content and mRNA expression of pyruvate carboxylase, cytosolic phosphoenolpyruvate carboxykinase, carnitine palmytoyltransferase 1A, and peroxisome proliferator-activated receptor-alpha. At 71 days following parturition, stage of ovarian cycles was synchronized and at day15 of the cycle oxytocin was injected i.v., blood samples were obtained at frequent intervals, and analyzed for 13,14 dihydro, 15-keto PGF(2alpha) (PGFM). Milk production and milk components were not different among treatment groups. Cows in PGLY gained body condition score (BCS) prepartum and net energy balance prepartum tended to be greater, but was not different postpartum from other groups. PGLY supplementation increased plasma insulin concentration prepartum, but not during the postpartum period. No significant differences were observed in plasma concentrations of glucose, NEFA, and insulin-like growth factor or hepatic triglyceride content, but all supplements tended to decrease beta hydroxybutyrate postpartum compared to CTL cows. Abundance of mRNA of gluconeogenic and lipid oxidation genes was not different among treatment groups. Days to first ovulation and uterine PGF(2alpha) production in response to an oxytocin treatment were not significantly different among treatment groups. Peripartum supplementation did not result in the substantial improvement of metabolic profile in early lactation nor significantly affect days to first ovulation and PGFM response to an oxytocin treatment.

Abstract:Studies of the non-enzymatic decarboxylation of /3-keto acids have shown that it is the keto form of the 8-keto acid which loses CO2 to form an enol of the product. This was demonstrated for dimethylacetoacetic acid by Pedersen (1) and for dimethyloxalacetic acid by Steinberger and Westheimer (2). These latter authors studied the catalytic effect of metal ions on the decarboxylation reaction and suggested that the non-enzymatic reaction be taken as a model for similar enzymatic decarboxylations. Kornberg et al. (3) observed the formation of an enol band during the enzymatic decarboxylation of oxalacetic acid and of another P-keto acid, oxalsuccinic acid. They regarded their results as evidence that an enol form of the P-keto acid was decarboxylated, although their data do not appear to preclude the possibility that an enol appears as a product of the decarboxylation (4). Recent studies in this laboratory have shown that wheat germ extracts catalyze an irreversible carboxylation reaction which is essentially the reverse of a decarboxylation. This reaction consists of the addition of CO2 to phosphoenolpyruvate to form oxalacetate and inorganic phosphate (5).’ If the mechanism for the non-enzymatic decarboxylation should apply in reverse to this reaction, then the enol, PEP, should add CO2 to give the keto form of oxalacetate. Experiments with deuterium described in this paper have provided unequivocal evidence that the oxalacetate formed enzymatically in the carboxylation reaction is in fact the keto form and not an enol. These results have been described in a preliminary publication (6).
The effect of cultural and environmental factors on potato seed tuber morphology and subsequent sprout and stem development

Abstract:Seed crops of the variety Estima were grown in each of 2 years using two planting dates, two harvest dates, two plant densities and two irrigation regimes to produce seed tubers which had experienced different cultural and environmental conditions. The effects of these treatments on tuber characteristics, sprout production and stem development in the ware crop were then determined in subsequent experiments using storage regimes of 3 and 10 °C. Time of planting the seed crop affected numbers of eyes, sprouts and above ground stems in the subsequent ware crop because environmental conditions around the time of tuber initiation appeared to alter tuber shape. Cooler, wetter conditions in the 7 days after tuber initiation were associated with tubers which were longer, heavier and had more eyes, sprouts and above ground stems. In contrast, the time of harvesting the seed crop did not affect tuber shape or numbers of above ground stems and there was no interaction with tuber size. The density of the seed crop had no effect on any character measured and irrigation well after tuber initiation did not affect tuber shape, numbers of sprouts or numbers of stems. Seed production treatments, which resulted in earlier dormancy break, were associated with tubers that produced more sprouts and above ground stems, in contrast to the conventional understanding of apical dominance. Storage at 3 °C gave fewer sprouts, a lower proportion of eyes with sprouts and fewer stems than storage at 10 °C. The major effects on stem production appear to result from environmental conditions at the time of tuber initiation of the seed crop and sprouting temperature.

Although most college students are healthy, they may be vulnerable to infectious diseases, which can cause substantial morbidity and mortality. The purpose of this article is to educate health care providers about proper vaccination for young adults preparing for college, focusing on individual diseases.
Influence of Ovarian Status and Steroid Hormone Concentration on Day of Timed Artificial Insemination (TAI) on the Reproductive Performance of Dairy Cows Inseminated with Sexed Semen

Abstract:This study aimed to evaluate the effect of the ovarian status and steroid hormone concentration on the day of TAI on the reproductive performance of dairy cows subjected to estrus synchronization treatment and timed artificial insemination with sexed semen. Seventy-eight cyclic Holstein cows pre-treated with PGF2α-GnRH were divided in two groups—I (Preselect-OvSynch, n = 38) and II (OvSynch+PRID-7-day+eCG, n = 40)—and inseminated with sexed semen. The presence of preovulatory follicle (PF) with or without corpus luteum (CL), the PF diameter, the estradiol (E2) and progesterone (P4) concentrations on the day of TAI, the pregnancy rate (PR) and embryo loss were determined. On the day of TAI, 78.4% of all the pregnant cows presented a PF (mean size 1.80 ± 0.12 cm) without CL, low P4 (0.59 ± 0.28 ng/mL) and high E2 (12.35 ± 2.62 pg/mg) concentrations. The positive correlation between the size of the PF and the level of E2 in the pregnant cows from group II was stronger than that of group I (R = 0.82 vs. R = 0.52, p < 0.05). The pregnancy rate on day 30 (57.5% vs. 36.8%) and day 60 (50% vs. 26.3%; p < 0.05) and the embryo losses (13% vs. 28.5%) showed better effects of treatment in group II. In conclusion, the ovarian status and the steroid hormone concentration on the day of TAI influence the pregnancy rates of dairy cows subjected to estrus synchronization and timed artificial insemination with sexed semen.
