Abstract:The oxidation of N-heterocycles is key to their functionalization since it enhances their reactivity in the ortho position. How-ever, an efficient post-reduction is often required to access the targeted compound. Here, a rhenium-based photocatalyst enables the versatile and fast deoxygenation of N-heterocyclic and alkyl N-oxides, avoiding sacrificial oxophilic reagents. The strong influence of the reaction conditions (solvent, H2O as additive, irradiation set-up) is demonstrated. The system shows a high functional group tolerance towards other reducible functions (free alcohol, carboxylic acid, ester, nitrile, halo-gen,…) and light-sensitive N-heterocycles.

Abstract:A meta-dehydrogenative alkylation of arenes with cyclic ethers, ketones, and esters catalyzed by ruthenium is achieved in the presence of a di-tert-butyl peroxide (DTBP) oxidant. Interestingly, when quinoline and isoquinoline are employed as the directing group, or a chain ether as alkylation reagent, the system produces Minisci reaction products. Mechanistic study indicates that meta-dehydrogenative alkylation is a radical process initiated by DTBP with the assistance of a C-Ar-Ru bond ortho/para-directing effect.

Abstract:Oxidative transformations utilizing molecular oxygen (O2 ) as the stoichiometric oxidant are of paramount importance in organic synthesis from ecological and economical perspectives. Alcohol oxidation reactions that employ O2 are scarce in homogeneous catalysis and the efficacy of such systems has been constrained by limited substrate scope (most involve secondary alcohol oxidation) or practical factors, such as the need for an excess of base or an additive. Catalytic systems employing O2 as the "primary" oxidant, in the absence of any additive, are rare. A solution to this longstanding issue is offered by the development of an efficient ruthenium-catalyzed oxidation protocol, which enables smooth oxidation of a wide variety of primary, as well as secondary benzylic, allylic, heterocyclic, and aliphatic, alcohols with molecular oxygen as the primary oxidant and without any base or hydrogen- or electron-transfer agents. Most importantly, a high degree of selectivity during alcohol oxidation has been predicted for complex settings. Preliminary mechanistic studies including (18) O labeling established the in situ formation of an oxo-ruthenium intermediate as the active catalytic species in the cycle and involvement of a two-electron hydride transfer in the rate-limiting step.

Abstract:Deoxygenative difunctionalization of carbonyls represents a convenient route to construct complex molecules considering the readily available aldehyde and ketone compounds. The present approaches typically rely on strategies via carbene, carbanion, or carbocation equivalents. Herein, combined with dual nickel/photoredox catalysis regime, we developed a strategy through a radical intermediate to achieve the deoxygenative arylboration of aldehydes. Compared with the known patterns of carbon–oxygen (C–O) bond transformations, this coupling of C-OBPin unit opens a direction for this chemistry. A wide variety of substrates bearing a diverse set of functional groups were compatible with this method under very mild conditions (visible light, ambient temperature, no strong base) to afford the benzylic boronic esters, which have important versatilities in organic synthesis.

Abstract:A versatile, selective, solvent (methanol vs ethanol)- and base (potassium vs lithium carbonate)-assisted switchable synthesis of saturated ketone and α-methyl saturated ketone from α,β-unsaturated ketone is developed. Mechanistic aspects, evaluated from spectroscopic studies, in situ monitoring of the reaction progress, control studies, and labeling studies, further indicate the involvement of a tandem dehydrogenation-condensation-hydrogenation sequence in the reaction, in which the interconvertible coordination mode (imino N → Ru and amido N-Ru) of coordinated imidazole with Ru(II)-para-cymene is crucial, without which the efficiency and selectivity of the catalyst are completely lost. The catalyst demonstrates good efficiency, selectivity, and functional group tolerance and displays a broad scope (69 examples) for monomethylation and hydrogenation of unsaturated chalcones, double methylation of ketones, and N-methylation of amines.

Abstract:Alkene carbonylations represent a major technology for the production of value-added bulk and fine chemicals. Nowadays, all industrial carbonylation processes make use of highly toxic and flammable carbon monoxide. Here we show the application of abundantly available carbon dioxide as C1 building block for the alkoxycarbonylations of industrially important olefins in the presence of a convenient and inexpensive ruthenium catalyst system. In our system, carbon dioxide works much better than the traditional combination of carbon monoxide and alcohols. The unprecedented in situ formation of carbon monoxide from carbon dioxide and alcohols permits an efficient synthesis of carboxylic acid esters, which can be used as detergents and polymer-building blocks. Notably, this transformation allows the catalytic formation of C–C bonds with carbon dioxide as C1 source and avoids the use of sensitive and/or expensive reducing agents (for example, Grignard reagents, diethylzinc or triethylaluminum).

Abstract:A novel protocol for ruthenium-catalyzed methoxycarbonylation reaction of alkenes is reported. Using easily available Ru3(CO)12 as catalyst in the presence of [Bmim]Cl as additive, industrial important esters are produced in yields up to 82%. Compared to traditional olefin carbonylation processes key to success is running the reaction at low carbon monoxide concentration. Notably, the ionic liquid phase which contains the catalyst can be reused for several runs.

Abstract:The green reduction of carboxylic acids to alcohols is a challenging task in organic chemistry. Herein, we describe a general protocol for generation of alcohols by catalytic hydrogenation of carboxylic acids. Key to success is the use of a combination of Ru(acac)(3), triphos and Lewis acids. The novel method showed broad substrate tolerance and a variety of aliphatic carboxylic acids including biomass-derived compounds can be smoothly reduced.

Abstract:Ruthenium-catalyzed oxidation of multisubstituted allyl alcohols in the presence of benzaldehyde gives enals or enones in good yields. Unlike the commonly reported ruthenium-catalyzed isomerization reaction of allyl alcohols to give saturated ketones, an intermolecular rather than intramolecular hydrogen transfer is involved in this transformation. This reaction offers an efficient, mild, and high-yielding method for the preparation of substituted α,β-unsaturated compounds.

Abstract:The rhodium-catalyzed deoxygenation and borylation of ketones with B2pin2 have been developed, leading to efficient formation of alkenes, vinylboronates, and vinyldiboronates. These reactions feature mild reaction conditions, a broad substrate scope, and excellent functional-group compatibility. Abstract:The synthesis of quinolines from alcohols and 2‐nitrobenzaldehydes is described. Unreactive alcohols and 2‐nitrobenzaldehydes were concurrently converted to the reactive reactants for a Friedländer annulation via a hydrogen transfer. Tris(triphenylphosphine)ruthenium(II) dichloride [Ru(PPh3)3Cl2] appeared to serve the dual role of an efficient hydrogen transfer catalyst and a suitable Friedländer annulation catalyst.

Abstract:An iridium-catalyzed, highly efficient, and site-selective deoxygenation of primary, secondary, and tertiary alcohols has been realized, under the assistance of a 4-(N-substituted amino)aryl directing group. Only the hydroxyl adjacent to the directing group can be deoxygenated. The deoxygenation is performed in water, with formic acid as both the promoter and hydride donor. Excellent yields and functionality tolerance, as well as high efficiency (S/C up to 1 000 000, TOF up to 445 000 h–1), are obtained. The kinetic isotope effect studies show that hydride formation is the rate-determining step, and the deoxygenation follows an SN1-type pathway. The deoxygenation protocol has been demonstrated useful in the structural modification of naturally occurring ketones and steroids.

Abstract:A previously elusive Ru(II) -catalyzed N-N bond-based traceless C-H functionalization strategy is reported. An N-amino (i.e., hydrazine) group is used for the directed C-H functionalization with either an alkyne or an alkene, affording an indole derivative or olefination product. The synthesis features a broad substrate scope, superior atom and step economy, as well as mild reaction conditions.

Abstract:C-O bond cleavage is often a key process in defunctionalization of organic compounds as well as in degradation of natural polymers. However, it seldom occurs regioselectively for different types of C-O bonds under metal-free mild conditions. Here we report a facile chemo-selective cleavage of the α-C-O bonds in α-carboxy ketones by commercially available pinacolborane under the catalysis of diazaphosphinane based on a mechanism switch strategy. This new reaction features high efficiency, low cost and good group-tolerance, and is also amenable to catalytic deprotection of desyl-protected carboxylic acids and amino acids. Mechanistic studies indicated an electron-transfer-initiated radical process, underlining two crucial steps: (1) the initiator azodiisobutyronitrile switches originally hydridic reduction to kinetically more accessible electron reduction; and (2) the catalytic phosphorus species upconverts weakly reducing pinacolborane into strongly reducing diazaphosphinane.

Abstract:Historically, deuteration reaction has been primarily utilized for mechanistic studies, but recently its pharmacological importance has been recognized. Many efforts have been made to fulfill the urgent need of deuteration tools since FDA approved the first deuterated drug, used to treat chorea-associated Huntington`s disease. Current deuteration strategies prevailingly involve reversible H/D exchange at the target C-H bond, which suffer from low functional group tolerance and deuterium content. Herein, a ruthenium-catalyzed deoxygenative deuteration of ketones is developed with 20 examples under mild conditions. The reaction proceeds via HOME chemistry (hydrazone as organometallic equivalent) followed by in situ deuteration, therefore enabling regiospecific deuteration at the methylene site with 91% to 96% deuterium content.
