Abstract:Nucleic acid‐based molecules are radiolabelled with beta‐plus, gamma or beta‐minus emitters for tracking and monitoring. The rise of nucleic acid‐based therapeutics continues apace. At the same time, the need for radiolabelled oligonucleotides for determination of spatial distribution is increasing. Complex molecular structures with mostly multiple charges and low solubility in organic solvents increase the challenge of integrating radionuclides. In preclinical research, it is important to understand the fate of new drug candidates in biodistribution studies, target binding or biotransformation studies. Depending on a specific question, the selection of a respective radiolabelling strategy is crucial. Radiometals for molecular imaging with positron emission tomography or single‐photon computed tomography generally require an attached chelating agent for stable complexation of the metal with the oligonucleotide, whereas labelling using carbon‐11/‐14 or tritium allows incorporation of the radioisotope into the native structure without altering it. Moreover, the suitability of direct radiolabelling of the oligonucleotide of interest or indirect radiolabelling, for example, by a two‐step pretargeting approach, for the study design requires consideration. This review focuses on the challenges of radiolabelling nucleic acid‐based molecules with beta‐plus, gamma and beta‐minus emitters and their use for tracking and monitoring.

Radiosynthesis of Tritium-Labeled Novel Nitromethylene Neonicotinoids Compounds with NaB3H4

Abstract:Paichongding and Cycloxaprid are two novel neonicotinoids with good industrialization prospects for their high activity against imidacloprid-resistant pest. In this paper, the radiosynthesis of [H-3(2)]-Paichongding and [H-3(2)]-Cycloxaprid was achieved using a (NaBH4)-H-3 reduction. The title compounds were obtained with chemical purities of 98.7 and 98.4%, and radiochemical purities of 97.3 and 98.6%, respectively. The labeled compounds could be used as radiotracers for further study of metabolism and toxicology.
Monitoring RNA base structure and dynamics using site-directed spin labeling.

Abstract:Site-directed spin labeling utilizes site-specific attachment of a stable nitroxide radical to probe the structure and dynamics of macromolecules. In the present study, a 4-thiouridine base is introduced at each of six different positions in a 23-nucleotide RNA molecule. The 4-thiouridine derivatives were subsequently modified with one of three methanethiosulfonate nitroxide reagents to introduce a spin label at specific sites. The electron paramagnetic resonance spectra of the labeled RNAs were analyzed in terms of nitroxide motion and the RNA solution structure. At a base-paired site in the RNA helix, where the nitroxide has weak or no local interactions, motion of the nitroxide is apparently dominated by rotation about bonds within the probe. The motion is similar to that found for a structurally related probe on helical sites in proteins, suggesting a similar mode of motion. At other sites that are hydrogen bonded and stacked within the helix, local interactions within the RNA molecule modulate the nitroxide motion in a manner consistent with expectations based on the known structure. For a base that is not structurally constrained, the mobility is higher than at any other site, presumably due to motion of the base itself. These results demonstrate the general utility of the 4-thiouridine/methanethiosulfonate coupling method to introduce nitroxide spin labels into RNA and the ability of the resulting label to probe local structure and dynamics.
Microfluidics for synthesis of peptide-based PET tracers.

Abstract:Positron emission tomography (PET) is a powerful noninvasive tool for acquisition of the physiological parameters in human and animals with the help of PET tracers. Among all the PET tracers, radiolabeled peptides have been widely explored for cancer-related receptor imaging due to their high affinity and specificity to receptors. But radiochemistry procedures for production of peptide-based PET tracers are usually complex, which makes large-scale clinical studies relatively challenging. New radiolabeling technologies which could simplify synthesis and purification procedures, are extremely needed. Over the last decade, microfluidics and lab-on-a-chip (LOC) technology have boomed as powerful tools in the field of organic chemistry, which potentially provide significant help to the PET chemistry. In this minireview, microfluidic radiolabeling technology is described and its application for synthesis of peptide-based PET tracers is summarized and discussed.

Abstract:The concept of biological receptors is now more than a century old with numerous receptor families being identified as they respond to the influence of a collection of agonist and antagonist substances. Many receptors have been fully studied and characterized. However, the imidazoline receptor family is one of the more intriguing groups for which unanswered biology questions still remain. To better understand the imidazoline receptors, this work describes the synthesis and characterization of several useful tritium labelled imidazoline radioligands.

Abstract:Natural compounds are important precursors for the synthesis of new drugs. The development of novel molecules that are useful for various diseases is the main goal of researchers, especially for the diagnosis and treatment of many diseases. Some pathologies need to be treated with radiopharmaceuticals, and, for this reason, radiopharmaceuticals that use the radiolabeling of natural derivates molecules are arousing more and more interest. Radiopharmaceuticals can be used for both diagnostic and therapeutic purposes depending on the radionuclide. β+- and gamma-emitting radionuclides are used for diagnostic use for PET or SPECT imaging techniques, while α- and β--emitting radionuclides are used for in metabolic radiotherapy. Based on these assumptions, the purpose of this review is to highlight the studies carried out in the last ten years, to search for potentially useful radiopharmaceuticals for nuclear medicine that use molecules of natural origin as lead structures. In this context, the main radiolabeled compounds containing natural products as scaffolds are analyzed, in particular curcumin, stilbene, chalcone, and benzofuran. Studies on structural and chemical modifications are emphasized in order to obtain a collection of potential radiopharmaceuticals that exploit the biological properties of molecules of natural origin. The radionuclides used to label these compounds are 68Ga, 44Sc, 18F, 64Cu, 99mTc, and 125I for diagnostic imaging.
Convergent F-18 radiosynthesis: A new dimension for radiolabelling

Abstract:The availability of radiolabelled probes is important for in vivo studies by positron emission tomography (PET). Among the frontier challenges in F-18-radiochemistry are the interconnected goals of increasing synthetic efficiency and diversity in the construction of F-18-labelled radiotracers. F-18-Radioretrosynthetic strategies implemented to date are typically linear sequences of transformations designed with the aim of introducing the F-18-label ideally in the last step or at least as late as possible. Here we report that convergent F-18-radiochemistry allows for the rapid assembly of functionalised F-18-radiotracers from readily accessible F-18-labelled prosthetic groups. Using multicomponent reactions for proof of concept, Ugi-4CR, Passerini-3CR, Biginelli-3CR and Groebke-3CR were performed successfully using F-18-benzaldehydes and these highly convergent reactions delivered, in high radiochemical yield (RCY), structurally complex F-18-radiotracers with the label positioned on an aryl motif not responsive to direct nucleophilic fluorination. These data establish an unprecedented connection between radiochemistry for PET and the field of multicomponent chemistry and demonstrate that convergent retroradiosynthesis is a powerful strategy expanding dramatically the scope of F-18-prosthetic group radiochemistry. The preparation of F-18-labelled prosthetic groups from [F-18]fluoride ion is commonly performed in many labelling laboratories, so the concept of convergent F-18-radiosynthesis can easily be applied immediately.

Abstract:Background: 211At-radiopharmaceuticals are currently the subject of growing studies for targeted alpha therapy of cancers, which leads to the widening of the scope of the targeting vectors, from small molecules to peptides and proteins. This has prompted, during the past decade, to a renewed interest in developing novel 211At-labelling approaches and novel prosthetic groups to address the diverse scenarios and to reach improved efficiency and robustness of procedures as well as an appropriate in vivo stability of the label. Main body: Translated from the well-known (radio)iodine chemistry, the long preferred electrophilic astatodemetallation using trialkylaryltin precursors is now complemented by new approaches using electrophilic or nucleophilic At. Alternatives to the astatoaryl moiety have been proposed to improve labelling stability, and the range of prosthetic groups available to label proteins has expanded. Conclusion: In this report, we cover the evolution of radiolabelling chemistry, from the initial strategies developed in the late 1970's to the most recent findings.

Abstract:Pulsed electron-electron double resonance (PELDOR) spectroscopy, X-ray scattering interferometry (XSI), and single-molecule Förster resonance energy transfer (smFRET) are molecular rulers that provide inter- or intramolecular pair-wise distance distributions in the nanometer range, thus being ideally suitable for structural and dynamic studies of biomolecules including RNAs. The prerequisite for such applications requires site-specific labeling of biomolecules with spin labels, gold nanoparticles, and fluorescent tags, respectively. Recently, site-specific labeling of large RNAs has been achieved by a combination of transcription of an expanded genetic alphabet containing A-T/G-C base pairs and NaM-TPT3 unnatural base pair (UBP) with post-transcriptional modifications at UBP bases by click chemistry or amine-NHS ester reactions. However, due to the bulky sizes of functional groups or labeling probes used, such strategies might cause structural perturbation and decrease the accuracy of distance measurements. Here, we synthesize an α-thiophosphorylated variant of rTPT3TP (rTPT3αS), which allows for post-transcriptional site-specific labeling of large RNAs at the internal α-phosphate backbone via maleimide-modified probes. Subsequent PELDOR, XSI, and smFRET measurements result in narrower distance distributions than labeling at the TPT3 base. The presented strategy provides a new route to empower the molecular rulers for structural and dynamic studies of large RNA and its complex.

Abstract:With low background radiation, tritiate compounds exclusively emit intense beta particles without structural changes. This makes them a useful tool in the drug discovery arsenal. Thanks to the recent rapid progress in tritium chemistry, the preparation and analysis of tritium-labeled compounds are now much easier, simpler, and cheaper. Pharmacokinetics, autoradiography, and protein binding studies have been much more efficient with the employment of tritium-labeled compounds. This review provides a comprehensive overview of tritium-labeled compounds regarding their properties, synthesis strategies, and applications.

Abstract:Introduction Antibiotic resistance is a burden on the healthcare system. In present study, we have labeled an antibiotic named Colistimethate sodium (CMS) with technetium-99m ( 99m Tc) to develop a SPECT based imaging tracer. Methods We standardised the labeling using 0.5–2 mg of CMS (in water) using stannous chloride dihydrate as a reducing agent followed by addition of 370 ± 74 MBq of 99m Tc. A group of mice were injected intravenously (in tail vein) with 4–6 MBq of [ 99m Tc]Tc-CMS diluted with saline and euthanized at various time intervals. microSPECT Imaging (Υ-eye) was acquired to study the biodistribution in the healthy mice. Results We standardised the labeling using 0.5 mg of colistin in 0.5 ml of saline with addition of 30 μg stannous chloride dihydrate. The retention factor value was 0.1–0.3 as compared to 0.9–1.0 for free 99m Tc by TLC and retention time was found to be 14.2 ± 1.3 min as evaluated by HPLC. The biodistribution data showed uptake in lungs, spleen, and liver at 30 min but the uptake decreased in lung at 60 min. The imaging data corroborated with the biodistribution data. Conclusions We could successfully label [ 99m Tc]Tc-CMS 99m Tc and we could study its biodistribution in healthy mice.

Abstract:A wide range of nano-objects is found in many applications of our everyday life. Recognition of their peculiar properties and ease of functionalization has prompted their engineering into multifunctional platforms that are supposed to afford efficient tools for the development of biomedical applications. However, bridging the gap between bench to bedside cannot be expected without a good knowledge of their behaviour in vivo, which can be obtained through non-invasive imaging techniques, such as positron emission tomography (PET). Their radiolabelling with [18F]-fluorine, a technique already well established and widely used routinely for PET imaging, with [18F]-FDG for example, and in preclinical investigation using [18F]-radiolabelled biological macromolecules, has, therefore, been developed. In this context, this review highlights the various nano-objects studied so far, the reasons behind their radiolabelling, and main in vitro and/or in vivo results obtained thereof. Then, the methods developed to introduce the radioelement are presented. Detailed indications on the chemical steps involved are provided, and the stability of the radiolabelling is discussed. Emphasis is then made on the techniques used to purify and analyse the radiolabelled nano-objects, a point that is rarely discussed despite its technical relevance and importance for accurate imaging. The pros and cons of the different methods developed are finally discussed from which future work can develop.

Abstract:The need for carbon-labeled radiotracers is increasingly higher in drug discovery and development (carbon-14, β-, t1/2 = 5730 years) as well as in positron emission tomography (PET) for in vivo molecular imaging applications (carbon-11, β+, t1/2 = 20.4 min). However, the structural diversity of radiotracers is still systematically driven by the narrow available labeled sources and methodologies. In this context, the emergence of carbon dioxide radical anion chemistry might set forth potential unexplored opportunities. Based on a dynamic isotopic equilibration between formate salts and [13C, 14C, 11C]CO2, C-labeled radical anion CO2•- could be accessed under extremely mild conditions within seconds. This methodology was successfully applied to hydrocarboxylation and dicarboxylation reactions in late-stage carbon isotope labeling of pharmaceutically relevant compounds. The relevance of the method in applied radiochemistry was showcased by the whole-body PET biodistribution profile of [11C]oxaprozin in mice.
A Facile Method for Attaching Nitroxide Spin Labels at the 5 ' Terminus of Nucleic Acids

Abstract:In site-directed spin labeling (SDSL), a nitroxide moiety containing a stable, unpaired electron is covalently attached to a specific site within a macromolecule, and structural and dynamic information at the labeling site is obtained via electron paramagnetic resonance (EPR) spectroscopy. Successful SDSL requires efficient site-specific incorporation of nitroxides. Work reported here presents a new method for facile nitroxide labeling at the 5' terminus of nucleic acids of arbitrary sizes. T4-polynucleotide kinase was used to enzymatically substitute a phosphorothioate group at the 5' terminus of a nucleic acid, and the resulting phosphorothioate was then reacted with an iodomethyl derivative of a nitroxide. The method was successfully demonstrated on both chemically synthesized and naturally occurring nucleic acids. The attached nitroxides reported duplex formation as well as tertiary folding of nucleic acids, indicating that they serve as a valid probe in nucleic acid studies.

Abstract:Technetium‐99m (99mTc) remains the cornerstone of nuclear medicine for single photon emission computed tomography (SPECT) due to its widespread availability and chemical and physical features. Its multiple oxidation states allow for the design and production of radiopharmaceuticals with versatile properties, namely in terms of pharmacokinetic profile. 99mTc(V) is the most common oxidation state, but 99mTc(I) gained traction after the pioneering work of Alberto and colleagues, which resulted in the introduction of the organometallic core fac‐[99mTc(CO)3(H2O)3]+. This core is readily available from [99mTcO4]‐ and displays three labile water molecules that can be easily swapped for ligands with different denticity and/or donor atoms in aqueous environment. This makes it possible to radiolabel small molecules as well as high molecular weight molecules, such as antibodies or other proteins, while assuring biological activity. Direct radiolabelling of those proteins with fac‐[99mTc(CO)3]+ under mild conditions is accomplished through incorporation of a polyhistidine tag (His‐tag), a commonly used tag for purification of recombinant proteins. This review aims to address the direct radiolabelling of His‐tagged macromolecules with fac‐[99mTc(CO)3]+ for development of molecular imaging agents and the impact of this technology in the discovery and development of imaging and/or therapeutic agents towards clinical application.

Detection of DNA synthesis in intact organisms with positron-emitting (methyl- 11 C)thymidine

Abstract:(11)CO(2) produced in the Brookhaven 152-cm cyclotron was converted to formaldehyde, which in turn was used for the enzymatic conversion of deoxyuridine-5'-phosphate to [(11)C]thymidylate. Enzymatic treatment of the nucleotide with alkaline phosphatase gave [(11)C]thymidine.The preparation of [(11)C]thymidine from cyclotron-generated (11)CO(2) required 110 min (about 5 half-lives): 35 min for the synthesis of H(11)CHO, 25 min for the enzymatic conversion to [(11)C]thymidylate, 20 min for column chromatography, 5 min for phosphatase treatment, 10 min for evaporation, 2 min for filtration through an anion-exchange resin, and 13 min for miscellaneous manipulations.Positron-emitting [(11)C]thymidine and [(11)C]thymidylate were used for in vivo tracer studies of DNA synthesis in mice for periods of up to 3 hr. Findings with carbon-11 were consistent with earlier studies in which carbon-14 and tritium-labeled thymidine were used. For example, 3 hr after injection of [(11)C]thymidine, spleen DNA was labeled to a much greater extent than was liver DNA.

Abstract:Micro- and nanoplastics are emerging pollutants with a concerning persistence in the environment. Research into their environmental impact requires addressing challenges related to sensitively and selectively detecting them in complex ecological media. One solution with great potential for alleviating these issues is using radiolabeling strategies. Here, we report the successful introduction of a 64Cu radiotracer into common microplastics, namely polyethylene, polyethylene terephthalate, polystyrene, polyamide, and polyvinylidene dichloride, which allows the sensitive detection of mere nanograms of substance. Utilizing a Hansen Solubility Parameter screening, we developed a swelling and in-diffusion process for tetraphenylporphyrin-complexed 64Cu, which permits one-pot labeling of polymer particles.

Enhanced TROSY Effect In [2‐19F, 2‐13C] Adenosine and ATP Analogs Facilitates NMR Spectroscopy of Very Large Biological RNAs in Solution

Abstract:Large RNAs are central to cellular functions, and yet characterizing such RNAs remains outside the reach of solution NMR. We present two labeling technologies based on [2-19F, 2-13C]-adenosine, which allow the incorporation of aromatic 19F-13C spin pairs. The labels when coupled with the powerful transverse relaxation optimized spectroscopy (TROSY) enable us to probe RNAs comprising up to 124 nucleotides. With our new [2-19F, 2-13C]-adenosine-phosphoramidite, all resonances of the human hepatitis B virus epsilon RNA could be readily assigned. With [2-19F, 2-13C]-adenosine triphosphate, the 124 nt pre-miR-17-NPSL1-RNA was produced via in vitro transcription and the TROSY spectrum of this 40 kDa [2-19F, 2-13C]-A-labeled RNA featured sharper resonances than the [2-1H, 2-13C]-A sample. The mutual cancelation of the chemical-shift-anisotropy and the dipole-dipole-components of TROSY-resonances leads to narrow linewidths over a wide range of molecular weights. With the synthesis of a non-hydrolysable [2-19F, 2-13C]-adenosine-triphosphate, we facilitate probing of co-factor binding in kinase complexes and NMR-based inhibitor binding studies in such systems. Our labels allow straightforward assignment for larger RNAs via a divide-and-conquer/mutational approach. The new [2-19F, 2-13C]-adenosine precursors are extremely valuable additions to the RNA NMR toolbox and will allow the study large RNAs/RNA protein complexes in vitro and in cells.

Abstract:Nucleic acids are considered as fundamental molecules of living systems, which serve as universal genetic information messengers and repositories. To uncover the multifaceted aspects of nucleic acid function and metabolism within cells, labeling has become indispensable. This labeling technique enables the visualization, isolation, characterization, and even quantification of specific nucleic acid species. This review delves into cellular metabolic approaches for nucleic acid labeling, wherein enzymatic steps are employed to introduce nucleic acid modifications before conjugation with a label for detection or isolation. The discussion begins with metabolic labeling for DNA, RNA with various reactive groups and post‐transcriptional RNA labeling for RNA methylation and acetylation sites, emphasizing recent advancements in the field and then, we spotlighted pertinent applications for cellular imaging and sequencing. of labeling.

Abstract:ConspectusThe unambiguous quantification of biomolecules is of great significance in fundamental biological research as well as practical clinical diagnosis. Due to the lack of a detectable moiety, the direct and highly sensitive quantification of biomolecules is often a “mission impossible”. Consequently, tagging strategies to introduce detectable moieties for labeling target biomolecules were invented, which had a long and significant impact on studies of biomolecules in the past decades. For instance, immunoassays have been developed with radioisotope tagging by Yalow and Berson in the late 1950s. The later languishment of this technology can be almost exclusively ascribed to the use of radioactive isotopes, which led to the development of nonradioactive tagging strategy-based assays such as enzyme-linked immunosorbent assay, fluorescent immunoassay, and chemiluminescent and electrochemiluminescent immunoassay.
