Quantum DFT analysis and molecular docking investigation of various potential breast cancer drugs

Abstract:Breast cancer is the most often diagnosed and deadliest type of cancer in the world. These days, the world is facing the threat of breast cancer. It is essential to find drugs for an effective treatment method against this disease. In this study, electronic and thermodynamic properties, molecular electrostatic potential (MEP) analysis, and molecular docking investigation of five different breast cancer drugs were studied with Density Functional Theory (DFT). Our finding demonstrated that Lapatinib drug is more stable and less reactive than anastrozole, letrozole, neratinib and tucatinib. Results disclosed that all studied drugs are needed to lower energy to donating/accepting an electron to become cation/anion due to these molecules have a higher value of the negative ionization potential and electron affinity. For optical properties, we found that all five anticancer drugs situated within the visible light region. Lapatinib shows maximum wavelength absorption with minimum transition energy as compared to other four drugs. The docking studies revealed the binding affinity of the Lapatinib shows maximum against the breast cancer target proteins and tucatinib and neratinib also shows good binding affinity and the type of interactions between the ligand and active site of proteins was analysed and the highest binding affinity was observed by lapatinib against PDK1 (PDB ID: 1UU7), PAK4(PDB ID: 2X4Z), GSK3 (PDBID:1GNG) with a docking score of -12.2Kcal/mol, 11.5 kcal/mol and 11.4 kcal/mol. The highest binding affinity was observed against HER-2 (PDB ID: 2IOK) with a docking score of -9.8 Kcal/mol by lapatinib drug. Depending on effective binding ability, strong hydrogen bond interactions and other bond interactions with the amino acids located at the target sites of the target proteins show lapatinib as an effective drug for the treatment of breast cancer.
