Abstract:The most promising class of heterocyclic compounds in medicinal chemistry are those with the quinolin-2-one nucleus. It is a versatile heterocyclic molecule that has been put together with numerous pharmaceutical substances and is crucial in the creation of anticancer medications. In this view, the present research work deals with design, synthesis, and characterization of various analogous of quinolin-2-one nucleus and evaluation of their anticancer activity against MCF-7 cells (adenoma breast cancer cell line). Fourteen new compounds have been synthesised using suitable synthetic route and are characterized by FTIR, 1 H NMR, 13 C NMR and Mass spectral data. Molecular docking studies of the title compounds were carried out using PyRx 0.8 tool in AutoDock Vina program. All the synthesised compounds were exhibited well conserved hydrogen bonding with one or more amino acid residues in the active pocket of EGFR tyrosine kinase (PDB ID: 1m17). The docking score of the derivatives ranged from − 6.7 to − 9.5 kcal mol −1 , standard drug Imatinib with − 9.6 kcal mol −1 and standard active ligand 4-anilinoquinazoline with − 7.7 kcal mol −1 . The designed compound IV-A1 showed least binding energy (− 9.5 kcal mol −1 ) against EGFR tyrosine kinase receptor. Further, top scored compound, IV-A1 found to be most significant against MCF-7 cells with IC 50 value of 0.0870 μM mL −1 , TGI of 0.0958 μM mL −1 , GI 50 of 0.00499 μM mL −1 , LC 50 of 1.670 μM mL −1 .

Abstract:Tetraheterocyclic compounds, derived from natural sources and contemporary pharmaceuticals, have shown promise as multitarget therapeutic agents. However, their mechanisms of action remain partially understood. In this study, we synthesized a series of 6H-thiochromeno[2,3-c]quinolin-12(12H)-one derivative, totaling 26 compounds, and assessed their potential for therapeutic application. We evaluated their effects on cell proliferation and conducted NCI-60 cell panel assays. MTT assays revealed that select compounds exhibited notable antiproliferative activity against two breast cancer cell lines (MCF-7 and MDA-MB-468). Notably, compounds 17 and 18 displayed the highest cytotoxicity against these cell lines. Furthermore, one-dose assays of the NCI-60 human tumor cell line screening program identified compounds 6 , 7 , 16 , 18 , 20 , 24 , 25 , and 30 for further investigation. Subsequent five-dose cytotoxicity studies focused on compounds 18 and 20 , which met the threshold inhibition criteria across a panel of cell lines. Our study highlights the effectiveness of compounds 18 and 20 in targeting breast cancer cell lines. Molecular docking simulations revealed that these compounds bind to the active sites of topoisomerase I (TOPO I). Our findings suggest that these novel compounds are promising anticancer agents, particularly against breast cancer, and are worthy of consideration as lead pharmacological candidates. Abstract:Quinoline- and acridine-based drugs are widely used as anti-breast cancer agents. These drugs act through various mechanisms of action; for example, neratinib acts on epidermal growth factor receptor 1 (EGFR) and human epidermal receptor type 2 (HER2) enzymes, whereas amsacrine and pyrazoloacridine act on topoisomerase II enzymes. The objective of the present study was to explore and compare the interaction mechanism of knowledge-based designed derivatives of FDA-approved quinoline- and acridine-based drugs. The above-stated cancer proteins were collected from PDB, prepared by correcting any broken chains, removing water and adding charges. Docking studies were performed in the PyRx docking tool, and Discovery Studio Visualizer was used to visualize the molecular interactions. The result obtained found that neratinib had a better binding affinity towards EGFR and HER2 but weaker binding affinity towards topoisomerase IIα and topoisomerase IIβ. On the other hand, amsacrine and pyrazoloacridine were found to have better binding scores in topoisomerase IIα and topoisomerase IIβ instead of EGFR and HER2, as already proved experimentally. All the designed molecules have shown better binding affinity to their favourable enzymes and other enzymes involved in breast cancer development. The outcome reveals that the designed quinoline- and acridine-based drugs derivatives could be examined as a potent inhibitory drug of breast cancer for their strong multi-targeted inhibition ability and reactivity. Further synthesis, in vitro and in vivo investigations of designed derivatives may be done to prove their therapeutic potential in breast cancer treatment.

Abstract:Natural compounds including bis-heterocyclic moieties displayed several applications in pharmaceutical, agrochemical, and industrial fields. Di-thiazoles are effective core of several anticancer drugs such as bleomycin. Derived from importance of di-thiazoles, herein, we synthesized novel series of di-thiazoles from condensation reaction of thiazole thiosemicarbazone derivative with α-haloketones. Physical features and chemical structures of new compounds formed were discussed and assured based on their spectral and conformational analysis. The cytotoxic potency of newly synthesized thiazole derivatives against colon (HCT-116) and hepatocellular (HepG2) carcinoma-cell lines was determined by MTT assay. Compounds 7a, 9a, 9c, and 9e revealed marked anticancer activity especially against (HCT-116). Molecular docking study was executed over chosen compounds referring to expected promising features against LDH-5, which appeared agree with biological results. This study was utilized to predict prior binding-efficiency of active thiazole compounds.

Abstract:Abstract Breast cancer is the most common illness among women, accounting for 25% of all diagnoses, as stated by the American Cancer Society. Current research focuses on 43 compounds of styrylquinoline derivatives as potential inhibitors of tubulin to design a new drug that could potentially be effective against breast cancer cells in humans. The target compounds were subjected to a three-dimensional quantitative structure-activity relationship/comparative molecular similarity indices analysis (CoMSIA) approach, where CoMSIA models were used; the best results obtained are ( Q 2 = 0.84, R 2 = 0.97, r ext 2 {r}_{\text{ext}}^{2} = 0.91), H-bond acceptor field was discovered to be important for increasing inhibitory activity by examining the contour maps (54%), and it plays a key role in the prediction of anticancer activity. Based on the contour maps of the CoMSIA models, we obtained information that allows us to propose four new molecules with higher cancer inhibitory than the 43 compounds found in the literature. The molecular docking was applied to determine the likely types of binding between the tubulin protein (PDB ID: 4O2B) and the proposed compounds, and the results show that M1 has a higher total score of 6.53 and two interactions with important conventional hydrogen bond type, followed by compound M2 with a total score of 5.74. Furthermore, the designed molecules showed better pharmacokinetic properties based on absorption, distribution, metabolism, excretion, and toxicity properties. Molecular dynamics simulations at 100 ns were conducted to confirm the binding stability of the selected ligands (M1 and M2) with tubulin protein. The simulation parameters used in the current study are root mean square deviation, root mean square fluctuation, H-bond, Rg, solvent accessible surface area, and binding energy. As a result, the designed compounds (ligands M1 and M2) have shown noteworthy potential as a drug candidate for experimental in vivo and in vitro testing due to their potential inhibition of breast cancer. Finally, the study of retrosynthesis in this work facilitates the synthesis of drug candidates.

Abstract:Current breast cancer therapy does not always work optimally, and cases of resistance have been reported. Therefore, it is imperative to develop new effective drugs with minimal side effects through predictions of the epidermal growth factor receptor (EGFR) signalling pathway inhibition. Among chemicals with the potential as anticancer candidates for breast cancer are phenylthiourea derivatives. In this study, two phenylthiourea derivatives were synthesized: N- (4-methoxy)-benzoyl-N'-phenylthiourea and N-(4-trifluoromethyl)-benzoyl-N'-phenylthiourea. These compounds were docked with the EGFR receptor (code: 1M17.pdb) to predict their cytotoxic activity in silico using AutoDock tools. Furthermore, the microculture tetrazolium technique (MTT) was used to investigate in vitro cytotoxicity against MCF-7 cells. The in silico test revealed that the compounds N-(4-trifluoromethyl)-benzoyl-N'-phenylthiourea and N- (4-methoxy)-benzoyl-N'-phenylthiourea had a binding score of -8.2 and -7.3 kcal/mol, respectively, and the in vitro cytotoxic activity testing showed IC50 values of 0.37 mM and 0.38 mM, demonstrating significant EGFR inhibitory activity in MCF-7 cells. Therefore, it can be concluded that N-(4-trifluoromethyl)-benzoyl-N'-phenylthiourea has better cytotoxic activity than N-(4-methoxy)-benzoyl-N'-phenylthiourea.
Exploring the antitumor potential of novel quinoline derivatives via tubulin polymerization inhibition in breast cancer; design, synthesis and molecular docking

Abstract:A series of quinoline derivatives was designed and synthesized as novel tubulin inhibitors targeting the colchicine binding site. All the rationalized compounds 3a–e , 4a–e , 5a–e , and 6a–e have been chosen for screening their cytotoxic activity against 60 cell lines by NCI. Compounds 3b , 3c , 4c , 5c and 6c demonstrated the most notable antitumor activity against almost all cell lines. Compound 4c emerged as the most potent compound as an antiproliferative agent. This compound was subsequently chosen for five-dose testing and it exhibited remarkable broad-spectrum efficacy with strong antitumor activity against several cell lines. Compound 4c significantly induced cell cycle arrest in MDA-MB-231 cells at G2 and M phases where the cell population increased dramatically to 22.84% compared to the untreated cells at 10.42%. It also increased the population in MDA-MB-231 cells at both early and late stages of apoptosis. Compound 4c can successfully inhibit tubulin polymerization with an IC 50 value of 17 ± 0.3 μM. The β-tubulin mRNA levels were notably reduced in MDA-MB-231 cells treated with compound 4c which is similar to the effect observed with colchicine treatment. Docking studies revealed that compound 4c interacted well with crucial amino acids in the active site.

Abstract:A novel series of 1-(5-((6-nitroquinazoline-4-yl)thio)-1,3,4-thiadiazol-2-yl)-3-phenylurea derivatives 8 were designed and synthesized to evaluate their cytotoxic potencies. The structures of these obtained compounds were thoroughly characterized by IR, 1 H, and 13 C NMR, MASS spectroscopy and elemental analysis methods. Additionally, their in vitro anticancer activities were investigated using the MTT assay against A549 (human lung cancer), MDA-MB231 (human triple-negative breast cancer), and MCF7 (human hormone-dependent breast cancer). Etoposide was used as a reference marketed drug for comparison. Among the compounds tested, compounds 8b and 8c demonstrated acceptable antiproliferative activity, particularly against MCF7 cells. Considering the potential VEGFR-2 inhibitor potency of these compounds, a molecular docking study was performed for the most potent compound, 8c , to determine its probable interactions. Furthermore, computational investigations, including molecular dynamics, frontier molecular orbital analysis, Fukui reactivity descriptor, electrostatic potential surface, and in silico ADME evaluation for all compounds were performed to illustrate the structure–activity relationship (SAR).

Abstract:Breast cancer is the predominant form of cancer among women and ranks as the second most prevalent cancer globally, affecting both developed and less developed countries. Presently, accessible cancer treatment methods either employ recently created, secure, and efficient chemotherapeutic medications or directly target innovative pathways that cause apoptosis. One of the indirect strategies for treating this fatal illness has mostly depended on its essential role in cell cycle arrest and apoptosis induction, as well as the antagonistic interaction between the Bcl-2 and Mcl-1 proteins, in order to avert major health repercussions. We reported that newly synthesized fused chromenopyrimidines (3a and 4a) showed potential cell cycle arrest and dual Bcl-2 and Mcl-1 inhibitory characteristics. Bcl-2 and Mcl-1 were the targets of a molecular docking procedure. The previous docking results are in line with the biological data and suggest that 3a may have promising anti-cancer activity.

Abstract:This review highlights the potential of halogen‐substituted 4(3H)‐quinazolinone as an emerging class of chemotherapeutic drugs with potential efficacy in treating breast cancer. In‐depth analysis of structure–activity relationships shows the significance of various substitution patterns. The 6‐iodo derivative stood out as a hit molecule since it inhibits cell proliferation and has the most promising anticancer activity of all the derivatives. Currently, cancer is a serious health challenge with predominance beyond restrictions. Breast cancer remains one of the major contributors to cancer‐related morbidity and mortality in women. Chemotherapy continues to be crucial in the treatment of all variants of cancer. Several antitumor drugs are presently in different phases of clinical trials, whereas many more have been approved for clinical use. However, these drugs have the potential to cause adverse effects, and certain individuals may become resistant to them, which would eventually reduce the drug's efficacy. Therefore, it is essential to discover, develop, and improve newer anticancer drug molecules that could potentially inhibit proliferative pathways. In recent years, quinazolinone derivatives, more specifically halogen‐substituted 4(3H)‐quinazolinone, have drawn attention as a promising new class of chemotherapeutic agents. In addition, these molecules showed significant inhibition in micromolar ranges when tested in vitro against the MCF‐7 cell line. Therefore, this study aims to emphasize the intriguing versatility of halogen atoms, providing an in‐depth summary and highlighting recent developments in the anticancer properties of halogenated 4(3H)‐quinazolinones. It also features a detailed discussion of the structure–activity relationship (SAR) of various functional groups and their interaction with amino acid residues utilizing molecular docking studies. The intent is to foster novel discoveries that can inspire innovative investigations in this domain. Hence, this study simplifies the drug design and development strategies by prolonging the array of pharmacologically active candidates.

Abstract:Aim: 22 derivatives of 7-hydroxy-4-methyl-3-substituted benzopyran-2-one were designed, synthesized and evaluated for their anticancer activity. Materials & methods: The prepared compounds were screened for their cytotoxicity against the MCF-7 breast cancer cell line. The best five were then evaluated against MCF10a to check their safety and then tested for their PI3K and Akt-1 inhibitory action. The best two derivatives were further analyzed through cell cycle analysis, caspase 3/7 activation, increasing BAX level and decreasing BCL-2. Docking and absorption, distribution, metabolism and excretion prediction studies were also performed. Results & conclusion: Compounds 3b , 3c , 3j , 7 and 8 were the most active. Compounds 3c and 8 showed remarkable inhibitory action against PI3K and Akt-1 enzymes, and both are promising candidates for treatment of breast cancer.

Anti-Cancer Agents in Medicinal Chemistry

Abstract:Background: Cancer is one of the most common reasons for mortality in the world. A continuous effort to develop effective anti-cancer drugs with minimum side effects has become necessary. The use of small-molecule drugs has revolutionized cancer research by inhibiting cancer cell survival and proliferation. Quinazolines are a class of bioactive heterocyclic compounds with active pharmacophores in several anti-cancer drugs. Such small molecule inhibitors obstruct the significant signals responsible for cancer cell development, thus blocking these cell signals to prevent cancer development and spread. Objective: In the current study, novel quinazoline derivatives structurally similar to erlotinib were synthesized and explored as novel anti-cancer agents. Methods: All the synthesized molecules were confirmed by spectroscopic techniques like 1 H NMR, 13 C NMR, and ESI-MS. Various techniques were applied to study the protein-drug interaction, DFT analysis, Hirshfeld surface, and target prediction. The molecules were screened in vitro for their anti-cancer properties against 60 human tumor cell lines. The growth inhibitory properties of a few compounds were studied against the MCF7 breast cancer cell line. Results: The activity of compounds 9f, 9o, and 9s were found to be active. However, compound 9f is more active when compared with other compounds. Conclusion: Some synthesized compounds were active against different cancer cell lines. The in-vitro study results were found to be in agreement with the predictions from in-silico data. The selected molecules were further subjected to get the possible mechanism of action against different cancer cells.

Abstract:A series of new compounds based on 8-methyl-2-phenyl quinazolinone Schiff's base were synthesized from 3-amino quinazolinone intermediates. These compounds were evaluated for their potential as anticancer and antimicrobial agents through docking analysis. Anticancer evaluation was done against sixty different cancer cell lines. Compounds 7b and 7 h showed strong efficacy against Melanoma (MDA-MB-435 Cell lines) and Non-Small Cell Lung Cancer (NCI-H522), respectively. Compound 7 g was found to be predominantly effective against both breast cancer (HS-578T) and central nervous system cancer (SNB-19). The antimicrobial activity results showed that the newly synthesized compounds 7d and 7j, which contain halogen, exhibited potential inhibiting action against selected bacterial and fungal microorganisms. The SAR study revealed that compounds with EDGs like hydroxyl and meta -substituted chloro group were found to be more potent in anticancer studies; while compounds with EWGs substituted in the para position (7d and 7j) demonstrated higher antimicrobial activity. Moreover, Antimicrobial docking analysis indicated that compounds 7d and 7j have a high affinity towards molecular targets found in both bacteria and fungi, with negative binding energies ranging from -8 to -12.5 kcal/mol. We endorse further evaluation of these compounds in combination with standard antibiotics to potentially increase their synergistic effect.

Abstract:A novel series of 2-methyl-3-phenylquinazolin-4-one derivatives were synthesized and biologically evaluated for their cytotoxic potential against MCF-7, HepG2, and PC-3 cancer cells. Most of the tested compounds showed reasonable safety in the normal human skin melanocyte HFB4 cell line. Compound 4 showed potent cytotoxicity on Hep-G2 cell lines, while compound 9 showed potent cytotoxicity on the MCF-7 cell line, whereas compounds 10 and 12 showed potent cytotoxicity on Hep-G2 cell lines using 5-fluorouracil as a reference standard. Cell division analysis on the tested cell lines revealed that compounds 4 , 9 , 10 , and 12 have potent antiproliferative properties. An in vitro enzymatic inhibition assay against EGFR-TK confirmed that those compounds have potent EGFR inhibitory activity. The target compounds arrested the cell cycle at the pre-G1 and G2/M phases. Abstract:Introduction: A new series of 4-hydroxy-8-methyl-2-oxo-1,2-dihydroquinoline-3-carboxamide derivatives has been synthesized in good yields, followed by complete characteri-zation using 1D-NMR, 2D-NMR, and IR techniques. Methods:: The final products contain amide, hydroxyl, and aromatic functional groups that usu-ally show significant bioactivity. The target products have been examined towards three cancer cell lines, namely colorectal cancer cell line (HCT116), breast cancer cell line (MCF-7), and leukemia cell line (K562) in addition to the fibroblast cells, that were used as a model for normal human tissue. Results:: The anticancer results signified that compound 6 showed the most activity in the series accomplished with IC50 values of 14.6, 5.3 and 12.8 μM, Conclusion:: Other compounds exhibited considerable activity, such as compounds 9 (IC50 3.5 and 19.0 μM), 10 (IC50 12.6 μM), and 11 (IC50 10.3 μM) against the three cancer cell lines HCT116, MCF-7 and K562, respectively.

Unveiling Novel Hybrids Quinazoline/Phenylsulfonylfuroxan Derivatives with Potent Multi‐Anticancer Inhibition: DFT and In Silico Approach Combining 2D‐QSAR, Molecular Docking, Dynamics Simulations, and ADMET Properties

Abstract:Biological activities of 29 novel quinazoline/phenylsulfonylfuroxan derivatives were in silico investigated as potential anticancer inhibitors against five cell lines, i.e., H1975, MCF‐7, Eca‐109, MGC‐803, and A549, which are involved in lung, breast, esophageal squamous carcinoma, and gastric cancer diseases. The combining QSAR predictive approach, molecular docking, and dynamics simulations allow the design of 70 new hybrids and predict their pIC50 activity. In this work, the biological activities of 29 novel quinazoline/phenylsulfonylfuroxan derivatives (1a–z, 1aa, 1ab, 2a, 2b, 2d, and 2f) were computationally investigated as potential anti‐cancer inhibitors against five cell lines, i.e., H1975, MCF‐7, Eca‐109, MGC‐803, and A549, which are involved in various diseases, including lung, breast, esophageal squamous carcinoma, and gastric cancer. The 2D‐QSAR predictive approach, exploiting multiple linear regression (MLR) models and rigorous internal and external cross‐validation, showed a correlation factor R2 of range: 0.68−0.82. Moreover, the MLR‐derived R2test and Y randomization (R2rand) values for the five cell lines are higher than 0.60 and less than 0.3, respectively, indicating a strong alignment with the internal and external validation data. New 70 quinazoline hybrids based on the most effective in vivo 1q inhibitor were designed, and their pIC50 activity was predicted. The best‐scoring 15 (N1–N15) compounds were further evaluated using molecular docking and dynamics simulations (100 ns) with the VEGFR‐2 kinase target (PDB code: 3U6J). All the data sets accurately predict the strongest binding affinity for the selected (N6, N7, N9, and N11) molecules, as evidenced by the highest docking score, hydrogen bond energy, and significant amino acid steric interactions. Furthermore, the RMS/RMSF/Rg dynamics parameters show that the formed complexes are satisfactorily stable. The ADMET properties indicate that the selected new ligands have shown a promising drug‐like profile and can be considered potential candidates for future anti‐cancer therapies, with perspective validating their anticancer activity by in vitro and in vivo studies.

Abstract:Cancer affects millions of people worldwide. PDK1 enzyme (co-crystallized with BIM-1) controls the proliferation of breast cancer cells. Aiming to resemble BIM-1's binding, quinoline-based pyrimidinediones and thiazolidinediones were synthesized starting from 2-chloro-3-formylquinoline. Compared with doxorubicin (reference), in vitro antiproliferative activity against MCF7 and HCT116 cancer cell lines showed the most potency of thiobarbiturate 3 and thiazolidinedione 4 . In silico molecular docking, DFT, and pharmacokinetics simulations supported the findings. The docking analysis toward PDK1 enzyme showed that most amino acids interacting with co-crystallized ligand (BIM-1) were successfully bonded to our docked substances, especially thiobarbiturate 3 with highest S-score closer to BIM-1. In DFT calculations, this compound exhibited the lowest energy gap and highest softness leading to more response to radical surface interactions. The compounds with significant antiproliferative activity exhibited high electrophilicity values. ADME analysis showed its desirable drug-likeness and oral bioavailability. This work may contribute to developing new potent antiproliferative agents.

Abstract:Various targets, such as estrogen receptor (ER), mammalian target ofrapamycin(mTOR), epidermal growth factor receptor (EGFR), androgen receptor (AR) andpoly adenosine diphosphate-ribose polymerase (PARP), are focused on the treatment of breast cancer. In this study, in silico tools such as pharmacophore modeling, 3D QSAR study, molecular docking, binding free energy determination and molecular dynamics were executed on a series of pyrazolopyrimidines derivatives. The pharmacophore modeling of forty-one anticancer derivatives was generated, and atom-based 3D QSAR was applied. Molecular docking, dynamics, binding energy and high-throughput virtual screening (HTVS) were conducted by the software Schrodinger. The best five featured pharmacophore hypotheses AHRRR_1 with a maximum survival score of 5.533 was subjected to rigorous scoring function analysis and 3D QSAR studies. Docking studies and binding free energy were carried out on 41 inhibitors for their anti-breast cancer activity against human estrogen, progesterone receptor, EGFR kinase and mammalian target of rapamycin mTOR. Molecular dynamics simulation of the docked complex-34/4WKQ validated the stability of this complex. HTVS was performed to determine the virtual hits with the best-fitted model AHRRR_1. New EGFR kinase inhibitors were designed based on the active compound templates. Molecular docking was carried out on virtual hits, and newly designed compounds and dynamics studies revealed that the binding modes obtained after MD simulation were more or less similar to that acquired post docking mode. Promiscuity assessments demonstrated that designed compounds would be specific rather than a promiscuous one. Based on these findings, we have designed four novel compounds as anti-breast inhibitors, with potent binding affinity and desirable ADME properties.

Abstract:Specific enzyme activities of the non-oxidative pentose phosphate pathway in Aspergillus oryzae mycelia grown on different carbon sources were determined. Mycelia grown on glucose, mannitol and ribose show the highest specific activities, ribose 5-phosphate isomerase being specially very enhanced. Moreover, transketolase, transaldolase, ribose 5-phosphate isomerase and ribulose 5-phosphate 3-epimerase were determined in different developmental stages of mycelia grown on glucose, mannitol and ribose. The non-oxidative pentose phosphate pathway is more active during conidiogenesis, except for ribulose 5-phosphate 3-epimerase, suggesting a fundamental role of this pathway during that stage to supply pentoses for nucleic acids biosynthesis. A general decrease of the enzyme activities was found in sporulated mycelia. Arabinose 5-phosphate was tested as metabolite of the pentose pathway. This pentose phosphate was not converted into hexose phosphates or triose phosphates and inhibits significantly the ribose 5-phosphate utilization, being therefore unappropriate to support the Aspergillus oryzae growth.

Abstract:In the presented work, a series of 22 hybrids of 8-quinolinesulfonamide and 1,4-disubstituted triazole with antiproliferative activity were designed and synthesised. The title compounds were designed using molecular modelling techniques. For this purpose, machine-learning, molecular docking, and molecular dynamics methods were used. Calculations of the pharmacokinetic parameters (connected with absorption, distribution, metabolism, excretion, and toxicity) of the hybrids were also performed. The new compounds were synthesised via a copper-catalysed azide-alkyne cycloaddition reaction (CuAAC). 8-N-Methyl-N-{[1-(7-chloroquinolin-4-yl)-1H-1,2,3-triazol-4-yl]methyl}quinolinesulfonamide was identified in in silico studies as a potential strong inhibitor of Rho-associated protein kinase and as a compound that has an appropriate pharmacokinetic profile. The results obtained from in vitro experiments confirm the cytotoxicity of derivative 9b in four selected cancer cell lines and the lack of cytotoxicity of this derivative towards normal cells. The results obtained from silico and in vitro experiments indicate that the introduction of another quinolinyl fragment into the inhibitor molecule may have a significant impact on increasing the level of cytotoxicity toward cancer cells and indicate a further direction for future research in order to find new substances suitable for clinical applications in cancer treatment.
