Abstract:<p>Protein, as the material basis of vita, is the crucial undertaker of life activities, which constitutes the framework and main substance of human tissues and organs, and takes part in various forms of life activities in organisms. Separating proteins from biomaterials and studying their structures and functions are of great significance for understanding the law of life activities and clarifying the essence of life phenomena. Therefore, scientists have proposed the new concept of proteomics, in which protein separation technology plays a momentous role. It has been diffusely used in the food industry, agricultural biological research, drug development, disease mechanism, plant stress mechanism, and marine environment research. In this paper, combined with the recent research situation, the progress of protein separation technology was reviewed from the aspects of extraction, precipitation, membrane separation, chromatography, electrophoresis, molecular imprinting, microfluidic chip and so on.</p>

Abstract:Food proteins are important nutrients for human health and thus make significant contributions to the unique functions of different foods. The modification of proteins through physical and biological processing could improve the functional and nutritional properties of food products; these changes can be attributed to modifications in particle size, solubility, emulsion stability, secondary structure, as well as the bioactivities of the proteins. Physical processing treatments might promote physical phenomena, such as combined friction, collision, shear forces, turbulence, and cavitation of particles, and lead to changes in the particle sizes of proteins. The objective of this review is to illustrate the effect of physical and biological processing on the structure, and physical and chemical properties of food-derived proteins and provide insights into the mechanism underlying structural changes. Many studies have suggested that physical and biological processes, such as ultrasound treatment, high pressure homogenization, ball mill treatment, and enzymatic hydrolysis could affect the structure, physical properties, and chemical properties of food-derived proteins. Some important applications of food-derived proteins are also discussed based on the relationships between their physical, chemical, and functional properties. Perspectives from fundamental or practical research are also brought in to provide a complete picture of the currently available relevant data.

Abstract:As complex biological macromolecules, protein plays a vital role in the activities of living activities. In recent years, more and more food products are based on protein as their raw materials and accessories. The properties of the protein have a great influence on the quality, production and processing, and the storage conditions of food. It can improve the function and application characteristics of protein by modifying the protein. At present, ultra-fine grinding is widely used in food processing because of its unique processing characteristics. Compared with the general pulverization technology, the ultra-fine grinding technology can crush the particle size of materials to 10 μm, or even 1 μm. It also has many characteristics, such as small nutrition loss of products, convenience, safety and high quality of products. This article mainly introduced the effects of many kinds of ultra-fine grinding techniques on the physicochemical and functional properties of proteins. It also provided some ideas for development of ultra-fine grinding technology to obtain protein products with specific characteristics.

Abstract:Protein is one of the most important components of food which significantly contributes to the structure, functionality, and sensory properties which may affect consumer acceptability of processed products. Conventional thermal processing affects protein structure and induce undesirable degradation of food quality. This review provides an overview of emerging pretreatment and drying technologies (plasma treatment, ultrasound treatment, electrohydrodynamic, radio frequency, microwave, and superheated steam drying) in food processing by assessing protein structural changes to enhance functional and nutritional properties. In addition, mechanisms and principles of these modern technologies are described while challenges and opportunities for the development of these techniques in the drying process are also critically analyzed. Plasma discharges can lead to oxidative reactions and cross-linking of proteins that can change the structure of proteins. Microwave heating contributes to the occurrence of isopeptide or disulfide bonds which promotes α-helix and β-turn formation. These emerging technologies can be adopted to improve protein surface by exposing more hydrophobic groups which restrict water interaction. It is expected that these innovative processing technologies should become a preferred choice in the food industry for better food quality. Moreover, there are some limitations for industrial scale application of these emerging technologies that need to be addressed.

Abstract:Background: In recent years, animal proteins are being replaced by plant-based proteins worldwide. Artificial meat and eggs made from plant-based proteins have been widely commercialized, especially in developed countries such as the United States. However, the traditional wet separation can damage the isolated proteins as well as have a negative impact on the environment. Therefore, there is a growing research interest in developing alternative methods, such as dry separation, for high quality plant protein separation.Scope and approach: Electrostatic separation, a dry technology based on the different triboelectric charging properties of materials, has been used for plant protein separation in recent years. This review summarizes the technical issues that need to be considered, from material preparation to equipment construction, for electrostatic separation. Moreover, the separation efficiency, energy consumption, combination with other separation methods, the functionality of the resulting protein concentrates, and prospects for electrostatic separation are also reviewed here.Key findings and conclusions: Factors influencing the outcomes of electrostatic separation include the pretreatment of materials, design of the electrostatic separator, and process parameters. The separation efficiency of electrostatic separation can be further improved by cyclic separation or a combination of electrostatic separation with other methods. However, although the proteins obtained by electrostatic separation maintain their native functional properties, they are affected by the presence of other components such as phenolics or starch. Thus, electrostatic separation has several limitations that need to be addressed before it can be used widely.

Abstract:Global populations are growing and ageing, with each generation expecting a higher standard of living than their predecessor. Proteins are essential in daily diets because of their nutritional value and roles in food structure, and proteins are generally expensive. Rising awareness of food security and meeting the needs of ageing populations motivate food researchers to explore alternative protein sources and sustainable and optimized use of proteins to increase their bioavailability and digestibility. Proteins behave differently as a function of pH, ionic strength, temperature, pressure or enzymatic mechanism, prompting approaches to modify their structure and functionality. Proteins may lose their native structure and impart sensory changes as a result of processing and storage. Modified proteins are considered as value-added food ingredients, i.e. specialty proteins prepared using enzymatic reactions, hydrolysis, fermentation, heat treatment, acidification, dehydration, emulsification and ultrafiltration. Food formulation influences proteins at both molecular and macro-polymer levels. This review presents case studies from previous research and demonstrates approaches for improving protein stability, extractability and bioactivity. The need for combining protein modification and smart food formulation is highlighted. Synergies achieved between conventional ingredients and added bioactives through careful food formulation and processing are emphasized. Comprehensive characterization of chemical composition, particle size, surface activity, microstructure, freezing-thawing properties of proteins and derived foods is required, to explain structure-function relationships. Data suggest that specialty proteins will play an increasing role in current and future global food sectors. Convenient concept foods for addressing the global food security and ageing population issues are proposed.

High quality isolation and purification is an essential step in the study of the structure and function of target proteins. Therefore, the development of protein purification technologies has great theoretical and practical significance in exploring the laws of life activities and guiding production practice. Up to now, there is no forthcoming method to extract any proteins from a complex system, and the field of protein purification still faces significant opportunities and challenges. Conventional protein purification generally includes three steps: pretreatment, rough fractionation, and fine fractionation. Each of the steps will significantly affect the purity, yield and the activity of target proteins. The present review focuses on the principle and process of protein purification, recent advances, and the applications of these technologies in the life and health industry as well as their far-reaching impact, so as to promote the research of protein structure and function, drug development and precision medicine, and bring new insights to researchers in related fields.

Abstract:This article reviews the influence of ultrasonic treatment on protein extraction, modification (structure and function) and application. Summary The advancement of the modern food industry has prompted a shift away from traditional protein extraction methods due to their inherent limitations. As a result, there is a growing interest in exploring alternative methods for protein extraction and modification such as ultrasonic (US) treatment. This shift towards more sustainable and efficient protein extraction methods is a key research focus in obtaining safe proteins in an environmentally friendly manner. Furthermore, enhancing the functional properties of proteins through modification can expand their application. This paper reviews recent research progress and applications of US treatment on protein extraction and modification as well as the impact on protein structure and functional properties. In addition, the latest applications of ultrasound‐modified‐proteins, including nano‐proteins, 3D printing and interactions between proteins and other food components, are reviewed. This review may provide some inspiration for the potential application of US technologies in food fields.

Abstract:Enzymatic hydrolysis (EH) of proteins relies essentially upon enhancing the functional and nutritional properties of proteins, such as antioxidant activity, solubility, oil holding capacity, water holding capacity, emulsification, foaming properties, and sensory properties. There is a big challenge for protein separation and purification due to the high production cost, the large number of amino acids, and the complex biological system of proteins. These biological structures are always presented in a multi-component mixture in native environments, which are usually similar to other molecular weights of other components, such as protein-lipid complexes. These difficulties present the importance of a combination of the hydrolysis process of protein with adequate technology for separation and purification to achieve better bioactive peptide recovery. There are many studies conducted dealing with the characteristics of the hydrolysis process of proteins, and other implications, such as the separation of protein and bioactive peptides. Therefore, this study aims to review the important and recent research papers that investigated the effect of various conditions of the enzymatic hydrolysis process (EHP) (type of enzyme, enzyme to substrate ratio, temperature, pH, and time) on the antioxidant capacity of protein hydrolysates and degree of hydrolysis, as well as to assess the recent studies about protein purification and potential separation techniques.

Abstract:Background: Protein is an essential component of human diet and can be applied to many aspects in food systems due to its abundant nutritional value and functional properties. There are many physical methods that have been used to modify the inherent structure of protein to expand its application areas in the food industry. Among them, electric fields and electromagnetic wave technologies have attracted increasing attention on their abilities to modify food protein structure and functionality, due to the advantages of energy efficiency, food safety and minimal loss of nutrients. Scope and approach: The current review presents the effects of electric fields and electromagnetic wave including pulsed electric field, microwave, radio frequency and gamma irradiation on the changes in food protein structure (primary, secondary, tertiary and quaternary) and functionality (solubility, apparent viscosity, emulsifying, foaming, and gelling properties). The affecting factors such as protein concentration and pH, and the strength and duration of electric fields and electromagnetic wave on the mechanisms and effectiveness of changes in protein structure and functional properties are introduced, and the advantages and limitations of these technologies for protein modification are also discussed. Key findings and conclusions: Applications of electric fields and electromagnetic wave can induce the conformational changes of protein via the creation of free radicals or larger or smaller molecules, damaging the primary, secondary, tertiary and quaternary structure of protein, and thus influence the functional properties. Therefore electric fields and electromagnetic wave are useful methods to modify food protein structure and functionality for the food industry, and active researches focus on multi-technology corporations for modifying protein structure are urgently required.

Abstract:Proteins are important food ingredients that possess both functional and nutritional properties. High hydrostatic pressure (HHP) is an emerging nonthermal food processing technology that has been subject to great advancements in the last two decades. It iswell established that pressure can induce changes in protein folding and oligomerization, and consequently, HHP has the potential to modify the desired protein properties. In this review article, the research progress over the last 15 years regarding the effect of HHP on protein structures, as well as the applications of HHP in modifying protein functionalities (i.e., solubility, water/oil holding capacity, emulsification, foaming and gelation) and nutritional properties (i.e., digestibility and bioactivity) are systematically discussed. Protein unfolding generally occurs during HHP treatment, which can result in increased conformational flexibility and the exposure of interior residues. Through the optimization of HHP and environmental conditions, a balance in protein hydrophobicity and hydrophilicity may be obtained, and therefore, the desired protein functionality can be improved. Moreover, after HHP treatment, there might be greater accessibility of the interior residues to digestive enzymes or the altered conformation of specific active sites, which may lead to modified nutritional properties. However, the practical applications of HHP in developing functional protein ingredients are underutilized and require more research concerning the impact of other food components or additives during HHP treatment. Furthermore, possible negative impacts on nutritional properties of proteins and other compounds must be also considered.

Abstract:The functional properties of peanut protein can be altered by different modification and processing methods. And the applications for peanut protein include meat and milk substitutes, edible films, nanoparticles, and some new technologies such as electrostatic spinning and three‐dimensional (3D) printing in recent years. Summary Peanuts produce millions of tons of high‐protein peanut meal (after oil extraction) annually that can be used for food. Peanut protein, as a plant‐based protein, is a great alternative to animal protein to address protein deficiencies and rising health problems in the world. It has a high nutritional value, while the globular structure results in poor functional properties, limiting its application. Better functional properties can be obtained through processing and modification. This paper elaborates the nutritional and functional properties of peanut protein and highlights the application of peanut protein in food industry in recent years. Applications for peanut protein include meat and milk substitutes, edible films, nanoparticles, and some new technologies such as electrostatic spinning and three‐dimensional (3D) printing in recent years. This paper could improve people's understanding of the molecular structure of peanut protein and the physicochemical properties associated with the structure changes during processing. It could also contribute to better utilisation of peanut protein, providing more varieties of peanut protein products for the food industry.
