This one-step synthesis of spirooxindole is both step- and atom-economic, proceeding with high regioselectivity in moderate to excellent yields.

Abstract:A facile and efficient approach for the synthesis of spirooxindoles has been developed via the coupling of spirocyclic C,C-palladacycles with CH2Br2. Abstract:A novel palladium-catalyzed [4 + 1] cycloaddition to give spirotetrahydrofuran oxindoles has been developed, in which diphenyl 2-oxoindolin-3-yl phosphates were used as the both electrophilic and nucleophilic C1 synthons at C-3 of the oxindole unit and 2-methylidenetrimethylene carbonate was used as the 1,4-dipole. The cycloannulation was performed at room temperature and provided the corresponding spirotetrahydrofuran oxindoles in good to excellent yields. The present method affords a new strategy for the construction of spirooxindole derivatives with unique three-dimensional structures.

Abstract:A palladium-catalyzed domino cyclization/cycloisomerization reaction of alkyne-tethered carbamoyl chlorides with (E)-β-chloroenones is reported. This reaction proceeds via a syn-carbopalladation of the alkyne, followed by a vinyl-PdII-catalyzed cycloisomerization of the (E)-β-chloroenone cascade, which provides an efficient method to synthesize furan-linked methylene oxindoles. The reaction features stereodefined vinyl-PdII species, high to excellent 5-exo/6-endo selectivity, excellent Z/E selectivity, and the sequential formation of three bonds and bis-heterocycles. The strategy for the synthesis of furan-containing benzofurans has also been demonstrated.

Abstract:A palladium-catalyzed decarboxylative asymmetric [4 + 2] annulation of methyleneindolinones with a zwitterionic oxo-1,4-dipole intermediate was successfully developed to access spirocyclic oxindoles bearing two vicinal stereocenters in good yields with highly diastereoselectivities and enantioselectivities. This strategy is featured with a broad substrate scope (28 examples), allowed for efficient scale-up. Further selective transformation of the product and preliminary mechanistic studies were well conducted.

Abstract:An efficient method for the practical formation of functionalized spirooxindoles, which was accompanied by generating a sterically congested spiroketal system and bearing two quaternary stereocenters via a sequential palladium, catalyzed decarboxylative [3 + 2] cycloaddition of vinylethylene carbonates (VECs) and isatins. Importantly, the redox‐neutral catalytic system possesses the characteristics of operational convenience, good functional group tolerance, and good regioselectivity and diastereoselectivity, which enables a diastereoselective synthesis of spiroketal oxindole skeletons in an excellent yield. An efficient method for the practical formation of functionalized spirooxindoles, which was accompanied by generating a sterically congested spiroketal system and bearing two quaternary stereocenters via a sequential palladium, catalyzed decarboxylative [3 + 2] cycloaddition of vinylethylene carbonates (VECs). This conversion is characterized by excellent reactivity and diastereoselectivity, mild reaction conditions, operational convenience, and wide substrate scope and provides an efficient and concise route to some complicated skeletons.

Abstract:A thermally induced, substrate-dependent reaction of alkynyl diazo compounds has been developed. This transformation produces spiro-4H-pyrazole-oxindoles and fused 1H-pyrazoles in good to high yields from the corresponding alpha-cyano and alpha-sulfonyl diazo compounds. The salient features of this reaction include excellent chemoselectivity and atom-economy, mild reaction conditions, simple purification and potential for large scale production.

Abstract:A facile promiscuous reaction catalyzed by Acylase ‘Amano’ was developed for the preparation of spirooxindole derivatives. At the optimized reaction condition, the acylase-catalyzed domino reactions could provide a series of different spirooxindole products in moderate yields and good diastereoselectivity. A chemo-enzymatic two-step process was also designed successfully, and several representative spirooxindole derivatives could be obtained in nearly quantitative yields. Additionally, one tentative mechanism for this enzyme-catalyzed process was proposed.

Abstract:A novel palladium catalyzed homodimerization of ortho-hydroxyphenyl substituted p-QMs has been developed via [4 + 2] cycloaddition/oxidative dehydrogenation coupling domino reactions. An interesting palladium catalyzed intramolecular benzyl C-H oxidation dehydrogenation to form a transannular C(sp3)-O bond was found. This protocol provided an efficient method to construct various dibenzodioxo[3.3.1]nonanes bearing spirocyclohexadienones.

Abstract:A novel palladium-catalyzed [4+3]-annulation of oxotryptamines with allyl dicarbonates has been developed. The reaction furnishes various spiro[azepane-4,3'-oxindoles] in moderate to excellent yields (30%-96%) with up to >20:1 Z/E ratio through a tandem allylic substitution strategy. This protocol enables the synthesis of a wide range of seven-membered spirocyclic oxindoles in high Z-selectivity. In addition, the gram-scale reaction and further derivatizations were also investigated, demonstrating the synthetic utility of this approach.

Abstract:Described herein is the development of a palladium(II)-catalyzed two- or three-component reaction of 2-(1-alkynyl)-2-alken-1-ones with nucleophiles and allylic chlorides. Various types of nucleophiles such as O-, N-, C-based nucleophiles and olefin-tethered O-, N-, C-based nucleophiles were investigated. The scope, mechanism and application of this Pd(II) -catalyzed domino reaction were studied. In these transformations, the palladium catalyst exhibits a dual role, serving simultaneously as a Lewis acid and a transition metal. Two possible reaction pathwavs (cross-coupling reaction vs. Heck reaction) from the same intermediate furanylpalladium species were observed. The reaction pathway is dependent on the property of the nucleophile and the length of the tethered chain as well. When olefin-tethered O- based nucleophiles were used., only the cross-coupling reaction pathway was observed, in contrast, both reaction pathways were observed when olefintethered C-based nucleophiles were employed. The product ratio is dependent on the length of the tethered chain. Furthermore, ring-closing metathesis (RCM) of corresponding furans with C=C bonds provides an easy method for the preparation of functionalized oxygen-heterocycles - 3,4-fused bicyclic furans. It is also noteworthy that allylic chloride can be as an oxidant besides its well known function as an alkylating reagent.

Abstract:The first organocatalytic asymmetric synthesis of a spirooxindole skeleton incorporated with a cyclobutane moiety has been successfully developed on the basis of H-bond-directing dienamine activation. Structurally complex spirocyclobutyl oxindoles, which possess four contiguous stereocenters, including one spiro quaternary center, were obtained in good yields (up to 83%) with excellent β,γ-regioselectivity (>19:1) and stereocontrol (up to >19:1 dr and 97% ee).

Abstract:The ubiquitous presence of spirooxindole architectures with several functionalities and stereogenic centers in bioactive molecules has been appealing for the development of novel methodologies seeking their preparation in high yields and selectivities. Expansion and refinement in the field of asymmetric organocatalysis have made possible the development of straightforward strategies that address these two requisites. In this review, we illustrate the current state-of-the-art in the field of spirooxindole synthesis through the use of non-covalent organocatalysis. We aim to provide a concise overview of very recent methods that allow to the isolation of unique, densely and diversified spirocyclic oxindole derivatives with high structural diversity via the use of cascade, tandem and domino processes.
