Abstract:The creation of a new metal-organic framework (MOF) with a hollow hierarchical porous structure has gained significant attention in the realm of enzyme immobilization. The present work employed a novel, facile, and effective combinatorial technique to synthesize modified MOF (N-PVP/HZIF-8) with a hierarchically porous core-shell structure, allowing for the preservation of the structural integrity of the encapsulated enzyme molecules. Scanning electron microscopy, X-ray diffraction, X-ray photoelectron spectroscopy, confocal laser scanning microscopy, and other characterization tools were used to fully explore the changes of morphological structure and surface properties in different stages of the preparation of immobilization enzyme CRL-N-PVP/HZIF-8, thus showing the superiority of N-PVP/HZIF-8 as an enzyme immobilization platform and the logic of the immobilization process on the carrier. Additionally, the maximum enzyme loading was 216.3 mg mL-1, the relative activity of CRL-N-PVP/HZIF-8 increased by 15 times compared with the CRL@ZIF-8 immobilized in situ, and exhibited quite good thermal, chemical, and operational stability. With a maximal conversion of 88.8%, CRL-N-PVP/HZIF-8 demonstrated good catalytic performance in the biosynthesis of phytosterol esters as a proof of concept. It is anticipated that this work will offer fresh concepts from several perspectives for the creation of MOF-based immobilized enzymes for biotechnological uses.

Abstract:Poly(vinyl alcohol)-sodium alginate composite hydrogels were first introduced to synthesize robust and well-intergrown zeolite imidazolate framework (ZIF)/polymer hollow fiber membranes. Through enough adsorption interaction with metal ions by chelation, sufficient nucleation sites for in situ metal-organic framework (MOF) preparation are provided. Using this method, we can not only easily prepare defect-free MOF membranes ignoring the complex modification process and seed deposition but also structurally fix crystalline MOF layers and greatly improve the stiffness and durability of MOF composite membranes. The strategy also gives the appropriate level of generality for synthesis of versatile dense MOF membranes on a variety of polymeric supports. The fabricated ZIF-8/polyethersulfone membrane presented remarkable gas separation performance with H2 permeance of as high as 9.66 × 10-7 mol m-2 s-1 Pa-1 and a high H2/CO2 separation factor of up to 29.0.

Abstract:Zeolitic imidazolate frameworks (ZIFs) have recently emerged as immobilization matrices for biomolecules, most notably enzymes. Understanding the key factors that dominate the enzyme's catalytic activity on/in ZIFs is crucial for the development of new immobilization matrices. In this work, a combination of the parallel tempering Monte Carlo simulation and all-atom molecular dynamics simulation is performed to study the orientation and conformation of the Candida rugose lipase (CRL) adsorbed on oppositely charged and neutral ZIF-8 (i.e., ZIF-8-COOH, ZIF-8-NH<sub>2</sub>, and ZIF-8-neutral) surfaces. The results show that CRL could adsorb on all ZIF-8 surfaces, with an ordered orientation obtained on charged ZIF-8 surfaces. ZIF-8-NH<sub>2</sub> is a good candidate for CRL immobilization since it can maximize the catalytic activity of CRL. The native conformation of CRL is well preserved on all three surfaces due to the partially water-containing surface of ZIF-8. The results could provide theoretical support for the application of porous materials in enzyme immobilization.

Abstract:This study investigates the immobilization of cellulase on zeolitic imidazolate frameworks (ZIFs) by physical adsorption, specifically the ZIF-8-NH 2 and Fe 3 O 4 @ZIF-8-NH 2 , to enhance enzymatic hydrolysis efficiency. The immobilization process was thoroughly analyzed, including optimization of conditions and characterization of ZIF carriers and immobilized enzymes. The impacts on the catalytic activity of cellulase under various temperatures, pH levels, and storage conditions were examined. Additionally, the reusability of the immobilized enzyme was assessed. Results showed the cellulase immobilized on Fe 3 O 4 @ZIF-8-NH 2 exhibited a high loading capacity of 339.64 mg/g, surpassing previous studies. Its relative enzymatic activity was found to be 71.39%. Additionally, this immobilized enzyme system demonstrates robust reusability, retaining 68.42% of its initial activity even after 10 cycles. These findings underscore the potential of Fe 3 O 4 @ZIF-8-NH 2 as a highly efficient platform for cellulase immobilization, with promising implications for lignocellulosic biorefinery.

Abstract:In this work, we have described the adsorption of dye acid blue 40 and methylene blue from aqueous solution onto isotypic zeolite imidazolate frameworks (ZIFs) such as ZIF-8 [Zn(mim)(2)] and ZIF-67 [Co(mim)(2)]. The results suggested that the charged sites at the external surface of zeolite imidazolate frameworks play a key role in dye adsorption. A similar adsorption capacity is observed for the uptake cationic dye methylene blue between ZIF-8 and ZIF-67, involving electrostatic interactions between the same N-extremities (belonging to mim linkers) and positively charged methylene blue. For anionic dye acid blue 40, the adsorption capacity for ZIF-67 is nearly three times higher than that of ZIF-8, involving electrostatic interactions between low-coordinated metal cations of zeolite imidazolate frameworks and negatively charged groups of acid blue 40 and Co cations in ZIF-67 have higher affinity with the SO3- groups of acid blue 40.

Abstract:Searching adsorbents for high recognition and adsorption capacities of active peptides has become the hotspot of scientific research in the screening and purification of peptides. In this work, we selected MOFs materials with good biocompatibility to systematically investigated for the selective adsorption of highly active ACE inhibitory peptides. Herein, five typical kinds of metal organic frameworks (MOFs) were initially screened for their adsorption capacities towards di-peptide (CW) having ultra-high angiotensin-converting enzyme (ACE) inhibitory activity (IC 50 = 0.16 μM). The effects of their pore sizes and surface potentials on CW adsorption capacity were explored in this work. Among these MOFs, ZIF-8 was chosen as a di-peptide adsorbent due to its excellent adsorption capacity of CW (0.382 mmol/g). Besides, the adsorption behavior of ZIF-8 for di-peptides in the mixed solution was systematically explored. Experimental results indicated that ZIF-8 exhibited highly specific adsorption towards CW while showing very low and almost non-adsorption for the two inactive peptides, DD and RR. In their ternary peptide solution (molar ratio: CW/DD/RR = 1/10/10) for simulated protein hydrolysate, ZIF-8 displayed ultra-high adsorption selectivity (α = 46.6) for CW and thus enhanced almost 70.9 times the ACE inhibitory activity for the enriched solution. Meanwhile, molecular simulation revealed that ZIF-8 had high recognition towards CW, which was primarily attributed to synergistic mechanisms of electrostatic attraction, NH/SH ··· π interaction, and pore size screening of ZIF-8 towards CW. Interestingly, ZIF-8 was discovered for the first time to have a distinct pH-responsive property towards CW enrichment. Based on this property, pH-responsive adsorption was carried out with desorption over ZIF-8, and a stable regeneration process for 5 continuous adsorption/desorption cycles was accomplished. Collectively, this work proved that MOFs have promising application prospects in the field of peptide adsorption.

Abstract:The development of high-performance protein-imprinted materials remains challenging due to defects concerning high mass transfer resistance and non-specific binding, which are crucial for protein purification and enrichment. In this paper, lysozyme-imprinted mesoporous Zr-based MOF (mesoUiO-66-NH 2 @MIPs) with specific and selective recognition of lysozyme (Lyz) were prepared by surface imprinting technology. In particular, the excellent hydrophilicity mesoporous MOFs (mesoUiO-66-NH 2 ) with a pore size of 10 nm was prepared as a carrier for Lyz immobilization by an auxiliary modulation strategy to regulate the microporous structure of UiO-66-NH 2 with the propionic acid solution, enabling massive loading of the macromolecular protein Lyz. The mesoUiO-66-NH 2 @MIPs reached a maximum saturation adsorption of 206.54 mg g −1 on Lyz in 20 min at 25 °C with an imprinting factor of 2.57 and selection factors of 2.02, 2.34, and 2.45 for cytochrome c (Cyt c ), bovine serum albumin (BSA) and bovine hemoglobin (BHb), respectively. More importantly, the mesoUiO-66-NH 2 @MIPs could specifically recognize Lyz from the mixed protein system. The adsorption capacity of Lyz could still reach 78.55% after 5 cycles with good cyclic regeneration performance. This provides a new research option for developing and applying novel porous MOF in biomolecule imprinting technology and the specific separation of biomolecules.

Abstract:Zeolitic imidazole frameworks materials-8 (ZIF-8), a member of the metal-organic framework (MOFs) series, have been extensively used as a host matrix for enzyme immobilization. However, the microporous structure and hydrophobicity, as well as the protonation of the precursor 2-methylimidazole (2-MeIm) of ZIF-8, remain considerable challenges in maintaining the activity of immobilized enzymes. Here, novel multivariate mesoporous MOFs (mMOFs) with regulatable hydrophilic/hydrophobic surfaces were designed by the multivariate competitive strategy and pore modification engineering. 3-Methyl-1H-1,2,4-triazole (3-MTZ) and 5-methyltetrazole (5-MTA) were employed to partially replace 2-MeIm. These were then combined with zinc sulfate heptahydrate (soft templates) to yield mMOFs in a methanol solution. As a proof-of-concept application, we used mMOFs as carriers for enzyme immobilization and investigated the properties of the immobilized enzymes. Benefiting from their mesoporous structure, hydrophilic surface, and improved microenvironment, multivariate mMOFs exhibit a strong ability to stabilize enzyme conformation and increase enzyme activity compared with traditional ZIF-8. Our study offers an avenue for the controllable preparation of well-designed MOF structures, which will further broaden the application opportunities of MOF materials for enzyme immobilization. Abstract:Proteases have been widely used in pharmaceutics and industry, and an economic, convenient and robust procedure for protease immobilization is of high interest. In this study, we report immobilization of proteases by the nanoflower-like zeolitic imidazole framework (nfZIF). Using trypsin, protease K, and Glu-C as model enzymes, we show nfZIF represents an excellent material for protease immobilization, and the resulting protease@nfZIF composites exhibit activities comparable to or even higher than the free proteases in solution. The protease@nfZIF composites are stable in high temperature and organic solvents, and retain almost fully active after several reaction cycles. Our studies demonstrate the great potential of nfZIF in the immobilization of proteases, which could be of interest in various industrial applications.
Pore size regulation of ZIFs for adsorptive separation of branched chain and aromatic amino acids

Abstract:This research endeavour aims to investigate the adsorptive separation of the branched chain (BCAA) and aromatic amino acids (AAA). Based on the different molecular sizes of BCAA and AAA, zeolitic imidazolate frameworks (ZIFs) with different pore structures were prepared by regulating the size of imidazole ligands. The structure and pore shape were characterized by Fourier transform infrared spectroscopy (FT‐IR), x‐ray diffraction (XRD), scanning electron microscope (SEM), and BET surface area (BET). The results indicated the successful synthesis of ZIF‐[Co(mIm)2], ZIF‐[Co(eIm)2], and ZIF‐[Co(pIm)2] with pore size distribution of 7.821, 6.943, and 9.394 Å, and particle size of these ZIFs was approximately 4 μm. ZIF‐[Co(eIm)2] was chosen as the optimal ZIFs for the separation of BCAA and AAA. The adsorption experiment was evaluated in the respective single and binary systems. The corresponding data in a single system demonstrated that ZIF‐[Co(eIm)2] showed good adsorption performance for BCAA and poor for AAA. Furthermore, the adsorption behaviour under experimental conditions conformed well to the pseudo‐second‐order kinetic model and Langmuir isotherm. ZIF‐[Co(eIm)2] had great separation ability for BCAA/AAA in their binary system. Finally, regeneration studies further manifested that ZIF‐[Co(eIm)2] exhibited fine reusability performance with adsorption efficiencies still higher than 70% after five cycles. This study exhibits a novel path for the design of adsorbents for efficient separation of BCAA/AAA.

Abstract:Zeolitic imidazolate frameworks (ZIFs) are hybrid organic-inorganic microporous materials that exhibit zeolite-like structures and can be synthesized with a wide range of pore sizes and chemical functionality. ZIFs as thin films and membranes are of interest for their applications in sensors and gas separation. Here, we report a method for ZIF film and membrane fabrication, based on support surface modification and in situ solvothermal growth, which has potential for general application to other ZIF membranes. Our simple surface modification method results in strong covalent bonds between α-Al(2)O(3) supports and imidazolate ligands, which promote the heterogeneous nucleation and growth of ZIF crystals. The microstructure of ZIF-8 films can be controlled by controlling the pH of the growth solution. ZIF-7 films were fabricated to demonstrate the potential for general applicability of our method. Finally, the separation performance of several ZIF-8 membranes was evaluated, revealing molecular sieving behavior with an ideal selectivity for H(2)/CH(4) of 13.

Abstract:Abstract A new method to engineer hierarchically porous zeolitic imidazolate frameworks (ZIFs) through selective ligand removal (SeLiRe) is presented. This innovative approach involves crafting mixed‐ligand ZIFs (ML‐ZIFs) with varying proportions of 2‐aminobenzimidazole (NH 2 ‐bIm) and 2‐methylimidazole (2‐mIm), followed by controlled thermal treatments. This process creates a dual‐pore system, incorporating both micropores and additional mesopores, suggesting selective cleavage of metal‐ligand coordination bonds. Achieving this delicate balance requires adjustment of heating conditions for each mixed‐ligand ratio, enabling the targeted removal of NH 2 ‐bIm from a variety of ML‐ZIFs while preserving their inherent microporous framework. Furthermore, the distribution of the initial thermolabile ligand plays a pivotal role in determining the resulting mesopore architecture. The efficacy of this methodology is aptly demonstrated through the assessment of hierarchically porous ZIFs for their potential in adsorbing diverse organic dyes in aqueous environments. Particularly striking is the performance of the 10%NH 2 ‐ZIF‐2 h, which showcases an astonishing 40‐fold increase in methylene blue adsorption capacity compared to ZIF‐8, attributed to larger pore volumes that accelerate the diffusion of dye molecules to adsorption sites. This versatile technique opens new avenues for designing micro/mesoporous ZIFs, particularly suited for liquid media scenarios necessitating efficient active site access and optimal diffusion kinetics, such as purification, catalysis, and sensing.

Abstract:The protective capacity and applications of biomimetically mineralized biomacromolecule zeolitic imidazolate framework (ZIF) composites are likely dependent on the localization of the biomolecule and the topology of the mineralized ZIF coating. Herein, we identify reaction conditions to reliably yield the porous ZIF-8 sodalite topology (high ZIF-8 precursor concentrations; high 2-methylimidazole:Zn2+ ratios) in preference to other more dense phases. Furthermore, protocols to universally prepare biocomposites with a range of biomacromolecules are canvassed. Through the use of fluorophore-tagged proteins and confocal laser scanning microscopy (CLSM), we further establish the positioning of biomolecules within ZIF-8 crystals. CLSM reveals subsurface localization with fluorescein-tagged bovine serum albumin (BSA) or full encapsulation with rhodamine B-tagged BSA. These observations allowed us to demonstrate that core-shell ZIF-8 growth strategies afford complete encapsulation with varying thicknesses of potentially active biocomposite or protective ZIF-8. The demonstrated control over ZIF topology (enabling mass transport) and biomacromolecule localization is critical for applications of MOF biocomposites in catalysis.

Abstract:ZIF-8, one kind of representative of metal-organic frameworks, has received much attention in various applications, due to its exceptional properties. Herein we reported the facially- controllable synthesis and post-modification of ZIF-8 nanocrystals with good colloidal stability. The research results demonstrated that the size of ZIF-8 nanocrystals synthesized in water using ZnI2 as zinc source could be controlled by adjusting the molar ratio of ligand (Hmim) to metal ion (Zn2+). Additionally, the zinc salts and solvents played an important role in controlling the size and morphology of ZIF-8 nanocrystals. Furthermore, the ZIF-8 nanocrystals modified with PAA showed good colloidal stability in PBS.

Abstract:In recent years, metal-organic frameworks (MOFs) have emerged as a promising support for immobilizing enzymes due to their high designability and structural diversity. Previous studies show that MOFs with single-crystal-ordered macroporous structures can effectively improve the accessibility of large-size enzyme and reduce the mass transfer resistance compared to conventional MOFs. In order to further enhance the reusability of lipase immobilized on macroporous MOFs, modification of MOFs through some magnetic particles could be an efficient approach. In this work, magnetic macroporous zeolitic imidazolate framework-8 (ZIF-8), referred to as m-M-ZIF-8 (with an average macropore size of about 140 nm), was synthesized and used for the immobilization of Thermomyces lanuginosus lipase (TLL). It was found that enzyme loading and the specific enzyme activity of the immobilized lipase were greatly enhanced through this magnetic modification. The enzyme loading of TLL@C-ZIF-8, T LL@M-ZIF-8, and TLL@m-M-ZIF-8 was 0.060, 0.074, and 0.076 mg/mg respectively. Besides, the activity of 93.5% was maintained after the immobilized lipase being repeatedly used for five batches, which was much higher than that of the immobilized lipase without magnetic modification, which was only 73.4%.
