Abstract:In the classical route of synthesizing of avanafil, the last step of the reaction uses EDCI and HOBT as an acid and amine binding agent. HOBT may contain residues of hydrazine, and potential mutagenic impurities containing acylhydrazide structures may be formed. The intermediate product (M6) of avanafil was synthesized by 6 steps using diethyl ethoxylmethylmalonate and S-methyl isothiourea semisulfide as starting materials. The potentially genotoxic impurity E (Imp-E) of avanafil was given by the reaction of M6 and hydrazine sulfate acid amine, yield of 20%. The single crystal of Imp-E was cultivated by solvent volatilization method, and its structure was analyzed by high resolution mass spectrometry (HRMS) and nuclear magnetic resonance spectroscopy (NMR). The Flack parameter of Imp-E was -0.03(3) by X-ray single crystal diffraction analysis, and the absolute configuration of Imp-E was determined as S configuration. The intramolecular / intermolecular hydrogen bonds of Imp-E make the molecules maintain a stable arrangement in space.

Abstract:Aims: To develop methods with complete validation according to ICH guidelines and to be applied for the determination of avanafil in pure form and in pharmaceutical formulation in the presence of its degradation products. Study Design: High performance thin layer chromatography (HPTLC) and different spectrophotometric methods (dual wavelength, first derivative, first derivative of ratio spectra and ratio difference are developed for simultaneous determination of avanafil in laboratory-prepared mixtures of avanafil with its degradation products and in pharmaceutical formulation. Place and Duration of Study: Sample: Department of Analytical Chemistry, Faculty of Pharmacy (Girls), Al-Azhar University, Cairo, Egypt, between May 2019 and September 2019. Methodology: Two techniques have been developed for the determination of avanafil in the presence of its degradation products. The first was HPTLC where separation was performed on silica gel 60 F254 plates, with chloroform: toluene: methanol: conc. ammonia (6:5:3:0.1, by volume) as a developing system and UV detection at 230 nm. The second one was UV- spectrophotometry which included dual wavelength between 267 and 292 nm, first derivative determination of the drug at 261 nm, first derivative of ratio of peak amplitudes at 275.6, 305.4 and 329 nm and the ratio difference with the amplitude difference between (266 and 250 nm). Results: HPTLC method was applied over the concentration range of 0.5-5. μg/spot, while spectrophotometric methods were linear over the concentration range 5-50 μg/mL for avanafil. Conclusion: Novel, simple and accurate method for the determination of avanafil in laboratory-prepared mixtures of avanafil with its degradation products and in pharmaceutical formulation.

Abstract:Avanafil (AVA), one of the most effective drugs prescribed for erectile dysfunction, is a pyrimidine-derivative PDE5 inhibitor. In the current work, new LC methods were developed and validated for quantitative determination of avanafil and qualitative determination of its degradation products. The quantitative determination of avanafil was carried out using liquid chromatography with photodiode array detection (LC-DAD) and liquid chromatography-tandem mass spectrometry LC-MS/MS methods, and fully validated according to the ICH Q2 (R1) guideline, while qualitative determination was performed using a liquid chromatography mass spectrometry-ion trap-time of flight (LCMS-IT-TOF) instrument. The separation of avanafil and its degradation products was carried out using the same reversed-phase chromatographic conditions, in which a second-generation C18-bonded monolithic silica column (Chromolith® High Resolution RP-18e, 100 × 4.6 mm, Merck KGaA) was used as stationary phase. Briefly, the methods enable quantitation of avanafil with high accuracy (recovery > 95%) and precision (RSD% < 2.0), within the ranges of 0.5–20 μg/mL for LC-DAD and 150–6000 ng/mL for LC-MS/MS. In the forced degradation studies, over and above currently existing data, a new oxidation-based degradation product, whose predicted m/z is 367.1168, was identified and its structure was confirmed by high-resolution mass spectrometric analysis. As the main advantage, either an LC-DAD or LC-MS/MS instrument can be chosen for interference-free quantitation of AVA, according to the facilities in quality-control laboratories.

Analytical Quality by Design Oriented Development of the UPLC Method for Analysing Multiple Pharmaceutical Process Intermediates: A Case Study of Compound Danshen Dripping Pills

Abstract:This work presented the integration of the Analytical Quality by Design concept in developing a UPLC method for the quantification of saponins and phenolic acids in two Compound Danshen Dripping Pills intermediates of a concentrated extract and a supernatant. Prior knowledge and systematic risk analysis were conducted to identify potential critical method parameters (CMPs) that affected critical quality attributes (CQAs). A definitive screening design was employed to investigate the quantitative relationship among the intermediate type, CPPs, and CQAs. Intermediate type was added in the experimental design table as a categorical variable. After the models were built, the method operable design region (MODR) was calculated with an exhaustive search-Monte Carlo method. The CPP combinations with a 95 % or higher probability of reaching the preset standard were collected in MODR. The MODR robustness test was conducted with a Plackett-Burman design. The separation of analytes was achieved with a Waters Acquity HSS T3 (100 x 2.1 mm, 1.8 mu m) column at 30 degrees C, using 0.05 % phosphoric acid and acetonitrile as the mobile phases in gradient elution at a flow rate of 0.40 mL/min. The elution was monitored at UV detection wavelengths of 280 nm and 203 nm. The optimized method was also validated. Good robustness, accuracy, precision, stability and repeatability, and linear regression (R > 0.9997) were obtained. All recoveries of the 9 compounds ranged from 97.8 % to 101.7 %, with an RSD of less than 3.49 %. Nine batches of Danshensu, protocatechualdehyde, rosmarinic acid, salvianolic acid B, salvianolic acid A, notoginsenoside R1, ginsenoside Rg1, ginsenoside Re, and ginsenoside Rb1 were analysed. Danshensu was found to be the compound with the highest content.

Abstract:Elvitegravir (ETV), drug substance, and its eleven process-related impurities have been identified and their structural identification study has been carried out with the aid of 1 H, 13 C NMR, and ESI–LC–MS spectroscopic techniques. Plausible fragments were also proposed for each impurity to ascertain its structure. Simple, facile, and selective, stability indicating, mass spectrometry compatible HPLC method has been developed and subsequently validated with the validation parameters of specificity, LOD, LOQ, precision at LOQ, linearity, accuracy at LOQ to 120% levels, method precision, intermediate precision studies, and solution stability has also been established. This method encompasses a simple gradient mode of separation with mobile phases—(A) 0.1% trifluoroacetic acid in water and (B) 0.1% trifluoroacetic acid in acetonitrile, the mass sectrometric compatible mobile phase has been chosen for the identification of known, unknown and degradation impurities. To assess the nature of each impurity, whether they are either process-related or degradation-induced, an intensive stress study has also been conducted. From this degradation assessment, all the impurities have been classified as process-related. Further, the assessment of three different manufacturers samples was also executed to show the method applicability and comparison of quality of the different manufacturers drug, and thus this method shall be engaged as a quality inferring tool for the marketed sample.

Abstract:Aims: In this study, a simple, green, and rapid capillary zone electrophoresis (CZE) method coupled with a diode array detector (DAD) was applied for the analysis of avanafil (AVA) and dapoxetine hydrochloride (DAP) as a binary mixture using vardenafil (VAR) as an internal standard (IS) in pure form and pharmaceutical formulation. Methodology: The separation was done using fused silica capillary (58.5 cm total length, 50 cm effective length, and 50 μm internal diameter) and the running background electrolyte (BGE) was 100 mM acetate buffer at pH 3.6. During the separation process, the applied voltage was 30 KV, while the temperature was 25 °C. The sample injection was applied at a pressure of 50 mbar for 10 s, and detection was carried out at 210 nm for DAP and 248 nm for AVA and VAR. Results: Analysis of the tested drugs and the internal standard was carried out in less than 6.5 min, where the migration times were 4.29, 4.90, and 6.02 min for IS, DAP and AVA respectively. The proposed method showed linearity in the concentration range 5-80 and 5-70 μg/mL with correlation coefficients 0.9996 and 0.9999 for AVA and DAP respectively. The limit of detection (LOD) was 0.523 and 0.531 for AVA and DAP respectively, while the limit of quantification (LOQ) was 1.585 and 1.608 in respective order. The Peak purity and identity in the proposed method were validated by DAD. Conclusion: The proposed CZE method was validated according to ICH guidelines and applied successfully for the estimation of AVA and DAP in their combined pharmaceutical preparation.
Development of a validated HPLC method for the quantitative determination of trelagliptin succinate and its related substances in pharmaceutical dosage forms

Abstract:Trelagliptin succinate is a dipeptidyl peptidase IV (DPP-4) inhibitor which is used as a new long-acting drug for once-weekly treatment of type 2 diabetes mellitus (DM). In the present study, a rapid, sensitive and accurate high-performance liquid chromatography (HPLC) method was developed and validated for separation and determination of trelagliptin succinate and its eight potential process-related impurities. The chromatographic separation was achieved on a Waters Xselect CSH™ C18 (250mm×4.6mm, 5.0μm) column. The mobile phases comprised of 0.05% trifluoroacetic acid in water as well as acetonitrile containing 0.05% trifluoroacetic acid. The compounds of interest were monitored at 224nm and 275nm. The stability-indicating capability of this method was evaluated by performing stress test studies. Trelagliptin succinate was found to degrade significantly in acid, base, oxidative and thermal stress conditions and only stable in photolytic degradation condition. The degradation products were well resolved from the main peak and its impurities. In addition, the major degradation impurities formed under acid, base, oxidative and thermal stress conditions were characterized by ultra-high-performance liquid chromatography coupled with linear ion trap-Orbitrap tandem mass spectrometry (UHPLC-LTQ-Orbitrap). The method was validated to fulfill International Conference on Harmonisation (ICH) requirements and this validation included specificity, linearity, limit of detection (LOD), limit of quantification (LOQ), accuracy, precision and robustness. The developed method in this study could be applied for routine quality control analysis of trelagliptin succinate tablets, since there is no official monograph.
Simultaneous Analysis of Indapamide and Related Impurities in Sustained-Release Tablets by a Single-Run HPLC-PDA Method

Abstract:The contents of indapamide and related impurities in generic indapamide sustained-release tablets were simultaneously detected by a single-run high performance liquid chromatography equipped with photodiode array detector (HPLC-PDA) method for the quality control in this paper. The results showed the method had a good selectivity and was validated through linearity, limits of detection and quantification, recovery, and precision. The linear ranges of indapamide, 2-methyl-1-nitroso-2,3-dihydro-1H-indole (impurity A, ImA), 4-chloro-N-(2-methyl-1H-indol-1-yl)-3-sulphamoyl-benzamide (impurity B, ImB) and 4-chloro-3-sulfamoylbenzoic acid (impurity 1, Im1) were 0.028-1.80 μg/mL ( R =0.999 95), 0.060-1.20 μg/mL ( R =0.999 6), 0.032 4-1.20 μg/mL ( R =0.999 85) and 0.060-1.20 μg/mL ( R =0.999 7) with detection limits of 0.009 3, 0.012, 0.012 and 0.006 μg/mL, respectively. ImA and Im1 were not detectable in the generic drug. The content of indapamide was 96.7% of the labeled amount with a relative standard deviation (RSD) of 1.30%, and the percentage of ImB relative to the labeled amounts of indapamide was 0.106% with an RSD of 1.82%. The content of other unspecified impurities all met the reference quality standards. The results provided references for the quality control and the quality standard study of generic indapamide sustained-release tablets.
Development of an analytical method for the determination of pimavanserin and its impurities applying analytical quality by design principles as a risk-based strategy

Abstract:Pimavanserin is an atypical antipsychotic indicated for the treatment of hallucinations and delusions associated with Parkinson's disease psychosis. As it is a relatively new drug on the market, limited number of pharmacokinetic information and analytical methods are available. This paper presents an ultra-high performance chromatography for the simultaneous determination of pimavanserin and its four process impurities. The method was developed applying analytical quality by design (AQbD) principles as a risk-based approach. Critical method attributes (CMAs) were selected as a resolution between the worst separated compounds (impurity B and impurity C), a duration of analysis defined by the retention time of the last eluting peak (impurity D), a capacity factor of the first eluted impurity (impurity A), a tailing factor and a theoretical plate number. Risk assessment in the early stage of method development pointed out critical method parameters (CMPs): column temperature, gradient time and pH-value of the mobile phase (water phase, eluent A). Design of experiments (DoE), using DryLab®4 software, was applied to evaluate the influence of CMPs on CMAs and to determine method operable design region (MODR). Based on the risk assessment and the results of robustness and precision tests, a control strategy with system suitability criteria was proposed. Developed method was validated according to ICH Q2 (R1) guideline with respect to the selectivity, LOD, LOQ, linearity, precision, accuracy, robustness and stability. A forced degradation study was performed to provide an evidence of the stability-indicating property of the method. Degradation products of pimavanserin were identified using ultra high-performance liquid chromatography coupled to high resolution mass spectrometry (UHPLC-qTOF). Additionally, potential degradation products were assessed in silico with the help of Zeneth® software and good agreement with experimentally identified degradation products was achieved. Main degradation products were formed during acid and base hydrolysis (m/z 223.16 [M+H]+ at RRT 0.37) and under oxidative stress conditions (m/z 444.26 [M+H]+ at RRT 0.57). The results revealed that the pimavanserin undergoes degradation through acid and base hydrolysis of urea and N-oxidation of aliphatic tertiary amine.
Analysis of Organic Impurities of Besifloxacin Hydrochloride by High-Performance Liquid Chromatography with Isocratic and Gradient Elution

Abstract:Introduction: Impurity analysis is an important step in the quality control of pharmaceutical ingredients and the final product. Impurities can arise from drug synthesis or excipients and even at small concentrations may affect product efficacy and safety. In this work, two methods using highperformance liquid chromatography (HPLC) were developed and validated for the evaluation of besifloxacin and its impurity synthesis, with isocratic elution and another with gradient elution. Method: The analysis by HPLC in isocratic elution mode was performed using a cyano column maintained at 25 °C. The mobile phase was composed of 0.5% triethylamine (pH 3.0): acetonitrile (88:12 v/v) eluted at a flow rate of 1.0 mL/min with detection at 330 nm. The gradient elution method was carried out with the same column and mobile phase components only modifying the rate between organic and aqueous phase during analysis. The procedures have been validated according to internationally accepted guidelines, observing results within acceptable limits. Results: The methods presented were found to be linear in the 140 to 260 μg/ mL range for besifloxacin and 0.3 to 2.3 μg/ mL for an impurity named A. The limits of detection and quantification were, respectively, 0.07 and 0.3 μg/ mL for impurity A, with a 20 μL injection volume. The precision achieved for all analyses performed provided RSD inter-day equal to 6.47 and 6.36% for impurity A with isocratic elution and gradient, respectively. The accuracy was higher than 99% and robustness exhibited satisfactory results. In the isocratic method, an analysis time of 25 min and 15 min was obtained for the gradient. For impurity A, the number of theoretical plates in the isocratic mode was about 5000 while in the gradient mode it was about 45000, hence, it made the column more efficient by changing the mobile phase composition during elution. In besifloxacin raw material and in the pharmaceutical product used in this study, other related impurities were present but impurity A was searched for and not detected. Conclusion: The proposed methods can be applied for the quantitative determination of impurities in the analysis of the besifloxacin raw material, as well as in ophthalmic suspension of the drug, considering the quantitation limit.

Impurity profiling of varenicline tartrate by LC-QTOF mass spectrometric techniques during drug development

Abstract:An LC-QTOF-MS method was developed for the separation and characterization of related substances in varenicline tartrate. The separation was established on an InertSustain C18 column (4.6 mm × 150 mm, 5 μm) by liner gradient elution using 0.05% trifluoroacetic acid as mobile phase A and acetonitrile as mobile phase B. The degradation studies were conducted under the ICH prescribed stress conditions. Varenicline tartrate was found to be unstable to alkaline, oxidative, thermal and photolytic stresses, while relatively stable under acid stress condition. Thirteen related substances were detected all together in varenicline tartrate and its stressed samples. Their structures were identified mainly through positive ESI high resolution QTOF mass spectrometric analysis of the parent and product ions' accurate masses and the calculated elemental compositions. Among the 13 related substances, seven were process-related and six were degradation products, and two of them were further verified by chemical synthesis and NMR spectroscopic determination. Their formation mechanisms were also discussed, and the key steps in the manufacturing processes were also determined to provide varenicline tartrate with high purity.
A validated green UPLC method for the quantification of synthetic pharmaceutical adulterants in the extract of the aphrodisiac herbal plant ashwagandha (Withania somnifera) and in spiked human plasma

Abstract:In this work, an ultra-performance liquid chromatographic method was developed and subjected to bioanalytical method validation for screening the root extract of Ashwagandha. The method was also extended for simultaneous determination of Sildenafil, Vardenafil and Avanafil in plant root extract and in human plasma samples to detect the possible counterfeiting of the plant's pharmaceutical products with other synthetic drugs that might endanger the life of their consumers, particularly elderly people, and those with chronic diseases. The method was developed using BDS Hypersil TM C18 column (250 × 4.6 mm and 3 μm particle size) as a stationary phase and a mobile phase consisting of acidified water and acetonitrile in a gradient elution program with a flow rate of 1.00 mL min 1. In the plant extract, the proposed method was found to be linear in the range 1.00–200.0 μg mL−1 for the Vardenafil and Avanafil and 5.00–200.0 μg mL−1 for Sildenafil with a recovery percentage in range of 98.18–101.07 % for the three drugs. Intra-day and inter-day precision ranges were 0.088–1.219 and 0.151–1.313, respectively. The limit of detection and the limit of quantification for Vardenafil and Avanafil were 0.166 μg mL−1 and 0.500 μg mL−1, respectively. Whereas, for Sildenafil 0.333 μg mL−1 and 1.000 μg mL−1. The ICH validation parameters were also demonstrated. Bioanalytical method validation was established following US-FDA recommendations and the method showed good linearity in the range 1.00–200.00 μg mL −1 for Vardenafil and Avanafil, and 5.00–200.00 μg mL −1 for Sildenafil. The results revealed high accuracy with % recovery ranging from 91.05 to 98.91 % for the three drugs. The intra-day and inter-day precision were also conducted, in terms of % relative standard deviation, and ranged from 0.119 to 4.037 % and from 0.223 to 5.062 %, respectively. Furthermore, other validation parameters were evaluated including extraction recovery and stability of the analytes throughout samples preparation and storage for both short- and long-term. The method greenness was evaluated using NEMI, GAPI, AGREE and analytical eco-scale.

Abstract:Two trace impurities in the bulk drug lisinopril were detected by means of high-performance liquid chromatography coupled with mass spectrometry (HPLC/MS) with a simple and sensitive method suitable for HPLC/MSn analysis. The fragmentation behavior of lisinopril and the impurities was investigated, and two unknown impurities were elucidated as 2-(6-amino-1-(1-carboxyethylamino)-1-oxohexan-2-ylamino)-4-phenylbutanoic acid and 6-amino-2-(1-carboxy-3-phenylpropylamino)-hexanoic acid on the basis of the multi-stage mass spectrometry and exact mass evidence. Abstract:A rapid and selective LC-MS/MS method is described for the simultaneous assay of Avanafil and Dapoxetine in human plasma via a protein precipitation (PP) sample preparation technique. Tadalafil was chosen as the internal standard reaching good recovery and reproducibility while diminishing the effects of the matrix. An Agilent Zorbax Eclipse XDB C18 column (4.6 × 50 mm, 1.8 μm) was used for the chromatographic separation and analysis, while 0.1% formic acid : acetonitrile (60 : 40, v/v) was utilized at a flow rate of 0.5 mL min-1. It was revealed that 6 min stop time accomplished the best separation. The assay was linear over the range of 10-6000 ng mL-1 for both drugs. The established bio-analytical method validation was demonstrated following US-FDA recommendations including sensitivity, selectivity, linearity, accuracy and precision. Furthermore, other validation parameters were assessed such as the dilution integrity, matrix effect, carryover, and analyte stability during both short- and long-term sample processing and storage. The adopted method was efficaciously applied to a clinical study for the concurrent determination of Avanafil and Dapoxetine in human plasma.

Abstract:Objective:To establish a quick,accurate,sensitive method for analysis of sildenafil citrate illegally added into traditional chinese medicinal preparation for invigorant by UPLC-MS-MS.Methods:The UPLC-MS-MS method was used.Electrospay ionization(ESI) source was applied and operated in both positive and negative ion modes.The qualitative and quantitative method of sildenafil citrate illegally added into traditional chinese medicinal preparations for invigorant were carried out.Results:Sildenafil citrate was found in three formulations of twenty-one.Conclusion:The method is selective and sensitive and can be used to detect the sildenafil citrate illegally added into traditional chinese medicinal preparations for invigorant.

In order to get the references for the process optimization and quality control of API, the eight compounds were synthesized. According to the possible formation pathways, a modified synthesis and the strategy for impurity control in the corresponding reactions were established.

Abstract:A high performance liquid chromatography-diode array detection-electrospray ionization tandem mass spectrometry method was developed for analysis and identification of impurities in lovastatin. Lovastatin and impurities were separated on a Diamonsil C-18 column (5 mu m,4. 6 mm x 250 mm) using acetonitrile-water (containing 0.1% acetic acid) (65:35) as mobile phase. The elutes were detected with diode array detector and ion trap mass spectrometer in positive mode. The ultraviolet (UV), MS and MS' spectra of each chromatographic peak were obtained. The UV spectra of the impurities are similar with those of lovastatin except for dihydrolovastatin. It indicated that they have the same basic structures. Then the [M+H](+) ions and their MS2 spectra were used for determining the molecule mass and side-chain. According to analyses of the UV, MS and MS' as well as the comparison with the published literature, the structures of ten impurities were identified. The results indicated that the described method could be effectively applied to perform identification of impurities in lovastatin.

Abstract:Five trace impurities were detected in the bulk drug citalopram using high-performance liquid chromatography with UV detection. A simple and sensitive method suitable for liquid chromatography tandem multistage mass spectrometry (HPLC/MS(n)) analysis was developed. Using this method, the fragmentation behavior of citalopram and the impurities was investigated. Four impurities were rapidly characterized, and an unknown impurity was elucidated as 3-(3-dimethylaminopropyl)-N-(1-(3-dimethylaminopropyl)-1-(4-fluorophenyl)-1,3-dihydroisobenzofuran-5-yl)-3-(4-fluorophenyl)-1,3-dihydroisobenzofuran-5-carboxamide on the basis of the MS(n) and exact mass evidence, and the proposed structure was further confirmed by nuclear magnetic resonance (NMR) experiments after preparative isolation.
