Abstract:OBJECTIVE:To develop an analytical method and quality control for determination of zolmitriptan and related substances.METHODS:Zolmitriptan and related substances were separated and determined on a shimadzu CLC-C(8) column (150 mm x 6 mm, 10 microm) with a mobile phase of acetonitrile-10 mmol/L phosphate buffer (25:75 pH 7.5) and a flow-rate of 1 ml/min; the UV-VIS detector was operated at 229 nm.RESULT:The limit of detection for the related substances was 0.5 ng on the zolmitriptan basis (S/N >3). Linear calibration curve was gene rated from 4 - 40 microg/ml with a correlation coefficient of 0.9999. The recovery rate of zolmitriptan was 99.1% with a standard deviation of 0.2%. The results of HPLC method were consistent with those of nonaqueous titration method.CONCLUSION:HPLC method is a rapid sensitive and accurate method for the determination of zolmitriptan and its related substances.
A Stability Indicating RP-HPLC Method Validation for Simultaneous Estimation of Metformin HCl, Dapagliflozin and Saxagliptin in Pharmaceutical Dosage Form

Abstract:Aims: Metformin HCl, Dapagliflozin and Saxagliptin is a new drug combination for the treatment of Diabetes Mellitus which is one of the oldest and lethal diseases of mankind. Aim of the research work was to develop and validate novel, rapid, sensitive, specific, robust stability indicating analytical method for simultaneous estimation of: Metformin HCl, Dapagliflozin and Saxagliptin in pharmaceutical dosage form as fixed dose formulation. Study Design: Method development and validation was performed as recommended in ICH guideline “Validation of analytical procedures: Test and Methodology Q2 (R1)”. Methodology: Method develop with chromatographic parameters as C18 column (250mm×4.6 mm, 5mm particle size), HPLC system with PDA detector and mobile phase contained a mixture of Phosphate Buffer pH 3.5 and Acetonitrile (80:20 v/v) + 1 ml triethylamine per 100 ml mobile phase. The flow rate was set to 1 ml/min with responses measured at 265 nm, injection volume was 20 μl, and run time of 15 mins. Results: The retention time of Metformin HCl, Dapagliflozin and Saxagliptin was 5.8 min, 6.8 mins and 8.4 min respectively with resolution of 3.5 between Metformin HCl and Dapagliflozin and 4.5 between Dapagliflozin and Saxagliptin. Linearity was established in the range of 250-1500 μg/ml for Metformin HCl, 1.25-7.5 μg/ml for Dapagliflozin and Saxagliptin with correlation coefficients more than 0.999. The percentage recoveries were between 98.39-101.66 for Metformin HCl, 99.01-101.77 for Dapagliflozin and 98.88-101.87 for Saxagliptin Validation parameters were evaluated according to the International Conference on Harmonization (ICH) Q2 R1 guidelines. The forced degradation studies were performed by using HCl, NaOH, H2O2, thermal and UV Radiation. The developed method was successfully applied for the quantification and hyphenated instrumental analysis. Conclusion: Significance of developed method is that it can be utilize for routine or unknown sample analysis of assay of Metformin HCl, Dapagliflozin and Saxagliptin in pharmaceutical dosage form developed by various Pharmaceutical Industry.
Stability-Indicating TLC-Densitometric and HPLC Methods for Simultaneous Determination of Teneligliptin and Pioglitazone in Pharmaceutical Dosage Forms with Eco-Friendly Assessment

Abstract:The combination of teneligliptin hydrobromide hydrate and pioglitazone hydrochloride in pharmaceutical formulations has improved type 2 diabetes management. Two chromatographic methods TLC-densitometry and RP-HPLC were developed for simultaneous quantification of teneligliptin hydrobromide hydrate and pioglitazone hydrochloride in pharmaceutical formulations, ensuring accuracy and stability assessment. The TLC method uses a mobile phase of methanol, toluene, ethyl acetate and triethylamine (1:7:2:0.1, v/v/v/v) on TLC silica gel plates, scanned at 268 nm. The RP-HPLC method employs isocratic elution with acetonitrile and sodium acetate buffer (adjust pH 3.6 with glacial acetic acid, 60:40 v/v) on a shimpack C18 column (250 × 4.6 mm i.d., 5 μm), detected at 235 nm. Both methods offer high accuracy and reliability, making them valuable for pharmaceutical quality control. Additionally, an environmental impact assessment was conducted using eco-scale, Analytical Greenness Metric Approach, Green Analytical Procedure Index, and national environmental method index to evaluate solvent consumption, waste generation and energy usage. Statistical comparisons (t-tests and F-tests) validate the outcomes of both methods, ensuring their effectiveness in drug formulation analysis. These methods can enhance pharmaceutical quality control while fulfilling environmental responsibilities.
A High Performance Liquid Chromatography Method for the Quantitative Determination Assay of Sitagliptin in Rat Plasma and Its Application in Pharmacokinetics Study

Abstract:A new high-performance liquid chromatography(HPLC)method for the quantitative determination of sitagliptin in rat plasma was developed and validated for pharmacokinetics study.The plasma was spiked with the internal standard(hydrocortisone,IS),treated with sodium hydroxide,and extracted with ethyl acetate.The extracted analyte was injected into an Agilent Zorbax Extend-C18 column(250 mm×4.6 mm,4 μm)maintained at 30 ℃ and monitored at 267 nm.The mobile phase consisting of methanol-water(60:40,v/v,containing 10 mM Tris and 10 mM triethylamine)was titrated to pH 9.0 using 1 mol/L hydrochloric acid.The flow rate was 1.0 mL/min.The method showed high specificity.Calibration curves of the peak area ratio of each analyte/IS versus sitagliptin concentration were linear in the range of 0.75-100.0 μg/mL(r>0.9957).The lower limit of quantification(LLOQ)was 0.75 μg/mL.The intra-day and inter-day coefficient of variation was lower than 10%.The accuracy(relative recovery)at three levels was 105.3%(0.75 μg/mL),99.8%(10.0 μg/mL)and 99.0%(100.0 μg/mL).The extraction recovery was 81.5%,82.4% and 84.5% at the concentrations of 0.75,10.0 and 100.0 μg/mL,respectively.The short-term,long-term,freeze-thaw storage stability of plasma samples and the stability of standard solutions were satisfactory.This HPLC method is suitable for determining the concentration of sitagliptin in rat plasma and it was applied to determine the concentration-time profiles of sitagliptin in rat plasma following oral administration of sitagliptin.

Abstract:Background: Sitagliptin phosphate monohydrate-dapagliflozin propanediol hydrate fixeddose combination (FDC) dual-layered tablet is used for type 2 diabetes treatment. Simultaneous quantitative analysis can shorten the analysis time of sitagliptin phosphate monohydrate-dapagliflozin propanediol monohydrate FDC dual-layered tablets and increase their efficiency. Objective: This study aimed to develop the simultaneous quantitative analysis for sitagliptin phosphate monohydrate-dapagliflozin propanediol monohydrate FDC dual-layered tablet, a type 2 diabetes treatment. Method: Simultaneous quantitative analysis using the rapid and selective reversed-phase highperformance liquid chromatography (RP-HPLC) method was developed and validated using method validation. RP-HPLC analysis was conducted using an ultraviolent absorption spectrophotometer and a Zorbax C18 column (4.6 x 150 mm, 5 μm). The flow rate and injection volume were set to 1.5 mL min-1 and 20 μL, respectively. The wavelength was set at 205 nm. Results: The retention times of sitagliptin phosphate monohydrate and dapagliflozin propanediol monohydrate were 2.28 mins and 10.65 mins, respectively. The relative standard deviations of the system suitability for validation of simultaneous quantitative analysis were 0.03% for sitagliptin phosphate monohydrate and dapagliflozin propanediol monohydrate. The chromatogram confirmed that there was no peak interference between the two main components and between the main component and the excipients. In addition, It revealed a favorable linearity with correlation coefficients of 0.9999 in the concentration range of 20–120% compared to the standard solution. Conclusion: The developed simultaneous quantitative analysis shortened the analysis time and high efficiency of the sitagliptin phosphate monohydrate-dapagliflozin propanediol monohydrate FDC bilayer tablet. The validity of the analytical method was verified through accuracy and precision, detection and quantitation limits, and solution stability tests. In addition, it was thought that it would be helpful in developing an analytical method by referring to the simultaneous quantitative analysis method for developing other FDC dual-layered tablets.

Abstract:A simple, rapid, and precise high-performance thin-layer chromatographic (HPTLC) method for simultaneous estimation of two antidiabetic drugs, metformin hydrochloride and sitagliptin phosphate, in tablet dosage form has been developed and validated. Chromatography was performed on silica gel 60 F254 plates with butanol : water : glacial acetic acid (6 : 2 : 2, v/v/v) as mobile phase. This system gave a good resolution for metformin hydrochloride ( R f value of 0.35 ± 0.01) and sitagliptin phosphate ( R f value of 0.75 ± 0.01). Detection and quantification were carried out at 227 nm. The linear regression data for the calibration plot showed a good relationship with r = 0.9995 and 0.9991 for metformin hydrochloride and sitagliptin phosphate, respectively. The method was validated for precision and recovery. The limits of detection and quantification were 13.05 and 39.56 ng/μL for metformin hydrochloride and 2.65 and 8.03 ng/μL for sitagliptin phosphate, respectively. The amounts of the drugs in the marketed formulation were 99.86% and 98.91% for metformin hydrochloride and sitagliptin phosphate, respectively.

Abstract:This study aimed to develop and validate a stability-indicating liquid chromatography method for the determination of tirofiban hydrochloride and two synthetic impurities (impurity A and impurity C). The method utilizes a RP-18 column (250 mm × 4.6 mm; 5 μm) with the PDA detector for quantitation. A mixture of triethylamine 0.1% (acidified to pH 5.5 with phosphoric acid) and acetonitrile was used as the mobile phase at a flow rate of 1 mL min−1 with gradient elution. The method presented satisfactory linearity, precision, accuracy and robustness, as well as low limits of detection and quantification, which demonstrate sensitivity in the determination of tirofiban and impurities A and C. It was selective for the determination of the drug and impurities analysed, without interference of the degradation products generated under forced conditions, demonstrating the stability-indicating capacity of the proposed method. Tirofiban showed to be practically stable to oxidative (30% H2O2 for 24 h) and thermal (75 oC for 24 h) conditions, but presented degradation to UVA light and acid hydrolysis, obeying the first order kinetics for both. In this way, it can be used as a stability-indicating method in the quality control of the raw material of tirofiban hydrochloride, as well as of the finished product. The obtained results demonstrate the importance of deepening the studies in this area, in order to guarantee the quality of commercialized pharmaceutical products.
Estimationof Sitagliptin Phosphate monohydrate in presenceof itsdegradantsusingRP-HPLC

Abstract:A simple, economic, selective, precise, and stability-indicating HPLC method has been developed and validated for estimation of Sitagliptin both in bulk drug and tablet dosage form. The drug was separated using a mobile phase acetonitrile: water, (5:95 v/v) on an Agilent, TC C18 (250 × 4.6 mm) 5µm column at flow rate of 1.0 mL min-1 at ambient temperature and detection was performed at 263 nm. The detector linearity was established in concentrations ranging from 10–80 µg mL−1, the regression coefficient was 0.9996. For stability study, the drug was exposed to the stress conditions such as acid, base, oxidation, neutral and sunlight as per the recommendations of ICH guidelines. The results of the analysis were validated in terms of specificity, limit of detection, limit of quantification, linearity, precision and accuracy. As per ICH guidelines results were found to be satisfactory. The method was proved to be robust with respect to changes in flow rate and temperature. The high recovery and low relative standard deviation confirm the suitability of these methods can be employed for the routine analysis of tablet containing Sitagliptin.

Abstract:Introduction: Trimetazidine and Metoprolol combination is more effective in the treatment of cardiac disorders as compared to single drug therapy.Background: Materials and Methods: A rapid, simple, and sensitive HPTLC method was developed for the simultaneous determination of Trimetazidine and metoprolol from its tablet dosage form and validated. In HPTLC method, standard and sample solutions of Trimetazidine hydrochloride and metoprolol succinate were applied on pre-coated silica gel G 60 F254 TLC plate, and developed by using mobile phase, n-butanol :water: methanol: ammonia as solvent (8.5:0.1:0.1: 0.85, v/v). The drugs on plate were scanned at 213 nm. The method produced compact and well-resolved bands at Rf of 0.32 ± 0.02 and 0.66 ± 0.02 for Trimetazidine Hydrochloride and Metoprolol succinate respectively. The range for linearity was observed as 500-2500 ng band-1 for Trimetazidine hydrochloride and 500-2500 ng band-1 for metoprolol succinate and correlation coefficient were 0.9991 and 0.9997 respectively. Conclusion: The developed method was validated according to the ICH guidelines for precision, accuracy, Limit of detection, Limit of quantitation, specificity and robustness. The method was checked for suitability in determination of Trimetazidine hydrochloride and Metoprolol succinate in their tablet dosage form. The assay result was found to be 99.64 % ± 0.45 and 99.94 % ± 0.53 of percentage label claim for Trimetazidine hydrochloride and Metoprolol succinate respectively.

Novel HPLC Method Validation for Quantification of Dapagliflozin in Bulk Drug and Formulation

Abstract:A new, steady, easy-to-use, and straightforward high-performance liquid chromatography (HPCL) technique was also tested to determine dapagliflozin concentration in medicinal and API forms. For the chromatographic extraction study, a Phenomenex Gemini-NX C18 Column was used. This column was filled with a mobile phase that was 70:30 v/v methanol to sodium 1-octanesulphonate. This experiment used a 20-liter syringe amount and a flow rate of 1-mL per minute. A bunch of factors were used to make sure the new method worked. The drug dapagliflozin was released at a wavelength of 203 nm after 6.5 ± 0.5 minutes. A correlation coefficient (r2) of 0.9994 between 15 to 55 μg/mL shows that the planned process is linear. The theoretical plates and peak tailing were found to be 4129 and 1.03, respectively. In terms of healing, the percent accuracy that was reached was within the acceptable range of 98 to 100. New suggestions say that the regular testing of dapagliflozin in large amounts must be done using a proven method that is unique and easy to repeat. Forced breakdown tests with drugs were used to show that the proposed method is stable. The product was tested according to the steps laid out in the ICH Q2R1 Guidelines. The test looked at many chemical processes, such as oxidation, photolysis, heat stress, and acids and alkaline hydrolysis.

Abstract:A simple, specific and accurate reverse phase high performance liquid chromatographic method was developed for the simultaneous determination Sitagliptinand Etruglifloxinin pharmaceutical dosage form. The column used was Discovery C18(250mm x 4.6 mm, 5m)in isocratic mode, with mobile phase containing phosphatebufferandacetonitrile (45:55v/v). The buffer is prepared by adding accurately weighed 1.36gm of PotassiumdihyrogenOrtho phosphate in a 1000ml of Volumetric flask add about 900ml of milli-Q water added and degas to sonicate and finally make up the volume with water then pH adjusted to 5.4with dil. Orthophosphoric acid solution.The flow rate was 1.0ml/ min and effluents were monitored at 260nm. The retention times of Sitagliptinand Etruglifloxinwere found to be 2.381min and 3.429min, respectively. The linearity for Sitagliptinand Etruglifloxinwere in the range of 25-150μg/mland 3.75-22.5μg/ml respectively. The recoveries of Sitagliptinand Etruglifloxinwere found to be 99.46to 101.19% and 99.36to 100.99%, respectively. The proposed method was validated and successfully applied to the estimation of Sitagliptinand Etruglifloxinin combined tablet dosage forms.

Abstract:Abstract Background Nateglinide and metformin HCl are used in combination for the treatment of type 2 diabetes. A simple, sensitive and reliable UPLC method was developed for simultaneous estimation of nateglinide and metformin HCl using Phenomenox C 18 (50*2.1 mm, 3.5 μm) column at ambient temperature as stationary phase in addition to mobile phase containing 75 volumes of ammonium formate buffer (pH = 3) along with 25 volumes of acetonitrile with a flow rate of 0.2 mL/min with UV detection at 260 nm and a run time of 3 min. The developed method was validated as per ICH Q2(R1) guidelines. Results The separation of metformin HCl and nateglinide was done at retention times of 1.014 min and 1.435 min, respectively. The mean % recovery for nateglinide and metformin HCl in the accuracy study was observed to be 99.9% and 99.2%, respectively. LOD and LOQ values were determined considering the S/N ratio and were found to be 0.09 μg/mL and 0.3 μg/mL, respectively, for nateglinide and 0.75 μg/mL and 2.5 μg/mL, respectively, for metformin HCl. The method was found to be precise with % RSD values of 0.58 and 0.45, respectively, for repeatability and intermediate precision of nateglinide and 0.43 and 0.43, respectively, for repeatability and intermediate precision of metformin HCl which were within acceptance criteria. The method was found to be linear in the range of 7.5–45 μg/mL and 62.5–375 μg/mL for nateglinide and metformin HCl, respectively. The regression equations for nateglinide and metformin HCl were found to be y = 17377 x + 6543.4 and y = 18439 x + 43,537, respectively. The method was found to be robust by deliberate changes in the method parameters like flow rate and mobile phase composition. Forced degradation studies were performed as per ICH Q1A(R2) and Q1B guidelines, and peak purity was observed in all types of degradation studies for both the drugs. Conclusion The developed method was found to be satisfactory as it is simple, sensitive, accurate, precise, robust, rapid, economical and yet stability indicating and can be applied successfully in the routine laboratory analysis for the simultaneous estimation of nateglinide and metformin HCl in the bulk and pharmaceutical dosage forms.

Abstract:A simple, precise, and accurate high-performance thin-layer chromatography (HPTLC) method was developed and authenticated to perform simultaneous estimation of dapagliflozin propanediol monohydrate (DAPA), vildagliptin (VILDA), and metformin (MET) in active pharmaceutical ingredient (API) and tablet dosage form. It was found that there is not any reported stability-indicating HPTLC method for the simultaneous determination of DAPA, VILDA and MET; hence, this method was developed. Merck HPTLC silica gel 60 F 254 plates were selected as the stationary phase. Acetonitrile, formic acid, and water (9:1:0.5, V/V ) were the chromatographic solvents used. A densitometric study was conducted at the wavelength of 215 nm. For DAPA, VILDA, and MET, the concentrations used to conduct the study were 0.2‒0.6 μg/band, 2‒6 μg/band, and 10‒30 μg/band, respectively. The peaks of DAPA, VILDA, and MET have retardation factors ( R F ) of 0.70, 0.45, and 0.34, respectively. The regression coefficients ( R 2 ) of DAPA, VILDA, and MET were 0.9955, 0.9958, and 0.9972, respectively. The limit of detection values of DAPA, VILDA, and MET were 0.0018 μg/band, 0.0094 μg/band, and 0.1434 μg/band. The limit of quantification of DAPA, VILDA, and MET were 0.0056 μg/band, 0.0285 μg/band, and 0.4346 μg/band, respectively. With respect to International Council for Harmonisation (ICH) prerequisites, the accuracy, precision, and robustness outcomes were within suitable limits. The outcomes of stability data revealed that the degradation by acid, base, hydrogen peroxide, and water produced distinct peaks of DAPA, VILDA, and MET, along with a few other peaks, having different R F values. The forced degradation study for DAPA, VILDA, and MET was performed. DAPA was found to be stable in acidic, oxidative, and neutral conditions. Under acidic, basic, and oxidative conditions, VILDA was observed to be stable. The proposed technique passes all the ICH criteria; hence, it is an accurate, easy, suitable, and stable method for the analysis of DAPA, VILDA, and MET in combined dosage form.

Abstract:Abstract The high sensitive HPLC-ESI/MS method for quantitative determination of a new antifungal drug – 2-[(1Z)-1-(3,5-diphenyl-1,3,4-thiadiazol-2(3Н)-ylidene)methyl]-3,5-diphenyl-1,3,4-thiadiazol-3-ium chloride (TDZ) – was developed and fully validated. TDZ was separated from plasma and urine samples by acetonitrile deproteinization and extraction without time-consuming sample preparation. The reversed-phase high-performance liquid chromatography on Kromasil 100–3.5 C8 column of TDZ in isocratic elution mode using 0.03% trifluoroacetic acid : acetonitrile (65:35, v/v) at a flow rate of 0.2 mL min−1 was performed. Determination of TDZ was carried out by a positive electrospray ionization in a selected ion monitoring mode for [M+]=489 m/z. The method of absolute calibration was used for quantification of TDZ in two concentrations ranges: 100–2500 pg mL−1 and 2500–30 000 pg mL−1. The established method showed a good linearity (R=0.999 for both ranges), the limits of determination and quantification were 50 and 100 pg mL−1, respectively. The Intra- and Inter-day precision values were measured by t-Distribution and Fisher′s Exact Test and were in accordance with the regulatory guidance. Low matrix effects and good recovery were found for TDZ. The present method was successfully applied to determine the pharmacokinetic parameters of TDZ by means of intravenous and oral administrations to rats at 5.0 mg kg−1 and 10.0 mg kg−1, respectively.

Abstract:Background: In June 2021, the Central Drug Standards Control Organization approved a fixed-dose combination tablet containing remogliflozin etabonate (100 mg) and teneligliptin (10 mg) to manage type II diabetes. Objective: This study aims to develop a stability-indicating RP-HPLC method for quantifying remogliflozin etabonate and teneligliptin in tablet formulations via analytical quality by design (AQbD) principles. Methods: Risk assessment, Plackett-Burman design, and central composite design were employed to understand the impact of independent variables on critical analytical attributes. The stationary phase was a HyperClone BDS C18 column, and the mobile phase consisted of acetonitrile and phosphate buffer (20 mM, pH 5) at a 45:55% (v/v) ratio. Results: The method, validated per ICH Q2 (R1), resulted in retention times of 3.395 and 12.308 min for teneligliptin and remogliflozin etabonate, respectively. Forced degradation studies confirmed robustness, with clear peak separation and no interference from degradation products. The AGREE score of 0.65 supports its green applicability for tablet analysis in quality control. Conclusion: The AQbD-assisted RP-HPLC method developed in this study offers environmental friendliness, efficient separation with well-defined peaks, and simple mobile phase combination.

Abstract:The present work aimed to develop and validate a simple, rapid, sensitive, accurate, and precise method for simultaneous determination of metformin hydrochloride and vildagliptin in tablet and biological samples. Isocratic elution of both the analytes was performed at 35 °C by injecting 20 μL into Thermo Hypersil ODS C18 column (5 μm, 4.6 mm× 250 mm), while the flow rate was set to 0.8 mL/min. The mobile phase comprised of methanol, acetonitrile, and phosphate buffer (5:30:65, v / v , pH 3.5), and wavelength was selected at 212 nm. The overall run time per sample was 7.0 min with a retention time of 3.36 and 5.41 min for metformin hydrochloride and vildagliptin, respectively. The calibration curve was linear from 10–140 μg/mL for metformin and 1–14 μg/mL for vildagliptin with a coefficient of determination ( R 2 ) ≤ 0.9919, while repeatability and reproducibility (expressed as relative standard deviation) were lower than 1.13 and 0.97%, respectively. Force degradation studies indicated a complete separation of the analytes in the presence of their degradation products providing a high degree of method specificity. The proposed reversed-phase high-performance liquid chromatography (RP-HPLC) method was demonstrated to be simple and rapid for the determination of metformin hydrochloride and vildagliptin in commercially available tablet and biological samples providing recoveries ranged between 100.13–100.29%.

Abstract:An accurate derivative spectrophotometric method was developed and validated for the determination of dipeptidyl peptidase inhibitor vildagliptin in the pharmaceutical dosage form. The second derivative of the UV spectra has enabled the estimation of vildagliptin absorbance at 217 nm without any interferences. Linearity, precision, accuracy, detection (LOD), and quantification (LOQ) limits were established for method validation. Calibration curve was linear in the range of 10-60 μg/mL with a regression coefficient of 0.998. The method was validated as per the International Conference on Harmonization (ICH Q2 (R1)). The limit of detection and the limit of quantification were found to be 2.06 μg/mL and 6.25 μg/mL, respectively. Intra and interday precision data illustrated that the method has acceptable reproducibility as the percentage relative standard deviation (RSD) was less than 2 %, which indicates the precision of the method. The recovery was 98.39 % by the standard addition method. The percentage assay of vildagliptin was 98.06 % which showed good applicability. The following results indicate that the procedure is accurate, precise, and reproducible while being simple and less time-consuming. The method was demonstrated to be adequate for routine analysis in quality control.

Abstract:A rapid, selective, and sensitive gradient HPLC method was developed for the analysis of dissolution samples of levothyroxine sodium tablets. Current USP methodology for levothyroxine (L-T(4)) was not adequate to resolve co-elutants from a variety of levothyroxine drug product formulations. The USP method for analyzing dissolution samples of the drug product has shown significant intra- and inter-day variability. The sources of method variability include chromatographic interferences introduced by the dissolution media and the formulation excipients. In the present work, chromatographic separation of levothyroxine was achieved on an Agilent 1100 Series HPLC with a Waters Nova-pak column (250 mm × 3.9 mm) using a 0.01 M phosphate buffer (pH 3.0)-methanol (55:45, v/v) in a gradient elution mobile phase at a flow rate of 1.0 mL/min and detection UV wavelength of 225 nm. The injection volume was 800 μL and the column temperature was maintained at 28°C. The method was validated according to USP Category I requirements. The validation characteristics included accuracy, precision, specificity, linearity, and analytical range. The standard curve was found to have a linear relationship (r(2)>0.99) over the analytical range of 0.08-0.8 μg/mL. Accuracy ranged from 90 to 110% for low quality control (QC) standards and 95 to 105% for medium and high QC standards. Precision was <2% at all QC levels. The method was found to be accurate, precise, selective, and linear for L-T(4) over the analytical range. The HPLC method was successfully applied to the analysis of dissolution samples of marketed levothyroxine sodium tablets.
