Abstract:Effective accumulation of nanoparticles (NPs) in tumors is crucial for NP-assisted cancer diagnosis and treatment. With the hypothesis that aggregation of NPs stimulated by tumor microenvironment can be utilized to enhance retention and cellular uptake of NPs in tumors, we designed a smart NP system to evaluate the effect of aggregation on NPs' accumulation in tumor tissue. Gold nanoparticles (AuNPs, ~16 nm) were facilely prepared by surface modification with mixed-charge zwitterionic self-assembled monolayers, which can be stable at the pH of blood and normal tissues but aggregate instantly in response to the acidic extracellular pH of solid tumors. The zwitterionic AuNPs exhibited fast, ultrasensitive, and reversible response to the pH change from pH 7.4 to pH 6.5, which enabled the AuNPs to be well dispersed at pH 7.4 with excellent stealth ability to resist uptake by macrophages, while quickly aggregating at pH 6.5, leading to greatly enhanced uptake by cancer cells. An in vivo study demonstrated that the zwitterionic AuNPs had a considerable blood half-life with much higher tumor accumulation, retention, and cellular internalization than nonsensitive PEGylated AuNPs. A preliminary photothermal tumor ablation evaluation suggested the aggregation of AuNPs can be applied to cancer NIR photothermal therapy. These results suggest that controlled aggregation of NPs sensitive to tumor microenvironment can serve as a universal strategy to enhance the retention and cellular uptake of inorganic NPs in tumors, and modifying NPs with a mixed-charge zwitterionic surface can provide an easy way to obtain stealth properties and pH-sensitivity at the same time.
Synthesis of g-C3N4 nanosheet/Au@Ag nanoparticle hybrids as SERS probes for cancer cell diagnostics

Abstract:Chemical sensing for the convenient detection of cancer cells has been widely explored with the use of various sensing materials and techniques, but it is still a challenge to achieve ultrasensitive, simple, rapid and inexpensive detection of cancer cells. Herein, we report a surface-enhanced Raman scattering (SERS) method for the detection of cancer cells in situ. In our work, ultrathin g-C3N4 nanosheet/Au@AgNP hybrids (g-C3N4/Au@AgNPs) were fabricated using a self-assembly strategy, in which poly(ethyleneimine) (PEI) was used to obtain cationic polyelectrolyte-modified ultrathin nanosheets and anchor the Au@AgNPs. The g-C3N4 nanosheets exhibited a strong enrichment ability and the self-assembled Au@AgNPs showed an excellent SERS activity, both of which led to an ultrahigh sensitivity. The hybrids were applied to detect folic acid (FA) with the sensitive detection limit of 2.41 nM. Importantly, after being modified with FA, which targeted cancer cells with folate receptors (FRs), the formed g-C(3)N(4)n/Au@AgNPs-FA was used as a SERS probe for the on-site monitoring of cancer cells with FA as the Raman reporter molecule.

Abstract:Theranostic agents based on near-infrared absorption which integrate both imaging and therapeutic functions have attracted considerable attention. However, because of the interference signal, indiscriminate treatment usually causes side effects on normal tissues during tumor treatment. To address this limitation, we propose a new synergistically triggered mechanism, release and self-assembly of Au nanospheres, for tumor theranostics based on the synergistic effect of H+ and glutathione on the tumor microenvironment. In vitro experiments reveal that Au nanospheres release from Au@ZIF-8 at a high concentration of H+ or glutathione. Importantly, Au aggregation only appears in the synergistic effect of glutathione and lower pH and exhibits strong coupling plasmonic resonance absorption in the near-infrared region and can be used as the theranostics agent. This statement was further verified by biological transmission electron microscopy and in vivo imaging. Au@ZIF-8 is stable and produces no photoacoustic signal in normal tissue; in contrast, in the presence of overexpressed glutathione and H+, Au nanospheres release from Au@ZIF-8, assemble to aggregates, and exhibit a strong signal at the tumor site for imaging and efficient photothermal therapy. This work provides a new strategy for designing theranostic agents with sequentially responsive steps to avoid interference diagnosis signals from normal tissues and reduce damage to normal tissue during treatment.

Abstract:There dimensional (3D) metallic nanohybrids have attracted attentions for the applications in nanophotonic devices, catalysis and biosensor. Here, the 3D suspended Ag nanoparticles/Carbon nanotubes (AgNPs/CNTs) nanohybrids, obtained by self-aggregating AgNPs onto the suspended CNT networks, are synthesized and used for the SERS application. The electric field distribution with the near-field and far-field coupling can be obtained by the optimization of the nanogaps of the AgNPs. Such suspended AgNPs/CNTs nanohybrids are studied to be used as the ultra-sensitive SERS biosensor, benefiting from the bio-compatibility and chemical absorption of the CNT networks and the EM enhancement of the 3D denser "hotspots". R6G molecules with a concentration as low as 10(-15) M can be clearly detected, demonstrating their excellent Raman enhancement.

Abstract:Two important features are required for promising radiosensitizers: one is selective tumor cell targeting to enhance the therapeutic outcome via lethal DNA damage and the other is rapid clearance to enable excellent biocompatibility for potential clinical application. Herein, ultrasmall gold nanoparticles (Au NPs) with diameter smaller than 5 nm were prepared and covered with a multifunctional peptide to endow them with selective tumor cell uptake capability. Combined with X-ray irradiation, the responsive Au NPs demonstrated superior radio-sensitizing toxicity and rapid renal clearance in vivo. Methods: A responsive peptide (Tat-R-EK) consists of three build blocks were used: a cell and even nuclear penetrating block derived from human immunodeficiency virus-1 transactivator of transcription protein (Tat), an cathepsin B cleavable linker, and a zwitterionic antifouling block. Ultrasmall Au NPs were prepared and then covered by the peptide via the Au-S bonds between gold and thiol groups from cysteine. The morphology, colloidal stability and the responsiveness of obtained Au@Tat-R-EK NPs were studied using transmittance electron microscopy and dynamic laser scattering. The selective cancer cell uptake and accumulation of Au@Tat-R-EK NPs in cancer tissue were studied via ICP-MS in vitro and in vivo, respectively. The cytotoxicity of Au@Tat-R-EK NPs on HepG2 cancer cells was evaluated in terms of cell viability, DNA damage, intracellular reactive oxygen species generation, and apoptosis analysis. Finally, the biocompatibility and tumor destruction ability against orthotopic LM3 liver cancers were verified in vivo. Results: Multifunctional peptide modified ultrasmall Au NPs were successfully prepared. The Au NPs exhibited enough colloidal stability and cathepsin B-responsive surface change, leading to selectively uptake by cancer cells in vitro and accumulation to tumor sites in vivo. Combined with X-ray irradiation, the responsive Au NPs demonstrated superior radio-sensitizing cytotoxicity in vitro and therapeutic outcome on mouse liver cancer in vivo. The ultrasmall size enables rapid clearance of the Au NPs, guarantees the biocompatibility in vivo for potential clinical applications. Conclusion: Some obstacles faced by the Au NPs-based radiotherapy, such as short circulation half-life, non-specific distribution, slow clearance and low radio-sensitizing effect, were effective solved through rational design of the smart nanomedicine. This work provides new insight in designing tumor microenvironment-responsive nanomedicine in cancer radiotherapy.

Abstract:Cellular imaging technologies employing metallic surface-enhanced Raman scattering (SERS) tags have gained much interest toward clinical diagnostics, but they are still suffering from poor controlled distribution of hot spots and reproducibility of SERS signals. Here, we report the fabrication and characterization of high narrow nanogap-containing Au@Au core-shell SERS nanoparticles (GCNPs) for the identification and imaging of proteins overexpressed on the surface of cancer cells. First, plasmonic nanostructures are made of gold nanoparticles (similar to 15 nm) coated with gold shells, between which a highly narrow and uniform nanogap (similar to 1.1 nm) is formed owing to polyA anchored on the Au cores. The well controlled distribution of Raman reporter molecules, such as 4,4'-dipyridyl (44DP) and 5,5'-dithiobis(2-nitrobenzoic acid) (DTNB), are readily encoded in the nanogap and can generate strong, reproducible SERS signals. In addition, we have investigated the size-dependent SERS activity of GCNPs and found that with the same laser wavelength, the Raman enhancement discriminated between particle sizes. The maximum Raman enhancement was achieved at a certain threshold of particle size (similar to 76 nm). High narrow nanogap-containing Au@Au core-shell SERS tags (GCTs) were prepared via the functionalization of hyaluronic acid (HA) on GCNPs, which recognized the CD44 receptor, a tumor-associated surface biomarker. And it was shown that GCTs have a good targeting ability to tumour cells and promising prospects for multiplex biomarker detection.

Abstract:The identification and detection of cancer biomarkers in early stages is an important issue for the therapy of cancer. However, most methods are time-consuming and have limited sensing sensitivity and specificity. In this work, we prepared a novel plasmonic multilayered core-shell-satellite nanostructure (Au@Ag@SiO2-AuNP) consisting of a gold nanosphere with a silver coating core (Au@Ag), an ultrathin continuous silica (SiO2) shell, and a high coverage of gold nanosphere (AuNP) satellites. The Au@Ag core is a prominent surface enhanced Raman scattering (SERS) platform, and the thin SiO2 layer exhibits a long-range plasmon coupling between the Au@Ag core to the AuNP satellites, further leading to enhanced Raman scattering. Meanwhile, the outer AuNP satellites have a high biocompatibility and long-term stability. Combining the above advantages, the well-designed metallic nanoassemblies would be a promising candidate for SERS-based applications in biochemistry. For specific detection of alpha-fetoprotein (AFP), we utilized the SERS-active core-shell-satellite nanostructures modified with AFP antibody as immune probes and nitrocellulose membrane (NC) stabilized captured anti-AFP antibodies as solid substrate. To improve the detection performance, we further systematically optimized the parameters, including the silver coating thickness of the Au@Ag core and the density and size of the satellite AuNPs. Under the optimized conditions, AFP could be detected by the SERS-based sandwich immunoassay with an ultralow detection limit of 0.3 fg/mL, and the method exhibited a wide linear response from 1 fg/mL to 1 ng/mL. The limit of detection (LOD) was considerably lower than conventional methods in the literature. This work relies on the unique Au@Ag@SiO2-AuNP nanostructures as the immune probe develops a new outlook for the application of multilayered nanoassemblies and demonstrates the great potential in early tumor marker detection.

Abstract:The acidic microenvironment of tumor tissues has proven to be one of the major differences from other normal tissues. The near-infrared (NIR) light irradiation of aggregated gold nanoparticles in a tumor acidic pH-induced manner could then provide an effect approach to treat solid tumors with the advantage of minimizing the undesired damage to normal tissues. Although it is well-known the aggregation of larger nanoparticles will result in a better NIR photothermal effect, the preparation of pH-sensitive gold nanoparticles in large sizes remains a big challenge because of their worse dispersive stability. In this paper, we introduce a facile way to endow large gold nanoparticles with tunable pH-aggregation behaviors by modifying the nanoparticle surface with mixed-charge self-assembly monolayers compromising positively and negatively charged thiol ligands. Four different size nanoparticles were used to study the general principle of tailoring the pH-induced aggregation behaviors of mixed-charge gold nanoparticles (MC-GNPs) by adjusting the surface ligand composition. With proper surface ligand composition, the MC-GNPs in four different sizes that all exhibited aggregation at tumor acidic pH were obtained. The biggest MC-GNPs showed the most encouraging aggregation-enhanced photothermal efficacy in vitro when they formed aggregates. The mixed-charge self-assembled monolayers were then proved as a facile method to design pH-induced aggregation of large gold nanoparticles for better NIR photothermal cancer therapy.
Raman Reporter-Coupled Ag-Core@Au-Shell Nanostars for in Vivo Improved Surface Enhanced Raman Scattering Imaging and Near-Infrared-Triggered Photothermal Therapy in Breast Cancers

Abstract:Noble-metal nanomaterials were widely investigated as theranostic systems for surface enhanced Raman scattering (SERS) imaging, and also for photothermal therapy (PTT) of cancers. However, it was still a major challenge to explore multifunctional nanoprobes with high performance, high stability, and low toxicity. In this work, Raman reporter (DTTC)-coupled Ag-core@Au-shell nanostars (Ag@Au-DTTC) were synthesized and investigated for in vivo improved SERS imaging and near-infrared (NIR)-triggered PTT of breast cancers. By the two-step coupling of DTTC, the SERS signal was improved obviously, and the cytotoxicity of nanoparticles was also decreased by coating Au nanostars onto Ag nanoparticles. The as-prepared Ag@Au-DTTC nanostars showed high photostability and excellent photothermal performance, in which the photothermal conversion efficiency was up to 79.01% under the irradiation of an 808 nm laser. The in vitro and in vivo SERS measurements of Ag@Au-DTTC nanostars showed that the many sharp and narrow Raman peaks located at 508, 782, 844, 1135, 1242, 1331, 1464, 1510, and 1580 cm1 could be obviously observed in MCF-7 cells and in MCF-7 tumor-bearing nude mice, compared with that in pure DTTC. In 14-day treatments, the tumor volume of MCF-7 tumor-bearing nude mice injected with Ag@Au-DTTC nanostars and irradiated by an 808 nm laser almost disappeared. This study demonstrated that the as-prepared Ag@Au-DTTC nanostars could be excellent multifunctional agents for improved SERS imaging and NIR-triggered PTT of breast cancers with low risk.
Homologous Gold Nanoparticles and Nanoclusters Composites with Enhanced Surface Raman Scattering and Metal Fluorescence for Cancer Imaging

Abstract:A large number of deaths from cancer can be attributed to the lack of effective early-stage diagnostic techniques. Thus, accurate and effective early diagnosis is a major research goal worldwide. With the unique phenomenon of localized surface plasmon resonance (LSPR), plasmonic nanomaterials have attracted considerable attention for applications in surface-enhanced Raman scattering (SERS) and metal-enhanced fluorescence (MEF). Both SERS and MEF are ultra-sensitive methods for the detection and identification of early tumor at molecular level. To combine the merits of the fast and accurate imaging of MEF and the stable and clear imaging of SERS, we propose a novel dual functional imaging nanoprobe based on gold nanoparticles and gold nanocluster composites (denoted AuNPC-RGD). The gold nanoparticles are used as LSPR substrates to realized enhancement of Raman or fluorescence signal, while the gold nanoclusters serve as a fluorophore for MEF imaging, and exhibit better biocompatibility and stability. Furthermore, target molecule of cyclic Arg-Gly-Asp (cRGD) is incorporated into the composite to improve delivery efficiency, selectivity and imaging accuracy. These integrated properties endow AuNPC-RGD composites with outstanding biocompatibility and excellent imaging abilities, which could be used to achieve accurate and effective diagnosis for early cancer.
Fabrication of Optoplasmonic Particles Through Electroless Deposition and the Application in SERS-based Screening of Nodule-Involved Lung Cancer.

Abstract:In this work, a core-satellite optoplasmonic particle containing a silica microsphere covered with gold nanoparticles (AuNPs) was developed through wet chemistry synthesis in aqueous phase. The electroless deposition and galvanic replacement were employed to anchor AuNPs onto silica sphere surface. The escalated as well as expanded electric field enhancement within the dielectric-metallic interface was analyzed through finite difference time domain (FDTD) simulation. The numerical models and the surface-enhancement Raman spectroscopy (SERS) measurements over blood serum both support that the equatorial plane is the preferred collecting plane for improved signal intensity and stability. The nanocomposite emerged lower relative standard deviation (RSD) in repetitive measurement compared to AuNPs. In practice, this hybrid structure was applied for lung cancer diagnosis based on serum SERS spectra analysis of the patients diagnosed with nodules. The prediction with the aid of principal component analysis (PCA) and support-vector machine (SVM) was attempted for the classification of healthy, benign and relatively malignant sample groups. The accuracy of distinguish benign samples from malignant ones reaches over 90%. These advantages make the structure a promising SERS substrate for the early screening of cancer based on the non-invasive biological samples.

Abstract:An ideal radiosensitizer holding an enhanced tumor retention can play an incredible role in enhancing tumor radiotherapy. Herein, a strategy of acid-triggered aggregation of small-sized gold nanoparticles (GNPs) system within tumor is proposed and the resulting GNPs aggregates are applied as a radiosensitizer in vitro and in vivo. The GNPs system with the acid-triggered aggregation achieves an enhanced GNPs accumulation and retention in cancer cells and tumors in the form of the resulted GNPs aggregates. As a consequence, the radiosensitization effect shows significant improvement in cancer radiotherapy, which is shown in the studies of DNA breakage and the comet assay, and the sensitizer enhancement ratio (SER) value of the GNPs system (1.730) with MCF-7 cancer cells is much larger than that of the single GNPs (1.16). In vivo antitumor studies reveal that the GNPs system also enhances the sensitivity of MCF-7 tumor xenograft to radiotherapy. Furthermore, the GNPs aggregates improve the signal of small GNPs in vivo photoacoustic imaging. This study provides a new strategy and insights into fabricating nanoaggregates to magnify the radiosensitive efficiency of nanosystems in cancer radiotherapy.

Abstract:Considering that each diagnostic method exhibits its disadvantages, such as the poor spatial resolution of SPECT and limited penetration depth of PA imaging, the development of a diagnosis integrated agent with dual-modality imaging performance is of great research interest. Herein, an interesting triggered mechanism, release, and aggregation of radiolabeled Au NPs were explored for tumor diagnosis based on the effect of H+ in the tumor microenvironment. In vitro experiments reveal that the pH-responsive Au NPs exhibit an obvious accumulation effect and local surface plasmon resonance (LSPR) coupling effect and generate strong NIR absorption and can be used as the PA imaging agent. Importantly, Au aggregation only appears in the acid tumor microenvironment and displays excellent tumor SPECT diagnostic and photoacoustic imaging performance in vivo. This work provides a new synergistic strategy for SPECT and PA bimodal imaging to further improve the diagnostic efficacy of tumors.

Abstract:Quantitative immunoassay of specific tumor markers in clinical serum is critical for early monitoring and treatment of cancer. Herein, a surface-enhanced Raman scattering (SERS)-active immunostructure, consisting of Fe3O4@TiO2@Ag core-shell nanoparticles as immunoprobes and Au nanowires (NWs)/polydimethylsiloxane (PDMS) film as the immunosubstrate, was proposed for the quantitative determination of prostate-specific antigen (PSA) and alpha-fetoprotein (AFP). In this method, the ultrasensitive detection of the two tumor markers could be achieved in parallel on the curved flexible AuNWs/PDMS substrate. A recyclable detection with limit of detections (LOD5) of 0.49 pg/mL for PSA and 0.72 pg/mL for AFP could be realized, attributing to the excellent photocatalytic property and magnetic separation of Fe3O4@TiO2@Ag core-shell NPs. Moreover, improved linear relationships with R-2 values of 0.986 and 0.996 in the detection were perfectly obtained by utilizing the intrinsic SERS peak of PDMS as an internal standard (IS). In addition, the trace two antigens of PSA and AFP in serum samples of cancer patients were monitored using the developed immunoassay protocol, which shows superiority than chemiluminescent immunoassay (CLIA). Such a novel quantitative and recyclable SERS-based immunoassay would provide an alternative reliable method for the early accurate diagnosis of cancer.
CuPc-Fe@BSA nanocomposite: Intracellular acid-sensitive aggregation for enhanced sonodynamic and chemo-therapy

Abstract:Due to their rigid π-conjugated macrocyclic structure, organic sonosensitizers face significant aggregation in physiological conditions, hindering the production of reactive oxygen species (ROS). An acid-sensitive nanoassembly was developed to address this issue and enhance sonodynamic therapy (SDT) and emission. Initially, copper phthalocyanine (CuPc) was activated using a H2SO4-assisted hydrothermal method to introduce multiple functional groups (-COOH, -OH, and -SO3H), disrupting strong π-π stacking and promoting ROS generation and emission. Subsequently, negatively charged CuPc-SO4 was incorporated into bovine serum albumin (BSA) to form CuPc-Fe@BSA nanoparticles (10 nm) with Fe3+ ions serving as linkers. In acidic conditions, protonation of CuPc-SO4 and BSA weakened the interactions, leading to Fe3+ release and nanostructure dissociation. Protonated CuPc-SO4 tended to self-aggregate into nanorods. This acidity-sensitive aggregation is vital for achieving specific accumulation within the tumor microenvironment (TME), thereby enhancing retention and SDT efficacy. Prior to this, the nanocomposites demonstrated cycling stability under neutral conditions. Additionally, the released Fe ions exhibited mimicry of glutathione peroxidase and peroxidase activity for chemotherapy (CDT). The synergistic effect of SDT and CDT increased intracellular oxidative stress, causing mitochondrial injury and ferroptosis. Furthermore, the combined therapy induced immunogenic cell death (ICD), effectively activating anticancer immune responses and suppressing metastasis and recurrence.

Abstract:Monodispersed, readily-grafted, and biocompatible surface-enhanced Raman spectroscopic (SERS) tagging materials are developed; they are composed of bimetalic Au@Ag nanoparticle (NPs) for optical enhancement, a reporter moleculae for spectroscopic signature, and a carbon shell for protection and bioconjugation. A controllable and convenient hydrothermal synthentic route is presented to synthesize the layer-by-layer triplex Au-Ag-C core-shell NPs, which can incorporate the Raman-active label 4-mercapto benzoic acid (4-MBA). The obtained goled seed-silver coated principles can be coated further with a thickness-controlled carbon shell to form colloidal carbon-encapsulated Au-core/Ag-shell spheres with a monodisperse size distribution. Furthermore, these SERS-active spheres demonstrated interesting properties as a novel Raman tag for quantitative immunoassays. The results suggest such SERS tags can be used for multiplex and ultrasensitive detection of biomolecules as well as nontoxic, in vivo molecular imaging of animal or plant tissues.
Ultrasensitive multiplex SERS immunoassay based on porous Au-Ag alloy nanoparticle-amplified Raman signal probe and encoded photonic crystal beads

Abstract:An ultrasensitive multiplex surface-enhanced Raman scattering (SERS) immunoassay was developed using porous Au-Ag alloy nanoparticles (p-AuAg NPs) as Raman signal amplification probe coupling with encoded photonic crystal microsphere. p-AuAg NPs were synthesized and modified with the second antibody (Ab2) and Raman tag (mercaptobenzoic acid, MBA) to prepare a Raman signal-amplified probe. The high porosity of the p-AuAg NPs enables significant coupling of the localized surface plasmon resonance and thus abundant inherent hotspots for Raman signal enhancement. 3D-ordered silver nanoparticles-coated silica photonic crystal beads (Ag/SPCBs) were prepared as encoded SERS substrate for multiplex detection using their reflection peaks. The signal-amplified probe was used for multiplex detection of tumor markers carcinoembryonic antigen (CEA) and alpha fetoprotein (AFP). The wide linear ranges of 10-7-103 ng/mL for CEA and 10-4-103 ng/mL for AFP with detection limits of 1.22 × 10-8 ng/mL and 2.47 × 10-5 ng/mL for CEA and AFP at a signal-to-noise ratio of 3 were obtained. The proposed multiplex SERS immunoassay method displays ultrahigh sensitivity, wide linear range, and excellent specificity, which can be successfully applied to measure clinical serum samples with satisfactory results. The research provides a novel SERS signal enhancement strategy for the multiplex bioassay.
