Abstract:The femtosecond laser is a type of laser that can produce pulses of light of extremely short duration. The application of femtosecond laser in surgery results in no thermal effect or shock wave, so that this laser is unlikely to cause tissue injuries outside the irradiation area of the laser beam. The femtosecond laser shows promising applications in refractive surgery and corneal transplantation due to its high precision and predictability. In this paper, we review the clinical application of the femtosecond laser in refractive surgery and corneal transplantation.

Abstract:Nonlinear microscopy (NM) enables us to investigate the morphology or monitor the physiological processes of the skin through the use of ultrafast lasers. Fiber (or fiber-coupled) lasers are of great interest because they can easily be combined with a handheld, scanning nonlinear microscope. This latter feature greatly increases the utility of NM for pre-clinical applications and in vivo tissue imaging. Here, we present a fiber-coupled, sub-ps Ti–sapphire laser system being optimized for in vivo, stain-free, 3D imaging of skin alterations with a low thermal load of the skin. The laser is pumped by a low-cost, 2.1 W, 532 nm pump laser and delivers 0.5–1 ps, high-peak-power pulses at a ~20 MHz repetition rate. The spectral bandwidth of the laser is below 2 nm, which results in a low sensitivity for dispersion during fiber delivery. The reduction in the peak intensity due to the increased pulse duration is compensated by the lower repetition rate of our laser. In our proof-of-concept imaging experiments, a ~1.8 m long, commercial hollow-core photonic bandgap fiber was used for fiber delivery. Fresh and frozen skin biopsies of different skin alterations (e.g., adult hemangioma, basal cell cancer) and an unaffected control were used for high-quality, two-photon excitation fluorescence microscopy (2PEF) and second-harmonic generation (SHG) z-stack (3D) imaging.

Abstract:Picosecond laser technology was cleared by Food and Drug Administration in 2012 and enhanced our ability to clear both benign pigmentation and dermal ink through a photomechanical impact created in the tissue. This impact created is greater than comparative nanotechnology and can be accomplished with lower fluences and smaller pulse durations. The addition of a diffractive lens array (FOCUS lens) has created a new category in skin rejuvenation by stimulating elastin and collagen through photomechanical and photoacoustic effects and not only through traditional photothermal tissue effects.

Abstract:Multiphoton tomography for in vivo high-resolution multidimensional imaging has been used in clinical investigations and small animal studies. The novel femtosecond laser tomographs have been employed to detect cosmetics and pharmaceutical components in situ as well as to study the interaction of drugs with intratissue cells and the extracellular matrix under physiological conditions. Applications include the intra-tissue accumulation of sunscreen nanoparticles in humans, the monitoring the metabolic status of patients with dermatitis, the biosynthesis of collagen after administration of anti-aging products, and the detection of porphyrins after application of 5-aminolevulinic acid. More than 2000 patients and volunteers in Europe, Australia, and Asia have been investigated with these unique tomographs. In addition, femtosecond laser nanoprocessing microscopes have been employed for targeted delivery and deposition in body organs, optical transfection and optical cleaning of stem cells, as well as for the optical transfer of molecular beacons to track microRNAs. These diverse applications highlight the capacity for multiphoton tomography and femtosecond laser nanoprocessing tools to advance drug delivery research.

Abstract:Purpose: To provide an update of novel applications for the femtosecond (FS) laser in ophthalmology. Design: Perspective, literature review, case report, and commentary. Methods: Literature review. Results: The many advantages of etching flaps with the FS laser for laser in situ keratomileusis (LASIK) have been well established. Alternative applications of the FS have been approved and are now used in clinical practice. In refractive ophthalmology, the FS laser can be used for lenticule extraction to correct myopia and intrastromal biochemical manipulation to correct presbyopia. This laser can be used for preparing host and donor tissue for both full thickness and lamellar keratoplasty. Research is underway, exploring ways to employ the FS laser for different stages of cataract surgery. Cosmetic procedures with FS-assisted tattooing serve to correct leukoria. Conclusions: Advancements in technology have allowed measurable improvements in the surgical safety, efficiency, speed, and versatility of FS lasers in ophthalmology.

Abstract:This article provides an overview of both established and innovative applications of femtosecond (fs)-laser-assisted surgical techniques in ophthalmology. Fs-laser technology is unique because it allows cutting tissue at very high precision inside the eye. Fs lasers are mainly used for surgery of the human cornea and lens. New areas of application in ophthalmology are on the horizon. The latest improvement is the high pulse frequency, low-energy concept; by enlarging the numerical aperture of the focusing optics, the pulse energy threshold for optical breakdown decreases, and cutting with practically no side effects is enabled.

Abstract:The successful application of lasers in the treatment of skin diseases and cosmetic surgery is largely based on the principle of conventional selective photothermolysis which relies strongly on the difference in the absorption between the therapeutic target and its surroundings. However, when the differentiation in absorption is not sufficient, collateral damage would occur due to indiscriminate and nonspecific tissue heating. To deal with such cases, we introduce a novel spatially selective photothermolysis method based on multiphoton absorption in which the radiant energy of a tightly focused near-infrared femtosecond laser beam can be directed spatially by aiming the laser focal point to the target of interest. We construct a multimodal optical microscope to perform and monitor the spatially selective photothermolysis. We demonstrate that precise alteration of the targeted tissue is achieved while leaving surrounding tissue intact by choosing appropriate femtosecond laser exposure with multimodal optical microscopy monitoring in real time.

Abstract:Pulsed lasers at the near infrared (NIR) range have been widely used in dermatology. Ultrashort pulsed picosecond lasers are found with the specific ability of very effective activation of skin repair and remodeling along with significant photodamage. Femtosecond lasers, with a shorter pulse width, may be a promising alternative to current NIR lasers in clinic. In this study, we performed optical micromachining by a femtosecond laser at 1,030 nm to skin of live mice in two modes of scanning of focused laser and direct irradiation by unfocused laser. The acute and one-day delayed immune molecular responses of the skin to the micromachining are studied by immunofluorescence microscopy of the skin sections. Our data shows the focused laser can activate remodeling of skin without any significant immune responses. In contrast, the direct irradiation by the unfocused laser activate significant immune responses in the deep dermis with high regulation of interleukin. Those results suggest focused femtosecond laser is of good promising potential in activation of skin remodeling and repairing with little immune or physical damage.

Abstract:We use an ultrafast diode-pumped semiconductor disk laser (SDL) to demonstrate several applications in multiphoton microscopy. The ultrafast SDL is based on an optically pumped Vertical External Cavity Surface Emitting Laser (VECSEL) passively mode-locked with a semiconductor saturable absorber mirror (SESAM) and generates 170-fs pulses at a center wavelength of 1027 nm with a repetition rate of 1.63 GHz. We demonstrate the suitability of this laser for structural and functional multiphoton in vivo imaging in both Drosophila larvae and mice for a variety of fluorophores (including mKate2, tdTomato, Texas Red, OGB-1, and R-CaMP1.07) and for endogenous second-harmonic generation in muscle cell sarcomeres. We can demonstrate equivalent signal levels compared to a standard 80-MHz Ti:Sapphire laser when we increase the average power by a factor of 4.5 as predicted by theory. In addition, we compare the bleaching properties of both laser systems in fixed Drosophila larvae and find similar bleaching kinetics despite the large difference in pulse repetition rates. Our results highlight the great potential of ultrafast diode-pumped SDLs for creating a cost-efficient and compact alternative light source compared to standard Ti:Sapphire lasers for multiphoton imaging.

Abstract:Objective\r\nTo build a low repetition rate femtosecond fiber laser with its pulse width narrower than 200 fs and pulse energy bigger than 50 nJ and realize making blasting holes on pig cornea by optical expanding and focusing femtosecond laser.\r\n\r\n\r\nMethods\r\nExperimental study. Introducing very long polarization maintaining fiber into the ring cavity, the mode-locking fiber laser under the similariton amplifier scheme was used to be the oscillator. The signal laser was amplified and compressed by propagating it through the construction of pre-chirp, amplifier and de-chirp to realize outputting high quality pulse train. The high power fiber laser was expanded by the lens group. The expanded laser beam was focused into the pig cornea by the plane objective with different numerical aperture (NA) to realize making high quality blasting holes.\r\n\r\n\r\nResults\r\nThis laser facility realized outputing a very stable 19.1 MHz, 109 fs, 1.4 W, 73.2 nJ mode-locking fiber laser. After five fold expansion, the laser beam was focused into the pig cornea by the NA=0.2 plane objective to get blasting holes whose aperture were 45~75 μm. Focusing the laser beam into the pig cornea by the NA=0.5 plane objective, the aperture of the blasting holes were about 30 μm.\r\n\r\n\r\nConclusion\r\nIntroducing very long Polarization Maintaining fiber into the similariton amplifier ring cavity can improve stability effectively. The construction of pre-chirp, amplifier and de-chirp can use the nonlinear affects to depress the third order dispersion to realize high quality amplify and compression. After expanded by lens group, using higher NA plane objective to focus the expanded laser beam into the cornea can realize making higher quality blasting holes.\r\n\r\n\r\nKey words: \r\nRefractive surgical procedures; Femtosecond laser; Keratomileusis, laser in situ; Mode-locking fiber laser

Abstract:Q-switched lasers have undeniably revolutionized the field of laser dermatology since four decades ago. Just as the first-generation laser emits its photonic signal in a few nanoseconds, the picosecond laser delivers pulse widths of at least ten times shorter. These devices offer a powerful tool for treating a wide range of skin conditions with a minimal downtime for visible improvement. For the current study, a literature research was performed on the dermatological applications of picosecond laser. The literature searched on this topic between 1999 and 2023 accessible through various platforms produce a result of 62 articles. The included studies have discussed the application of picosecond laser technology in tattoo removal, treatment of epidermal and dermal pigmentation, and collagen remodeling. After sifting the data from the articles into tables, the results were discussed in detail. The study shows a lot of evidence towards the efficacy of picosecond laser, yet it draws attention to its downsides.
Micro-processing of Polymers and Biological Materials Using High Repetition Rate Femtosecond Laser Pulses

Abstract:High repetition rate femtosecond laser micro-processing has been applied to ophthalmological hydrogel polymers and ocular tissues to create novel refractive and diffractive structures. Through the optimization of laser irradiation conditions and material properties, this technology has become feasible for future industrial applications and clinical practices. A femtosecond laser micro-processing workstation has been designed and developed. Different experimental parameters of the workstation such as laser pulse duration, focusing lens, and translational stages have been described and discussed. Diffractive gratings and three-dimensional waveguides have been fabricated and characterized in hydrogel polymers, and refractive index modifications as large as + 0.06 have been observed within the laser-irradiated region. Raman spectroscopic studies have shown that our femtosecond laser micro-processing induces significant thermal accumulation, resulting in a densification of the polymer network and increasing the localized refractive index of polymers within the laser irradiated region. Different kinds of dye chromophores have been doped in hydrogel polymers to enhance the two-photon absorption during femtosecond laser micro-processing. As the result, laser scanning speed can be greatly increased while the large refractive index modifications remain. Femtosecond laser wavelength and pulse energy as well as water and dye concentration of the hydrogels are optimized. Lightly fixed ocular tissues such as corneas and lenses have been micro-processed by focused femtosecond laser pulses, and refractive index modifications without any tissue-breakdown are observed within the stromal layer of the corneas and the cortex of the lenses. Living corneas are doped with Sodium Fluorescein to increase the two-photon absorption during the laser micro-processing, and laser scanning speed can be greatly increased while inducing large refractive index modifications. No evidence of cell death has been observed in or around the laser-induced refractive index modification regions. These results support the notion that femtosecond laser micro-processing method may be an excellent means of altering the refraction or higher order aberration content of corneal tissue without cell death and short-term tissue damage, and has been named as Intra-tissue Refractive Index Shaping (IRIS). The femtosecond laser micro-processing workstation has also been employed for laser transfection of single defined cells. Some preliminary results suggest that this method can be used to trace individual cells and record their biological and morphological evolution, which is quite promising in many biomedical applications especially in immunology science. In conclusion, high repetition rate femtosecond laser micro-processing has been employed to fabricate microstructures in ophthalmological hydrogels and ocular tissues. Fiber-optic nonlinear wavelength converter for adaptive femtosecond biophotonics

Abstract:Broad and safe access to ultrafast laser technology has been hindered by the absence of optical fiber-delivered pulses with tunable central wavelength, pulse repetition rate, and pulse width in the picosecond-femtosecond regime. To address this long-standing obstacle, we developed a reliable accessory for femtosecond ytterbium fiber chirped pulse amplifiers, termed as fiber-optic nonlinear wavelength converter (FNWC), as an adaptive optical source for the emergent field of femtosecond biophotonics. This accessory embowers the fixed-wavelength laser to produce fiber delivered ~20 nJ pulses with central wavelength across 950-1150 nm, repetition rate across 1-10 MHz, and pulse width across 40-400 fs, with a long-term stability of >2000 hrs. As a prototypical label-free application in biology and medicine, we demonstrate the utility of FNWC in real-time intravital imaging synergistically integrated with modern machine learning and large-scale fluorescence lifetime imaging microscopy.

Abstract:The effects of different lasers on biological tissues are various. With short duration time, great instantaneous power and small focus dimension, the femtosecond laser pulses have expansive application future on the ultra-fast, the ultra-intense and the ultra-fine fields. And the study on tiny cell's dynamics, such as mitosis, metabolism, and apoptosis, is of great importance to understand the biological and developmental properties of cells. Meanwhile the biological big molecules and the water do not absorb the light in the near-infrared region, so it is considered to do experiments on the biological cells with the near-infrared femtosecond laser pulses. This laser technique has been applied to dissect and ablate intracellular structures. Some applications of this technology on cells, such as nanosurgery, gene transfection and chromosomal dissection are introduced. It has higher precision than the traditional technologies, and can be used without compromising the cell's viability.

Abstract:Abstract The development of the femtosecond laser (fs laser) with its ability to provide extremely rapid athermal ablation of materials has initiated a renaissance in materials science. Sample milling rates for the fs laser are orders of magnitude greater than that of traditional focused ion beam (FIB) sources currently used. In combination with minimal surface post-processing requirements, this technology is proving to be a game changer for materials research. The development of a femtosecond laser attached to a focused ion beam scanning electron microscope (LaserFIB) enables numerous new capabilities, including access to deeply buried structures as well as the production of extremely large trenches, cross sections, pillars and TEM H-bars, all while preserving microstructure and avoiding or reducing FIB polishing. Several high impact applications are now possible due to this technology in the fields of crystallography, electronics, mechanical engineering, battery research and materials sample preparation. This review article summarizes the current opportunities for this new technology focusing on the materials science megatrends of engineering materials, energy materials and electronics.

Abstract:Understanding the photodamage mechanism underlying the highly nonlinear dynamic of femtosecond laser pulses at the second transparent window of tissue is crucial for label-free microscopy. Here, we report the identification of two cavitation regimes from 1030 nm pulses when interacting with the central nervous system in zebrafish. We show that at low repetition rates, the damage is confined due to plasma-based ablation and sudden local temperature rise. At high repetition rates, the damage becomes collateral due to plasma-mediated photochemistry. Furthermore, we investigate the role of fluorescence labels with linear and nonlinear absorption pathways in optical breakdown. To verify our findings, we examined cell death and cellular responses to tissue damage, including the recruitment of fibroblasts and immune cells after irradiation. These findings contribute to advancing the emerging nonlinear optical microscopy techniques and provide a strategy for inducing precise, and localized injuries using near-infrared femtosecond laser pulses.
