Identification of lactic acid bacteria profiles from red Oncom, an Indonesian traditional mixed fermented tofu waste and peanut press cake

Abstract:Oncom is one of Indonesia’s fermented food originating from West Java. Oncom using Neurospora sp. as inoculum will produce red oncom while oncom using Rhizopus sp. will produce black oncom. Since oncom is not produced aseptically, another microorganism such as lactic acid bacteria (LAB) possibly grows. This research aimed to isolate and detect antibacterial potentials producing LAB through hemolysis activities, antibiotic resistance, molecular identification and antibacterial activities towards medical and foodborne pathogenic bacteria. This research employed red oncom collected from six traditional markets in Bandung. This study successfully isolated 103 isolates. Five isolates were proved catalase-positive. Thus, only 98 remaining isolates were tested. They showed γ-hemolysis activities except isolate Pa G which showed α-hemolysis activities. Most of the isolates had antibacterial activities towards pathogenic bacteria tested, both gram-positive and gram-negative bacteria. Moreover, 24 isolates were selected for the antibiotic resistance test because they had the highest antibacterial activities. All isolates were sensitive to chloramphenicol and 21 isolates showed sensitive results to ampicillin. Meanwhile, four isolates with the most sensitivities to antibiotic tested were successfully identified using 16S rRNA sequencing. Isolate Bo O and Bo S showed the highest similarity towards Lactobacillus paracasei subsp. tolerans NBRC 15906. Meanwhile, isolate Pm A and Ch A showed the highest similarity towards Lactobacillus plantarum NBRC 15891.
