Abstract:For the domestic yogurt culture starter cost is high, the culture starter was monopolized by foreign coun-tries. In this study, the high flavor content strains were screened from the commercially available yoghurt products and culture starter based on the characteristic flavored acetaldehyde and diacetyl, and the strain was prepared by complex and sensory evaluation. A total of 58 lactic acid bacteria were identified, including Lactobacillus aci-dophilus(9 strains), Lactobacillus rhamnosus(2 strains), Lactobacillus delbrueckii(4 strains), Streptococcus ther-mophilus (39 strains), Lactococcuslactis subsp. Lactis/Lactococcuslactis (4 Strain). The ability to produce acetalde-hyde showed Lactobacillus delbrueckii>Streptococcus thermophilus>Lactococcuslactis>Lactobacillus acidophilus>Lacto-bacillus rhamnosus, up to 25.57μg/g. The ability to produce diacetyl is Lactobacillus rhamnosus>Lactobacillus acidophilus>Streptococcus thermophilus>Lactococcuslactis>Lactobacillus delbrueckii, up to 3.57μg/g. Lactobacillus delbrueckii and Lactobacillus rhamnosus have strong ability to produce acid. After the strain was mixed, the flavor content increased, under certain conditions, showing the higher proportion of acetaldehyde/diacetyl, the higher the content of acetaldehyde, the better the sensory flavor.
Characterization and antimicrobial activities of lactic acid bacteria isolated from selected Nigerian traditional fermented foods

Abstract:For production and preservation of traditional fermented foods, the genera, lactic acid bacteria (LAB) have been used. This study was carried out to determine the characteristics and the antimicrobial activities of lactic acid bacteria isolated from selected Nigerian traditional fermented foods. Changes in pH and titratable acidity (TA) of the samples were investigated for a period of four days (96 h). Eleven tentative LAB from fermented maize and cassava (Ogi and Fufu, respectively) were isolated and characterized. The spoilage organisms from fish were aseptically identified and the antimicrobial activity was determined by agar well diffusion method against three isolated food spoilage organisms (Pseudomonas aeruginosa, Enterobacter aerogene and Bacillus cereus). The isolates were selected and further identified as Lactobacillus amylolyticus strain L6, Lactobacillus plantarum strain ci-4w and Lactobacillus sakei strain MLS1 by the aide of genotypic characteristics (16S rRNA gene sequences). These strains were screened for their EPS producing activity, resistance to low pH and bile salts as well as bacteriocin activity. These strains can be used as starter culture or protective cultures to improve the hygiene, quality and increased safety of the food products by inhibiting the food borne pathogens and spoilage microorganisms. Developing Starter Culture from Lactic Acid Bacteria Isolated from Cow Milk for the Production of Nigerian “Nunu”

Abstract:Starter cultures are mostly organisms deliberately introduced into food as a single culture or mixture of cultures to bring about fermentation and production of desired characteristics in food through metabolic and enzymatic activities. Organisms in this category have attained the generally regarded as safe (GRAS) status, are majorly lactic acid bacteria with food acidification and production of natural preservatives potential. This research was aimed at developing a starter culture for the production of local dairy drink “Nunu” Cow milk was obtained from “Gaa Mobolohunduro”, Tanke, Ilorin, Kwara State, Nigeria. Potential lactose fermenters were isolated on MRS agar in pour plate technique. The isolates were characterized and identified using standard procedures. Lactic Acid Bacteria isolated were inoculated as a starter culture for milk fermentation and the products were subjected to pH, titratable acid and organoleptic assessments following standard procedures. Two of the isolated lactic acid bacteria with the best fermentation products were identified through molecular technique. A total of six isolates were obtained and identified as Lactococcus sp., Leuconostoc sp., Pediococcus sp. and Lactobacillus spp. The pH ranges from 5.46±0.01 to 6.03±0.01while the titratable acidity ranged from 0.244±0.004 to 0.526±0.006. The moisture content ranged from 90.50±0.30 to 91.75±0.35. The fermentation product from the isolated Lactobacillus sp. scored 80 % during the organoleptic assessment. Molecular evidence confirmed the two isolates as Lactic Acid Bacteria. In conclusion, isolated species could be used as potential organisms for milk fermentation.
Genotypic characterization and safety assessment of lactic acid bacteria from indigenous African fermented food products

Abstract:BackgroundIndigenous fermented food products play an essential role in the diet of millions of Africans. Lactic acid bacteria (LAB) are among the predominant microbial species in African indigenous fermented food products and are used for different applications in the food and biotechnology industries. Numerous studies have described antimicrobial susceptibility profiles of LAB from different parts of the world. However, there is limited information on antimicrobial resistance profiles of LAB from Africa. The aim of this study was to characterize 33 LAB previously isolated from three different African indigenous fermented food products using (GTG)5-based rep-PCR, sequencing of the 16S rRNA gene and species-specific PCR techniques for differentiation of closely related species and further evaluate their antibiotic resistance profiles by the broth microdilution method and their haemolytic activity on sheep blood agar plates as indicators of safety traits among these bacteria.ResultsUsing molecular biology based methods and selected phenotypic tests such as catalase reaction, CO2 production from glucose, colonies and cells morphology, the isolates were identified as Lactobacillus delbrueckii, Lactobacillus fermentum, Lactobacillus ghanensis, Lactobacillus plantarum, Lactobacillus salivarius, Leuconostoc pseudomesenteroides, Pediococcus acidilactici, Pediococcus pentosaceus and Weissella confusa. The bacteria were susceptible to ampicillin, chloramphenicol, clindamycin and erythromycin but resistant to vancomycin, kanamycin and streptomycin. Variable sensitivity profiles to tetracycline and gentamicin was observed among the isolates with Lb. plantarum, Lb. salivarius, W. confusa (except strain SK9-5) and Lb. fermentum strains being susceptible to tetracycline whereas Pediococcus strains and Lb. ghanensis strains were resistant. For gentamicin, Leuc. pseudomesenteroides, Lb. ghanensis and Ped. acidilactici strains were resistant to 64 mg/L whereas some W. confusa and Lb. plantarum strains had a MIC value of 16 mg/L and 32 mg/L respectively. No β-haemolytic activity was observed, however, α-haemolytic activity was observed in 27% (9) of the strains comprising Lb. salivarius (6), W. confusa (2) and Lb. delbrueckii (1) isolates.ConclusionsThe resistance to kanamycin and vancomycin is probably an intrinsic feature since similar observations were reported in the literature for LAB. Low prevalence of pathogenicity indicator traits were observed among the isolates especially with the presence of poor haemolytic activities and they could therefore be considered as interesting candidates for selection of starter cultures or probiotics for different applications.

Probiotic Potential of Lactic Acid Bacteria from Traditional Fermented Dairy and Meat Products: Assessment by In Vitro Tests and Molecular Characterization

Abstract:The aim of the present study was to evaluate the probiotic potential of lactic acid bacteria (LAB) isolated from Greek traditional fermented products. A series of In vitro tests that included survival in simulated gastrointestinal conditions (resistance to low pH, bile salts resistance and bile salts hydrolysis) and safety assessment (resistance to antibiotics, haemolytic and antimicrobial activity) were performed to select potential probiotic candidates, while Lactobacillus rhamnosus GG and Lactobacillus casei Shirota were used as reference strains. Initially, a total of 255 isolates of LAB have been recovered and screened for their survival in simulated gastrointestinal tract conditions and 133 isolates that exhibited moderate or good behavior in these tests were subsequently differentiated and characterized at species level with molecular tools. Pulsed Field Gel Electrophoresis was applied for strain differentiation, while species differentiation was based on restriction analysis of the amplified 16S rRNA gene. Specific multiplex PCR assay targeting the recA genes was applied to resolve the species level of the isolates, belonged to Lb. plantarum group. From the 133 isolates, 47 different strains were recovered and were assigned to Lactobacillus sakei(14), Lactobacillus curvatus (4), Leuconostoc mesenteroides (4), Lactococcus lactis (4), Lactobacillus casei group (1), Lactobacillus brevis (1), Lb. plantarum (10), Lb. pentosus (7) and Lb. paraplantarum (2). The identified strains with good behavior to the gastrointestinal tract tests were selected and further evaluated for their safety aspect. In conclusion, 19 out of the 47 identified strains were assessed as well-behaved, under simulated gastrointestinal conditions and also considered as safe, possessing thus desirable In vitro probiotic properties similar or better to that of the reference strains. These strains may be considered as good candidates for further investigation at in vivo and in situ studies to assess their potential health benefits and their performance as novel probiotic starters or adjunct cultures.

Abstract:Lactobacillus contribute to the organoleptic quality of foods during the fermentation process in which they are involved. They also have the ability to inhibit the growth of undesirable germs by producing metabolites such as bacteriocins. The aim of this study was to isolate and identify the different Lactobacillus species from various fermentation processes and from the Doderlein flora. Lactobacillus were isolated from raw cow milk, curdled milk, cassava ferment (Magnan) and Doderlein flora on MRS agar. Strains were identified by mass spectrometry (MALDI-TOF). 213 Lactobacillus strains were isolated and identified from the different matrices. The main species isolated from raw cow's milk and curdled milk were Lactobacillus plantarum (31.58%), Lactobacillus casei (26.31%), Lactococcus lactis (21.05%), Lactobacillus delbrueckii spp bulgaricus (31%), Lactobacillus helveticus (30%) and Lactobacillus fermentum (25%) respectively. In the cassava ferment, Lactobacillus plantarum (37.5%), Lactobacillus rhamnosus (25%) and Lactobacillus reuteri (25%) were the most isolated species. The main species of Doderlein flora were Lactobacillus acidophilus (50%) and Lactobacillus plantarum (25%). This study showed a diversity of Lactobacillus present and involved in the different fermentation processes of foods and Doderlein flora (Commensal flora of the vagina). The qualitative and quantitative proportions of the different Lactobacillus species depend on the matrix. These Lactobacillus can potentially produce substances (bacteriocins) that inhibit the growth of pathogenic bacteria and multi-resistant bacteria.

Abstract:Lactic acid bacteria (LAB) have long been consumed by people in several fermented foods such as dairy products. A study was conducted on lactating dairy cows to isolate and characterize LAB from dairy products found in and around Bahir-Dar city, North Western Ethiopia. Milk and milk products were randomly collected from dairy farms, milk vending shops, individual households, and supermarkets for bacteriological investigations. A total of sixteen samples were taken from different sources and cultured on different selective media: de Man, Rogosa, and Sharpe (MRS) agar for Lactobacillus spp.; M17 agar for Lactococcus spp.; Rogasa SL agar for Streptococci spp.; and MRS supplemented with cysteine (0.5%) for Bifidobacteria spp. Different laboratory techniques were implemented for LAB isolation and identification. A total of 41 bacterial isolates were grouped under five different genera of LAB and Bifidobacteria spp. were identified based on the growth morphology on the selective media, growth at a different temperature, gas production from glucose, carbohydrate fermentation, and other biochemical tests. LAB genera such as Lactobacillus, Lactococcus, Leuconostoc, Pediococcus, Streptococcus, and Bifidobacterium spp. were isolated and identified from raw milk, cheese, and yogurt. Based on the current study, the majority of the LAB (24.38%) was isolated from cheese and yogurt. Among these, Lactobacillus, Lactococcus (21.94%), Streptococcus (19.51%), Leuconostoc (14.64%), Bifidobacteria (12.19%), and Pediococcus (7.31%) spp. were also identified from these products. Furthermore, based on the bacterial load count and different identification methodologies, our study revealed that Lactobacillus spp. were the dominant LAB isolated from milk and milk products. As a result, since there are few studies on the isolation and identification of lactic acid bacteria from dairy products in Ethiopia, more research studies are needed to complete the identification and characterization to species level and their possible role as probiotics.

Abstract:The taxonomic differentiation among lactic acid bacteria (LAB) using only phenotypic methods is inadequate. In the past, several phenotypic techniques have been applied in identification of different species of LAB from Nigerian indigenous fermented foods. In this study, two less-expensive and rapid molecular techniques were employed in the identification and differentiation of LAB species isolated from some Nigerian fermented foods: gari (cassava mash), fufu (cassava dough) and ogi (maize gruel) to avoid the common errors that are synonymous with the use of conventional methods. The size of the amplified products for the LAB species using universal primers fD1 and rD1 was about 1500 bp. The differentiation of the three predominant LAB, Lactobacillus plantarum, Pediococcus pentosaceus and Weissella confusa to species level was performed by intergenic transcribed spacer (ITS-PCR) and amplified ribosomal DNA restriction analysis (ARDRA) using three restriction endonucleases: HinfI, HaeIII and RsaI. In summary, the study demonstrated that the ARDRA and ITS-PCR analysis are simple, fast and efficient methods that can be used for the routine identification and rapid differentiation of LAB species which are often regularly used as probiotics, starter culture and biopreservative in fermented foods consumed since they are generally regarded as safe (GRAS).

Abstract:It is well known that consumers are keen to try fermented milk products with different flavors and starter cultures are important in determining the resulting fermented dairy products. Here, we present the phenome of 227 Lactococcus lactis subsp. lactis isolates from traditionally fermented dairy products and the selection of potential starter strains. Large-scale phenotyping revealed significant technological diversity in fermentation characteristics amongst the isolates including variation in fermentation time, viscosity, water holding capacity (WHC) and free amino nitrogen (FAN) production. The 16 isolates with the best fermentation characteristics were compared, in a sensory evaluation, with the commercial starter Chr. Hansen R-704 as excellent fermentation characteristics to identify potential starter isolates and find the isolate which can product good flavors. From these, and from solid phase micro extraction (SPME) – gas chromatography (GC)-mass spectrometry (MS) analysis, we identified IMAU11823 and IMAU11919 as producing 3-methyl butanal and 3-methyl-2-butanone which contribute to the malt aroma. This study expands the characterization of L. lactis subsp. lactis phenotypic dataset and technological diversity and identified isolates with potential culture starter in the fermentation industry.

Abstract:Probiotic microorganisms are incorporated in foods due to their numerous health benefits. We investigated lactic acid bacteria (LAB) and yeasts isolated from goat milk in Nigeria for novel probiotic strains. In this study, a total of 27 LAB and 23 yeast strains were assessed for their probiotic potentials. Only six LAB strains ( Weissella cibaria GM 93m3, Weissella confusa GM 92m1, Pediococcus acidilactici GM 18a, Pediococcus pentosaceus GM 23d, Lactiplantibacillus pentosus GM 102s4, Limosilactobacillus fermentum GM 30m1) and four yeast strains ( Candida tropicalis 12a, C. tropicalis 33d, Diutina rugosa 53b, and D. rugosa 77a) identified using partial 16S and 26S rDNA sequencing, respectively, showed survival at pH 2.5, 0.3% bile salt, and simulated gastrointestinal conditions and possessed auto-aggregative and hydrophobic properties, thus satisfying key in vitro criteria as probiotics. All LAB strains showed coaggregation properties and antimicrobial activities against pathogens. Pediococcus pentosaceus GM 23d recorded the strongest coaggregation percentage (34–94%) against 14 pathogens, while W. cibaria GM 93m3 showed the least (6–57%) against eight of the 14 pathogens. The whole cell and extracellular extracts of LAB and yeast strains, with the exception of D. rugosa 77a, had either 2,2-diphenyl-1-picryl-hydrazyl and/or hydroxyl radical scavenging activity. In conclusion, all six LAB and four yeast strains are important probiotic candidates that can be further investigated for use as functional starter cultures.

Abstract:Lactic acid bacteria (LAB) are renowned for the potential of producing antimicrobial compounds. This study focuses on isolation and antimicrobial effect of lactic acid bacteria isolated from locally fermented cow milk products (Nono, Kindirmo and Maishanu). The antimicrobial effect of the LAB isolates against clinical test isolates was also investigated. Fifty (50) samples each of Nono and Kindirmo, and twenty-five (25) of Maishanu were collected from cow milk products sellers in Keffi, Nasarawa State Nigeria. Standard microbiological methods were used for the isolation and identification of Lactic acid bacteria from fermented cow milk products. The percentage isolation rate of Lactobacillus species from Nono and Kindirmo showed that 52.0% were isolated from Nono and 58.0% from kindirmo. The highest percentage occurring Lactobacillus species from Nono was Lactobacillus plantarum (14.0%) and the highest percentage occurring Lactobacillus species from kindirmo was Lactobacillus delbrueckii (16.0%). The Lactobacillus species isolated were screened for antibacterial activity and it was observed that seven (7) were able to show inhibition zones. The antimicrobial effect of the LAB culture supernatant against clinical isolates of Staphylococcus aureus, Escherichia coli and Shigella dysenteriae was investigated using agar diffusion technique. The highest 22.00mm was observed by Lactobacillus delbrueckii and Lactobacillus delbrueckii against Shigella dysenteriae, while the least inhibition zone of 15.00mm was observed by Lactobacillus delbrueckii and Lactobacillus acidophilus against E. coli. In conclusion, the zones of inhibition observed in this research strongly suggest that various antimicrobial compounds (lactic acid, acetic acid, propionic acid and bacteriocins) produced during the fermentation process are potent in the control of growth of the test bacteria.
Isolation and characterization of potential probiotic lactic acid bacteria isolated from cow milk and milk products

Abstract:Probiotics refer to the living microorganisms that exhibit beneficial effect on the health of human by the intestinal microbial balance. Most widely used probiotics are lactic acid bacterial group found in milk and milk products. This study was aimed to characterize the probiotic properties of lactic acid bacterial (LAB) strains isolated from cheddar cheese, yoghurt and cow milk. Bacterial strains naturally grown in milk, cheddar cheese and yoghurt were isolated using De Mann Rogosa Sharpe (MRS) agar medium and incubated at 37oC for 48 h separately under aerobic and anaerobic conditions. There were eight strains grown under aerobic conditions and they were isolated, purified and characterization was done based on the morphological and biochemical analysis such as gram staining, catalase test and motility test. All the eight isolates were either rod or cocci shaped, gram positive, catalase negative, non-motile and non-spore formers. These eight isolated strains were identified as lactic acid bacteria. When screening of the 8 isolates was done to determine their antimicrobial activities against five human pathogenic strains such as E.coli, Klebsiella pneumoniae, Pseudomonas eaeruginosa, Salmonella sp and Staphylococcus aureus, three isolates (M6, C1 and Y1) showed wide spectrum antimicrobial activity. To determine the probiotic properties of these three isolates, different tests such as tolerance to acid, NaCl and bile, lactose utilization and antibiotic resistance were done. Though all the three isolates showed resistance to stomach pH (pH 3.0), the strain C1 showed significantly higher tolerance to stomach pH than the other strains. Though the three isolates grew well in the presence of NaCl and 0.3% bile, the isolate M6 showed significantly higher growth with NaCl and 0.3% bile than the others. Even though all the three isolates had the capacity of utilizing lactose, the isolate M6 showed prominent colour change in the lactose utilization test than the other two strains. When antibiotic susceptibility of the isolated LABs were evaluated using four antibiotics such as Ampicillin, Streptomycin, Bacitracin and Gentamycin, the strain M6 showed significantly higher resistance to Ampicillin and Bacitracin but sensitive to Streptomycin and Gentamycin. Other two isolates (C1 and Y1) were sensitive to all the four antibiotics used. Based on the antibiotic sensitivity tests and the analysis of probiotic properties, the isolated strain M6 was confirmed as a potential probiotic lactic acid bacterium. This strain was identified as Lactobacillus plantarum strain CIP 103151 through16S rDNA sequence analysis.

Abstract:A selection of 36 commercial probiotic fermented dairy products from UK and Europe markets were evaluated for the numbers, types, and viability of Lactobacillus strains against the stated information on their packages. A comparative study was carried out on selectivity of MRS-Clindamycin, MRS-Sorbitol, and MRS-IM Maltose, to select the right medium for enumeration of probiotic Lactobacillus. Based on selectivity of medium for recovery of the targeted lactobacilli, and also simplicity of preparation, MRS-Clindamycin was chosen as the best medium for enumeration of probiotic Lactobacillus in fermented milks. The results of enumeration of lactobacilli showed that 22 out of a total 36 tested products contained more than 106 colony-forming units/g at the end of their shelf life, which comply with the recommended minimum therapeutic level for probiotics. Rep-PCR using primer GTG-5 was applied for initial discrimination of isolated strains, and isolates, which presented different band profile, were placed in different groups. The isolated Lactobacillus spp. were identified mainly as Lactobacillus acidophilus, Lactobacillus casei, and Lactobacillus paracasei by analysis of partial sequences of the 16S ribosomal RNA and rpoA genes.

Abstract:Nono is a traditional cultured dairy product consumed across West Africa. In this study, five cultures isolated from Nigerian-produced nono and three purified lactic acid bacteria from the USDA-NRRL were examined for use in preparing nono starter cultures. Isolated cultures were characterized using microbiological and biochemical tests, including 16s rDNA sequencing to identify the genotype. Each isolated strain was cultured and inoculated into UHT milk (1% v/v) and allowed to ferment for 24 h at 25 °C. Fermented products were evaluated for pH, moisture content, water activity, and viscosity, and their descriptive sensory properties were noted. The isolate that resulted in sensory properties most similar to traditional nono was then used as the primary strain for subsequent starter culture blends made with the NRRL cultures. These blends were used for the fermentation of nono and compared with commercial nono samples. Isolates obtained from nono were as follows: Lactobacillus fermentum, Lactobacillus paracasei, and, surprisingly, Lactobacillus rhamnosus, which has not been previously reported as a part of the nono microflora. There was no significant difference in the physical parameters of nono made from the individual indigenous isolates and a similar pattern was observed for the organisms from NRRL, except that their total titratable acidity and viscosities were significantly (p < 0.05) higher than those of the indigenous organisms. Compounded starter made with L. rhamnosus and NRRL cultures was then used to make nono that showed significantly (p < 0.05) different pH and viscosity values than commercially purchased nono, while sensory evaluation showed that nono made from the new starter culture had a high overall consumer acceptance score.

Abstract:This review deals with recent research advances on the biodiversity of Chinese traditional foods lactic acid bacteria (LAB). Traditional foods like yoghourt, kumiss, kefir and fermented camel milk are abundant in LAB. The distribution of LAB taxa is highly variable from one traditional food to another. Isolation of LAB taxa mainly depends on the sample plating methods, incubation media and conditions. It is therefore difficult to define associations between population composition and either the type of traditional food or the geographic location. Identification of isolated strains needs a ployphasic approach, including a combination of phenotypic and genotypic methods, among which the latter often based on the polymerase chain reaction (PCR) and encompassing 16S rRNA sequencing and DNA-DNA hybridizations. The main obstacle in current identification approach of LAB strains is the lack of a robust and exchangeable identification system for all LAB species. Some proposal are come up in this paper : 1) to establish a reliable and scientific system in sample collection, strain isolation and identification. 2) to isolate new characteristic strains. 3) to explore and develop new products contained LAB. 4) to intensify the research on the function and mechanism of isolated strains.

Abstract:Lactic acid bacteria (LAB) are important microorganisms for the food industry due to their functional activity, as starters and potential probiotic strains. With that in mind, we explored the LAB diversity in raw buffalo milk, screening for novel potential probiotic strains. A total of 11 strains were identified by combination of MALDI-TOF and partial 16S rDNA sequencing and selected as potential probiotic candidates. Bacteria innocuity assessment was performed by determining antimicrobial susceptibility and the presence of virulence factors. Antagonism activity against Escherichia coli, Pseudomonas aeruginosa, Listeria monocytogenes and Staphylococcus aureus was assessed, as well as milk proteolytic activity and exopolysaccharides production. Seven strains were identified as innocuous and two of them, Lactobacillus rhamnosus LB1.5 and Lactobacillus paracasei LB6.4 were selected for further probiotic potential analyses. Both strains demonstrated adhesion ability to Caco-2 cells, coaggregated with S. aureus and E. coli and maintained cell viability after gastrointestinal simulation in vitro, suggesting their probiotic potential. Furthermore, the transcriptional response of Lact. rhamnosus LB1.5 and Lact. paracasei LB6.4 to in vitro acid stress was assessed by RT-qPCR targeting seven genes related to adhesion, aggregation, stress tolerance, DNA repair and central metabolism. The association between the transcriptional responses and the maintenance of cell viability after gastrointestinal simulation highlights the genetic ability as probiotic of the two selected strains. Finally, we have concluded that Lact. rhamnosus LB1.5 and Lact. paracasei LB6.4 are important probiotic candidates to further in vivo studies.

Abstract:Lactic acid bacteria (LAB) commonly used in food as starter cultures are known to produce antimicrobial substances such as bacteriocins and have great potential as food biopreservatives. LAB isolated from traditional fermented foods (appam batter and pickles) were screened for bacteriocin production. Two lactobacilli, LABB and LABP (one from each source) producing bacteriocins were characterized. Both the bacilli were homo-fermentative, catalase negative and micro-aerophilic in nature. LABB was found to be a thermobacterium growing at 45 degrees C while LABP was a streptobacterium growing at 15 degrees C. Both were able to grow at pH 4.5-8.6 but were intolerant to high salt concentration. They failed to produce gas from glucose as well as ammonia from arginine. Among the sugars examined they could not ferment arabinose, raffinose, rhamnose or xylose. Additionally, LABB could not ferment esculin, gluconate or mannose. LABB is identified as Lactobacillus acidophilus while LABP as Lb. casei. Their bacteriocins showed a broad inhibitory spectrum against the indicator organisms tested. They were active below pH 8.0 and after autoclaving as well. There was a complete loss of activity when treated with proteolytic enzymes such as trypsin indicating the proteinaceous nature of the active molecules. SDS-PAGE of partially purified bacteriocins indicated the molecular mass of the bacteriocin as 3.8 and 4.5 kDa for LABB and LABP respectively.

Abstract:Purpose: The current study’s aim is to evaluate the probiotic potential of lactic acid bacteria strains isolated from traditional "Tchoukou" milk cheeses produced in a selected region of Niger. Methodology: Nine Samples were collected in selected regions of Niger (Tahoua, Maradi, and Zinder).Probiotic properties of isolated LAB were identified based on their acid tolerance, bile salt tolerance, auto-aggregation ability, simulated stomach and duodenum passage, simulated gastric juice survivability and their antimicrobial activities. Findings: A total of eighteen strains were analysed in vitro for acid tolerance, bile tolerance, survival under simulated gastro-intestinal tract conditions and antimicrobial activity against index organisms. The results indicated that all seventeen strains exhibited a high viability after twenty-four hours of incubation at pH 2.5 and pH 3, but a decreased viability at pH 2.0 in which only eight strains were able to survive, strain C13 failed to grow at the three different pH. In this study, the isolates generally survived better after exposure to 0.3% bile salt. Also were able to survive exposure to simulated stomach and duodenum passage (SSDP) for three hours ranging from (89%-100%).All strains were able to survive under simulated gastric juice at different pH (2, 2.5 and 3). for auto-aggregation Only fifteen isolates showed the best auto-aggregation abilities ranging from (15-83%) and the other two had less auto-aggregation ability (2-11%). The isolates showed diverse antimicrobial activity against the index organisms. Among the isolates, only three (C1, C2 and C9) could not inhibit any of the selected pathogens. Unique contribution to theory, practice and policy: This study was conducted to characterize the probiotic properties of LAB isolate which could serve as a potential source for industries and commercial applications.
