Abstract:The power output and stability of enzyme-based biofuel cells (BFCs) is greatly dependent on the properties of both the biocathode and bioanode, which may be adapted for portable power production. In this paper, a novel highly uniform three-dimensional (3D) macroporous gold (MP-Au) film was prepared by heating the gold "supraspheres", which were synthesized by a bottom-up protein templating approach, and followed by modification of laccase on the MP-Au film by covalent immobilization. The as-prepared laccase/MP-Au biocathode exihibited an onset potential of 0.62 V versus saturated calomel electrode (SCE, or 0.86 V vs NHE, normal hydrogen electrode) toward O2 reduction and a high catalytic current of 0.61 mAcm(-2). On the other hand, mutated glucose dehydrogenase (GDH) surface displayed bacteria (GDH-bacteria) were used to improve the stability of the glucose oxidation at the bioanode. The as-assembled membraneless glucose/O2 fuel cell showed a high power output of 55.8 ± 2.0 μW cm(-2) and open circuit potential of 0.80 V, contributing to the improved electrocatalysis toward O2 reduction at the laccase/MP-Au biocathode. Moreover, the BFC retained 84% of its maximal power density even after continuous operation for 55 h because of the high stability of the bacterial surface displayed GDH mutant toward glucose oxidation. Our findings may be promising for the development of more efficient glucose BFC for portable battery or self-powered device applications.
Controllable Display of Sequential Enzymes on Yeast Surface with Enhanced Biocatalytic Activity Toward Efficient Enzymatic Biofuel Cells.

Abstract:A precisely localized enzyme cascade was constructed by integrating two sequential enzymes, glucoamylase (GA) and glucose oxidase (GOx), on a yeast cell surface through an a-agglutinin receptor as the anchoring motif with cohesin-dockerin interaction. The overall catalytic activities of the combinant strains were significantly dependent on the assembly method, enzyme molecular size, enzyme order, and enzyme stoichiometry. The combinant strain with GA-DocC initially bound scaffoldin prior to GOx-DocT exhibited a higher overall reaction rate. The highest overall reaction rate (29.28 +/- 1.15 nmol H2O2 min(-1) mL(-1)) was achieved when GA/ GOx ratio was 2:1 with enzyme order: yeast-GA-GOx-GA, 4-fold enhancement compared to free enzyme mixture. Further, the first example of starch/O-2 enzymatic biofuel cells (EBFCs) using codisplayed GA/GOx based bioanodes were assembled, demonstrating excellent direct biomass-to-electricity conversion. The optimized EBFC registered an open-circuit voltage of 0.78 V and maximum power density (P-max) of 36.1 +/- 2.5 mu W cm(-2), significantly higher than the P-max for other starch/O-2 EBFCs reported so far. Therefore, this work highlights rational organization of sequential enzymes for enhanced biocatalytic activity and stability, which would find applications in biocatalysis, enzymatic biofuel cells, biosensing, and bioelectro-synthesis.

Abstract:Enzymatic biofuel cells (EBFCs) provide a new strategy to enable direct biomass-to-electricity conversion, posing considerable demand on sequential enzymes. However, artificial blend of multi-enzyme systems often suffer biocatalytic inefficiency due to the rambling mixture of catalytic units. In an attempt to construct a highperformance starch/O-2 EBFC, herein we prepared a starch-oxidizing bioanode based on displaying a sequential enzyme system of glucoamylase (GA) and glucose dehydrogenase (GDH) on E.coli cell surfaces in a precise way using cohesin-dockerin interactions. The enzyme stoichiometry was optimized, with GA & GDH (3:1)-E.coli exhibiting the highest catalytic reaction rate. The bioanode employed polymerized methylene blue (polyMB) to collect electrons from the oxidation of NADH into NAD(+), which jointly oxidized starch together with co displayed GA and GDH. The bioanode was oxygen-insensitive, which can be combined with a laccase based biocathode, resulting in a membranless starch/O-2 EBFC in a non-compartmentalized configuration. The optimal EBFC exhibited an open-circuit voltage (OCV) of 0.74 V, a maximum power density of 30.1 +/- 2.8 mu W cm(-2), and good operational stability.

Abstract:An electrocatalytic glucose oxidation reaction (GOR) is crucialfor building a high-efficiency biofuel cell for a more sustainable society andconstructing a sophisticated device to precisely detect trace amounts of glucosein blood and food for a more healthy life. Yet, the reported GOR catalysts sufferlow activity (current induced by catalysts toward 1 mM glucose in a 1 cm-2electrode,mu AmM-1cm-2), slow response, and poor response to trace glucose.Herein, we fabricate noble-metal-free WO3-decorated carbon nanotubes withstrong W-C bonds (WO3/CNT60) (CNT, carbon nanotube). In the GOR,WO3/CNT60 triggers an activity as high as 1960 mu AmM-1cm-2, a responsetime of only 3 s, and a response minimum of only 5 mu M and exhibitsoutstanding stability, anti-interference ability to impurities, and reusability. TheGOR performance of WO3/CNT60 is much better than the WO3/CNTcatalyst without W-C bonds (1320 mu AmM-1cm-2,10s,40 mu M) and the widely used noble-metal catalyst (270 mu AmM-1cm-2,10s, 25 mu M). The strong W-C bonds create more C-W-O-W bridge sites active for catalyzing the GOR, thus enhancing the GORperformance of WO3/CNT60. These results open a new way for fabricating a noble-metal-free high-efficiency biofuel cell andsophisticated device to precisely detect trace amounts of glucose and are also helpful for detecting and converting complex moleculeslike polyols and poly(carboxylic acid)s

Abstract:The appearance and evolution of biofuel cells can be categorized into three groups: microbial biofuel cells (MBFCs), enzymatic biofuel cells (EBFCs), and enzyme-like nanomaterial (nanozyme)-based biofuel cells (NBFCs). MBFCs can produce electricity from waste; however, they have significantly low power output as well as difficulty in controlling electron transfer and microbial growth. EBFCs are more productive in generating electricity with the assistance of natural enzymes, but their vulnerability under diverse environmental conditions has critically hindered practical applications. In contrast, because of the intrinsic advantages of nanozymes, such as high stability and robustness even in harsh conditions, low synthesis cost through facile scale-up, and tunable catalytic activity, NBFCs have attracted attention, particularly for developing wearable and implantable devices to generate electricity from glucose in the physiological fluids of plants, animals, and humans. In this review, recent studies on NBFCs, including the synthetic strategies and catalytic activities of metal and metal oxide-based nanozymes, the mechanism of electricity generation from glucose, and representative studies are reviewed and discussed. Current challenges and prospects for the utilization of nanozymes in glucose biofuel cells are also discussed.

Abstract:Glucose biofuel cell (GBFC) is a power supply device which has attracted considerable attention because of its green environmental protection and high economic benefits. Fuels like glucose and oxygen are ubiquitous in physiological fluids, allowing the direct harvest of energy from human bodies. Compared with conventional batteries such as Li-Po, GBFC is a more promising alternative to power medical devices without the need to be replaced or refueled. However, the energy conversion efficiency of the existing GBFCs still needs to be further improved for practical applications. In this paper, the performance of the GBFC was studied based on single-walled carbon nanotubes (SWCNTs), which have relatively high conductivity and large specific surface area that could improve the activity of enzymes immobilized on the electrode surface and thus realize the direct electron transfer (DET). After optimization of the catalysts’ amount, the GBFC based on SWCNTs performed well with two Pt layers sprayed on one side of the proton exchange membrane (PEM) and 1.5 mL glucose oxidase (GOx) dropped on the other side, which attained the highest open-circuit potential (OCP) of 0.4 V. After being encapsulated with a flexible porous enclosure made by polydimethylsiloxane (PDMS), the biological compatibility of the completed GBFC has been successfully improved, which provides great potential for powering wearable or implantable devices.

Abstract:A non-enzymatic glucose biofuel cell (GBFCs) with high-power density and adequate open-circuit potential in physiological environment is based on the improved hydrogen template electrodeposition of the platinum-nickel (PtNi) nanoalloys (the third step adopted a potential-step electrochemical synthesis instead of the conventional method). The morphology tests demonstrate that the highly porous PtxNi1-x nanoalloys exhibit broader pore-size distribution and larger specific electrochemically active surface area than the Pt monometallic nanostructures. Combined with cyclic voltammograms, polarization parameters and cell tests, degradation behavior measurements prove that the nanoalloy products display the excellent long-term stability and high electrocatalytic activity. Controlling the preparation conditions of the highly porous PtxNi1-x nanoalloys could control the morphology and nanostructure of the as-synthesis nanoalloys in order to improve the catalytic performance of these nanoalloys, which grants it great potentialities for controllable synthesis of electrocatalysts in the application of GBFCs.Graphical abstractDescriptionThe highlyporous platinum nickel nanoalloys are applied as bothanode and cathode in the non-enzymatic glucose biofuel cell withhigh-power density and adequate open-circuit potential inphysiological environment.

Abstract:This study demonstrated a novel nanographene platelets (NGPs)-based glucose/O2 biofuel cell (BFC) with the glucose oxidase (GOD) as the anodic biocatalysts and the laccase as the cathodic biocatalysts. The GOD/NGPs-modified electrode exhibited good catalytic activity towards glucose oxidation and the laccase/NGPs-modified electrode exhibited good catalytic activity towards O2 electroreduction. The maximum power density was ca. 57.8μWcm−2 for the assembled glucose/O2 NGPs-based BFC. These results indicated that the NGPs were very useful for the future development of novel carbon-based nanomaterials BFC device.

Abstract:Rhodium single-atom nanozymes (Rh SANs), mimicking the function of natural glucose oxidase (GOD) for both biometabolism and electrometabolism of glucose oxidation, were designed in this work. Here, Rh SANs were proved to have the same biocatalytic mechanism as natural GOD, as well as a similar electrocatalytic path, namely two-electron electrooxidation from glucose to gluconic acid. On this basis, by coupling a bilirubin oxidase (BOD) biocathode, the Rh SANs-based glucose/O2 enzymatic biofuel cell (EBFC) was developed, and a maximum power density of 135.0 ± 3.0 μW cm–2 was obtained, which was nearly 2-fold enhanced over that of natural GOD-based EBFC. In addition to gathering fuels from soft drinks available in daily life, this EBFC could be designed as a novel, low-cost, and miniaturized bio-energy device. With an ingenious catalyst and flexible configuration, the device not only accelerates the development of nanozymes but paves avenues for powering portable and wearable devices.

Abstract:This study demonstrates a new kind of miniature glucose/O2 biofuel cells (BFCs) based on carbon fiber microelectrodes (CFMEs) modified with single-walled carbon nanotubes (SWNTs). SWNTs are used as a support both for stably confining the electrocatalyst (i.e., methylene green, MG) for the oxidation of NADH and the anodic biocatalyst (i.e., NAD+-dependent glucose dehydrogenase, GDH) for the oxidation of glucose and for efficiently facilitating direct electrochemistry of the cathodic biocatalyst (i.e., laccase) for the O2 reduction. The prepared micro-sized GDH-based bioanode and laccase-based biocathode exhibit good bioelectrocatalytic activity toward the oxidation of glucose and the reduction of oxygen, respectively. In 0.10M phosphate buffer containing 10mM NAD+ and 45mM glucose under ambient air, the power density of the assembled miniature compartment-less glucose/O2 BFC reaches 58μWcm−2 at 0.40V. The stability of the miniature glucose/O2 BFC is also evaluated.

Abstract:Achieving efficient and reasonable renewable energy utilization is crucial for sustainable socio-economy development. Herein, an ingenious glucose/O2 bio-photoelectrochemical system (BPECS) is developed to enhance glucose metabolism process for high-efficiency biofuels utilization. Capturing the intermittent nature of sunlight, glucose metabolism is divided into two-step reasonably. Under dark condition, bio-metabolism catalyzed by glucose oxidase (GOD), is conducted firstly to enrich gluconic acid and H2O2. Benefiting from specific electrocatalytic selectivity of BiVO4 photoanode and hemin cathode towards gluconic acid oxidation and H2O2 reduction, respectively, electro-matabolism occurs subsequently under light illumination. Moreover, profiting from prominently catalytic stability, a nanozyme-enhanced BPECS is constructed for the first time by integrating GOD-like Au nanoparticles, and a maximum power density of 139.17 μW cm−2 with open-circuit voltage of 0.62 V, is obtained. Such a BPECS provides a guiding significance for exploration of renewable energy sources and opens up a new path to expand nanozymes in extensive research fields.

Abstract:Abstract Presently, one of the most important aspects for the development of enzymatic biofuel cells (EBFCs) is to synthesize the novel electrode materials that possess high current density, low open-circuit voltage (OCV) and long-term stability. To achieve the above attributes, lots of new strategies are being used by the researchers for the development of advanced materials. Nowadays, nanomaterials and nanocomposites are the promising material that has been utilized as effective electrode material in solar cells, supercapacitors and biofuel cells application. Herein, we account for a novel electrocatalyst as electrode material that comprised ZnO nanoparticles decorated on the surface of polyindole (PIn)-multi-walled carbon nanotube (MWCNT), for the immobilization of glucose oxidase (GOx) enzyme and mediator (Ferritin). The PIn-MWCNT scaffold is prepared via in situ chemical oxidative polymerization of indole on the surface of MWCNT and assessed by myriad techniques. The micrograph of scanning electron microscopy (SEM) designated the interconnected morphology of MWCNTs in the polymer matrix. X-ray diffraction spectroscopy (XRD) and Fourier transform infrared spectroscopy (FTIR), confirm the crystallinity and different functional groups available in the synthesized material, respectively. The electrochemical assessment demonstrates that the ZnO/PIn-MWCNT/Frt/GOx nanobiocatalyst exhibits much higher electrocatalytic activity towards the oxidation of glucose with a maximum current density of 4.9 mA cm −2 by consuming 50 mM glucose concentration in phosphate buffer saline (PBS) (pH 7.4) as the testing solution by applying 100 mVs −1 scan rates. The outcomes reflect that the as-prepared ZnO/PIn-MWCNTs/Frt/GOx biocomposite is a promising bioanode for the development of EBFCs.

Abstract:3,4,9,10‐perylenetetracarboxylic dianhydride (PD), with a highly conjugated structure and high electron mobility, is discovered to exhibit both glucose oxidase‐like and catalase‐like activities. A self‐cascade catalytic system based on the dual enzyme activities of PD is constructed for application in the anode of biofuel cells (BFCs), which can consume H2O2 in situ and effectively improve the performance of BFCs. Carbon‐based nanozymes have received considerable attention due to their superior biosafety, enhanced tolerance to extreme conditions, and ease of chemical modification. However, due to limited material diversity and unconfirmed molecular structure, carbon‐based nanozymes face challenges such as relatively low enzyme activity and unclear catalytic mechanisms. The development of materials with well‐defined structures and controllable properties is crucial for promoting the rapid progress of nanozymes. Herein, 3,4,9,10‐perylenetetracarboxylic dianhydride (PD) exhibits both glucose oxidase (GOx)‐like and catalase (CAT)‐like activities, which may be due to the fact that PD possesses the features of highly conjugated structure and high electron mobility. In addition, it is demonstrated that the enzymatic activity is related to the degree of PD aggregation via the characterization of its morphology and size. Based on the excellent GOx‐like and CAT‐like activities of PD, a self‐cascade catalytic system is constructed for application in biofuel cells (BFCs). It is worth mentioning that such BFC still maintains high stability after working for 30 days. Therefore, this study expands the enzyme‐like systems and discovers that nanomaterials with highly conjugated structures and high electron mobility can mimic enzymes. Additionally, the multi‐enzyme activities are utilized to construct self‐cascade systems, which can effectively improve the performance of BFCs.

Abstract:This study demonstrates a novel carbon nanotubes-hydroxyapatite (CNTs–HA) nanocomposite-based compartment-less glucose/O2 biofuel cell (BFC) with the glucose oxidase (GOD) as the anodic biocatalysts and the laccase as the cathodic biocatalysts. CNTs–HA nanocomposite prepared by the self-assembly method via an aqueous solution reaction has been used as the co-immobilization matrix to incorporate biocatalysts, i.e. GOD and laccase successfully. Moreover, the three-dimensional configuration of the CNTs–HA films electrode would be advantageous to the glucose oxidation on the bioanode and O2 electroreduction on the biocathode of BFC. The maximum power density delivered by the assembled glucose/O2 BFC could reach 15.8μWcm−2 at a cell voltage of 0.28V with 10mM glucose. The results indicate that the CNTs–HA nanocomposite is believed to be very useful for the development of novel BFC device.

Abstract:Here, an electron transfer enhanced redox mediator is presented that significantly enhances electron transfer between the host electrode and enzymes, as well as between adjacent enzymes, promoting seamless interfaces through multiple interactions. Biofuel cells (BFCs) offer an eco‐friendly route to convert biochemical energy into electricity. However, their performance is hindered by insufficient enzyme immobilization as well as limited electron transfer within the enzymatic electrode. While the incorporation of redox mediators (RMs) into enzyme layers has been shown to improve BFC performance through enhanced electron transfer, progress has plateaued in the last decade. Herein, a major breakthrough is presented realized by a novel strategy that exploits electron transfer‐intensified RM layers. Metal nanoparticles covalently bridged between neighboring RMs facilitate electron transfer ubiquitously. Electron transfer characteristics are enhanced not only within the RM layers themselves, but also at the glucose oxidase (GOx)/host electrode and GOx/GOx interfaces. This leads to a remarkable performance boost in the enzymatic anode. A hybrid BFC constructed with innovative anode and Pt‐based cathode exhibits a striking combination of high power output (2.3 and 8.5 mW cm−2 at 10 and 300 mmol L−1 glucose, respectively) and exceptional operational stability (≈80% and 47% power retention after 10 days and 1 month, respectively), outperforming all previously reported BFCs by a significant margin.

Abstract:Constructing an integrated system with high-efficiency and stable energy conversion and storage (ECS) is of great significance but remains challenges. Herein, we report a biodevice consisting of a bioanode of tetrathiafulvalene/glucose oxidase (TTF/GOx) immobilized on asphalt-derived porous carbon (APC) for catalyzing glucose oxidation and a biocathode of 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonate)/bilirubin oxidase (ABTS/BOD) immobilized on APC for catalyzing oxygen reduction , to achieve ECS with high-efficiency and excellent stability. The redox mediators , TTF and ABTS, used at the bioanode and biocathode, respectively, simultaneously mediate electron transfer to release electric current on discharging and serve as pseudocapacitors to achieve charge storage at open-circuit conditions, enabling high power density and self-charge capability. The biodevice of the above configuration delivers a peak power output of 5.01 mW cm −2 in the pulse discharge mode with cell voltage remaining 93.7 % of its initial value after 100 pulse discharge/self-charge cycles. This work provides a highly efficient solution to address the low power density and poor stability of the conventional enzymatic biofuel cells, showing great implications for small, implantable and wearable electronics.

Construction of a Cascade Catalyst of Nanocoupled Living Red Blood Cells for Implantable Biofuel Cell

Abstract:The broad applications of implantable glucose biofuel cells (GBFCs) have become very attractive in biomedical sciences. The key challenge of GBFCs is eliminating the inevitable product H2O2 generated from the oxidation of glucose when glucose oxidase (GOx) is used as a catalyst while improving the performance of GBFCs. In this work, the cascade electrocatalyst, was obtained through the in situ polymerization of dopamine to form nanopolydopamine (NPDA) on the surface of red blood cells (RBCs). The can catalyze both fuels of H2O2 and O2, so as to generate a high cathodic current (0.414 mA cm–2). Furthermore, when was used as a cathodic catalyst in the membraneless GBFC, it exhibited the cascade catalytic activity in the reduction of O2–H2O2 and minimized the damage to RBCs caused by the high concentration of H2O2. The mechanism research indicates that integrates the property of NPDA and RBCs. Specifically, NPDA plays a catalase-like role in catalyzing the decomposition of H2O2, while RBCs play a laccase-like role in electrocatalyzing the O2 reduction reaction. Thermal Annealing‐Enhanced Bioelectrocatalysis in Membraneless Glucose/Oxygen Biofuel Cell based on Hydrophilic Carbon Fibers

Abstract:The electrode material is a pivotal component in achieving efficient electron transfer in an enzymatic biofuel cell (EBFC). Herein, novel hydrophilic carbon fibers (HCFs) with a helical structure are synthesized through chemical vapor deposition and are used as an electrode material for the immobilization of biocatalysts to fabricate bioelectrodes, where glucose dehydrogenase, serving as the anodic enzyme, is co‐immobilized with 1,4‐naphthoquinone on the HCFs for the catalysis of the glucose oxidation reaction (GOR), and bilirubin oxidase, as the cathodic enzyme, is co‐immobilized with protoporphyrin IX for the catalysis of the oxygen reduction reaction (ORR). It is found that thermal annealing the HCFs improves the bioelectrocatalytic performances of the as‐fabricated bioanode for the GOR and biocathode for the ORR, owing to the enhanced electron transfer kinetics. Subsequently, a membraneless glucose/O2 EBFC assembled by using the as‐synthesized bioelectrodes is able to generate a high open circuit voltage of 0.8 V and a maximum power density of 0.7 mW cm−2 at 0.44 V with good operational stability. The practicability in fabricating membraneless EBFCs makes the synthesized HCFs promising in miniature bioenergy devices. Trying to cell it: Hydrophilic carbon fibers with a helical architecture are synthesized to immobilize redox enzymes in a membraneless glucose/O2 biofuel cell. The enhanced electron transfer kinetics and the hydrophilic property show affinity toward glucose oxidation and oxygen reduction reactions, resulting in superior cell performance.

Abstract:In this paper, we present a neutral non-enzymatic glucose biofuel cell (GBFC) based on a Pt/Au nano-alloy anode. We synthesize Pt/Au alloy anodes by the electrochemical deposition of a Pt/Au nano-alloy on the surface of a thin polyethylene terephthalate membrane with a uniformly distributed microhemispheric array. The experimental results demonstrate that the proposed GBFC can generate a maximum power density of 0.32 mW/cm2, an open-circuit voltage (Voc) of 0.42 V, and a short-circuit current (Isc) density of 2.67 mA/cm2 with 5 mM of glucose in a 0.1-M PBS (pH 7.4) solution while the cathode is being supplied with 7% saturated oxygen. The synergistic effect of the Pt/Au bimetallic alloy is found to improve the performance of the proposed GBFC. The proposed neutral non-enzymatic GBFC shows promising feasibility as an implantable power supply system for the provision of continuous and consistent electricity to a continuous glucose monitoring system.
