Abstract:Background: Plant height is an important architecture trait which is a fundamental yield-determining trait in crops. Variety with dwarf or semi-dwarf phenotype is a major objective in the breeding because dwarfing architecture can help to increase harvest index, increase planting density, enhance lodging resistance, and thus be suitable for mechanization harvest. Although some germplasm or genes associated with dwarfing plant type have been carried out. The molecular mechanisms underlying dwarfism in oilseed rape (Brassica napus L.) are poorly understood, restricting the progress of breeding dwarf varieties in this species. Here, we report a new dwarf mutant Bndwarf2 from our B. napus germplasm. We studied its inheritance and mapped the dwarf locus BnDWARF2. Results: The inheritance analysis showed that the dwarfism phenotype was controlled by one semi-dominant gene, which was mapped in an interval of 787.88 kb on the C04 chromosome of B. napus by Illumina Brassica 60 K Bead Chip Array. To fine-map BnDWARF2, 318 simple sequence repeat (SSR) primers were designed to uniformly cover the mapping interval. Among them, 15 polymorphic primers that narrowed down the BnDWARF2 locus to 34.62 kb were detected using a F2:3 family population with 889 individuals. Protein sequence analysis showed that only BnaC04.BIL1 (BnaC04g41660D) had two amino acid residues substitutions (Thr187Ser and Gln399His) between ZS11 and Bndwarf2, which encoding a GLYCOGEN SYNTHASE KINASE 3 (GSK3-like). The quantitative real-time PCR (qRT-PCR) analysis showed that the BnaC04.BIL1 gene expressed in all tissues of oilseed rape. Subcellular localization experiment showed that BnaC04.BIL1 was localized in the nucleus in tobacco leaf cells. Genetic transformation experiments confirmed that the BnaC04.BIL1 is responsible for the plant dwarf phenotype in the Bndwarf2 mutants. Overexpression of BnaC04.BIL1 reduced plant height, but also resulted in compact plant architecture. Conclusions: A dominant dwarfing gene, BnaC04.BIL1, encodes an GSK3-like that negatively regulates plant height, was mapped and isolated. Our identification of a distinct gene locus may help to improve lodging resistance in oilseed rape.
A BIN2-like1 Protein Confers Dwarf by Interacting with BZR1 of Brassinosteroid Signaling in Allotetraploid Brassica Napus

Abstract:Abstract Brassinosteroids (BRs) are steroid hormones that play essential roles in plant growth and development. In this study, we identified a new dwarf mutant in Brassica napus. By map-based cloning, BnaC04.BIL1 (BnaC04g41660D) gene, a BIN2-like1 (BIL1) encoding a GLYCOGEN SYNTHASE KINASE 3 (GSK3-like) protein kinase, was isolated. To date, how BIL1 involves in BR signal transduction remains uncovered. Genetic transformation experiments confirmed that the BnaC04.BIL1 is responsible for the plant dwarf phenotype in the Bndwarf2 mutants. Overexpression of BnaC04.BIL1 not only reduced plant height, but also resulted in compact plant architecture. Using CRISPR/Cas9, two sgRNAs were designed to target BnaC04.BIL1 gene. The gene editing experiments generated mutations of BnaC04.BIL1 sequence, which were stably transmitted to successive generations, and lead to restoration of plant height and plant architecture. The molecular mechanism of Bndwarf2 dwarfing was further verified by Y2H and BiFC assays. Results shown that a Thr187Ser amino acid substitution residing in the conserved region promotes the interaction between BnaC04.BIL1Mut with BnaBZR1, thus enhances the negative regulation of plant growth. The genetic and molecular evidence clarifies first the BnaC04.BIL1 can sharply change plant architecture in natural plant accessions in allotetraploid, and provides new insights into the molecular mechanisms of BR signaling.

Abstract:Plant architecture is the determining factor of yield in many crops. Rapeseed (Brassica napus) is an important oilseed crop worldwide, but the genetic basis underlying plant architecture in this species remains elusive. Here, we characterized the dwarf mutant dwarf and compact 1 (dac1) from an ethyl methanesulfonate (EMS) mutagenized population of rapeseed cv. Zhongshuang 11 (ZS11). dac1 has multiple morphological defects, such as dwarfism, narrow branch angles, delayed flowering, and dark-green, crinkled leaves. We identified the underlying gene, which encodes a glycogen synthase kinase 3 (GSK3)-like protein homologous to Arabidopsis thaliana BRASSINOSTEROID-INSENSITIVE 2 (BIN2)-LIKE 1 (BIL1), and named this gene BnaC04. BIL1. A C-to-T mutation in BnaC04. BIL1 changes proline at position 314 of the encoded protein to leucine (P314L), affecting the degradation of BnaC04. BIL1, and which represses plant architecture in a dosage-dependent manner via brassinosteroid signaling. We identified an 11-bp core sequence and one pivotal base in the promoter region of BnaC04. BIL1 that affect its expression. Importantly, heterozygotes of dac1 displayed single-locus overdominance, resembling a proposed ideotype for rapeseed. Our findings demonstrate that a gain-of-function mutation in BnaC04. BIL1 contributes to ideal plant architecture, representing a valuable resource for rapeseed breeding.
Cloning and functional analysis of BnMAPK2 gene in Brassica napus

Abstract:: The mitogen-activated protein kinase (MAPK) cascade is involved in plant growth and development and it is in response to a variety of biotic and abiotic stresses. In this study, a BnMAPK2 (BnaC01g28210D) gene was isolated and cloned from Brassica napus . The cDNA and its coding sequence were 1516 bp and 1113 bp in length, respectively, encoding 371 amino acids. Bioinformatics analysis revealed that the molecular weight of BnMAPK2 protein was 42,497.0 kD, the isoelectric point was 6.36, protein instability coefficient was 38.74, it was a hydrophobic protein, and it had STKc_TEY_MAPK_ plant (cd07858) conserved structure domain unique to MAPKs protein, protein secondary level. The alpha helix accounted for the largest proportion of 44.05% in the secondary structure of protein, and there was no signal peptide, which was more closely related to the C group AtMAPK2 of Arabidopsis . The core element prediction indicated that BnMAPK2-P contained related cis-acting elements in response to salicylic acid hormone, heat stress, and light, including TCA-element, HSE, AAAC-motif, and MYB binding sites. Real-time quantitative PCR (qRT-PCR) demonstrated that BnMAPK2 was expressed in various tissues and organs in Brassica napus , which was induced by methyl jasmonate, salicylic acid, H 2 O 2 , injury, high temperature, and Sclerotinia sclerotiorum. The phenotypic data of transgenic Arabidopsis lines expressing BnMAPK2 heterologously showed that compared with the wild type, the overexpression of BnMAPK2 made the bolting period of Arabidopsis plants earlier, and significantly increased plant height, the effective length of main inflorescence, and the number of siliques. Abstract:Key message A novel mutation in the BnaA03.IAA7 protein reduces plant height and enhances gibberellin signaling in Brassica napus L. Abstract Rapeseed ( Brassica napus ) is an excellent and important source for vegetable oil production, but its production is severely affected by lodging. Lodging hinders mechanization and decreases yield, and an ideal solution is semidwarf breeding. Limited by germplasm resources, semidwarf breeding developed slowly in rapeseed. In the current study, a mutant called sdA03 was isolated from EMS-mutagenized lines of Zhongshuang 11 (ZS11). The inheritance analysis showed that phenotypes of sdA03 were controlled by a single semidominant gene. Genetic mapping, RNA-seq and candidate gene analysis identified BnaA03.IAA7 as a candidate gene, and a function test confirmed that the mutated BnaA03.iaa7 regulates plant architecture in a dose-dependent manner. Yeast two-hybrid and transient expression experiments illustrated the P87L substitution in the GWPPV/I degron motif of BnaA03.iaa7 impaired the interaction between BnaA03.IAA7 and TIR1 proteins, and BnaA03.iaa7 prevented ARF from activating the auxin signaling pathway.The gibberellin (GA) content was higher in sdA03 hypocotyls than in those of ZS11. Further expression analysis showed more active gibberellin signaling in hypocotyl and richer expression of GA synthetic genes in root and cotyledon of sdA03 seedlings. Finally, a marker was developed based on the SNP found in BnaA03.iaa7 and used in molecular breeding. The study enriched our understanding of the architectural regulation of rapeseed and provided germplasm resources for breeding.
Identification and Cloning of GOLDEN2-LIKE1 (GLK1), a Transcription Factor Associated with Chloroplast Development in Brassica Napus L.

Abstract:Photosynthesis is the process by which dry matter accumulates, which affects rapeseed yield. In this study, we identified GOLDEN2-LIKE1 (GLK1), located on chromosome A07 and 59.2 kb away from the single nucleotide polymorphism marker SNP16353A07, which encodes a transcription factor associated with the rate of photosynthesis in leaves. We then identified 96 GLK1 family members from 53 species using a hidden Markov model (HMM) search and found 24 of these genes, which were derived from 17 Brassicaceae species. Phylogenetic analysis showed that 24 Brassicaceae proteins were classified into three subgroups, named the Brassica family, Adenium arabicum, and Arabidopsis. Using homologous cloning methods, we identified four BnaGLK1 copies; however, the coding sequences were shorter than the putative sequences from the reference genome, probably due to splicing errors among the reference genome sequence or different gene copies being present in the different B. napus lines. In addition, we found that BnaGLK1 genes were expressed at higher levels in leaves with more chloroplasts than were present in other leaves. Overexpression of BnaGLK1a resulted in darker leaves and siliques than observed in the control, suggesting that BnaGLK1 might promote chloroplast development to affect the rate of photosynthesis in leaves. These results will help to elucidate the mechanism of chloroplast biogenesis by GLK1 in B. napus.
Genome-Wide Association Mapping Reveals the Genetic Control Underlying Branch Angle in Rapeseed (brassica Napus L.).

Abstract:Plant architecture is vital not only for crop yield, but also for field management, such as mechanical harvesting. The branch angle is one of the key factors determining plant architecture. With the aim of revealing the genetic control underlying branch angle in rapeseed (Brassica napus L.), the positional variation of branch angles on individual plants was evaluated, and the branch angle increased with the elevation of branch position. Furthermore, three middle branches of individual plants were selected to measure the branch angle because they exhibited the most representative phenotypic values. An association panel with 472 diverse accessions was estimated for branch angle trait in six environments and genotyped with a 60K Brassica Infinium® SNP array. As a result of association mapping, 46 and 38 significantly-associated loci were detected using a mixed linear model (MLM) and a multi-locus random-SNP-effect mixed linear model (MRMLM), which explained up to 62.2 and 66.2% of the cumulative phenotypic variation, respectively. Numerous highly-promising candidate genes were identified by annotating against Arabidopsis thaliana homologous, including some first found in rapeseed, such as TAC1, SGR1, SGR3, and SGR5. These findings reveal the genetic control underlying branch angle and provide insight into genetic improvements that are possible in the plant architecture of rapeseed.

Abstract:Enhancing photosynthetic efficiency is a major goal to improve crop yields under agricultural field conditions and is associated with chloroplast biosynthesis and development. In this study, we demonstrate that GOLDEN2-LIKE 1a (BnGLK1a) plays an important role in the regulating chloroplast development and photosynthetic efficiency. Overexpressing BnGLK1a resulted in significant increases in chlorophyll content, the number of thylakoid membrane layers and photosynthetic efficiency in B. napus, while knocking down BnGLK1a transcript levels through RNA interference (RNAi) had the opposite effects. A yeast two-hybrid screen revealed that BnGLK1a interacts with the abscisic acid receptor PYRABACTIN RESISTANCE 1-LIKE 1-2 (BnPYL1-2) and CONSTITUTIVE PHOTOMORPHOGENIC 9 SIGNALOSOME 5A subunit (BnCSN5A), which play essential roles in regulating chloroplast development and photosynthesis. In agreement with this, BnGLK1a-RNAi lines of B. napus display hypersensitivity to ABA response. Importantly, overexpression of BnGLK1a resulted in a 10% increase in thousand-seed weight, whereas seeds from BnGLK1a-RNAi lines were 16% lighter than wild type. We propose that BnGLK1a could be a potential target in breeding for improving rapeseed productivity. Our results not only provide the insight into the mechanisms of BnGLK1a function, but also offer a potential approach to improving the productivity of Brassica species.
Genome-wide Identification and Characterization of SnRK Family Genes in Brassica Napus

Abstract:Background Sucrose non-fermenting 1 related protein kinases ( SnRK) play crucial roles in responding to biotic and abiotic stresses through activating protein phosphorylation pathways. However, little information of SnRK genes was available in Brassica napus , one of important oil crops. Recently, the released sequences of the reference genome of B.napus provide a good chance to perform genome-wide identification and characterization of BnSnRK gene family in the rapeseed. Results Totally 114 SnRK genes distributed on 19 chromosomes were identified in the genome of B.napus and classified into three subfamilies on the basis of phylogenetic analysis and the domain types. According to gene structure and motif composition analysis, the BnSnRK sequences showed obvious divergence among three subfamilies. Gene duplication and synteny between the genomes of the rapeseed and Arabidopsis were also analyzed to provide insights into the evolutionary characteristics of BnSnRK family genes. C is- element analysis revealed that BnSnRKs may response to diverse environmental stresses. Moreover, the expression patterns of BnSnRKs in various tissues and under diverse abiotic stresses were distinct difference. Besides, Single Nucleotide Polymorphisms (SNP) distribution analysis suggests the function disparity of BnSnRK family genes in different genotypes of the rapeseed. Conclusion We examined genomic structures, evolution features, expression patterns and SNP distribution of 114 BnSnRKs . The results provide valuable information for functional characterization of BnSnRK genes in future studies.
Integrating GWAS, linkage mapping and gene expression analyses reveal the genetic control of first branch height in Brassica napus L

Abstract:Rapeseed (Brassica napus L.) is a crucial oil crop cultivated worldwide. First branch height, an essential component of rapeseed plant architecture, has an important effect on yield and mechanized harvesting; however, the underlying genetic mechanism remains unclear. In this study, based on the 60K single nucleotide polymorphism array and a recombinant inbred lines population derived from M083 and 888-5, a total of 19 QTLs were detected in five environments, distributed on linkage groups A02, A09, A10, C06, and C07, which explained phenotypic variation ranging from 4.87 to 29.87%. Furthermore, 26 significant SNPs were discovered on Chr.A02 by genome-wide association study in a diversity panel of 324 re-sequencing accessions. The major QTL of the first branch height trait was co-located on Chr.A02 by integrating linkage mapping and association mapping. Eleven candidate genes were screened via allelic variation analysis, inter-subgenomic synteny analysis, and differential expression of genes in parental shoot apical meristem tissues. Among these genes, BnaA02g13010D, which encodes a TCP transcription factor, was confirmed as the target gene according to gene function annotation, haplotype analysis, and full-length gene sequencing, which revealed that TATA insertion/deletion in the promoter region was closely linked to significantly phenotypic differences BnaA02.TCP1 M083 overexpression resulted in decreased branch height and increased branch number in Arabidopsis. These results provide a genetic basis for first branch height and the ideal architecture of B. napus.

Abstract:Abstract Background Plant height is an important plant characteristic closely related to yield performance of many crops. Reasonable reduction of plant height of crops is beneficial for improving yield and enhancing lodging resistance. Results In the present study, we described the Brassica napus dwarf mutant bnd2 that was isolated using ethyl methanesulfonate (EMS) mutagenesis. Compared to wild type (WT), bnd2 exhibited reduced height and shorter hypocotyl and petiole leaves. By crossing the bnd2 mutant with the WT strain, we found that the ratio of the mutant to the WT in the F 2 population was close to 1:3, indicating that bnd2 is a recessive mutation of a single locus. Following bulked segregant analysis (BSA) by resequencing, BND2 was found to be located in the 13.77–18.08 Mb interval of chromosome A08, with a length of 4.31 Mb. After fine mapping with single nucleotide polymorphism (SNP) and insertion/deletion (InDel) markers, the gene was narrowed to a 140-Kb interval ranging from 15.62 Mb to 15.76 Mb. According to reference genome annotation, there were 27 genes in the interval, of which BnaA08g20960D had an SNP type variation in the intron between the mutant and its parent, which may be the candidate gene corresponding to BND2 . The hybrid line derived from a cross between the mutant bnd2 and the commercial cultivar L329 had similar plant height but higher grain yield compared to the commercial cultivar, suggesting that the allele bnd2 is beneficial for hybrid breeding of lodging resistant and high yield rapeseed. Conclusion In this study, we identified a novel dwarf mutant of rapeseed with a new locus, which may be useful for functional analyses of genetic mechanisms of plant architecture and grain yield in rapeseed.

Abstract:Glucosinolates are secondary metabolites found mainly in Cruciferae plants.The CYP79 family,belongs to cytochromes P450,had been shown to catalyze the conversion of amino acids to their corresponding oximes in the biosynthesis of cyanogenic glucosides.In the present study,one of the glucosinolates regulator genes,named as BnCYP79B1(GenBank JX535391.1)was cloned by homologous cloning from three stains of Brassica napus.The cDNA sequence of its open reading frame(ORF)was as long as 1 625bp,encoding aputative protein of 541 amino acid residues with a theoretical isoelectric point of 8.88.Sequence comparison with Brassica oleracea var.botrytis CYP79B1 showed that,BnCYP79B1gene had 98.83%similarity of nucleotide sequence and 99.26% similarity of the amino acid sequence.Three different lines,w678(high-stemed and high glucosinolates content in seed),w673(high-stemed and low glucosinolates content in seed),and w578(dwarf mutant with high glucosinolates content in seed),were estimated for BnCYP79B1 expression by real-time quantitative PCR analysis.The results showed that:BnCYP79B1 gene mainly expressed in roots of w678and in leaves of w578,but with the highest expression of BnCYP79B1 in w678.In conclusion,wild-type line had higher BnCYP79B1 expression level than the dwarf mutant,and also the high glucosinolates content line had rather higher BnCYP79B1 expression level than the low glucosinolates content line.
A gain-of-function mutation in BnaIAA13 disrupts vascular tissue and lateral root development in Brassica napus

Abstract:Rapeseed ( Brassica napus ) is an important oilseed crop worldwide. Plant vascular tissues are responsible for material transport and provide mechanical support. The lateral roots (LRs) absorb sufficient water and nutrients. The genetic basis of vascular tissues and LRs development in rapeseed remains unknown. This study characterized an EMS-mutagenized rapeseed mutant, T16 , which showed dwarf stature, reduced LRs, and leaf wilting. Scanning electron microscopy observations showed that the internode-cell shortened. Observations of the tissue sections revealed defects in the development of vascular bundles in the stems and petioles. Genetic analysis revealed that the phenotypes of T16 were controlled by a single semi-dominant nuclear gene. Map-based cloning and genetic complementarity confirmed that BnaA03.IAA13 is the functional gene, a G-to-A mutation in second exon changed the glycine at the 79 th position to glutamic acid, disrupting the conserved degron motif VGWPP. Transcriptome analysis in roots and stems showed that auxin and cytokinin signaling pathways were disordered in T16 . Evolutionary analysis showed that AUXIN/INDOLE‐3‐ACETIC ACID was conserved during plant evolution. The heterozygote of T16 significantly reduced the plant height while maintaining other agronomic traits. Our findings provide novel insights into the regulatory mechanisms of vascular tissues and LRs development, and provide a new germplasm resource for rapeseed breeding.

Abstract:The leucine-rich repeat receptor-like protein kinase ( LRR-RLK ) family represents the largest group of RLKs in plants and plays vital roles in plant growth, development and the responses to environmental stress. Although LRR-RLK families have been identified in many species, they have not yet been reported in B. napus . In this study, a total of 444 BnLRR-RLK genes were identified in the genome of Brassica napus cultivar “Zhongshuang 11” (ZS11), and classified into 22 subfamilies based on phylogenetic relationships and genome-wide analyses. Conserved motifs and gene structures were shared within but not between subfamilies. The 444 BnLRR-RLK genes were asymmetrically distributed on 19 chromosomes and exhibited specific expression profiles in different tissues and in response to stress. We identified six BnBRI1 homologs and obtained partial knockouts via CRISPR/Cas9 technology, generating semi-dwarf lines without decreased yield compared with controls. This study provides comprehensive insight of the LRR-RLK family in B. napus . Additionally, the semi-dwarf lines expand the “ideotype” germplasm resources and accelerate the breeding process for B. napus .

Genome Analysis of BnCNGC Gene Family and Function Exploration of BnCNGC57 in Brassica Napus L.

Abstract:The cyclic nucleotide-gated ion channel (CNGC), as a non-selective cation channel, plays a pivotal role in plant growth and stress response. A systematic analysis and identification of the BnCNGC gene family in Brassica napus is crucial for uncovering its biological functions and potential applications in plant science. In this study, we identified 61 BnCNGC members in the B. napus genome, which are phylogenetically similar to Arabidopsis and can be classified into Groups I-IV (with Group IV further subdivided into IV-a and IV-b). Collinearity analysis with other species provided insights into the evolution of BnCNGC. By homology modeling, we predicted the three-dimensional structure of BnCNGC proteins and analyzed cis-acting elements in their promoters, revealing diverse roles in hormone regulation, growth, and stress response. Notably, overexpression of BnCNGC57 (BnaC09g42460D) significantly increased seed size, possibly through regulating cell proliferation via the MAPK signaling pathway. Our findings contribute to a better understanding of the BnCNGC gene family and highlight the potential regulatory role of BnCNGC57 in the seed development of B. napus.
Homologous Cloning of BnGS3 and BnGhd7 Genes in Brassica Napus and Their Relationship with Rapeseed Yield-related Traits

Abstract:Brassica napus is an allotetraploid with complex genomic structure,but Brassica genome has a collinearity with that of rice. In this study, the homologous genes BnGS3 and BnGhd7 of Brassica napus were obtained by homologous cloning of the yield related genes GS3 and Ghd7 of rice.BnGS3 has six exons with a 666 bp of ORF and encodes 222 amino acids.BnGS3 pro-tein has VWF structure,one of the four conserved domains of rice GS3,and belongs to type A.BnGhd7 contains one exon having ORF with a full-length of 1014 bp and encoding 337 amino acids. BnGhd7 protein has two important domains, the N-terminal B-Box and the C-terminal CCT. BnGS3 and BnGhd7 were located in linkage groups A2 and A10, respectively. Polymorphism markers brgs-16 of BnGS3 and polymorphism markers brghd-3 and ghd7-7 of BnGhd7 were obtained by comparative sequencing, among which brghd-3 with thousand kernel weight (P < 0.05) and ghd7-7 with plant height (P <0.01) were positively correlated, and ghd7-7 was negatively correlated with flowering stage (P < 0.05). The results indicate that it is feasible to clone the homolo-gous genes of Brassica napus using rice functional gene sequence information,which provides an effective way in Brassica napus functional gene research.
Natural Variation and Artificial Selection at the BnaC2.MYB28 Locus Modulate Brassica Napus Seed Glucosinolate.

Abstract:The degradation products of glucosinolates (GSLs) greatly lower the nutritional value of rapeseed (Brassica napus) meal; thus, reduction of seed GSL content (SGC) has become an important objective of rapeseed breeding. In our previous study, we finely mapped a major QTL (qGSL-C2) for SGC to a 49-kb collinear region on B. rapa chromosome A2. Here, we experimentally validated that BnaC2.MYB28, encoding an R2R3-MYB transcription factor, is the causal gene of qGSL-C2. BnaC2.MYB28 is a nucleus-localized protein mainly expressed in vegetative tissues. Knockout of BnaC2.MYB28 in the high-SGC parent G120 reduced SGC to a value lower than that in the low-SGC parent ZY50, while overexpression of BnaC2.MYB28 in both parental lines (G120 and ZY50) led to extremely high SGC, indicating that BnaC2.MYB28 acts as a positive regulator of SGC in both parents. Molecular characterization revealed that BnaC2.MYB28 forms a homodimer and specifically interacts with BnaMYC3. Moreover, BnaC2.MYB28 can directly activate the expression of GSL biosynthesis genes. Differential expression abundance resulting from the polymorphic promoter sequences, in combination with the different capability in activating downstream genes involved in aliphatic GSL biosynthesis, caused the functional divergence of BnaC2.MYB28 in SGC regulation between the parents. Natural variation of BnaC2.MYB28 was highly associated with SGC in natural germplasm and has undergone artificial selection in modern low-GSL breeding. This study provides important insights into the core function of BnaC2.MYB28 in regulating SGC and a promising strategy for manipulating SGC in rapeseed.
Cloning and expression analysis of a G protein β subunit gene in Brassica napus L.

Abstract:A full-length cDNA encoding a heterotrimeric GTP-binding proteinβsubunit(Gβ),designated as BnAGB1,was isolated from Brassica napus L.by RT-PCR.BnAGB1 was predicted to encode a precursor protein of 378 amino acid residues.Comparative and bioinformatic analyses revealed that BnAGB1 showed high degrees of homology with Gβproteins from other plant species and contained conserved WD-40 motifs and motifs for contact Gαand Gγ.The expression of BnAGB1 in different B.napus tissues and stags in both the dwarf mutant and its wild-type parent was analyzed using real-time PCR.The results showed that BnAGB1 gene expression was detected in all tissues analyzed in both the dwarf mutant and its wild-type parent.Stage expression pattern analysis revealed that BnAGB1 expressed strongly at the cotyledon stage,the bolting stage and fruiting stage,weak at the two leaves stage,four leaves stage and flowering stage.The expression in dwarf mutant was significantly higher than that in the wild-type. These suggest that BnAGB1 may be involved in the regulation of growth and development of B. napus.and associated with dwarf mutant.
Brcd1 is Associated with Plant Height Through the Gibberellin Pathway in Brassica Rapa L.

Abstract:In crops such as Brassica rapa, the agronomic trait plant height affects the leaf light absorption rate, benefits substance accumulation, and increases production by increasing the biological yield of the population. The mechanism of plant height was adequately studied in Brassica napus and Arabidopsis, while Brassica rapa had been rarely reported. Herein, we studied a B. rapa dwarf mutant Brcd1(YS). Compared with its wild-type, Yellow sarson, Brcd1(YS) showed shorter hypocotyls and slow growth, with inhibited root elongation and reduced numbers of lateral roots. Chlorophyll content determination and pollen viability detection indicated that the mutant's chlorophyll content was higher than that of the wild-type; however, its pollen was inactive. Cytological identification showed that the number of cells in Brcd1(YS) leaves was significantly reduced and were arranged irregularly compared with those of the wild-type. Bulked Segregant RNA sequencing combined with conventional linkage mapped the dwarf mutation Brcd1(YS) to Chromosome A06, at position 21,621,766 to 24,683,923 bp. Application of exogenous gibberellic acid (GA) partially rescued the dwarf phenotype of Brcd1(YS). GA-related genes BraA06g034110.3C and BraA06g033010.3C were identified as the most promising candidate genes. These results make a valuable contribution to our understanding of the mechanism of plant height determination in B. rapa, paving the way for further breeding of dwarf B. rapa.
The Transcription Factor BnaWRKY10 Regulates Cytokinin Dehydrogenase BnaCKX2 to Control Cytokinin Distribution and Seed Size in Brassica Napus.

Abstract:Cytokinins (CKs) are phytohormones that promote cell division and differentiation. However, the regulation of CK distribution and homeostasis in Brassica napus is poorly understood. Here, the endogenous CKs were first quantified by LC-ESI-MS/MS in rapeseed tissues and visualized by TCSn::GUS reporter lines. Interestingly, the cytokinin oxidase/dehydrogenase BnaCKX2 homologs were mainly expressed in reproductive organs. Subsequently, the quadruple mutants of the four BnaCKX2 homologs were generated. Endogenous CKs were increased in the seeds of the BnaCKX2 quadruple mutants, resulting in a significantly reduced seed size. In contrast, overexpression of BnaA9.CKX2 resulted in larger seeds, probably by delaying endosperm cellularization. Furthermore, the transcription factor BnaC6.WRKY10b, but not BnaC6.WRKY10a, positively regulated BnaA9.CKX2 expression by binding directly to its promoter region. Overexpression of BnaC6.WRKY10b rather than BnaC6.WRKY10a resulted in lower concentration of CKs and larger seeds by activating BnaA9.CKX2 expression, indicating that the functional differentiation of BnaWRKY10 homologs might have occurred during B. napus evolution or domestication. Notably, the haploid types of BnaA9.CKX2 were associated with 1000-seed weight in the natural B. napus population. Overall, the study reveals the distribution of CKs in B. napus tissues, and shows that BnaWRKY10-mediated BnaCKX2 expression is essential for seed size regulation, providing promising targets for oil crop improvement.
