Abstract:We have developed a method of introducing biological oxime ether fragments into peptides by CuI-catalyzed late-stage modification and functionalization of peptides, utilizing their acid moiety and varied 2H-azirines. As a result of its mild conditions, high atom economy, moderate yield, and excellent functional-group tolerance, the method can provide access to late-stage peptide modification and functionalization at their acid sites both in the homogeneous phase and on resins in SPPS, providing a new tool kit for peptide functionalization, diversification, and fluorescent labeling.

Abstract:1-{6-(2-Methylbenzoyl)- N -ethylcarbazole-3-yl}-ethane-1-one oxime O -acetate, an excellent two-photon carbazole photoinitiator, was synthesized with moderate yield (76.9%) starting from the successive o -methyl benzoylation and acetylation of N -ethylcarbazole, followed by oximation and esterification in the article. The structure was confirmed by 1 H-NMR, 13 C-NMR, and IR. The by-product acetic acid in the neutralization of hydroxylamine hydrochloride is effectively utilized in esterification with the aid of the one-pot process. In particular, the phase transfer technique is applied to overcome the deficiency of low reaction rate caused by heterogeneous reaction conditions in esterification.

Abstract:A copper‐catalysed (diacetoxyiodo)benzene‐promoted aerobic esterification reaction of acetoacetamides was developed for the synthesis of oxamates, which are useful precursors in synthetic organic chemistry. This practical and mild synthetic approach proceeded at 25 °C under open‐air conditions and afforded methyl 2‐oxo‐2‐(phenylamino)acetates in good to excellent yields combined with C−C σ‐bond cleavage and formal oxidative C−H bond functionalization. A mechanism is proposed.

Abstract:Herein, we disclose a general and practical iminoarylation of alkenes by nickel-catalyzed reductive cross-coupling of unsaturated oxime esters with readily available aryl halides, providing an expedient approach for constructing pyrroline derivatives. The absence of organometallic reagents enables the reaction to occur under mild conditions with a broad substrate scope and good functional group tolerance. Moreover, other C-based electrophiles, including alkenyl, alkynyl and alkyl halides, or pseudohalides, were also competent substrates for this reaction.

Abstract:Use of oxime forming reactions has become a widely applied strategy for peptide and protein bioconjugation. The efficiency of the reaction and robust stability of the oxime product has led to the development of a growing list of methods to introduce the required ketone or aldehyde functionality site specifically into proteins. Early methods focused on site-specific oxidation of an N-terminal serine or threonine and more recently transamination methods have been developed to convert a broader set of N-terminal amino acids into a ketone or aldehyde. More recently, site-specific modification of protein has been attained through engineering enzymes involved in posttranslational modifications in order to accommodate aldehyde-containing substrates. Similarly, a growing list of unnatural amino acids can be introduced through development of selective amino-acyl tRNA synthetase/tRNA pairs combined with codon reassignment. In the case of glycoproteins, glycans can be selectively modified chemically or enzymatically to introduce aldehyde functional groups. Finally, the total chemical synthesis of proteins complements these biological and chemoenzymatic approaches. Once introduced, the oxime ligation of these aldehyde and ketone groups can be catalyzed by aniline or a variety of aniline derivatives to tune the activity, pH preference, stability and solubility of the catalyst. Abstract:We describe a simple and robust oxidation strategy for preparing N-terminal thiazolidine-containing peptide thioesters from peptide hydrazides. Abstract:The transformations of unactivated alkenes through C & xe001;C bond double cleavage are always attractive but very challenging. We report herein a chemoselective approach to valuable cyclic imides by a novel Cu-catalyzed geminal amino-oxygenation of unactivated C & xe001;C bonds. O-2 was successfully employed as the oxidant as well as the O-source and was incorporated into alkenyl amides via C & xe001;C bond cleavage for the efficient preparation of succinimide or glutarimide derivatives. Moreover, the present strategy under simple conditions can be used in the late-stage modification of biologically active compounds and the synthesis of pharmaceuticals, which demonstrated the potential application.

Abstract:The development of new catalytic systems that enable regio- and chemoselective construction of diversely functionalized oxazocines is an important topic in organic synthesis and pharmacochemistry. Herein, a novel Pd/Mengphos complex was designed and applied in a palladium-catalyzed high-order [4+4] cycloaddition of 2-substituted allylic carbonates to α,β-unsaturated imines, allowing facile access to versatile oxazocines in good yields with excellent b / l and Z / E selectivities (up to 92% yield and complete b / l and Z / E selectivities). The reaction exhibited a broad substrate scope, mild reaction conditions, and good functional group compatibility. In addition, an asymmetric version has also been tested, affording the desired oxazocines in moderate to good enantioselectivity. Abstract:Oxalic acid monothioester (OAM), an easily accessible and storable reagent, was reported herein as a thioester synthetic equivalent for palladium-catalyzed decarboxylative thiocarbonylation of organohalides and hydrothiocarbonylation of unsaturated carbon-carbon bonds at room temperature with high chemo- and regioselectivity. The reaction is applicable to the synthesis of cysteine-derived thioesters, thus allowing chemical modification of cysteine-containing peptides. Decarboxylation of OAM proceeds through oxidative addition of Pd(0) to the acyl-S bond, which accounts for the very mild reaction conditions.

Abstract:Thioacetals are important compounds because they can be considered as both useful protecting groups of carbonyl compounds in the synthesis of multi-functional complex molecules and acyl carbanion equivalents in C-C bond forming reactions. Since many reactions have been developed to prepare oximes from non carbonyl compounds, to lead to a novel and efficient method for thioacetal preparation, transthioacetalization of oximes has received more and more attention. Unfortunately, the transformation usually suffers from the use of harmful, odorous thiols which can lead to serious safety and environment problems. From the green chemistry point of view, an efficient and odorless transthioacetalization of oximes involving an environment friendly reagent is of great importance and necessity. In this work, using odorless and stable alpha-oxo ketene dithioacetals 1 as thiol equivalents, the thioacetalization reaction of oximes 2 were studied. In the reaction system of MeCOCl-EtOH (95%) or 4-dodecylbenzenesulfonic acid (DBSA)-H2O, the thioacetalization reaction were carried out in reflux temperature. It is noteworthy that the odor of thiols can not be perceived during either the reaction or workup.

Abstract:Nowadays, peptidomimetics are widely studied, being useful tools in drug discovery and medicinal chemistry. The coupling between a carboxylic acid with an amine to form a peptide bond is the most common reaction to obtain peptides/peptidomimetics. In this work, we have investigated an innovative metal‐free photoredox‐catalyzed carbamoylation to form peptidomimetics thanks to the reaction between dihydropyridines functionalized with amino acids (or peptide sequences) and differently functionalized imines. As the organic photocatalyst, we used 4CzIPN, a donor‐acceptor cyanoarene vastly used in photoredox catalysis. By easily modulating the amino acid (or peptide sequence), which is directly attached to the dihydropyridine, we proposed this key‐reaction as new valuable method to obtain peptidomimetics, in situ building the not‐natural portion of the sequence. Moreover, we successfully employed this methodology in solid phase peptide synthesis, both inserting the new photoredox‐generated amino acid at the end or in the middle of the sequence. Peptides with different lengths and secondary structures were prepared, proving the success of this approach, even in sterically hindered environment. Herein, to the best of our knowledge, we describe the first photocatalytic protocol which allows the building of the peptide backbone, with the possibility of simultaneously inserting a non‐coded amino acid in the sequence.

Abstract:An unprecedented copper-catalyzed oxidative dehydrogenative [3 + 2] annulation of O-acyl oximes containing α-C(sp3)-H bonds with α-amino ketones for the preparation of 3-aminopyrroles is developed. This method handles O-acyl oximes as both internal oxidants and three-atom-unit components combined with the copper catalyst to drive catalytically generated oxime nitrogen-centered radicals and oxidative dehydrogenation of the α-amino ketones to the key 2-iminoethan-1-one intermediates, enabling site-selective cleavage of the multiple α-C(sp3)-H bonds in O-acyl oximes and α-amino ketones to create two new bonds, including a C(sp2)-C(sp2) bond and a C(sp2)-N bond, between the carbon atoms and the nitrogen atom in a cross-selective fashion.

Abstract:A promising method for the synthesis of cyclic peptides through the direct aminolysis of peptide thioesters is presented. The cyclization step was carried out in a mixture of acetonitrile and 1.5 M aqueous imidazole solution with no observable oligomers. Studies on the N- and C-terminal residues show that the choice of C-terminal residue has a more significant effect on the success rate of cyclization than the choice at the N-terminal residue.

However, a notable limitation is the need for a substantial excess of the thiol reagent, restricting usage to simple thiols. Here, we introduce an adapted procedure that significantly enhances thioester production with just a minimal thiol excess, facilitating the use of advanced thiol nucleophiles.

Abstract:A simple and efficient approach for the synthesis of privileged oxime esters by employing donor-acceptor cyclopropane diesters (DACs) as one of the potential precursors is reported. The strategy involves Lewis acid catalyzed ring-opening of DACs, resulting in an open-chain intermediate followed by the base-mediated construction of the corresponding oxime esters in a one-pot reaction. Moreover, the process also features the synthesis of diethyl 4-(4-methoxyphenyl)azete-2,2(3 H )-dicarboxylate. Abstract:Oxazole is an important pharmacophore and exists in the backbone of many bioactive peptide natural products and peptidomimetics. Efficient methods for the synthesis and direct functionalization of complex oxazole-containing peptides are in high demand. Herein, we report the late-stage site-selective functionalization of oxazole-containing peptides via palladium-catalyzed d-C(sp(2))-H olefination of phenylalanine, tryptophan, and tyrosine residues. This strategy utilizes oxazole motifs as internal directing groups and provides access to oxazole-containing peptide macrocycles with bioactivities.

Abstract:Palladium- and rhodium-catalyzed selective reactions of 2H-azirines and N-(acyloxy)amides leading to alpha-amino oxime esters were described. The regioselectivities of the reactions are catalyst-controlled, and two different alpha-amino oxime esters were selectively obtained in high yields. These reactions involved the construction of new N-O and C-N bonds, which are characterized by 100 % atom economy, good to excellent yields, and mild reaction conditions.
