Biosynthesis of Platinum Nanoparticles: Evaluation of Their Activity Against Streptococcus mutans and In Silico Study

Abstract:This work looks at the production, description, and biological assessment of platinum nanoparticles that were created using an ablation pulse laser, an effective physical strategy for creating nanotechnology, especially when solids are ablated in liquid conditions. This process yields highly pure nanoparticles and is far less complicated than chemical approaches. The goal of this work was to create novel platinum nanoparticles (PtNPs) with distinct biological properties by Nd-YAG (1064 nm) laser pulses by striking a platinum plate in double deionized water (DDW) and its role in anti-biofilm against Streptococcus mutans bacteria which cause tooth decay. PtNP samples were characterized using, UV/Vi spectrophotometer, transmission electron microscopy (TEM), X-ray diffraction, atomic force microscopy (AFM), and Fourier-transform infrared (FTIR) spectroscopy. Results show TEM the nanoparticles were spherical shaped with a size distribution 0 to 45 nm, the average nanoparticle size was 21 nm, while XRD analysis of PtNPs showed diffraction peaks at 40.01°, 46.28°, and 66.50° corresponding to the (111), (200), and (220). To evaluate the biological activity, the PtNPs possess significant antibacterial activity against S. mutans ; in comparison with antibiotic (amoxicillin) and ant biofilm activity, PtNPs were stronger than the ant biofilm activity of amoxicillin along, as well as the PtNP nanoparticles, and were potent than the amoxicillin preventing the formation of biofilm by crystal violet. This consequence was confirmed by studied the result of PtNPs in microbial biofilm creation in dental bridges maked ceramic using atomic force microscopy (AFM) method. The amoxicillin and PtNPs showed up exceptional inhibitory properties against microbial isolates, with its inhibition zone diameters attainment 22.25 ± 0.30, in S. mutans . The antioxidant abilities of amoxicillin and PtNPs were tested by their DPPH assay. A therapeutic model design which exhibited the lowest binding energy of − 0.73 kcal/mol and the lowest inhibition constant (Ki) of 289.63 mM. This binding energy indicates a weak interaction between the PtNPs and the target receptor, resulting in a delicate inhibition effect with S. mutans bacteria which cause tooth decay. The findings of this study demonstrate the potential of laser-synthesized PtNPs as a promising therapeutic agent for the management of dental caries and related oral health issues, warranting further investigation and development.
