Abstract:Current report is paramount contribution via nanotechnology to the existing remedies of health diseases. The lag in application of capped metallic oxide nanoparticles in restorative dentistry exist which is covered by this promising study. The uncapped and chitosan encapsulated ZnO nanoparticles were fabricated by facile co-precipitation method, and characterized using various biophysical strategies including X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), Scanning electron microscopy (SEM), Transmission electron microscopy (TEM) and Energy dispersive x-ray (EDX). ZnO nanoparticles and ZnO-Citosan nanoparticles were estimated to be <30 nm and <25 nm in size on respective basis. Significant in vitro antibacterial, antioxidant, cytotoxic and antidiabetic activity of ZnO nanoparticles has been elucidated that is enhanced by capping with chitosan polymer. 90% cytotoxicity against brine shrimps, 69.6% antidiabetic activity against α-amylase, and noteworthy antioxidation power by chitosan decorated ZnO nanoparticles has been effectively illustrated. Furthermore, the effective secondary caries remediation approach has been established by an amalgamation of ZnO nanoparticles and ZnO-Chitosan nanoparticles into dentine bonding agents. A remarkable reduction in Streptococcus mutans and Lactobacillus acidophillus strains has been observed, in-specific boosted by chitosan capped ZnO nanoparticles reinforced dental adhesive discs. Additionally, augmented mechanical properties, greater resistance to water sorption and solubility, notably high release profile, and slight variation of shear bond strength values have been obtained. In short, the prepared nanoparticles reported are detected to be auspicious theranostic agents for combating wide array of human pathogens in healthcare system.

Abstract:Dental caries is a major lifestyle concern as dental components affect the face of an individual. The issue of tooth decay occurs in every age group throughout the globe. Researchers are probing incipient implements and techniques to develop filling agents for decayed teeth. Zinc oxide (ZnO) powder is utilized mostly as a filling agent. Nanotechnology enhanced the efficiency of compounds of metal oxides utilized for dental caries. The present study aims to investigate the properties of ZnO nanoparticles (NPs) synthesized chemically (using ZnCl2 and NaOH) as well as biologically (using aqueous leaf extract of Murraya paniculata). The XRD patterns confirm that ZnO NPs have a hexagonal crystalline structure with particle sizes of 47 nm and 55 nm for chemically and biologically synthesized NPs, respectively. The FE-SEM data confirm the nanorod and spherical/cubical shape morphologies for the chemically and biologically synthesized ZnO NPs, respectively. FTIR data show the peaks between 4000 and 450 cm−1 of the functional groups of –OH, C-O, –C-H-, and Zn-O bonds. The UV–Vis absorption study indicates a peak around 370 nm and a hump around 360 nm corresponding to the chemically and biologically synthesized ZnO NPs, respectively. An antibacterial bioassay was performed and compared with commercially available ZnO bulk powder against tooth decaying pathogens, viz., Streptococcus mutans, Staphylococcus aureus, E. coli, and Lactobacillus fermentum, and found that both ZnO NPs had results closer to those of the standard drug (rifampicin). Thus, the synthesized ZnO NPs may be utilized as nano-drugs for the application of tooth decaying filling agents. Even biologically synthesized ZnO NPs may be considered more environmentally friendly and less toxic to human health concerns.

Abstract:The medical application of chitosan (Cs) has been for about half a century, but the molecular mechanism has not been elucidated yet. This study is to explore the antibacterial mechanism of chitosan nanoparticles (Csnp) in dentin at the atomic resolution level. Extracted tooth specimen was prepared in three groups: A. control group; B. Csnp treatment under ultrasonic agitation (UA); C. Csnp treatment without UA. A scanning electron microscope (SEM) was used to observe the Csnp distribution on the dentin surface. The incubations of Enterococcus faecalis ( E. faecalis ) were performed. Further, we explored the protection mechanism of chitosan polymers to collagen type I, using molecular docking technique and crystal structure superimposition analysis. We revealed that Csnp under UA was evenly distributed on the dental surface and the Csnp-pretreated dentin had great antibacterial activity for E. faecalis . Our work demonstrated that Csnp occupied the grooves of the triple-helical collagen surface, strengthened by crosslinking, and interfered with the bond of collagen adhesin through steric hindrance effect and interrupting hydrophobic interaction. Csnp protects dentin against E. faecalis by interacting and crosslinking with collagen type I and prevents bacterial collagen adhesin binding through steric hindrance effect and interrupting hydrophobic interaction. Abstract:Dental caries is a chronic, progressive disease caused by plaque, influenced by multiple factors and can damage the hard tissues of the teeth. In severe cases, it can also lead to the onset and development of other oral diseases, seriously affecting patients' quality of life. The creation of effective biomaterials for the prevention and treatment of dental caries has become one of the relentless goals of many researchers, with a focus on inhibiting the production of cariogenic plaque and retaining beneficial bacteria, guiding and promoting the reconstruction of dental hard tissues, and delaying the progression of existing caries. Chitosan is a natural cationic polymer extracted from the shells of crustaceans and shellfish. Since its discovery, chitosan has shown to have various biological functions such as antibacterial, biomimetic mineralization, drug delivery, etc. , making it one of the most promising biopolymers for new caries prevention and materials of prostheses. Therefore, this article provides an overview of the anti-caries applications of chitosan, which mainly covers the basic research on the application of chitosan in caries prevention and treatment since 2010, with a focus on categorizing and summarizing the following characteristics of chitosan as a caries prevention material, including its antibacterial effect, biomimetic mineralization effect and delivery ability of caries prevention drugs and vaccines. It also explores the limitations of current research on chitosan as a caries prevention biomaterial and the difficulties that need to be focused on and overcome in the future to provide theoretical reference for the clinical implementation of chitosan as a caries prevention biomaterial.

Abstract:Background: Dental caries is the most prevalent bacterial biofilm-induced disease. Current clinical prevention and treatment agents often suffer from adverse effects on oral microbiota diversity and normal tissues, predominately arising from the poor biofilm-targeting property of the agents. Methods: To address this concern, we herein report dual-sensitive antibacterial peptide nanoparticles pHly-1 NPs upon acid and lipid-binding for treatment of dental caries. Amino acid substitutions were performed to design the peptide pHly-1. The potential, morphology and secondary structure of pHly-1 were characterized to elucidate the mechanisms of its pH and lipid sensitivity. Bacterial membrane integrity assay and RNA-seq were applied to uncover the antimicrobial mechanism of peptides under acidic condition. The in vitro and ex vivo antibiofilm assays were used to determine the antibiofilm performance of pHly-1 NPs. We also carried out the in vivo anti-caries treatment by pHly-1 NPs on dental caries animal model. Oral microbiome and histopathological analyses were performed to assess the in vivo safety of pHly-1 NPs. Results: The pHly-1 peptide underwent the coil-helix conformational transition upon binding to bacterial membranes in the acidic cariogenic biofilm microenvironment, thereby killing cariogenic bacteria. Under normal physiological conditions, pHly-1 adopted a β-sheet conformation and formed nanofibers, resulting in negligible cytotoxicity towards oral microbes. However, in acidic solution, pHly-1 NPs displayed reliable antibacterial activity against Streptococcus mutans, including standard and clinically isolated strains, mainly via cell membrane disruption, and also suppressed in vitro and human-derived ex vivo biofilm development. Compared to the clinical agent chlorhexidine, in vivo topical treatment with pHly-1 NPs showed an advanced effect on inhibiting rat dental caries development without adverse effects on oral microbiota diversity and normal oral or gastric tissues. Conclusion: Our results demonstrated the high efficacy of dual-sensitive antimicrobial peptides for the selective damage of bacterial biofilms, providing an efficient strategy for preventing and treating dental caries.
Dextran-Coated Iron Oxide Nanoparticles As Biomimetic Catalysts for Localized and Ph-Activated Biofilm Disruption.

Abstract:Biofilms are surface-attached bacterial communities embedded within an extracellular matrix that create localized and protected microenvironments. Acidogenic oral biofilms can demineralize the enamel-apatite on teeth, causing dental caries (tooth decay). Current antimicrobials have low efficacy and do not target the protective matrix and acidic pH within the biofilm. Recently, catalytic nanoparticles were shown to disrupt biofilms but lacked a stabilizing coating required for clinical applications. Here, we report dextran-coated iron oxide nanoparticles termed nanozymes (Dex-NZM) that display strong catalytic (peroxidase-like) activity at acidic pH values, target biofilms with high specificity, and prevent severe caries without impacting surrounding oral tissues in vivo. Nanoparticle formulations were synthesized with dextran coatings (molecular weights from 1.5 to 40 kDa were used), and their catalytic performance and bioactivity were assessed. We found that 10 kDa dextran coating provided maximal catalytic activity, biofilm uptake, and antibiofilm properties. Mechanistic studies indicated that iron oxide cores are the source of catalytic activity, whereas dextran on the nanoparticle surface provided stability without blocking catalysis. Dextran-coating facilitated NZM incorporation into exopolysaccharides (EPS) structure and binding within biofilms, which activated hydrogen peroxide (H2O2) for localized bacterial killing and EPS-matrix breakdown. Surprisingly, dextran coating enhanced selectivity toward biofilms while avoiding binding to gingival cells. Furthermore, Dex-NZM/H2O2 treatment significantly reduced the onset and severity of caries lesions (vs control or either Dex-NZM or H2O2 alone) without adverse effects on gingival tissues or oral microbiota diversity in vivo. Therefore, dextran-coated nanozymes have potential as an alternative treatment to control tooth decay and possibly other biofilm-associated diseases.
Bioinspired green synthesis of ZnO nanoparticles by marine-derived Streptomyces plicatus and its multifaceted biomedicinal properties

Abstract:The present study explores the bioinspired green synthesis of zinc oxide nanoparticles (ZnONPs) using marine Streptomyces plicatus and its potent antibacterial, antibiofilm activity against dental caries forming Streptococcus mutans MTCC and S. mutans clinical isolate (CI), cytotoxicity against oral KB cancer cells, hemolysis against blood erythrocytes and artemia toxicity. The bioinspired ZnONPs showed a distinctive absorption peak at 375 nm in UV-Vis spectra, the FT-IR spectra divulged the active functional groups, and XRD confirmed the crystalline nature of the nanoparticles with an average grain size of 41.76 nm. SEM analysis evidenced hexagonal morphology, and EDX spectra affirmed the presence of zinc. The ZnONPs exerted higher antagonistic activity against S. mutans MTCC (Inhibitory zone: 19 mm; MIC: 75 μg/ml) than S. mutans CI (Inhibitory zone: 17 mm; MIC: 100 μg/ml). Results of biofilm inhibitory activity showed a concentration-dependent reduction with S. mutans MTCC (15 %-95 %) more sensitive than S. mutans CI (13 %-89 %). The 50 % biofilm inhibitory concentration (BIC50) of ZnONPs against S. mutans MTCC was considerably lower (71.76 μg/ml) than S. mutans CI (78.13 μg/ml). Confocal Laser Scanning Microscopic visuals clearly implied that ZnONPs effectively distorted the biofilm architecture of both S. mutans MTCC and S. mutans CI. This was further bolstered by a remarkable rise in protein leakage (19 %-85 %; 15 %-77 %) and a fall in exopolysaccharide production (34 mg-7 mg; 49 mg-12 mg). MTT cytotoxicity of ZnONPs recorded an IC50 value of 22.06 μg/ml against KB cells. Acridine orange/ethidium bromide staining showed an increasing incidence of apoptosis in KB cells. Brine shrimp cytotoxicity using Artemia salina larvae recorded an LC50 value of 78.41 μg/ml. Hemolysis assay substantiated the biocompatibility of the ZnONPs. This study underscores the multifaceted application of bioinspired ZnONPs in dentistry.
Silica nanoparticles containing nano-silver and chlorhexidine respond to pH to suppress biofilm acids and modulate biofilms toward a non-cariogenic composition

Abstract:Objectives: Dental caries is caused by acids from biofilms. pH-sensitive nanoparticle carriers could achieve improved targeted effectiveness. The objectives of this study were to develop novel mesoporous silica nanoparticles carrying nanosilver and chlorhexidine (nMS-nAg-Chx), and investigate the inhibition of biofilms as well as the modulation of biofilm to suppress acidogenic and promote benign species for the first time. Methods: nMS-nAg was synthesized via a modified sol-gel method. Carboxylate group functionalized nMS-nAg (COOH-nMS-nAg) was prepared and Chx was added via electrostatic interaction. Minimal inhibitory concentration (MIC), inhibition zone, and growth curves were evaluated. Streptococcus mutans (S. mutans), Streptococcus gordonii (S. gordonii), and Streptococcus sanguinis (S. sanguinis) formed multispecies biofilms. Metabolic activity, biofilm lactic acid, exopolysaccharides (EPS), and TaqMan real-time polymerase chain reaction (RT-PCR) were tested. Biofilm structures and biomass were observed by scanning electron microscopy (SEM) and live/dead bacteria staining. Results: nMS-nAg-Chx possessed pH-responsive properties, where Chx release increased at lower pH. nMS-nAg-Chx showed good biocompatibility. nMS-nAg-Chx exhibited a strong antibacterial function, reducing biofilm metabolic activity and lactic acid as compared to control (p < 0.05, n = 6). Moreso, biofilm biomass was dramatically suppressed in nMS-nAg-Chx groups. In control group, there was an increasing trend of S. mutans proportion in the multispecies biofilm, with S. mutans reaching 89.1% at 72 h. In sharp contrast, in nMS-nAg-Chx group of 25 μg/mL, the ratio of S. mutans dropped to 43.7% and the proportion of S. gordonii and S. sanguinis increased from 19.8% and 10.9 to 69.8% and 56.3%, correspondingly. Conclusion: pH-sensitive nMS-nAg-Chx had potent antibacterial effects and modulated biofilm toward a non-cariogenic tendency, decreasing the cariogenic species nearly halved and increasing the benign species approximately twofold. nMS-nAg-Chx is promising for applications in mouth rinse and endodontic irrigants, and as fillers in resins to prevent caries.
Functional nanomaterials and their potentials in antibacterial treatment of dental caries

Abstract:Dental caries is one of the most common bacteria-related oral diseases, and the cariogenic bacteria play critical roles in its pathogenesis. Antibacterial therapy is regarded as an important approach for treating dental caries. However, the low local drug concentration and the potential generation of drug resistance limit the traditional antibacterial efficacy. Nanomaterials, such as polymeric nanoparticles and liposomes, have been widely used as functional carriers for drug delivery. The antibiotic-loaded nanomaterials have shown great potentials in enhancing the efficacy of antibacterial therapy. In addition, some functionalized nanomaterials, such as metal nanoparticles and graphene-based nanomaterials, can be used as direct antibacterial agents for physical antibacterial therapy, photothermal therapy and photodynamic therapy. Herein, we aim to provide a comprehensive understanding of dental caries and the related pathogenic bacteria, and summarize the recent progress in nanomaterials-based antibacterial treatment. Also, the antibacterial mechanisms and the concerns of nanomaterials in practical use are discussed.

The research was studying the effects of Nanoparticles of Zinc Oxide on Streptococcus mutans isolate from dental caries. In this research, Different ZnO NPs concentrations were studied as its anti-bacterial effects on Streptococcus mutans was isolated from dental caries cases. 27 Streptococcus mutans isolates were obtained, which is equivalent to 20% of the total number of 135 isolates. This nanomaterial was chosen because it has a high affinity for human cells and does not cause harm compared to other nanoparticles. The nanoparticles are those very fine materials that can be produced so that their dimensions or the dimensions of their grains range from (1-100) nanometers, and because of the small size of the nanomaterials, which made them behave differently from large-sized materials whose size exceeds 100 nanometers, and because of the small size of the particles. Nanoparticles, which gave them electrical and magnetic properties that differ from large particles of the same compounds. The well diffusion method and the MIC experiment were conducted, and the results were good, as in the well diffusion method the efficacy increased with increasing the concentration of ZnO NPs. In the MIC experiment, it was observed that MIC in the sixth tube, meaning that the MIC of ZnO NPs towards Streptococcus mutans equals 0. 312 mg / ml.
Synthesis of hybrid copper-chitosan nanoparticles with antibacterial activity against cariogenic Streptococcus mutans

Abstract:Hybrid nanoparticles (CuChNP) comprising of copper nanoparticles with a chitosan shell were synthesized. Antimicrobial properties of CuChNP were assessed against Streptococcus mutans (S. mutans), one of the main bacterium that causes tooth decay. Antibacterial activity of CuChNP against S. mutans was comparable to that of oral antimicrobial agents, such as chlorhexidine, and cetylpyridinium chloride. Particularly, CuChNP exhibited superior capacity to prevent the S. mutans growth on human tooth surface as well as disrupt and kill the bacterial cells in an established dental biofilm. Chitosan may interact with both tooth hydroxyapatite and bacterial cell wall, which improves the adherence of copper to the tooth surface and potentiates their anti-biofilm action. The antimicrobial properties exhibited by CuChNP could be useful for the future development of more effective treatments for the control of dental plaque biofilms.
Antimicrobial Effect of Copper Oxide Nanoparticles on Some Oral Bacteria and Candida Species

Abstract:Statement of problem: Acid producing bacteria including Streptococcus mutans and lactobacilli cause tooth demineralization and lead to tooth decay. Also, oral colonization of the species of Candida has been reported in many studies that are resistant to antifungal agents. Objectives: In this study, antibacterial and antifungal effects of nano-CuO were studied against some oral bacteria and yeast fungi. Materials and methods: The minimum inhibitory concentrations (MICs) of copper oxide nanoparticles (CuO NPs) for oral bacterial and fungal test strains were determined in 96-well microtiter plate technique. The agar diffusion test (ADT) was employed to assess the antifungal properties of nystatin. Results: The MIC50 value of CuO NPs was determined at the range of 1-10 µg/ml for S. mutans, < 1 µg/ml for L. acidophilus, and 10 µg/ml for L. casei. Higher concentrations of CuO NPs (100-1000 µg/ml) were effective on the bacterial cell growth, resulting in 100% reduction in the optical density in TSB medium. The cells of Candida albicans, C. krusei and C. glabrata were treated with CuO NPs and the results showed a decrease in fungal growth at a concentration of 1-1000 µg/ml in TSB medium. The MIC50 value of CuO NPs was determined 1000 µg/ml for three species of Candida. The diameter of growth inhibition zones of 1100 µg/ml nystatin was obtained 15-21 mm for clinical isolates of three species of Candida. Conclusions: With respect to the potential bactericidal activity of CuO NPs on various cariogenic bacteria examined in this study, these NPs could be introduce as a candidate control agent for preventing dental caries or dental infections. In our study, on the other hand, Nano copper oxide had a weak effect on the candida species.
Antimicrobial Effect of Copper Nanoparticles on Relevant Supragingival Oral Bacteria

Abstract:Copper nanoparticles (Cu NPs) show promise in dentistry for combating bacterial dysbiosis and tooth decay. Understanding their effects on commensal versus pathogenic bacteria is vital for maintaining oral health balance. While Cu NPs demonstrate antibacterial properties against various oral bacteria, including common pathogens associated with tooth decay, their impact on commensal bacteria requires careful examination. In our work, we analyzed three types of Cu NPs for their effects on the growth, viability, and biofilm formation of representative caries-associated and commensal oral bacteria. S. sanguinis showed high tolerance to all Cu NPs, while L. rhamnosus was highly sensitive. Oxide-Cu NPs exhibited a stronger inhibitory effect on pathobionts compared with commensal bacteria. Moreover, the biofilm formation of the key cariogenic bacteria S. mutans was reduced, with minimal negative effects on commensal species’ biofilm formation. All our results showed that CuO nanoparticles (CuO NPs) exhibit reduced toxicity toward commensal bacteria growth and development but have a strong impact on pathogens. This suggests their potential for targeted treatments against pathogenic bacteria, which could help in maintaining the balance of the oral bacterial community.

Well-Orientation Strategy Biosynthesis of Cefuroxime-Silver Nanoantibiotic for Reinforced Biodentine™ and Its Dental Application against Streptococcus mutans

Abstract:Dental caries results from the bacterial pathogen Streptococcus mutans (S. mutans) and is the maximum critical reason for caries formation. Consequently, the present study aims to evaluate the antibacterial activity of a newly synthesized nanoantibiotic-Biodentine formulation. The silver nanoparticles (ROE-AgNPs) were biosynthesized from the usage of Rosmarinus officinalis L. extract (ROE) and conjugated with cefuroxime to form Cefuroxime-ROE-AgNPs. Using Biodentine™ (BIOD), five groups of dental materials were prepared, in which Group A included conventional BIOD, Group B included BIOD with ROE-AgNPs, Groups C and D included BIOD with Cefuroxime-ROE-AgNPs at concentrations of 0.5% and 1.5% cefuroxime, respectively, and Group E included BIOD with 1.5% cefuroxime. The synthesized ROE-AgNPs or Cefuroxime-ROE-AgNPs were characterized for conjugating efficiency, morphology, particle size, and in vitro release. Minimum inhibitory concentration (MIC) of the cefuroxime, ROE-AgNPs, and Cefuroxime-ROE-AgNPs were additionally evaluated against cefuroxime resistant S. mutans, which furthered antibacterial efficacy of the five groups of dental materials. The UV-Visible spectrum showed the ROE-AgNPs or Cefuroxime-ROE-AgNPs peaks and their formation displayed through transmission electron microscopy (TEM), X-ray diffraction (XRD) pattern, and Fourier transforms infrared (FTIR) analysis. The end result of Cefuroxime-ROE-AgNPs showed conjugating efficiency up to 79%. Cefuroxime-ROE-AgNPs displayed the highest antibacterial efficacy against S. mutans as compared to cefuroxime or ROE-AgNPs alone. Moreover, the MIC of ROE-AgNPs and Cefuroxime-ROE-AgNPs was detected against S. mutans to be 25 and 8.5 μg/mL, respectively. Consequently, Cefuroxime-ROE-AgNPs displayed that a decrease in the MIC reached to more than three-fold less than MIC of ROE-AgNPs on the tested strain. Moreover, Cefuroxime-ROE-AgNPs/BIOD was employed as a novel dental material that showed maximum antimicrobial activity. Groups C and D of novel materials showed inhibitory zones of 19 and 26 mm, respectively, against S. mutans and showed high antimicrobial rates of 85.78% and 91.17%, respectively. These data reinforce the utility of conjugating cefuroxime with ROE-AgNPs to retrieve its efficiency against resistant S. mutant. Moreover, the nanoantibiotic delivered an advantageous antibacterial effect to BIOD, and this may open the door for future conjugation therapy of dental materials against bacteria that cause dental caries.

Abstract:AbstractThe aim of this study was to prepare zinc oxide nanoparticles from chitosan and Piper longum and to assess the antimicrobial activity. The chitosan is a biocompatible polymer and also used as a polymeric nanoparticle. P. longum is a flowering vine which is commonly used as a spice to season food. It is also being used as a traditional medicine that treats asthma, viral hepatitis, cough, and respiratory infections. Dental varnishes are usually applied on the tooth surface and are similar to fluoride varnishes that prevent tooth decay. The zinc oxide nanoparticles were prepared using the P. longum plant extract and color change was noted. The nanoparticle formation was confirmed using UV–Vis spectroscopy and the solution was centrifuged for 10 min and the nanoparticles were collected. The chitosan was prepared using 1% acetic acid with chemical reaction. In a centrifuge tube, 100 µL of prepared nanoparticles, with the addition of chemical ingredients the varnish, were prepared. The study showed Staphylococcus aureus to be very sensitive with a maximum zone of inhibition followed by Sterptococcus mutants, Enterococcus faecalis, and Candida albicans. P. longum- and chitosan-assisted nano zinc oxide–based dental varnishes will be a better choice for infections caused by S. aureus and S. mutans.

Abstract:Background: Dental caries is still a major public health problem. The use of fluoride is one of the most effective ways to prevent tooth decay. Objective: The purpose of this research was to investigate the effectiveness of fluoride entrapped in chitosan nanoparticles in vivo. Methods: Sodium fluoride was loaded in chitosan via ionic gelation of tripolyphosphate nanoparticles. Characterization of nanoparticles was investigated by using the zeta potential, size of particles, loading capacities, encapsulation efficiency, and Fourier Transforms Infrared Spectroscopy. Chitosan/ fluoride nanoparticles were fabricated by a method of fluoride/chitosan cross-linking with tripolyphosphate. Results: The size of nanoparticles was 219 nm. According to the zeta potential results, by adding sodium fluoride to chitosan/tripolyphosphate nanoparticles reducing the number of positive charges of chitosan, the result was diminished zeta potential from +30.8 mV to +14.9 mV. The optimum drug loading and percentage of entrapment efficiency were 70% and 30% respectively. Fourier transform infrared spectroscopy confirmed linked among tripolyphosphate, chitosan and fluoride nanoparticles. In vitro characterization of nanoparticles demonstrated higher fluoride uptake ability and smooth releasing profile. Conclusion: It is suggested that fluoride/chitosan nanoparticles synthesized in our study may be a promising means of delivering fluoride for the early prevention of tooth decay.

Abstract:These coincidences caught Zoghbi’s attention at first, but her desire to help Rett syndrome patients and their families exerted lasting effects on her career. Two years after first encountering patients with this disorder, Zoghbi sidelined her clinical career and formally trained as a researcher in genetics in the hopes of understanding the causes of diseases like Rett syndrome. Nearly 20 years later, in 2004, she was elected to the National Academy of Sciences for her research on the molecular genetics of neurological disorders, including her 1999 discovery of the gene responsible for Rett syndrome. Zoghbi’s Inaugural Article (2), published in a recent issue of PNAS, continues her explorations of the disease’s mechanisms.

Dental caries is one of the most prevalent human diseases resulting from tooth demineralization caused by acid production of bacteria plaque. It remains challenges for current practice to specifically identify, intervene and interrupt the development of caries while restoring defects. In this study, inspired by natural dental plaque, a stimuli-responsive multidrug delivery system () has been developed to prevent tooth decay and promote enamel restoration. Classic spherical core-shell structures of micelles dual-loaded with antibacterial and restorative agents are self-assembled into bacteria-responsive multidrug delivery system based on the pH-cleavable boronate ester bond, followed by conjugation with salivary-acquired peptide (SAP) to endow the nanoparticle with strong adhesion to tooth enamel. The constructed specifically adheres to tooth, identifies cariogenic conditions and intelligently releases drugs at acidic pH, thereby providing antibacterial adhesion and cariogenic biofilm resistance, and restoring the microarchitecture and mechanical properties of demineralized teeth. Topical treatment with effectively diminishes the onset and severity of caries without impacting oral microbiota diversity or surrounding mucosal tissues. These findings demonstrate this novel nanotherapy has potential as a promising biomedical application for caries prevention and tooth defect restoration while resisting biofilm-associated diseases in a controlled manner activated by pathological bacteria. A bifunctional nanocomplex with remineralizing and antibacterial activities to interrupt dental caries

Abstract:Dental caries result from an imbalance between the demineralization and remineralization of dental hard tissues, primarily caused by biofilm accumulation. According to the theory of interrupting dental caries, effective anticaries materials and techniques should possess both remineralizing and antibacterial properties. However, current anticaries materials fail to mimic the process of amelogenesis to achieve remineralization while inhibiting the adhesion of cariogenic bacteria and the formation of biofilms. In this study, silk fibroin (SF) loaded with benzalkonium chloride (BZC) successfully formed an SF-BZC composite. This composite stabilized amorphous calcium phosphate (ACP), creating an ACP@SF-BZC dual-functional nanocomplex with both remineralizing and antibacterial properties. ACP@SF-BZC demonstrated significant anti-adhesion and biofilm inhibitory effects against Streptococcus mutans and Streptococcus sobrinus. Moreover, compared to fluoride, ACP@SF-BZC significantly enhanced the remineralization of demineralized enamel surfaces, forming a stable remineralized layer with improved mechanical properties, both in vitro and in vivo. In summary, the dual-function ACP@SF-BZC nanocomplex, with its remineralizing and antibacterial effects, offers a promising alternative for preventing and arresting enamel caries.
